The effects of nutrient enrichment and herbivory on resource allocation patterns among morphology, reproduction, and chemical content of the brown alga, Turbinaria conoides (J. Agardh) Kützing were tested in the shallow subtidal zone of the Gulf of Thailand. The field experimental design comprised 36 plots (50 × 50 cm2) with and without herbivores, and two nutrient levels. Cages (uncaged and fully caged plots) were used to exclude herbivorous fishes and two nutrients levels were achieved by experimental enrichment above ambient nutrient concentrations. For morphology of Turbinaria, the maximum length (holdfast to frond apex), the width of the base of the stipe, number of blades, holdfast and stipe diameter were measured. Biomass, reproductive output, and tissue nutrient (carbon: nitrogen: phosphorus) content of T. conoides were examined. Phlorotannin concentrations were examined using a modified Folin‐Ciocalteu method. The results showed that herbivory had no effect on morphology, reproduction, or phlorotannin concentrations. This could be due to the structural and morphological deterrents of alga, which might minimize grazing effects from herbivorous fishes. Nutrient enrichment had no effect on morphology and reproduction of T. conoides, possibly due to low nutrient demand in Turbinaria. However, nutrient enrichment did affect phlorotannin concentrations, as phlorotannins in the enriched plots were lower than the ambient plots. These results support, in part, the carbon–nutrient balance hypothesis that states algae will allocate fewer resources to production of anti‐herbivore chemical compounds when enriched with potential growth‐limiting nutrients.  相似文献

In recent years, sulfated polysaccharide (fucoidan) is being marketed as a nutraceutical and food supplement because of its various established pharmacological activities. Hence this study was designed to investigate the protective effect of fucoidan extracted from Turbinaria conoides against isoproterenol induced myocardial injured rats. Biochemical assessment of myocardial injury was done by measuring the activities of creatine kinase, lactate dehydrogenase, membrane bound triphosphatases and minerals, which were significantly altered in isoproterenol administered rats. In addition, the enzyme mapping assay on the size of myocardial infarct also correlated with these biochemical parameters. Thus, the observed protective effects of sulfated polysaccharides of T. conoides against ISO induced myocardial injury are due to membrane stabilizing property and this property might be due to decreased lipid peroxidation. These results may trigger a renewed interest in the use of T. conoides fucoidan for myocardial injury.  相似文献

The ability of the brown seaweed, Turbinaria conoides to absorb rhodamine B (RB) from aqueous solution was investigated in a batch system. The effects of various experimental parameters (e.g., morphology, initial dye concentration, temperature, and pH) were investigated and optimal experimental condition was ascertained. The results revealed that sorption capacity of T. conoides increased with increasing temperature, and decreasing initial dye concentration and pH. Sorption equilibrium studies using Langmuir and Freundlich models demonstrated that RB biosorption fitted well to the Langmuir isotherm. This study suggested that T. conoides can be used as a potential biosorbent for the removal of RB from wastewater.  相似文献

In screening for novel photosensitizers for photodynamic therapy, 14 seaweed samples from Port Dickson in Malaysia were collected. Methanolic extracts of these samples were prepared and evaluated for phototoxicity using a short-term cell viability assay, where promyelocytic leukemia cells, HL60 were incubated with the extracts prior to irradiation with a broad spectrum light at 9.6?J?cm?2 (equivalent to 10.5?mW?cm?2 for 10?min). Four of the methanolic extracts demonstrated moderate to strong phototoxicity and bioassay-guided isolation of photosensitizers was carried out on two selected seaweeds to yield a total of eight cyclic tetrapyrrolic compounds which are derivatives of chlorophyll-a and -b. Seven of these compounds showed >50% phototoxicity at 5?μg?mL?1 while exhibiting minimal cytotoxicity in the dark, which is an important characteristic of an ideal photosensitizer.  相似文献

