Screening of microorganisms producing a new kind of flocculating substance was carried out. A fungus that produced a large amount of a strong microbial cell flocculant was isolated from soil. The fungus was classified as a Paecilomyces species. The substance produced showed very interesting and unique flocculation characteristics. It could efficiently flocculate all tested suspended solids in aqueous solution, and flocculation was practically not affected by ionic strength, pH or temperature.

The influence of components of the culture medium upon the flocculant production by Paecilomyces sp. 1-1 was studied. Addition of hydrolysates of casein such as casamino acid and polypeptone stimulated the production of the flocculant. A high concentration of calcium ions also significantly increased the mycelial growth and the production of the flocculant. Under the optimum cultural conditions, a 500-fold dilution of the culture filtrate could efficiently flocculate and sediment E. coli cells.  相似文献

Malic acid consumption by Saccharomyces cerevisiae was studied in a synthetic medium. The extent of malic acid degradation is affected by its initial concentration, the extent and the rate of deacidification increased with initial malate concentration up to 10 g/l. For malic acid consumption, an optimal pH range of 3–3.5 was found, confirming that non-dissociated organic acids enter S. cerevisiae cells by simple diffusion. A full factorial design has been employed to describe a statistical model of the effect of sugar and malic acid on the quantity of malate degraded (g/l) by a given amount of biomass (g/l). The results indicated that the initial malic acid concentration is very important for the ratio of malate consumption to quantity of biomass. The yeast was found to be most efficient at higher levels of malate.  相似文献

A continuous process to deacidify apple juices and cider was developed by entrapping Oenococcus oeni in LentiKats, a new polyvinyl alcohol hydrogel. For a residence time of 0.55 h, malic acid was completely converted into lactic acid when the LentiKats bioreactor was fed with apple juice at pH 4.46 and 3.95 and thirty three percent of initial malic acid (6.7 g l–1) was converted when the initial apple juice pH was 2.30. The optimal malolactic activity of this bioreactor was obtained at 30°C and a 50% reduction in malic acid conversion was measured between 15°C and 20°C, at a residence time of 0.3 h. The LentiKats bioreactor gave better performance than continuous reactor with Oenococcus oeni immobilised in alginate beads (specific malic acid consumption increased by a factor of 4.6) due to the increase of the ratio external surface to volume, allowing better mass transfer.  相似文献

Optimization of microalgal biomass harvesting is essential to produce effective and optimum outcomes that can contribute towards a feasible and economical harvesting technique. Two Chlorella species were used, namely, C. vulgaris and C. sorokiniana UKM3. Two essential factors affecting microalgal biomass harvesting via flocculation, namely, the initial pH of the microalgal broth and flocculant (chitosan) concentration were studied. The optimization process was conducted by using a response surface methodology (RSM) based on the model of face-centered-central composite design (FC-CCD). The potential for biofuel application of the harvested biomass was evaluated based on the production of fatty acid methyl esters (FAMEs) by transesterification. The quadratic models obtained from the RSM significantly fitted the experiment data as the p-values were less than 0.05. The initial pH of the microalgal suspension was found to have a more significant effect on the flocculation process than flocculant concentration. For C. vulgaris, the highest flocculant efficiency of 98.7% was obtained at a chitosan concentration of 0.2 g L?1 and an initial pH of 12.0, whereas for C. sorokiniana UKM3, at 0.15 g L?1 of chitosan and initial pH of 12.0 produced the highest efficiency of 97.1%. The harvested biomass of both species exhibited a high content of palmitic acid (C16:0) with 29.74 wt% and 11.81 wt% of dry biomass for C. vulgaris and C. sorokiniana UKM3, respectively. This study showed that Chlorella species can be harvested efficiently using the flocculation process and manifested an excellent potential for biodiesel production where palmitic acid (C16:0) is one of the main compounds for high-acid oil-biodiesel.

