A method is described for counting viable units of Mycobacterium microti (vole bacillus) suspensions after 21 days of culture on an oleic-albumin agar medium containing 5% defibrinated horse blood, with a tight seal to maintain humidity. This method is important because vole bacillus is an alternative to BCG in the prevention of tuberculosis.  相似文献

Despite their remarkable genetic homology, members of the Mycobacterium tuberculosis complex express very different phenotypes, most notably in their spectra of clinical presentation. For example, M. tuberculosis is regarded as pathogenic to humans, whereas members having deleted RD1, such as Mycobacterium microti and Mycobacterium bovis BCG, are not. The dassie bacillus, an infrequent variant of the M. tuberculosis complex characterized as being most similar to M. microti, is the causative agent of tuberculosis (TB) in the dassie (Procavia capensis). Intriguingly, the dassie bacillus is not pathogenic to rabbits or guinea pigs and has never been documented to infect humans. Although it was identified more than a half-century ago, the reasons behind its attenuation are unknown. Because large sequence polymorphisms have presented themselves as the most obvious genomic distinction among members of the M. tuberculosis complex, the DNA content of the dassie bacillus was interrogated by Affymetrix GeneChip to identify regions that are absent from it but present in M. tuberculosis H37Rv. Comparison has led to the identification of nine regions of difference (RD), five of which are shared with M. microti (RDs 3, 7, 8, 9, and 10). Although the dassie bacillus does not share the other documented deletions in M. microti (RD1(mic), RD5(mic), MID1, MID2, and MID3), it has endured unique deletions in the regions of RD1, RD5, N-RD25, and Rv3081-Rv3082c (virS). RD1(das), affecting only Rv3874-Rv3877, is the smallest natural deletion of the RD1 region uncovered and points to genes within this region that are likely implicated in virulence. Newfound deletions from the dassie bacillus are discussed in relation to their evolutionary and biological significance.  相似文献

Decline and long-term depression of mean densities of the grey-sided vole (Myodes rufocanus) and the field vole (Microtus agrestis) have occurred in managed forest landscapes of Sweden since the 1970s. Generally poor over-winter survival during a period with mild winters suggested a common climatic driver, but other explanations exist. Here we explore the response of the grey-sided vole, preferring forested habitats, and the field vole, preferring open habitats, to clear-cutting of old forest in Sweden. The cumulated impact from long-term clear-cutting explained local disappearances of the grey-sided vole. Maintained connectivity of old forest to stone fields was important for local population survival, since no such populations disappeared. For the grey-sided vole, it is probable that climate is not the dominating driver due to different timing of the decline in our study area. Instead, habitat loss is concluded as being a potential cause of the decline in mean density and depression of grey-sided vole densities. The long-lasting depression of field vole densities, despite favourable landscape changes, suggests action of another strong driver. A recent field vole recovery, essentially back to pre-decline densities and distribution, coincided with favourable winter/snow conditions, suggesting a climatic driver in this case.  相似文献

Dynamic changes in the lipid composition of the cell wall occur in pathogenic mycobacteria that are often intended for adaptation to the host environment. Dormant mycobacteria should have evolved efficient maneuvers for cohabitation, allowing the microbes to persist for years within the host. Glycerol monomycolate (GroMM) has been implicated as a specific immune target in human individuals with latent, but not active, tuberculosis, but the in vivo response to GroMM and the relevance of it to latent infection remain poorly understood. Here, we immunized guinea pigs with bacillus Calmette–Guerin (BCG) expressing high levels of GroMM and then, monitored skin reactions at the site of challenge with GroMM-containing liposome. We found that BCG-immunized guinea pigs mounted enhanced skin reactions to GroMM with prominent local infiltration by eosinophils. Consistent with this, GroMM-stimulated lymph node cells upregulated the expression of T helper (Th)2-type cytokines, such as interleukin (IL)-5 and IL-10, that could potentially counteract the microbe-eliminating Th1-type cytokine response. On the basis of these observations, we predict that the host response to GroMM produced by dormant mycobacteria would contribute to their long-term survival in the host.  相似文献

