Nitrogen (N) transfer from one species to another is important for the N cycling in low-input grassland. In the present work, estimates obtained by an indirect 15N dilution technique were compared with estimates obtained by a direct 15N leaf feeding technique over two complete growing seasons in red clover-ryegrass and white clover-ryegrass mixtures under field conditions. The direct technique confirmed that N transfer between clovers and ryegrass is a bi-directional process. The transfer of N from both clovers to ryegrass occurred within 25 days upon the first labelling event. A very high N transfer occurred from white clover to the associated ryegrass, 4.5 and 7.5 g m−2 in the 1st and 2nd production year, respectively. The corresponding values for transfer from red clover to the associated ryegrass were 1.7 and 3.6 g m−2. Quantified relatively to the total above-ground N content of white clover- ryegrass and red clover-ryegrass mixtures, the N transfer exceeded 50% and 10%, respectively, in three out of seven harvests. The N transfer from 15N labelled grass to associated clovers constituted a relatively constant proportion of approx. 8% of the above-ground N content of the mixtures. Estimates based on the soil 15N dilution technique generally underestimated the net N transfer by more than 50% compared to the direct 15N labelling technique. Furthermore, the indirect 15N dilution technique estimated only marginal differences between red and white clover in the quantities of N transferred, whereas the direct 15N labelling technique showed the N transfer from white clover to the associated ryegrass to be significantly higher than that involving red clover. It is concluded that N transfer is a much more dynamic and quantitatively important process in grassland than previously recognised. Virtually complete sequence specific 1H and 15N resonance assignments are presented for acid denatured reduced E. coli glutaredoxin 3. The sequential resonance assignments of the backbone rely on the combined use of 3D F1-decoupled ROESY-15N-HSQC and 3D 15N-HSQC-(TOCSY-NOESY)-15N-HSQC using a single uniformly 15N labelled protein sample. The sidechain resonances were assigned from a 3D TOCSY-15N-HSQC and a homonouclear TOCSY spectrum. The presented assignment strategy works in the absence of chemical exchange peaks with signals from the native conformation and without 13C/15N double labelling. Chemical shifts, 3J(H, NH) coupling constants and NOEs indicate extensive conformational averaging of both backbone and side chains in agreement with a random coil conformation. A plant mixture of white clover (Trifolium repens L.), red clover (Trifolium pratense L.), and ryegrass (Lolium perenne L.) was established in the spring of 1991 under a cover-crop of barley. Treatments were two levels of nitrogen (400 and 20 kg N ha-1) and two cutting intensities (3 and 6 cuts per season). Fixation of atmospheric derived nitrogen was estimated by two 15N dilution methods, one based on application of 15N to the soil, the other utilising small differences in natural abundance of 15N.Both methods showed that application of 400 kg N ha-1 significantly reduced dinitrogen fixation, while cutting frequency had no effect. Atmospheric derived nitrogen constituted between 50 and 64% of harvested clover nitrogen in the high-N treatment, while between 73% and 96% of the harvested clover nitrogen was derived from the atmosphere in the low-N treatment. The amounts of fixed dinitrogen varied between 31–72 kg N ha-1 and 118–161 kg N ha-1 in the high-N and low-N treatment, respectively. The highest values for biological dinitrogen fixation were estimated by the enriched 15N dilution method.Estimates of transfer of atmospheric derived nitrogen from clover to grass obtained by the natural 15N abundance method were consistently higher than those obtained by the enriched 15N dilution method. Neither mineral nitrogen application nor defoliation frequency affected transfer of atmospheric derived nitrogen from clover to grass.Isotopic fractionation of 14N and 15N (B value) was estimated by comparing results for nitrogen fixation obtained by the enriched 15N dilution and the natural 15N abundance method, respectively. B was on average +1.20, which was in agreement with a B value determined by growing white clover in a nitrogen free media.  相似文献

