Selected chemical and physical properties of 12 different pruning waste compost (PWC) samples were evaluated to assess their suitability as substrates for ornamental plants. Samples were taken periodically from the same composting facility over 18 months in order to determine if there was any seasonal variability. In addition to the PWC samples, a Canadian Sphagnum peat and a commercial growing medium (CGM) were used as standard materials. With respect to PWC properties, pH values were above 8, significantly higher than the pH of peat and the commercial substrate. All samples showed adequate levels of organic matter and correspondingly high cation-exchange capacity (CEC) values. The C/N ratio varied between 22 and 48, significantly higher than the optimal values of 15-20. Although composts were sampled from piles established in different seasons, no significant differences were found in their chemical properties. However, water retention characteristics were affected by seasonal changes in components entering the facility. Comparing the properties of PWC to those of peat and commercial growing medium, this material appears to be an acceptable component of a substrate for container-grown ornamental plants.  相似文献

Secreta from the palm and forearm was sampled for 1-min periods by a new technique, using a glass cylinder. Subjects exercised for 10-min periods at successive intensities of 40%, 50% and 65% VO2max with a leg ergometer operated in the supine position. Changes in the concentrations (values) of Na+, K+ and Cl- in their secreta during exercise were investigated. Significant positive correlations were found between the values of any two electrolytes in samples from the palm or the forearm, but the correlations between values for any one of the three electrolytes from the two sites were not significant. Values for concentrations of the electrolytes were significantly higher in samples from the palm than in those from the forearm at rest, 10 min after the beginning of exercise and at the end of exercise. No significant correlation was found between values for electrolytes in samples from the palm and the exercise intensity, but values for Na+ in samples from the forearm increased stepwise with increase in exercise intensity, and similar tendencies were observed for values of K+ and Cl-. The values for the three electrolytes in samples from the forearm, but not the palm, were significantly correlated with values for blood lactate, the percentage of VO2max and the heart rate. These results suggest that the present technique is suitable for successive samplings of secreta from the forearm, and that values for the electrolytes in samples are useful indices of exercise intensity.  相似文献

Honey has been widely used to treat several human pathogens. However, all honeys may not have equal potencies against different human pathogens. The purpose of the current work was to investigate the physico-chemical and antimicrobial qualities of some mono-floral honeys from Ethiopia against some human pathogen bacteria and fungi. In the study, seven different botanical origin honeys were used of which some were from plants known for their medicinal properties. The samples were tested for their major physico-chemical properties (sugar profiles, total free acids, pH, color, electric conductivity and total soluble substances) and their medicinal values as total antioxidant capacity, total phenolic content and antimicrobial properties as minimum inhibitory concentration against some human pathogens, following standard protocols. Generally, the average values of the physico-chemical properties of the samples were within the acceptable ranges of world honey quality values. The average total antioxidant value of the samples was 320.3 ± 15.1 with range of 225.4 ± 12.8–465.7 ± 21.8 μM Fe(II)/100g. Relatively higher values 421.5 ± 23.4 and 465.7 ± 21.8μM Fe(II)/100g recorded for Croton macrostachyus and Vernonia amygalina honeys respectively. The average phenolic contents of the samples varied from 233.3 ± 24.0 to 693.3 ± 26.8 mgGAE/kg and relatively higher values recorded for C. macrostachys and V. amygdalina honeys. The significant proportion of the tested samples showed strong antimicrobial qualities inhibiting the growth of tested pathogens at concentration of 10.5%–28.6% of MIC (% v/v). Honeys from medicinal plants (C. macrostachys and V. amygdalina) relatively showed more antimicrobial properties which could be due to the presence of plant specific phytochemicals which require further investigations.  相似文献

Recent developments in technology and soil science have drawn particular attention to peatlands and their behaviour, properties and utilisation. Further, characteristics of peat are very important in evaluating it as a plant growth medium. The objective of this study was to characterise a number of samples collected from different depths of five different profiles of Yeni?a?a peat (carex) by measuring the organic carbon, total nitrogen, and extinction values (Er) of their humic acid fractions (HAs) in comparison with decomposition degrees. Peat samples having a high decomposition degree were near to the Er45 axis in the ordinate and had high Er67 values, whereas peats with low decomposition degree were far from the Er45 axis and had low Er67 values.  相似文献

