Pearl millet synergids from unpollinated pistils at 1.5–2 and 2.5–3 days postmaturity (dpm) were examined using transmission electron microscopy following antimonate fixation to precipitate loosely-bound calcium (Ca). With increasing age of synergids, the gap above filiform apparatus (FA) and the coalesced vacuoles in midchalazal core extended and merged. The FAs became compressed and precipitates along their common wall were dispersed. The matrix material in numerous small chalazal vacuoles changed from dense to flocculent. Precipitates in vacuoles appeared mainly as clumps without or with a halo in the dense matrix and mostly finely distributed in the flocculent matrix. Eventually, vacuoles became free of both matrix and precipitates. Precipitates bound to nucleus, nucleolus, and micropylar cytoplasm increased initially, but then seemed to decrease, while the nucleus became disorganized and the nucleolus disappeared. Precipitates in the embryo sac wall and nucellar cells also increased initially, but then decreased. At very late stages, egg apparatus and ES lost structural integrity and lacked precipitates. Differences in degeneration and Ca levels of sister synergids were smaller at 1.5–2 dpm than at 2.5–3 dpm. A logical and correlative scheme of degenerative events and Ca distributional changes occurring in pearl millet synergids from maturity to 2.5–3 dpm is presented. Calcium in the synergid cells and other regions of pearl millet ovaries   总被引:8,自引:2,他引:6  
Summary The synergids and other cells of mature, unpollinated pearl millet ovaries were investigated using: (1) freeze-substitution fixation in conjunction with scanning electron microscope observations and energy-dispersive X-ray microanalysis to localize total calcium (Ca) and other elements, and (2) antimonate precipitation to selectively localize loosely sequestered, exchangeable calcium (Ca++). In freeze-fixed ovaries, the synergid cells, ovary wall, nucellus, and other regions of the ovary displayed, respectively and relatively, extremely high, high, moderate, and low levels of Ca. In antimonate-fixed ovaries, Ca-containing antimonate precipitates exhibited similar distribution patterns. In ovaries fixed using the conventional 2% (w/v) antimonate in fixatives, the synergids were disrupted due to precipitate overload. In the ovary wall, precipitates were mainly located in the intercellular spaces. Some precipitates were observed at the micropyle and along the outer ovule integument, associated with diffuse extracellular material, and in the cell walls of nucellar cells proximal to the micropyle. Examination of precipitate distribution inside the synergids was possible in ovaries fixed using 0.5% (w/v) antimonate in the fixatives. Cytoplasmic organelles of all synergids examined exhibited variable states of disintegration. The amount of precipitates associated with the degenerated organelles appeared to be proportional to the degree of their degeneration. Distinct precipitates were localized in contiguous regions of the nucellar cells fused with the embryo sac, the micropylar half of the embryo sac wall, and the filiform apparatus. 用焦锑酸盐沉淀法对莴苣助细胞中的钙分布进行了观察。结果表明,开花前3天刚形成的助细胞中的钙颗粒很少:开花前2天助细胞壁中的钙颗粒增加;开花前1天助细胞珠孔端细胞壁加厚,其中积累了许多钙颗粒:开花当天助细胞珠孔端的丝状器中聚集了大量的钙颗粒。授粉后1h时两个助细胞的结构和钙分布发生差异,一个呈退化状,其中的钙颗粒明显增多,另一宿存助细胞中的钙分布与授粉前相似。去雄不授粉1天后两个助细胞均保持完好,且两助细胞中的钙分布没有明显差异,表明由花粉管引起一个助细胞中钙含量增加进而导致了助细胞退化。退化助细胞在卵细胞与中央细胞之间形成一薄层。助细胞退化后不同部位的钙颗粒呈现出与受精作用密切有关的变化:授粉后1h时,钙主要聚集在近合点端部位;授粉后2.5h卵细胞即将受精,这时许多细小的钙颗粒主要聚集在卵细胞与中央细胞之间的薄层中;授粉后4h精、卵细胞已融合,这时退化助细胞合点端的钙颗粒明显减少,而在其珠孔端又聚集了较多的钙。上述助细胞中的钙含量变化与吸引花粉管进入胚囊和促使精卵细胞融合密切有关。  相似文献

