Unground triticale kernels contaminated with 6.63 mg deoxynivalenol (DON) per kg dry matter were stored for up to 63 days at total moisture contents of 13 and 15% in order to study the time-dependent kinetics of DON concentration in dependence on graded levels of sodium metabisulfite [0, 1, 2, 3, 4 and 5 g Na2S2O5 (SBS) per kg], and in the absence and presence of 10 g propionic acid (PA) per kg. The DON concentration decreased with increasing amounts of supplemented SBS and with increasing duration of the preservation period in a bi-exponential fashion when SBS addition was ≥3 g/kg. Lower SBS concentrations yielded inconsistent results. The maximum measured DON reductions after adding 5 g SBS/kg were 3 and 4% of the initial DON concentration after 63 days in the absence and presence of PA at moisture contents of 15%, while the corresponding recovery for the variants preserved at 13% amounted to 21 and 11%, respectively. The 12 variants preserved without PA supplementation were more frequently contaminated by moulds and yeasts (n = 5) than the corresponding variants stored together with PA (n = 1). The overall results and regressive evaluations do suggest that the highest SBS addition of 5 g/kg triticale at a moisture content of 15% preserved for 63 days would be necessary for a maximum DON reduction. Although PA did not exert a direct decontaminating effect, an additional supplementation together with SBS seemed to be advantageous with regard to the prevention of yeast and mould contamination and favouring the decontamination reaction by the acid milieu.  相似文献

Fusarium toxin-contaminated ground maize was hydrothermally treated in the presence of different combinations of chemicals in order to simultaneously reduce zearalenone (ZEA) and deoxynivalenol (DON) concentrations. Treatments were carried out in a laboratory conditioner at 80 °C and 17 % moisture. Six different treatments were performed, consisting of 3 doses of methylamine (MMA; 2.5, 5 and 10 g/kg maize) at a constant dose of 5 g sodium metabisulfite (SBS)/kg, either with or without the addition of 20 g calcium hydroxide (Ca(OH)2)/kg. The used maize was contaminated with approximately 45.99 mg DON/kg and 3.46 mg ZEA/kg. Without the addition of Ca(OH)2, DON reductions reached approximately 82 % after 1-min treatment and the toxin disappeared nearly completely after 10 min when 2.5 or 5 g MMA were applied. ZEA concentrations were only marginally affected. In the presence of Ca(OH)2, reductions in DON concentrations were lower, but were enhanced by increasing doses of MMA. ZEA concentrations were reduced by 72, 85 and 95 % within the first 5 min of the treatment at MMA dosages of 2.5, 5 and 10 g/kg maize, respectively. The application of SBS in combination with a strong alkaline during hydrothermal treatment seems to be a promising approach to simultaneously decontaminate even high amounts of DON and ZEA in ground maize and may contribute to reduce the toxin load of diets  相似文献

Abstract

A 35-day feeding experiment was performed with weaned piglets (initial live weight of 7.7±1 kg) to test the effects of a Fusarium toxin contamination of the diet (4.44 mg deoxynivalenol [DON] per kg diet) compared to an uncontaminated control diet. Both diet types were fed either unsupplemented or supplemented with a polymeric glucomannan mycotoxin adsorbent (GMA) to examine the claimed efficacy of detoxification of this feed additive through unspecific performance parameters and the specific proof of the prevention of DON absorption. Feeding of the mycotoxin-contaminated diets resulted in a decrease in feed intake and live weight gain by 28% and 14% when compared to the control group. These effects were independent of GMA addition. Based on the lack of significant interactions between diet mycotoxin-contamination and GMA supplementation for the performance data, and because of the inefficiency of GMA in decreasing DON absorption, it was concluded that the supplementation of DON-contaminated piglet diets with GMA cannot be recommended as an effective tool for alleviation of adverse DON effects.  相似文献

