A map-L-system is described which simulates the developmentof the two-dimensional patterns of cell walls displayed at thesurfaces of shoot apices of Psilotum nudum. The simulation ofthese cellular patterns commences with the division of a triangularcell and continues until a complete set of ten different cells,including new triangular cells, is formed amongst the descendantsof each merophyte. The triangular cells generated by means ofthis division pathway, P1, are, in their three-dimensional aspect,four-sided apical cells. In the plant, they have the potentialityto support the development of a shoot apex. The generation ofnew triangular cells by pathway P1 therefore seems to be a preconditionfor the branching of the shoot. Observed variations upon thecellular pattern developed by pathway P1 have also been analysed.Two of these variant pathways, P2 and P3, suggest the typesof controls which are required to bring about all three (P1–P3)patterns of cells. These controls may involve the participationof the plant cytoskeleton and may also require an influencefrom the apical cell itself. The triangular shoot apical cellsof Psilotum are autoreproductive cells: that is, at each division,one of the daughters is a new triangular cell, the other daughterhas some other shape. This example of triangular cell autoreproductionand self-maintenance and its relation to organogenesis is discussedin light of the views on reproduction and self-maintenance expressedby Agnes Arber (1950) in her book The natural philosophy ofplant form(Cambridge: Cambridge University Press). The developmental anatomy of the shoot apex, rhizophore and root ofSelaginella uncinata was examined by the semi-thin section method. The shoot apex has a single, lens-shaped apical cell with two cutting faces. Rhizophore primordia are initiated exogenously at the branching point of the second youngest lateral shoot. The rhizophore apex has a tetrahedral apical cell with three cutting faces. A pair of root primordia is initiated endogenously from inner cells of the rhizophore apex, after the rhizophore apical cell becomes unidentifiable losing its activity, and subsequently a root cap is formed from the distal face of the root apical cell. During the course of successive root branching the apical cell in an original root apical meristem becomes unidentifiable and then a new apical cell is initiated in each of the bifurcated root apical meristems. The root branching mode seems to be equivalent to the described dichotomous branching mode of fern shoots. Our results demonstrate a distinct morphogenetical difference between the rhizophore and the root, and confirm the exogenous origin of the rhizophore, as described for other species ofSelaginella. This evidence indicates that the rhizophore is not an aerial root but a leafless, root-producing axial organ.  相似文献

After a critical evaluation of the concept of dichotomous branching in Cormophytes the shoot apical meristems ofPsilotum triquetrum andSelaginella speciosa are described. InPsilotum only the terminal meristems of the cryptophilic shoots have a three sided apical cell. Those of the photophilic shoots lack a typical apical cell.Selaginella has a two sided apical cell. The process of branching is independent from apical cells. It is due to an equal or unequal fractionation of the initial zone of the shoot apex which embraces all tissues above the leaf producing zone of the apical meristem.

One of the important factors responsible for recalcitrance of maize tissue towards Agrobacterium-mediated transformation is the presence of 2,4-dihydroxy-7-methoxy-2H-1,4-benzoxazin-3(4H)-one (DIMBOA), an inhibitory metabolite found in maize cells. DIMBOA-resistant strains of Agrobacterium tumefaciens were used to transfer genes coding for GUS (-glucuronidase) and NPTII (neomycine phosphotransferase II) in maize shoot apical meristems derived from 20 day-old seedlings and immature embryos. GUS expression was higher (21–34%) in the apical meristem and was dependent on the type of infecting strain and explant-age. The PCR analysis of selected tissues confirmed the presence of GUS gene in the transformed cells.  相似文献

