Weaned piglets were fed a wheat based diet either non-supplemented or supplemented with a multi-enzyme preparation or a xylanase mono-enzyme preparation, respectively. Both enzyme preparations increased live weight gain nonsignificantly, but only animals of the xylanase group showed a trend (p = 0.076) for an improved feed conversion. Only precaecal digestibility of total amino acids was improved significantly when the mono-enzyme preparation was added. Improvements of digestibility of crude fat, crude protein and starch did not reach the significance level. Both enzyme preparations reduced jejunal viscosity, however viscosity in the colon was only reduced by the mono-enzyme preparation. Both enzymes significantly reduced Lactobacillus spp. cell numbers as well as bacterial metabolites in the stomach and showed similar nonsignificant modifications in jejunum contents except for acetate in the mono-enzyme group. Total jejunal bile acids were unchanged. Compared to the control, the ratio of the main conjugated to the main deconjugated bile acid was significantly higher in the mono-enzyme group. This study has shown that the mono- and multi-enzyme preparation can lead to improved performance in wheat based diets for piglets. Like in poultry, the main mode of action seems to be the reduction of small intestinal viscosity. However, the generation of fermentable carbohydrates by the multi-enzyme preparation may mask beneficial effects on performance due to the development of an active bile acid deconjugating microbiota in the small intestine.  相似文献

Effects of field beans with various tannin content and exogenous enzyme mixture containing tannase, pectinase and xylanase activities on N-corrected dietary apparent metabolisable energy (AMEn), coefficients of dry matter (DMR) and nitrogen retention (NR), fat digestibility, gastrointestinal tract (GIT) development, jejunal villus morphometry, ileal digesta viscosity and sialic acid were examined. Birds’ growth performance and energy conversion ratio (ECR) were also measured. Birds were fed one of eight mash diets. The Control diet contained as major ingredients wheat (400 g/kg) and soybean meal (SBM) (127 g/kg and 221 g crude protein/kg and 12.83 MJ AMEn/kg. To reduce nutrient density, the Control diet also contained washed sand at 119 g/kg. Another three diets containing 300 g/kg of each of three experimental field bean cultivar samples in replacement for SBM and sand were also mixed. Each diet was fed to nine pens with two male Ross 308 broilers. Diets high in tannin had low AMEn, ECR, DMR and NR (p < 0.001). Feeding field beans increased (p < 0.001) the weights of the pancreas and the proventriculus and gizzard (PG) of the birds. Supplementing diets with the enzyme mixture improved (p < 0.001) feed conversion efficiency, AMEn and all nutrient utilisation coefficients despite the tannins in diets. The enzyme mixture reduced ileal digesta viscosity (p < 0.001) and the weight of pancreas, total GIT and PG (p < 0.05) of the birds. It can be concluded that the feeding value of field beans with different tannin contents may vary when fed to broilers. The supplementation of the enzyme mixture improved the feeding value of diets for broilers. The beneficial effect of the addition of the enzyme mixture seems to be mediated through reduced ileal digesta viscosity and improved nutrient availability.  相似文献

The effects of age of growing turkeys and β-glucanase-xylanase activity-containing feed enzyme supplementation on digestibility and feeding value of pelleted maize, wheat, barley and oats were investigated in growing turkeys using excreta collection and ileal sampling by slaughter. Excreta were collected and turkeys were slaughtered at 4, 8 and 12 weeks of age. Viscosity of jejuno-duodenal digesta, caecal volatile fatty acid concentration, ileal crude protein digestibility, total tract fat digestibility and AMEN were assayed using titanium dioxide as an indigestible marker. The highest viscosities were observed in barley and wheat. Viscosity of wheat, barley and oats digesta decreased while caecal volatile fatty acid concentration, fat digestibility and AMEN increased with age. Ileal crude protein digestibility was highest in wheat and lowest in barley. Ileal crude protein digestibility significantly declined with age in most feeding treatments. Enzyme reduced digesta viscosity most efficiently in wheat and barley and improved ileal crude protein digestibility, total tract fat digestibility and AMEN in wheat, barley and oats, but interactions occurred, the effect of enzyme on viscosity being the most remarkable for wheat and barley and for the young birds.  相似文献