The objective was to estimate the possible influence of season or season and sex on the selenium, glutathione peroxidase (GSH-Px), copper, zinc, and superoxide dismutase (SOD) values in plasma with a view to establishing reference values. For this purpose, 55 healthy volunteers (36 women and 19 men) were selected and their whole-blood samples were collected four times a year at the beginning of each season. In the population as a whole, no statistically significant seasonal differences were detected in copper, selenium, and GSH-Px values in plasma, whereas zinc and SOD values in plasma depended on the season. In general, the variability was higher among the women. In view of the differences detected according to sex and/or season in the parameters studied, we recommend taking samples from men and women and from different seasons to establish reference values.  相似文献

The mechanism of transport of trace elements from the mother to the newborn is still not well known. The aim of present study was to compare the status of trace toxic elements, arsenic (As), cadmium (Cd), and lead (Pb) in biological samples (whole blood, urine and scalp hair) of insulin-dependent diabetic mothers (age ranged 30-40) and their newly born infants (n = 76). An age and socioeconomics matched 68 nondiabetic mothers and their infants, residing in the same locality, who were selected as referents. The elemental concentrations in all three biological samples were determined by an electrothermal atomic absorption spectrometer, prior to microwave-assisted acid digestion. The mean values of As, Cd, and Pb in all biological samples of diabetic mothers and their infants were significantly higher as compared to the referent mother-infant pair samples (p < 0.01). The high levels of As, Cd, and Pb in biological samples of diabetic women may play a role in the pathogenesis of diabetes mellitus and impacts on their neonates.  相似文献

The aim of this study was to investigate the lubrication potential of 2 grades of magnesium stearate (MS) blended with a mix of dicalcium phosphate dihydrate and microcrystalline cellulose. Force-displacement, force-time, and ejection profiles were generated using an instrumented rotary tablet press, and the effect of MS mixing time (10, 20, and 30 minutes) and tableting speed (10.7, 13.8, and 17.5 rpm) was investigated. The packing index (PI), frictional index (FI), and packing energy (PE) derived from the force-displacement profiles showed that MS sample I performed better than sample II. At higher lubricant mixing times, the values of PI were observed to increase, and values of FI and PE were observed to decrease for both MS samples. Lower values of area under the curve (AUC) calculated from force-time compression profiles also showed sample I to be superior to sample II in lubrication potential. For both the samples, the values of AUC were observed to decrease with higher lubricant mixing times. Tapping volumetry that simulates the initial particle rearrangement gave values of parameter a and C(max) that were higher for sample I than sample II and also increased with lubricant mixing time. The superior lubrication potential of sample I was also established by the lower values of peak ejection force encountered in the ejection profile. Lower ejection forces were also found to result from higher tableting speeds and longer lubricant mixing times. The difference in lubrication efficacy of the 2 samples could be attributed to differences in their solid-state properties, such as particle size, specific surface area, and d-spacing.  相似文献

Calcium carbonate is added to fermentation media in biosynthesis of tetracyclines for providing definite pH values and binding tetracycline into insoluble complexes. Seven different samples were studied with respect to their physical properties, such as the microscopic size of the particles, their form, capacity for agglomeration, specific volume, rate of the particle precipitation and chemical properties, such as purity, buffer capacity, effect on the medium pH before and after sterilization. The above properties were studied in comparison with activity chlortetracycline biosynthesis. Microfine calcium carbonate proved to be the best from the point of view of productivity of Str. aureofaciens. With its use the activity of the culture fluid increased by 20 per cent as compared to the other samples. The titration curve of the sample had the lowest bend.  相似文献

