Summary The concentrations of pyruvate, lactate, oxalo-acetate, aceto-acetate -hydroxybutyrate, -ketoglutarate, glutamate, NH 4 + , NAD+ and NADH were measured in goldfish tissues after previous conditioning to normal and anoxic (12h) conditions. For 11 different metabolites efficiency of different extraction methods was tested by means of internal standards. The recoveries were generally over 80%. The substrate/product couples of the reactions catalysed by lactate dehydrogenase, malate dehydrogenase, -hydroxybutyrate dehydrogenase and glutamate dehydrogenase were used as redox parameters. In the lateral red muscle the redox state did not change during 12 h of anoxia. In the dorsal white muscle only the cytoplasmic redox state underwent a change, as indicated by the increase of the lactate/pyruvate ratio from 20 to 110. In liver both cytoplasm and mitochondria were reduced during anoxia. From the measured values the NAD+/NADH ratio was found to change only in white muscle, while the calculated free NAD+/NADH ratios were reduced in anoxic white muscle cytoplasm, anoxic liver mitochondria, and anoxic liver cytoplasm. Oxalo-acetate concentrations calculated from the equilibrium constants of lactate dehydrogenase and malate dehydrogenase were at least one order of magnitude smaller than the measured values. The data obtained from anoxic goldfish are in contrast to available data on other animals and support earlier reports which indicate that this animal has a special anaerobic metabolism. Previous work showed that goldfish learn to avoid a region of an aquarium where they receive a discrete shock to the flank. This avoidance is reduced if another fish is present next to the shock zone, suggesting fish trade-off avoidance against a tendency to associate [Dunlop, R., Millsopp, S., Laming, P., 2006. Avoidance learning in goldfish (Carassius auratus) and trout (Oncorhynchus mykiss) and implications for pain perception. App. Anim. Behav. Sci. 97 (2–4), 255–271]. Only shock intensity was adjusted in those experiments, here we report results where a requirement for food was also altered.Fish were trained to feed at a region of the aquarium where subsequently they would receive a discrete shock to the flank. One experimental group had different shock intensities applied, whereas in the other group shock intensity was consistent but food deprivation was varied.We show the number of feeding attempts and time spent in the feeding/shock zone decreased with increased shock intensity and with increased food deprivation the number and the duration of feeding attempts increased as did escape responses as this zone was entered. These data suggest that goldfish balance their need for food against avoidance of an acute noxious stimulus.  相似文献

Subcellular modifications in hepatocytes of Carassius carassius var. auratus subjected to 24 hr and 48 hr sublethal acute lead (5mg·1−1) exposure were studied by electron microscopy. Cytological alterations were observed after 24 hr of treatment and became more evident after 48 hr. Lead induced an increase in nuclear heterochromatin and alterations in mitochondria, endoplasmic reticulum and Golgi complex ultrastructure. Glycogen granula decreased, and secondary lysosomes and lipid droplets increased. Furthermore, intracytoplasmic lumina with microvilli-bearing surfaces and numerous autophagic vacuola were observed after 48 hr of exposure.  相似文献

Summary Goldfish acclimated to normal oxygen levels and to 20°C were made anoxic and injected i.p. with U-14C-glucose, 6-14C-glucose, U-14C-lactate, 3-14C-lactate, 1-14C-acetate or 3,4-14C-glutamate. Radioactivity released into the water (total14C and14CO2) was monitored over a period of about 12 h. With the exception of 3,4-14C-glutamate from which only 4% was released, the release of14C from the other compounds was found to be over 30%. The fraction of the radioactivity released as CO2 varied with the compound injected but was high during the first 4 h after injection. It is argued that the acid-stable14C component is ethanol, which arises by the combined action of a modified pyruvate dehydrogenase and of alcohol dehydrogenase in muscle (Shoubridge and Hochachka 1980; Mourik et al. 1982).14CO2 release from 3-14C-lactate, 6-14C-glucose, 3,4-14C-glutamate and 1-14C-acetate cannot be explained by ethanol fermentation. Neither was there a stoichiometric relation between14CO2 and14C-ethanol release after U-14C-glucose and U-14C-lactate injection. It is concluded that at least 20% of the CO2 released is produced by Krebs cycle activity.  相似文献