The influence of enzyme supplementation on performance and intestinal viscosity of male broiler chickens fed with diets containing high amount of wheat was examined in three experiments. In the first experiment, addition with an enzyme preparation including different cell wall degrading enzymes to diets containing 63 g kg−1 and 72 g kg−1 of wheat improved (P<0.05) feed conversion efficiency in the 72 g kg−1 wheat diet. In addition, intestinal viscosity of chickens fed with the 72 g kg−1 wheat diet was reduced (P<0.05). Weight gain and feed intake were not influenced by enzyme addition. In Experiments 2 and 3, the inclusion level of wheat in the diets was more than 80 g kg−1 and four different enzyme preparations were used (two xylanase preparations, two mixed preparations). Overall, a significant effect on performance and intestinal viscosity of chickens was obtained as a result of enzyme supplementation in both experiments. In the first 21 days, improvements (P<0.05) in weight gain and feed conversion efficiency were found to be on average 5% and 6% in Experiment 2 and 7% and 8% in Experiment 3, respectively. When weight gain and feed conversion efficiency were examined on a weekly basis it was shown that the significant response of enzyme addition was confined to the first 4 weeks. However, the effect of enzyme supplementation was still significant in the whole period from 21–42 days. Feed intake was not influenced by enzyme addition. The viscosity of intestinal content in both the jejunum and ileum was in general reduced (P<0.05) with enzyme supplementation, the xylanase preparations proving to be the most efficient. It was concluded that enzyme supplementation of wheat-based diets resulted in improved performance of broiler chickens, which was related to a concomitant reduction in intestinal viscosity. However, the response of enzyme supplementation was most pronounced in diets with a wheat content higher than 80 g kg−1.  相似文献

Several studies were carried out to investigate the influence of dietary cereals differing in soluble non starch polysaccharides (NSP) content and a xylanase preparation on selected bacterial parameters in the small intestine of broiler chicken. Compared to a maize diet colony forming units (CFU) of mucosa associated bacteria were higher in a wheat/rye diet, most notably for enterobacteria and enterococci. Xylanase supplementation to the wheat/rye diet generally led to lower CFU, especially in the first week of life. However, xylanase supplementation also displayed higher in vitro growth potentials for enterobacteria and enterococci. Bacterial growth of luminal samples in minimal media supplemented with selected NSP showed that the wheat/rye diet enhanced bacterial capacities to utilize NSP only in ileal samples. The xylanase application generally shifted respective maximum growth to the proximal part of the small intestine. The presence of soluble NSP from wheat or rye in the diet per se did not enhance bacterial NSP hydrolyzing enzyme activities in the small intestine, but xylanase supplementation resulted in higher 1,3-1,4- g - glucanase activity. Compared to a maize diet the activity of bacterial bile salt hydrolases in samples of the small intestine was not increased due to inclusion of wheat/rye or triticale to the diet. However, xylanase supplementation led to a reduction with a corresponding increase of lipase activity. It was concluded that dietary cereals producing high intestinal viscosities lead to increased overall bacterial activity in the small intestine. The supplementation of a xylanase to cereal based diets producing high intestinal viscosity, changes composition and metabolic potential of bacterial populations and may specifically influence fat absorption in young animals.  相似文献

