目的建立简便的人乳腺癌裸鼠移植模型,并探讨其部分生物学特性。方法采用雌激素受体阴性的MDA-MB-231和SK-BR-3人乳腺癌细胞株,分别接种于10只裸鼠左侧腋窝皮下,移植细胞总数为1×107/只。观察肿块生长情况,第42天处死荷瘤鼠,切除肿块作病理切片。结果 MDA-MB-231接种后第5d在接种部位可见结节,成瘤率为90%(9/10),接种42 d肿瘤体积426.6±333.8,瘤重0.417±0.276,病理学检查为浸润性导管癌;SK-BR-3接种后第11天在接种部位可见结节,成瘤率为80%(8/10),接种42 d肿瘤体积357.5±246,瘤重0.325±0.167,病理学检查为浸润性导管癌。结论该方法建立的人乳腺癌裸鼠移植模型,皮下移植方法简单,易于操作,成功率较高,肿瘤可部分保持人乳腺癌生物学特性,为研究人乳腺癌提供了重要工具。  相似文献

As neovascularization is essential for tumor growth and metastasis, controlling angiogenesis is a promising tactic in limiting cancer progression. Melatonin has been studied for their inhibitory properties on angiogenesis in cancer. We performed an in vivo study to evaluate the effects of melatonin treatment on angiogenesis in breast cancer. Cell viability was measured by MTT assay after melatonin treatment in triple-negative breast cancer cells (MDA-MB-231). After, cells were implanted in athymic nude mice and treated with melatonin or vehicle daily, administered intraperitoneally 1 hour before turning the room light off. Volume of the tumors was measured weekly with a digital caliper and at the end of treatments animals underwent single photon emission computed tomography (SPECT) with Technetium-99m tagged vascular endothelial growth factor (VEGF) C to detect in vivo angiogenesis. In addition, expression of pro-angiogenic/growth factors in the tumor extracts was evaluated by membrane antibody array and collected tumor tissues were analyzed with histochemical staining. Melatonin in vitro treatment (1 mM) decreased cell viability (p<0.05). The breast cancer xenografts nude mice treated with melatonin showed reduced tumor size and cell proliferation (Ki-67) compared to control animals after 21 days of treatment (p<0.05). Expression of VEGF receptor 2 decreased significantly in the treated animals compared to that of control when determined by immunohistochemistry (p<0.05) but the changes were not significant on SPECT (p>0.05) images. In addition, there was a decrease of micro-vessel density (Von Willebrand Factor) in melatonin treated mice (p<0.05). However, semiquantitative densitometry analysis of membrane array indicated increased expression of epidermal growth factor receptor and insulin-like growth factor 1 in treated tumors compared to vehicle treated tumors (p<0.05). In conclusion, melatonin treatment showed effectiveness in reducing tumor growth and cell proliferation, as well as in the inhibition of angiogenesis.  相似文献

目的:用人体胰腺癌细胞株,建立小鼠胰腺癌细胞株(pGHAM-1)移植性胰腺癌模型,并研究其生物学特性。方法:将pGHAM-1细胞培养后配成1×107/ml,取0.2mL接种于小鼠皮下。于接种后20、30、40天观察肿瘤的生长、转移及腹水量。接种40天后处死动物,解剖取出移植瘤,用游标卡尺测量肿瘤大小,再进行HE染色的组织病理学检查,免疫组织化学检测血管内皮细胞生长因子(VEGF)及肿瘤转移相关蛋白(nm23-H1)的表达。结果:分别于20、30、40天测量肿瘤体积为84.1±21.9 mm3,413.7±208.4 mm3,2187.3±1882.8 mm3,后者与10 d前比较差异均具有统计学意义(P<0.01)。HE染色结果显示肿瘤组织呈条索状或小梁状排列,肿瘤组织与正常组织交界处有少量淋巴细胞浸润,肿瘤组织中血管生成罕见。免疫组织结果显示VEGF和nm23-H1蛋白在肿瘤组织中均呈阴性表达。结论:异位裸鼠人胰腺癌移植瘤模型易复制,时间短,成功率高,为胰腺癌体内研究提供了理想的动物模型。  相似文献