Summary Changes in the concentrations of ammonia, glutamate, alanine, aspartate, -ketoglutarate, oxaloacetate and succinate were measured in freeze-clamped lateralred muscle, dorsal white muscle and liver, and in rapidly cooled blood of goldfish after 12 h of anoxia. Alanine accumulation, succinate accumulation and aspartate depletion are observed in all tissues examined; in the liver the concentrations of glutamate increase and those of ammonia decrease. The mass-action ratio of the glutamate-pyruvate transaminase-catalyzed reaction stays within one order of magnitude from thermodynamic equilibrium in the direction of alanine formation. The mass-action ratio of the glutamate-oxaloacetate transaminase reaction is far from equilibrium when measured oxaloacetate concentrations are used. When levels of free oxaloacetate are calculated from LDH and MDH equilibrium constants, the mass-action ratio of glutamate-oxaloacetate transamination is close to equilibrium in the direction of aspartate formation. Since neither alanine nor glutamate decreases, and since ammonia gradients suggest a continuous ammonia production in all tissues examined, anaerobic proteolysis is assumed. In this study we investigated the viable but non-culturable (VBNC) state of Aeromonas hydrophila and its virulence in goldfish. Aeromonas hydrophila cultured in a 0.35% NaCl solution at pH 7.5 and at 25 °C for 50 days showed the VBNC state. In the VBNC state we were unable to detect viable bacteria by the plate count method but we did find 104 cells/ml by the direct viable count microscopical method after staining with fluorescein diacetate and ethidium bromide. The virulence comparison in goldfish showed that bacteria cultured at 25 °C for 1 day in a 0.35% NaCl solution were more virulent than bacteria cultured for 28 days. VBNC bacteria showed lower virulence in goldfish compared to 28-day-cultured bacteria by intraperitoneal injection.The results from the study suggest that A. hydrophila can remain in the aquatic environment for prolonged periods in the VBNC state but those cells are not pathogenic to goldfish.  相似文献

Summary Goldfish (Carassius auratus) were acclimated for 5 months at temperatures of either 2°C or 31°C. Natural actomyosin was prepared from white myotomal muscle and its Mg2+Ca2+ ATPase activity determined. Temperature acclimation results in adaptations in substrate turnover number and thermodynamic activation parameters of the ATPase. When assayed at 31°C the Mg2+Ca2+ ATPase of natural actomyosin was 4 times higher in 31°C than 2°C acclimated fish. Arrhenius plots of natural actomyosin ATPase from cold acclimated fish show a break in slope at 15–18°C. In contrast, the temperature dependence of warm acclimated actomyosin was linear. Activation enthalpy (H ) of the ATPase, calculated over the range 0–16°C, was approximately 8,000 cal/mole lower in 2°C than 32°C acclimated fish.In contrast, desensitised actomyosins from which the calcium regulatory proteins have been removed show a linear temperature dependence in the range 0–32°C and have similar properties in 2°C and 31°C acclimated fish. Cross-hybridisation of regulatory proteins (tropomyosin-troponins complex) from cold-acclimated fish to desensitised actomyosin from warm-acclimated fish alters the ATPase towards that of cold-acclimated natural actomyosin and vice versa. The results suggest that the regulatory proteins can influence the kinetics of the ATPase and, furthermore, that they are involved in the acclimation of the actomyosin to different cell temperatures.  相似文献

1. 1.|The mitochondrial population in hypothalamic and hypophysial brain tissue from warm (30°C) and cold (5°C) acclimated goldfish (Carassius auralus L.) was analyzed using sterological techniques.

2. 2.|It was revealed that there is a significantly larger volume density (Vv) in the cold acclimated tissue, with no significant difference in either of the surface densities (Svext and Svint) from either of the brain areas.

3. 3.|The hypothalamic brain tissue has a significantly lower specific surface (S/V) in the cold acclimated tissue but there is not a significant difference in this parameter for the hypophysial brain tissue.

4. 4.|The values for these three parameters (Vv, Svext and SVint, and S/V) indicate that mitochondria from acclimated brain tissue undergo shape changes in response to thermal stress.

5. 5.|We suggest that the shape changes may be related to the change in the phospholipid composition of the inner mitochondrial membrane with acclimation temperature.

Summary Goldfish (Carassius auratus L.) were acclimated to environmental temperatures of 3 °C, 18 °C and 31 °C for a period of three months. Cytochemical techniques were used to study the metabolism and myofibrillar ATPase activities of individual muscle fibres. Fish muscle is composed of three basic fibre types each with distinct contractile and metabolic characteristics. Cold acclimation resulted in a shift to a more aerobic type of metabolism, particularly in the red and pink fibres. In addition, environmental temperature was found to affect the size and relative distribution of the different fibre types in the myotome. The total number of pink and red fibres increased significantly with cold acclimation. Mechanisms of environmentally-induced adaptation of muscle fibre phenotype are discussed.In addition to changes in the metabolism and distribution of muscle-fibre types, biochemical studies have provided evidence for different kinetic forms of Mg2+Ca2+ myofibrillar ATPase at different environmental temperatures. Activities of myofibrillar ATPase assayed at 31 °C were 2–3 times higher in fish acclimated to the higher environmental temperature. Activation enthalpy (H ) of the ATPase was also signficantly reduced in the cold adapted enzyme. Reduction of H in the cold acclimated ATPase is thought to reduce the temperature sensitivity of the activation process thus partly compensating for the reduced cell temperature.  相似文献