Recombinant xylanase preparations from Nonomuraea flexuosa (Nf Xyn, GH11) and Thermoascus aurantiacus (Ta Xyn, GH10) were evaluated for their abilities to hydrolyze hydrothermally pretreated wheat straw. The GH family 10 enzyme Ta Xyn was clearly more efficient in solubilizing xylan from pretreated wheat straw. Improvement of the hydrolysis of hydrothermally pretreated wheat straw by addition of the thermostable xylanase preparations to thermostable cellulases was evaluated. Clear synergistic enhancement of hydrolysis of cellulose was observed when cellulases were supplemented even with a low amount of pure xylanases. Xylobiose was the main hydrolysis product from xylan. It was found that the hydrolysis of cellulose increased nearly linearly with xylan removal during the enzymatic hydrolysis. The results also showed that the xylanase preparation from T. aurantiacus, belonging to GH family 10 always showed better hydrolytic capacity of solubilizing xylan and acting synergistically with thermostable cellulases in the hydrolysis of hydrothermally pretreated wheat straw.  相似文献

Thermophilic Humicola sp. secreted thermostable xylanases when grown on wheat bran medium at 50°C. DEAE-Sephadex A-50 column chromatography of the crude xylanase separated three fractions of xylanase (I, II and III), xylanase I being homogeneous in polyacrylamide gel electrophoresis after CM-Sephadex column chromatography. The respective xylanases, including the crude xylanase, increased pulp brightness but xylanases II and III decreased the viscosity of the pulp due to CMCase activity. The crude xylanase contained lower CMCase activity than xylanases II and III.  相似文献

The experiment consisted of a 2 × 2 × 2 factorial design testing the two D+ and D- chicken lines selected for divergent digestion efficiency (fifth selection generation), xylanase (with or without) and ampicillin and collistin (with or without) supplementations. From 8 to 22 days, 144 chickens (18 birds per treatment) were fed a diet containing 55% wheat from a high-viscosity cultivar (Rialto). Effects of treatments were evaluated on individual growth performance (8 to 19 days), digestibilities of lipids and dry matter, dietary energy value (apparent metabolisable energy corrected to zero-nitrogen retention (AMEn)), digestive organ and breast sizes, and intestinal bile acids at 3 weeks of age. Individual variabilities were much lower in D+ than in D- birds for feed : gain ratios, digestibilities and AMEn values. In all cases, feed : gain ratios were lower in the D+ than in the D- line (P < 0.001), and D+ birds showed 22% to 86% higher values than in D- birds (P < 0.001) for digestibilities and AMEn. In D- birds, antibiotics but not xylanase supplementation had significant effects on lipid digestibility (P < 0.01) and AMEn (P < 0.05), whereas both supplements improved these parameters in D+ birds (P < 0.001 for both additives on lipids digestibility, P < 0.05 for xylanase and P < 0.01 for antibiotics on AMEn). Relative weights of gizzard and proventriculus, and gizzard : intestine weight ratio were higher in D+ than in D- birds, while relative weight of intestine was increased in D- birds compared with D+ birds. Antibiotics reduced intestine relative weight in D+ (P < 0.001) and D- (P < 0.01) lines. AMEn variations were efficiently predicted by the gizzard : intestine weight ratio. In conclusion, antibiotics were very efficient for improving growth performance, AMEn and digestibility values in both chicken lines. Xylanase was less efficient than antibiotics. Because of their low individual variabilities, D+ birds were much more efficient than D- ones for the detection of significant effects induced by xylanase supplementation. Differences between lines in feed : gain ratio, digestibilities and AMEn were reduced when xylanase and antibiotics were added together. Effects of xylanase supplementation and animal genetics on lipid digestibility could not be entirely explained in terms of intestinal bile acids. Other factors should be involved, especially for the lipid digestibility difference induced by animal genetics. The gizzard : intestine weight ratio was an efficient parameter for predicting AMEn variations due to animal genetics and additives.  相似文献