目的建立一种操作简便、性能稳定的新生大鼠高氧肺损伤动物模型。方法①设计制造能自动控制氧浓度的高氧动物饲养舱。②将孕期满21 d出生的SPF级新生大鼠随机分成4组,即:高氧组Ⅰ(入高氧舱中饲养1~7 d),高氧组Ⅱ(入高氧舱中饲养14 d),以及相应的空气对照组Ⅰ、Ⅱ,每组14只。舱内氧浓度≥90%,每天23 h。计算肺系数,HE染色与病理学观察。结果高氧组与相应的对照组比较:高氧组大鼠体重轻,肺系数大,HE染色显示部分肺泡萎缩、肺间质及肺泡腔有水肿、出血,中性粒细胞浸润;肺泡发育明显滞后,辐射状肺泡计数明显少。结论本动物饲养舱性能稳定,操作简便;复制的新生大鼠的肺病理变化符合高氧肺损伤的改变。  相似文献

目的建立人子宫内膜异位症裸鼠模型。方法通过开腹手术方法将人子宫内膜组织块种植于裸鼠盆腹腔,观察其生长情况,并进行病理检查。结果成功建立人子宫内膜异位症裸鼠模型,种植病灶能保持原有内膜组织的形态结构,并可见血管生成明显。结论子宫内膜异位症裸鼠模型的建立是子宫内膜异位症早期临床研究的理想模型。  相似文献

Multiple myeloma (MM), a malignant plasma cell disease, remains incurable and novel drugs are required to improve the prognosis of patients. Due to the lack of the bone microenvironment and auto/paracrine growth factors human MM cells are difficult to cultivate. Therefore, there is an urgent need to establish proper in vitro and in vivo culture systems to study the action of novel therapeutics on human MM cells. Here we present a model to grow human multiple myeloma cells in a complex 3D environment in vitro and in vivo. MM cell lines OPM-2 and RPMI-8226 were transfected to express the transgene GFP and were cultivated in the presence of human mesenchymal cells and collagen type-I matrix as three-dimensional spheroids. In addition, spheroids were grafted on the chorioallantoic membrane (CAM) of chicken embryos and tumor growth was monitored by stereo fluorescence microscopy. Both models allow the study of novel therapeutic drugs in a complex 3D environment and the quantification of the tumor cell mass after homogenization of grafts in a transgene-specific GFP-ELISA. Moreover, angiogenic responses of the host and invasion of tumor cells into the subjacent host tissue can be monitored daily by a stereo microscope and analyzed by immunohistochemical staining against human tumor cells (Ki-67, CD138, Vimentin) or host mural cells covering blood vessels (desmin/ASMA).In conclusion, the onplant system allows studying MM cell growth and angiogenesis in a complex 3D environment and enables screening for novel therapeutic compounds targeting survival and proliferation of MM cells.  相似文献

目的建立人泌尿系肿瘤无限细胞系,为泌尿系肿瘤研究提供实验模型.方法无菌取下肿瘤标本后,将标本剪成大小约1.0mm3的组织块,在裸鼠右后肢皮下包埋,当皮下肿瘤块发生明显增殖并长到一定程度后,再行裸鼠体内传代两次,最后取下组织块进行原代培养.培养细胞传代超过20代后按建系标准[2]进行检测.结果共取40例标本,裸鼠体内传代F1代成功6例,F3代成功3例,该3例标本行原代培养后建成3个无限细胞系人肾透明细胞癌RCC-9863,人膀胱癌BC-6,人前列腺癌PC-98106,全部细胞传代1年以上,生长稳定,传代周期固定,其形态结构,分化程度与原发瘤保持一致,染色体形态仍为人类核型.结论裸鼠肿瘤皮下种植法是泌尿系肿瘤建系的一个较好方法.  相似文献

Squamous cell carcinoma of the cervix, highly prevalent in the developing world, is often metastatic and treatment resistant with no standard treatment protocol. Our laboratory pioneered the patient-derived orthotopic xenograft (PDOX) nude mouse model with the technique of surgical orthotopic implantation (SOI). Unlike subcutaneous transplant patient-derived xenograft (PDX) models, PDOX models metastasize. Most importantly, the metastasis pattern correlates to the patient. In the present report, we describe the development of a PDOX model of HER-2-positive cervical cancer. Metastasis after SOI in nude mice included peritoneal dissemination, liver metastasis, lung metastasis as well as lymph node metastasis reflecting the metastatic pattern in the donor patient. Metastasis was detected in 4 of 6 nude mice with primary tumors. Primary tumors and metastases in the nude mice had histological structures similar to the original tumor and were stained by an anti-HER-2 antibody in the same pattern as the patient’s cancer. The metastatic pattern, histology and HER-2 tumor expression of the patient were thus preserved in the PDOX model. In contrast, subcutaneous transplantation of the patient’s cervical tumors resulted in primary growth but not metastasis.  相似文献