Nine gilts weighing 80 kg at the beginning of the trial were fed a mycotoxin contaminated diet containing 2 mg deoxynivalenol (DON) and 0.4 mg zearalenone (ZON) per kg (Diet M). Their daily weight gain until 103 kg BW was reduced in comparison to the nine control animals fed an uncontaminated diet (Diet C) (763 vs. 912 g; p = 0.02). There was no treatment effect on the age at first observed oestrus. Seven and eight gilts receiving Diet M and C, respectively, became pregnant after being mated once or being again mated three weeks later. The examination of the uteri of gilts slaughtered 35–61 days after mating showed that the exposure to DON and ZON had no effect on the number of foetuses per gilt (p = 0.54), but increased their growth rate (p = 0.003). Thus, low dietary DON and ZON levels had no negative effects on the reproductive parameters examined. The hypothesis that the bulbourethral gland weight of barrows can be used for the bioassay of low dietary ZON levels was rejected since feeding Diet M from 80–103 kg BW did not increase the weight of that accessory sex gland (p = 0.51).  相似文献

The objective of the present study was to demonstrate the efficiency of the decontamination process applied to deoxynivalenol (DON)-contaminated maize by sodium sulphite (Na2SO3) treatment in vivo. Additionally, in vitro characterisation of the toxicity of the DON sulphonates (DONS 1, 2 and 3 denote structurally different forms), the resulting DON metabolites, on peripheral blood mononuclear cells (PBMC) should substantiate the inactivation of DON. In a piglet experiment, both DON-contaminated maize and -uncontaminated control maize either untreated (DON?, CON?) or Na2SO3-treated (DON+, CON+) were mixed into feed and fed for 42 d starting from weaning. The results showed that feed intake and daily weight gain of animals fed DON? were significantly lower compared to animals fed CON? and CON+, whereas group DON+ reached the control level or even exceeded it. The feed-to-gain ratio was unaffected (p = 0.45). Furthermore, DON concentrations in plasma markedly reflected the diets’ DON concentrations. These were < 0.1, < 0.1, 5.4 and 0.8 mg/kg feed for CON?, CON+, DON? and DON+, and amounted to 0.3, 0.4, 33.0 and 9.3 ng/ml in plasma, respectively. Whereas DONS 2 and 3 were detected in the DON+ diet, only DONS 2 was recovered in plasma. Regarding the toxicity of DONS, no or much lower toxicity was found compared to DON. DONS 1 and Na2SO3 did not affect the viability of PBMC. At 32.71μM DONS2 the viability was reduced by 50% and thus this compound was less toxic than DON by a factor of 73. Consequently, wet preservation of maize with Na2SO3 was an effective tool to avoid the adverse effects of DON on performance of piglets.  相似文献

Pigs might be exposed to lipopolysaccharides (LPS) and deoxynivalenol (DON) at the same time, and both toxins are thought to interactively affect the intestinal barrier, the innate immune system, and the xenobiotics metabolism. Hence, we aimed at examining the single and combined effects of both toxins on nutrient digestibility and DON metabolism. For this purpose, barrows (26?±?4 kg) were fed restrictedly either a control diet (CON) or a diet contaminated with 3.1 mg DON/kg (DON) for 37 days. At day 37 of the experiment, pigs were infused intravenously for 60 min either with 100 μg DON/kg body weight (BW) (CON-DON), 7.5 μg LPS/kg BW (CON-LPS, DON-LPS) or a combination of both substances (CON-DON?+?LPS), or physiological saline (CON-CON, DON-CON). Blood samples were collected frequently until 3.25 h before the pigs were sacrificed for bile, liver, and kidney collection. The apparent digestibility of N-free extractives was significantly increased by 1 % when the DON-contaminated diet was fed. The total DON content in blood was significantly higher in endotoxemic pigs (34.8 ng/mL; CON-DON?+?LPS) when compared to the pigs infused with DON alone (18.8 ng/mL; CON-DON) while bile concentrations were not influenced by LPS. DON residue levels in liver and kidney closely reflected the treatment effects as described for blood. In contrast to DON infusion, the LPS challenge resulted in a significantly lower total DON concentration (13.2 vs. 7.5 ng/mL in groups DON-CON and DON-LPS, respectively) when the pigs were exposed to DON through the diet. The conjugation degree for DON in blood and bile was not influenced by treatments. In conclusion, endotoxemic pigs are characterized by higher DON residue levels in blood, liver, and kidney, probably by a compromised elimination.  相似文献