Turbinaria foliosasp. nov. is described on the basis of several collections made from Dhofar, Sultanate of Oman. The new species is known only from southern Oman, a region of the northern Arabian Sea that is strongly impacted by the upwelling from the summertime monsoon. It is distinguished from other species of the genus by the shape of the leaves and by their loose, non‐congested arrangement. Air vesicles, embedded in the leaves, may be present, but more often are absent. A census of the currently recognized species in the genus Turbinaria is provided. Reference is made to Sargassum turbinarioides and Sargassum turbinati‐folium. These species possess discrete blades and spherical vesicles and thus conform to Sargassum.  相似文献

Several species of the genus Turbinaria coexist along the coasts of islands in the Indian and Pacific Oceans. Among these brown algae, Turbinaria ornata and T. conoides are sister species that are difficult to differentiate using exclusively morphological characters. Based on in vivo nuclear magnetic resonance and chromatographic techniques, i.e., liquid and gas chromatography‐mass spectrometry analysis, combined with phylogenetic data, we successfully identified turbinaric acid in T. conoides samples from several Indian and Pacific Ocean islands. This nonvariable discriminant molecule was only identified in T. conoides specimens, but not in the two allied species T. ornata and T. decurrens. Results are discussed with regard to turbinaric acid as an interesting chemomarker isolated from T. conoides and the rapid discrimination of Turbinaria specimens using chemical assays.  相似文献

From the lipophilic extract of the brown alga Bifurcaria bifurcata collected off the Atlantic coast of Southern Brittany (Quiberon, France), five polar linear diterpenoids have been isolated. These metabolites have been identified as hydroxylated derivatives of 13-oxo- and 13-hydroxygeranylgeraniol. Their structures were characterized on the basis of chemical and spectral evidence including two-dimensional NMR experiments and mass spectrometric techniques. The absolute configuration of the 13-position has been determined, for the 13-hydroxygeranylgeraniol derivatives, to be R by means of a modified Mosher's method and therefore that of 13-hydroxygeranylgeraniol (eleganediol) has been revised. Along with these compounds, three related known geranylgeraniol derivatives were also identified, and these data were used for chemotaxonomical purposes.  相似文献

Spatial variation in polyphenolic content in annual shoots of the brown alga Ascophyllum nodosum was quantified using a hierarchical sampling design. Three sampling levels, covering distances of 100–106 m, were used. Comparisons were made between two areas, Tjärnö on the Swedish west-coast and the Isle of Man in the Irish Sea, with very different types of environmental conditions. No significant differences in mean polyphenolic levels were found between the two study areas (6.6% of dry mass at Tjärnö and 9.2% at the Isle of Man), whereas significant and substantial differences were found among sites within areas (range 5.7%–11.4%) and among quadrats within sites (range 3.7%–13.1 %). The extensive variation at the smaller spatial scales points out the importance of using thorough sampling procedures at all levels in large-scale studies on algal polyphenolics, e.g. biogeographical comparisons, which have been neglected in several previous studies. Moreover, the results imply that experiments on causal factors of polyphenolic variation should be designed to explain the spatial scales on which the factors are impportant. This study also investigated the relationship between polyphenolic concentration and both plant size and mean area of annual shoots. The mean area was used as an estimate of the mean growth rate of the annual shoots within an individual. No significant relationships were found between shoot growth rate, or plant size, and polyphenolic levels in annual shoots at any of the three spatial scales that were investigated.  相似文献

The mature female conceptacle of Sargassum horneri (Turner) C. Agardh has an ostiole filled with a gelatinous plug. The oogonium in the conceptacle has cell walls that can be differentiated into a dense outer and a less dense inner microfibrillar layer. Just prior to egg release, stalk material is produced inside the outer layer and the inner layer disappears. At this stage the gelatinous plug is extruded and mucilage is released through the ostiole. The released eggs are retained on the receptacle by the stalk and are surrounded by a large amount of the mucilage. Three-celled germlings form a primary wall with a polylamellated structure of microfibril layers. In multicellular germlings that have differentiated into thallus and rhizoids, the peripheral thallus cells have an outer cell wall consisting of a microfibril layer under the primary wall, while the cell wall of the rhizoid tip has an amorphous structure. The germlings are released from the stalk and become attached to the substratum by an adhesive substance secreted from rhizoidal cells.  相似文献

