Four experiments were done to determine: (1) the effectiveness of early detection and treatment of vesiculitis in bulls; (2) whether antibiotic treatment at recommended dosages will result in adequate vesicular gland tissue concentrations of antibiotics to prevent in vitro bacterial growth; (3) whether intraglandular injection of antibiotics can be a successful alternative to systemic antibiotic treatment; and (4) the effectiveness of tilmicosin versus tulathromycin for treatment of clinical vesiculitis. In Experiment 1, there was a high rate of spontaneous remission from vesiculitis detected at 9-12 mo of age. Furthermore, there was no advantage for early antibiotic treatment versus no treatment for bulls of this age. In Experiment 2, bacteria on agar plates were exposed to fluid extracted from vesicular gland biopsies after antibiotic treatment of normal, healthy bulls. Although inadequate concentrations of antibiotics were achieved to inhibit bacterial growth when recommended dosages of various antibiotics were administered, doubling the antibiotic dosage increased in vitro bacterial growth inhibition. In Experiment 3, relatively nonirritating antibiotics were injected directly into the glands of bulls with clinical vesiculitis, demonstrating that intraglandular injections of antibiotic could be used as a successful alternative to systemic antibiotic treatment. Experiment 4 was a clinical field trial to compare the efficacy of tilmicosin versus tulathromide at recommended dosages for the treatment of clinical vesiculitis. Although the results favored tulathromycin, both antibiotics resulted in clinical cures of vesiculitis.  相似文献

The objective of this study was to determine whether early detection and treatment with tilmicosin would reduce the number of yearling bulls culled due to vesicular gland disease at the time of their first semen test. Bulls (n=2207) of 15 breeds at 17 performance test stations were examined by transrectal palpation at 9-12 months of age and assigned to three treatment groups: (1) positive treated group, receiving subcutaneous injections of tilmicosin every second day for three treatments (2) positive untreated control group, (3) negative untreated control group. Transrectal palpation of the glands was done again at a pre-sale semen test 28-70 d (mean=42.8 d) after the first examination. Semen was evaluated for the presence of pus and/or leukocytes by light microscopy. The proportion of bulls with vesiculitis was 4.4% (97/2207). At the pre-sale semen test the number of bulls with vesiculitis had decreased to 1.3% (29/2207); however, seven of these were new cases that developed after the first examination. Therefore, there was a recovery rate of 75/97 (77.3%) in the original group of bulls positive for vesiculitis. At the pre-sale test, there was no difference in the proportion of bulls with enlarged glands in the positive treated group (15/66) versus the positive untreated group (7/31) and no difference in the proportion of bulls with leukocytes or pus in their semen samples. The results of this experiment did not indicate any advantage in detection and treatment of vesiculitis before a year on age due to a high rate of spontaneous recovery.  相似文献

Crossbred (Bos taurus) yearling beef bulls were assessed for breeding soundness and physical traits prior to multi-sire natural mating at pasture. Bulls (n = 60) were assigned to six groups of nine or 10 bulls and two bull-groups were rotated on 14-day intervals during a 63-day mating season in each breeding herd (n = 3) of 191-196 cows. The remaining bulls (n = 14) were maintained under similar environmental conditions without mating exposure. Bulls were observed during mating and assessed for breeding soundness and changes following mating. Bulls used for breeding (UFB) lost 77 kg of body weight and declined from body condition scores of 6 to 4.5, whereas bulls not used for breeding (NUB) maintained body condition scores of 6 and gained 27 kg. The UFB bulls incurred a 75% total injury rate with 63% incidence of lameness and 12% incidence of reproductive injuries, resulting in a 22% attrition rate. Only 45% were physically sound at the end of mating. Scrotal circumference declined in UFB bulls (-4.58%) and increased in NUB bulls (2.49%). From the 98% BSE-satisfactory rate (UFB) prior to breeding, only 61% were BSE-satisfactory post-breeding. The NUB bulls declined from 57 to 36% satisfactory. The BSE classification was influenced by significant increases in abnormal spermatozoa (primary and secondary), which was significantly associated with injuries incurred during mating. Group and breed differences in injury rates and BSE-status following mating were evident. Environmental conditions and mating activity influenced bull seminal quality and physical condition. Pregnancy rates in all three breeding herds (91-96%) were similar, with insignificant differences between bull-groups; the effects of physical and reproductive changes on individual bull fertility were immeasurable.  相似文献