Abstract

Orally supplemented lactoferrin derived from bovine milk is purported to have beneficial effects on gut health of animals. Bovine lactoferrin (0, 60, or 120 mg/d) was fed to ileal cannulated, adult dogs in a replicated 3 × 3 Latin square design with 14 d periods. Control dogs tended (p = 0.06) to have higher fecal DM concentrations compared with dogs supplemented with 120 mg/d lactoferrin (34.5 vs. 32.9%). Fecal scores ranged from 3.0 – 3.3, suggesting that feces of all dogs was near the desired consistency, with dogs supplemented with 120 mg/d lactoferrin tending (p = 0.08) to have higher fecal scores. Ileal azoreductase activity tended (p < 0.10) to be higher in dogs supplemented with 60 or 120 mg/d lactoferrin (609 vs. 592 nmol/h per g ileal DM, respectively) as compared with unsupplemented dogs (272 nmol/h per g ileal DM). The following bacterial groups were measured: bifidobacteria, Campylobacter spp., Clostridium spp., eubacteria, Escherichia coli, Lactobacillus spp., Staphylococcus spp., and Streptococcus spp. Fecal streptococci concentrations were lower (p < 0.05) for dogs receiving 60 mg/d lactoferrin (8.60 log10 cfu/g fecal DM) as compared with unsupplemented dogs (9.19 log10 cfu/g fecal DM) or dogs receiving 120 mg lactoferrin/d (9.43 log10 cfu/g fecal DM). Dogs supplemented with 120 mg/d lactoferrin tended (p = 0.08) to have higher fecal indole concentrations as compared to unsupplemented dogs (1.80 vs. 1.46 μmol/g fecal DM). Because most bacterial groups measured were unaffected, it appears that lactoferrin did not exhibit prebiotic activity, and based on the data collected, lactoferrin also did not appear to have major effects on indices of health in the dog.  相似文献

Experiments were carried out to examine the effects of a Fusarium contaminated wheat (10mg deoxynivalenol and 0.76mg zearalenone, ZON, perkg dry matter) and of a detoxifying agent (Mycofix ®Plus, Biomin GmbH, Herzogenburg, Austria) on the growing performance of bulls, carry-over of ZON and its metabolites into body fluids and tissues, and on nutrient digestibility in wethers. The experiments were designed according to a complete two by two factorial approach which meant that both the uncontaminated control wheat and the Fusarium toxin contaminated wheat were tested both in the absence and presence of Mycofix ®Plus. The growing experiment with bulls (n = 14 per treatment) covered the live weight range between 244kg and 460kg. The respective wheat batches were included in the concentrate portion at 65%. Concentrates were fed according to plan whereas maize silage was offered for ad libitum consumption. Daily dry matter intake and live weight gain [kg per animal and day] were 7.40, 7.52, 7.51 and 7.49 and 1.367, 1.296, 1.380 and 1.307 for bulls fed the unsupplemented control wheat, the supplemented control wheat, the unsupplemented and Fusarium toxin contaminated wheat and the supplemented Fusarium toxin contaminated wheat, respectively. ZON and its metabolites were not detected in edible tissues. The most striking effects of feeding the Fusarium toxin contaminated wheat on carcass characteristics were a reduced dressing percentage, an increased weight of the emptied gastro-intestinal tract and a reduced weight of the testicles. No effect of the detoxifying agent was seen for these parameters whereas heart weight increased independently of Fusarium toxin contamination of the concentrates. Nutrient digestibility of the two wheat batches, unsupplemented or supplemented with Mycofix ®Plus was evaluated according to the difference method using wethers. Presence of Fusarium toxins in wheat did not influence its feeding value. The effects of the addition of the detoxifying agent were mycotoxin unspecific and resulted in an increase in apparent digestibility of crude protein and a decrease in crude fiber digestibility. It is concluded that feeding of Fusarium toxin contaminated wheat did not adversely affect performance of growing bulls (approximately 2.2mg DON and 0.1mg ZON perkg complete ration at a reference dry matter content of 88%) or nutrient digestibility in wethers. The effects of the detoxifying agent Mycofix ®Plus on growing performance and on nutrient digestibility were rather Fusarium toxin unspecific. The slightly negative effects on growing performance needs to be examined further.  相似文献