Two experiments were conducted to investigate the effect of combined supplementation of α-galactosidase and xylanase on nutrient digestibility and growth performance in growing pigs. Experiment 1 had a 2 × 2 Latin square design, where eight barrows (45.0 ± 0.52 kg body weight [BW]) were fitted with a simple T-cannula in the distal ileum and received a basal diet without or with supplementation of α-galactosidase (12 U/kg diet) and xylanase (15 AXC/kg diet) within two periods of 10 d. The apparent ileal digestibility (AID) and apparent total tract digestibility of nutrients, pH, viscosity of digesta and digestive enzyme activities were assessed. In Experiment 2, a total of 432 growing pigs (initial BW 44.7 ± 0.66 kg) were allocated to four treatments. Diets were based on corn and soybean meal and had a normal or reduced nutrient level (reduced by 0.42 kJ digestible energy [DE] per kg and 0.8% crude protein). Both diets were offered without or with supplementation of α-galactosidase and xylanase. The growth performance was assessed within a 43-d feeding period, where at the end, biochemical serum indices were estimated. In Experiment 1, the enzyme-supplemented diet had a greater contents of DE and DE/gross energy ratio (p < 0.05), and a higher AID of Arg, raffinose, stachyose and arabinoxylan (p < 0.05). In Experiment 2, the low nutrient level caused lower daily gain (p < 0.05), which was partially compensated by enzyme addition. Enzyme addition also increased the serum concentration of Lys (p < 0.05). Moreover, it appears that the tested enzyme supplementation could increase dietary DE, serum total amino acid concentrations and decrease serum urea nitrogen.  相似文献

Experiments were carried out to examine the effects of a Fusarium contaminated wheat (10mg deoxynivalenol and 0.76mg zearalenone, ZON, perkg dry matter) and of a detoxifying agent (Mycofix ®Plus, Biomin GmbH, Herzogenburg, Austria) on the growing performance of bulls, carry-over of ZON and its metabolites into body fluids and tissues, and on nutrient digestibility in wethers. The experiments were designed according to a complete two by two factorial approach which meant that both the uncontaminated control wheat and the Fusarium toxin contaminated wheat were tested both in the absence and presence of Mycofix ®Plus. The growing experiment with bulls (n = 14 per treatment) covered the live weight range between 244kg and 460kg. The respective wheat batches were included in the concentrate portion at 65%. Concentrates were fed according to plan whereas maize silage was offered for ad libitum consumption. Daily dry matter intake and live weight gain [kg per animal and day] were 7.40, 7.52, 7.51 and 7.49 and 1.367, 1.296, 1.380 and 1.307 for bulls fed the unsupplemented control wheat, the supplemented control wheat, the unsupplemented and Fusarium toxin contaminated wheat and the supplemented Fusarium toxin contaminated wheat, respectively. ZON and its metabolites were not detected in edible tissues. The most striking effects of feeding the Fusarium toxin contaminated wheat on carcass characteristics were a reduced dressing percentage, an increased weight of the emptied gastro-intestinal tract and a reduced weight of the testicles. No effect of the detoxifying agent was seen for these parameters whereas heart weight increased independently of Fusarium toxin contamination of the concentrates. Nutrient digestibility of the two wheat batches, unsupplemented or supplemented with Mycofix ®Plus was evaluated according to the difference method using wethers. Presence of Fusarium toxins in wheat did not influence its feeding value. The effects of the addition of the detoxifying agent were mycotoxin unspecific and resulted in an increase in apparent digestibility of crude protein and a decrease in crude fiber digestibility. It is concluded that feeding of Fusarium toxin contaminated wheat did not adversely affect performance of growing bulls (approximately 2.2mg DON and 0.1mg ZON perkg complete ration at a reference dry matter content of 88%) or nutrient digestibility in wethers. The effects of the detoxifying agent Mycofix ®Plus on growing performance and on nutrient digestibility were rather Fusarium toxin unspecific. The slightly negative effects on growing performance needs to be examined further.  相似文献

