Summary Patch-clamp measurements were made on osteoblast-like cells isolated from embryonic chick calvaria. Cell-attachedpatch measurements revealed two types of high conductance (100–250 pS) channels, which rapidly activated upon 50–100 mV depolarization. One type showed sustained and the other transient activation over a 10-sec period of depolarization. The single-channel conductances of these channel types were about 100 or 250 pS, depending on whether the pipettes were filled with a low K+ (3mm) or high K+ (143mm) saline, respectively. The different reversal potentials under these conditions were consistent with at least K+ conduction. Whole-cell measurements revealed the existence of two types of outward rectifying conductances. The first type conducts K+ ions and activates within 20–200 msec (depending on the stimulus) upon depolarizing voltage steps from <–60 mV to >–30 mV. It inactivates almost completely with a time constant of 2–3 sec. Recovery from inactivation is biphasic with an initial rapid phase (1–2 sec) followed by a slow phase (>20 sec). The second whole-cell conductance activates at positive membrane potentials of >+50 mV. It also rapidly turns on upon depolarizing voltage steps. Activation may partly disappear at the higher voltages. Its single channels of 140 pS conductance were identified in the whole cell and did conduct K+ ions but were not highly Cl or Na+ selective. The results show that osteoblasts may express various types of voltage controlled ionic channels. We predict a role for such channels in mineral metabolism of bone tissue and its control by osteoblasts.  相似文献

Summary Ionic current steps were recorded with the patch-clamp technique from algal cells that had been prepared without enzyme treatment. Inward current steps with different conductance levels occurred, the lowest level being 7 pS. There were complex transitions between levels indicating either a lack of independence between single channels, or sublevels of a much larger conductance unit. The reversal potential was consistent with the permeant ion being Cl. Furthermore, when a different concentration of Cl was used in the patch electrode the reversal potential of the inward current shifted in a manner consistent with a Nernstian change in the Cl reversal potential. The frequency of the current steps was voltage dependent and suggestive of the hyperpolarization-activated Cl currents reported in voltage-clamp studies. Outward current steps, with conductances of 38 pS, were recorded when the membrane patch was depolarized by more than +120 mV. Their amplitude and frequency increased at more positive potentials. The current was probably carried by an efflux of cations through a different set of channels. The resting membrane potential, measured unambiguously without contamination from the tonoplast, was –190±5 mV.  相似文献

The single channel properties of recombinant gamma-aminobutyric acid type A (GABA(A))alphabetagamma receptors co-expressed with the trafficking protein GABARAP were investigated using membrane patches in the outside-out patch clamp configuration from transiently transfected L929 cells. In control cells expressing alphabetagamma receptors alone, GABA activated single channels whose main conductance was 30 picosiemens (pS) with a subconductance state of 20 pS, and increasing the GABA concentration did not alter their conductance. In contrast, when GABA(A) receptors were co-expressed with GABARAP, the GABA-activated single channels displayed multiple, high conductances (> or =40 pS), and GABA (> or =10 microM) was able to increase their conductance, up to a maximum of 60 pS. The mean open time of GABA-activated channels in control cells expressing alphabetagamma receptors alone was 2.3 +/- 0.1 ms for the main 30-pS channel and shorter for the subconductance state (20 pS, 0.8 +/- 0.1 ms). Similar values were measured for the 30- and 20-pS channels active in patches from cells co-expressing GABARAP. However higher conductance channels (> or =40 pS) remained open longer, irrespective of whether GABA or GABA plus diazepam activated them. Plotting mean open times against mean conductances revealed a linear relationship between these two parameters. Since high GABA concentrations increase both the maximum single channel conductance and mean open time of GABA(A) channels co-expressed with GABARAP, trafficking processes must influence ion channel properties. This suggests that the organization of extrasynaptic GABA(A) receptors may provide a range of distinct inhibitory currents in the brain and, further, provide differential drug responses.  相似文献

Summary Patch-clamp methods were used to study single-channel events in isolated oxyntic cells and gastric glands fromNecturus maculosa. Cell-attached, excised inside-out and outside-out patches from the basolateral membrane frequently contained channels which had conductances of 67±21 pS in 24% of the patches and channels of smaller conductance, 33±6 pS in 56% of the patches. Channels in both classes were highly selective for K+ over Na+ and Cl, and shared linear current-voltage relations. The 67-pS channel was activated by membrane depolarization, whereas the activity of the 33-pS channel was relatively voltage independent. The larger conductance channels were activated by intracellular Ca2+ in the range between 5 and 500nm, but unaffected by cAMP. The smaller conductance channels were activated by cAMP, but not Ca2+. The presence of K+ channels in the basolateral membrane which are regulated by these known second messengers can account for the increase in conductance and the hyperpolarization of the membrane observed upon secretagogue stimulation.  相似文献

