We describe three new species of the genus Gromia from bathyal and abyssal depths in the Weddell Sea. The new species are characterized by a combination of morphological and molecular criteria. All three species possess a distinct oral capsule and a layer of ‘honeycomb membranes’, which form the inner part of the organic test wall. Both these features are typical of gromiids. Their identification as gromiids is confirmed by analyses of partial small subunit ribosomal DNA (SSU rDNA) gene sequences. Gromia marmorea sp. nov. is a rounded species with a prominent oral capsule and a characteristically mottled appearance. In Gromia melinus sp. nov. , the test surface exhibits a polygonal pattern of ridges, with a layer of clay particles coating the surface between the ridges. Gromia winnetoui sp. nov. represents an elongate morphotype in which the organic test is enclosed within an agglutinated case, a feature previously unknown in gromiids. Phylogenetic analysis using the maximum‐likelihood method revealed that all three species form distinct clades, reflecting the morphological differences among Weddell Sea species, as well as between deep‐water Southern Ocean Gromia and previously described gromiids. © 2009 The Linnean Society of London, Zoological Journal of the Linnean Society, 2009, 157 , 451–469.  相似文献

SYNOPSIS. Oocysts of Eimeria nieschulzi from the laboratory rat, Rattus, norvegicus , were studied by scanning and transmission electron microscopy. Oocysts had a rough outer wall with apparent random depressions. The oocyst wall is composed of 2 layers: an osmiophilic outer layer consisting of a rough external and smooth internal surface, and a relatively thick, electron-lucent inner layer. The outer layer is composed of a dense, coarsely granular matrix. The inner layer consists of homogeneous fine granular material interspersed with coarse osmiophilic granules and contains one closely applied membrane on the outermost surface. Several raised lenticular areas are seen on the coarse outer surface of the inner layer. These layers are 102 (75–128) and 176 (135–204) nm thick, respectively.
The sporocyst wall is thin, consisting of 3 to 4 unit membranes, and measures 27 (18–34) nm thick.  相似文献

SYNOPSIS. Mature macrogamonts were present in the small intestine of rats 5.5 to 7.5 days postinoculation with Eimeria nieschulzi oocysts; oocysts were present at 6 to 7.5 days. Types I and II wall-forming bodies in macrogamonts began to undergo ultrastructural changes within zygotes to form the outer and inner layers of the oocyst wall. Before and during oocyst wall formation a total of 5 membranes (M1–5) were formed at or near the surface of the zygote. The outer and inner oocyst wall layers formed between M2 and M3, and M4 and M5, respectively. The mature oocyst was loosely surrounded by M1 and M2, had an electron-dense outer layer, 100–275 nm thick, and an electron-lucent inner layer, 160–180 nm thick. It also contained an electron-lucent line consisting of M3 and M4 interposed between the outer and inner layers of the oocyst wall. The micropyle, measuring 935 × 47 nm, was located in the outer layer of the oocyst wall and consisted of 10–14 alternating layers of electron-dense and lucent material. The sporont of mature oocysts was covered by M5, immediately beneath which were M6 and M7. The sporont contained a nucleus and nucleolus, lipid and amylopectin bodies, mitochondria, ribosomes, as well as smooth and rough endoplasmic reticulum. Lepidodendralean lycopsids, a dominant component in Late Palaeozoic wetland plant communities, possess a diversity of reproductive structures that are primarily known from the Late Palaeozoic floras of Europe and North America. Here we document an anatomically preserved lepidodendralean lycopsid sporophyll with attached megasporangium from the Lower Permian Taiyuan Formation in Shanxi Province, northern China. The sporophyll has a pedicel onto which the sporangium is attached, and the sporangium is dorsiventrally flattened, proximally dehiscent and longitudinal ridged. The megasporangial wall comprises three zones: an outer uniseriate layer of columnar cells, a middle layer 1–3 cells thick comprising isodiametric parenchymatous cells, and an inner zone 1–3 cells thick of thick-walled cells. The vascular system comprises a single xylem strand surrounded by zone of parenchyma that continues through the pedicel into the lamina. Within the megasporangium a single functional megaspore and three abortive megaspores occur. Features of this specimen conform to Achlamydocarpon Schumacher-Lambry, and comparisons with other species show it shares similarities with A. takhtajanii (Sni.) Schumacher-Lambry and A. varius Taylor and Brack-Hanes. Although the morphology and anatomy of the specimen we describe overlaps with these two species, it is distinct from both leading to the erection of the new species A. intermedium sp. nov. The evolutionary significance of A. intermedium sp. nov. and the identity of its parent plant are considered, and the status and systematic position of “Oriental lepidophytes” from the Cathaysian floras are discussed.  相似文献

