The central part of the mitochondrial coxII gene was amplified from 38 different dicots and two monocots using polymerase chain reaction. In 30 of the 40 plants studied, the amplified coxll gene-fragment contains an intron, ranging from 930 bp in Capsicum (pepper) in Solanaceae to 1,635 bp in Ampelamus albidans (climbing milkweed) in Asclepiadaceae. The composition of this intron varies as revealed by Southern hybridizations using oligonucleotide probes specific to the coxII intron-regions in maize, wheat, and rice. In the Apocynaceae, Calharanthus roseus (Madagascar periwinkle) and Vinca minor (common periwinkle) lack the coxII intron, while other members of the same family (various Mandevilla species, Nerium oleander and Apocynum cannabinum) and members of the closely related Asclepiadaceae (Asclepias incarnata, Ampelamus albidans and Asclepias tuberosa) retain the intron. Analysis of these data suggest a selective loss of the coxII intron from a plant, ancestral to both Catharanthus and Vinca, after the divergence of the Asclepiadaceae and Apocynaceae. The remaining eight plants from the Brassicaceae, Cucurbitaceae, Fabaceae, and Onagraceae lacking the intron fall into a single group or clade using the phylogenetic tree proposed by Chase et al. (Annals of the Missouri Botanical Garden: 80: 528–580, 1993) based on sequence of the chloroplast rbcL gene.  相似文献

The family Asclepiadaceae (Dicotyledones) was created by Brown in 1810 by splitting in two the family Apocynaceae of Jussieu established in 1789. The morphological characters used to make this distinction were mainly palynological, such as presence of tetrads or pollinia and number and orientation of pollinia. Those characters, still used in higher taxonomic delimitation (families, subfamilies, and tribes), are here critically reexamined and compared to a molecular phylogeny obtained with one of the more variable plastid genes (matK) of 46 species in the order Gentianales. In this molecular phylogeny, Asclepiadaceae form a monophyletic group derived from within Apocynaceae. Each of the subfamilies of Asclepiadaceae is monophyletic and based on reliable palynological characters, but palynological characters are not useful to delimit tribes of the subfamily Asclepiadoideae. Based on the molecular data, these tribes have undergone parallelisms in several reproductive traits.  相似文献

SYNOPSIS. A total of 4,866 plants belonging to the family Asclepiadaceae was examined in the eastern part of the United States; 291 (6%) of 11 species naturally infected with Phytomonas elmassiani; 9 of these were new host records. Eight of the 9 infected species belonged to the genus Asclepias, one to Cynanchum. Infections were recorded between 25.5° and 41.0° N latitude, and from 74.0° to 90.5° west longitude. At the southernmost latitude Asclepias curassavica, an exotic annual, perennates, harbors phytomonads and is infested the year round with Oncopeltus. Usually but not always infection is correlated with the presence and abundance of insect vectors. The annual die-back of the usually infected perennial plant hosts, the uninfected emerging vernal growth, the reservoir nidus of infected plants in the southern latitudes, and the known proclivity of O. fasciatus to migrate suggest the spread of flagellosis of Asclepiadaceae thru migration of the vector hemipteron.  相似文献

Plants of the Apocynaceae family produce a wide range of terpenoid indole alkaloids (TIAs) which have important pharmaceutical applications. Studies of the molecular mechanisms controlling TIA biosynthesis may eventually provide possibilities to improve product yield by genetic modification of plants or cell cultures. However, these studies suffer from the lack of transformation/regeneration protocols for Apocynaceae plants. We chose to study the feasibility of Agrobacterium tumefaciens-mediated transformation of Tabernaemontana pandacaqui, because of the availability of an efficient regeneration procedure for this member of the Apocynaceae family. A procedure to produce transgenic T. pandacaqui plants was established, albeit with low efficiency. Transgenic expression was demonstrated of an intron-containing β-glucuronidase reporter gene and of a gene coding for the TIA biosynthetic enzyme strictosidine synthase from Catharanthus roseus, another Apocynaceae species. Forty different medicinal plants were investigated for arbuscular mycorrhizal association in the Rajshahi University Campus in Bangladesh. The results indicated that 35 different plants were infected by AM (arbuscular mycorrhizal) fungi as found by trypan blue staining procedure. The percentage of root colonization by AM fungi varied from 13.3% to 100%. Mangifera indica and Morus indica have maximum percentage of colonization (100%). The intensity of root colonization were abundant in the plants belonging to the families Anacardiaceae, Asclepiadaceae, Moraceae, Leguminosae and Apocynaceae whereas the intensity of colonization of crop roots were moderate and poor belonging to Gramineae and Leguminosae. The presence of greater number of spore in soil was always associated with the incidence of abundant mycelia. In plant roots the formation of spore and mycelia was restricted by low pH. Number of mycorrhizal fungus spores ranged between 35 to100 per 100g air dried soil in different family respective soils. The frequency of mycorrhizal fungus infection showed positive correlation with soil pH, moisture, water holding capacity, texture, total nitrogen, organic carbon, phosphorus, calcium, potassium, and magnesium. Especially phosphorus and nitrogen in the soil greatly influenced the plant root infection by AM fungi.  相似文献