The species Perna viridis is a highly consumed species, which fast growth makes it an interesting aquaculture alternative for Venezuelan and Trinidad coasts. With the aim to contribute with its nutritional value information, this study analyzed lipid and fatty acid contents from samples taken in five locations from Eastern Venezuela and three from Trinidad West Coast. Total lipids were extracted and quantified, from a pooled sample of 100 organisms per location, by standard gravimetric methods, and their identification and quantification was done by TLC/FID (Iatroscan system). Furthermore, the esterified fatty acids of total lipid, phospholipids and triacylglycerols were identified and quantified by gas chromatography. Eastern Venezuela samples from Los Cedros, La Brea and Chaguaramas showed the highest total lipid values of 7.92, 7.74 and 7.53, respectively, and the minimum values were obtained for La Restinga (6.08%). Among lipid composition, Chacopata samples showed the lowest phospholipid concentration (48.86%) and the maximum values for cholesterol (38.87%) and triacylglycerols (12.26%); besides, La Esmeralda and Rio Caribe samples exhibited maximum phospholipids (88.71 and 84.93 respectively) and minimum cholesterol (6.50 and 4.42%) concentrations. Saturated fatty acids represented between 15.04% and 65.55% within total lipid extracts, with maximum and minimum values for La Esmeralda and Chacopata, respectively. Polyunsaturated results resulted between 7.80 and 37.18%, with higher values in La Brea and lower values in La Esmeralda. For phospholipids, saturated fatty acids concentrations varied between 38.81 and 48.68% for Chaguaramas and Chacopata samples, respectively. In the case of polyunsaturated fatty acids, these varied between non detected and 34.51%, with high concentrations in Los Cedros (27.97%) and Chaguaramas (34.51%) samples. For the triacylglycerols, the saturated fatty acids composition oscillated between 14.27 and 53.80% with low concentrations for Chacopata and high concentration for La Restinga; the polyunsaturated fatty acids were between 4.66 and 35.55% with lower values for Chacopata and higher values for Chaguaramas samples. P. viridis is recommended for human being consumption, according to the high content of unsaturated fatty acids found for this species.  相似文献

The study was performed to compare real-time PCR after nucleic acid extraction directly from stool samples as well as from samples stored and transported on Whatman papers or flocked swabs at ambient temperature in the tropics. In addition, the possible suitability for a clear determination of likely aetiological relevance of PCR-based pathogen detections based on cycle threshold (Ct) values was assessed. From 632 Tanzanian children <5 years of age with and without gastrointestinal symptoms, 466 samples were subjected to nucleic acid extraction and real-time PCR for gastrointestinal viral, bacterial and protozoan pathogens. Equal or even higher frequencies of pathogen detections from Whatman papers or flocked swabs were achieved compared with nucleic acid extraction directly from stool samples. Comparison of the Ct values showed no significant difference according to the nucleic acid extraction strategy. Also, the Ct values did not allow a decision whether a detected pathogen was associated with gastrointestinal symptoms.  相似文献

Sixty seven sediment samples were collected from Kyeonggi Bay, Korea, including the mouth of Han River, Incheon Harbor, the Namdong industrial complex, and the open sea. Collections were conducted in December, 1995 and samples were maintained frozen (-20 °C) until analysis. Dichloromethane extracts were analysed for their content of CYP1A1-inducing compounds with a P450RGS (reporter gene system) assay, and for polycyclic aromatic hydrocarbons (PAHs). Sediment samples were also analysed for organic carbon (OC) content and grain size, to aid in evaluating the relationship between contamination and physical nature of the sediments. The responses of the P450RGS assay to the sediment extracts were expressed as μg of benzo[a]pyrene toxic equivalents per g dry weight (μg g-1 BaPTEQ), and these values correlated well (r2 = 0.624 with total PAHs. BaPTEQ values were also highly correlated with the OC content of the sediments. The determination of P450RGS BaPTEQ is a useful tool, because it is both a rapid and inexpensive means of assessing the potential toxicity of organic compounds in environmental sediment samples. These values represent an estimate of the levels of compounds in the sediment that are potentially available to organisms through chronic exposure to pore water or ingestion of benthic species. We believe BaPTEQ values are more useful than tables of specific PAH concentrations, if the purpose of the investigation is to either obtain a rapid screening of an area or to develop some form of ecological or human health risk assessment.  相似文献