【背景】我国禽型结核菌素(avian tuberculin)的制造用菌株为CVCC 68201、CVCC 68202和CVCC 68203株,但目前仍未明确这3株菌的生物学特性及对豚鼠致病性的情况。【目的】探究禽分枝杆菌(Mycobacterium avium)的生物学特性及对动物机体的致病性,为禽结核病和牛结核病的防控工作提供技术支撑。【方法】对3株禽分枝杆菌基因组进行鉴定分析及核酸相似度分析;用3株禽分枝杆菌分别感染豚鼠,观察感染后的临床症状、病理学变化、体重增重情况分析、皮内变态反应结果、脏器系数变化等,进而分析3株禽分枝杆菌对豚鼠的致病力。【结果】种型鉴定和进化分析结果表明,CVCC 68201、CVCC 68202和CVCC 68203均为禽分枝杆菌,基因组与Mycobacterium avium subsp. avium FDAARGOS_1608最为相近;在感染前期、中期、后期对3株禽分枝杆菌感染豚鼠的体重增重情况分析发现,感染禽分枝杆菌影响豚鼠增重,主要表现为生长迟缓,感染第5周时,CVCC 68201、CVCC 68202组豚鼠的平均体重明显轻于未感染组;皮内变态反应试验结果显示,感染CVCC 68201组豚鼠的皮肤红肿面积明显大于其他2个感染组,CVCC 68201可引起机体更为强烈的迟发型变态反应;3株禽分枝杆菌感染后,豚鼠脾脏和肺脏存在不同程度的肿大与出血,其中感染CVCC 68201豚鼠的肺脏系数与未感染组相比差异显著(P<0.01);病理学观察结果显示,豚鼠肺脏可见不同程度病变,其中CVCC 68201组更为严重,表现为肿大和轻微出血。各感染组豚鼠肺脏和脾脏组织切片抗酸染色均可见红色的分枝杆菌散在浸润。【结论】3株禽分枝杆菌对豚鼠均有一定程度的致病性,可引发局部病变。本研究为禽分枝杆菌的制备和鉴定提供依据,也为牛结核病的鉴别诊断方法研究提供参考。  相似文献

Blood parasites of small mammals living in Białowieża Forest (eastern Poland) were investigated between 1996 and 2002. The following haemoparasite species were found:Trypanosoma (Herpetosoma) evotomys in bank voleClethrionomys glareolus; T. (H.) microti in root voleMicrotus oeconomus; Babesia microti in root vole;Hepatozoon erhardovae in bank vole andHepatozoon sp. in root vole. Some non-identifiedBartonella species were found in bank vole, root vole, field voleMicrotus agrestis, yellow-necked mouseApodemus flavicollis, common shrewSorex araneus, Eurasian water shrewNeomys fodiens, and Mediterranean water shrewN. anomalus. The prevalence and diversity of blood parasites were lower in shrews than small rodents. Totally, 52.0% of bank voles, 50.0% of root voles, 32.5% of common shrews, and 41.2% of Eurasian water shrews were infected with any of the blood parasites. Mixed infections were seldom observed in bank vole (17.3% of investigated individuals) and root vole (14.7%). No animals were infected with three or four parasites simultaneously. Infection of Białowieża small mammals with haemoparasites seemed to be similar to those described in other temperate forest regions rather than boreal ones. Infection rates of rodent species seem to be higher in their typical habitats: for bank vole it was the highest in mixed forest, whereas for root vole in sedge swamp. The results suggest that Arvicolidae play a greater role than Muridae or Soricidae in maintenance ofBabesia andHepatozoon foci in natural environments of central Europe.  相似文献

The phenomenon of interspecific hybridization accompanied by transfer of the mitochondrial genome from the northern red-backed vole (Clethrionomys rutilus) to the bank vole (Cl. glareolus) in northeastern Europe is well known already for 25 years. However, the possibility of recombination between homologous segments of maternal and paternal mtDNAs of the voles during fertilization was not previously studied. Analysis of data on variability of nucleotide sequences of the mitochondrial gene for cytochrome b in populations of red-backed and bank voles in the area of their sympatry has shown that as a result of interspecific hybridization, the mitochondrial gene pool of bank voles contains not only mtDNA haplotypes of red-backed vole females, but also mtDNA haplotypes of bank voles bearing short nucleotide tracts of red-backed vole mtDNA. This finding supports the hypothesis that an incomplete elimination of red-backed vole paternal mtDNA during the interspecific hybridization between bank vole females and red-backed vole males leads to the gene conversion of bank vole maternal mtDNA tracts by homologous ones of mtDNA of red-backed vole males.  相似文献

