We studied the diel expression pattern of PAX6 (a structural gene that is commonly involved in the eye development and photoreception of eye forming animals) and the effects of light and covering behavior on PAX6 expression in the sea urchin Strongylocentrotus intermedius. We confirmed that aphotic condition significantly reduced covering behavior in S. intermedius. The diel expression pattern of PAX6 was significantly different in S. intermedius under photic and aphotic conditions. The gene expression of PAX6 significantly deceased in covered S. intermedius both under natural light and in darkness. The present finding provides valuable insight into the probable link between covering and PAX6 expression of sea urchins. Further studies are required to investigate the detailed expression network of light detection involved genes in order to fully reveal the molecular mechanism of the light-induced covering behavior of sea urchins.  相似文献

In this study, we investigated effects of 24 weeks of food deprivation on covering behavior of Glyptocidaris crenularis and Strongylocentrotus intermedius. Neither G. crenularis nor S. intermedius significantly reduced covering behavior during both short- and long-term food deprivation. However, G. crenularis and S. intermedius had significantly different behavioral patterns over the 24 weeks. Covering behavior of G. crenularis greatly increased from the first to the second week, with decreasing reaction time and increasing covering ability, while the covering behavior of S. intermedius generally remained unchanged during the experiment of 24 weeks. As prolonged food deprivation does not significantly affect covering behavior of G. crenularis and S. intermedius, the present study highlights the innate nature and biological significance of covering behavior in sea urchins, providing new insights into its behavioral mechanism.  相似文献

研究了较长时间生活在3种不同实验遮蔽条件下海刺猬(Glyptocidaris crenularis)的遮蔽行为特点。结果表明:不同生活环境下海刺猬都保持着遮蔽行为。生活在以贝壳作为遮蔽材料环境下(遮蔽组)和以砖块作为掩蔽材料环境下(掩蔽组)的海刺猬初次遮蔽耗时要显著短于生活在无遮蔽(或掩蔽)材料环境下(空白组)的海刺猬(N=3,P<0.05)。3组海刺猬用于遮蔽的贝壳总数和有遮蔽行为的海刺猬总数都呈现先增加后趋于平稳的趋势。掩蔽组有遮蔽行为的海刺猬总数要显著多于遮蔽组和空白组(P<0.05),后两者差异不显著。3组海刺猬用于遮蔽的贝壳总数差异不显著(P>0.05)。海刺猬遮蔽时对两种贝壳(菲律宾蛤仔和贻贝)存在显著的选择差异(P<0.05)。生活环境中一段时间内遮蔽物的缺失并不会使其失去这种行为,但是会在一定程度上影响该行为的强度。因此,光照很可能是海刺猬遮蔽行为的一个进化压力,该行为也许只是作为一种避光策略。海刺猬对遮蔽材料具有显著的选择性,这可能与遮蔽材料自身特征和海刺猬的生理状态相关。  相似文献

海胆遮蔽行为对于海胆的生长和发育的影响目前还不清楚,在实验室条件下,研究了遮蔽行为对海刺猬(Glyptocidariscrenularis)摄食、生长和性腺性状的影响。实验设两个对照组和一个处理组:对照组为无任何遮蔽条件的空白组和以堆积砖块创造的黑暗空间来满足海胆的掩蔽行为的掩蔽组,遮蔽组则以菲律宾蛤仔(Ruditapes philippinarum)的贝壳作为遮蔽物来满足海胆的遮蔽行为。结果显示:遮蔽组的海胆周摄食量要显著大于掩蔽组(P<0.05),而空白组与遮蔽组和掩蔽组之间的差异不显著(P>0.05);各组海胆的存活率没有显著差异(P>0.05);空白组和遮蔽组海胆的壳径、壳高、体重要显著大于掩蔽组(P<0.05),而空白组和遮蔽组之间没有显著性的差异(P>0.05);空白组和遮蔽组的性腺湿重、性腺干重、性腺指数、壳湿重、壳干重、亚氏提灯湿重、亚氏提灯干重要显著大于掩蔽组(P<0.05),而空白组和遮蔽组之间没有显著性的差异(P>0.05);空白组的提灯指数要显著小于遮蔽组和掩蔽组(P<0.05),而遮蔽组和掩蔽组之间没有显著的差异(P>0.05)。结果证明,遮蔽和掩蔽行为对海刺猬生长及发育的影响是有差别的,掩蔽行为对海刺猬的生长与发育造成了负面影响,而遮蔽行为在一定的生活环境条件下不会对其生长造成影响。研究表明,海刺猬的遮蔽行为反映了其对外界不利环境条件的一种适应性,且这种行为对其生长和发育的影响与掩蔽行为有所不同。至于海刺猬遮蔽行为的进化选择压力,目前的研究还不足以下定论,需要进一步深入的研究。  相似文献