A total of 92 range beef bulls (Hereford = 60; Angus = 32) were given a breeding soundness examination (BSE) and two assessments for sex drive prior to their use in 23 breeding trials employing estrous synchronized females. Bulls were in three age groups: yearlings (n=29), two year olds (n=36), and three year olds and older (n=27). All yearling bulls were virgins, but the majority of the older bulls had previous mating experience. Angus bulls were superior (P<0.01) to Herefords in spermatozoal morphology and BSE score. Scrotal circumference increased with age beyond two years in Angus bulls but not in Herefords. Spermatozoal abnormalities generally decreased with age. BSE scores did not differ significantly among age groups. Apart from number of mounts, measures of sex drive did not differ with age or breed of bulls. This represents qualified justification for the current practice of using the same sex-drive assessment procedures for Bos taurus bulls of various ages and breeds.  相似文献

Objectives in this study were to determine the accuracy of different methods of evaluating libido score (L), service rate (SR) and reaction time to service (RTS) in yearling Bos taurus bulls. Using restrained, non-estrus females, 26 yearling Bos taurus bulls were evaluated a total of eight times (four sessions, two tests per session) over 2 consecutive months for L, SR, and RTS. Individual bull variation influenced both L (P<0.0001) and SR (P<0.003). Repeatability was moderate for L (R=0.64) and low for both SR (R=0.12) and RTS (R=0.04). Under the conditions of this study and characteristics of these tests, variance was reduced to 69-73% for L and 26-23% for SR with four to eight repetitions, respectively. Bulls that scored highly in the first session, however, tended to score highly throughout. Although, three bulls did not serve in any test, RTS was independent of effects. However, the tendency of RTS to decrease, and for L and SR to both increase with consecutive tests, suggests influences other than genetic, such as learning and/or environmental factors. These tendencies were most evident in bulls which obtained low L scores at the first session. It was concluded that, despite the important degree of individual variability observed in L and SR, accurate quantitative evaluation of the sex-drive in young Bos taurus beef bulls would benefit from use of test procedures of greater repeatability.  相似文献

Two experiments were conducted in successive years on a Kansas ranch to determine 1) if serving capacity could be evaluated in virgin yearling beef bulls under field conditions, 2) the influence of sire on serving capacity, and 3) the effect of sexual experience on subsequent serving capacity for low serving capacity bulls. In Experiments 1 and 2, 70 and 78 Angus bulls 13 to 15 mo old representing four and six different sires, respectively, were used. In both experiments, a pretest session that measured reaction time to service preceded two successive serving capacity tests. Following the first serving capacity test, in Experiment 2, all low serving capacity bulls (n = 23) underwent sexual experience school, which consisted of exposure to nonrestrained estrus-induced females over a 4-d period. Five days after sexual experience school (9 d after the first serving capacity test), all Experiment 2 bulls were retested. In both experiments, sire influenced mean serving capacity scores of sons (P < 0.01, Experiment 1; P < 0.05, Experiment 2). Characteristics measured, including age, weight, scrotal circumference, and reaction time, were all unrelated to bulls serving capacity in both experiments, as well as to breeding soudness examination scores for bulls in Experiment 1. There was a dramatic impact of sexual experience school on subsequent serving capacity, since 85% of sexual experience school bulls moved into the medium or high serving capacity category at the second serving capacity test. It is concluded that serving capacity can successfully be evaluated in yearling beef bulls under field conditions, that sire has a profound impact on bulls serving capacity, and that providing sexual experience to low serving capacity bulls can improve their serving capacity.  相似文献