Among Metazoa, sponges (phylum Porifera) are the richest source for different bioactive compounds. The availability of the raw material is, however, restricted. To obtain enough of the bioactive compounds for application in human therapy, sponges have to be cultured in in vitro systems. One technique for the establishment of a long-term cell culture from sponges has recently been elaborated. Here, we present a procedure to cultivate tissue samples from sponges in an open system. The species Geodia cydonium, which produces bioactive compounds, has been selected. Tissue samples of approximately 10 g were attached to the bottoms of cultivation trays. After 2 to 3 days, the tissue samples formed a robust contact with the metal support. Subsequently, sets of trays, called tray batteries, either remained in huge aquaria at the Center for Marine Research or were transferred to the vicinity of a fish and mussel farm. The growth rates of the samples remained unchanged within the first month; however, after 3 and 6 months, they increased to 147% and 189%, respectively. In parallel, extracts were prepared from the tissue samples and tested for cytotoxicity in a mouse lymphoma cell assay system. Extracts from cultured tissue initially had a low inhibitory potency; however, after cultivation for 3 or 6 months, values comparable to those of extracts from sponges taken from the biotope were found. In addition, a molecular marker was applied to document the response (health state) of the tissue and the identity of the material in culture. The CD63 molecule was chosen because the expression of this molecule in mammalian systems changes with the age of the animals. The corresponding complementary DNA was isolated from Geodia cydonium. With this probe, the level of expression in cultured tissue samples decreased immediately after starting cultivation; after a cultivation period of 6 months, however, values were similar to those found in controls. These data show that sponge species that produce bioactive compounds can be cultivated in open systems, in which they retain their potency to produce bioactive compounds as well as their health state. Pigment concentration, in vivo absorption, and photosynthetic parameters of the coralline alga Hydrolithon onkodes (Heydrich) Penrose and Woelkerling were compared among samples from a lagoon and from a reef crest of Tahiti Island. Four groups of specimens were considered, differing in their natural exposure to PAR. For specimens collected from the lagoon, the tissues from low-light samples had significantly higher pigment concentration, particularly chl a and phycobilins, compared with the high-light exposed plants that contained more total carotenoids. The in vivo absorption spectra normalized to chl a (called a* values) also revealed differences. The low-light samples had a reduced absorption capacity and a well-marked phycobilin absorption signature, whereas sunlit samples showed a greater absorption at wavelengths absorbed mainly by chl a and carotenoids. The decrease of a* when pigment concentration increased is interpreted as a consequence of the pigment packaging. Significantly lower α (chlorophyll basis) and higher Ek values were found in the shaded plants. The values of P max for the four groups of specimens were not significantly different. The samples showed various degrees of photoinhibition depending on the light exposure during growth, and this effect was more pronounced in the shaded plants. The specimens from the reef crest deviate from the general model presented for the lagoon samples and show a mix of sun- and shade-exposed characteristics. We have shown that the coralline alga H. onkodes responds to its light environment, probably by acclimation rather than ecotypic genetic variation, by adjusting its physiology, but some morphological differences are also involved. Photoacclimation can explain partly the wide distribution of this species over the reef ecosystem and its major contribution to the building of the reef.  相似文献

Hematological, differential, and blood chemistry values were determined in four Chinese pangolins (Manis pentadactyla), both at the time of their arrival and after more than 1 year in captivity. The data presented were derived from two samples obtained from each of four animals, and they provide a preliminary basis for comparison with clinical blood values from this and other pangolin species. No peculiar features were identified that distinguish this species from other mammals.  相似文献

We employed ovalbumin (a simulant used for ricin and botulism toxins in biodefense applications) and its high affinity polyclonal antibody as a model system to examine the sensitivity, dynamic range, linearity, and reproducibility of forward-phase array results in comparison to suspension arrays. It was found that protein microarrays had a dynamic range of 4 orders of magnitude and a sensitivity of less than 1 pg/mL, respectively. The dynamic range and sensitivity of suspension arrays were close to 2 orders of magnitude and 0.25 ng/mL, respectively. The sensitivity we observed for the suspension arrays is comparable to that reported for enzyme-linked immunosorbent assays (ELISAs) in the literature. We used ovalbumin samples with two different purities, 38.0% and 76.0% (w/w), as determined by polyacrylamide gel electrophoresis (PAGE). These samples were used to evaluate the effect of impure samples on detection. The data obtained from the forward-phase protein arrays gave values that were consistent with the PAGE data. The data from the suspension arrays were not as consistent and may indicate that this format may not give as reliable data with impure samples. Knowledge of the advantages and disadvantages of the two proteomic methods would allow their more rational use in clinical diagnosis.  相似文献