近年来的分子系统学把狭义萝藦科和狭义夹竹桃科合并为广义夹竹桃科,包括5个亚科和25个族,但亚科和族间的亲缘关系较为复杂,亟待多学科证据澄清。本文利用常规石蜡切片技术观察了马利筋亚科南山藤属中的中国特有植物苦绳(Dregea sinensis var. sinensis)的孢子发生和配子体发育,结合已有资料比较了5个亚科的胚胎学特征。结果表明:(1)苦绳的花药由一对侧生并列药室组成,各有一个花粉团。(2)花药壁有6层,由外至内分别为表皮、2层药室内壁、中层和2层绒毡层,花药壁发育模式属于多层型。(3)绒毡层细胞单核,排成2列,为腺质型; 在小孢子四分体形成时期,药室内壁发生明显纤维状加厚; 花药成熟时,位于药室远轴最外侧处的花药壁发生断裂,准备散粉。(4)小孢子母细胞减数分裂中,胞质分裂方式为连续型,小孢子四分体排列方式为左右对称; 成熟花粉粒为3-细胞型,排列紧密,形成花粉团。(5)雌蕊含有两枚离生心皮,具边缘胎座,胚珠倒生,单珠被,薄珠心,蓼型胚囊。本文观察到的这些胚胎学特征为牛奶菜族提供了新资料。同时,胚胎学特征在5个亚科间的区别和联系,支持广义夹竹桃科的成立。  相似文献

Tomato yellow leaf curl virus (TYLCV) is one of the most important plant viruses belonging to the genus Begomovirus of the family Geminiviridae. To identify natural weed hosts that could act as reservoirs of TYLCV, 100 samples were collected at a TYLCV-affected tomato farm in Iksan from 2013 to 2014. The sample weeds were identified as belonging to 40 species from 18 families. TYLCV was detected in 57 samples belonging to 28 species through polymerase chain reaction using root samples including five species (Eleusine indica, Digitaria ciliaris, Echinochloa crus-galli, Panicum dichotomiflorum, and Setaria faberi) from the family Poaceae. Whitefly Bemisia tabaci-mediated TYLCV transmission from TYLCV-infected E. indica plants to healthy tomatoes was confirmed, and inoculated tomatoes showed typical symptoms, such as leaf curling and yellowing. In addition, TYLCV was detected in leaf and root samples of E. indica plants inoculated by both whitefly-mediated transmission using TYLCV-viruliferous whitefly and agro-inoculation using a TYLCV infectious clone. The majority of mastreviruses infect monocotyledonous plants, but there have also been reports of mastreviruses that can infect dicotyledonous plants, such as the chickpea chlorotic dwarf virus. No exception was reported among begomoviruses known as infecting dicots only. This is the first report of TYLCV as a member of the genus Begomovirus infecting monocotyledonous plants.  相似文献

The insects that feed on the related plant families Apocynaceae and Asclepiadaceae (here collectively termed “milkweeds”) comprise a “component community” of highly specialized, distinctive lineages of species that frequently sequester toxic cardiac glycosides from their host plants for defense against predators and are thus often aposematic, advertising their consequent unpalatability. Such sets of specialized lineages provide opportunities for comparative studies of the rate of adaptation, diversification, and habitat-related effects on molecular evolution. The cerambycid genus Tetraopes is the most diverse of the new world milkweed herbivores and the species are generally host specific, being restricted to single, different species of Asclepias, more often so than most other milkweed insects. Previous work revealed correspondence between the phylogeny of these beetles and that of their hosts. The present study provides analyses of near-complete DNA sequences for Tetraopes and relatives that are used to establish a molecular clock and temporal framework for Tetraopes evolution with their milkweed hosts.  相似文献

The Asclepiadaceae, as traditionally defined, have repeatedly been shown to be an apomorphic derivative of the Apocynaceae. It has often been recommended that the Asclepiadaceae be subsumed within the Apocynaceae in order to make the latter monophyletic. To date, however, no comprehensive, unified classification has been established. Here we provide a unified classification for the Apocynaceae, which consists of 424 genera distributed among five subfamilies: Rauvolfioideae, Apocynoideae, Periplocoideae, Secamonoideae, and Asclepiadoideae. Keys to the subfamilies and tribes are provided, with lists of genera that (as far as we have been able to ascertain) are recognized in each tribe.  相似文献