The relationships between natural service fertility of beef bulls and the components of breeding soundness evaluation, age, preweaning average daily gain, yearling weight and scores of two libido tests were studied in two seedstock herds. In one of the herds (YBH), 15 yearling bulls were rotated during six-week breeding season so that each group of five bulls served the cows for a period of one week and rested for two weeks before the next exposure. Five older bulls (one 3-year old and four 2-year old) were assigned to the other herd (MBH) during the entire breeding season (six weeks). The progeny of each bull were identified by blood typing.Calf crop was 6.4% higher (P=0.14) in the MBH compared with that in the YBH. There were large differences in fertility between the bulls in the MBH. The 3-year old bull sired 40.9% of the calves, presumably due to his high social dominance order. The third group of yearling bulls, which served the herd during the third and sixth weeks of breeding, sired the highest number of calves (49.4%) compared with the other two groups (28.4% and 22.2%). In each of the groups one and two of the yearling bulls, one bull did not produce any progeny, while another one sired 44% of the calves. The correlation coefficients between fertility of the bulls and the traits used to predict their fertilizing capacity were generally small and inconsistent in different groups, when the effect of age in the mixed age group was removed. The correlation coefficients of bull fertility with scrotal circumference and reproductive system score were higher and more consistent in different groups as compared with the other traits studied.  相似文献

Yearling, grass-fed, beef bulls at the USDA Subtropical Agricultural Research Station, Brooksville, Florida, were assessed for physical and semen traits in January, April, July and October of 1991 (Trial 1) and 1992 (Trial 2). Bulls were given a breeding soundness evaluation (BSE) using revised semen and scrotal circumference (SC) criteria. In Trial 1, the bulls consisted of Angus (n = 15), Brahman (n = 14), Hereford (n = 15) and Senepol (n = 14). In Trial 2, the breeds were Angus (n = 15), Brahman (n = 16), Romosinuano (n = 13) and Nellore x Brahman (n = 9). Trial bulls generally showed delayed growth compared with grain-fed bulls in temperate environments. Breed influenced semen traits (percentage sperm motility, normal spermatozoa and those with primary abnormalities) in both trials. Temperate Bos taurus breeds (Angus, Hereford) were generally superior to Bos indicus breeds (Brahman, Nellore x Brahman). Tropically-adapted Bos taurus breeds (Senepol, Romosinuano) were intermediate for those traits tested. In general, tropically-adapted Bos taurus breeds were more similar in reproductive development to temperate Bos taurus than to Bos indicus breeds. Breed by test period interactions occurred and were mainly influenced by delayed sexual maturity of Bos indicus bulls. Qualitative semen traits increased with bull age, particularly from 12 to 18 mo. Scrotal circumference development was slower in the Bos indicus breeds. Bulls of satisfactory BSE status at 18.1 to 22 mo of age were 73.9% in Trial 1 and 58.5% in Trial 2. Brahman bulls had the least satisfactory BSE scores in both years (Trial 1, 44.4%; Trial 2, 22.2%). Most bulls failed to achieve satisfactory BSE status due to a small SC relative to age (Trial 1, 66%; Trial 2, 72%). The most efficacious use of the BSE was > or = 15 mo in Bos taurus bulls and > 18 mo for Bos indicus bulls. Although the BSE has proven to be useful for the assessment of young, pasture-raised bulls in semi-tropical environments, use of SC thresholds linked more with growth traits than with calendar age would improve comparisons of relative reproductive development in such bulls, particularly those of Bos indicus derivation.  相似文献

Three methods of assessing sex-drive were compared in 113 yearling beef bulls. These were the serving capacity score (SC), the libido score (L) and reaction time to first service (R). Ovariectomized heifers restrained in service crates were the stimulus for all tests. Bulls were assessed twice by each method. For the first libido and serving capacity tests (L1 and SC1), the heifers were induced to show estrus. For the second tests (L2 and SC2), the heifers were not induced to show estrus. On a non test day, single blood samples were taken from all bulls and assayed for LH and testosterone. Reaction times to first service (R1 and R2) in the two serving capacity tests were not significantly correlated. Although the numbers of services (SC1 and SC2) in both serving capacity tests, were significantly correlated (r = .67), 57% of the bulls did not achieve a service in both the tests with heifers in heat and with heifers not in heat. Libido scores between the test with heifers in heat and heifers not in heat were significantly correlated (r = .67). The libido score method had the advantage that more bulls received a positive score and the test duration was shorter than in the serving capacity test. Of the three scoring procedures compared, libido score appeared to have most advantage in assessing sex-drive in yearling beef bulls. The total number of services achieved in the first serving capacity tests (using 'estrus' heifers) did not differ from that achieved in the second tests (using non-estrus heifers). The total services achieved in 10-min compartments of the 30-min serving capacity tests showed no difference either within or between tests. It is concluded that a 10-min test provides as much comparative information on the sex-drive of yearling beef bulls as longer tests. Further, the use of females in estrus appears unnecessary to satisfactorily assess bull sex-drive, provided proper restraint and presentation of stimulus females is employed. LH and testosterone values were not significantly correlated or were poorly correlated with sex-drive measurements.  相似文献