A chalcone synthase (CHS)-like gene, MpCHSLK1, was isolated from liverwort, Marchantia paleacea var. diptera. Phylogenetic analysis revealed that MpCHSLK1 is closely related to stilbene synthase of the whisk fern, Psilotum nudum. Southern blot analysis using an MpCHSLK1 probe revealed that the gene belongs to a small gene family. Northern blot analysis indicated that CHS-like genes were expressed in either the mother plants or photoautotrophic cells. Summary The osphradium of Aplysia californica, a sensory organ, is a small yellow-brown epithelial patch located in the mantle cavity immediately anterior to the rostral attachment of the gill. Scanning electron microscopy reveals a round ellipsoid structure of 0.6–1 mm in diameter with a central, occasionally folded, sensory epithelium. The central area is covered with microvilli and surrounded by a densely ciliated epithelium. Transmission electron micrographs show that the columnar supporting cells in the sensory epithelium contain an abundance of apical pigment granules and microvilli. Between the epithelial-supporting cells, the putative sensory elements consist of thin neurites (0.4–1.5 m in diameter) that reach the sea-water side of the osphradium. The neurites contain many neurotubules, mitochondria, vesicles and cilia in their apices. The nerve endings originate from cell bodies up to 40 m below the epithelium or in the osphradial ganglion itself, as revealed by electron microscopy and retrograde labeling with Lucifer yellow. There appear to be two populations of putative sensory cells, a large population of heavily stained cell bodies 4–10 m in diameter and a few scattered cells of large diameter (25–60 m). Following lanthanum impregnation, septate junctions can be seen between all types of cells in the epithelium, 3–5 m below the sea-water surface. This study provides new information for further investigation of osmo- and mechanosensation in Aplysia californica.  相似文献

Summary We compared the size distributions of leaves on naturally-occurring crowded and experimentally thinned uncrowded individuals of Impatiens pallida in southeastern Pennsylvania. Crowding decreased the number of leaves on individual plants and altered the distribution of leaf size. Crowded individuals had smaller leaves, but the size (length) inequality of the leaf population did not change. The relationships between the height of a plant and the mean and maximum length of its leaves were significantly different for crowded and uncrowded plants. There were weak positive relationships between height and total leaf area, and height and total number of leaves for uncrowded plants, whereas crowded plants showed tighter but curvilinear relationships between these variables. Our results point out the strengths and the limitations of viewing canopies as populations of modules.  相似文献

A unicellular alga which can grow in the light without a combined nitrogen source was isolated from a hot spring. The cells were almost spherical, usually 5–10 m in diameter. Absorption spectra of the watersoluble pigments and of the acetone-extracted ones revealed the existence of chlorophyll a and b and the absence of phycobilins. Thin sections examined by electron microscopy revealed an eukaryotic organization with features typical of the coccoid green algae (the Chlorococcales). Cells divided by internal cytokinesis and subsequent liberation of daughter cells from the parental wall, in a way similar to Chlorella. The alga reduced acetylene to ethylene and incorporated 15N2 into cell protoplasm when incubated in a low oxygen atmosphere. Nitrogenase activity was light-dependent, microaerophilic and thermophilic. A morphogenetic investigation was made of the rhizophore of three large-sized tropical Selaginella species. The rhizophores of Selaginella delicatula, S. caudata, and S. plana arise exogenously at the points of branching of the main stems. In S. delicatula they are initiated at the junction of the second youngest branching. The rhizophore apical meristem has a tetrahedral apical cell and is capless. The rhizophores are usually three or four times dichotomously branched in S. delicatula and S. plana and four or five times in S. caudata. In S. delicatula, dichotomous branching of the rhizophore involves formation of two new apical cells subsequent to loss of an original apical cell. A pair of roots is formed endogenously from inner cells below the dermal layer at the apex of ultimate rhizophore branches. The finding that the rhizophore is an autonomously branched, leafless, and capless axis leads us to argue that Selaginella rhizophores, like lepidodendrid rhizomorphs, are fundamental axial organs that coordinate with the stem and root.  相似文献

Summary The two human homologues of the fission yeast cell cycle protein p13 suc1 displayed structural characteristics consistent with their existing in solution as differently folded monomers despite 81% identity with respect to their primary structures and both being capable of fulfilling the functions of their homologues in fission and budding yeasts. Carboxyfluorescein-labelled p9 CksHs1 and p9 CksHs2 retained their native structures. When microinjected into live stamen hair cells ofTradescantia virginiana, the labelled proteins accumulated in the nuclei of the cells. Medium type, its water status and the relative humidity in the culture vessel modified carnation leaf development in vitro. Carnation shoot apices cultured on liquid or on 0.8% agar solidified media developed into plantlets having succulent and translucent leaves which are not transplantable to non-aseptic conditions. Increasing the agar and/or sucrose concentration in the medium as well as decreasing the relative humidity in the culture vessel by a desiccant promoted glaucous leaf production. Increased water status (H2O and relative humidity) increased shoot proliferation and translucency of leaves. Decreased water status reduced shoot proliferation but induced the formation of glaucous leaves. The culture of apices for 5–6 days on liquid medium prior to their sub-culture to 1.5% agar medium improved shoot proliferation and normal leaf development. An agar slant prevented the submergence of apices in water accumulating on the medium and thus reduced leaf translucency. Survival was further increased by the transfer of plantlets in uncapped culture vessels to a desiccator for 1–2 weeks prior to transplanting to soil.  相似文献