Chopped wheat straw was homogeneously mixed with urine of horses (5.75 gN per 1, 16.88 atom‐% 15N‐excess) and airtightly stored in plastic containers for 6 months. Three rumen fistulated sheep and goats each were fed with untreated or urine treated straw. Concentrate was added to straw. Untreated and urine treated straw were given in nylon bags and incubated in the rumen of sheep and goats for 1, 3, 6, 12, 24, 48 and 72 hours. A three compartment exponential function was used to fit the measurements of 15N‐ex‐cess and 15N‐amount of bag content. The curves and the calculated partial Y‐values of the three compartments show the inflow and outflow of 15N into or from the bags and allow conclusions about the binding of urine N. Most N of urine was not compactly bound by straw during storage. Primarily micro‐bial N was attached to the straw in the rumen. About 6% of urine N were bound more compact to the straw. Similar curves were calculated for 15N‐excess and 15N‐amount of nylon bags. The curves allow conclusions about tracer flows without quantitative knowledge. The use of 13C isotope tracer techniques in terrestrial ecology has been restricted by the technical requirements and high costs associated with the production of 13C enriched plant material by 13CO2 release in labelling chambers. We describe a novel, simple and relatively inexpensive method for the small-scale production of 13C and 15N labelled plant material. The method is based on foliar feeding of plants with a urea solution (97 atom% 13C, 2 atom% 15N) by daily misting. Maize was grown in a greenhouse in a compost–soil mixture and enclosed in clear polythene bags between urea applications. Final enrichment in 27 d old maize shoots was 211 13C (1.34 atom% 13C) and 434 15N (0.52 atom% 15N). Enrichments of hot-water extractable fractions (289 13C, 469 15N) were only slightly higher than those observed in plant bulk material, which suggests that daily urea applications ensured fairly uniform labelling of different biochemical fractions and plant tissues. Recovery of applied excess 13C and 15N in plant shoots was 22% and 42%, respectively. Roots were less enriched (21 13C and 277 15N), but no attempts were made to recover roots quantitatively.  相似文献

Two animal slurries either labelled with 15N in the urine or in the faeces fraction, were produced by feeding a sheep with unlabelled and 15N-labelled hay and collecting faeces and urine separately. The slurries were applied (12 g total N -2) to a coarse sand and a sandy loam soil confined in lysimeters and growing spring barley (Hordeum vulgare L). Reference lysimeters without slurry were supplied with15 NH4 15NO3 corresponding to the inorganic N applied with the slurries (6 g N m-2). In the second year, all lysimeters received unlabelled mineral fertilizer (6 g N m-2) and grew spring barley. N harvested in the two crops (grain + straw) and the loss of nitrate by leaching were determined. 15N in the urine fraction was less available for crop uptake than mineral fertilizer 15N. The first barley crop on the sandy loam removed 49% of the 15N applied in mineral fertilizer and 36% of that applied with urine. The availability of fertilizer 15N (36%) and urine15 N (32%) differed less on the coarse sand. Of the15 N added with the faeces fraction, 12–14% was taken up by the barley crop on the two soils. N mineralized from faeces compensated for the reduced availability of urine N providing a similar or higher crop N uptake in manured lysimeters compared with mineral fertilized ones.About half of the total N uptake in the first crop originated from the N applied either as slurry or mineral fertilizer. The remaining N was derived from the soil N pool. Substantially smaller but similar proportions of15 N from faeces, urine and fertilizer were found in the second crop. The similar recoveries indicated a slow mineralization rate of the residual faeces N since more faeces was left in the soil after the first crop.More N was lost by leaching from manured lysimeters but as a percentage of N applied, losses were similar to those from mineral fertilizer. During the first and second winter, 3–5% and 1–3%, respectively, of the 15N in slurry and mineral fertilizer was leached as nitrate. Thus slurry N applied in spring just before sowing did not appear to be more prone to loss by nitrate leaching than N given in mineral fertilizer. Slurry N accounted for a higher proportion of the N leached, however, because more N was added in this treatment.  相似文献