Ninety-three beef bulls and 2316 females were used to determine the relationships between breeding assessments of bulls and subsequent mating performance. Each bull was given a breeding soundness examination (BSE) and two 10-min libido/serving capacity (L/SC) tests. Breeding potential of each bull was classified as satisfactory (BSE score = 60 to 100) or questionable (BSE score = 30 to 59); libido was classified as either high (mean score = 9.0 to 10) or medium (mean score = 6.0 to 8.5). Bulls were then joined, single-sire, with groups of females which had received one of two treatments to synchronize estrus. Bull-to-female ratios ranged from 1:7 to 1:51. Continuous observations were conducted on the mating activity of each group. One bull was removed from the experiment due to a severe breeding problem. Bulls of satisfactory breeding potential (n = 80) did not differ (P > 0.10) from bulls of questionable breeding potential (n = 12) in measurements of mating activity. However, by the end of the synchronized breeding period, bulls classified as satisfactory breeders achieved approximately a 9% higher (P < 0.10) pregnancy rate than did bulls of questionable breeding status (45.6 +/- 2.1% vx 36.5 +/- 5.3%). Bulls with a high libido (n = 69) serviced more (P < 0.01) estrous females (81.3 +/- 3.1% vs 63.5 +/- 4.2%) than bulls with a medium libido (n = 23). However, pregnancy rates achieved by bulls of either libido classification did not differ significantly. Individual components of the BSE as well as mean libido score were poorly correlated with pregnancy rates (r = -0.22 to 0.18). It was concluded that classification of bulls by mean libido score can aid in identifying groups of bulls that service more estrus synchronized females, whereas classification by BSE score aids in identifying groups of bulls that impregnate more females.  相似文献

Young (9 to 10 mo) Aberdeen-Angus bulls (n = 5) in excellent body condition and pastured with approximately 35 other bulls of similar age on a farm in southeastern Alabama had fever, harsh pulmonary sounds, increased respiratory rate and variable amounts of hindlimb and scrotal edema. Bulls had mild microcytic, normochromic anemia. Numerous eperythrozoon organisms were identified on blood smears. Indirect hemagglutination inhibition test results for Eperythrozoon suis antibodies were negative either because E. suis antigens do not cross react with cattle eperythrozoon organism antibodies or blood was collected before there was sufficient time for seroconversion. Bulls had swelling of the scrotal wall, soft testes, and poor semen quality, characterized by low progressive motility and a high percentage of spermatozoa with primary and secondary abnormalities. Some of these abnormalities may be attributed to the age of the bulls. However, loss of scrotal thermoregulation was a major cause of testicular degeneration leading to poor semen quality. Other bulls in the herd had fever, increased respiratory rate, and swollen hindlimbs or scrotum. Subsequent to administration of oxytetracycline, parasitemia resolved rapidly, and clinical signs gradually abated. Four of five bulls successfully passed breeding soundness examinations six months after initial clinical illness. The remaining bull failed twice due to the presence of testicular inflammation.  相似文献

Sixteen yearling bulls were utilized to investigate administration of ergotamine tartrate on semen parameters, fertilization, and endocrinology. Bulls were allotted to a control diet of cracked corn, corn silage, and soybean meal (CON, n = 8) or a diet supplemented daily with 40 microg/kg body weight of ergotamine tartrate (ET, n = 8). Blood samples, average daily gain, scrotal circumference and rectal temperatures were collected every 14 day. Semen samples were obtained every 60 day and evaluated for motility and morphology. Scrotal temperatures were obtained by thermography immediately before electroejaculation. Semen from a subset of bulls from each treatment was also evaluated for in vitro fertilization. Administration of ET increased rectal temperature and resulted in lower scrotal temperatures compared to CON bulls (P < 0.05). However, prolactin, scrotal circumference, testosterone, and semen motility and morphology did not differ between groups throughout the experimental period (224 day). Cleavage rates of embryos derived from in vitro fertilization (IVF) with semen of bulls, fed with ET, were reduced compared to CON (P < 0.05); however, development of cleaved embryos to blastocyst did not differ between treatments. In conclusion, extended exposure of bulls to ET appeared to reduce fertilization potential of sperm.  相似文献