The wheat supply is known to have levels of deoxynivalenol (DON) from fungal infections. The fungal infections can be suppressed with benomyl (BEN). Under certain exposure laboratory conditions DON and BEN are developmental toxins. Since use of BEN may result in concurrent dietary exposure to both compounds, evaluation of the relative developmental risks is needed. The dietary NOAELs for pup decreased body weight for DON and BEN are 0.375 mg/kg/day in mice and 5 mg/kg/day in rat, respectively. Human exposure to DON can be estimated from surveys of wheat grain and finished products which gave a mean DON concentration of 0.86 ppm. The tolerance level for BEN in grain is 0.2 ppm. Presence of DON and BEN in wheat at these concentrations would not impact wheat consumption. A modified margin of exposure (modMOE) was developed using “a”; kg (food) /kg (body weight) as the constant for wheat consumption. The modMOE for DON is 28/a and that for BEN is 1667/a. Comparative relative risks can be expressed as the ratio of BEN risks to DON risks (60). Treatment with BEN at the most efficacious time decreased DON levels to 34% of controls, and decreased the comparative risk ratio from 60 to 18. The potential for fungicides to decrease mycotoxin health risks is important in the safety assessment for foodstuffs. This approach improves on current practices for food safety assessment by including the expected reductions in risks from naturally occurring mycotoxins in the regulatory analysis.  相似文献

Wheat infected naturally withFusarium, contaminated mainly with 18.53 mg DON per kg, was added to a total constant wheat proportion of 400g/kg diet. Control and DON contaminated feed was fed for 11 weeks underad libitum and restrictive feeding conditions to 48 pigs of both sexes, which were randomly divided into 4 groups. Effects on performance (live weight range between 26 and 100kg), duration of feed intake and blood parameters were investigated. Parallel to this study, a balance study was carried out to examine the effects on nutrient digestibility and DON metabolism. The group fed the DON contaminated rationad libitum consumed 15% less feed and gained 14% less live weight compared to thead libitum control group, while the feed to gain ratio was unaffected. Under restrictive feeding conditions (DON and control) pigs exhibited 33%, 25% and 10% lower feed consumption, live weight gain and feed to gain ratio, respectively, than the control group fedad libitum. Metabolizability of energy, digestibility of organic matter, crude protein, crude fat and N-retention were significantly increased by 4, 3, 6, 11 and 10%, respectively, in the DON group of the restrictively fed pigs. In average up to 43.2% of the ingested DON, as the parent toxin in both groups, was eliminated with the urine and up to 3.0% with faeces. DON fed animals needed more time to consume the restrictive ration than the control group. Presented at the 26th Mykotoxin-Workshop in Herrsching, Germany, May 17–19, 2004. The effect of diets differing in enzyme supplements, mineral phosphorus (P) and microwave wheat treatment on phytate hydrolysis and lower inositol phosphate isomers (InsPs) appearance in broiler crops was studied. The broilers (16- and 15-day-old) were assigned to 48 pens of 15 or 20 birds each (n = 8 pens per treatment) in Experiments 1 and 2, respectively. In Experiment 1, birds received a low-P wheat-soybean meal diet where the wheat was either microwave treated or not. These diets were offered without further supplementation or with added phytase (500 FTU/kg diet), alone or in combination with a xylanase (16,000 BXU/kg diet). In Experiment 2, two maize-soybean meal-based diets were fed, without or with monocalcium phosphate supplementation. Furthermore, these diets were offered without further supplementation or with phytase at 500 or 12,500 FTU/kg diet. On day 23 or 24 (Experiments 1 and 2, respectively), crop digesta were pooled per pen, freeze-dried and analysed for InsPs and the marker TiO2. Microwaving reduced the intrinsic phytase activity and InsP6 hydrolysis, but increased the concentration of Ins(1,2,3,4,5)P5 and Ins(1,2,4,5,6)P5 in the digesta of crop (Experiment 1). Microwave treatment significantly interacted with enzyme supplementation for Ins(1,2,5,6)P4 concentration, indicating a synergistic effect of intrinsic and supplied phytase in the crop. Xylanase tended to support phytase hydrolysis in diets with microwave-treated wheat. Phytase addition increased InsP6 hydrolysis up to 79% (Experiment 2). Thus, wheat phytase activity can cause high InsP6 hydrolysis in the crop. Treatment differences in lower InsPs indicated that hydrolysis of the first InsP6 phosphate group is not the only step in the degradation cascade in the crop of broilers that is influenced by dietary factors.  相似文献