Nitrogen (N) is often the most limiting nutrient in organic cropping systems. N2 fixing crops present an important option to improve N supply and to maintain soil fertility. In a field experiment, we investigated whether the lower N fertilization level and higher soil microbial activity in organic than conventional systems affected symbiotic N2 fixation by soybean (Glycine max, var. Maple Arrow) growing in 2004 in plots that were since 1978 under the following systems: bio-dynamic (DYN); bio-organic (ORG); conventional with organic and mineral fertilizers (CON); CON with exclusively mineral fertilizers (MIN); non-fertilized control (NON). We estimated the percentage of legume N derived from the atmosphere (%Ndfa) by the natural abundance (NA) method. For ORG and MIN we additionally applied the enriched 15N isotope dilution method (ID) based on residual mineral and organic 15N labeled fertilizers that were applied in 2003 in microplots installed in ORG and MIN plots. These different enrichment treatments resulted in equal %Ndfa values. The %Ndfa obtained by NA for ORG and MIN was confirmed by the ID method, with similar variation. However, as plant growth was restricted by the microplot frames the NA technique provided more accurate estimates of the quantities of symbiotically fixed N2 (Nfix). At maturity of soybean the %Ndfa ranged from 24 to 54%. It decreased in the order ORG > CON > DYN > NON > MIN, with significantly lowest value for MIN. Corresponding Nfix in above ground plant material ranged from 15 to 26 g N m-2, with a decreasing trend in the order DYN = ORG > CON > MIN > NON. For all treatments, the N withdrawal by harvested grains was greater than Nfix. This shows that at the low to medium %Ndfa, soybeans did not improve the N supply to any system but removed significant amounts of soil N. High-soil N mineralization and/or low-soil P availability may have limited symbiotic N2 fixation.  相似文献

Plant species collected from tundra ecosystems located along a north-south transect from central Alaska to the north coast of Alaska showed large and consistent differences in 15N natural abundances. Foliar 15N values varied by about 10% among species within each of two moist tussock tundra sites. Differences in 15N contents among species or plant groups were consistent across moist tussock tundra at several other sites and across five other tundra types at a single site. Ericaceous species had the lowest 15N values, ranging between about –8 to –6. Foliar 15N contents increased progressively in birch, willows and sedges to maximum 15N values of about +2 in sedges. Soil 15N contents in tundra ecosystems at our two most intensively studied sites increased with depth and 15N values were usually higher for soils than for plants. Isotopic fractionations during soil N transformations and possibly during plant N uptake could lead to observed differences in 15N contents among plant species and between plants and soils. Patterns of variation in 15N content among species indicate that tundra plants acquire nitrogen in extremely nutrient-poor environments by competitive partitioning of the overall N pool. Differences in plant N sources, rooting depth, mycorrhizal associations, forms of N taken up, and other factors controlling plant N uptake are possible causes of variations in 15N values of tundra plant species.  相似文献

A triple-resonance pulse scheme is described which records15N, NH correlations of residues that immediately follow amethyl-containing amino acid. The experiment makes use of a15N, 13C and fractionally deuterated proteinsample and selects for CH2D methyl types. The experiment isthus useful in the early stages of the sequential assignment process as wellas for the confirmation of backbone 15N, NH chemical shiftassignments at later stages of data analysis. A simple modification of thesequence also allows the measurement of methyl side-chain dynamics. This isparticularly useful for studying side-chain dynamic properties in partiallyunfolded and unfolded proteins where the resolution of aliphatic carbon andproton chemical shifts is limited compared to that of amide nitrogens.  相似文献