Eighteen yearling beef bulls were used in a study to determine the effects of diets containing gossypol on spermatogenic tissues, Sertoli cells and Leydig cells. Bulls were randomly assigned to one of three (n = 6) isonitrogenous diet groups formulated from alfalfa hay and corn (gossypol-free control), control plus whole cottonseed and cottonseed hulls, or control plus cottonseed meal and cottonseed hulls as sources of gossypol. Testicular tissues collected were examined histologically, and tissues from bulls fed gossypol-free diets were compared with those fed diets containing cottonseed products. Following a 2-mo period (P(1)) when bulls were fed diets containing gossypol, one-half of the bulls were placed on a gossypol-free diet for an additional 2 mo (P(2)) to determine if gossypol effects were reversible. At the end of P(1), bulls fed whole cottonseed and cottonseed meal had larger (P<0.01) lumens in their seminiferous tubules, decreased (P<0.001) wall thickness in their seminiferous tubules, and a reduced (P<0.0001) number of cell layers in their seminiferous tubles, when compared with bulls fed a gossypol-free diet. These histological changes indicate detrimental effects to the spermatogenic tissues and associated cells. Following the initial two months (P(1)) of receiving feed containing gossypol, herd mates that were fed a gossypol-free diet for 2 mo (P(2)) showed improvement in histological characteristics, indicating that gossypol-induced effects were partially reversible.  相似文献

Thirty crossbred bulls, 12 to 13 mo of age, were used to examine the relationship of testosterone and progesterone concentrations and testosterone: progesterone ratio to measurements of testicular function. Bulls were allotted to 1 of 2 groups based on scrotal circumferences (SC) as follows: the Small SC (n=20) group had scrotal circumference less than 28 cm while the Large SC (n=10) group had scrotal circumference greater than 28 cm. All bulls were administered GnRH (100 mug, im), and blood was obtained immediately prior to injection (t=0), 30 min after injection (t=30) and 2 to 3 h after injection (t=150). Serum was assayed for concentrations of testosterone and progesterone. Semen was evaluated for the percentage of morphologically normal spermatozoa. Testicular parenchyma was sectioned and stained, and 300 cross sections per testis of seminiferous tubules were examined under a light microscope and classified as either active (spermatocytes and spermatids present) or inactive (no spermatocytes or spermatids present). Although progesterone concentrations varied widely (range: 21 pg/ml to 1070 pg/ml), repeated measurements from individual bulls were highly correlated (r(2)=0.74) and did not change significantly (P > 0.1) in response to GnRH treatment. Small SC bulls had a higher percentage of inactive seminiferous tubules (P < 0.001) and a lower percentage morphologically normal spermatozoa (P < 0.001) than Large SC bulls, but no differences in testosterone or progesterone concentrations or in the ratio of testosterone: progesterone were detected. Mean serum testosterone concentration increased (P < 0.0001) by 30 min after GnRH treatment and continued to increase (P < 0.0001) through t=150 but did not differ (p > 0.1) between groups. Normal testosterone secretion in response to GnRH injection suggested that no biochemical lesions in the testosterone production pathway were present in bulls with very small scrotal circumference.  相似文献

In bull calves an early transient increase in circulating concentrations of LH occurs between 6 and 20 weeks of age. This has been shown to influence reproductive development and performance later in life. In an attempt to hasten the onset of sexual maturity, bull calves (Hereford x Charolais) were treated (im) with 120 ng/kg of GnRH (n=6) twice every day from 4 to 8 weeks of age; control calves received saline (n=6). Injection of GnRH resulted in an LH pulse in all animals. GnRH treated bulls displayed more rapid testicular growth rates between 22 and 44 weeks of age. Sexual maturity (SC>or=28 cm) was achieved earlier in GnRH treated bulls compared to saline treated bulls (41.7+/-2.22 and 47.0+/-0.45 weeks of age, respectively) and this was confirmed by age of sexual maturity based on ejaculate characteristics (>50 million spermatozoa, >10% motility; 45.0+/-0.86 and 49.0+/-1.13 weeks of age for GnRH and control treated bull calves, respectively; P<0.05). We concluded that treatment with GnRH, twice daily, from 4 to 8 weeks of age, prior to the endogenous early increase in plasma LH concentrations, could increase in plasma LH concentrations, advance testicular development and reduce age at puberty in beef bull calves. This may provide the basis for a simple regimen to hasten sexual development in the bull calf.  相似文献

