Biomass production and carbohydrate reduction were determined for a two-stage continuous fermentation process with a simulated potato processing waste feed. The amylolytic yeast Saccharomycopsis fibuligera was grown in the first stage and a mixed culture of S. fibuligera and Candida utilis was maintained in the second stage. All conditions for the first and second stages were fixed except the flow of medium to the second stage was varied. Maximum biomass production occurred at a second stage dilution rate, D(2), of 0.27 h (-1). Carbohydrate reduction was inversely proportional to D(2), between 0.10 and 0.35 h (-1).  相似文献

1. Changes in dry weight, DNA, RNA, protein and reserve and structural carbohydrate were measured during the aerobic growth of yeast on 0.9% glucose in an aerobic synthetic medium. 2. After glucose had been consumed and during the growth of yeast on ethanol and acetate, the rate of formation of DNA remained about the same but the rate of increase of dry weight was greatly diminished. 3. During the second stage of growth the ratios dry weight/DNA, protein/DNA, RNA/DNA and carbohydrate/DNA decreased to about 30% of the corresponding values during the first stage of growth. 4. A higher fraction of the dry weight of the yeast cells could be accounted for by the reserve carbohydrate content of the cells during the second stage of growth. 5. By the end of the first stage of growth an increase in the reserve carbohydrate content of the cells was observed. Part of this reserve carbohydrate was consumed by the cells in the beginning of the second stage of growth. The possibility of adaptation of cells at the expense of their reserves is discussed.  相似文献

A leucine auxotroph strain of Saccharomyces cerevisiae was used to study plasmid stability and expression using a recombinant plasmid, which contained a foreign gene for firefly luciferase (luc). This recombinant yeast was tested in a series of continuous cultures in semi-defined media with varying concentrations of yeast extract in order to study its effect on stability. While the biomass concentration and luciferase activity increased with increasing concentrations of yeast extract, the plasmid stability declined. An analysis of the growth rates showed that the recombinants enjoyed a growth rate advantage over the plasmid-free cells at critically low yeast extract concentrations, possibly due to leucine starvation in the media. A two-stage cultivation strategy was designed in order to create a yeast extract limited environment so that plasmid-free cells could not grow and overtake the recombinant cells. The cells were cultivated in selective media in the first stage, and then transferred continuously to the second stage where the media was enriched by feeding yeast extract. The feed rate was kept low in order to ensure yeast extract and hence leucine starvation, thereby selecting against the plasmid-free cells. This strategy resulted in a stable existence of recombinant cells, which stabilized around 60% at steady state during the tested period of cultivation. The complex nitrogen feed helped in increasing the cell density and volumetric activity by approximately 9 and 18-fold respectively with respect to that achieved in minimal medium. The experimental data was used to formulate a mathematical model to predict cell growth and plasmid stability in two-stage cultivation, which correctly explained the experimental data.  相似文献

