The sucrose transporter SUT1 functions in phloem loading of photoassimilates in solanaceous plant species. In the present study, wildtype and transgenic potato plants with either constitutive overexpression or antisense inhibition of SUT1 were grown under high or low phosphorus (P) fertilization levels in the presence or absence of the arbuscular mycorrhizal (AM) fungus Glomus intraradices. At a low soil P fertilization level, the extent of AM fungal root colonization was not different among the genotypes. In all plants, the AM symbiosis contributed significantly to P uptake under these conditions. In response to a high soil P fertilization level, all genotypes showed a decrease in AM fungal root colonization, indicating that the expression level of SUT1 does not constitute a major mechanism of control over AM development in response to the soil P availability. However, plants with overexpression of SUT1 showed a higher extent of AM fungal root colonization compared with the other genotypes when the soil P availability was high. Whether an increased symbiotic C supply, alterations in the phytohormonal balance, or a decreased synthesis of antimicrobial compounds was the major cause for this effect requires further investigation. In plants with impaired phloem loading, a low C status of plant sink tissues did apparently not negatively affect plant C supply to the AM symbiosis. It is possible that, at least during vegetative and early generative growth, source rather than sink tissues exert control over amounts of C supplied to AM fungi.  相似文献

As toxic pollutants commonly found in tobacco (Nicotiana tabacum L.) products, lead (Pb) and cadmium (Cd) can enter the human body via smoking and thus pose a potential health risk to smokers. We conducted a greenhouse experiment to study the effects of arbuscular mycorrhizal (AM) inoculation with Glomus intraradices BEG 141 and organic amendment with cattle manure, alone or in combination, on the growth, P nutrition, and heavy-metal uptake by tobacco plants grown in soil to which was added Pb-Cd at 0/0, 350/1, 500/10, and 1,000/100?mg?kg?1, respectively. In general, AM colonization and plant growth were greatly reduced by Pb-Cd contamination, whereas organic amendment alleviated Pb-Cd stress and showed some beneficial effects on AM symbiosis and some soil parameters. AM inoculation, alone or in combination with organic amendment, increased plant dry weights and improved P nutrition significantly at all Pb-Cd addition levels, and, in most cases, it decreased Pb and Cd concentrations in tobacco plants and DTPA-extractable concentrations in soil. AM inoculation increased total glomalin-related soil protein (GRSP) concentrations in soil to which Pb-Cd was added. The higher soil pH and GRSP contents and the lower DTPA-extractable Pb and Cd concentrations contributed by AM inoculation and/or organic amendment may be contributing factors that lead to higher growth promotion and lower metal toxicity and uptake by plants. Our findings suggest that AM inoculation in combination with organic manure may be a potential method for not only tobacco production but phytostabilization of Pb-Cd-contaminated soil.  相似文献

We investigated the growth and nutrient uptake of the Lycopersicon esculentum symbiosis mycorrhiza-defective plant mutant rmc, challenged with arbuscular mycorrhiza (AM) fungal propagules, in the presence or absence of roots of the commercial wild-type tomato cv. Golden Queen (GQ). Two plants shared the middle (combi) compartment of a horizontal three-compartment split-root pot with one part of their root system; the other part was grown separately in an outer (solo) pot. Combinations of rmc and GQ plants were grown together in soil that was either mycorrhiza-free (-M) or prepared with AM fungal inoculum (+M). Surface colonization of rmc roots was strongly increased in the presence of (+M) GQ roots. AM fungal inoculation increased phosphorus uptake of GQ plants, but decreased growth and P uptake of rmc plants. Growth and P uptake of (+M) GQ plants were reduced when plants were grown in combination with rmc rather than another GQ plant. AM fungi in the (combi) compartment may have preferentially formed hyphae spreading infection rather than functioning in P uptake in (+M) GQ plants grown in combination with rmc. Surface colonization of (+M) rmc roots, in the presence of GQ roots, was probably established at the expense of carbohydrates from associated GQ plants. Possible reasons for a decreased P uptake of rmc plants in response to AM fungal inoculation are proposed.  相似文献