Background  

Anther smuts of the basidiomycetous genus Microbotryum on Caryophyllaceae are important model organisms for many biological disciplines. Members of Microbotryum are most commonly found parasitizing the anthers of host plants in the family Caryophyllaceae, however they can also be found on the anthers of members of the Dipsacaceae, Lamiaceae, Lentibulariaceae, and Portulacaceae. Additionally, some members of Microbotryum can be found infecting other organs of mainly Polygonaceae hosts. Based on ITS nrDNA sequences of members of almost all genera in Microbotryaceae, this study aims to resolve the phylogeny of the anther smuts and their relationship to the other members of the family of plant parasites. A multiple analysis strategy was used to correct for the effects of different equally possible ITS sequence alignments on the phylogenetic outcome, which appears to have been neglected in previous studies.  相似文献

Although orchid bees (Apidae: Euglossini) are known as key pollinators in tropical ecosystems, knowledge of their floral sources is still scarce, especially for those species commonly found in urban environments. We aimed to identify the pollen, nectar and resin sources used by the widespread species Euglossa cordata in an urban area in south-eastern Brazil. The residual pollen from 81 brood cells of nine nests reactivated between October 2013 and December 2014 was acetolysed and analysed. A total of 50 pollen types belonging to 20 botanical families were identified in the samples. Pollen sources included species from seven families; five of them were plants with poricidal anthers (Bixaceae, Commelinaceae, Fabaceae, Melastomataceae and Solanaceae). The mass-flowering trees Handroanthus chrysotrichus and H. heptaphyllus (Bignoniaceae) are firstly reported as important pollen sources to an orchid bee species. Nectar was collected primarily from plants with long, tubular corolla as Acanthaceae, Apocynaceae, Bignoniaceae and Convolvulaceae. The vine species Dalechampia stipulacea (Euphorbiaceae) acted as a floral resin source. All pollen sources consisted of native plants whereas some exotic plant species were visited for nectar collection. Although nesting in an area encompassed by a high proportion of invasive plant species, Euglossa cordata females preferred to visit native plants to gather floral resources.  相似文献

Using variously enriched media, axenic cultures were obtained of Phytomonas spp. associated with Allamanda cathartica, Mandevilla scabra, and Rhabdadenia biflora (Apocynaceae), and Blepharodon nitidus (Asdepiadaceae) and Euphorbia hirta (Euphor-biaceae). Cultures were usually subcultured once every week for many weeks in succession. Attempts to grow the flagellates from Bonafousia tetrastachya (Apocynaceae), Cecropia palmata (Cecropiaceae), and Euphorbia hyssopifolia (Euphorbiaceae) were unsuccessful. The “phytomonads” in latex of different host plants showed morphological differences, whereas the cultured flagellates were similar in morphology whatever the host plant source or culture medium.  相似文献

Family Apocynaceae is an economically important family grown as ornamental plants and many wild species have medicinal uses as well. The aim of the present study was to understand the level and pattern of genetic variability among the selected individuals of Apocynaceae. For this purpose, three species of different genera of Apocynaceae, Thevetia peruviana, Alstonia scholaris and Catharanthus roseus, were collected from Rawalpindi and Quaid-i-Azam University forest, Islamabad. To evaluate the level of polymorphism within the species and members of different species, randomly amplified polymorphic DNA (RAPD) markers were used. A series of OPC RAPD primers were used; only six primers of OPC series gave amplification. Highest genetic variation at interspecific and intraspecific levels was shown by OPC 9 and the lowest polymorphism was observed in OPC 4. The data was analyzed by using software Statistica 5.5. In total 105 monomorphic and 272 polymorphic bands were produced from all primers. Therefore, out of 322 amplified products, 26% were monomorphic and 68% were polymorphic. Low genetic diversification was observed both at intraspecific and interspecific level. At the molecular level Alstonia scholaris and Catharanthus roseus (subfamily Plumerioideae) appeared in a group and Thevetia peruviana (subfamily Rauvolfoideae) formed another group, confirming the classification based on morphological characters.  相似文献

The larvae of the hawk moth, Nephele densoi Keferstein, feed on the leaves of Cryptostegia grandiflora R. Br. in Madagascar. Host-specificity tests showed that feeding by N. densoi is restricted to plants in the families Asclepiadaceae (subfamily Periplocoideae), Apocynaceae and Moraceae, but only plants in the subfamily Periplocoideae supported larval development. The comparatively wide host range contrasts with the narrow host range demonstrated for the Australian moth N. subvaria (Walker). The possible evolution of host plant specialization in this genus is discussed.  相似文献