Sargassum is a cosmopolitan brown algal genus spanning the three ocean basins of the Atlantic, Pacific and Indian Oceans, inhabiting temperate, subtropical and tropical habitats. Sargassum has been postulated to have originated in the Oligocene epoch approximately 30 mya according to a broad phylogenetic analysis of brown macroalgae, but its diversification to become one of the most widespread and speciose macroalgal genera remains unclear. Here, we present a Bayesian molecular clock study, which analyzed data from the order Fucales of the brown algal crown radiation (BACR) group to reconstruct a time-calibrated phylogeny of the Sargassum clade. Our phylogeny included a total of 120 taxa with 99 Sargassum species sampled for three molecular markers – ITS-2, cox3 and rbcLS – calibrated with an unambiguous Sargassaceae fossil from between the lower and middle Miocene. The analysis revealed a much later origin of Sargassum than expected at about 6.7 mya, with the genus diversifying since approximately 4.3 mya. Current geographic distributions of Sargassum species were then analyzed in conjunction with the time-calibrated phylogeny using the dispersal-extinction-cladogenesis (DEC) model to estimate ancestral ranges of clades in the genus. Results strongly support origination of Sargassum in the Central Indo-Pacific (CIP) region with subsequent independent dispersal events into other marine realms. The longer history of diversification in the ancestral CIP range could explain the much greater diversity there relative to other marine areas today. Analyses of these dynamic processes, when fine-tuned to a higher spatial resolution, enable the identification of evolutionary hotspots and provide insights into long-term dispersal patterns.  相似文献

13C nuclear magnetic resonance spectroscopy (NMR) has been used to study the metabolic flux of carbon through the intracellular pools of the isomeric hexitols d-altritol and d-mannitol in Himanthalia elongata. Natural abundance 13C NMR spectra of freshly collected plant material showed altritol as the dominant intracellular low-molecular-weight organic solute, with mannitol present at less than 20% of the altritol concentration. Plant material incubated in seawater medium with added 13C-enriched bicarbonate showed a rapid increase in the 13C signal due to mannitol, with an increase of more than 12-fold in under 48 h, in contrast to the altritol signal, which increased by approximately 2-fold over the same period. The intracellular mannitol signal decreased rapidly when 13C-enriched plant material was transferred to a non-enriched medium, while altritol showed a slower decline. These results are consistent with previous observations on the effects of salinity on the intracellular hexitol pools of H. elongata, suggesting that mannitol is more rapidly metabolised than altritol. Estimates of the half-time for tracer exchange support this view, with a half-time for the turnover of altritol (> 400 h) which is an order of magnitude greater than that for mannitol turnover (≈ 50h).  相似文献

Spontaneous formation and development of adventive embryos were observed in cauline leaves of Sargassum macrocarpum in laboratory culture. Semi-spherical swellings, which were 200–250 μm in diameter, arose from the surface of cauline leaves of thalli cultured for 4 months from zygotic embryos. Swellings became cylindrical protuberances and grew into ‘daughter’ thalli with one or two small cauline leaves. These thalli detached from ‘mother’ thalli and attached to the surface of culture vessels by rhizoids produced within 1 week after detachment. Each daughter thallus developed into an individual thallus exhibiting the same morphological processes as zygotic embryos.  相似文献

The relative growth rate of the brown alga Sargassum horneri (Turner) C. Agardh in the sea at Maizuru Bay, Japan, was investigated from summer to winter to examine the impact of declining solar radiation and temperature. The observed change in the growth rate between summer and winter was insignificant, in spite of a major decline in solar radiation and water temperature. The mean relative growth rate was 4.6% per day, equivalent to the daily net photosynthetic production previously reported for this species.  相似文献