This study was conducted to elucidate the effect of P fertilisation on the function of field communities of arbuscular mycorrhizal fungi (AMF) measured as P transport to flax. Two methods were applied to soil from a long-term field experiment with NaHCO3-extractable soil P levels of 24 and 50 mg kg-1in an experiment under controlled conditions: i) Measurement of plant growth and P uptake in the presence or absence of the fungicide benomyl and ii) measurement of hyphal P transport from a root-free compartment labelled with 32P. Benomyl successfully prevented mycorrhizal function. The absolute contribution of AMF to plant P uptake was of the same magnitude with or without P fertilisation at 27 days after sowing. Therefore, even though plants grown at the higher soil P level had greater P uptake, the relative contribution of AMF to P uptake was greater at the lower P level than at the higher P level (77 and 49% of total P uptake, respectively). The AMF in P-fertilized soil transported less P32 from the root-free compartment to the plant after 23 days than the AMF in unfertilized soil, but this difference disappeared in plants harvested after 27 and 32 days. The production of hyphae was largely similar in both fertilization treatments, indicating that the capacity for P uptake and transport by hyphae of the two AMF communities was similar. An experiment was set up to investigate the role of arbuscular mycorrhiza (AM) in utilization of P from organic matter during mineralization in soil. Cucumber (Cucumis sativus L.) inoculated with one of two AM fungi or left uninoculated were grown for 30 days in cross-shaped PVC pots. One of two horizontal compartments contained 100 g soil (quartz sand: clay loam, 1:1) with 0.5 g ground clover leaves labelled with32P. The labelled soil received microbial inoculum without AM fungi to ensure mineralization of the added organic matter. The labelling compartment was separated from a central root compartment by either 37 m or 700 m nylon mesh giving only hyphae or both roots and hyphae, respectively, access to the labelled soil. The recovery of32P from the hyphal compartment was 5.5 and 8.6% for plants colonized withGlomus sp. andG. caledonium, respectively, but only 0.6 % for the non-mycorrhizal controls. Interfungal differences were not related to root colonization or hyphal length densities, which were lowest forG. caledonium. Both fungi depleted the labelled soil of NaHCO3-extractable P and32P compared to controls. A 15–25% recovery of32P by roots was not enhanced in the presence of mycorrhizas, probably due to high root densities in the labelled soil. The experiment confirms that AM fungi differ in P uptake characteristics, and that mycorrhizal hyphae can intercept some P immobilization by other microorganisms and P-sorbing clay minerals.  相似文献

The seasonality of arbuscular mycorrhizal (AM) fungi–plant symbiosis in Lotus glaber Mill. and Stenotaphrum secundatum (Walt.) O.K. and the association with phosphorus (P) plant nutrition were studied in a saline-sodic soil at the four seasons during a year. Plant roots of both species were densely colonized by AM fungi (90 and 73%, respectively in L. glaber and S. secundatum) at high values of soil pH (9.2) and exchangeable sodium percentage (65%). The percentage of colonized root length differed between species and showed seasonality. The morphology of root colonization had a similar pattern in both species. The arbuscular colonization fraction increased at the beginning of the growing season and was positively associated with increased P concentration in both shoot and root tissue. The vesicular colonization fraction was high in summer when plants suffer from stress imposed by high temperatures and drought periods, and negatively associated with P in plant tissue. Spore and hyphal densities in soil were not associated with AM root colonization and did not show seasonality. Our results suggest that AM fungi can survive and colonize L. glaber and S. secundatum roots adapted to extreme saline-sodic soil condition. The symbiosis responds to seasonality and P uptake by the host altering the morphology of root colonization.  相似文献