Summary K+, Cl, and Ca2+ channels in the vacuolar membrane of tobacco cell suspension cultures have been investigated using the patch-clamp technique. In symmetrical 100mM K+, K+ channels opened at positive vacuolar membrane potentials (cytoplasmic side as reference) had different conductances of 57 pS and 24 pS. K+ channel opened at negative vacuolar membrane potentials had a conductance of 43 pS. The K+ channels showed a significant discrimination against Na+ and Cl. The Cl channel opened at positive vacuolar membrane potentials for cytoplasmic Cl influx had a high conductance of 110pS in symmetrical 100mM Cl. When K+ and Cl channels were excluded from opening, no traces were found of Ca2+ channel activity for vacuolar Ca2+ release induced by inositol 1,4,5-trisphosphate or other events. However, we found a 19pS Ca2+ channel which allowed influx of cytoplasmic Ca2+ into the vacuole when the Ca2+ concentration on the cytoplasmic side was high. When Ca2+ was substituted by Ba2+, the conductance of the 19 pS channel became 30 pS and the channel showed a selectivity sequence of Ba2+Sr2+Ca2+Mg2+=10.60.60.21. The reversal potentials of the channel shifted with the change in Ca2+ concentration on the vacuolar side. The channel could be efficiently blocked from the cytoplasmic side by Cd2+, but was insensitive to La3+, Gd3+, Ni2+, verapamil, and nifedipine. The related ion channels in freshly isolated vacuoles from red beet root cells were also recorded. The coexistence of the K+, Cl, and Ca2+ channels in the vacuolar membrane of tobacco cells might imply a precise classification and cooperation of the channels in the physiological process of plant cells.  相似文献

Using an 125I efflux assay, we have studied the expression of various types of chloride channels in isolated neonatal rat cardiomyocytes. Three different classes of anion conductances were distinguished: (1) a Cal2+-sensitive Cl conductance, triggered upon stimulation of the cells with endothelin-1 or Ca2+-ionophore; (2) a CAMP/protein kinase A-operated Cl conductance, activated by addition of forskolin. This anion channel could be identified as the Cystic Fibrosis Transmembrane conductance Regulator (CFTR-CI channel) by Western blotting as well as by its enhanced activity in cultures pretreated with the tyrosine kinase inhibitor genistein; (3) a distinct class of cell volume-regulated Cl channels, potentiated in the presence of endothelin-1 or the phosphotyrosine phosphatase inhibitor pervanadate. The potential role of each class of Cl channels in the generation and/or modulation of action potentials as well as in maintaining cell volume is discussed.  相似文献

Summary Properties of the single Cl channels were studied in excised patches of surface membrane from molluscan neurones using single-channel recording technique. These channels are controlled by Ca2+ and K+ acting on cytoplasmic and outer membrane surfaces, respectively, and by the membrane potential. The channels display about 16 intermediate conductance sublevels, each of them being multiples of 12.5 pS. The upper level of the channel conductance is about 200 pS. The channel behavior is consistent with an aggregation of channel-forming subunits into a cluster.  相似文献

Summary K+ channels in cultured rat pancreatic islet cells have been studied using patch-clamp single-channel recording techniques in cell-attached and excised inside-out and outside-out membrane patches. Three different K+-selective channels have been found. Two inward rectifier K+ channels with slope conductances of about 4 and 17 pS recorded under quasi-physiological cation gradients (Na+ outside, K+ inside) and maximal conductances recorded in symmetrical K+-rich solutions of about 30 and 75 pS, respectively. A voltage- and calcium-activated K channel was recorded with a slope conductance of about 90 pS under the same conditions and a maximal conductance recorded in symmetrical K+-rich solutions of about 250 pS. Single-channel current recording in the cell-attached conformation revealed a continuous low level of activity in an apparently small number of both the inward rectifier K+ channels. But when membrane patches were excised from the intact cell a much larger number of inward rectifier K+ channels became transiently activated before showing an irreversible decline. In excised patches opening and closing of both the inward rectifier K+ channels were unaffected by voltage, internal Ca2+ or externally applied tetraethyl-ammonium (TEA) but the probability of opening of both inward rectifier K+ channels was reduced by internally applied 1–5mm adenosine-5-triphosphate (ATP). The large K+ channel was not operational in cell-attached membrane patches, but in excised patches it could be activated at negative membrane potentials by 10–7 to 10–6 m internal Ca2+ and blocked by 5–10mm external TEA.  相似文献