章丘大葱(Allium fistulosum L.cv.Zhangqiu)的卵器由1个卵细胞及2个助细胞组成,观察到不少卵器没有卵细胞,只有2个助细胞。卵细胞的核及大部分细胞质位于细胞的合点端,1个大液泡占据了细胞其他部位。卵细胞含有很多的核糖体及多聚核糖体、嵴明显的线粒体、粗面内质网、高尔基体具小泡,卵细胞似是一个活跃的细胞。细胞外被细胞壁,其合点端及侧方与助细胞共同壁不连续,助细胞有一较大的核,位于细胞膨大的部位,众多的小液泡遍布细胞中。核糖体及聚合核糖体、线粒体,粗面内质网及风心圆环状粗面内质丰富,高尔基体及小泡常见,反映了其活跃的代谢作用。助细胞合点端及侧方与卵细胞、中央细胞的共同壁不连续,与卵细胞共同壁含胞间连丝,壁不连续处,有不状多层膜结构伸入卵细胞质,显示助细胞可能对卵细胞提供营养,伟粉后,一个助细胞退化,宿存助细胞至随胚胚期尚存在,它经历了一个缓慢的退化过程,出现质壁分离,细胞质变稀,液泡扩大,细胞器逐渐减少,在椭形胚期,宿存助细胞核内的染色质及核仁消失,有细胞质侵入核内,因宿存助细胞壁变厚,细胞质出现现脂滴,宿存助细胞可能仍有合成功能,宿存助细胞壁出现若干无壁部位,细胞内的营养物质可能通过无壁部位向胚乳转运,供游离核胚乳及胚乳细胞化初期的发育。  相似文献

在野外居群调查的启示下,本文以组件观点对柳叶野豌豆复合种和歪头菜幼苗亚单位的时序变化与开花关系进行了分析。结果发现在柳叶野豌豆复合种栽培居群中存在打破物种间形体结构特征的个体,即在复叶由一对小叶组成的植株就已开花而进入生殖时期。另外,在歪头菜的野生居群中发现由三或四枚小叶组成复叶的个体,因此,我们推测这种形体结构的变化可能暗示着柳叶野豌豆复合种和歪头菜有着共同的祖先。  相似文献

The cytoskeletal organization of the embryo sac of tobacco (Nicotiana tabacum L.) was examined at maturity and during synergid degeneration, pollen-tube delivery and gamete transfer using rapid-frozen, freeze-substituted and chemically fixed material in combination with immunofluorescence and immunogold electron microscopy. Before fertilization, the synergid is a highly polarized cell with dense longitudinally aligned arrays of microtubules adjacent to the filiform apparatus at the micropylar end of the cell associated with major organelles. The cytoskeleton of the central cell is less polarized, with dense cortical microtubules in the micropylar and chalazal regions and looser, longitudinally oriented cortical microtubules in the lateral region. In the synergid and central cell, F-actin is frequently found at the surface of the organelles and co-localizes with either single microtubules or microtubule bundles. Egg cell microtubules are frequently cortical, randomly oriented and more abundant at the chalazal end of the cell; actin filaments are associated with microtubules and the cortex of the egg cell. At 48 h after pollination and before the pollen tube arrives, the onset of degeneration is evident in one of the two synergids: the electron density of cytoplasmic organelles and the ground cytoplasm increases and the nucleus becomes distorted. Although synergids otherwise remain intact, the vacuole collapses and organelles degenerate rapidly after pollen-tube entry. Abundant electron-dense material extends from the degenerated synergid into intercellular spaces at the chalazal end of the synergid and between the synergids, egg and central cell. Rhodamine-phalloidin and anti-actin immunogold labeling reveal that electron-dense aggregates in this region contain abundant actin forming two distinct bands termed coronas. 水稻(Oryza sativa L.)胚囊成熟时,卵细胞的合点端无细胞壁,核居细胞中部,细胞器集中在核周围,液泡分散于细胞周边区域。助细胞珠孔端有丝状器,合点端无壁,核位于细胞中部贴壁处,细胞器主要分布在珠孔端,液泡主要分布在合点端。开花前不久,一个助细胞退化。中央细胞为大液泡所占,两个极核靠近卵器而部分融合,细胞器集中在极核周围和靠近卵器处,与珠心相接的胚囊壁上有发达的内突。反足细胞多个形成群体,其增殖主要依靠无丝分裂与壁的自由生长,反足细胞含丰富活跃的细胞器,与珠心相接的壁上有发达的内突。开花后6小时双受精已完成,合子和两个助细胞合点端均形成完整壁。合子中开始形成多聚核糖体、液泡减小。退化助细胞含花粉管释放的物质,其合点端迴抱合子。极核已分裂成数个胚乳游离核,中央细胞中细胞器呈活化状态。反足细胞仍在继续增殖。讨论了卵细胞的极性、助细胞的退化、卵器与中央细胞间界壁的变化、反足细胞的分裂特点等问题。  相似文献