Non-starch polysaccharides (NSP) present in wheat and barley can act as anti-nutrients leading to an increase in digesta viscosity and a reduction in nutrient digestibility. Xylanase, an NSP-degrading enzyme, has been shown to increase nutrient digestibility in pigs. The objectives of this study were: (1) to identify the optimum inclusion level of xylanase in grower pig diets by measuring the effect of increasing enzyme levels on growth performance, the concentration of volatile fatty acids (VFA) and peptide YY concentration in portal and peripheral blood of grower pigs and (2) to increase our understanding of the interrelationships between xylanase inclusion, VFA production and peptide YY secretion. A total of 512 grower pigs ((Large White×Landrace)×MAXGRO) were allocated to pens creating 32 replicates of four pigs per pen per treatment. Pigs were allocated to trial weighing 14.2±0.31 kg and remained on trial until ~41.5±3.31 kg. The experiment was a dose response design with four inclusion levels (0, 8000, 16 000 or 32 000 BXU/kg) of xylanase (Econase XT). Diets were cereal-based wheat, barley mix formulated to meet or exceed the nutrient requirements of grower pigs. Body weight and feed intake were recorded to calculate growth performance. Pen faecal samples were collected to estimate DM, organic matter (OM) and crude fibre (CF) apparent total-tract digestibility. At the end of the trial 16 pigs per treatment were euthanised by schedule 1 procedures. Peripheral and portal blood samples were collected for peptide YY and VFA analysis. The addition of xylanase to the diet had no effect on growth performance, DM, OM or CF total-tract digestibility; however, xylanase tended to have a quadratic effect on ileum pH with higher pH values recorded for pigs fed a diet supplemented with 8000 and 16 000 BXU/kg xylanase (P<0.1). Xylanase had no effect on peptide YY levels or VFA concentration. Total VFA concentration was higher in portal compared with peripheral blood (P<0.05). In conclusion, the addition of xylanase had no effect on grower pig performance, nutrient digestibility, VFA concentration or peptide YY concentration when fed up to 32 000 BXU/kg over a 35-day period. Pig performance was good for all treatments throughout the trial suggesting that diet quality was sufficient thus there were no beneficial effects of adding xylanase.  相似文献

The ability of a crude enzyme preparation ofAureobasidium pullulans containing xylanase [61 units (U)/ml] and xylosidase (3 U/ml) activity to remove pentosans from unbleached sulphite pulps was investigated. Greater amounts of pentosans and reducing sugars were released from the pulp when the enzyme dosages and incubation times were increased. A combination of enzyme hydrolysis and alkali extraction resulted in a greater removal of pentosans than using the enzyme preparation alone. By treatment with a xylanase loading of 450 U/g pulp for 24 h followed by alkaline extraction, 35% of the pentosans were removed. The kappa number decreased up to 30% whereas viscosity was only slightly affected by these treatments. Enzymatic hydrolysis released mainly xylose whereas xylobiose was the main product liberated by alkaline extraction. Scanning electron micrographs indicated improved fibrillation and flexibility of the fibre structure by enzyme treatment.  相似文献

The aim of the experiment on 180 weaned piglets (8.9 kg body weight) was to investigate the influence of high fibre diets formulated with different fibrous ingredients on performance, nutrient digestibility, diarrhoea incidence and numbers of faecal microbiota. The dietary treatments included a Control diet and five high fibre diets formulated with different fibre sources including wheat bran, soybean hulls, naked oat hulls, palm kernel expeller and bamboo fibre. The high fibre diets averaged 14.6% neutral detergent fibre with different non-starch polysaccharides (NSP) components and were fed ad libitum for 28 d. Faecal samples were collected during the last 3 d of the experiment and the apparent total tract digestibility of nutrients and fibre components were determined. Pigs fed the Control and wheat bran diets had a higher (≤ 0.05) average daily gain (ADG) than pigs fed the palm kernel expeller and bamboo meal diets. The reduced ADG for pigs appeared to be related to reductions in the digestibility of gross energy and dry matter, respectively. The feed-to-gain ratio was significantly higher (≤ 0.05) for pigs fed the fibre diets. The digestibility of NSP components was different among the treatments. The diarrhoea incidence was not affected by treatments. The abundance of faecal bifidobacteria was significantly higher (≤ 0.05) for pigs fed the wheat bran diet than for pigs fed the bamboo meal diet. It was concluded that the diets formulated with different fibre sources when fed to weaned piglets have different effects on pig performance, nutrient digestibility and numbers of faecal microbiota. The wheat bran diet rich in arabinoxylans enabled a better performance than the other tested diets with fibre addition.  相似文献

