The ability of a natural enemy to tolerate starvation increases its chances to survive in the absence of food, what is an important factor for its success in storage grain environment. The objective of the present work was to assess the survival of Acarophenax lacunatus (Cross & Krantz) in the absence of food. The experiment used individualized physogastric females of A. lacunatus placed in petri dishes (5 cm diameter) and maintained at 20, 25, 28, 30 and 32 degrees C, 50+/-5 % R.H. and 24h scotophase. The number of live mites was recorded every 6h thus assessing the progeny survival without food at different temperatures. The mites died within 60h at the temperatures 30 degrees C and 32 degrees C, while they survived for up to 108h at 20, 25 and 28 degrees C. The mean lethal time for death was 58.6h for the lowest temperatures and 39.3h for the highest temperatures. Thus, A. lacunatus subjected to starvation lived longer under lower temperatures, what is probably due to its lower metabolism. In contrast, the mites survived for about 90h at 28 degrees C, temperature commonly observed in tropical and subtropical climates, what may favor their use as control agents of stored product insects in these regions.  相似文献

Action of adrenaline on ATPase activity of ground squirrel synaptosomes in vitro at 37 degrees and 17 degrees C was studied. It has been shown in experiments in vitro at 37 degrees C that adrenaline in a concentration of 5.10(-4) M influenced Mg and Na, K-ATPase of the synaptosomes in ground squirrel brain. The inhibition (42-72%) of Na, K-ATPase in the synaptosomes of the brain was seen during hibernation and in summer. The inhibition of Mg-ATPase (50%) was observed only in summer. The effect of adrenaline on the activity of Na, K-ATPase of synaptosome was seen in vitro as well as at 17 degrees (a 50% inhibition). It was shown that adrenaline in vitro at a concentration of 5.10(-4) M inhibited ATPases more than noradrenaline.  相似文献

Six Campylobacter jejuni and six Campylobacter coli strains were isolated from cows and pigs, and their survival in lake water was compared by viable counts. Campylobacter jejuni survived longer in culturable form than C. coli in untreated and membrane-filtered water both at 4 and 20 degrees C. This difference in survival time may be a reason why C. jejuni is generally isolated from surface waters more frequently than C. coli. Both species survived better in filtered than in untreated water. This suggests that predation and competition for nutrients affect the survival of both Campylobacter species in the aquatic environment.  相似文献

Ethyl methanesulfonate-treated autosomes were screened for the presence of dominant cold-sensitive (DCS) lethal mutations in Drosophila melanogaster. None was found among 6,552 treated and 168 untreated third chromosomes. Twenty-three DCS-L chromosomes which caused death at 17 degrees C but survived at 22 degrees C and 29 degrees C were recovered from 5,046 mutagenized chromosome 2's.-The DCS-L mutations all mapped around dp and appeared to be functionally allelic. Lethality of heterozygotes for most of the DCS-L's occurred over a prolonged interval from the embryonic through the larval instars. Prolonged incubation at 17 degrees C did not demonstrate any maternal effect on zygotic survival.  相似文献