The ultrastructure and composition of the synergids of Capsella bursa-pastoris were studied before and after fertilization. The synergids in the mature embryo sac contain numerous plastids, mitochondria, dictyosomes and masses of ER and associated ribosomes. Each synergid contains a large chalazal vacuole, a nucleus with a single nucleolus and is surrounded by a wall. This wall is thickest at the micropyle end of the cell where it proliferates into the filiform apparatus. At the chalazal end of the cell the wall thins and may be absent for small distances. The pollen tube grows into one of the two synergids through the filiform apparatus and extends one-third the length of the cell before it discharges. Following discharge of the pollen tube, mitochondria and plastids of the tube can be identified in the synergid as can hundreds of 0.5 μ polysaccharide spheres liberated by the tube. The method by which the sperm or sperm nuclei enter the egg or central cell is not known although an apparent rupture was found in the wall of the egg near the tip of the pollen tube. The second synergid changes at the time the pollen tube enters the first synergid. These changes result in the disorganization of the nucleus and loss of the chalazal wall and plasma membrane. Eventually this synergid loses its identity as its cytoplasm merges with that of the central cell.  相似文献

Potassiam antimonate was used to localize Ca2+ in the micropyle and embryo sac of Brassica napus L. before and after pollination. To identify the nature of the pyroantimonate deposits, energy-dispersive X-ray microanalysis (EDXA) was employed and the deposits were proved to contain calcium pyroantimonate. Image processing system was employed to measure the volume density and the diameter of the deposits. Before and after pollination, calcium was more abundant in the exostome and endostome as compared with the other regions of the integuments, and was concentrated at the apoplast system, i.e. the intercellular matrix of the micropyle canal and the cell wall. Before pollination, each of the two sister synergids accumulated more calcium than the other embryo sac cells. Although the mean diameter of the deposits in the synergid was only two-thirds as that in the egg cell and central cell, the volume density of the deposits in the synergid was about 2.5 times and 1.9 times as that in the egg cell and the central cell respectively. The filiform apparatus and the nucleus had the most abundant calcium within a synergid. After pollination both sister synergids degenerated conspicuously and were characterized by much more deposited calcium (about 2.4 times more than before); and the diameter of the deposits decreased dramatically, which was less than one-third as before. The relationship between calcium distribution and synergid degeneration as well as its functions was discussed.  相似文献

Changes in actin organization in the living egg apparatus of Torenia fournieri from anthesis to post-fertilization have been investigated using microinjection and confocal microscopy. Our results revealed that the actin cytoskeleton displays dramatic changes in the egg apparatus and appears to coordinate the events of synergid degeneration, pollen tube arrival and gametic fusion during fertilization. Synergid degeneration occurs after anthesis and is accompanied by actin fragmentation and degradation. The actin cytoskeleton becomes organized with numerous aggregates in the chalazal end of the degenerating synergid, and some of the actin infiltrates into the intercellular gap between synergids, egg and central cell, forming a distinct actin band. An actin cap is present near the filiform apparatus after anthesis and disappears after pollen tube arrival. In the egg cell, actin filaments initially organize into a network and after pollination become fragmented into numerous patches in the cortex. These structures, along with the actin in the degenerating synergid and intercellular spaces form two distinct actin coronas during fertilization. The actin coronas vanish after gametic fusion. This is the first report of changes in actin organization in the living egg apparatus. The reorganization of the actin cytoskeleton in the egg apparatus and the presence of the actin coronas during fertilization suggest these events may be a necessary prelude to reception of the pollen tube and fusion of the male and female gametes. Studies of ultrastructure and ATPase localization of the mature embryo sac in Vicia faba L. show that the egg cell has no cell wall at thechalazal end, it has a chalazally located nucleus and a large micropylar vacuole. There are many nuclear pores in the nuclear membrane. The cytoplasm is restricted around the nucleus. Dictyosome and mitochondria are few. There are some starch grains and lipid grains in the egg cytoplasm. There are no obvious differences between two synergids. No cell wall is seen at the chalazal end either, but there are some vesicles which project to vacuole of the central cell and fuse with its vacuolar membrane. Plasmodesmata connections occur within the synergid wall where it is adjacent to the central cell. The synergid has a micropylarly located nucleus and a chalazal vacuole, the nucleus is irregularly shaped. The synergid cytoplasm is rich in organelles. The filiform aparatus is of relatively heterogeneous structure. The central cell is occupied by a large vacuole and its cytoplasm is confined to a thin layer along the empryo sac wall, but is rich in various organelles, starch grains and lipid bodies. Nucleolar vacuoles are often present two polar nuclei. The nuclear membranes of two polar nuclei have partly fused. ATPase reactive product was located obviously at the endoplasmic reticulum in cytoplasm of the egg cell and central cell. The embryo sac wall consists of different density of osmiophilic layer. There are some wall ingrowths in chalazal region of the embryo sac. The long-shaped and cuneate cells of chalazal region are peculiar. Special tracks of ATPase reactive products are visible at their intercellular space which may be related to transportation of nutrients.  相似文献

