The upstream region of the metA gene in Escherichia coli contains two promoters. We have identified by lacZ fusion an additional promoter in this region, and showed that it is transcribed in the opposite orientation from the metA gene. The putative translation product corresponds to a peptide of 147 amino acids – ORF19 by molecular mass. This peptide is probably not essential for growth, as an insertion mutant is viable.  相似文献

We report the nucleotide sequence of the human chromosomal gene which encodes the interleukin-1 beta protein (IL-1 beta). The gene spans a region of 7.5 kb and the coding part is divided into seven exons. Comparison with the homologous mouse gene reveals that the structural organization is conserved through evolution. In addition to this, human and murine IL-1 beta genes show extensive sequence homology within the intervening sequences.  相似文献

MCM3 acetylase (MCM3AP) and germinal-centre associated nuclear protein (GANP) are transcribed from the same locus and are therefore confused in databases because the MCM3 acetylase DNA sequence is contained entirely within the much larger GANP sequence and the entire MCM3AP sequence is identical to the carboxy terminus of GANP. Thus, the MCM3AP and GANP genes are read in the same reading frame and MCM3AP is an N-terminally truncated region of GANP. However, we show here that MCM3AP and GANP are different proteins, occupying different locations in the cell and transcribed from different promoters. Intriguingly, a promoter for MCM3AP lies within an intron of GANP. This report is an interesting example in nature of two separate gene products from the same locus that perform two entirely different functions in the cell. Therefore, to avoid further confusion, they should now be referred to as separate but overlapping genes.  相似文献

Elevation of cAMP has been considered to be an important downregulative signal in the production of interleukin-1(IL-1). This study demonstrates that this phenomenon is dependent on the signal used to activate the IL-1 production. The IL-1 beta production of lipopolysaccharide activated human monocytes was readily inhibited by dibutyryl cAMP. This took place without a significant change in the steady-state levels of IL-1 beta mRNA. By contrast, in PMA activated monocytes 100 microM dibutyryl cAMP increased in IL-1 beta production ca. 4-fold. The steady-state levels of IL-1 beta mRNA were also simultaneously increased.  相似文献

IL-1 was localized within the cytoplasm of human blood monocytes by indirect immunofluorescence using polyclonal rabbit antibodies. After LPS stimulation first IL-1-positive cells appeared at 2 hours and maximal intracellular IL-1 concentration was observed at 10-24 hours when nearly 90% monocytes were labeled with subsequent decline at 48 hours. The highest intracellular IL-1 content preceded its maximal level in cell supernatants.  相似文献