Decorin, a member of the small leucine-rich proteoglycan gene family, exists and functions wholly within the tumor microenvironment to suppress tumorigenesis by directly targeting and antagonizing multiple receptor tyrosine kinases, such as the EGFR and Met. This leads to potent and sustained signal attenuation, growth arrest, and angiostasis. We thus sought to evaluate the tumoricidal benefits of systemic decorin on a triple-negative orthotopic breast carcinoma xenograft model. To this end, we employed a novel high-density mixed expression array capable of differentiating and simultaneously measuring gene signatures of both Mus musculus (stromal) and Homo sapiens (epithelial) tissue origins. We found that decorin protein core modulated the differential expression of 374 genes within the stromal compartment of the tumor xenograft. Further, our top gene ontology classes strongly suggests an unexpected and preferential role for decorin protein core to inhibit genes necessary for immunomodulatory responses while simultaneously inducing expression of those possessing cellular adhesion and tumor suppressive gene properties. Rigorous verification of the top scoring candidates led to the discovery of three genes heretofore unlinked to malignant breast cancer that were reproducibly found to be induced in several models of tumor stroma. Collectively, our data provide highly novel and unexpected stromal gene signatures as a direct function of systemic administration of decorin protein core and reveals a fundamental basis of action for decorin to modulate the tumor stroma as a biological mechanism for the ascribed anti-tumorigenic properties.  相似文献

利用RT-PCR方法从PHA活化的人外周血单个核细胞(PBMCs)中克隆hIL-17F基因,亚克隆至逆转录病毒载体pSIV-1,与辅助病毒载体pHIT456和pHIT60脂质体法共转染293T包装细胞,获得的成熟重组逆转录病毒(RV-hIL-17F)再感染SMMC-7721人肝癌细胞,并经G418筛选建立hIL-17F转基因肝癌细胞。PCR、RT-PCR和Westernblot结果表明hIL-17F基因在肝癌细胞中能成功整合、转录和表达。MTT和FCM结果表明hIL-17F不能改变SMMC-7721肝癌细胞的增殖活力和细胞周期,但ELISA结果表明其能明显下调肝癌细胞IL-6、IL-8和VEGF的表达。转基因肝癌细胞rhIL-17F表达上清具有抑制ECV304人脐静脉内皮细胞生长的作用。裸鼠皮下成瘤试验结果表明hIL-17F转基因肝癌细胞裸鼠致瘤能力明显减弱,VEGF和CD34表达降低,血管形成显著减少。hIL-17F可通过减少肿瘤血管形成显著抑制裸鼠人肝癌移植瘤的生长,为其进一步开展肿瘤血管靶向基因治疗和开发抗血管新药提供了一定的实验依据。  相似文献

Circulating tumor cells (CTCs) are the potential precursors of metastatic disease. Most assays established for the enumeration of CTCs so far–including the gold standard CellSearch—rely on the expression of the cell surface marker epithelial cell adhesion molecule (EpCAM). But, these approaches may not detect CTCs that express no/low levels of EpCAM, e.g. by undergoing epithelial-to-mesenchymal transition (EMT). Here we present an enrichment strategy combining different antibodies specific for surface proteins and extracellular matrix (ECM) components to capture an EpCAMlow/neg cell line and EpCAMneg CTCs from blood samples of breast cancer patients depleted for EpCAM-positive cells. The expression of respective proteins (Trop2, CD49f, c-Met, CK8, CD44, ADAM8, CD146, TEM8, CD47) was verified by immunofluorescence on EpCAMpos (e.g. MCF7, SKBR3) and EpCAMlow/neg (MDA-MB-231) breast cancer cell lines. To test antibodies and ECM proteins (e.g. hyaluronic acid (HA), collagen I, laminin) for capturing EpCAMneg cells, the capture molecules were first spotted in a single- and multi-array format onto aldehyde-coated glass slides. Tumor cell adhesion of EpCAMpos/neg cell lines was then determined and visualized by Coomassie/MitoTracker staining. In consequence, marginal binding of EpCAMlow/neg MDA-MB-231 cells to EpCAM-antibodies could be observed. However, efficient adhesion/capturing of EpCAMlow/neg cells could be achieved via HA and immobilized antibodies against CD49f and Trop2. Optimal capture conditions were then applied to immunomagnetic beads to detect EpCAMneg CTCs from clinical samples. Captured CTCs were verified/quantified by immunofluorescence staining for anti-pan-Cytokeratin (CK)-FITC/anti-CD45 AF647/DAPI. In total, in 20 out of 29 EpCAM-depleted fractions (69%) from 25 metastatic breast cancer patients additional EpCAMneg CTCs could be identified [range of 1–24 CTCs per sample] applying Trop2, CD49f, c-Met, CK8 and/or HA magnetic enrichment. EpCAMneg dual-positive (CKpos/CD45pos) cells could be traced in 28 out of 29 samples [range 1–480]. By single-cell array-based comparative genomic hybridization we were able to demonstrate the malignant nature of one EpCAMneg subpopulation. In conclusion, we established a novel enhanced CTC enrichment strategy to capture EpCAMneg CTCs from clinical blood samples by targeting various cell surface antigens with antibody mixtures and ECM components.  相似文献