The influence of five yeast strains on the nitrogen fractions, amino acids, peptides and proteins, during 12 months of aging of sparkling wines produced by the traditional or Champenoise method, was studied. High-performance liquid chromatography (HPLC) techniques were used for analysis of the amino acid and peptide fractions. Proteins plus polypeptides were determined by the colorimetric Bradford method. Four main stages were detected in the aging of wines with yeast. In the first stage, a second fermentation took place; amino acids and proteins plus polypeptides diminished, and peptides were liberated. In the second stage, there was a release of amino acids and proteins, and peptides were degraded. In the third stage, the release of proteins and peptides predominated. In the fourth stage, the amino acid concentration diminished. The yeast strain used influenced the content of free amino acids and peptides and the aging time in all the nitrogen fractions. A Pseudomonas luteola strain possessing azoreductase activity was utilized to decolorize a reactive azo dye (C. I. Reactive Red 22) with fed-batch processes consisting of an aerobic cell growth stage and an anaerobic fed-batch decolorization stage. The fed-batch decolorization was conducted with different agitation and aeration rates, initial culture volumes, dye loading strategies, and yeast extract to dye (Y/D) ratios, and the effect of those operation parameters on azo dye decolorization was evaluated. Dissolved oxygen strongly inhibited the azo reduction activity; thus aeration should be avoided during decolorization but slight agitation (around 50 rpm) was needed. With the periodical feeding strategy, the specific decolorization rate (v(dye)) and overall decolorization efficiency (eta(dye)) tended to increase with increasing feeding concentrations of dye, whereas substrate inhibition seems to arise when the feeding concentration exceeded 600 mg dye/L. In the continuous feeding mode, higher initial culture volume resulted in better eta(dye) due to higher biomass loading, but lower v(dye) due to lower dye concentration in the bioreactor. With a volumetric flow rate (F) of 25 mL/h, both v(dye) and eta(dye) increased almost linearly with the increase in the loading rate of dye (F(dye)) over the range of 50-200 mg/h, while further increase in F(dye) (400 mg/h) gave rise to a decline in v(dye) and eta(dye). As the F was doubled (50 mL/h), the v(dye) and eta(dye) increased with F(dye) only for F(dye) < 80 mg/h. The best v(dye) (113.7 mg dye g cell(-)(1) h(-)(1)) and eta(dye) (86.3 mg dye L(-)(1) h(-)(1)) were achieved at F(dye) = 200 mg/h and F = 25 mL/h. The yield coefficient representing the relation between dye decolorized and yeast extract consumed was estimated as 0.8 g/g. With F(dye) = 75 mg/h, the Y/D ratio should be higher than 0.5 to ensure sufficient supply of yeast extract for stable fed-batch operations. However, performance of the fed-batch decolorization process was not appreciably improved by raising the Y/D ratio from 0.5 to 1.875 but was more sensitive to the changes in the dye loading rate.  相似文献

Suspensions of yeast cell (zygo Saccharomyces bailii) in a phosphate buffer solution were subjected to conventional (hot water) and ohmic (electric current) heating under identical temperature histories. Experiments were also conducted with cells of Escherichia coli to compare the lethal effect of combination of sublethal electrical preteatment and conventional heating with conventional heating. The kinetic parameters (D,Z,K and E(a)) were determined for both organisms during different treatments. There was no significant difference in the death rate of yeast cells during conventional and ohmic heating at the voltage range used in this study. Results of electrical pretreatment and conventional heating on E. coli indicated differences under certain conditions when compared with pure conventional heating. Thus it is concluded that microbial death during ohmic heating was due primarily to thermal effects with no significant effect of electric current per se. Sublethal electrical pretreatment appears to offer potential for increased bacterial inactivation in certain cases.  相似文献

As part of the process optimization of a two-stage continuous culture system, the effect of growth rate mu(2) (app) on the performance of the second stage (production stage) was studied in a recombinant Escherichia coli K12 (DeltaH1Deltatrp/pPLc23trpA1). Important parameters considered were specific gene expression rate, plasmid content, and plasmid stability, all of which were closely related to the cell growth rate and the production rate of the cloned gene product (trpalpha). When operating conditions were maintained constant (T(1) = 35 degrees C, D(1) = 0.9 h(-1), T(2) = 40 degrees C, and D(2) = 0.7 h(-1)) and mu(2) (app) was varied, plasmid content in the second stage showed its maximum at mu(2) (app) = 0.4 h(-1) and decreased thereafter. Specific gene expression rate linearly increased with increasing mu(2) (app), while plasmid stability decreased. Optimum cell growth rate giving the maximum value in overall productivity was observed at around mu(2) (app) = 0.4 h(-1). The contribution or role of the three parameters, specific gene expression rate, plasmid content, and plasmid stability in exhibiting the maximum productivity at the optimal mu(2) (app) is discussed.  相似文献

The effect of yeast hulls (yeast ghosts) on sluggish or stuck white wine fermentations was studied. The enhancing effect on yeast growth and fermentation rate displayed by the hulls was shown to be similar to the effect provided by lipid extract from the same hulls. Unsaturated fatty acids and sterols were incorporated into the yeast from lipid extracts during fermentation carried out under oxygen-limited conditions. Adsorption of toxic medium-chain fatty acid (decanoic acid) onto the yeast hulls took place through a dialysis membrane. However, when the hulls were placed inside a dialysis bag, the increase in yeast growth and fermentation rate seen when freely suspended hulls were used did not occur. Accordingly, the effect of yeast hulls in preventing stuck fermentations cannot be attributed only to the adsorption and consequent removal of medium-chain fatty acids from the juice.  相似文献