Summary The feral pigeon in New York City was found to serve as a mechanical carrier of pathogenic strains ofCryptococcus neoformans. Of 94 feral pigeons freshly trapped in the city, 7 were found to carryC. neoformans on their beaks and feet, while their rectal swabs were negative. Following crop instillation ofC. neoformans in 3 feral pigeons, the fungus survived passage through the gastrointestinal tract and appeared in the fresh feces within one hour after inoculation and was still present 24 hours later. The internal body (rectal) temperatures of 57 feral pigeons recorded soon after capture in two seasons of the year, ranged from 41.5° C to 43.3° C and averaged 42.5° ± 0.39° C. The birds were able to maintain their high temperatures in the face of sustained cold, indicating the presence of a strong thermoregulating mechanism. A study of the growth and survival of 60 human strains ofC. neoformans on Sabouraud dextrose yeast extract agar slants revealed that 100 % of the cultures were able to grow at 39° C, 92 % at 40° C, 30 % at 41° C, 17 % at 42° C and 8 % at 43° C. Despite exposure to these temperatures for 7 days, 100 % of the human strains survived at 40° C, 97 % at 41° C, 95 % at 42° C, 91 % at 43° C, 88 % at 44° C, 47 % at 45° C and none at 47° C. Seventy strains ofC. neoformans of pigeon excreta origin showed a similar pattern of heat resistance, except that 11 % of these strains survived 47° C. The inability ofC. neoformans to grow at 44° C, a property shared by all 130 strains, provides a new species characteristic.C. neoformans was found to multiply rapidly in moist pigeon excreta extract, reaching counts up to 60 million per ml within 3 weeks, and to be still viable in moist as well as dessicated pigeon excreta extract after more than two years of storage at room temperature.Presented in part at the 26th VA-Armed Forces Pulmonary Disease Research Conference, Cleveland, Ohio, Jan. 23–26, 1967, and the 67th Annual Meeting American Society for Microbiology, New York, April 30–May 4, 1967. Cerebral cortical slices from rat brain were incubated at 37 degrees C for 2 h in the presence of isoproterenol, noradrenaline, or adrenaline, and binding affinities and densities of adrenoceptor subtypes were subsequently examined in homogenized tissue. The density of alpha 2- and total beta-adrenoceptors was estimated using the radioligands [3H]rauwolscine and [3H]dihydroalprenolol (DHA), respectively. The percentages of beta 1- and beta 2-adrenoceptors were defined by inhibiting the binding of [3H]DHA with the beta 1-selective antagonist metoprolol. Exposure of slices to noradrenaline and adrenaline significantly decreased the maximal number of binding sites (Bmax) of alpha 2-adrenoceptors (48 and 37% respectively) without significantly affecting affinity; isoproterenol had no effect. Exposure to isoproterenol, noradrenaline, and adrenaline significantly decreased the Bmax of beta-adrenoceptors (by 60, 34, and 24%, respectively) but did not affect the affinity. Isoproterenol and adrenaline significantly decreased the density of beta 1-adrenoceptors by 75 and 24% and beta 2-adrenoceptors by 23 and 28%, respectively. Noradrenaline significantly decreased the density of beta 1-adrenoceptors by 42% without affecting the number of beta 2-adrenoceptors. These findings indicate that subtypes of adrenoceptors in rat cerebral cortex are differentially regulated by adrenergic agonists.  相似文献

In 5-6-week-old cockerels the circulating corticosterone concentration was significantly increased in birds i.m. injected 30 and 60 min previously with adrenaline (0.33 mg/kg), noradrenaline (0.33 mg/kg) or a beta-adrenergic agonist (isoprotorenol, 1 mg/kg), but was reduced in birds pretreated with an alpha-adrenergic agonist (phenylephrine, 1 mg/kg). The stimulation of corticosterone secretion induced by a 30 min period of forced exercise (0.04 km/hr; 0 degree incline) was potentiated by noradrenaline, isoprotorenol and phentolamine pretreatment. In response to exogenous adrenocorticotrophin (ACTH, 8 i.u./kg), administered i.v., the increase in the plasma corticosterone concentration was elevated above that in the controls in birds pretreated with adrenaline, noradrenaline, isoprotorenol or phentolamine (an alpha antagonist administered at 1 mg/kg). The corticosterone response to ACTH was suppressed by phenylephrine pretreatment. These results demonstrate that both basal and stimulated levels of adrenocortical activity may be subtly regulated by adrenergic mechanisms acting at a site(s) within the hypothalamo-pituitary-adrenal axis.  相似文献

Spores of Encephalitozoon cuniculi were exposed to various temperature or to disinfectants, and their infectivity was then tested on monolayer cultures of canine kidney cells. The maximum survival time for spores suspended in medium 199 was 1 day at -20 degrees C, 98 days at 4 degrees C, 6 days at 22 degrees C, and 2 days at 37 degrees C. Only 2.5% survived 30 min at 56 degrees C. Boiling for 5 min or autoclaving at 120 degrees C for 10 min killed all spores. Dry spores survived less than a week at 4 degrees C but at least 4 weeks at 22 degrees C. Exposure for 30 min to recommended working concentrations of 9 of the 11 disinfectants tested killed all spores. The growth-inhibition effect of 7 antibiotics and chemotherapeutics was studied on canine kidney cell culture inoculated with E. cuniculi. None could completely inhibit growth. The most effective was chloroquine phosphate which, at a concentration of 12.5 mg per 1000 ml culture medium and during a test period of 8 weeks, reduced the harvest of E. cuniculi to 31% of that from inoculated, untreated cultures.  相似文献