Summary The patch-clamp technique for the recording of single-channel currents was used to investigate the activity of ion channels in the intact epithelium of the toad urinary bladder. High resistance seals were obtained from the apical membrane of tightly stretched tissue. Single-channel recordings revealed the activity of a variety of ion channels that could be classified in 4 groups according to their mean ion conductances, ranging from 5 to 59 pS. In particular, we observed highly selective, amiloridesensitive Na channels with a mean conductance of 4.8 pS, channels with a similar conductance that were not Na-selective and channels with mean conductance values of 17–58 pS that were mostly seen after stimulation of the tissue with vasopressin or cAMP. When inside-out patches from the apical membrane were exposed to 110mm fluoride, large conductances (86–490 pS) appeared.  相似文献

GABA-gated Cl channels were studied in the nervous system of the larval tobacco hawk moth, Manduca sexta, using electrophysiology, 36Cl uptake into membrane microsacs and immunocytochemistry. A GABA-induced increase in Cl conductance was recorded from a visually identifiable neurone (fg1) in the desheathed frontal ganglion. The response was insensitive to the vertebrate GABAA receptor antagonist, bicuculline, but was blocked by picrotoxinin. Bicuculline-insensitive, picrotoxinin-sensitive, GABA-stimulated 36Cl uptake was also detected in membrane microsacs prepared from the isolated larval M. sexta nervous system. Such receptors appear to be the major type of GABA receptor in larval nervous system membrane microsac preparations. An antibody raised against a 17 amino acid peptide, based on the predicted C-terminus of the Drosophila GABA receptor subunit (RDL), stained not only cell bodies, including that of fg1, but also the neuropile in the frontal ganglion, indicating the existence of RDL-like GABA receptor subunits in neurones of this ganglion. Thus, bicuculline-insensitive GABA-gated Cl channels are present in the larval nervous system of M. sexta.  相似文献

Summary Chloride channels from rat colonic enterocytes were studied using the patch-clamp technique. After removal of mucus, inside-out patches were excised from the apical membrane of intact epithelium located at the luminal surface. They contained spontaneously switching Cl channels with a conductance of 35–40 pS. The channels were blocked reversibly by anthracene-9-carboxylic acid (1mm).In excised patches from single enterocytes, isolated by calcium removal, the Cl channels were studied in more detail. TheI–V relation was linear between ±80 mV. The selectivity was I>Br>Cl=NO 3 >F=HCO 3 .Thirty pS Cl channels were also found on the basolateral membrane of crypts isolated by brief calcium removal. TheI–V curve of these Cl channels was also linear.The results provide direct evidence for the existence of Cl channels in the apical membrane of surface cells in colonic mucosa. The properties of these channels are similar to those previously observed when incorporating membrane vesicles into planar lipid bilayers. Both results support the validity of the theoretical models describing intestinal secretion.  相似文献

Large conductance channels were observed in the membrane of cultured cardiac cells of newborn rats studied with the patch-clamp technique in cell-attached and inside-out configurations. These channels were observed in 4% of the patches. In the cell-attached configuration they exhibited outward rectification and partial inactivation. In the inside-out configuration no rectification occurred but inactivation was present, mainly during hyperpolarizations. Two channels with large single unit conductances (400–450 pS) and one with a smaller conductance (200–250 pS) were frequently observed in the same patch. The two large channels generally had different kinetics. Under steady-state conditions the opening probability of the faster channel appeared to be voltage-independent. The slower channel was activated by depolarization. In asymmetrical solutions the permeability ratios P Na/P Cl were 0.03 and 0.24 for the larger and smaller channels, respectively; corresponding values for P Ba/P Cl were 0.04 and 0.09. It is proposed that in cardiac membranes the chloride permeability system is composed of widely dispersed microclusters forming grouped channels of different types and sizes.  相似文献

Summary Whole-cell patch-clamp recordings were made from freshly isolated human platelets. The pipette contained a high concentration of divalent cations, which permitted easy disruption of cell-attached membrane patches by suction. Single-channel currents were measured when the pipette contained isotonic BaCl2 or MgCl2 saline; over 30 sec –5 min an increasing number of channels appeared until conductance steps through individual channels could no longer be distinguished. The current-voltage relationship was curvilinear; chord conductance at –35 mV was 25 pS increasing to 45 to 52 pS at +45 mV. Ion substitution experiments showed the current to be primarily carried by Cl.E rev was shifted 30 mV/10-fold change in external Cl (replaced by gluconate), was similar with BaCl2 or MgCl2 in the pipette and was not significantly shifted by replacing external Na+ with K+. Addition of 1mm BAPTA to the MgCl2 pipette saline prevented activation of Cl currents; with isotonic CaCl2 internal saline, current appeared immediately upon patch rupture, suggesting that the Cl channels are dependent on internal Ca2+, 5-nitro-2-(3-phenylpropylamino)-benzoate, reported to block a Cl conductance in studies of rat epithelial cells, caused a potent flickery block and may be a useful tool with which to investigate the physiological role of Cl currents in human platelets.  相似文献