Summary The production of xylanase from Bacillus coagulans has been studied with respect to the environmental parameters, the carbon source and the concentration of carbon source at the shake flask level. Among the various carbon sources used, wheat straw powder favoured higher enzyme production. Xylan isolated from wheat straw gave higher enzyme production as compared to the birchwood xylan. Maximum enzyme activity of 165 IU/ml was obtained with 2% wheat straw xylan in a shake flask study. Improvement of xylanase production was achieved by increasing the wheat straw powder concentration up to 3%. Enzyme has optimum activity at a temperature of 55 °C and pH of 7. The concentrated crude enzyme was found to reduce the kappa number of enzyme-treated eucalyptus pulp by␣5.45% with a marginal increase in the CED viscosity of the enzyme treated pulp as compared to the non-enzymatically treated pulp.  相似文献

Cellulase-free xylan-degrading enzyme preparations from Acrophialophora nainiana, Humicola grisea var. thermoidea and two Trichoderma harzianum strains were used as bleaching agents for Eucalyptus kraft pulp, prior to a chlorine dioxide and alkaline bleaching sequence. In comparison to the control sequence (performed without xylanase pretreatment), the sequence incorporating enzyme treatment was more effective. Removal of residual lignin was indicated by a reduction in kappa number. Overall, enzyme preparations from T. harzianum were marginally more effective in reducing pulp viscosity and chlorine chemical consumption and improving the brightness of the kraft pulp. However, the highest reduction in pulp viscosity was mediated by the xylanase preparation from A. nainiana. Xylanase pretreatment compares very favorably with that of chemical pulping. Pigs were fed diets containing 40% wheat bran incubated with a water.‐acetic acid mixture (control, C) and a cellulase (Cel‐i) or xylanase (Xyl‐i) preparation or with addition of the cellulase (Cel‐a) or xylanase (Xyl‐a) preparation immediately before feeding. Stomach and ileal samples were analysed for pH, osmolality, soluble saccharides, volatile fatty acids (VFA) and lactic acid. Incubation of wheat bran resulted in a small reduction of NDF and an increase in the amount of soluble starch, ß‐glucans and saccharides (glucose, xylose and arabinose), especially after incubation with the cellulase preparation. Two hours after feeding, significantly higher arabinose and xylose concentrations were present in the stomach for diets Cel‐i, Cel‐a and Xyl‐i. In the ileum xylose and arabinose concentrations were higher 2 to 4 and 6 to 8 hours after feeding the enzyme‐treated diets. In stomach and ileum there were no differences between the diets in pH, osmolality, VFA and lactic acid concentrations, but ileal VFA concentration from 4 h after feeding tended to be higher for diets Cel‐i and Xyl‐i. It can be concluded that the amount of soluble saccharides in stomach and small intestine and the ileal VFA concentration may be increased by cell wall degrading enzyme preparations.  相似文献