Phoretic stages of the exuviotrophic apostome Gymnodinioides pacifica were examined using transmission and scanning electron microscopy (TEM and SEM). TEM revealed that the mature cyst wall possesses 2 or 3 layers differing by the presence or absence of the third inner layer. This inner layer may represent a different form of the middle wall material. The inner cyst layer is approximately 0.15 microm thick and has striations with a periodicity of approximately 19 nm. The middle cyst layer has a variable thickness and the outer dense layer is approximately 0.1 microm thick. The 3 layered cyst wall had a thickness of 0.3-0.7 microm and averaged 0.5 microm. Advanced phoront stages were enclosed by fully formed cyst walls or by cyst walls thinned to approximately 0.1 microm, as the phoronts prepared to excyst prior to host ecdysis. Additionally, we report the fine structure of the rosette, trichocysts, nuclei, food plaquettes, oral fiber, and other cytoplasmic inclusions. SEM revealed an outer cyst wall layer connected to the secreted peduncle material, which was observed to extend over a wide (15 microm) area on the host setae. Parasitological examination of bearded false chameleons Chamaeleolis barbatus freshly imported from Cuba revealed the presence of 2 species of coccidia that are described as new. Oocysts of Isospora chamaeleolidis n. sp. are spherical to slightly subspherical, 16.1 (13-21) x 15.6 (13-19) microm, with a brownish and bilayered wall approximately 1.0-1.5 microm thick; outer layer markedly pitted. 0.75-1.0 microm thick. One, rarely 2, globular polar granules, 1.5 in diameter are present in the sporulated oocysts. Sporocysts are ellipsoidal, 10.8 (10-13) x 7.8 (7-9) microm, with a smooth, colorless, and unilayered sporocyst wall. Stieda body and substieda bodies are present. A sporocyst residuum is present, consisting of small granules of irregular size scattered among the sporozoites. Oocysts of Eimeria chamaeleolidisbarbati n. sp. are broadly oval, 19.0 (17-21) x 15.7 (15-17) microm, with a bilayered, colorless oocyst wall approximately 0.75 thick; outer layer of oocyst wall is smooth, 0.5 microm thick. One or 2, rarely 4, globular, irregular polar granules, approximately 1.5 microm in diameter, are present in sporulated oocysts. Sporocysts are broadly oval, 7.4 (7-8.5) x 6.1 (5.5-7) microm, with a smooth, colorless, and unilayered sporocyst wall, composed of 2 valves joined by suture; Stieda body and substieda bodies are absent.  相似文献

Progress in understanding the structure-function relationships of the mycobacterial cell wall has been hampered by its complex architecture as well as by the lack of sensitive, high-resolution probing techniques. For the first time, we used atomic force microscopy (AFM) to image the surface topography of hydrated Mycobacterium bovis bacillus Calmette Guérin cells and to investigate the influence of the antimycobacterial drug ethambutol on the cell wall architecture. While untreated cells showed a very smooth and homogeneous surface morphology, incubation of cells in the presence of ethambutol caused dramatic changes of the fine surface structure. At 4 micro g mL(-1), the drug created concentric striations at the cell surface and disrupted a approximately 8 nm thick cell wall layer, attributed to the outer electron-opaque layer usually seen by electron microscopy, while at 10 micro g mL(-1) an underlying approximately 12 nm thick layer reflecting the thick electron-transparent layer was also altered. These noninvasive ultrastructural investigations provide novel information on the macromolecular architecture of the mycobacterial envelope as well as into the destructuring effects of ethambutol.  相似文献