Abstract

Fifteen Murrah buffalo calves (age about 10 months, 163 – 176 kg BW) were divided into three groups. Group I (Control) was fed a complete feed mixture consisted of 50% wheat straw and 50% concentrate mixture (contained per kg: maize 330 g, groundnut cake 210 g, mustard cake 120 g, wheat bran 200 g, de-oiled rice bran 110 g, mineral mixture 20 g and common salt 10 g) along with 2 kg green oats per animal and day to meet the vitamin A requirements. Calves of Groups II and III were fed with the Control diet supplemented with Orpinomyces sp. C-14 and Piromyces sp. WNG-12 cultures, respectively. The digestibility of DM was significantly highest with Piromyces sp. WNG-12 in Group III (62.2%) followed by Orpinomyces sp. C-14 in Group II (60.3%), and Control (53.5%). A similar pattern of increase in digestibility of crude protein and cell-wall contents was observed in treatment groups. The digestible energy in terms of percent total digestible nutrients was also significantly enhanced in Groups II (56.6%) and III (59.9%) when compared to Control (49.2%). The rumen fermentation parameters such as pH and NH3-N were found to be lower, whereas total nitrogen, tricarboxylic acid precipitable-nitrogen, total volatile fatty acids and zoospore counts per millilitre of rumen liquor were significantly higher in fungal administered groups. After administration of fungal cultures, improvements of animal growth rate (i.e. body weight gain) and feed efficiency were also observed.  相似文献

This study aimed to investigate a potential modulatory effect of E. coli lipopolysaccharide (LPS) on the kinetics of deoxynivalenol (DON) and zearalenone (ZEN) after pre- or post-hepatic LPS administration to unravel the putative role of the liver. Fifteen barrows were fed a diet containing mycotoxin-contaminated maize (4.59 mg DON/kg feed, 0.22 mg ZEN/kg feed) for 29 days and equipped with pre-hepatic catheters (portal vein, “po”) and post-hepatic catheters (jugular vein, “ju”), facilitating simultaneous infusion of LPS (“LPS group”, 7.5 μg/kg body weight) or 0.9% sterile NaCl solution (control, “CON group”, equivolumar to LPS group) and blood sampling. This resulted in three infusion groups, depending on infusion site: CONju-CONpo, CONju-LPSpo, and LPSju-CONpo. On day 29, pigs were fed their morning ration (700 g/pig) (?15 min), and blood samples were collected at regular intervals relative to infusion start. At 195 min, pigs were sacrificed and bile, urine, liquor, and liver samples collected. DON concentrations in jugular and portal blood decreased in both LPS-infused groups, whereas the ZEN concentrations increased, regardless of the treatment site. In liver tissue, a decrease of both toxin concentrations was observed in endotoxaemic pigs as well as a drop in hepatic conjugation, regardless of LPS entry site. In contrast to our hypothesis, DON and ZEN were not differently altered depending on the LPS-entry site. Neither the absorption nor the accumulation of DON and ZEN in different tissues differed significantly between animals which were infused with LPS via either the jugular or portal vein.  相似文献