 Root colonization by arbuscular mycorrhizal fungi (AMF) may affect protein and lipid composition of plants by altering P nutrition or by eliciting other metabolic responses in the host plant. This study was conducted to determine the effects of an AMF and soil P on seed protein and lipid contents and yield of two genotypes of durum wheat (Triticum durum L.). Plants were grown in a greenhouse using soil: sand mixes with different levels of P, and with or without the AMF Glomus mosseae [(Nicol. and Gerd.) Gerd. and Trappe]. Percentage AMF root colonization decreased as P added to soil increased. The wheat genotype CR057 had higher AMF root colonization but lower seed P and protein concentrations than CR006. Without added soil P, protein concentration was significantly lower and lipid concentration and seed dry weight higher in arbuscular mycorrhizal (AM) than in nonAM plants. Seed lipid and protein contents were highly correlated with P content of plants. In nonAM plants, seed lipid and protein contents were low with no added soil P and did not differ with added soil P. Seed protein/lipid (Pro/L) concentration ratios of AM plants were higher than those of nonAM plants only when no P was added to the soil. The data indicate different patterns of seed P accumulation and different relationships between seed P and protein and lipid in AM and nonAM plants. Arbuscular mycorrhizal (AM) mycelia persist in soil over winter. Functioning of the AM symbiosis very early in the spring when the soil temperature is low may be of ecological significance for perennial and biannual plants in cool climates. An indoor experiment was conducted to investigate the effects of low root-zone temperatures on 32P uptake by 10-week-old leek plants (Allium porrum L.) inoculated or not with the AM fungus Glomus intraradices Schenck & Smith. Plants were grown in a greenhouse at approximately 23 degrees C prior to exposing their roots to 23 degrees C, 15 degrees C or 0 degree C. Mycorrhizal colonization increased 32P activity of leek leaves at a root-zone temperature of 23 degrees C seven days after injection of 32P into the soil, whereas 14 days after injection, 32P increases were measured at both 23 degrees C and 15 degrees C. The lack of difference in 32P activity between AM and non-AM plants at 0 degree C, both 7 and 14 days after injection, suggests that the AM fungus is not functional at this low root-zone temperature.  相似文献

The effects of soil P amendments and time of application on the formation of external mycelium by different arbuscular mycorrhizal (AM) fungi were studied. In the first experiment the external mycelium produced in the soil by the AM fungus Glomus etunicatum Beck. and Gerd., during the early stages of root colonization (7 and 14 days after inoculation), was quantified by the soil-agar film technique. A Brazilian Oxisol was used with three different phosphate levels, varying from deficient to supra-optimal for the plant. Significant differences were observed in the phosphate and inoculation treatments for plant dry weight, P content in the tissue, root length and root colonization, at fourteen days after planting. At 7 days, mycelium growth, root colonization and their relationship were reduced at supra-optimal P concentrations. Applications of P one week after planting reduced mycelium growth and root colonization more than when applied to the soil before planting. In a second experiment the arbuscular mycorrhizal (AM) fungi, Scutellospora heterogama (Nicol. and Gerd.) Walker and Sanders and E3 were tested and compared with Glomus etunicatum. For the species studied, the length of external hyphae per unit of colonized root length was affected by small P additions but no further significant differences were observed at high P levels. The three AM endophytes showed marked differences in their response to P in the soil: Scutellospora heterogama, although producing external mycelium more profusely than the Glomus spp., showed a higher sensitivity to soil P supply.  相似文献