The appearance of cavities and holes in fronds of commercially cultivated Chondrus crispus is described. These ultimately arise from the ravages of a green spot or green rot disease system in which several biotic agents can participate. Nematodes capable of bacterial grazing were recovered from necrotic lesions and we suggest that the nematodes can facilitate wound healing in diseased tissues. Bacteria isolated from disease lesions and from the surfaces of healthy fronds were screened for pathogenic strains. A particularly virulent one, the DOR isolate, was purified from small dark orange colored colonies grown on dilution plates. It was present in necrotic tissue and also recovered from surface scrapings of old healthy fronds, but not from their apical regions. Growth of the DOR isolate appeared to be inhibited by other bacteria colonizing the algal surface. It was shown to be a facultative pathogen, the virulence of which depended on the availability of ammonium or constituents easily metabolized to ammonium. It induced green rot disease in healthy C. crispus and was recoverable in pathogenic form from experimentally infected frond apices. A wound, disease and recovery cycle is discussed to illustrate potential interrelationships involving animal grazers, algal endophytes, Petersenia pollagaster, bacteria and nematodes.  相似文献

The ontogenetic change taking place in the facultative photoperiodic response of A. retroflexus to inductive short-day (SD) conditions was studied by exposing plants to continuous induction after different initial exposures to long-days (LD), and comparing the kinetics of their developmental responses (cumulative number of plants with reproductive apices, flowering stage, and height of the apical dome). As the plants progressed from emergence to autonomous flowering (i.e., in non-inductive conditions), their response to continuous induction became progressively more rapid. Reproductive development was initiated following a progressively shorter lag-phase after the start of induction, but its subsequent rate remained unchanged. Until the onset of reproductive development, the undifferentiated upper part of the shoot apex (apical dome) elongated much more rapidly in SD than in LD. However, in both cases reproductive development was initiated when the apex had elongated to about the same extent, after which its elongation accelerated considerably, but to similar rates in both photoperiods. The data indicate that progress towards reproductive development takes place in inductive (SD), as well as in non-inductive (LD) photoperiods, but one cycle of the latter is as effective as 0.20–0.25 of a cycle of the former. —Plants induced at different stages in ontogeny started to change their subsequent branching pattern (ratio of leafy to leafless branches) as soon as induction was delayed beyond autonomous flowering.Abbreviations LD long-days - SD short-days - RGR Relative Growth Rate  相似文献

Some species of Cactaceae from the Sonoran Desert are characterized by a determinate growth pattern of the primary root, which is important for rapid lateral-root formation and seedling establishment. An analysis of the determinate root growth can be helpful for understanding the mechanism of meristem maintenance in plants in general. Stenocereus gummosus (Engelm.) Gibson & Horak and Pachycereus pringlei (S. Watson) Britton & Rose are characterized by an open type of root apical meristem. Immunohistochemical analysis of 5-bromo-2-deoxyuridine incorporation into S. gummosus showed that the percentage of cells passing through the S-phase in a 24-h period is the same within the zone where a population of relatively slowly proliferating cells could be established and above this zone in the meristem. This indicated the absence of the quiescent center (QC) in S. gummosus. During the second and the third days of growth, in the distal meristem portion of P. pringlei roots, a compact group of cells that had a cell cycle longer than in the proximal meristem was found, indicating the presence of the QC. However, later in development, the QC could not be detected in this species. Summary Arbuscular mycorrhizal (AM) fungi significantly improve plant growth in soils with low phosphorus availability and cause many changes in root morphology, similar to those produced by increased P nutrition, mainly depending on root apex size and activity. The aim of this work was to discriminate between the morphogenetic role of AM fungi and P in leek (Allium porrum L.) by feeding mycorrhizal and nonmycorrhizal plants with two nutrient solutions containing 3.2 or 96 M P and examining specific parameters related to adventitious root apices (apex size, mitotic cycle, and RNA synthesis). The results showed that AM fungi blocked meristem activity as indicated by the higher percentages of inactive apices and metaphases in the apical meristem of mycorrhizal plants, whereas the high P supply lengthened the mitotic cycle without blocking the apices, resulting in steady, slow root growth. Summary Root differentiation could be elicited on carrot discs by transformation with the agropine Ri plasmid rolB gene cloned in the binary vector Bin19, provided two conditions were met. Firstly, an adequate auxin supply had to be provided. This was achieved by co-inoculation with a strain carrying only the auxin synthetic genes of the TR-DNA. Most of the resulting roots were then shown to harbour only rolB and no aux genes. Secondly, an extended non-coding region (1200 bp) at the 5 end of rolB had to be included in the construction. A shorter (300 bp) 5 region, including TATA and CCAAT boxes, was not sufficient to trigger root differentiation. Both the extended (B1185) and reduced (B310) 5 regions of rolB were then cloned upstream of the -glucuronidase (GUS) reporter gene and infections carried out both on the apical and on the basal side of carrot discs. Strong expression of GUS, visualized histochemically as an intense blue colouring of transformed cells was observed with B1185-GUS constructions on the apical side of the discs. Only occasionally could coloured cells be observed on the basal side of the discs with B1185-GUS and on both apical and basal sides with B310-GUS constructions. Strong GUS expression was, on the contrary, achieved on cells of both auxin-rich (apical) and auxin-depleted (basal) sides of the discs with the strong constitutive viral promoter, CaMV35S. These results indicate the presence of an upstream regulatory region which confers polar expression to the rolB gene and suggest a role for auxin in its activation.  相似文献