Myobacterium tuberculosis is the most common infectious cause of death in the world, with up to one-third of the population infected. In industrial countries infection with M. tuberculosis and tuberculosis disease has been decreasing since the 19th century. Now, however, tuberculosis disease is on the increase again, with resistance of the bacillus to available drugs spreading rapidly. This resurgence can be seen from the ecological and evolutionary point of view, where human hosts are the niche of the tuberculosis bacillus.  相似文献

The goal of this study is to determine whether a parasitic nematode may regulate, or destabilise by inducing demographic cycles, its host populations. We explore three host–parasite systems through population dynamic models. The hosts considered are the fossorial water vole, Arvicola terrestris, the common vole Microtus arvalis and the bank vole Myodes (Clethrionomys) glareolus and the parasitic nematode is Trichuris arvicolae. Three differential equation-based mathematical models are developed including host immunity and the existence of trade-off between immunity and host survival. Using parameters estimated from field data and laboratory observations, all these models show that T. arvicolae can induce host population regulation but not demographic cycles. The regulation effect of the nematode is un-ambiguous for the water vole (reduction of 50.2% of the host population size), but less obvious for the common vole (5.9%) and even less for the bank vole (1.4%). Important biological parameters to be taken into account in such models are discussed. Experimental confirmation of the regulatory potential of the nematode and of the costs of mounting an immune response against this nematode are now required. Communicated by W. Lutz  相似文献

Three species of British voles, the bank vole Clethrionomys glareolus, the field vole Microtus agrestis and the water vole Arvicola terrestris were maintained on diets of seed and plant leaf material to investigate changes in gut anatomy. C. glareolus and M. agrestis showed significant changes in most regions of the gut; they developed longer and heavier tracts when on a highfibre diet. This response may be important in enabling these animals to withstand seasonal changes in diet quality.  相似文献

The biology of the gray-sided voleClethrionomys rufocanus in Hokkaido, concerning taxonomy, morphology, phylogeny, distribution, and natural history, is reviewed. Applied issues in forest management (damage, control and census) are also mentioned. AlthoughClethrionomys rufocanus of Hokkaido was originally identified as a distinct species,Evotomys (=nowClethrionomys) bedfordiae Thomas, 1905, current literature generally refers to the gray-sided vole of Hokkaido asClethrionomys rufocanus or asC. rufocanus bedfordiae (vernacular name, the Bedford’s red-backed vole). The gray-sided vole is the most common small mammal in Hokkaido. It inhabits open areas as well as forests, and mainly feeds on green plants. The gray-sided vole has a high reproductive potential; litter size: 4–7; gestation period: 18–19 days; maturation age: 30–60 days old. Although spring-born individuals usually attain sexual maturity in their summer/fall of birth, their maturation is sometimes suppressed under high densities. The breeding season is generally from April to October, but with some regional variation.Clethrionomys rufocanus has a rather specialized diet (folivorous), particularly during winter when it feeds on bamboo grass. Many predators specialize on the grey-sided vole in Hokkaido; even the red fox, which is a typical generalist predator, selectively feeds on this vole. Damage by voles’ eating bark used to be sever on forest plantations in Hokkaido. Censuses of small rodents have been carried out for management purpose since 1954.  相似文献

Three wild red-legged partridges (Alectoris rufa) from intensively managed hunting areas in Spain were received for necropsy. They showed granulomatous lesions in different parts of the body, mainly in liver and spleen. Microscope examination of the granulomas showed central caseous necrosis and large amounts of acid-fast bacilli, surrounded by epitheloid cells, giant cells, and lymphocytes. Attempts to isolate and culture the bacillus in Colestsos medium were unsuccessful, but the polymerase chain reaction technique revealed the presence of microorganisms belonging to the Mycobacterium avium complex in one of the partridges. This is the first report of avian tuberculosis in free-living red-legged partridges.  相似文献