Survival of the cystophorous cercariae of Halipegus occidualis (Trematoda: Hemiuridae) from naturally infected Helisoma anceps (Gastropoda: Planorbidae) was determined following constant temperature storage at 4, 16, 22, 26 and 30 degrees C. At 4 degrees C, 90% of cercariae survived for 14 weeks and 50% survived for 30 weeks. Survival varied inversely with temperature, and at 30 degrees, 90% survived for 1.5 weeks and 50% survived for 2.5 weeks. At the time of release from snails, cercariae excysted within 0.5 s of stimulation. Excystment times ultimately increased to 1-3 s at all temperatures as length of storage increased, but no connection was detected between excystment time and the activity of the cercaria. Excystment times began increasing while survival was still above 90%. Temperature specific mortality rates of cercariae were estimated, and used to simulate the survival of cercariae released naturally at various times of the year. The results of these simulations suggest that cercariae of H. occidualis are present in the environment at all times of the year, but that cercariae present in late winter (January to mid-March) may have an impaired excystment mechanism that reduces their infectivity.  相似文献

The purpose of this study was to investigate the effect of cool exposure on lipolytic response during prolonged intermediate-intensity exercise in humans. Eight male subjects participated in this study; they performed 120-min cycle ergometer exercise at 60% of maximal oxygen uptake (VO2max) in a climatic chamber at 10 degrees C (C) and 30 degrees C (WH). There were no significant differences in oxygen uptake and respiratory exchange ratio between the two conditions during the prolonged exercise. Significant influences of cool exposure were observed in the changes in both heart rate and rectal temperature (p<0.01). Although cool exposure had no significant effects on plasma triglyceride, free fatty acid, and glycerol levels, changes in adrenaline and noradrenaline levels at C were significantly lower than WH during the prolonged exercise (p<0.01). Changes in the ratio of glycerol to noradrenaline (Gly/Nad), as an index of lipolytic efficiency, were significantly high at C as compared with WH (p<0.01). These results suggest that cool exposure has an influence on lipid metabolism during prolonged intermediate-intensity exercise, from the viewpoint of efficiency in lipolysis.  相似文献

This paper reports the cryopreservation of an immortalized human endothelial cell line (ECV304), either as a single cell suspension or as a confluent layer on microcarrier beads. Cell suspensions were exposed to 10% w/w dimethyl sulfoxide in a high-potassium solution (CPTes) at 0 degrees C. The cells were then cooled to -60 degrees C at controlled rates between 0.3 and 500 degrees C/min and stored below -180 degrees C. Samples were thawed in a 37 degrees C water bath and the cryoprotectant was removed by serial dilution at 22 degrees C over 6 min. The recovery of cell suspensions was assayed by culturing aliquots in 24-well plates for 7-9 days and counting the number of colonies that contained >25 cells. Maximum survival was 45-50% at cooling rates of 0.3, 1.0, and 10 degrees C/min, but decreased to 20% at 50 degrees C/min and to <1% at 500 degrees C/min. Biosilon microcarrier beads were used for the attached cells. Confluent beads were cryopreserved by exactly the same technique and cell function was assayed by measuring active amino acid (leucine) transport at 37 degrees C. Control, untreated confluent beads gave approximately 73% of control uptake and negative controls (frozen without cryoprotectant) gave approximately 4% uptake. The cells attached to beads showed percentage uptakes that were numerically similar to the survival of cells in suspension at cooling rates between 10 and 500 degrees C/min, but at lower cooling rates the recovery of attached cells increased to 70% at 1 degrees C/min and to 85% at 0.3 degrees C/min. These results indicate a marked difference in the effect of cooling rate on ECV304 cells depending upon attachment.  相似文献

The influence of antibiotic decontamination of Pseudomonas contamination of the GI tract prior to whole-body neutron or gamma irradiation was studied. It was observed that for fission neutron doses greater than 5.5 Gy, cyclotron-produced neutron doses greater than 6.7 Gy, and 137Cs gamma-ray doses greater than 14.4 Gy, the median survival time of untreated rats was relatively constant at 4.2 to 4.5 days, indicating death was due to intestinal injury. Within the dose range of 3.5 to 5.5 Gy of fission neutrons, 4.9 to 6.7 Gy of cyclotron-produced neutrons, and 9.6 to 14.4 Gy of gamma rays, median survival time of these animals was inversely related to dose and varied from 12 to 4.6 days. This change in survival time with dose reflects a transition in the mechanisms of acute radiation death from pure hematopoietic, to a combination of intestinal and hematopoietic, to pure intestinal death. Decontamination of the GI tract with antibiotics prior to irradiation increased median survival time 1 to 5 days in this transitional dose range. Contamination of the intestinal flora with Pseudomonas aeruginosa prior to irradiation reduced median survival time 1 to 5 days in the same radiation dose range. Pseudomonas-contaminated animals irradiated within this transitional dose range had maximum concentrations of total bacteria and Pseudomonas in their livers at the time of death. However, liver bacteria concentration was usually higher in gamma-irradiated animals, due to a smaller contribution of hematopoietic injury in neutron-irradiated animals. The effects of both decontamination of the GI tract and Pseudomonas contamination of the GI tract were negligible in the range of doses in which median survival time was dose independent, i.e., in the pure "intestinal death" dose range. Finally, despite the marked changes in survival time produced by decontamination or Pseudomonas contamination in the "transitional dose range," these treatments had little effect on ultimate survival after irradiation as measured by the LD50/5 day and the LD50/30 day end points. The implications of these results with respect to treatment of acute radiation injury after whole-body irradiation are discussed.  相似文献