Abstract

The experiment was conducted to study the effects of fermentation of barley, using two different strains of lactic acid bacteria, a Lactobacillus plantarum/pentosus strain isolated from spontaneously fermented rye sourdough (AD2) and a starch-degrading Lactobacillus plantarum (AM4), on contents of mixed-linked (1 → 3) (1 → 4)-β-glucans, α-amylase inhibitor activity, inositol phosphates, and apparent digestibility of macronutrients in mink. Effects of fermentation were compared with effects of gamma irradiation (γ-irradiation: 60Co γ-rays at 25 kGy). The diets were fed to mink with and without a supplementary enzyme preparation. Both lactic acid fermentation and γ-irradiation followed by soaking and incubation, reduced concentrations of soluble β-glucans, phytate and α-amylase inhibitor activity. Dietary enzyme supplementation increased significantly digestibility of crude protein, fat, starch and crude carbohydrate (CHO). Fermentation of the barley increased digestibility of starch and CHO. Fermentation with lactic acid bacteria AD2 resulted in higher starch and CHO digestibility than strain AM4, and had greater effect than γ-irradiation, soaking and incubation. The highest digestibility of starch and CHO was obtained after AD2 fermentation followed by enzyme supplementation. An experiment examined the effects of two field bean cultivar samples with different tannin contents, the effect of heat treatment (micronising) and the effect of dietary supplementation of a proprietary enzyme preparation containing tannase, pectinase, and xylanase activities on metabolisable energy (ME), total tract dry matter digestibility (DMD) and ether extract digestibility (EED), nitrogen retention (NR), tannin degradability, gastrointestinal tract (GIT) development, and endogenous mucin losses excretion in broiler chickens. The Control diet contained per kg 221 g crude protein and 12.83 MJ ME. Four additional diets contained 300 g/kg of each of the two untreated or micronised experimental field bean cultivar samples. Each diet was then split into two batches and one of them was supplemented with 3400 units tannase per kg diet resulting in 10 diets in total. Each diet was fed to seven pens with two randomly selected male broilers each. Birds fed the high tannin bean sample had a lower weight gain (p < 0.001), and a lower determined apparent ME (p < 0.05), and DMD (p < 0.001) but a higher tannin degradability (p < 0.001). Compared to the Control diet, feeding field beans increased (p < 0.001) the weights of the proventriculus and gizzard of the birds, and also increased endogenous mucin losses (p < 0.05). Supplementing diets with the tannase-containing enzyme preparation improved dietary ME (p < 0.001), DMD (p < 0.001), NR (p < 0.001) and DEE (p < 0.05), but did not change tannin degradability. Heat treatment of the beans reduced the degradability of condensed tannins and increased endogenous mucin losses (p < 0.05). The differences in the feeding value of the different field bean samples were not improved by heat treatment, but enzyme supplementation improved the feeding value of all diets regardless of the bean samples or heat treatment. Further research is warranted to study the effectiveness of tannase supplementation in poultry diet formulations by dose response trials with purified tannase preparations.  相似文献

The xylanase inhibitor TAXI‐III has been proven to delay Fusarium head blight (FHB) symptoms caused by Fusarium graminearum in transgenic durum wheat plants. To elucidate the molecular mechanism underlying the capacity of the TAXI‐III transgenic plants to limit FHB symptoms, we treated wheat tissues with the xylanase FGSG_03624, hitherto shown to induce cell death and hydrogen peroxide accumulation. Experiments performed on lemmas of flowering wheat spikes and wheat cell suspension cultures demonstrated that pre‐incubation of xylanase FGSG_03624 with TAXI‐III significantly decreased cell death. Most interestingly, a reduced cell death relative to control non‐transgenic plants was also obtained by treating, with the same xylanase, lemmas of TAXI‐III transgenic plants. Molecular modelling studies predicted an interaction between the TAXI‐III residue H395 and residues E122 and E214 belonging to the active site of xylanase FGSG_03624. These results provide, for the first time, clear indications in vitro and in planta that a xylanase inhibitor can prevent the necrotic activity of a xylanase, and suggest that the reduced FHB symptoms on transgenic TAXI‐III plants may be a result not only of the direct inhibition of xylanase activity secreted by the pathogen, but also of the capacity of TAXI‐III to avoid host cell death.  相似文献