With 500 000 cases of multidrug‐resistant tuberculosis there is an urgent need for attractive targets to enable the discovery of novel antimycobacterials. The biosynthesis of essential cofactors is of particular interest as these pathways are absent in man and their inhibition is expected to affect the metabolism of Mycobacterium tuberculosis at multiple sites. Our data demonstrate that the pathogen synthesizes pyridoxal 5‐phosphate (PLP), the bioactive form of vitamin B6, by a heteromeric PLP synthase composed of Pdx1 (Rv2606c) and Pdx2 (Rv2604c). Disruption of the pdx1 gene generated a strictly B6 auxotrophic M. tuberculosis mutant, Δpdx1. Removal of the cofactor during exponential growth or stationary phase demonstrated the essentiality of vitamin B6 biosynthesis for growth and survival of the pathogen in culture. In a tuberculosis dormancy model based on gradual oxygen depletion, de novo biosynthesis of PLP was required for regrowth of the bacillus after direct oxygen exposure. The Δpdx1 mutant showed a severe growth defect in immunocompetent mice: bacilli applied intranasally failed to persist in host tissues and were quickly cleared. We conclude that vitamin B6 biosynthesis is required for survival of M. tuberculosis in vivo and thus might represent a candidate pathway for the development of new antitubercular agents.  相似文献

Arvicolinae voles are small herbivores relying on constant food availability with weak adaptations to tolerate prolonged food deprivation. The present study performed a comparative analysis on the responses to 4–18 h of food deprivation in the common vole (Microtus arvalis) and the tundra vole (Microtus oeconomus). Both species exhibited rapid decreases in the plasma and liver carbohydrate concentrations during phase I of fasting and the decline in the liver glycogen level was more pronounced in the tundra vole. The plasma thyroxine concentrations of the common vole decreased after 4 h. Lipid mobilization (phase II of fasting) was indicated by the increased plasma free fatty acid levels after 8–18 (the common vole) or 4–18 h (the tundra vole) and by the elevated lipase activities. In the tundra vole, the plasma ghrelin concentrations increased after 12 h possibly to stimulate appetite. Both species showed increased liver lipid concentrations after 4 h and plasma aminotransferase and creatine kinase activities after 12–18 h of food deprivation implying liver dysfunction and skeletal muscle damage. No signs of stimulated protein catabolism characteristic to phase III of fasting were present during 18 h without food.  相似文献

  • 1 Martino's vole Dinaromys bogdanovi is the only living member of the Tertiary genus Dinaromys, and probably also the only surviving member of the Pliomys lineage. The range of the genus Dinaromys has historically been small and its rate of evolution has been low.
  • 2 Martino's vole shows all three attributes of rarity in accordance with Rabinowitz's ‘seven forms of rarity’ model: (i) its range is estimated at 43 545 km2 but the area of occupancy is <5200 km2; (ii) its habitat requirements are narrow and the species is strictly tied to exposed, karstified bedrock; and (iii) current populations are invariably small and frequently isolated.
  • 3 The Pleistocene range of Martino's vole exceeded the recent one, at least in the north‐western part of the Balkans, and its shrinkage continued into the Holocene.
  • 4 Martino's vole may be in competition with the European snow vole Chionomys nivalis, which has a very similar morphology and presumably identical habitat requirements, but is shifted towards an r‐selected life‐history strategy. Long‐term sympatry of these voles has probably resulted in competitive exclusion of the relatively K‐selected Martino's vole by the relatively r‐selected European snow vole.
  • 5 Martino's vole consists of two deeply divergent (about one million years ago) phylogeographical lineages, which may represent distinct cryptic species. Rarity is particularly pronounced in the north‐western lineage to the west of the Neretva River, where rocky habitats are largely occupied by the European snow vole.
  • 6 In the IUCN Red List of Threatened Animals, Martino's vole is classified as ‘near threatened’. However, the north‐western lineage, which is phylogeographically most divergent and has the greatest genetic diversity, is classed as a ‘vulnerable’ evolutionary significant unit on the basis of its small area of occupancy (<2000 km2). Long‐term population monitoring is an essential step in evaluating the conservation needs of Martino's vole.