Ovules of Nicotiana tabacum L. were cryofixed with a propane-jet freezer and freeze-substituted in acetone to examine technique-dependent changes in pre- and post-fertilization embryo sacs using rapidly frozen material. Freezing quality was acceptable in 10% of the embryo sacs in the partially dissected ovules, with ice-crystal damage frequently evident in vacuoles and nuclei. One of the two synergids begins to degenerate before pollen-tube arrival in cryofixed material, with breakdown of the plasma membrane and large chalazal vacuole delayed until the penetration of the pollen tube. Early synergid degeneration involved characteristic increases in cytoplasmic electron density and the generation of cytoplasmic bodies to the intercellular space through “pinching-off”. Upon pollen-tube arrival, the male gametes are released through a terminal aperture into the degenerate synergid. Sperm cells undergo morphological alteration before gametic fusion: their mitochondrial electron density increases, the endoplasmic reticulum dilates, cytoplasm becomes finely vacuolated and the surrounding pollen plasma membrane is lost, causing the sperm cells and vegetative nucleus to dissociate. Discharge of the pollen tube results in the formation of numerous enucleated cytoplasmic bodies which are either stripped or shed from sperm cells and pollen-tube cytoplasm. Two so-called X-bodies are found in the degenerate synergid after pollen-tube penetration: the presumed vegetative nucleus occurs at the chalazal end and the presumed synergid nucleus near the micropylar end.  相似文献

The ultrastructure of synergids of watermelon (Citrullus Lanatus L.) was investigated using transmission electron microscopy at following stages of embryo sacs: 1. Unpollination, on the first flowering day. 2. Unpollination, on 2nd day after anthesis (DAA). 3. Fertilization, on DAA 2. The synergids with distinct filiform apparatus at the micropylar end have abundant organelle, such as mitochondria, endoplasmic reticulum, and plastids in cytoplasm, which indicate that they are active on the first flowering day. No wall is present at the chalazal part of synergid, and there are some flocculent materials and vesicles in the spaces of cytoplasma membranes among synergid, egg cell and central cell in embryo sacs at the first and the second stages. On DAA 2, in unpollinated embryo sacs, the central large vacuole of synergid is divided into several smaller ones and the starch grains decrease in cytoplasm. There is no newly synthesized wall at the chalazal end of persistent synergid in fertilized embryo sacs. The contents of degenerated synergid, in the form of electron dense granules, are located in the wide space among central cell, zygote and persistent synergid, and some of them migrate into central cell through cytoplasma membrane. Therefore, it is deduced that the contents of synergid might serve as a nutrient supplement to the development of endosperm, but not embryo.  相似文献