Abstract

An experiment was conducted to study the effects of graded levels of choline addition (0, 500, 1000 and 4000 mg/kg diet) in laying hen diets prepared either with degummed or refined rape oil on the performance, sensory properties and trimethylamine (TMA) contents of the eggs. Furthermore, the diets containing no supplemented choline or 4000 mg choline/kg diet were tested with adequate or inadequate methionine supply (4.2 vs. 2.8 g methionine/kg diet). TMA metabolism and N-balance were measured for the latter diet types, but only with the diets containing refined rape oil. Therefore, a total of 12 and 4 diets were tested in the feeding (n = 60) and balance study (n = 9). Laying performance (23 – 75 weeks of age) was not significantly influenced by increasing choline additions with the exception of feed-to-egg mass ratio which decreased significantly linearly (p linear = 0.003). However, a significant interaction between choline addition and laying month was detected which was caused by a depression of performance of the unsupplemented control group occurring from the sixth laying month. The most obvious effect of an inadequate methionine supply was a temporary drop in performance between the third and sixth laying months. The mean TMA-concentration in pooled egg yolks [μg/g] increased with dietary choline concentration [mg/kg] in an exponentially related fashion (y = 1.14 + 4E?10 ? x2.71, r2 = 0.962) and suggested only a minor influence of total dietary choline on TMA content up to approximately 2000 mg choline/kg. Individual TMA-concentrations varied greatly from 0.4 – 1.5 μg/g, from 2.2 – 34 μg/g and from 18.4 – 75 μg/g for eggs with a normal, aberrant and heavily aberrant odour, respectively. It is concluded that a total choline concentration of at least approximately 1500 mg/kg is necessary to maintain a maximal laying performance. An inadequate methionine supply cannot be compensated by an increased addition of choline. Neither degummed nor refined rape oil influenced the TMA content of eggs.  相似文献

Six pregnant sows of 180.6 ± 5.6 kg were fed either a Fusarium-contaminated (4.42 mg DON and 48.3 μg ZON per kg, DON per os, n = 3) or a control diet (0.15 mg DON and 5 μg ZON/kg) in the period of days 63 and 70 of gestation. On day 63 of gestation, sows fed the control diet were implanted with an intraperitoneal osmotic minipump (delivery rate of 10 μL/h, for 7 days) containing 50 mg pure (98%) DON in 2 ml 50% DMSO (DON ip, n = 3). Frequent plasma samples were taken to estimate the kinetics after oral and ip DON exposure. The intended continuous delivery of DON by the intraperitoneal minipump could not be shown, as there was a plasma peak (Cmax) of 4.2–6.4 ng DON/mL either immediately (sow IP-2+3) or 2.5 h (sow IP-1) after implantation of the pump followed by a one-exponential decline with a mean half-time (t1/2) of 1.75–4.0 h and only negligible DON plasma concentrations after 12 h. Therefore, the DON ip exposure has to be regarded as one single dose 1 week before termination of experiment. The DON per os sows showed a mean basis level (after achieving a steady state) of DON plasma concentration of about 6–8 ng/mL, as also indicated by the plasma DON concentration at the termination of the experiment. On day 70, caesarean section was carried out, the fetuses were killed immediately after birth, and samples of plasma, urine, and bile were taken to analyze the concentration of DON and its metabolite de-epoxy-DON. At necropsy there were no macroscopic lesions observed in any organ of either sows or piglets. Histopathological evaluation of sows liver and spleen revealed no alterations. The proliferation rate of peripheral blood mononuclear cells (PBMC) with or without stimulation was not affected by the kind of DON treatment. The exposure of pregnant sows at mid-gestation (days 63–70, period of organogenesis) to a Fusarium toxin-contaminated diet (4.42 mg DON and 0.048 mg ZON per kg) or pure DON via intraperitoneal osmotic minipump did not cause adverse effects on health, fertility, maintenance of pregnancy, and performance of sows and their fetuses. However, DON was detected in fetus plasma, indicating that this toxin can pass the placental barrier and may cause changes in the proportion of white blood cells (lower monocyte and neutrophil and higher lymphocyte proportion in DON per os fetuses).  相似文献