Summary Calcium channels in GH3 cells exhibit at least five conductance levels when examined in cell-attached or outside-out patches. These channels resemble the high threshold Ca2+ current in their range of activation and inactivation, and in their sensitivity to dihydropyridines (DHP). Mean open times for the five levels were brief (<1 msec) in control solutions but increased in the presence of BAY K 8644. In 100mm Ba2+ and BAY K 8644, the five predominant slope conductances were 8–9, 12–13, 16–18, 23–24, and 28 pS. The present study is the first report of multiple levels of the DHP-sensitive Ca2+ channel occurring with high frequency in native membranes. The range of conductance levels that we observed encompasses the range of conductances found for two other different types of Ca2+ channels and indicates that unit conductance should be used with caution as a distinguishing characteristic for identification of different channel types.  相似文献

Patch clamp studies show that there may be as many as seven different channel types in the plasma membrane of protoplasts derived from young leaves of the halophytic angiosperm Zostera muelleri. In whole-cell preparations, both outward and inward rectifying currents that activate in a timeand voltage-dependent manner are observed as the membrane is either depolarized or hyperpolarized. Current voltage plots of the tail currents indicate that both currents are carried by K+. The channels responsible for the outward currents have a unit conductance of approximately 70 pS and are five times more permeable to K+ than to Na+. In outside-out patches we have identified a stretch-activated channel with a conductance of 100 pS and a channel that inwardly rectifies with a conductance of 6 pS. The reversal potentials of these channels indicate a significant permeability to K+. In addition, the plasma membrane contains a much larger K+ channel with a conductance of 300 pS. Single channel recordings also indicate the existence of two Cl channels, with conductances of 20 and 80 pS with distinct substates. The membrane potential difference of perfused protoplasts showed rapid action potentials of up to 50 mV from the resting level. The frequency of these action potentials increased as the external osmolarity was decreased. The action potentials disappeared with the addition of Gd3+, an effect that is reversible upon washout.We would like to thank K. Morris and D. Summary Transmitter receptor ion channels from previously identified rabbit olfactory bulb neurons were studied by using a thin slice preparation in combination with patch-clamp measurements. PG cells, which closely resembled previously described periglomerular interneurons in their morphology, responded to microapplication of GABA, acetylcholine, norepinephrine and glycine with the activation of distinct ionic currents. JG cells, which belong either to the class of short axon cells or external tufted cells, never showed GABA responses. In mitral cells ionic currents activated by GABA, acetylcholine, norepinephrine and glutamate could be elicited. Further measurements of GABA-activated currents of PG cells were made and indicated that these cells expressed two different types of GABA receptors: one which showed fast desensitization with a decay time constant of about 5 s, and one which slowly desensitized with a decay time constant of about 20–30 s. Both types were completely inhibited by bicuculline methiodide (50 M). GABA receptors were not blocked by Zn2+ (0.1 mM). From the dose-response relationship of the peak GABA-activated currents, an apparent dissociation constant of 50 M was derived. From single channel measurements in excised outside-out patches, a single channel conductance of GABA-activated Cl currents of 24 pS was obtained during continuous application of the agonist. The measurements of unitary outward ion currents in unidentified neurons of the snailHelix pomatia with the patch-clamp technique in a cell-attached configuration showed the presence of several types of K+ channels. We investigated three types of K+ channels: with big (75 pS, BKC), medium (22 pS, MKC), and small (6.2 pS, SKC) unitary conductance. BKC and MKC were activated at a membrane potential of about –30 mV, whereas SKC were activated at more negative potentials, with opening probability of the latter channels significantly decreasing at potentials more positive than –30 mV. Pharmacological investigation showed that BKC and MKC channel activity disappeared after 8–10 min of cell patching with a pipette solution containing 60 mM Cs+, whereas MKC channels remained unaffected. BKC and MKC were proved to be more sensitive to TEA (20 mM), whereas SKC were selectively sensitive to 4-AP (10 mM). Cd2+ (100 µM) in the pipette solution decreased the unitary conductance of BKC channels by 55 % and that of MKC channels by about 31 %. In contrast, the unitary conductance of SKC channels was not changed by the above blocker. Bath application of 10 µM 5-HT showed that MKC were suppressed by 5-HT, whereas SKC and BKC were insensitive to this transmitter. It is supposed that BKC can be classified as big-conductance Ca2+-dependent K+ channels (KCa) or to 5-HT-sensitive K+ channels (S-type channels), while MKC correspond to intermediate-conductance KCa, and SKC channels comply well with the characteristics of A-type K+ current.Neirofiziologiya/Neurophysiology, Vol. 28, No. 6, pp. 250–259, November–December, 1996.  相似文献