Summary The oxygen uptake of Mytilus californianus between 12 and 24° C. varies greatly with different samples, from approximately 0.3 to 2.0 cc. per 10 gms. of wet weight.. Similar values are found for the carbon dioxide production. Respiratory quotients as high as 2.2 were found; they are explained by the buffering action of the shell. Simultaneous profiling analysis of urinary amino acids (AAs) and carboxylic acids (CAs) was combined with retention index (I) analysis for graphic recognition of abnormal metabolic state. The temperature-programmed I values of the AA and CA standards measured as ethoxycarbonyl (EOC)/methoxime (MO)/tert-butyldimethylsilyl (TBDMS) derivatives were used as the reference I values. Urine samples were subjected to the sequential EOC, MO and TBDMS reactions for the analysis by gas chromatography (GC) and GC-mass spectrometry. The complex GC profiles were then transformed into their respective I patterns in bar graphic forms by plotting the normalized peak area ratios (%) of the identified AAs and CAs against their reference I values as the identification numbers. When the present method was applied to infant urine specimens from normal controls and patients with inherited metabolic diseases such as phenylketonuria, maple syrup urine disease, methylmalonic aciduria or isovaleric aciduria, each I pattern of bar graph more distinctly displayed quantitative abundances of urinary AAs and CAs in qualitative I scale, thus allowing graphic discrimination between normal and abnormal states.  相似文献

Hyaluronic acid (HA) is a natural polysaccharide with importance in the pharmaceutical, medical and cosmetic industries. Determining factors in its final applications are its physicochemical properties, particularly molecular mass. A high molecular mass HA was degraded using five different hydroxyl free-radical starting concentrations chemically produced from ascorbic acid and hydrogen peroxide. The aims of the study were to investigate the effect of different hydroxyl free-radical concentrations on the chain length of HA and compare the molecular masses obtained from analytical ultracentrifugation using sedimentation equilibrium experiments and size exclusion chromatography/multi-angle laser light scattering (SEC/MALLS). The results indicated that their molecular masses varied, depending on the degree of hydroxyl free-radical starting concentration. Molecular mass values obtained from sedimentation equilibrium experiments for each sample showed the same trend as those obtained from the SEC/MALLS in the range of molecular masses studied. The molecular masses obtained from sedimentation equilibrium for high molecular mass samples from reciprocal plots of apparent weight average molecular mass against concentration gave values similar to those obtained by SEC/MALLS. In contrast, the molecular mass from conventional plots for high molecular mass samples were much lower than those from SEC/MALLS, even when high ionic strength buffers were used.  相似文献

Summary To assess the status of copper in Egyptian soils, surface and subsurface soil samples were collected from various geographical regions of Egypt and of various genesis. The samples were analyzed for the total Cu, water-soluble plus exchangeable as well as the acid-extractable and EDTA-extractable Cu. The total Cu varied between 3.5 and 72.3 ppm. Less than 2% of this copper was in the soluble plus exchangeable form. The highest values of total and soluble plus exchangeable copper were found in the alluvial soils while the sandy soils had the least amounts. This was attributed to the high clay and organic matter content of the alluvial soils in addition to the dominance of montmorillonitic clay minerals in their fine fraction. The calcareous soils showed intermediate values of total and soluble plus exchangeable Cu due to their lower organic matter and clay mineral content. The clay mineralogy of these soils revealed the dominance of illitic and kaolinitic minerals which are relatively poor in Cu and other trace elements.Because of their relatively higher organic matter content, the alluvial soils had a large percentage (up to 43%) of their total Cu in the EDTA extractable form. The calcareous soils, on the other hand, had a large percentage (up to 52%) of their Cu in the acid-extractable form. The EDTA extractable Cu was correlated with the organic matter content of the soils. Since the pH of the EDTA extract was found to be dependent on the CaCO3 content of the soils, it was suggested that the method be modified so that the final pH of the extract is constant for all soils.  相似文献

Hormonal analysis of urine from free-ranging primates has been limited due to the difficulty of preserving samples under field conditions. Drying urine on filter paper is an alternative for field preservation. This study describes a new laboratory method for eluting steroids from filter paper with methanol, along with a series of experiments used to develop and validate this method. The overall elution recovery of estrone sulfate (ES) from filter paper was 86.4%. Estrone conjugate (E1C) levels in humans and captive orangutans were analyzed by radioimmunoassay (RIA). Values from samples dried on filter paper were significantly correlated with values from matched frozen samples, with elution efficiencies ranging between 97.1% and 102.4%. Creatinine (Cr) measurements from frozen urine compared to urine dried on filter paper were also significantly correlated (r=0.96) with an elution efficiency of 101.7%. After the samples were stored for 2 years, the absolute values of E1C and Cr were significantly lower but were still significantly correlated with frozen urine values. These data demonstrate the effectiveness of filter paper as a medium for preserving urinary steroid samples, and the efficiency of methanol as a solvent for eluting E1C and Cr. This method thus provides a viable alternative to the traditional procedure of freezing urine for field studies, where freezers are not available.  相似文献