Scrotal circumference (SC) was measured both before and after 2- to 3-yr-old beef bulls were trucked either medium (138 km) or long (565 km) distances. Two SC measurement techniques were used, one recommended by the Society for Theriogenology and the “slip-off” technique. No differences (P > 0.05) were observed between pre-and post- trucking SC measurements using the Theriogenology technique when bulls were trucked either medium (n = 12; 35.9 ± 0.8 vs 35.8 ± 0.7 cm) or long (n = 16; 36.5 ± 0.5 vs 36.6 ± 0.4 cm) distances. However, when the SC of the same bulls was measured using the “slip-off” technique, there was a small, but significant (P < 0.05), reduction in SC for bulls trucked medium (38.0 ± 0.7 vs 37.5 ± 0.7 cm) and long (38.9 ± 0.4 vs 38.6 ± 0.4 cm) distances. Bulls having SC at or below the recommended minimum standard reacted no differently to the stress of trucking than did bulls having a SC well above the standard. Errors in measurement technique, severe weather conditions and testicular degeneration over an extended interval are discussed in terms of their potential contribution to the reduction in SC observed by some cattlemen. These data support the use of the SC measurement technique recommended by the Society for Theriogenology because its accuracy is not affected by trucking of bulls. The suggested implementation of an adjustment period between the trucking of beef bulls and measurement of their SC using the Theriogenology technique is not supported.  相似文献

In Experiment I, 38 crossbred suckled beef cows grazing fescue pastures and 34 crossbred beef cows grazing bluestem pastures were randomly allocated at the time of calving into a group with 4 teaser bulls or no bulls. Two blood samples were collected 7 d apart from the cows to determine cyclic activity 67 and 76 d after calving in the fescue and bluestem pastures, respectively. Progesterone greater than 1.0 ng/ml in one or both samples indicated cyclic activity. There was no difference in the percentage of cows cyclic among the different groups. The number of cyclic cows in the fescue pasture with bulls was 16/19 (84%); in the fescue pasture with no bulls, 14/19 (74%); in the bluestem pasture with bulls, 17/17 (100%); and in the bluestem pasture with no bulls, 16/17 (94%). Overall cyclic activity among all cows for teaser bull-exposed and no bull was similar, 33/36 (91%) and 30/36 (83%). Overall cyclic activity was greater (P < 0.05) in cows grazing bluestem (33/34), 97% than fescue pastures (30/38), 80%. Measurements of cyclic activity were initiated too late in the postcalving period to quantify differences in estrous activity between the bull and no bull treatment groups. Another trial was planned for the following year with a modified protocol. In Experiment II, blood samples were collected for progesterone concentrations soon after calving and were repeated at intervals to characterize both the occurrence and duration of estrous cycles. In this experiment, 29 crossbred suckled beef cows grazing fescue pastures were randomly allocated 12 d after calving (Day 0) into 1 of 2 groups with teaser bulls or without bulls. Nineteen crossbred beef cows grazing bluestem pastures were allocated similarly 10 d after calving (Day 0). Bulls were added to the groups with bulls in fescue and bluestem pastures on day 6 after the initial allocations. Blood samples were collected from all cows on Day 0 and every 3 d until Day 46. Means (+/- SEM) of the cumulative progesterone concentrations (ng/ml) per cow for the 16 samples from cows grazing fescue were 12.5 +/- 3.5 for cows exposed to bulls, 2.5 +/- 0.16 for cows not exposed to bulls, 27.6 +/- 4.42 for cows grazing bluestem pastures and exposed to bulls, and 16.0 +/- 2.75 for cows without exposure to bulls. Progesterone concentrations were higher in cows exposed to bulls (P < 0.01). The percentages of both short and normal cycles increased (P < 0.01) in groups exposed to bulls (88%, 21/24 and 63%, 15/24) when compared with the no bull groups (29%, 7/24 and 21%, 5/24), respectively. Electroejaculated semen from Santa Gertrudis bulls was used to study the effect of centrifugation (600 x g for 5 min) or dialysis [molecular weight cutoff <14,000 Daltons (Da)] on post-thaw motility and on fertility in beef cattle. Analysis of post-thaw motility showed that the main effects (bulls and semen treatment) were significant (P<0.05). Dialysis significantly improved post-thaw motility in four of seven bulls. Initial volume of seminal plasma in the ejaculate was negatively correlated to post-thaw motility (r = -0.73). No significant improvement in post-thaw motility was observed for bulls with high volumes of ejaculate. In a fertility trial, calving rates of heifers synchronized with PGF(2)alpha and inseminated at 72 and 96 h after the second PGF(2)alpha injection with dialyzed or commercial semen were not statistically different (P>0.05; 54.4% vs 55.4%). These results show that dialysis could be used to improve post-thaw motility of electroejaculated bull semen without altering its fertilizing capacity. However, a high initial volume of seminal plasma seems to have a deleterious effect on sperm freezability that cannot be reversed by dialysis.  相似文献