Live (Rose Bengal stained) and dead benthic foraminiferal communities (hard-shelled species only) from the Pakistan continental margin oxygen minimum zone (OMZ) have been studied in order to determine the relation between faunal composition and the oxygenation of bottom waters. Samples were taken from 136 m to 1870 m water depth during the intermonsoon season of 2003 (March–April). Live foraminiferal densities show a clear maximum in the first half centimetre of the sediment only few specimens are found down to 4 cm depth. The faunas exhibit a clear zonation across the Pakistan margin OMZ. Down to 500 m water depth, Uvigerina ex gr. U. semiornata and Bolivina aff. B. dilatata dominate the assemblages. These taxa are largely restricted to the upper cm of the sediment. They are adapted to the very low bottom-water oxygen values (≈ 0.1 ml/l in the OMZ core) and the extremely high input of organic carbon on the upper continental slope. The lower part of the OMZ is characterised by cosmopolitan faunas, containing also some taxa that in other areas have been described in deep infaunal microhabitats. The contrast between faunas typical for the upper part of the OMZ, and cosmopolitan faunas in the lower part of the OMZ, may be explained by a difference in the stability of dysoxic conditions over geological time periods. The core of the OMZ has been characterised by prolonged periods of stable, strongly dysoxic conditions. The lower part of the OMZ, on the contrary, has been much more variable over time-scales of 1000s and 10,000 years because of changes in surface productivity and a fluctuating intensity of NADW circulation. We suggest that, as a consequence, well-adapted, shallow infaunal taxa occupy the upper part of the OMZ, whereas in the lower part of the OMZ, cosmopolitan deep infaunal taxa have repeatedly colonised these more intermittent low oxygen environments.  相似文献

The antifungal effects of an imidazole-antimyeotic omoconazole nitrate (OMZ) on the morphology and ultrastructure of Candida albicans yeast cells were studied using scanning and transmission electron microscopy. The treatment of growing Candida cultures with fungistatic doses (0.4 to 4 μg/ml) of OMZ produced the formation of a chain or cluster of cells. Thickening of the cell wall and accumulation of electron-dense vesicles in the wall were clearly observed. Development of Golgi-like complex membranous structures in the cytoplasm was the most prominent finding. The cytological alteration induced by exposure to a higher concentration (40 μg/ml) of the drug was characterized by disruption of the intracytoplasmic organelles. Our results confirm the strong antifungal activity of OMZ against fungal cells.  相似文献

Oocysts and sporocysts of Eimeria funduli sp. n. are described from the Gulf killifish, Fundulus grandis, on the basis of light microscopy, transmission and scanning electron miscroscopy, and location in the liver of infected hosts. The spherical sporulated oocysts of E. funduli isolated from liver tissue measure 20-31 (25) micrometer across with ovoid sporocysts 9-11 X 5-7 (10 X 6) micrometer. A micropyle, polar granule, and oocyst residuum are absent, but sporocysts have Stieda and substieda bodies, a few residual granules, and 10-25 (15) unique projecting structures with expanded distal portions that we term "sporopodia". Sporopodia 1-3 (2) micrometer high support a transparent membrane that completely surrounds the sporocyst. Sporozoites have one large posterior refractile body. Ultrastructurally, the oocyst wall consists of two thin layers of granular material: an electron-dense outer layer with a rough external surface and an electron-lucent inner one of approximately equal thickness. One or two unit membranes line the inner surface of the inner layer. Each layer is 40-60 (55) nm thick. The sporocyst wall, 78-130 (110) nm thick, consists of an electron-lucent material with the outer surface being more electron dense and giving rise to osmiophilic sporopodia; closely associated with these and the outer surface are one or two unit membranes. A thin osmiophilic layer of fine granular material lines the inner surface.  相似文献