We investigated uptake of Cd by arbuscular mycorrhizal (AM) maize inoculated with Glomus mosseae from a low-P sandy calcareous soil in two glasshouse experiments. Plants grew in pots containing two compartments, one for root and hyphal growth and one for hyphal development only. Three levels of Cd (0, 25 and 100 mg kg–1) and two of P (20 and 60 mg kg–1) were applied separately to the two compartments to assess hyphal uptake of Cd. Neither Cd nor P addition inhibited root colonization by the AM fungus, but Cd depressed plant biomass. Mycorrhizal colonization, P addition and increasing added Cd level led to lower Cd partitioning to the shoots. Plant P uptake was enhanced by mycorrhizal colonization at all Cd levels studied. When Cd was added to the plant compartment and P to the hyphal compartment, plant biomass increased with AM colonization and the mycorrhizal effect was more pronounced with increasing Cd addition. When P was added to the plant compartment and Cd to the hyphal compartment, plant biomass was little affected by AM colonization, but shoot Cd uptake was increased by colonization at the low Cd addition rate (25 mg kg–1) and lowered at the higher Cd rate (100 mg kg–1) but with no difference in root Cd uptake. These effects may have been due to immobilization of Cd by the fungal mycelium or effects of the AM fungus on rhizosphere physicochemical conditions and are discussed in relation to possible phytostabilization of contaminated sites by AM plants.  相似文献

在温室盆栽条件下,分别模拟单作、间作和尼龙网分隔种植,比较接种丛枝菌根(arbuscular mycorrhizal, AM)真菌Glomus intraradicesGlomus mosseae对菌根植物玉米和非菌根植物油菜生长和磷吸收状况的影响,并分析土壤中各无机磷组分的变化。结果发现,接种AM真菌可以促进土壤中难溶性磷(Ca10-P和O-P)向有效态磷转化,并显著降低总无机磷含量 (P<0.05),显著提高菌根植物玉米的生物量和磷吸收量(P<0.05),特别是在间作体系中使玉米的磷营养竞争比率显著提高了45.0%-104.1% (P<0.05),显著降低了油菜的生物量和磷吸收量(P<0.05),从而增强了了菌根植物的竞争优势,降低了非菌根植物与菌根植物的共存能力。揭示了石灰性土壤中AM真菌对植物物种多样性的影响,有助于更加全面地理解AM真菌在农业生态系统中的作用。  相似文献

Two experiments with soil cores were carried out to investigate the effects of arbuscular mycorrhizal (AM) fungal colonization on mobility of phosphorus (P) during leaching of repacked columns of a soil with a loamy sand texture. Trifolium subterraneum plants inoculated with an AM fungus or not inoculated were grown in cores with low or high P concentrations for 8 or 10 weeks in the glasshouse. Cores were then irrigated with 2500 mL water and the leachate collected. Plant growth and the amounts of P removed by plants, remaining in soil as available P and removed dissolved in leachate were measured. Mycorrhizal fungal colonization and development of external hyphae were also determined. Inoculation and/or P application significantly increased plant growth and plant P removal and decreased P leaching. In low P soils AM fungal colonization significantly increased plant P uptake and decreased soil available P and total dissolved P in leachates. Lower P leaching from cores with AM plants under low P conditions was related to enhancement of plant growth and to scavenging and removal of P from the soil by roots and/or external hyphae. When P was applied AM effects were not observed and available P remaining in the soil after leaching was much higher, regardless of AM fungal colonization.  相似文献

Previously, tillage has been found to reduce early-season phosphorus (P) uptake from soil in continuous maize cropping systems. This reduced P uptake has often been associated with delayed colonization of roots by arbuscular mycorrhizal (AM) fungi. Our aim was to determine if similar responses occur in maize-soybean rotations, which are more typical of current farming in Ontario, Canada. Similar responses were expected because both are AM crops, and the mechanism by which tillage reduces P uptake is thought to be a negative impact on the development of effective mycorrhizae. Simultaneous field experiments with either maize-soybean-maize or soybean-maize-soybean rotations were conducted in 1992–4. Treatments imposed were no-till (NT), ridge-tillage (RT), and conventional tillage using a moldboard plow (MP). In 1993, early-season dry mass of maize was similar among treatments, but colonization of maize roots by AM fungi and P uptake of maize were stimulated by NT and RT, compared with MP. In 1994, early growth was more rapid overall than in 1993, but it was reduced in the NT and RT treatments compared with MP for reasons not related to P. For soybean, AM colonization in NT and RT systems was higher than with MP, but P uptake was unchanged. As was found for maize in 1994, early-season shoot dry mass of soybean was higher in the MP treatment than with NT, but both in 1993 and 1994. We conclude that colonization of both maize and soybean by AM fungi is susceptible to slower development in tilled systems, and that for maize, stimulation of P uptake under reduced tillage can occur in rotations with soybean just as easily as it does with continuous maize. Taken with other studies, the data here suggest that responses to tillage of colonization of roots by AM fungi and of P uptake could apply to many cropping systems. The slow early-season shoot growth seen in some years in response to reduced tillage is discussed.  相似文献