目的建立整合素αvβ3高表达的黑色素瘤细胞肺转移模型。方法通过将不同数目的M21细胞经尾静脉接种裸鼠,适时处死后计数肺表面癌结节。通过实时定量PCR和明胶酶谱的方法比较肺转移灶细胞与亲代M21细胞差异。结果M21细胞1×10^6、2×10^6、5×10^6尾静脉接种裸鼠,均能形成肺转移灶,癌结节数目均值分别为:84±8、70±6、88±12,三组之间没有明显差异,阴性对照组未形成转移灶。M21肺转移灶细胞与亲代细胞相比,增殖增快,MMP-2活性增高,整合素αv和β3mRNA表达水平明显增高。结论M21细胞1×10^6经尾静脉接种裸鼠50d内即可100%成瘤。M21肺转移灶细胞具有更快的增殖能力,整合素αvβ3和MMP-2表达水平明显增高。本实验建立了稳定的肺转移模型,为黑色素瘤和整合素αvβ3的研究提供重要的动物模型。  相似文献

将人肺巨细胞癌PLA-801D裸鼠皮下移植瘤作为瘤源,建立了PLA-801D-AS裸鼠腹水鼠模型,其转移率及转移程度明显增高;以PLA-801D-AS腹水瘤模型的肺转移灶细胞进行裸鼠皮下连续传代,建立了肺转移率及程度高于PLA-801D的PLA-801DL皮下移植瘤株。本文从肿瘤的异质性以及肿瘤与宿主相互作用的角度讨论了其肺转移提高的原因。  相似文献

Tumor cell microenvironment defines cancer development, also in hepatocellular carcinoma (HCC). Hepatic stellate cells (HSCs) are believed to be the key contributors to tumor microenvironment in HCC, yet their precise role in cancer progression is still unclear. The aim of this study was to determine the effect of human HSCs on progression of HCC using a subcutaneous xenograft nude mouse model. Nude mice were stratified to receive subcutaneous injections of human HCC cell line HepG2 and human HSC line LX-2 (HepG2 + LX-2), HepG2 alone, LX-2 alone, or phosphate-buffered saline. Tumor growth was assessed by measuring tumor size. After 30 days, final tumor size, weight, and histology were assessed. Compared with mice that were only injected HepG2 cells, mice injected with HepG2 + LX-2 exhibited more rapid tumor growth, increased tumor size and weight, higher tumor cell numbers due to increased proliferation and reduced apoptosis, increased fibrotic bands containing LX-2 cells, and increased tumor angiogenesis. In conclusion, HSCs play a significant role in promotion of HCC growth.  相似文献

目的-建立人乳腺癌MDA-MB-231细胞株裸小鼠模型,研究其生物学特性,观察MDA-MB-231乳腺癌细胞在移植前后的形态学变化。方法-将人乳腺癌细胞MDA-MB-231接种于裸鼠腋窝处皮下,每3天测量肿瘤大小,第30天处死小鼠。肿瘤组织及相关脏器送病理切片。皮下肿瘤组织细胞及细胞株培养HE染色。结果-肿瘤生长较快,成功率为72%,病理检查符合人乳腺癌细胞特征。肿瘤组织细胞及培养细胞形态学未见显著差异。结论-人乳腺癌细胞株MDA-MB-231裸小鼠模型建立方法较简便,细胞形态无明显差异,且保持了人乳腺癌的生物学特性。  相似文献