In embryos of the gastropod Ilyanassa obsoleta, the first-quartet micromeres of the A, B and C lineages (1a, 1b, and 1c) are each competent to form an eye in response to signaling from the 3D cell. The first-quartet micromere of the dorsal D lineage (1d) is smaller than the others, divides at a slower rate, and lacks the ability to form an eye. These properties of 1d all depend on inheritance of vegetal polar lobe cytoplasm by its mother cell D at second cleavage. I show that they depend also on the presence of cells adjacent to D during the late four-cell stage: after ablation of the A and/or C cells before this stage, 1d inherits more cytoplasm than normal, divides more rapidly, and frequently forms an eye. In non-D lineages, cleavage plane positioning and micromere division rates are relatively insensitive to cell contacts. Compressing whole embryos during third cleavage also leads to an increase in 1d volume correlated with abnormal eye formation; this suggests that the normal effect of cell contacts is to position the D cell cleavage furrow closer to the animal pole, and the enhanced division asymmetry of the D cell contributes to the suppression of eye development.  相似文献

Using either permeabilized cells or microsomes we have reconstituted the early events of the yeast secretory pathway in vitro. In the first stage of the reaction approximately 50-70% of the prepro-alpha-factor, synthesized in a yeast translation lysate, is translocated into the endoplasmic reticulum (ER) of permeabilized yeast cells or directly into yeast microsomes. In the second stage of the reaction 48-66% of the ER form of alpha-factor (26,000 D) is then converted to the high molecular weight Golgi form in the presence of ATP, soluble factors and an acceptor membrane fraction; GTP gamma S inhibits this transport reaction. Donor, acceptor, and soluble fractions can be separated in this assay. This has enabled us to determine the defective fraction in sec23, a secretory mutant that blocks ER to Golgi transport in vivo. When fractions were prepared from mutant cells grown at the permissive or restrictive temperature and then assayed in vitro, the acceptor Golgi fraction was found to be defective.  相似文献

The dinuclear micro-okso vanadium (III) complex compound H(4)V(2)OCl(4)(Ad)(2) synthesized in our laboratory was investigated as a potential cytotoxic agent against yeast cells. The results of these studies could be helpful in the explanation of the mechanism governing the V (III) compound action on yeast as a simple model of eukaryotic cells. The important factors influencing the toxicity of this complex compound are: the stage of the yeast life cycle, the rate of growth and the pH of reaction mixture. The lethal effect was distinctly stronger when the reaction mixture was slightly acidic (pH = 4). In such solutions V(III) mononuclear species with adenine was relatively stable, and during the time of experiment possibly only a slow oxidation process to V(IV) occurred. In the solutions with pH = 7, several hydrolytic, perhaps mixed-valence, polynuclear species were present and their action on the yeast cells was rather weak. The increased lethal activity of this compound in acidic solutions may be useful in specific treatment against cancer cells whose cytoplasm and/or closest surrounding has lower pH value. The next important result was an observation that the killing activity of this compound was enhanced for yeast cells being in log phase. Also these which had a slower rate of growth (possessing some auxotrophic mutations) were more resistant than those growing faster. The extent of yeast mutagenesis caused by the complex compound is negligible, as the number of mutants found in experiments was within the limit of experimental error. These results are promising and the investigated complex can be considered as a potential anti cancer agent.  相似文献

To determine the effect of organics (yeast extract) on microbial community during chalcopyrite bioleaching at different temperature, real-time polymerase chain reaction (PCR) was employed to analyze community dynamics of major bacteria applied in bioleaching. The results showed that yeast extract exerted great impact on microbial community, and therefore influencing bioleaching rate. To be specific, yeast extract was adverse to this bioleaching process at 30°C due to decreased proportion of important chemolithotrophs such as Acidithiobacillus ferrooxidans and Acidithiobacillus thiooxidans. However, yeast extract could promote bioleaching rate at 40°C on account of the increased number and enhanced work of Ferroplasma thermophilum, a kind of facultative bacteria. Similarly, bioleaching rate was enhanced under the effect of yeast extract at 50°C owing to the work of Acidianus brierleyi. At 60°C, bioleaching rate was close to 100% and temperature was the dominant factor determining bioleaching rate. Interestingly, the existence of yeast extract greatly enhanced the relative competitiveness of Ferroplasma thermophilum in this complex bioleaching microbial community.  相似文献