The shoot apical meristem of higher plants consists of a population of stem cells at the tip of the plant body that continuously gives rise to organs such as leaves and flowers. Cells that leave the meristem differentiate and must be replaced to maintain the integrity of the meristem. The balance between differentiation and maintenance is governed both by the environment and the developmental status of the plant. In order to respond to these different stimuli, the meristem has to be plastic thus ensuring the stereotypic shape of the plant body. Meristem plasticity requires the ZWILLE (ZLL) gene. In zll mutant embryos, the apical cells are misspecified causing a variability of the meristems size and function. Using specific antibodies against ZLL, we show that the zll phenotype is due to the complete absence of the ZLL protein. In immunohistochemical experiments we confirm the observation that ZLL is solely localized in vascular tissue. For a better understanding of the role of ZLL in meristem stability, we analysed the genetic interactions of ZLL with WUSCHEL (WUS) and the CLAVATA1, 2 and 3 (CLV) genes that are involved in size regulation of the meristem. In a zll loss-of-function background wus has a negative effect whereas clv mutations have a positive effect on meristem size. We propose that ZLL buffers meristem stability non-cell-autonomously by ensuring the critical number of apical cells required for proper meristem function.Edited by G. The organogenetic potential from callus of three tomato land races from the Canary Islands adapted to semi-arid environment (Salvaje, Rusa and Especial), and one tomato cultivar (Meltine), were examined. The response of four explant types (cotyledon, shoot apex, hypocotyl and root) to nine PGR regimes (BAP at 1 or 2 or 5 mg/l) + either IAA (0.5 mg/l) or 2,4-D (0.5 or 1 mg/l) were measured. BAP at 5 mg/l+IAA at 0.5 mg/l induced most organogenesis in all the explant types for all genotypes. A combination tapetum consisting of a cellular, parietal component and a plasmodial component occurs inSchizaea pectinata. A single, tapetal initial layer divides to form an outer parietal layer which maintains its cellular integrity until late in spore wall development. The inner tapetal layer differentiates into a plasmodium which disappears after the outer exospore has developed. In the final stages of spore wall development, granular material occurs in large masses and is dispersed as small granules throughout the sporangial loculus. No tapetal membrane develops. Genetically engineered pacemaking in ventricular cells has been achieved by down-regulation of the time independent inward rectifying current (I K1), or insertion of the hyperpolarisation-activated funny current (I f). We analyse the membrane system (i.e. ionic concentrations clamped) of an epicardial Luo-Rudy dynamic cell model using continuation algorithms with the maximum conductance () of I K1 and I f as bifurcation parameters. Pacemaker activity can be induced either via Hopf or homoclinic bifurcations. As K1 is decreased by ≈74%, autorhythmicity emerged via a homoclinic bifurcation, i.e., the periodicity first appear with infinitely large periods. In contrast, the insertion of f induced periodicity via a subcritical Hopf bifurcation at f≈ 0.25 mSμF−1. Stable autorhythmic action potentials occurred at f > 0.329 mSμF−1.  相似文献