Summary Measurements of electric potential and of sodium and chloride effluxes were carried out on a freshwater fish, the goldfishCarassius auratus. The potential was affected by a number of inorganic ion species but that with the most significant influence on transepithelial potential was calcium. The potential was about 10 mV negative in the absence of calcium and about 10 mV positive in the presence of 10 mM/l calcium. Sodium and chloride efflux rates were greatly reduced upon the addition of calcium to the medium. The origin of the transepithelial potential is discussed in terms of the relative permeability of the branchial membranes to sodium and chloride and the factors influencing these permeabilities.  相似文献

Summary The presence of immunoreactive pro-opiomelanocortin (POMC)-derived peptides (adrenocorticotropin hormone, β-endorphin, α-melanocyte-stimulating hormone) and of cytokine-like molecules [interleukin (IL)-1α, IL-1β, IL-2, IL-6, tumour necrosis factor-α] was demonstrated in periodic acid-Schiff-positive epithelial cells in the thymus of the goldfish (Carassius c. auratus) using immunocytochemical procedures. POMC-derived peptide- and cytokine-like molecules were localized in the same cell type. Lymphocytes were negative for all the above mentioned molecules. Despite the smaller number of cells positive for neuropeptide- and cytokine-like molecules, our findings suggest that immune-neuroendocrine interactions are likely to occur in the thymus of goldfish.  相似文献

【目的】了解水环境中重金属铜对异育银鲫肠道微生物组成及多样性的影响。【方法】采用试剂盒提取异育银鲫肠道总DNA,然后对总DNA进行16S r RNA进行扩增,构建异育银鲫肠道微生物16S r RNA基因克隆文库,最后进行数据分析。【结果】厚壁菌门、变形菌门和拟杆菌门为异育银鲫肠道中主要的细菌类群,在不同浓度的重金属铜胁迫处理后,厚壁菌门的含量明显降低。稀释性曲线、Venn图和多样性指数分析结果表明,重金属铜胁迫处理后异育银鲫肠道微生物多样性明显降低。【结论】重金属铜会使异育银鲫肠道微生物组成及多样性降低。此结果为研究重金属污染对异育银鲫健康状况的影响及异育银鲫养殖过程中病害的诊断奠定基础。  相似文献

This study reports the effect of aqueous, ethanol and methanol triherbal solvent extract from Azadirachta indica, Ocimum sanctum and Curcuma longa on innate immune mechanisms such as phagocytosis activity, respiratory burst activity, alternative complement activity and lysozyme activity and disease resistance in goldfish (Carassius auratus) against Aeromonas hydrophila. Fish were intraperitoneally injected with different doses of 0, 5, 50 and 100 mg kg−1 body weight of each triherbal solvent extracts. The functional immunity in terms of percentage mortality and Relative Percent Survival (RPS) and innate immune response was assessed on week 1, 2 and 4 by challenging with live A. hydrophila (1 × 107 cells ml−1). All the chosen innate immune parameters were enhanced in the ethanol and methanol triherbal solvent extract treatment after week 2. However, the aqueous triherbal extract was enhanced only after week 4. The ethanol and methanol triherbal solvent extracts administration preceding the challenge with live A. hydrophila decreased the percentage mortality in the experimental groups with the consequence increase in RPS values. The study indicates that all the doses of ethanol or methanol triberbal solvent extracts could be positively influence the immune response and protect the heath status of goldfish against A. hydrophila infection.  相似文献

The exploratory locomotor behaviour of twenty-one naïve goldfish in a homogeneous open field during 20-hr periods was monitored and analysed into spatial and temporal characteristics. The fish did not randomly explore the entire field. Locomotion is organized spatially and temporally. The fish moves about in one specific area during a specific time after which it displaces the centre of its activity to another area. The time spent in earch area is a function of rate of activity, expressed by a linear equation and, during the initial 4 hr, the elapsed time in the novel environment (novelty content). The area-by-area exploration was never replaced by random exploration throughout the environment during the 20-hr periods of observation.  相似文献