* This study investigated effects of plant density and arbuscular mycorrhizal (AM) colonization on growth and phosphorus (P) nutrition of a cultivar of wheat (Triticum aestivum) that often shows early AM-induced growth depressions. * Two experiments were conducted. Expt 1 had three plant densities and one soil P concentration. Expt 2 had two plant densities and two P concentrations. Plants were grown in calcareous P-fixing soil, inoculated with Glomus intraradices or Gigaspora margarita, or noninoculated (nonmycorrhizal (NM)). Glomus intraradices colonized well and caused a growth depression only in Expt 1. Gigaspora margarita caused large growth depressions in both experiments even though it colonized poorly. * The results showed that growth depressions were mitigated by changes in relative competition for soil P by NM and AM plants, and probably by decreasing carbon costs of the fungi. * The different effects of the two fungi appear to be attributable to differences in the balance between P uptake by the fungal pathway and direct uptake via the roots. These differences may be important in other AM symbioses that result in growth depressions. The results show that mycorrhizal growth responses of plants grown singly may not apply at the population or community level.  相似文献

通过30μm尼龙网将根盒分成根室和菌丝室,菌丝室中的低磷土壤施加75 mg P/kg土壤的植酸钙,研究了菌丝室土壤中丛枝菌根(AM)真菌Glomus intraradices和解磷细菌Bacillus megaterium C4对有机磷的矿化和吸收.结果表明,在试验条件下,植酸钙的溶解性很低,对土壤溶液有机磷的贡献不大.接种解磷细菌C4提高了土壤中磷酸酶的活性,减少了土壤中有机磷的含量.但是,由于存在解磷细菌与AM真菌对磷的竞争,解磷细菌矿化出的磷大部分被自身利用,AM真菌的生长受到抑制,解磷细菌对植物磷营养的改善没有表现出显著的贡献.  相似文献

A pot experiment was conducted to investigate the uptake of Zn from experimentally contaminated calcareous soil of low nutrient status by maize inoculated with the arbuscular mycorrhizal (AM) fungus Glomus caledonium. EDTA was applied to the soil to mobilize Zn and thus maximize plant Zn uptake. The highest plant dry matter (DM) yields were obtained with a moderate Zn addition level of 300 mg kg?1. Plant growth was enhanced by mycorrhizal colonization when no Zn was added and under the highest Zn addition level of 600 mg kg?1, while application of EDTA to the soil generally inhibited plant growth. EDTA application also increased plant Zn concentration, and Zn accumulation in the roots increased with increasing EDTA addition level. The effects of inoculation with Gcaledonium on plant Zn uptake varied with Zn addition level. When no Zn was added, Zn translocation from roots to shoots was enhanced by mycorrhizal colonization. In contrast, when Zn was added to the soil, mycorrhizal colonization resulted in lower shoot Zn concentrations in mycorrhizal plants. The P nutrition of the maize was greatly affected by AM inoculation, with mycorrhizal plants showing higher P concentrations and P uptake. The results indicate that application of EDTA mobilized soil Zn, leading to increased Zn accumulation by the roots and subsequent plant toxicity and growth inhibition. Mycorrhizal colonization alleviated both Zn deficiency and Zn contamination, and also increased host plant growth by influencing mineral nutrition. However, neither EDTA application nor arbuscular mycorrhiza stimulated Zn translocation from roots to shoots or metal phytoextraction under the experimental conditions. The results are discussed in relation to the environmental risk associated with chelate-enhanced phytoextraction and the potential role of arbuscular mycorrhiza in soil remediation.  相似文献