Domain 5 (D5) and domain 6 (D6) are adjacent folded hairpin substructures of self-splicing group II introns that appear to interact within the active ribozyme. Here we describe the effects of changing the length of the 3-nucleotide segment joining D5 to D6 [called J(56)3] on the splicing reactions of intron 5 gamma of the COXI gene of yeast mitochondrial DNA. Shortened variants J(56)0 and J(56)1 were defective in vitro for branching, and the second splicing step was performed inefficiently and inaccurately. The lengthened variant J(56)5 had a milder defect-splicing occurred at a reduced rate but with correct branching and a mostly accurate 3' splice junction choice. Yeast mitochondria were transformed with the J(56)5 allele, and the resulting yeast strain was respiration deficient because of ineffective aI5 gamma splicing. Respiration-competent revertants were recovered, and in one type a single joiner nucleotide was deleted while in the other type a nucleotide of D6 was deleted. Although these revertants still showed partial splicing blocks in vivo and in vitro, including a substantial defect in the second step of splicing, both spliced accurately in vivo. These results establish that a 3-nucleotide J(56) is optimal for this intron, especially for the accuracy of 3' splice junction selection, and indicate that D5 and D6 are probably not coaxially stacked.  相似文献

A human leukaemic cell line (REH) was subjected to various temperatures approximately greater than 42 degrees C for various time intervals; the cells were stained with a mixture of ethidium bromide and acridine orange, and red and green fluorescence were analysed by flow cytometry. Nontreated cells appeared as one cluster (V) in the biparameter histograms, but with time of heat treatment, two further discrete clusters (D1,D2) of cells appeared successively. They were distinguished by both the degree of red and green fluorescence. The kinetics of transit from one cluster to the other was dependent on temperature, the time lag between both steps becoming shorter with higher temperatures. It was shown previously that the same effect occurred during incubation with various cytostatic agents, and that only the D2 stage correlated with the stage of cell death monitored by the usual trypan blue exclusion test. Therefore the ethidium bromide technique seems to monitor an earlier stage of cell death. The decrease in the number of dye-excluding (V) cells during heat exposure occurred in two phases. After an initial decrease a plateau of number of dye-excluding cells was reached; the duration and level of this plateau depended on the temperature. The plateau was followed by a second phase where the remaining cells ceased to exclude the dye.  相似文献

Inoculum size has been found to affect significantly the maximum attainable specific growth rate during batch cultivation ofCandida utilis. Lower inoculum size resulted in an increased growth rate and relatively longer lag. The culture is found to be most active in the beginning of the exponential phase as regards its RNA synthesis rate. Batch data were used for predicting the conditions of the yeast population in single-stage continuous culture system. Predicted and the experimental values showed a reasonable agreement. In single-stage chemostat the physiology of the yeast was studied on the basis RNA, DNA and protein synthesis rates at various growth rates. The results indicate that the productivity of cells and the rate of synthesis of macromolecules is highest at the dilution rate values of 0.33 to 0.35 hr−1. In order to attain so-called unrestricted conditions of growth a pluristage pluristream continuous system was employed. It is assumed that under such conditions the specific growth rate and the synthetic activity of yeasts may reach its maximum on a given medium. The results presented do not show such conditions of growth under the experimental conditions employed (D 1=0.35 hr−1 andD 2=0.2 to 1.7 hr−1) withCandida utilis cultivated on beet molasses medium. Second stage of a two-stage two-stream continuous system is constantly fed with the cells from the foregoing stage; this category of cells on entering the new conditions of the second stage is expected to show some adaptation period. Continuous culture in a cascade of vessels with the addition of supplemental nutrients to any stage permits adjustment of the physiological state of the culture in each stage to best achieve a desired performance goal. The yeast Saccharomyces cerevisiae in two-stage continuous cultivation was selected as a model system. With conditions in the first stage held constant- at a selected glucose concentration in the feed stream, dilution rate for the second stage was varied. Cell numbers, dry weight, glucose concentration, respiration coefficient, and titers of several enzymes were determined. The seed rate was defined as the ratio of glucose concentration in the feeds to stage 1 and to stage 2. At low seed rates, the calculated specific growth rate in the second stage was proportional to dilution rate. At higher seed rates, the specific growth rate based on dry weight behaved differently from that based on cell numbers, and the dependence on dilution rate was not linear.  相似文献