先捕后放是一种重要的鱼类资源保护策略和渔业管理手段。经历先捕后放的被钓鱼类个体的生理与行为会发生变化。当这些个体再次面对人工垂钓诱饵时,可能会呈现更加谨慎的摄饵或鱼钩回避行为。为考察重复垂钓对鱼类易钓性、能量代谢和个性的影响,本研究以鲤科鱼类鲫(Carassius auratus)幼鱼为实验对象,在(25.3±0.1)℃水温条件下对垂钓组和对照组进行3次的能量代谢(标准代谢率,SMR;最大代谢,MMR)和个性(勇敢性、活跃性)测定。其中,垂钓组需进行垂钓实验,而对照组不进行任何垂钓操作。研究发现:(1)重复垂钓显著降低鲫幼鱼的垂钓率,但明显增加单尾平均垂钓时间和整体死亡率。(2)重复垂钓降低鲫幼鱼的体重、肥满度、SMR、MMR、代谢空间(AS)和相对代谢空间(FAS),但整体上不影响勇敢性和活跃性。(3)在测定Ⅰ中,高易钓性个体的体长和开阔区运动时间比大于低易钓性个体,但前者的肥满度小于后者。然而,在测定Ⅱ和Ⅲ中,高易钓性个体的形态参数、能量代谢、个性(勇敢性和活跃性)与低易钓性个体并无差异。研究表明:重复垂钓明显降低鲫幼鱼的易钓性,并且种内个体易钓性的变化方向不尽相同;重复垂钓对鲫幼...  相似文献

为考察鲫(Carassius auratus)幼鱼标准代谢的个体差异与运动力竭后代谢特征和行为的关系,在(25.0±0.5)℃条件下测定80尾鲫幼鱼标准代谢率(SMR),筛选出40尾实验鱼[体重(13.54±0.20)g,体长(8.05±0.07)cm],其中包括20尾高SMR个体和20尾低SMR个体,测定运动至力竭实验鱼的最大代谢率(MMR),并计算代谢空间(AS=MMR-SMR)、相对代谢空间(FAS)和过量耗氧(EPOC)总量,随后测定单尾鱼的快速启动行为[反应时间(RT)、最大线速度(Umax)、最大线加速度(Amax)和120ms移动距离(S120)]以及个性行为(勇敢性和活跃性)。结果显示鲫幼鱼的SMR与AS、FAS和EPOC总量均呈负相关,而与MMR不相关;MMR与AS、FAS和EPOC总量均呈正相关;协方差分析显示高SMR个体组的代谢恢复速率与低SMR个体组无显著差异。鲫幼鱼的SMR与快速启动行为的RT呈正相关,与Amax呈负相关,而与Umax和S120均不相关。在快速启动行为中,鲫幼鱼的RT与Umax、Amax和S120均呈负相关,而Umax与Amax和S120呈正相关。鲫幼鱼的SMR与其勇敢性指标如潜伏期(L)、曝露时间(ET)和探头频率(AF)以及活跃性指标如运动时间比(PTM)、撞墙频率(FHW)均不相关,但鲫幼鱼的PTM与FHW和ET呈正相关。研究表明在实验室条件下鲫幼鱼的能量代谢特征、快速启动和个性行为存在明显的个体差异现象,并且部分表型特征之间存在权衡,提示这些权衡可能是鲫幼鱼的生理、行为与栖息环境相互作用的综合结果。  相似文献

Summary The effect of hypoxia was studied in cold (15°C) and warm (30°C) acclimated goldfish. The hypoxic thresholds, defined as the lowest sustainablePO2 were found to be 1.6 and 4.0 kPa O2 at, respectively, 15°C and 30°C. At these levels the fish did not loose either weight or appetite over a 2-months period. While during starvation under normonic conditions a significant weight loss and breakdown of lactate dehydrogenase (90%) was observed, no such changes were found in fed hypoxic animals. In red lateral muscle, white epaxial muscle and liver of goldfish from 4 differently acclimated groups the maximal activities were measured of: glycogen phosphorylase, hexokinase, malate dehydrogenase, glycerol-3-P dehydrogenase, glucose-6-P dehydrogenase, malic enzyme, succinate oxidase, pyruvate carboxylase, phosphoenol-pyruvate carboxykinase, fructose-bisphosphatase and glucose-6-phosphatase. Thermal compensation, according to Precht's typology, was predominantly observed in red muscle and to a lesser extent in white muscle. The liver glucose-6-P dehydrogenase showed a strong inverse response, which points to enhanced synthetic activity at the higher temperature. Hypoxia acclimation exerted weaker responses at 15°C than at 30°C. Changes in liver enzyme activities suggest depressed protein synthesis and enhanced gluconeogenesis in hypoxic animals. In muscle of 30°C-acclimated goldfish hypoxia induces a significant increase of succinate oxidase activity, indicating adaptation of the aerobic energy metabolism. The occurrence of pyruvate carboxylase, never before observed in vertebrate muscle, probably plays an important role in pyruvate catabolism. Because its action produces oxalo-acetate, the enzyme may stimulate pyruvate oxidation and thus prevent early lactate accumulation. Since all gluconeogenic enzymes were shown to be active in goldfish muscle, the possible occurrence of gluconeogenesis in muscle (albeit at low rate) must be accepted. Enzyme activities in goldfish muscle were compared with literature data for a number of other fish species. This comparison indicates that maximal glycolytic flux in goldfish muscle tissue is rather low, although muscular glycogen levels are very high. It is suggested that this is part of the gold-fish's strategy to cope with hypoxia.  相似文献