The aim of the present experiment was to test the effects of a wet preservation of triticale contaminated mainly with deoxynivalenol (DON) with sodium metabisulphite (Na2S2O5, SBS) on growth performance, liver function, clinical-chemical plasma parameters and organ histopathology of piglets. For this purpose both the uncontaminated control triticale and the DON contaminated triticale were included in the piglet diet either untreated (CON, FUS) or SBS-treated (CON-SBS, FUS-SBS) and fed for 28 d starting from weaning. The dietary concentrations of DON and DON sulfonate (DONS), the DON derivative resulting from the SBS treatment, amounted to 0.156, 0.084, 2.312 and 0.275 mg DON per kg CON, CON-SBS, FUS and FUS-SBS diet, and to <0.05, <0.05, <0.05 and 1.841 mg/kg diet, respectively. Feeding the FUS diet significantly reduced the feed intake compared to the other three groups as indicated by the significant interactions between triticale source and SBS treatment when the whole experimental period of 28 d was considered (p = 0.014) while live weight gain and feed to gain ratio remained unaffected. The total plasma protein concentration was significantly depressed due to feeding the contaminated diets whereas SBS treatment exerted an increasing effect at the same time (45.4, 49.5, 40.7 and 46.5 g/l for piglets fed the CON, CON-SBS, FUS and FUS-SBS diet, respectively). The liver function was tested by the 13C-methacetin breath test (MBT) allowing evaluation of the cytochrome P4501A2 activity. MBT results, expressed as cumulative percentage dose recovery after 360 min (cPDR360) revealed a slight stimulation of liver function due to SBS treatment (p = 0.052) (37.5, 39.4, 37.4 and 55.1% for piglets fed the CON, CON-SBS, FUS and FUS-SBS diet, respectively). Liver weight and histopathological scoring were only weakly related to the MBT results. Further histopathological examinations of kidneys, pancreas and heart revealed no treatment effects. It was concluded that the SBS treatment of the contaminated triticale restored the performance of piglets to the level of the piglets fed the control diet while the effects on liver function, clinical-chemical plasma parameters - excepting the protein concentration - and organ histopathology were only marginal.  相似文献

Effects of the mycotoxin deoxnivalenol (DON) on weaned piglets were investigated. Two feeding trials were conducted with naturally contaminated wheat in the ration. In the first trial as preliminary study weaned piglets were fed for one week with 7.7 mg DON per kg ration, intake of DON contaminated diets was not associated with any obvious negative health effects. Also in the main feeding trial with 3 mg DON per kg ration no vomiting and other negative clinical symptoms could be registered. The feed intake and the daily weight gains were significantly lower in the DON fed group when compared with control group, but the feed conversion rates were slightly improved in group fed DON.

Four diets contaminated with 1.1 to 5.0 mg/kg deoxynivalenol (DON) and 0.4 to 2.4 mg/kg zearalenone (ZEA) were fed to four groups of six growing Large White pigs. Urine samples were collected after 3 to 4 days and again after 6 to 7 days on the diets. On each sampling day, half of the animals were sampled in the morning, after an 8-h fast, and the other half were sampled in the afternoon, after 7 h of ad libitum access to feed. The urinary concentrations of DON, DON-glucuronide, DON-3-sulphate, de-epoxy-DON, as well as of ZEA, ZEA-14-glucuronide, α-zearalenol and α-zearalenol-14-glucuronide, analysed using LC-MS/MS, were used to calculate urinary DON and ZEA equivalent concentrations (DONe and ZEAe). The urinary concentration of DONe (P?<?0.001), but not of ZEAe (P?=?0.31), was lower in the fasted than that in the fed animals. The urinary DONe/creatinine and ZEAe/creatinine ratios were highly correlated with DON and ZEA intake per kg body weight the day preceding sampling (r?=?0.76 and 0.77; P?<?0.001). The correlations between DON intake during the 7 h preceding urine sampling in the afternoon and urinary DONe/creatinine ratio (r?=?0.88) as well as between mean ZEA intake during 3 days preceding urine sampling and urinary ZEAe/creatinine ratio (r?=?0.84) were even higher, reflecting the plasma elimination half-time of several hours for DON and of more than 3 days for ZEA. ZEAe analysed in enzymatically hydrolysed urine using an ELISA kit was highly correlated with the LC-MS/MS data (r?=?0.94). The urinary DONe and ZEAe to creatinine ratios, analysed in pooled urine samples of several pigs fed the same diet, can be used to estimate their exposure to DON and ZEA.  相似文献

The commercially available dog food samples (29 dry foods and 11 wet foods) were analysed for deoxynivalenol (DON) and ochratoxin A (OTA) using ELISA.