Cell swelling has been shown to increase the permeability of the plasma membrane to ions such as K+, Na+, Ca2+ or Cl in many types of cells. In cardiac cells, swelling has been reported to increase Cl conductance, but whether cation-selective currents are activated by swelling is not known. Low Cl or Cl-free solutions were used to study the presence of such currents. Lowering the osmolarity of the extracellular medium from 299 to 219 mOsm resulted in cell swelling and concurrent activation of a cation-selective whole-cell current. When cell-attached patches were formed on swollen cells, opening of bursting single channel currents were observed in 18% of the patches studied. Ion substitution experiments indicated that the channel discriminated poorly among monovalent cations, and was impermeable to Cl. The channel was permeable to Ca2+. In symmetrical 140 mM K+, the current-voltage relation was linear with a single channel conductance of 36 ± 3 pS. Depolarization increased channel open probability. Interestingly, depending on the membrane patch studied, application of negative pressure to the pipette caused either an increase or a decrease in the open probability of the channel already activated by swelling. Thus, the sensitivity to tension of the swelling-activated channel was different from those of previously reported stretch-activated channels. Summary Cl channels from basolaterally-enriched rabbit outer renal medullary membranes are activated either by increases in intracellular Cl activity or by intracellular protein kinase A (PKA). Phosphorylation by PKA, however, is not obligatory for channel activity since channels can be activated by intracellular Cl in the absence of PKA. The PKA requirement for activation of Cl channels in certain secretory epithelia is, in contrast, obligatory. In the present studies, we examined the effects of PKA and intracellular Cl concentrations on the properties of Cl channels obtained either from basolaterally-enriched vesicles derived from highly purified suspensions of mouse medullary thick ascending limb (mTALH) segments, or from apical membrane vesicles obtained from two secretory epithelia, bovine trachea and rabbit small intestine. Our results indicate that the Cl channels from mTALH suspensions were virtually identical to those previously described from rabbit outer renal medulla. In particular, an increase in intracellular (trans) Cl concentration from 2 to 50 mm increased both channel activity (P o) and channel conductance (g Cl, pS). Likewise, trans PKA increased mTALH Cl channel activity by increasing the activity of individual channels when the trans solutions were 2 mm Cl. Under the latter circumstance, PKA did not activate quiescent channels, nor did it affect g Cl. Moreover, when mTALH Cl channels were inactivated by reducing cis Cl concentrations to 50 mm, cis PKA addition did not affect P o. These results are consistent with the view that these Cl channels originated from basolateral membranes of the mTALH.Cl channels from apical vesicles from trachea and small intestine were completely insensitive to alterations in trans Cl concentrations and demonstrated markedly different responses to PKA. In the absence of PKA, tracheal Cl channels inactivated spontaneously after a mean time of 8 min; addition of PKA to trans solutions reactivated these channels. The intestinal Cl channels did not inactivate with time. Trans PKA addition activated new channels with no effect on basal channel activity. Thus the regulation of Cl channel activity by both intracellular Cl and by PKA differ in basolateral mTALH Cl channels compared to apical Cl channels from either the tracheal or small intestine.We acknowledge the able technical assistance of Steven D. Summary Vesicles derived from epithelial cells of the colonic mucosa of the rat were fused to planar phospholipid bilayer membranes, revealing spontaneously switching anion-conducting channels of 50 pS conductance (at-30 mV with 200mm Cl each side). The equilibrium selectivity series was I (1.7)/Br (1.3)/Cl (1.0)/F (0.4)/HCO 3 (0.4)/Na (<0.11.). Only one dominant open-state conductance could be resolved, which responded linearly to Cl concentrations up to 600mm. The singlechannel current-voltage curve was weakly rectifying with symmetrical solutions. When 50 mV were exceeded at the highconductance branch of the curve, switching was arrested in the closed state. At more moderate voltages (±40 mV) kinetics were dominated by one open state of about 35-msec lifetime and two closed states of about 2 and 9-msec lifetime. Of these, the more stable closed state occurred less often. At these voltages one additional closed state of significantly longer lifetime (>0.5 sec) was observed.  相似文献
