The curves of UV (254 nm) induced pyrimidine dimers (endonuclease sensitive sites) vs. photoreactivating blacklight (365 nm) dose for cultured chick embryo fibroblasts reveal several new features. When the cells are incubated in the dark at 37 degrees following UV (254 nm) treatment, the efficiency of subsequent photorepair increases for the first few hours post-UV. The efficiency then remains approximately constant for several hours. Photorepair data obtained during this later period were plotted as the logarithm of dimer-enzyme complexes available for photoreactivation vs. blacklight (365 nm) dose. For a fixed damaging UV (254 nm) dose, the resulting curve has a shoulder of approximately 6-10 kJ/m2 followed by a straight line portion with a slope of magnitude about 1.5 X 10(-4) m2/J for UV doses up to 15 J/m2. For higher UV doses the shoulder remains about the same, but the slope decreases in magnitude. The shoulder is interpreted to indicate that a light-dependent step is necessary to activate the enzyme. The decrease in slope with increased UV dose together with some split photoreactivation dose experiments suggests that some site-to-site motion and multiple site function of the photorepair enzyme molecules may come into play at the higher levels of damage, but the evidence indicates that these complications are relatively unimportant at low UV doses.  相似文献

In darkness, sporalation of Penicillium claviforme Bainier CBS strain 126–23 was uniform. A single 10 J m−2 blue light pulse (in the range 360 to 520 nm) was sufficient to elicit an endogenous zonation rhythm (coremia formation); the higher the fluence, the longer the rhythm expression. The period was 30 to 36 h as long as sufficient fluence rate (3 μW m−2 for broad band blue light and 18.1 nmol m−2 s−1 for wavelengths 433, 457 or 465 nm) was continuously maintained; this rhythm became desynchronized in about a week. The period increased rapidly and reached 72 to 120 h as soon as the fluence rate was too low or dark was established. The 445,479 and 495 nm radiations evoked the rhythm in pulse experiments, whereas the rhythm was immediately desynchronized in continuous light. The participation of two photosensitive reactions in the rhythm regulation of P. claviforme is postulated.  相似文献

This work reports on the significance of UV-B absorbing compounds and DNA photorepair in protecting bean plants from UV-B radiation under nitrogen restriction. Bean plants grown in sterile vermiculite and irrigated periodically with a nutrient solution containing 12 or 1 mM of nitrate were irradiated with 22 μW cm−2 of UV-B, 4 h daily during 10 days after the first trifoliate leaf was developed. This intensity was equivalent to 3.2 kJ m−2 per day, approximately. PAR fluence rate was 350 ± 50 μmol quanta m−2 s−1. Control plants did not receive UV-B irradiation. Leaf expansion was negatively affected by both nitrate restriction and UV-B irradiation. This decrease was paralleled by a significant increase in starch, which was exacerbated by the combined action of both factors. Combined action of low nitrogen and UV-B also negatively affected the CO2 assimilation rate and the stomatal conductance. Formation of UV-B absorbing compounds was significantly increased by both UV-B irradiation and nitrogen restriction and this increase was exacerbated by the combination of both factors. No significant increase in dimer formation was detected in irradiated plants at the UV-B dose used. Significant dimer formation was only obtained by using very high UV-B intensities. This suggests that under an irradiation level of 22 μW cm−2 of UV-B, which is close to natural conditions, protective mechanisms such as pigment screening and DNA photorepair were probably sufficient to prevent any dimer formation in leaves.  相似文献

The induction and photorepair of pyrimidine dimers in DNA have been measured in the ultraviolet-irradiated, corneal epithelium of the marsupial, Monodelphis domestica, using damage-specific nucleases from Micrococcus luteus in conjunction with agarose gel electrophoresis. We observed that FS-40 sunlamps (280-400 nm) induced 7.2 +/- 1.0 X 10(-5) pyrimidine dimers per kilobase (kb) of DNA per J/m2. Following 100 J/m2, 50% and greater than 90% of the dimers were photorepaired during a 10- and 30-min exposure to photoreactivating light (320-400 nm), respectively. In addition, approximately 70% and approximately 60% of the dimers induced by 300 and 500 J/m2, respectively, were repaired by a 60-min exposure to photoreactivating light. The capacity of the corneal epithelium of M. domestica to photorepair pyrimidine dimers identifies this animal as a potentially useful model with which to determine whether pyrimidine dimers are involved in pathological changes of the irradiated eye.  相似文献