The study was conducted to evaluate the effects of scrotal insulation on semen samples collected from bulls on embryonic development after IVF. Semen samples were obtained and cryopreserved from four Holstein bulls before and after a scrotal insulation period of 48 h (Day 0). Three types of samples were used for IVF: (1) semen from the test bulls collected 5 d prior to scrotal insulation (pre-insult); (2) semen from Day 13 (2-week post-insult; 2-week PI); and (3) semen from Day 20 (3-week PI). After 18 h of sperm-oocyte co-incubation, the zygotes were cultured for 8 d when a developmental score (0=degenerate, 1=2-cell embryo through 5=blastocyst) was assigned to each embryo. The post-thaw morphological evaluation of sperm samples revealed a decrease (P<0.01) in the percentages of normal spermatozoa in the 3-week PI samples in comparison with the pre-insult samples for Bulls I and III (74-22.3% and 67.7-0.5 %, respectively). The percentage of vacuolated spermatozoa increased significantly for Bull II. The cleavage and blastocyst formation rates and embryo development scores were affected (P<0.01) by the interaction of bull by sample collection time. For Bulls I and III (severe responders) the scrotal insulation effects persisted from the time of cleavage through blastocyst formation. In contrast, the cleavage and blastocyst formation rates for Bulls II and IV were unaffected, despite high percentages of vacuolated spermatozoa present in the post-insult samples for Bull II. In conclusion, the use of scrotal insulation to elevate scrotal temperature was an effective method to obtain semen samples with high percentages of abnormal spermatozoa. The decrease in embryonic development after IVF when using spermatozoa with morphological abnormalities seemed to be multifaceted and related to changes in head morphology.  相似文献

The objectives of this study were to determine if the response to luteinizing hormone releasing hormone (LHRH) could be used to select bull calves capable of early sexual maturation and to establish the optimum route and dose of LHRH. In Trial 1, at 4, 10 and 20 week of age, 20 calves were treated iv with 2 microg/kg body weight of LHRH 1 and 5h after commencing a 9-h period of blood sampling. Bulls were separated into early and late maturing (n=10), based on age at puberty (scrotal circumference (SC) of >or=28 cm). At 4 and 20 week of age, peak serum LH concentrations and area under the LH response curve in response to LHRH were lower (P<0.05) in early- versus late-maturing bulls. In Trial 2, calves at 20 week of age were given LHRH as follows: 2 microg/kg body weight iv (n=6), im (n=6) or sc (n=6); 5 microg/kg im (n=6), or ischio-rectally (ir, n=6) or sc (n=6); and 10 microg/kg im (n=6) or sc (n=6). Serum LH concentrations were at a plateau from 30 to 165 min after treatment with 5 microg/kg of LHRH (im or ir; P>0.05). We concluded that the LH responses to LHRH in calves at 4 and 20 week of age could facilitate the development of a simple test (one blood sample prior to treatment with LHRH and a second during the period of sustained response to LHRH) to select early-maturing bulls.  相似文献