The skeleton of spines and tests of the species of sea urchins Strongylocentrotus intermedius, Mesocentrotus nudus, Scaphechinus mirabilis, and Echinocardium cordatum from the Sea of Japan is composed of a spongy stereom, consisting of calcite with a high content of magnesium. It was found that the tests and spines of the skeletons of sea urchins are composed of calcium–organic composite materials inlaid with other metals: Mg, Fe, Zn, and Rb. In the four species of sea urchins studied, the strength and other mechanical properties of the tests and spines differ and depend on the chemical composition and structural organization of their components. It was shown that the content of volatile substances correlates with their fragility or elasticity. It is revealed that the chemical composition of the tests of two species of the spherical sea urchins S. intermedius and M. nudus indicates significant differences between these two species of sea urchins.  相似文献

Many sea urchin genera exhibit cryptic covering behaviors. One such behavior has been documented in the sea urchin Tripneustes gratilla, and previous studies have theorized that this behavior serves as protection from UV radiation. However, other hypotheses have been presented such as protection from predators or added weight to help T. gratilla resist strong currents. A field study was conducted in October-November 2015 in Moorea, French Polynesia to assess urchin covering behavior in natural habitats. The study found that urchins partially underneath rocks covered more, and with more algae, than urchins totally underneath rocks. To test if this behavior was driven by light intensity, a series of 30-minute experimental trials were run on 10 individuals in bright and dim conditions. Individuals were given red and clear plastic, and percent cover of each was recorded. These tests were repeated once fifty percent of spines had been removed from the urchin, in order to determine whether spine loss affects T. gratilla covering behavior. The study found that urchins had a distinct preference for cover that best protects them from UV radiation. Spine loss did not significantly affect urchin ability to cover, and urchins with removed spines still preferred opaque cover. Additionally, covering behavior was mapped onto a phylogeny of echinoderms to determine how it might have evolved. Understanding urchin covering behavior more fully is a step towards an understanding of the evolution of cryptic behavior across species.  相似文献

The formation of sea urchin ‘barrens’ on shallow temperate rocky reefs is well documented. However there has been much conjecture about the underlying mechanisms leading to sea urchin barrens, and relatively little experimentation to test these ideas critically. We conducted a series of manipulative experiments to determine whether predation mortality is an important mechanism structuring populations of the sea urchin Heliocidaris erythrogramma in Tasmania. Tethered juvenile and adult sea urchins experienced much higher rates of mortality inside no-take marine reserves where sea urchin predators were abundant compared to adjacent fished areas where predators were fewer. Mortality of tagged (but not tethered) sea urchins was also notably higher in marine reserves than in adjacent areas open to fishing. When a range of sizes of sea urchins was exposed to three sizes of rock lobsters in a caging experiment, juvenile sea urchins were eaten more frequently than larger sea urchins by all sizes of rock lobster, but only the largest rock lobsters (> 120 mm CL) were able to consume large adult sea urchins. Tagging (but not tethering) juvenile and adult sea urchins in two separate marine reserves indicated that adult sea urchins experience higher predation mortality than juveniles, probably because juveniles can shelter in cryptic microhabitat more effectively. In a field experiment in which exposure of sea urchins to rock lobster (Jasus edwardsii) and demersal reef fish predators was manipulated, rock lobsters were shown to be more important than fish as predators of adult sea urchins in a marine reserve. We conclude that predators, and particularly rock lobsters, exert significant predation mortality on H. erythrogramma in Tasmanian marine reserves, and that adult sea urchins are more vulnerable than smaller cryptic individuals. Fishing of rock lobsters is likely to reduce an important component of mortality in H. erythrogramma populations.  相似文献