Pyrimidine dimer-DNA glycosylase activity prepared from Micrococcus luteus has been used to develop an enzyme-sensitive site assay for the detection and quantification of closely opposed pyrimidine dimers in the nuclear DNA of UV-irradiated yeast. With this assay, closely opposed dimers were found to be induced as a linear function of dose from 0 to 200 J/m2 (254 nm). Closely opposed dimer frequencies decreased during the incubation of UV-irradiated, excision repair-proficient cells under liquid-holding conditions in the dark and during post-irradiation exposure of excision-deficient cells to photoreactivating light. Incubation of excision-deficient cells in the dark had no effect on the frequency of closely opposed dimers for up to 16 h. These results indicate that closely opposed dimers in UV-irradiated yeast are subject to repair by enzymatic photoreactivation and/or by dark-repair processes dependent, at least in part, upon functions necessary for normal excision repair. The genetic and biochemical implications of these results are discussed.  相似文献

The development of effective purification protocols has permitted evaluation of the resistance of isolated scrapie prions to inactivation by UV irradiation at 254 nm. Prions were irradiated on ice with doses of UV light ranging up to 120,000 J/m2. UV dosimetry experiments, performed with Saccharomyces cerevisiae plasmid DNA or eucaryotic cells, indicated that under these experimental conditions an incident UV dose of 10 J/m2 formed 2 thymine dimers per 5.1 X 10(6) daltons of eucaryotic cell DNA. The D37 values for scrapie prions ranged from 17,000 to 22,000 J/m2; D37 values were also determined for virus, viroid, and enzyme controls. The number of pyrimidine dimers formed was correlated with the D37 values obtained for irradiated prions and target nucleic acids. The D37 value for bacteriophage M13, 6.5 J/m2, occurred at a dose that would form 0.56 dimers per target genome; the D37 for potato spindle tuber viroid, 4,800 J/m2, occurred at a dose that would form about 24 dimers per target viroid. The D37 value for an EcoRI restriction site, a target of 12 bases, occurred at a dose that would correspond to the formation of 0.89 thymine dimers per target site. The D37 value for prions occurred at a dose that would form 1 dimer in every 4 bases of single-stranded target nucleic acid. If the putative scrapie nucleic acid were double-stranded and readily repairable after UV damage, then the prion D37 value could reflect a nucleic acid molecule of 30 to 45 base pairs. While the D37 value for prions fell within the range of pure protein targets, our experiments cannot eliminate the possibility that a prion contains a small, highly protected nucleic acid molecule.  相似文献

Pimephales promelas (fathead minnow) embryos were used to show a correlation between induction of pyrimidine dimers in DNA and embryo death. Embryo killing was measured by a lack of heart-beat and blood circulation at 48 h post-ultraviolet radiation (UVR). When the embryos were exposed to various doses of UVR from a FS-40 sunlamp followed by exposure to photoreactivating light (PRL) (320-400 nm), the number of pyrimidine dimers decreased significantly. The photorepair of dimers was accompanied by a substantial increase in embryo survival. When embryo killing was examined as a function of the number of dimers present, dimers were identified as a major lesion involved in UVR-induced killing in these fish embryos. This in vivo study on photoreactivation treatment of fish embryos shows a direct association between UVR-induced pyrimidine dimers and embryo killing. In addition, when embryos were held in the dark for 9 h after UVR, 50% of the dimers were removed by excision repair.  相似文献

In crude extracts of the unicellular green alga Chlorella kessleri Fott et Novákóva grown in red light the activity of the glycolytic enzyme phosphofructokinase (PFK, EC 2.7.1.11) is about 40% higher compared to white light conditions giving the same dry matter production. Application of cycloheximide and density labelling with D2O indicate that this increase depends on the de novo synthesis of the enzyme: Twelve h of illumination at a fluence rate of 7 × 1018 quanta m−2 s−1 (11.6 μmol m−2 s−1) suffice to saturate the effect. In autotrophically grown algae maximal increase in enzyme saturate the effect. In autotrophically grown algae maximal increase in enzyme activity is reached in light of 680 nm, while in 3-(3,4-dichlorophenyl)-1,1-dimethyl urea (DCMU)-poisoned, glucose-fed cells, light of wavelengths around 727 nm is most effective. Involvement of a phytochrome-like photoreceptor is discussed.  相似文献