DL-Malic acid-grown cells of the yeast Hansenula anomala formed a saturable transport system that mediated accumulative transport of L-malic acid with the following kinetic parameters at pH 5.0: Vmax, 0.20 nmol.s-1.mg (dry weight)-1; Km, 0.076 mM L-malate. Uptake of malic acid was accompanied by proton disappearance from the external medium with rates that followed Michaelis-Menten kinetics as a function of malic acid concentration. Fumaric acid, alpha-ketoglutaric acid, oxaloacetic acid, D-malic acid, and L-malic acid were competitive inhibitors of succinic acid transport, and all induced proton movements that followed Michaelis-Menten kinetics, suggesting that all of these dicarboxylates used the same transport system. Maleic acid, malonic acid, oxalic acid, and L-(+)-tartaric acid, as well as other Krebs cycle acids such as citric and isocitric acids, were not accepted by the malate transport system. Km measurements as a function of pH suggested that the anionic forms of the acids were transported by an accumulative dicarboxylate proton symporter. The accumulation ratio at pH 5.0 was about 40. The malate system was inducible and was subject to glucose repression. Undissociated succinic acid entered the cells slowly by simple diffusion. The permeability of the cells by undissociated acid increased with pH, with the diffusion constant increasing 100-fold between pH 3.0 and 6.0.  相似文献

The pH, viability and flocculation during the growth of Saccharomyces cerevisiae NCYC 1214 in rich medium (YEPD) and in chemically defined medium (YNB) were compared. As the pH fell from 5.3 to 2.2, the viability fell to 20% and no flocculation was observed in YNB medium, despite the high flocculation in buffer. With the addition of 50 mM succinic acid and 3.1 mM Ca2+, similar growth to YEPD, with 99% of viability and flocculation in culture medium, was obtained.  相似文献

Due to their small size and low concentration in the culture medium, cost-efficient harvesting of microalgae is a major challenge. We evaluated the potential of cationic starch as a flocculant for harvesting microalgae using jar test experiments. Cationic starch was an efficient flocculant for freshwater (Parachlorella, Scenedesmus) but not for marine microalgae (Phaeodactylum, Nannochloropsis). At high cationic starch doses, dispersion restabilization was observed. The required cationic starch dose to induce flocculation increased linearly with the initial algal biomass concentration. Of the two commercial cationic starch flocculants tested, Greenfloc 120 (used in wastewater treatment) was more efficient than Cargill C*Bond HR 35.849 (used in paper manufacturing). For flocculation of Parachlorella using Greenfloc 120, the cationic starch to algal biomass ratio required to flocculate 80% of algal biomass was 0.1. For Scenedesmus, a lower dose was required (ratio 0.03). Flocculation of Parachlorella using Greenfloc 120 was independent of pH in the pH range of 5 to 10. Measurements of the maximum quantum yield of PSII suggest that Greenfloc 120 cationic starch was not toxic to Parachlorella. Cationic starch may be used as an efficient, nontoxic, cost-effective, and widely available flocculant for harvesting microalgal biomass.  相似文献

The marine myxobacterium strain NU-2, which can grow on high concentrations (up to 7%) of NaCl, was isolated from a salt soil sample collected from the coast of the Huanghai Sea, China. Morphological properties and 16S rDNA sequence analysis indicated that the isolate is a novel species related to the genus Nannocystis. Nannocystis sp. NU-2 produced a new kind of flocculating substance in a starch medium with a yield of 14.8 g l–1. The NU-2 flocculant was composed of 40.3% proteins and 56.5% polysaccharides, of which glucose, mannose and glucuronic acid were the principal constituents in the relative proportions of 5:4:1. The flocculation activity of the NU-2 flocculant depends strongly on cations such as Fe3+ and Al3+. When a 30 mg l–1 FeCl3 solution is present in kaolin clay suspension, 30 mg l–1of the flocculant produced a high flocculating activity value of 90%, which remained unchanged over an extensive pH range (pH 2.0–13.0). The flocculant was tested for its ability to bleach dyeing liquors, and the bleaching activities were 98.2% for acid red in 100 mg l–1of the flocculant and 99.0% for direct emerald blue in 50 mg l–1of the flocculant under test conditions. Summary The flocculation of Chlorella vulgaris by Lactobacillus casei was studied to determine whether the latter could act as a suitable flocculant for the removal of Chlorella from algal ponds. The flocculating activity of the Lactobacilli was caused by the bacterial cells themselves, and not by diffusible products of bacterial metabolism. Diffusible products of algal metabolism inhibited flocculation. For algae resuspended in water, the best flocculation occurred at pH values less than 3.5 where the charges on the bacterial and algal cells were opposite. For flocculation at least one bacterium was required for every algal cell; in terms of cell concentrations,10 mg/l of bacteria were required to flocculate an algal suspension of 1,000 mg/l. The mechanism of flocculation implied by the results is that positively charged cells of L. casei adsorb to the surface of negatively charged cells of C. vulgaris neutralizing the charge and thus destabilizing the algal suspension. Because of the low pH required and because diffusible products of algal metabolism inhibit the flocculation, it is unlikely that L. casei could be usefully employed as a flocculant of Chlorella from algal ponds.  相似文献