Nutritional attractiveness of the brown alga Laminaria japonica for the sea urchin Strongylocentrotus intermedius was studied. The composition of L. japonica was analyzed after one and two years of its life under natural conditions, in its seedlings, and in the alga partially degraded by natural factors. Substances extracted with various solvents were tested for the presence of inhibitors and activators of 1,3-β-D-glucanase, a digestive enzyme of the sea urchin. Ethanolic extract of freshly harvested L. japonica was found to suppress the enzyme activity. Substances present in ethanolic extracts of the alga after one or two years of its life cycle and in the alga, partly degraded by natural factors, activated the sea urchin enzyme. This fact is in agreement with earlier natural observations concerning the nutritional attractiveness of such L. japonica samples for Strongylocentrotus intermedius.  相似文献

Heat shock proteins (HSPs) expression is commonly used as indicators of cellular stress in animals. However, very little is known about either the expression patterns of HSPs or their role in the stress-tolerance phenomenon in early life stages of fish. To this end, we examined the impact of food-deprivation (12 h), reduced oxygen levels (3.5 mg/L for 1 h) and heat shock (HS: + 5 °C for 1 h) on HSP70 and HSP90 protein expression in early life stages of the gilthead sea bream (Sparus aurata), a warm-water aquaculture species. Also, we investigated HSP70 and HSP90 response to food-deprivation (7 days) in early life stages of rainbow trout (Oncorhynchus mykiss), a cool-water aquaculture species, and the tolerance of this larvae to heat shock (either + 5 or + 10 °C for 1 h). Our results clearly demonstrate that food-deprivation enhances HSP70 and HSP90 protein expression in larvae of both species. In gilthead sea bream larvae, the stressors-induced HSP70 and HSP90 (only in the reduced oxygen group) protein expression returned to unstressed levels after 24 h recovery. In fed trout larvae, a + 5 °C heat shock did not elevate HSP70 and HSP90 expression, whereas 100% mortality was evident with a + 10 °C HS. However, food-deprived trout larvae, which had higher HSP70 and HSP90 protein content, survived HS and showed HS-dependent increases in HSP70, but not HSP90 expression. Overall, HSP70 and HSP90 protein expression in early life stages of fish have the potential to be used as markers of nutritional stress, while elevation of the tissue HSPs content may be used as a means to increase stress tolerance during larval rearing.  相似文献

The gonad condition of the sea urchin Strongylocentrotus intermediuscollected in August 1997 at two stations in Peter the Great Bay was examined. One of the stations was located in a polluted area (Alekseev Bight, Popov Island) and the other, in a relatively clean area (the Verkhovskii Islands). The results were compared with analogous data for 1984, 1985, and 1989. In 1997, the gonad condition of sea urchins inhabiting the two areas differed significantly. The mean value of the gonad index (GI) for sea urchins from Alekseev Bight was less than half and the maturity index was about twice that of sea urchins from the Verkhovskii Islands. The GI of sea urchins from Alekseev Bight decreased by a factor of 1.5 between 1984 and 1997. Pronounced histopathological changes were found in sea urchin gonads at this station: granular and hydropic dystrophy of oocytes, resorption and a sharp decrease in the number of gametes (in about 20% of the sea urchins, hardly any gametes were found in the gonads), changes in the morphology of accessory cells (hypertrophy, atrophy, and necrosis), and accumulation of lipofuscin in the cytoplasm of accessory cells and oocytes, in the hemal sinuses and mesentery. The suppressed gonad condition of the sea urchin S. intermediusin Alekseev Bight may be a consequence of the unfavorable environmental situation that formed in the bight in the 1980s–1990s. The main negative factor is anthropogenic pollution of Amurskii Bay.  相似文献