A method was developed for applying strains of Actinoplanes spp. that are hyper-parasites of oospores of Pythium ultimum to soil for reducing Pythium damping-off of plants. The method is based on the augmentation of soil with sporangia of a strain of Actinoplanes spp. borne on clay granules. In vitro sporulation of strains K30, W57, W257 and 25844 was: (1) greater for most strains on dilute Czapek-Dox agar than on four other agar media; (2) inhibited by continuous exposure to fluorescent light of intensity 4-150 μEm-2s-1, but not by exposure to 1 μEm-2s-1 or darkness; (3) greater at 20-307deg;C than at 10°C;and (4) greater at pH 6-7 than at pH 5 or 8. On solid carriers treated with dilute Czapek-Dox broth (pH 7) and incubated in the dark at 30°C for 3 weeks, strains sporulated poorly or not at all on vermiculite, perlite and rice hulls, but sporulated abundantly (107-109 colony-forming units (CFU) g-1 of granules) on montmorillonite clay granules. When strains 25844, W57 and W257 were applied as granules (4 107 - 4 × 108 CFU g-1) at 5% (w/w) to field plots infested with 750-1000 oospores of P. ultimum g-1 of soil, only strain 25844 consistently increased emergence and reduced root rot of table beets 8- 1 at 24-28 days after planting compared with controls. Strain 25844 (108 CFU g-1 of granules) at 1% (w/w) also increased the emergence of bush beans at 28 days after planting in P. ultimum-infested plots, but lower rates were ineffective. The inoculum viability of strain 25844 on clay granules declined 100-fold during 2 months of storage at 5-35°C, but thereafter remained stable for another 4 months. Strain 25844 on 6-month-old granules retained a high degree of hyper-parasitic activity toward oospores of P. ultimum. Augmentation of field soil with sporangia of Actinoplanes spp. is a valid approach to the biological control of pythium damping-off.  相似文献

An action spectrum for photoinduction of perithecial formationafter a prior 72 h dark growth period was determined in theUV region with apically growing mycelia of a sordariaceous fungus,Gelasinospora reticulispora. The spectrum exhibited a peak at280 nm. Quantum effectiveness of 280 nm irradiation was ca.1.7 times higher than that of 450 nm light. The number of peritheciainduced by UV radiation was saturated at a lower level as comparedwith blue light. UV radiation having a fluence greater thanthe saturation level decreased the number of induced perithecia.UV radiation that was given after a saturating exposure to inductiveblue light inhibited the inductive effect of blue light. Anaction spectrum for this inhibition exhibited a peak between260 and 270 nm. Monochromatic light beyond 350 nm had no inhibitoryeffect. Inhibitory effects of UV radiation given after inductiveblue light irradiation were observed in the fluence range wherephotoinductive effects of UV radiation became obvious. Therefore,the true height of the UV peak in the photoinduction actionspectrum,when free of distortion from the inhibitory effect, should behigher than the peak obtained in this study. Maize ( Zea mays L. hybrid ZP-704) and black pine ( Pinus nigra Arn.) were grown for five days at low fluence rate (0.4–4.0, μmol m–2 s−1) in blue or red light. Compared to red light of the same fluence rate, blue light effects in maize were repressive for the accumulation of Chita, b , carotenoids and light-harvesting complex-2 (LHC-2) proteins. The maximal reduction of proteins bound to the light-harvesting complex of photosystem 2 and pigments was attained at different fluence rate levels. In black pine, blue light compared to the red of the same fluence rate level either activated or reduced accumulation of pigments and LHC proteins, the effect being dependent on its fluence rate level. At fluence less than 3.0 μmol m−2 s−1 blue light was more efficient for the synthesis of Chi a, b and carotenoids, hut for LHC-2 complexes, fluence rates between 0.4 and 1.5 [μmol m−2 s−1 were more effective. In pine the effects of the two lights on the accumulation of pigments and LHC proteins were demonstrated separately and were dependent on fluence rate level. This suggests irradianoe-controlled activation/deactivation of the photoreceptor at the level of the cell.  相似文献

Abstract.  [His7]-corazonin is a neuropeptide that induces dark coloration in locusts. This study examined the effect of temperature on body colour in the migratory locust, Locusta migratoria . L. Injection of a low dose (1 pmol) of [His7]-corazonin caused albino nymphs to develop dark coloration at 25–34 °C, but little darkening occurred at 38 and 42 °C. However, injection of a high dose (10 pmol) induced darkening even at these high temperatures. Transfer of nymphs injected with 1 pmol of [His7]-corazonin from 30 to 42 °C, and vice versa, indicated that temperature influenced darkening at any time after injection. Measurements of the luminance of the pronotum were made using commercially available computer software to follow continuous changes in darkening of the body. The body colour of nymphs injected with [His7]-corazonin was reddish-brown at 25 °C, blackish at 30 and 34 °C, and greyish or whitish at 38 and 42 °C. High temperature also suppressed darkening in a normal (pigmented) strain. Most nymphs transferred from 30 to 42 °C during the first three stadia developed a light colour in the fifth stadium without the striking black patterns that are typically manifested in gregarious nymphs at lower temperatures. Such individuals developed black patterns in the fifth stadium when injected with [His7]-corazonin at a mid stage of the previous stadium. These results indicate that high temperature may induce light body coloration by suppressing the release of [His7]-corazonin in normal locusts.  相似文献