The effect of sucrose and trehalose on the viability of one- and two-cell rabbit embryos was investigated. A significant decrease in the viability of one- and two-cell embryos exposed for 30 min. at 20 degrees C was observed. At 38 degrees C none of the two-cell embryos in a sucrose solution survived after 30 min exposure, while approximately 50% of the embryos survived in a trehalose solution. The cleveage rate in culture of two-cell embryos exposed both to 2.0 M or 1.45 M trehalose was significantly lower in comparison with the control group. However the survival rate after transfer of two-cell embryos exposed to 1.45 M trehalose solution at 20 degrees C remained the same as that of the control group.  相似文献

Temperature strongly influenced percent mortality and time to death of salamanders exposed to the Ambystoma tigrinum virus (iridovirus) (ATV). Most salamanders survived when exposed at 26 degrees C, whereas all died at 18 degrees C and nearly all died at 10 degrees C. Some asymptomatic salamanders that survived 60 d at 10 or 26 degrees C were found to be carrying virus. Polymerase chain reaction (PCR) confirmed the presence of virus in ATV-exposed salamanders but was found to be less sensitive than cell culture in detecting ATV at low concentrations. PCR products were 100% identical to ATV in the major capsid protein sequence. Virus titer was higher in salamanders held at 10 degrees C than at 18 degrees C but little virus, if any, was present in the small number of salamanders that died at 26 degrees C. These results may help explain periodic viral epizootics in field populations of A. tigrinum where water temperatures fluctuate widely.  相似文献

The upper limiting temperature of growth of Staphylococcus aureus MF31 in heart infusion broth (HI) was about 44 degrees C but addition of monosodium glutamate (MSG) and soy sauce permitted the organism to grow above this temperature. This effect is similar to that of NaCl. Tomato ketchup, Worcestershire and HP sauces added to HI did not allow growth at the non-permissive temperature of 46 degrees C but death was delayed. Staphylococcus aureus died in unsupplemented chicken meat slurry at 46 degrees C but grew at 48 degrees C in slurry supplemented with 5.8% NaCl and survived incubation for 18 h at 50 degrees C in slurry supplemented with 5.8% NaCl and 5% MSG. Cultures grown at 37 degrees C had a D60 value of 2 min in 50 mmol/l Tris (pH 7.2) buffer. Cultures grown at 46 degrees C in HI containing 5.8% NaCl had a D60 value of 8 min in Tris buffer. Addition of 5.8% NaCl plus 5% MSG to the buffer increased the D60 by a factor of about 7 for both cultures. In storage experiments at room temperature, the culture grown at 37 degrees C and at 46 degrees C plus 5.8% NaCl died at about the same rate in salami. In milk powder, however, the count of 37 degrees C culture decreased from 10% g to 10(6)/g in 5 weeks while the count of 46 degrees C culture remained unchanged. In cottage cheese, freeze-dried rice and macaroni, the 37 degrees C cultures also died more rapidly. It is suggested that cultures grown at 46 degrees C plus 5.8% NaCl may be suitable for experiments with artificially contaminated foods.  相似文献

Basal metabolic rate (MR), body temperature (T(b)) and wet thermal conductance (C(wet)) of three tropical species of fruit-doves were investigated at ambient temperatures (T(a)) of 11-33 degrees C in activity (alpha) and rest (rho) phases to investigate the possible effect of obligate frugivory on the physiology of columbids. The basal metabolic rates of Ptilinopus melanospila (black-naped fruit-dove, 94 g), Drepanoptila holosericea (cloven-feathered dove, 198 g) and Ducula pinon (Pinon's imperial pigeon, 748 g) are 20-38% lower than predicted for all birds, including granivorous columbid species from temperate and tropical regions. The MR was minimal at a T(a) value of approximately 30 degrees C (=lower critical temperature, T(lc)) for all three species, indicating that these rainforest birds are not able to withstand high ambient temperatures as well as arid-adapted members of the pigeon family. Minimal wet-thermal conductance was, on average, higher than expected, indicating poor insulation in these tropical birds. Body temperatures were as expected; however, below T(lc) the body temperatures decreased to levels of 35-36 degrees C (T(a)=12 degrees C).  相似文献