This study examines the molecular species composition and heat-induced crystalline-liquid crystalline phase transitions of phosphatidylcholine (PC) and phosphatidylethanolamine (PE) from the muscle tissues of six species of echinoderms that were collected during the summer: the starfishes Distolasterias nipon and Asterias amurensis, the sea urchin Strongylocentrotus intermedius, and the holothurians Eupentacta fraudatrix, Cucumaria frondosa japonica, and Apostichopus japonicus. Phospholipids (PLs) were in the liquid crystalline state, which is optimal for the functioning of the cell membranes. The use of data on the molecular species composition of PLs for the interpretation of their thermotropic behavior indicated that homeoviscous adaptation is achieved by various rearrangements in the composition of the aliphatic groups of PLs. The phase transitions of PC and PE of echinoderms (except holothurians) were symbatic. The presence of a high-temperature peak on the PC thermograms of C. frondosa japonica and A. japonicus is attributable to the melting of the phospholipid domain, which is composed of molecular species with saturated aliphatic groups. Such domains are responsible for a significant shift in the temperature ranges of the phase transitions of phospholipids of holothurians and sea urchin towards temperatures above 0°C.  相似文献

Temporally consistent individual differences in behavior impact many ecological processes. We simultaneously examined the effects of individual variation in prey activity level, covering behavior, and body size on prey survival with predators using an urchin–lobster system. Specifically, we tested the hypothesis that slow‐moving purple sea urchins (Strongylocentrotus purpuratus) and urchins who deploy extensive substrate (pebbles and stones) covering behavior will out‐survive active urchins that deploy little to no covering behavior when pitted against a predator, the California spiny lobster (Panulirus interruptus). We evaluated this hypothesis by first confirming whether individual urchins exhibit temporally consistent differences in activity level and covering behavior, which they did. Next, we placed groups of four urchins in mesocosms with single lobster and monitored urchin survival for 108 hr. High activity level was negatively associated with survival, whereas urchin size and covering behavior independently did not influence survival. The negative effect of urchin activity level on urchin survival was strong for smaller urchins and weaker for large urchins. Taken together, these results suggest that purple urchin activity level and size jointly determine their susceptibility to predation by lobsters. This is potentially of great interest, because predation by recovering lobster populations could alter the stability of kelp forests by culling specific phenotypes, like foraging phenotypes, from urchin populations.  相似文献

应用实时定量PCR技术对主要卵黄蛋白(Major yolk protein,MYP)基因在不同饥饿时期中间球海胆的体腔细胞、性腺、肠、胃中的转录表达差异进行了分析。结果表明,在正常状态下,MYP基因在体腔细胞、性腺、肠、胃等不同组织中的转录表达差异明显,肠中的表达量最高,其他组织中的表达量均较低。随饥饿时间的延长,MYP基因在体腔细胞中的表达量先迅速下降,而后稳定在较低水平,实验结束时下降至对照组的1.58%;在性腺中的表达量持续上升,实验结束时上升至对照组的679.75%;在肠中的表达量持续下降,实验结束时下降至对照组的33.33%;在胃中的表达量呈上升趋势,实验结束时上升至对照组的106.52倍。综合来看,饥饿状况下,中间球海胆肠中的MYP表达量持续下降,但仍是MYP的主要合成部位;性腺中MYP表达量持续上升,致使其MYP表达比重上升;胃、体腔细胞中表达量在饥饿过程中虽有变化,但总表达量很少,对MYP的整体表达影响不大。  相似文献