Three groups, each of 6 Hereford cattle, were infected by the i.v. inoculation of 1010, 108 or 106 Anaplasma marginale-infected erythrocytes. The mean time taken to reach a 1% parasitaemia was 7.3, 13.9 and 19.9 days in the 1010, 108 and 106 infection dose groups, respectively. The rates of increase in parasitaemias during the exponential phase of parasite multiplication were similar for the 3 groups (doubling time 0.9 days). The exponential increase of the parasitaemia in the 1010 dose group extended to a higher level than in the 108 or 106 dose groups (to approximately 10% compared with 3%). The mean maximum parasitaemia attained in the 1010, 108 and 106 infection dose groups was 23.7, 14.7 and 8.7%, respectively. The time taken to reach the treatment criterion (packed cell volume decrease to 15% or lower) from a 1% parasitaemia was similar for the 3 groups. These results showed that the pathological outcome (anaemia) of anaplasmosis were similar over the 10 000-fold infective dose range tested.  相似文献

Metathetical exchange between carbon dioxide and the tin(II) dimer, {Sn[N(SiMe3)2](μ-OBu1)}2 (3) has been observed to cleanly produce the two new heteroleptic tin(II) dimers, Sn[N(SiMe3)2](μ-OBut)2Sn(OSiMe3) (6) and [Sn(OSiMe3)](μ-OBut)]2 (7]). In addition, reaction of 3 with I equiv, of tert-butylisocyanate (8), at 25°C, quantitatively provides 6, and with 2 equiv., quantitatively provides 7. Likewise 6 reacts with 1 equiv, of 8 to quantitatively provide 7. The mechanism for these latter processes has been investigated by low temperature 1H NMR spectroscopy which reveals that metathetical exchange does not involve the tri-coordinate tin(II) centers of the dimeric structures, but rather, it occurs, in each case, via the transient monomeric tin(II) species, Sn[N(SiMe3)2](μ-OBut) (4), that undergoes metathesis to produce, initially the open dimer intermediate, Sn(OCNBut)(OSiMe3)(μ-OBut)Sn(OBut) (OSiMe3) (12), that is observed at −10°C. Subsequent redistribution reactions then generate the final products that are observed. Together, these mechanistic details provide additional support for the ‘monomeric tin(II)’ hypothesis proposed earlier for metathetical exchange between XCO and Sn[N (SiMe3)2]2 (1).  相似文献

The infrared, visible and nuclear magnetic resonance spectra of protochlorophyll a and vinylprotochlorophyll a in dry non-polar solvents (carbon tetrachloride, chloroform, cyclohexane) are presented and interpreted in terms of dimer interaction.

The infrared spectra in the 1600–1800 cm−1 region clearly show the existence of a coordination interaction between the C-9 ketone oxygen function of one molecule and the central magnesium atom of another molecule. Infrared spectra in the OH stretching region (3200–3800 cm−1) provide a valuable test of the water content in the samples.

The analysis of the absorption and circular dichroism spectra of protochlorophyll a and vinylprotochlorophyll a in carbon tetrachloride demonstrates the existence of a monomer-dimer equilibrium in the concentration range from 10−6 to 5 · 10−4 M. The dimerization constants are (6±2) · 105 1 · M−1 for protochlorophyll a and (4.5±2) · 105 1 · M−1 for vinylprotochlorophyll a at 20 °C. The deconvolution of visible spectra in the red region has been performed in order to obtain quantitative information on the dimer structure. Two models involving a parallel or a perpendicular arrangement of the associated molecules are considered.