Allamanda exhibits an unusual type of gynoecial development in which the two carpels are free at initiation, but fuse completely during development, resulting in a unilocular ovary with parietal placentation at maturity. Whereas the majority of the Apocynaceae are characterized by an advanced type of gynoecium that is secondarily apocarpous, in Allamanda gynoecial evolution has proceeded one step further to secondary syncarpy. This condition is not known to occur in any other genus in the Apocynaceae and provides further evidence of the isolated position of Allamanda within the family.  相似文献

A new papain-like cysteine peptidase isolated from latex of Philibertia gilliesii Hook. et Arn., Apocynaceae (formerly Asclepiadaceae) has been purified and characterized. The enzyme, named philibertain g I, is the most basic component present in latex extracts and was purified by acetone fractionation followed by cation exchange chromatography (SP-Sepharose HR) using FPLC system. Homogeneity was confirmed by SDS-PAGE and mass spectroscopy (MS). Molecular mass of the enzyme was 23,530 Da (MALDI-TOF MS), its isoelectric point was >10.25, and maximum proteolytic activity (casein) was achieved at pH 7–8. The new protease was inhibited by E-64 a cysteine peptidases inhibitor. Km was 0.15 mM, using PFLNA as substrate. The N-terminal sequence of philibertain g I (LPASVDWRKEGAVLPIRHQGQCG) was compared with those of twenty plant proteases. Philibertain g I showed the higher degree of identity (73%) with caricain, one of the Carica papaya endopepetidases.  相似文献

The bushhopper Phymateus leprosus (Fabricius) in the field shows a special appetite for the milkweed Asclepias fruticosa. Asclepiadaceae, like Apocynaceae and Scrophulariaceae, contain cardiac glycosides. Raw and purified extracts of these plants phagostimulate larval and adult P. leprosus. We also screened natural and half-synthetic compounds found in those plant extracts. While saponins and sapogenins did not stimulate the animals, many cardiac glycosides and aglycones, offered on filter paper, proved to be phagostimulants.  相似文献

Twenty-nine laticiferous taxa of Apocynaceae, Asclepiadaceae, and Sapotaceae were screened for suitability as alternative sources of renewable energy, rubber, and phytochemicals and to select the most promising ones for large-scale cultivation. Of these,Allamanda violacea (14.9% protein, 13.8% polyphenol, 8.6% oil, 3.2% hydrocarbon),Catharanthus roseus (15.4% protein, 10.4% polyphenol, 11.5% oil, 1.9% hydrocarbon), andHolarrhena antidysenterica (14.2% protein, 16.4% polyphenol, 5.4% oil, 4.8% hydrocarbon) of Apocynaceae;Asclepias curassavica (19.3% protein, 6.5% polyphenol, 3.9% oil, 2.0% hydrocarbon), Calotropis gigantea (18.5% protein, 6.8% polyphenol, 7.0% oil, 2.8% hydrocarbon) of Asclepiadaceae;Mimusops elengi (11.3% protein, 9.7% polyphenol, 7.2% oil, 4.0% hydrocarbon) of Sapotaceae show promising potential for future petrochemical plantations; of all these taxa,Holarrhena antidysenterica yielded an unusually high percentage (4.8%) of hydrocarbon fraction followed byMimusops elengi (4.0%). NMR spectra confirmed the presence of cis-polyisoprene in all species studied exceptNerium indicum (white-flowered var.). These data indicate that the majority of the species under investigation may be considered for large-scale cultivation as an alternative source of rubber, intermediate energy, and other phytochemicals.  相似文献

RNA介导的基因沉默是近年来在生物体中发现的一种基于核酸水平高度保守的特异性降解机制.病毒诱导的基因沉默(virus induced gene silencing, VIGS)是指携带植物功能基因cDNA的病毒在侵染植物体后,可诱导植物发生基因沉默而出现表型突变,进而可以研究该目的基因功能.至今,已经建立了以RNA病毒、DNA病毒、卫星病毒和DNA卫星分子为载体的VIGS体系,这些病毒载体能在多种寄主植物(包括拟南芥、番茄和大麦)上有效抑制功能基因的表达.VIGS已开始应用于N基因和Pto基因介导的抗性信号途径中关键基因的功能研究、抗病毒相关的寄主因子研究以及植物代谢和发育调控研究.在当前植物基因组或EST序列大量测定的情况下,VIGS为植物基因功能鉴定提供了有效的技术平台.  相似文献