The structure gene FLO1 from Saccharomyces cerevisiae W303-1A encoding a flocculation protein and the G418 resistance gene kanMX from plasmid pUG6 were amplified by PCR method. The expression vector pYX212 harboring FLO1 gene and kanMX gene was transformed into Angel yeast. The transformant Angel yeast F6 was obtained and showed strong and stable flocculation ability during 20 batches inoculation. And the flocculation ability of the transformant Angel yeast F6 showed no difference in the medium with the initial pH ranging from 3.5 to 6.0. Noteworthily, the flocculation onset of the transformant strain was in the early stationary growth phase, not coincident with the glucose depletion in the cultural medium. And in the experiment the ethanol yield and other properties of the transformant Angel yeast F6 were similar to those of the wild-type strain, although its fermentation time was a little slower comparing with the wild-type strain. Those would be potential application for yeast cells to separate and recycle in the fuel ethanol industry.  相似文献

The aim of this work was to optimize acid stress conditions for induction of acid tolerance response (ATR) in the biocontrol agent Pantoea agglomerans and study the effect of ATR induced on the ability to survive under acidic conditions. Initially, Pantoea agglomerans was grown in mild acidic conditions (pH 6.0, 5.5, 5.0 and 4.0) in order to induce ATR. The highest ATR was induced at initial pH of 5 using malic or citric acid. A first in vitro experiment was carried out. Thus, basal liquid medium at different pHs (3.0, 3.5, 4.0 and non-acidified) were then inoculated with acid-adapted and non-adapted inocula of P. agglomerans and survivals were examined during incubation at 25 or 4 °C. It was found that acid adaptation enhanced the survivals of Pantoea agglomerans CPA-2 cells at pH levels at which the cells were unable to grow (<3.5 and 4.0, at 25 and 4 °C, respectively). In contrast, in pH levels at which the cells were able to grow (pH 4.0 at 25 °C and non-acidified medium at 25 and 4 °C) no-differences were found between adapted and non-adapted cells. In in vivo tests, adapted and non-adapted cells were inoculated in wounds on mandarins and pome fruits. No differences were found between adapted and non-adapted cells and biocontrol efficacy was maintained. The present study demonstrated that exposure of Pantoea agglomerans to mild acidic conditions could induce acid resistance in this biocontrol agent.  相似文献

The effects of different metal ions, carbohydrates, heat and enzymatic treatments on the flocculation of yeast cells caused by a flocculant type of Lactobacillus fermentum were investigated. Calcium ion was required at pH 3.0, 4.5 and 6.2 for complete flocculation. Some flocculation was detected at pH 4.5 even if no calcium was added to the system. Manganese and magnesium ions were capable of partly replacing calcium at pH 6.2. Mannose had an inhibitory effect on flocculation, while other sugars had no effect. Protease is capable of inhibiting the flocculating ability of bacterial cells. Heat treatment of bacterial cells also destroyed the flocculating ability and the effectiveness of this treatment was pH dependent. No effect of protease or heat treatment on yeast cells was found. The results suggest that a cell wall component of L. fermentum, mannan residues of yeast cells and divalent ions were involved in this phenomenon.  相似文献