Grazing‐induced changes in plant quality have been suggested to drive the negative delayed density dependence exhibited by many herbivore species, but little field evidence exists to support this hypothesis. We tested a key premise of the hypothesis that reciprocal feedback between vole grazing pressure and the induction of anti‐herbivore silicon defenses in grasses drives observed population cycles in a large‐scale field experiment in northern England. We repeatedly reduced population densities of field voles (Microtus agrestis) on replicated 1‐ha grassland plots at Kielder Forest, northern England, over a period of 1 year. Subsequently, we tested for the impact of past density on vole life history traits in spring, and whether these effects were driven by induced silicon defenses in the voles’ major over‐winter food, the grass Deschampsia caespitosa. After several months of density manipulation, leaf silicon concentrations diverged and averaged 22% lower on sites where vole density had been reduced, but this difference did not persist beyond the period of the density manipulations. There were no significant effects of our density manipulations on vole body mass, spring population growth rate, or mean date for the onset of spring reproduction the following year. These findings show that grazing by field voles does induce increased silicon defenses in grasses at a landscape scale. However, at the vole densities encountered, levels of plant damage appear to be below those needed to induce changes in silicon levels large and persistent enough to affect vole performance, confirming the threshold effects we have previously observed in laboratory‐based studies. Our findings do not support the plant quality hypothesis for observed vole population cycles in northern England, at least over the range of vole densities that now prevail here.  相似文献

Several LINE-1s have been isolated and characterized from genomic DNA of the vole, Microtus kirgisorum. Blot hybridization revealed specific restriction patterns of L1 elements in vole genomes. Rehybridization of the genomic blot with a cloned 5′-end fragment revealed two major bands indicating the presence of two different L1 subfamilies. The copy numbers are estimated for different parts of M. kirgisorum L1 elements. Data also demonstrate that most vole L1 elements are truncated at the 5′-end; however, in contrast to mouse, the ORF1 copy number is higher in vole. A difference between the substitution rates of the ORF1 5′-region (approximately 330 nucleotides) and the rest of the L1 coding regions is revealed. In a short but intense field investigation, surface activity of common voles in a cropped lucerne field was assayed by live-trapping while, simultaneously, hunting activity and yield of three species of raptors were recorded by continuous observation. Pronounced short-term rhythms in trappability of the vole population ran parallel with fluctuations in yield per hour of flight-hunt of hen harriers and kestrels. These raptors, as well as Rough-legged buzzards, hunted more at times of increased vole surface activity; hen harriers saved c. 15 hours of flight-hunt per day by such temporal adjustment, corresponding to about 12% of their daily energy intake. Voles suffered a predation rate of an estimated 0 2% per day; under such heavy predation and temporal concentration of raptor hunting at times of increased vole activity, surface feeding in synchrony with the vole majority was associated with increased risk of predation.  相似文献

Although large human populations have been safely immunized against tuberculosis with two live vaccines, Mycobacterium bovis BCG or Mycobacterium microti, the vole bacillus, the molecular basis for the avirulence of these vaccine strains remains unknown. Comparative genomics has identified a series of chromosomal deletions common to both virulent and avirulent species but only a single locus, RD1, that has been deleted from M. bovis BCG and M. microti. Restoration of RD1, by gene knock-in, resulted in a marked change in colonial morphology towards that of virulent tubercle bacilli. Three RD1-encoded proteins were localized in the cell wall, and two of them, the immunodominant T-cell antigens ESAT-6 and CFP-10, were also found in culture supernatants. The BCG::RD1 and M. microti::RD1 knock-ins grew more vigorously than controls in immunodeficient mice, inducing extensive splenomegaly and granuloma formation. Increased persistence and partial reversal of attenuation were observed when immunocompetent mice were infected with the BCG::RD1 knock-in, whereas BCG controls were cleared. Knocking-in five other RD loci did not affect the virulence of BCG. This study describes a genetic lesion that contributes to safety and opens new avenues for vaccine development.  相似文献