The mature embryo sac of barley consists of an egg, two synergids, a central cell, and up to 100 antipodal cells. At shedding the male gametophyte is 3-celled, consisting of a vegetative cell with a large amount of starch and two sperms having PAS+ boundaries. Before pollination the nucleus and cytoplasm of each synergid appear normal. After pollination the nucleus and cytoplasm of one synergid undergo degeneration. The pollen tube grows along the surface of the integument of the ovule, passes through the micropyle, and enters the degenerate synergid through the filiform apparatus. The pollen tube discharges the vegetative nucleus, two cellular sperms, and a variable amount of starch into the degenerate synergid. Soon after deposition the sperms migrate by an unknown mechanism to the chalazal end of the degenerate synergid. Sperm nuclei then enter the cytoplasm of the egg and central cell, ultimately resulting in the formation of the zygote and primary endosperm nucleus, respectively. Sperm boundaries do not enter egg or central cell, but it was not possible to determine the fate of other sperm components. Degenerate vegetative and synergid nuclei remain in the synergid after fertilization, constituting what are considered to be X-bodies in barley. With light microscopy maize megagametophytes stained with Alcian blue-periodic acid-Schiff (AB-PAS) reveal acid or neutral polysaccharides in various cell walls. Comparative fine structural studies were made of permanganate- or OsO4-fixed material. Organelle distribution is random in the vacuolate and multinucleate antipodal cells; organelles are abundant; starch is scarce. Antipodal cell walls have large openings forming several syncytia. Some walls are papillate. In the central cell (primary endosperm cell) a thin peripheral layer of cytoplasm surrounds the large vacuole; organelle number is moderate; starch is abundant. The central cell wall is also papillate adjacent to the antipodals and around the egg apparatus. In the synergids organelle distribution is non-random; nuclei and numerous organelles occupy the micropylar cytoplasm of each synergid; vacuoles dominate the chalazal cytoplasm of these cells. The filiform apparatus stains with AB-PAS and is composed of both lightly and darkly stained amorphous material. In the egg, organelle distribution is perinuclear with vacuoles proximal to the micropyle; mitochondria are large, abundant and polymorphic; starch is abundant. Nucleolar diameter is five times greater in the central cell and egg than in the antipodal cells and ten times greater than in the synergids. Plasmodesmata occur in all cell walls within the gametophyte, but none appear in the gametophyte wall itself. It is suggested that the antipodals and synergids might be secretory, the latter probably being involved in pollen tube attraction, and that stored metabolites in the central cell and egg cytoplasm support rapid increase in metabolism following fertilization.  相似文献

Summary Sister synergids of wheat were examined for post-pollination calcium distribution using freeze-substitution fixation, scanning electron microscopy, and energy-dispersive X-ray microanalysis. At 15–30 min postpollination (before pollen-tube penetration and discharge), the sister synergids were similar in size and in elemental concentrations. Both synergids measured about the same as the egg and contained relatively high concentrations of calcium, potassium, and magnesium. The sister synergids remained identical even in an aberrant case where a pollen tube had penetrated, but not discharged into a synergid. After 30 min post-pollination (after pollen-tube discharge), typically one of the sister synergids was reduced in size and devoid of high concentrations of elements. The other synergid initially remained unaltered and only later underwent slight size reduction; however, it retained high concentrations of elements. The results indicate that high concentrations of calcium, potassium, and magnesium are rapidly lost from a synergid following pollen-tube discharge. The structure of embryo sac before and after fertilization, embryo and endosperm development and transfer cell distribution in Phaseolus radiatus were investigated using light and transmission electron microscopy. The synergids with distinct filiform apparatus have a chalazal vacuole, numerous mitochondria and ribosomes. A cell wall exists only around the micropylar half of the synergids. The egg cell has a chalazally located nucleus, a large micropylar vacuole and several small vacuoles. Mitochondria and plasrids with starch grains are abundant. No cell wall is present at its chalazal end. There are no plasma membranes between the egg and central cell in several places. The zygote has a complete cell wall, abundant mitochondria and plastids containing starch grains. Both degenerated and persistent synergids migh.t serve as a nutrient supplement to proembryo. The wall ingrowths occur in the central cell, basal cell, inner integumentary cells, suspensor cells and endosperm cells. These transfer cells may contribute to embryo nutrition at different developmental stages of embryo.  相似文献

Ultrastructure of the embryo sac lacking antipodals in prefertilization stages in Arabidopsis thaliana has been examined 2 hr before and 5 hr after manual cross pollination. The cytoplasm of both synergids before fertilization is rich in ribosomes, mitochondria, and rough endoplasmic reticulum, and also contains several microbodies and spherosomes. The filiform apparatus includes electron-dense material and a fibrous part. Many cortical microtubules appear in the filiform apparatus area. One of the two synergids degenerates before fertilization. The synergids, the egg cell, and central cell have a rich cytoskeleton of microtubules; only the synergids appear to contain microfilaments. At the chalazal end, the antipodals are initially present but degenerate by the time of pollination in most embryo sacs in the starchless line studied. The embryo sac is completely surrounded by a wall containing an electron-dense layer, separating it from the nucellus, including the chalazal end. When the antipodals have degenerated, the electron-dense layer disappears at the chalazal end only, and the wall between the central cell and the nucellus is homogeneous. Between the central cell and nucellar cells no plasmodesmata are found. The membranes of both antipodal cells at the chalazal end of the embryo sac appear sinuous, like those of transfer cells. The central cell has plastids preferentially distributed around the nucleus, but the other organelles are randomly distributed. The central cell in the embryo sac and the adjacent chalazal nucellar cells show a transfer-cell function in the embryo sac after the antipodals degenerate.  相似文献