Nipah virus (NiV) is a member of the genus Henipavirus (family Paramyxoviridae) that causes severe and often lethal respiratory illness and encephalitis in humans with high mortality rates (up to 92%). NiV can cause Acute Lung Injury (ALI) in humans, and human-to-human transmission has been observed in recent outbreaks of NiV. While the exact route of transmission to humans is not known, we have previously shown that NiV can efficiently infect human respiratory epithelial cells. The molecular mechanisms of NiV-associated ALI in the human respiratory tract are unknown. Thus, there is an urgent need for models of henipavirus infection of the human respiratory tract to study the pathogenesis and understand the host responses. Here, we describe a novel human lung xenograft model in mice to study the pathogenesis of NiV. Following transplantation, human fetal lung xenografts rapidly graft and develop mature structures of adult lungs including cartilage, vascular vessels, ciliated pseudostratified columnar epithelium, and primitive “air” spaces filled with mucus and lined by cuboidal to flat epithelium. Following infection, NiV grows to high titers (107 TCID50/gram lung tissue) as early as 3 days post infection (pi). NiV targets both the endothelium as well as respiratory epithelium in the human lung tissues, and results in syncytia formation. NiV infection in the human lung results in the production of several cytokines and chemokines including IL-6, IP-10, eotaxin, G-CSF and GM-CSF on days 5 and 7 pi. In conclusion, this study demonstrates that NiV can replicate to high titers in a novel in vivo model of the human respiratory tract, resulting in a robust inflammatory response, which is known to be associated with ALI. This model will facilitate progress in the fundamental understanding of henipavirus pathogenesis and virus-host interactions; it will also provide biologically relevant models for other respiratory viruses.  相似文献

目的:探讨分子靶向药物格列卫与多西紫杉醇联合对人乳腺癌细胞株MCF-7的凋亡及其裸鼠皮下移植瘤生长的影响。方法:采用流式细胞检测仪检测MCF-7细胞在格列卫与多西紫杉醇单独处理及共同处理条件下的凋亡率;建立人乳腺癌细胞株MCF-7裸鼠皮下移植瘤模型,观察格列卫与多西紫杉醇单独治疗组及联合治疗组移植瘤的生长,计算抑瘤率。结果:MCF-7细胞在格列卫与多西紫杉醇共同处理条件下的凋亡率(50.86%)远高于多西紫杉醇单独处理(22.06%)及同剂量格列卫组单独处理(8.13%)条件下的凋亡率;高剂量多西紫杉醇与格列卫联合组肿瘤质量与单独多西紫杉醇组比较,差异虽然没有显著性,但联合组抑瘤率高达99.55%,高于多西紫杉醇组(97.43%),q值为1.014;中剂量多西紫杉醇与格列卫联合组肿瘤质量与单独多西紫杉醇组比较,差异有高度显著性,联合组抑瘤率达96.53%,高于多西紫杉醇组(92.01%),q值为1.02;低剂量多西紫杉醇与格列卫联合组肿瘤质量与单独多西紫杉醇组比较,差异有高度显著性,联合组抑瘤率达68.20%,高于多西紫杉醇组(58.40%),q值为1.004。结论:格列卫与凋亡诱导剂多西紫杉醇共同处理MCF—7细胞能达到协同诱导凋亡的效果;高、中、低剂量的多西紫杉醇与格列卫联合对人乳腺癌细胞株MCF-7裸鼠移植瘤增殖的抑制具有相加作用。  相似文献

用人喉癌手术切除标本建立了裸鼠移植瘤模型,已传至13代。原代移植成功率为66.7%,潜伏期30~70d;鼠间传代移植成功率为100%,潜伏期14~19d,生长稳定。经光学显微镜检查,各代移植瘤组织结构与原人喉癌组织基本一致。电镜检查证明,具有人喉癌特征,癌细胞间有大量桥粒,细胞浆中可见张力原纤维,有的张力原纤维与桥粒相连。细胞表面有较大的指状突,核膜较规则,胞浆中线粒体较多。  相似文献