Conspecific populations with different life‐cycle seasonality, particularly of their reproductive season, have been reported for many seaweed species. However, the number of genetic analyses of such seasonal populations is limited. Herein, based on the mitochondrial cytochrome oxidase subunit 3 gene (cox3) haplotype, we evaluated the genetic diversity of winter (January–March) and spring (April–June) fertile populations of the brown alga Sargassum horneri in Sado Island, Japan. Obviously, dominant cox3 haplotypes were different between seasons (even at a single site), and the N ST values indicated the presence of a strong genetic differentiation between the two seasonal populations. AMOVA analyses confirmed that most of the genetic diversity was between seasons rather than between regions. Herein, the possibility of allochronic isolation is discussed. Considering the phylogeny of the mitochondrial cox3 DNA sequence, autumn or winter seasonal populations such as those in this investigation have evolved independently at least twice in the species. The present study shows that a shift in breeding season could be expected to conserve the genetic diversity of S. horneri in a certain region.  相似文献

The release of the C11-hydrocarbon spermatozoid attractant hormosirene by eggs of the Australian fucoid Hormosira banksii has been determined quantitatively using a closed-loop extraction technique. The eggs secrete erotactin for more than 2 weeks with an exponentially decreasing rate. The initial secretion rate was estimated to be 75 fmol h?1 egg?1 and the total amount of hormosirene produced per egg to be 2.35 pmol (0.35 ng). The results were used to calculate the threshold range for spermatozoid chemotaxis in the pheromone gradient around eggs.  相似文献

Habitat-forming macroalgae play a central role in the ecology of temperate reefs worldwide but there exists a critical lack of knowledge about important processes such as dispersal and gene flow. Understanding dispersal and gene flow of habitat-forming seaweeds is particularly pertinent given that loss of habitat-forming algae is an increasingly prevalent problem worldwide. Here, we develop 10 microsatellite markers for the monotypic Phyllospora comosa, an important habitat-forming macroalga that has undergone massive declines on urbanised coastlines of Sydney, Australia. We characterise population genetic diversity and structure and estimate levels of dispersal and gene flow between the geographically isolated northern and southern populations in this subtidal macroalga.  相似文献

A new species, Sargassum boreale Yoshida et Horiguchi is described. It belongs to the subgenus Bactrophycus section Teretia, with cylindrical receptacles and is distinct from Sargassum confusum C. Agardh, S. pallidum (Turner) C. Agardh and Sargassum microceratium (Turner) C. Agardh in having a rather elongated stem with smooth surface and distantly issuing main branches, with narrow leaves. The distinction between S. boreale and these species is also revealed by a difference in internal transcribed spacer 2 (ITS‐2) sequences. In addition to the base substitutions, the existence of a large gap in S. boreale distinguishes this species from others. Sargassum boreale is distributed around Hokkaido and Saghalien to 50°N latitude. A key to the species of section Teretia is provided.  相似文献

Summary Processes of fertilization and zygote development inFucus distichus were studied by indirect immunofluorescence microscopy using anti- tubulin antibody and electron microscopy. Just after plasmogamy, sperm aster formation occurs during migration of a sperm nucleus toward an egg nucleus at the center of cytoplasm. Only sparse microtubules (MTs) exist around the egg nucleus. The sperm aster can be observed till karyogamy, but afterwards vanishes. Accompanying sperm aster formation, cortical MTs which are reticulately arranged develop further in the zygotes. In 4 h-old zygotes, characteristic structures which are composed of fine granular masses and consist of intermixed dense and lighter staining areas appear around the nucleus. These structures cannot be detected with anti- tubulin immunofluorescence microscopy. The two centrioles derived from the sperm separate and migrate to both poles. In 4 h-and 8 h-old zygotes, there are no defined MT foci around the zygote nucleus and MTs radiate from the circumference of it. In 12 h-old zygotes, each centriole has migrated to the poles and derivative centrioles are generated. The fine granular masses also migrate to both poles and finally disappear accompanying the appearance of numerous MTs radiating from the poles. Therefore, two distinct MT foci appear from 12 h onwards. Progressive stages of nuclear division were also examined with electron and immunofluorescence microscopy in 16 h-old zygotes. The sperm chloroplast with an eyespot and the sperm mitochondria with an intercristal tubular structure, which are distinctive from those of egg, can be detected after plasmogamy and karyogamy. The sperm chloroplast is still present in 16 h-old zygotes.  相似文献