Summary Plants from agricultural and natural upland ecosystem were investigated for15N content to evaluate the role of symbiotic N2-fixation in the nitrogen nutrition of soybean. Increased yields and lower δ15N values of nodulating soybeansvs, non-nodulating isolines gave semi-quantitative estimates of N2 fixation. A fairly large discrepancy was found between estimations by δ15N and by N yield at 0 kg N/ha of fertilizer. More precise estimates were made by following changes in plant δ15N when fertilizer δ15N was varied near15N natural abundance level. Clearcut linear relationships between δ15N values of whole plants and of fertilizer were obtained at 30 kg N/ha of fertilizer for three kinds of soils. In experimental field plots, nodulating soybeans obtained 13±1% of their nitrogen from fertilizer, 66±8% from N2 fixation and 21±10% from soil nitrogen in Andosol brown soil; 30%, 16% and 54% in Andosol black soil; 7%, 77% and 16% in Alluvial soil, respectively. These values for N2 fixation coincided with each corresponding estimation by N yield method. Other results include: 1)15N content in upland soils and plants was variable, and may reflect differences in the mode of mineralization of soil organics, and 2) nitrogen isotopic discrimination during fertilizer uptake (δ15N of plant minus fertilizer) ranged from −2.2 to +4.9‰ at 0–30 kg N/ha of fertilizer, depending on soil type and plant species. The proposed method can accurately and relatively simply establish the importance of symbiotic nitrogen fixation for soybeans growing in agricultural settings.  相似文献

Summary Six pasture grasses,Paspalum notatum cv batatais,P. notatum cv pensacola,Brachiaria radicans, B. ruziziensis, B. decumbens andB. humidicola, were grown in concrete cylinders (60 cm diameter) in the field for 31 months. The soil was amended with either a single addition of15N labelled organic matter or frequent small (2 kg N. ha–1) additions of15N enriched (NH4)2SO4. In the labelled fertilizer treatment soil analysis revealed that there was a very drastic change in15N enrichment in plant-available nitrogen (NO 3 +NH 4 + ) with depth. The different grass cultivars recovered different quantities of applied labelled N, and evidence was obtained to suggest that the roots exploited the soil to different depths thus obtaining different15N enrichments in soil derived N. This invalidated the application of the isotope dilution technique to estimate the contribution of nitrogen fixation to the grass cultivars in this treatment. In the labelled organic matter treatment the15N label in the plant-available N declined at a decreasing rate during the experiment until in the last 12 months the decrease was only from 0.274 to 0.222 atom % excess. There was little change in15N enrichment of available N with depth, hence it was concluded that although the grasses recovered different quantities of labelled N, they all obtained virtually the same15N enrichment in soil derived N. Data from the final harvests of this treatment indicated thatB. humidicola andB. decumbens obtained 30 and 40% respectively of their nitrogen from N2 fixation amounting to an input of 30 and 45 kg N.ha–1 year–1 respectively.  相似文献

A laboratory scale working model that could detect the 15N enrichment in cyanobacterial biomass and extracellular ammonia, using 15N gas under in vitro conditions was designed and fabricated. Using the model, 15N enrichment of 0.48% atom excess was detected in the cyanobacterial biomass on the 30 d after inoculation. The 15N enrichment increased linearly in the extracellular ammoniacal fraction from the 20 d onward. The model would prove to be a useful tool to quantify the extent of 15N enrichment under in vitro conditions using 15N gas.  相似文献

The fate of15N-ammonium sulphate fertilizer that was applied to four lysimeters in the 1990/91 summer was studied over three consecutive growing seasons during which either maize or wheat was grown. Aboveground portions of15N-labelled maize plants from the first harvest were applied to four other lysimeters at 5 t ha–1. Two lysimeters in each of the sets of four were assigned a low and a high moisture treatment using irrigation. In both moisture treatments, plant recovery of fertilizer-15N in the first season was 27% and a further 2% was recovered by plants during the next two seasons. During the second and third seasons, total recovery of15N by aboveground plant portions from lysimeters that received15N-labelled maize material was equivalent to 2.5% of applied fertilizer-15N. This corresponded to ca. 18% recovery of the15N added in maize material. Leaching of fertilizer-N over the three growing seasons did not exceed 0.3% in total. During the first season, a maximum of 0.25 kg N ha–1, equivalent to 0.25% of the applied fertilizer-N, was leached in the high moisture treatment. This represented 1.8% of the nitrate load in leachates. Less than 0.002% of the applied fertilizer-N was leached in the low moisture treatment during the first season.  相似文献