Schizosaccharomyces yeasts can be used for deacidification of grape musts. To this aim, we studied malic acid degradation by yeasts included in double layer alginate beads in a bubble column reactor. Use of immobilized micro-organisms allowed a continuous process with high dilution rates giving a deacidification capacity of 0.032 g of malate/hour/dm3/g of beads. Summary The effect of various culture conditions on growth kinetics of a Schizosaccharomyces pombe strain were investigated in order to facilitate the potential use of this organism in enology. The effect of ethanol was characterized as a 80 g·l–1 critical concentration. An optimal pH range of 3.5–4.0 was found. Growth was slightly affected by malic acid at pH 3.0 but was shown to decrease drastically at pH 5.5. Aerated cultures had increased rates of growth and malate degradation than those grown under anaerobiosis, although the specific rate of malate degradation remained fairly constant. Taking into account biomass synthesis data it would seem likely that under anaerobiosis malate can replenish the Krebs' cycle for amphibolic precursor synthesis and hence avoid potential growth limitation. Summary A synthetic medium containing 9 g/l sucrose was hydrolyzed in a novel hybrid reactor. A minimum hydraulic retention time (HRT) of 9.9 h, with a gas production rate of 1.07 m3/m3·d, was obtained without continuous neutralization. A viable anaerobic cell count of 109 organisms/ml was obtained in the reactor fluid. The results showed that both pH and temperature significantly influenced the type and concentration of the various metabolites formed. These include ethanol, formic, acetic, propionic and butyric acids as primary metabolites and caproic acid as secondary metabolite. From the results obtained, it is suggested that to obtain the energetically most favourable products, a substrate pH of 6.5 and a temperature of 35°C must be used in anaerobic acidogenic digesters.  相似文献

Summary Transport and utilization of malic acid by the yeast Hansenula anomala are subject to glucose repression. Derepressed diploid mutant strains were obtained by hybridization of derepressed haploid mutant strains of opposite mating type. Six diploid mutant strains displayed derepressed behaviour with respect to malic acid utilization in the presence of glucose up to 30% (w/v). Three of these diploid mutant strains, as compared with the parent strain, were able to degrade completely malic acid in grape juice without fermenting the sugars. In addition, using one diploid mutant strain together with a strain of the wine yeast Saccharomyces cerevisiae, it was possible to carry out a mixedmicrovinification in which deacidification occurred simultaneously with alcoholic fermentation.  相似文献

A biopolymer flocculant-producing bacterium, strain PY-90, was isolated and considered to belong to Bacillus subtilis. For the production of biopolymer flocculant by strain PY-90, a medium containing 2 to 5% l-glutamic acid as a nitrogen source was suitable. The biopolymer flocculant was a homopolymer composed of glutamic acid residues and was presumed to be poly(γ-glutamic acid). In kaolin suspension, the highest flocculating activity was attained at the biopolymer flocculant concentration of 20 mg/l. The flocculating activity was increased by the addition of Ca2+, and the optimum concentration of which was about 2 to 8 mM. The flocculating activity was high in an acidic pH range of 3.0 to 5.0, and decreased upon heating at 100°C.  相似文献

The influence of pH on growth, and lactic acid and bacteriocin production byLactococcus lactis subsp.lactis 140 NWC was studied during batch fermentation in a lactose-based complex medium. Growth and lactic acid production were modelled using a simple logistic equation while substrate consumption was found to be a function growth and lactic acid production rate. The optimal pH for growth and lactic acid production was between 6.0 and 6.5. Bacteriocin production showed primary metabolite kinetics. pH had a dramatic effect on the production of the bacteriocin, lactococcin 140. A maximum activity of 15.4 × 106 AU (arbitrary units) 1–1 was obtained after 7 h at pH 5.5. Maximum bacteriocin activity was achieved before the end of growth and was followed by a decrease in activity, which was due to adsorption to the cells of the producing organism, possibly followed by degradation by specific proteases. Both bacteriocin production and degradation rates were higher at pH 5.0 and 5.5, resulting in sharper activity peaks than at pH 6.0 or 6.5. On the basis of the experimental results a qualitative model for bacteriocin production is proposed.  相似文献

Citrobacter freundii DSM 30040 immobilized on modified polyurethane carrier particles PUR 90/16 was used for continuous glycerol fermentation in an anaerobic fixed bed reactor with effluent recycle and pH control (fixed bed loop reactor). The fermentor was run with buffered mineral medium under growth conditions resulting in the permanent renewal of active biomass. The effects of glycerol concentration in the feed, dilution rate (D), pH and temperature (T) were investigated to optimize the process. With 400 mm glycerol in the feed, pH 6.9, T = 36°C and D = 0.5 h–1 the maximum productivity could be determined as 8.2 g/l per hour of 1,3-propanediol.  相似文献