Degradation of alginate and its constituents, polymannuronate (polyM) and polyguluronate (polyG), by gut bacteria isolated from sea urchins and abalones in the northern part of Japan, were investigated. Bacterial counts in the guts of sea urchin S. intermedius, were 105 to 108 CFU/g, and in abalone H. discus hannai, counts ranged from 106 to 109 CFU/g. More than 80% of total 600 isolates were found to have alginolytic activity. The alginolytic bacteria were predominantly fermentative, but some differences were observed in their substrate specificity as well as between the flora in the gut of sea urchins and the abalones. Seventy percent of the alginolytic bacteria from the sea urchins showed no degrading preference for polyM or polyG blocks, and were able to degrade both the substrates simultaneously. Most of the alginolytic bacteria (96.6%) from sea urchins belonged to the genus Vibrio. The majority of alginolytic bacteria (68.0% on average) from abalones only degraded polyG and they were predominantly non-motile fermenters. From these results, it appeared that a different type of association exists between alginolytic gut microflora and the marine algal feeders with respect to the level of contribution by bacteria to the host's digestion of alginate. Sea urchins have been used as experimental organisms for developmental biology for over a century. Yet, as is the case for many other marine invertebrates, understanding the development of the juveniles and adults has lagged far behind that of their embryos and larvae. The reasons for this are, in large part, due to the difficulty of experimentally manipulating juvenile development. Here we develop and validate a technique for injecting compounds into juvenile rudiments of the purple sea urchin, Strongylocentrotus purpuratus. We first document the distribution of rhodaminated dextran injected into different compartments of the juvenile rudiment of sea urchin larvae. Then, to test the potential of this technique to manipulate development, we injected Vivo-Morpholinos (vMOs) designed to knock down p58b and p16, two proteins involved in the elongation of S. purpuratus larval skeleton. Rudiments injected with these vMOs showed a delay in the growth of some juvenile skeletal elements relative to controls. These data provide the first evidence that vMOs, which are designed to cross cell membranes, can be used to transiently manipulate gene function in later developmental stages in sea urchins. We therefore propose that injection of vMOs into juvenile rudiments, as shown here, is a viable approach to testing hypotheses about gene function during development, including metamorphosis.  相似文献

A mannan-binding lectin activity was revealed in the coelomic fluid of the following echinoderm species inhabiting the coastal areas of the Sea of Japan, the holothurian Eupentacta fraudatrio, sea urchins Echinocardium cordatum, Strongylocentrotus nudus and S. intermedius, brittle star Amphipholis kochii, sea stars Asterina pectinifera, Lethasterias fusca, Lysastrosoma anthosticta, and Distolasterias nipon. It was shown that, concurrently with the general pattern of lectin interaction with branched bacterial mannans, there were also distinctions caused by the fine carbohydrate specificity of lectins. The obtained data preconditioned the further study of physical and chemical properties and structural features of the echinoderm MBL and the revelation of their role in the formation of the adaptive immune response and in other biological processes.  相似文献

In this study the gene encoding HSP70 was isolated from Phascoloma esculenta by homologous cloning and rapid amplification of cDNA ends (RACE). The full-length of cDNA (2520 bp) consists of a 5′-terminal untranslated region (UTR) (125 bp), a 3′-terminal UTR (421 bp) with a canonical polyadenylation signal sequence (AATAAA), a poly (A) tail, and an open reading frame (ORF) (1974 bp). The predicted molecular mass and isoelectric point for HSP70 is 71.6 kDa and 5.15, respectively. BLAST analysis showed that P. esculenta HSP70 gene shared high similarity. Classical HSP signature motifs, ATP/GTP-Binding Site Motif A, Bipartite Nuclear Targeting Sequence, the cytosolic HSP70 could be expressed in Escherichia coli BL21. After purification, the recombinant pET-HSP70 protein was used to produce the polyclonal antibody in mice and the specificity of the antibody was confirmed by Western blot analysis. Fluorescent real-time quantitative PCR analysis showed that expression of Hsp70 in sipuncula was increased significantly after exposure to 10 mM Zn for12 h, Cd for 24 h, Cu for 48 h, and was exposure to 37 °C for 24 h sea water.  相似文献