The emerald ash borer, Agrilus planipennis Fairmaire (Coleoptera: Buprestidae), a phloem-feeding insect from Asia, was identified in 2002 as the cause of widespread ash (Fraxinus sp.) mortality in southeastern Michigan and Essex County, Ontario. Most larvae overwinter as nonfeeding prepupae in the outer sapwood or thick bark of large trees. In a series of studies, we evaluated effects of grinding, chipping, and heat treatment on survival of A. planipennis prepupae in ash material. Heavily infested ash bolts containing roughly 8,700 prepupae were processed by a horizontal grinder with either a 2.5- or 10-cm screen. There was no evidence of A. planipennis survival in chips processed with the 2.5-cm screen, but eight viable prepupae were recovered from chips processed with the 10-cm screen. We chiseled additional sentinel chips with prepupae from ash logs and buried 45 in each chip pile. In total, six prepupae in sentinel chips survived the winter, but we found no sign of adult A. planipennis emergence from the processed chips. Subsequently, we assessed prepupal survival in chips processed by a chipper or a horizontal grinder fit with 5-, 10-, or 12.7-cm screens. An estimated 1,565 A. planipennis prepupae were processed by each treatment. Chips from the chipper were shorter than chips from the grinder regardless of the screen size used. No live prepupae were found in chips produced by the chipper, but 21 viable prepupae were found in chips from the grinder. Infested wood and bark chips chiseled from logs were held in ovens at 25, 40, or 60 degrees C for 8, 24, or 48 h. Prepupal survival was consistently higher in wood chips than bark chips at 40 degrees C, whereas no prepupae survived exposure to 60 degrees C for eight or more hours. In a second study, prepupae in wood chips were exposed to 40, 45, 50, 55, or 60 degrees C for 20 or 120 min. Some prepupae survived 20 min of exposure to all temperatures. No prepupae survived exposure to 60 degrees C for 120 min, but 17% survived exposure to 55 degrees C for 120 min, suggesting that some fraction of the population may survive internationally recognized phytosanitary standards (ISPM-15) for treatment of wood packing material.  相似文献

Staphylococcus aureus isolated from milk and grown in milk was heated in milk. The phenomena of death as well as injury was investigated in the range of 50 to 75 degrees C. The D60 value (decimal reduction time on salt-free medium) was 0.87 min, the D'60 value (decimal reduction time in salt-containing medium) was 0.62 min. Cultures were injured as soon as heating started. This initial thermal shock increased with increasing temperature. At 50-60 degrees C injury was more rapid than death. At greater than 60 degrees C death became faster than injury and the two processes coincided at 70 degrees C. The Z value was 9.46 degrees C and the Z' value was 9.93 degrees C.  相似文献

1. Blood flow and the concentrations of noradrenaline, adrenaline and dopamine were determined in the skins of cattle and goats, before, at the onset of and 3 hr after commencement of sweating induced by heat exposure (40 degrees C). 2. The onset of sweating in both cattle and goats was associated with a rise in cutaneous blood flow, which was thus independent of sweat pattern. Cutaneous blood flow was also higher at 40 degrees C than at 15 degrees C. 3. The predominant catecholamine in the skin of both species was dopamine, which in the goat increased in concentration in the warm environment. 4. There was no clear evidence of a change in the amount of any of the cutaneous catecholamines during exposure to 40 degrees C, although there was a consistent tendency for the concentrations of adrenaline in the calf and noradrenaline in the goat, to fall during the onset of sweating.  相似文献

Summary Enkephalin (ENK)- and neurotensin (NT)-immunoreactivities (IR) were localized in cat adrenal medulla using immunocytochemistry. ENK was localized mainly in adrenaline cells, 70%–80% of which were heavily labelled. About 5%–10% of the noradrenaline cells were ENK-immunoreactive (IR). A dense network of ENK-IR nerves was localized among adrenaline cells. NT was localized only in the noradrenaline cells, 60%–70% of which were immunoreactive. NT- and ENK-IR were localized in separate noradrenaline cells. A dense network of NT-IR nerves was restricted among noradrenaline cells.The present findings confirm that ENK- and NT-like peptides are localized in separate adrenal medullary cells and demonstrate that also the preganglionic nerves are divided in subpopulations according to their neuropeptide content. These nerves have a selective localization among adrenaline and noradrenaline cells and may have physiological significance in the control of secretion of catecholamines and neuropeptides from adrenal medulla.  相似文献