The influence of temperature (17 and 31 degrees) on the maternal effect of mutation Puffed (Pu) in Drosophila hybrids has been studied. In the hybrids female D. littoralis +/+ x male D. virilis Pu/Pu, the stage of formation of black ring on anterior spiracles in the 3rd larval instar is sensitive to 31 degrees. In the hybrids female D. virilis Pu/Pu x male D. littoralis +/+, the expression of Pu gene manifests the maternal effect and as a result, two temperature-sensitive stages are found. The first one--onset of embryogenesis (2 to 4 hrs). At the temperature 17 degrees, the penetrance of Pu increases. The second stage is sensitive to 31 degrees and coincides with the period of black rings formation on anterior spiracles in the 3rd laval instar. It has been shown that at least two genetic systems take part in the formation of this feature. One group of genes controls the maternal effect and is sensitive to low temperature in the early embryogenesis of the hybrids female D. virilis x male D. littoralis. The second one--Pu gene and its modifiers--is active during the 2nd half of the 3rd larval instar and is heat-sensitive.  相似文献

Adult dietary yeast modulates mortality rate and reproduction of the Mediterranean fruit fly, Ceratatis capitata. In the medfly, a sugar-only diet leads to low mortality rates and reduced reproduction; addition of dietary yeast increases both mortality and egg laying. In Drosophila melanogaster low availability of dietary yeast is known to increase life span and reduce the rate of reproduction. Despite these similarities, because of differences in experimental design it remains unclear whether a common physiological mechanism modulates the effect of diet on survival. Here, we investigate how mortality rate and reproduction in D. melanogaster respond to the treatment regime used to study the medfly: no-yeast versus full diet. We find that adult medfly and D. melanogaster have opposite responses to the absence of yeast: D. melanogaster have high mortality when on no-yeast diet; when switched to full diet, D. melanogaster reduce mortality rates to the level presented by females continuously maintained on yeast. This reduction in mortality is accompanied by increased fecundity. These patterns are observed in all tested wildtype stocks, but flies made sterile by mutation in the gene oo18 RNA-binding protein (orb) lack this response. D. melanogaster, unlike medflies, appear to require adult dietary yeast to maintain maximal survival, and the capacity to assimilate yeast for somatic processes is one wildtype function of the gene orb.  相似文献

Glutathione reductase was extracted from toluene-treated baker's yeast cells by a two-stage buffer autolysis method. The yeast cells were treated with toluene for 1 h at 40 degrees C. After removal of the toluene, the cells were then allowed to autolysis in buffer for 72 h at 4 degrees C. The cells were collected and resuspended in buffer. A second stage autolysis was carried out for another 96 h at 4 degrees C. The enzyme was purified to 786-fold from the second stage cell autolysate by using two steps of affinity chromatography with triazine dyes (Yellow H-E4G and Yellow H-E6G) coupled to Sepharose CL-4B. By using this simplified method, 1.44 mg (165 units/mg) of glutathione reductase was obtained from 65 g (wet weight) of yeast cells, equivalent to 80% enzyme recovery.  相似文献

The effect of different yeast variants of the yeast Saccharomyces cerevisiae on the outcome of competition between Drosophila simulans and D. melanogaster was investigated. In all experiments differential birth rate was the effective mode of species competition. Addition of live yeast and especially mixtures of yeast strains improved the competition situation of a species, but could not prevent the extinction.  相似文献