The effects of inoculation with arbuscular mycorrhizal (AM) fungi, organic fertilizer (F) applications, and soil sterilization on maize growth were evaluated in a pot experiment. The experiment was in a completely randomized factorial design (2 × 4 × 2) with six replicates for each treatment. There were two soil treatments (sterilized soil, SS and unsterilized soil, US), four organic fertilizer treatments (0.0, 0.5, 1.0 and 2.0 g kg-1 soil), and two AM fungi treatments (inoculation with Glomus mosseae, +AM and uninoculated control, -AM). Inoculated plants generally had greater AM colonization, plant height, dry weight and phosphorus (P) uptake than uninoculated controls, and these parameters were significantly increased as the organic fertilizer application increased up to 0.5 g kg-1 but decreased or had no significant effect compared to the uninoculated plants at the highest fertilizer rate (2.0 g kg-1). Plant growth, P uptake and AM colonization of root system were significantly higher in sterilized soil compared to the unsterilized control. Our results indicated that the inoculation of AM fungi in field soil with optimal organic fertilizer application greatly improved maize growth and nutrient uptake, and the effect was greater under sterilized soil condition.  相似文献

The hypothesis of this study was that cumulative P fertilization decreases the contribution of arbuscular mycorrhiza (AM) to crop growth and nutrient uptake in Northern European field conditions. The modes of action of P fertilization were evaluated through effects on mycorrhization, crop dependence on AM, and AM fungal (AMF) community. Field studies were carried out within long-term experiments on soils with low and intermediate initial content of extractable P, where no P fertilization and 45 kg ha–1 a–1 P were applied for 20 years. AM effectiveness in terms of growth and nutrient uptake of flax, red clover and barley, percentage root length colonized by AMF, P response of flax, and spore densities and species composition of the AMF communities, were assessed. In the soil with low initial P supply, cumulative P fertilization decreased AM contribution to crop growth and nutrient uptake. The higher AM effectiveness in soil with no added P compensated the cumulative P fertilization (soil PH2O 2.5 v. 9.5 mg kg–1) for flax, but not completely for clover. In contrast, barley obtained no benefit from AM at harvest and only a slight benefit from cumulated P. In the soil with intermediate initial P supply, AM reduced growth of flax and barley, especially with no added P, and no response to AM was obtained on clover due to retarded mycorrhization. Cumulative P fertilization reduced yield losses of flax by AM (PH2O 18.8 v. 5.4 mg kg–1), because fertilization inhibited mycorrhization. In both soils, root colonization and spore density were decreased by cumulative P fertilization, but no changes in AMF species composition were observed.  相似文献

We investigated the physiology that underlies the influence of arbuscular mycorrhizal (AM) colonization on outcomes of interactions between plants. We grew Medicago truncatula A17 and its AM‐defective mutant dmi1 in intragenotypic (two plants per pot of the same genotype, x2) or intergenotypic (one plant of each genotype, 1 + 1) combinations, inoculated or not with Rhizophagus irregularis (formerly Glomus intraradices) or Gigaspora margarita. We measured plant growth, colonization, contributions of AM and direct P uptake pathways using 32P, and expression of plant Pi transporter genes at two levels of P supply. A17 (x2) responded positively to inoculation only at low P. The response was enhanced with 1 + 1 even at high P where colonization in A17 was reduced. With R. irregularis P uptake by the AM pathway was unaffected by P supply, whereas with G. margarita, the AM pathway was lower at high P, and direct uptake higher. Gene expression varied and was unrelated to P uptake through the two pathways. There was no evidence of plant control of P uptake via R. irregularis at high P but there was via G. margarita. Importantly, growth responses of plant genotypes grown alone did not predict outcomes of intergenotypic interactions.  相似文献