Based on the HSQC scheme, we have designed a 2D heterocorrelated experiment which combines constant time (CT) 13C and variable time (VT) 15N chemical shift labelling. Although applicable to all carbons, this mode is particularly suitable for simultaneous recording of methyl-carbon and nitrogen chemical shifts at high digital resolution. The methyl carbon magnetisation is in the transverse plane during the whole CT period (1/JCC=28.6 ms). The magnetisation originating from NH protons is initially stored in the 2HzNz state, then prior to the VT chemical shift labelling period is converted into 2HzNy coherence. The VT -15N mode eliminates the effect of 1 J N,CO and 1,2 J N,CA coupling constants without the need for band-selective carbon pulses. An optional editing procedure is incorporated which eliminates signals from CH2 groups, thus removing any potential overlap with the CH3 signals. The CT-13CH3,VT-15N HSQC building block is used to construct two 3D experiments: 3D NOESY-CH3NH and 3D 13C,15N HSQC-NOESY-CH3NH. Combined use of these experiments yields proton and heteronuclear chemical shifts for moieties experiencing NOEs with CH3 and NH protons. These NOE interactions are resolved as a consequence of the high digital resolution in the carbon and nitrogen chemical shifts of CH3 and NH groups, respectively. The techniques are illustrated using a double labelled sample of the CH domain from calponin.  相似文献

Translocation of carbon and nitrogen within a single source-sink unit, comprising a trifoliated leaf, the axillary pod and the subtending internode, and from this unit to the rest of the plant was examined in soybean (Glycine max L. cv. Akishirome) plant by feeding 13CO2 and 15NO3. The plants were grown at two levels of nitrogen in the basal medium, i.e. low-N (2 g N m–2) and high-N (35 g N m–2) and a treatment of depodding was imposed by removing all the pods from the plant, except the pod of the source sink unit, 13 days after flowering. The plants at high-N accumulated more biomass in its organs compared to low-N and pod removal increased the weight of the vegetative organs. When the terminal leaflet of the source-sink unit was fed with 13CO2, almost all of the radioactive materials were retained inside the source-sink unit and translocation to rest of the plants was insignificant under any of the treatments imposed. Out of the13C exported by the terminal leaflet, less than half went into the axillary pod, as the lateral leaflets claimed equal share and very little material was deposited in the petiole. Pod removal decreased 13C export at high-N , but not at low-N. Similar to 13C, the source-sink unit retained all the 15N fed to the terminal leaflet at high-N. At low-N, the major part of 15N partitioning occurred in favour of the rest of the plant outside the source-sink unit, but removal of the competitve sinks from the rest of the plants nullified any partitioning outside the unit. Unlike the situation in 13C, no partitioning of 15N occurred in favour of the lateral leaflets from the terminal leaflet inside the unit. It is concluded that sink demand influences partitioning of both C and N and the translocation of carbon is different from that of nitrogen within a source-sink unit. The translocation of the N is more adjustive to a demand from other sink units compared to the C.  相似文献

Abstract

The objective of this study was to evaluate dry matter (DM), nitrogen yield, N2 fixation (Ndfa) and soil N uptake (Ndfs) in the shrubby medic (Medicago arborea) and saltbush (Atriplex halimus) grown in pots either solely or in a mixture on a salt-affected soil, using 15N dilution method. The combined DM of both species was considerably higher than that of solely grown shrubs. The inclusion of saltbush in the mixed cropping system decreased Ndfs by shrubby medic and enhanced % Ndfa without affecting amounts of N2 fixed. It can be concluded that the use of mixed cropping system of shrubby medic and saltbush could be a promising bio-saline agricultural approach to utilize salt affected soils in terms of forage yield and N2 fixation.  相似文献