From 1H NMR spectra, it appears that the region of overlap occurs near ring V, in agreement with the interpretation of the infrared spectra.  相似文献

Low-molecular-weight chitosan were prepared using 85% phosphoric acid at different reaction temperatures and reaction time. At room temperature, the viscosity average-molecular weights (Mv) of chitosan decreased to 7.1×104 from 21.4×104 after 35 days treatment. The degradation rate decreased with increasing hydrolysis time. The yields of chitosan also continuously decreased from 68.4 to 40.2% after 35 days. At 40, 60 and 80 °C, the molecular weight decreased to 3.70×104, 3.50×104 and 2.00×104 on 8 h hydrolysis, respectively. The yields of chitosan remain at a high level compared with that at room temperature and were 86.5, 71.4 and 61.3% at 40, 60 and 80 °C treatment, respectively. The different reaction time gave chitosan with different molecular weights. At 60 °C, the molecular weight of products decreased to 7.40×104 from 21.4×104 within 4 h, then decreased slowly to 1.90×104 in 15 h. It was also found that the water-solubility of chitosan increased as the molecular weight decreased. Results show the changes in yields and molecular weight of chitooligomers were strongly dependent on the reaction temperature and reaction time.  相似文献

The structure of the 7S globulin from Phaseoulus vulgaris L in dilatue solutions has been studied by small angle X-ray scattering (SAXS), by quasi-elastic light scattering (Q ELS), by circular dichroism spectroscopy (c.d.), and by precise density measurements. The molar mass, the radius of gyration, the volume, the maximum dimension and the diffusion coefficient were determined as M = 1.45 × 105 g mol−1, RG = 4.05 nm, V = 300- nm3, L = 13.0 nm and D20,w0 = 4.5 × 10−7 cm2 s−1, respectively. The molecule has an asymmetrical shape with the dimensions 12.5 × 12.5 × 3.75 nm. The secondary structure of the 7S globulin is characterized by a small portion of -helical structure (14%) and a marked content of β-structure (18%).  相似文献

The green macroalga Ulva pertusa Kjellman produced UV‐B absorbing compounds with a prominent absorption maximum at 294 nm in response only to UV‐B, and the amounts induced were proportional to the UV‐B doses. Under a 12:12‐h light:dark regime, the production of UV‐absorbing compounds occurred only during the exposure periods with little turnover in the dark. There was significant reduction in growth in parallel with the production of UV‐B absorbing compounds. The polychromatic action spectrum for the induction of UV‐B absorbing compounds in U. pertusa exhibits a major peak at 292 nm with a smaller peak at 311.5 nm. No significant induction was detected above 354.5 nm, and radiation below 285 nm caused significant reduction in the levels of UV‐B absorbing compounds. After UV‐B irradiation at 1.0 W·m?2 for 9 h, the optimal photosynthetic quantum yield of the samples with UV‐B absorbing compounds slightly increased relative to the initial value, whereas that of thalli lacking the compounds declined to 30%–34% of the initial followed by subsequent recovery in dim light of up to 84%–85% of the initial value. There was a positive and significant relationship between the amount of UV‐B absorbing compounds with antioxidant activity as determined by the α,α‐diphenyl‐β‐picrylhydrazyl scavenging assay. In addition to mat‐forming characteristics and light‐driven photorepair, the existence and antioxidant capacity of UV‐B absorbing compounds may confer U. pertusa a greater selective advantage over other macroalgae, thereby enabling them to thrive in the presence of intense UV‐B radiation.  相似文献

Photoreactivation of Escherichia coli after inactivation by a low-pressure (LP) UV lamp (254 nm), by a medium-pressure (MP) UV lamp (220 to 580 nm), or by a filtered medium-pressure (MPF) UV lamp (300 to 580 nm) was investigated. An endonuclease sensitive site (ESS) assay was used to determine the number of UV-induced pyrimidine dimers in the genomic DNA of E. coli, while a conventional cultivation assay was used to investigate the colony-forming ability (CFA) of E. coli. In photoreactivation experiments, more than 80% of the pyrimidine dimers induced by LP or MPF UV irradiation were repaired, while almost no repair of dimers was observed after MP UV exposure. The CFA ratios of E. coli recovered so that they were equivalent to 0.9-, 2.3-, and 1.7-log inactivation after 3-log inactivation by LP, MP, and MPF UV irradiation, respectively. Photorepair treatment of DNA in vitro suggested that among the MP UV emissions, wavelengths of 220 to 300 nm reduced the subsequent photorepair of ESS, possibly by causing a disorder in endogenous photolyase, an enzyme specific for photoreactivation. On the other hand, the MP UV irradiation at wavelengths between 300 and 580 nm was observed to play an important role in reducing the subsequent recovery of CFA by inducing damage other than damage to pyrimidine dimers. Therefore, it was found that inactivating light at a broad range of wavelengths effectively reduced subsequent photoreactivation, which could be an advantage that MP UV irradiation has over conventional LP UV irradiation.  相似文献