Labelled urea or ammonium nitrate was applied to winter wheat growing on a loamy soil in Northern France. Two applications of fertilizer were given: 50 kg N ha–1 at tillering (early March) and 110 kg N ha–1 at the beginning of stem elongation (mid-April). The kinetics of urea hydrolysis, nitrification of ammonium and the disappearance of inorganic nitrogen were followed at frequent intervals. Inorganic nitrogen soon disappeared, mainly immobilized by soil microflora and absorbed by the crop. Net immobilization of fertilizer N occured at a very similar rate for urea and ammonium nitrate. Maximum immobilization (16 kg N ha1) was found at harvest for the first dressing and at anthesis for the second dressing (23 kg N ha1). During the nitrification period, the labelled ammonium pool was immobilized two to three times faster than the labelled nitrate pool. No significant net15N remineralization was found during the growth cycle.The actual denitrification and volatilization losses were probably more important than indicated from calculations made by extrapolation of fluxes measured over short intervals. However microbial immobilization was the most important of the processes which compete with plant uptake for nitrogen.  相似文献

Nitrogen dynamics in semi-natural environments is crucial for the development and ecological stability of these systems. The present paper shows the results of the reinvestigation of a 15N-tracer experiment, which was established in the Grossglockner massif in Austria at 2300ma.s.l. in 1974/1975. We show that large quantities of nitrogen introduced by a single pulse labelling (amounting to approximately 1.7% of the nitrogen in the system) into an alpine grassland remain in the soil–plant system, with only 55% being lost during 27–28 years. In the first 10cm of the four investigated soil profiles 40% of 15N was recovered, being mainly bound in organic forms. A simple site specific model was established on the basis of the results considering a biological, residual and labile N-pool, the latter being the source for N-losses. By the model a long mean residence time close to 100 years was derived for the remaining 15N.  相似文献

Preliminary attempts to make retrospective studies of N balances and water stress in forest fertilization experiments by analyzing changes in the abundances of 15N and 13C, respectively, are discussed. Most evidence is from the Swedish Forest Optimum Nutrition Experiments, which have been running for two decades. Annual additions of N have been given either alone or in combination with other elements, notably P and K, every third year. Processes leading to loss of N, e.g. volatilization of ammonia, nitrification followed by leaching or denitrification, and denitrification alone, discriminate against the heavy isotope 15N. A correlation was found between fractional losses of added N and the change in 15N () during 19 years in current needles in a Scots pine forest, irrespective of source of N. Isotope effects were larger on urea than on ammonium nitrate plots (2 as compared to 9 15N ()) because of ammonia volatilization and higher rates of nitrification. They developed gradually over time, which opens possibilities to analyse the development of N saturation. However, the analysis may be confounded by shifts in 15N abundance of fertilizer N. In another trial, N isotope effects could be seen in both plants and soils 10 years after the last fertilization; they were smaller in soils because of a large pretreatment memory effect, but we expect them to persist there for decades.The enzyme RuBisCo discriminates strongly against the heavy isotope 13C during photosynthesis, but this effect becomes less expressed as stomata close because of water stress. The supply of N may also affect the 13C () via effects on rates of photosynthesis, and the source of N may have an influence directly via non-RubisCo carboxylations, and indirectly via effects on water use efficiency. In a trial with Norway spruce, the effect of N fertilization on the 13C () of current needles was strongly correlated with production and weakly so with foliar biomass a dry year, but not a wet year. This suggested that these variations are primarily related to induced differences in the balance between supply and demand for water. Hence, studies of {au13}C abundance can disentangle the role of water as such from its effects on mineralization of N and flow of N.  相似文献