Scolecopteris Zenker, a kind of anatomically-preserved fertile foliage of Late Paleozoic Marattiales, has been well studied in Euramerican Flora. It is composed of 28 species which can be divided into four forms (groups) mainly based on modified or umodified pinnules, the variation of the outer facing sporangial walls, and with or without a prominent central parenchyma area. In contrast, Scolecopteris Zenker in Cathaysian Flora has been poorly studied so far, and only one species S. sinensis Zhao, was reported in 1991 which was considered as a member of Minor Group. The paper reports a second species of Scolecopteris, i.e.S, shanxiensis sp. nov., which differs from the above four groups in that its outer facing wall of the sporangia is thick at the base and top (2 ~ 3 layers of cells), and a little thinner ( 1~2 layers of cells) at the midlevel of the synangia. So a new group, Shanxiensis group, is set for the new species. The other characteristics of Shanxiensis group is comparable with Minor group. The new species comes from the coal balls in Coal Seam No. 7 in the upper part of Taiyuan Formation (early Early Permian) from Taiyuan, Shanxi, China. The identification of Scolecopteris shanxiensis sp. nov. :The fertile pinnule probably peeopterids, 5.5 ~ 6.0 mm in length and 2.0 ~ 2.2 mm in width. The lateral extensions of the lamina of the pinnule bend abaxially and above the synangia. The synangia arrange along the sides of the midrib of the pinnnle and there are about 10 synangia in each row. The synangium is elliptical in longitudinal section and radial in cross section, 0.7 ~ 0.8 mm in height and 0.6 ~ 0.7 mm in diameter. Each synangium has 5-7 (mostly 6) fusiform sporangia fused at the base and attached to the top of the synangial pedicel. The outer facing wall of the sporangia consists of 2~3 layers of cells at the base and becomes thinner at the midlevel (1~2 layers of cells), and at the top of the synangia the wall become thicker again. The cells of the outer facing wall of the sporangia are elongate in the longitudinal sections. The lateral and inner facing walls of the sporangia are one cell thick. The synangial pedicel is small. Spores in situ are small, generally 11~14 µm in diameter, spherical or rounded-triangular, trilete and smooth-walled.  相似文献

Summary Glucuronoxylans (GXs), the main hemicellulosic component of hardwoods, are localized exclusively in the secondary wall of Japanese beech and gradually increase during the course of fiber differentiation. To reveal where GXs deposit within secondary wall and how they affect cell wall ultrastructure, immuno-scanning electron microscopy using anti-GXs antiserum was applied in this study. In fibers forming the outer layer of the secondary wall (S1), cellulose fibrils were small in diameter and deposited sparsely on the inner surface of the cell wall. Fine fibrils with approximately 5 nm width aggregated and formed thick fibrils with 12 nm width. Some of these thick fibrils further aggregated to form bundles which labelled positively for GXs. In fibers forming the middle layer of the secondary wall (S2), fibrils were thicker than those found in S1 forming fibers and were densely deposited. The S2 layer labelled intensely for GXs with no preferential distribution recognized. Compared with newly formed secondary walls, previously formed secondary walls were composed of thick and highly packed microfibrils. Labels against GXs were much more prevalent on mature secondary walls than on newly deposited secondary walls. We describe two new pillotinaceous spirochetes (Canaleparolina darwiniensis, Diplocalyx cryptotermitidis) and identify for the first time Hollandina pterotermitidis from both the subterranean termite Cryptotermes cavifrons and the wood-eating cockroach Cryptocercus punctulatus based on morphometric analysis of transmission electron micrographic thin sections. C. darwiniensis, gen. nov., sp. nov., limited to near Darwin, Australia, invariably is present on the surface of the treponeme-studded trichomonad Mixotricha paradoxa, a consistent inhabitant of the hindgut of healthy termite Mastotermes darwiniensis. The spirochete both attached to the surface of protists and free-swimming in the paunch (hindgut) lumen of the insect has 16 periplasmic flagella (16:32:16) and imbricated wall structures that resemble flattened crenulations of Pillotina. The flagella surround half the protoplasmic cylinder. C. darwiniensis is the largest (0.5 microm diameter x 25 microm length) of the three epibiotic bacteria (two spirochetes, one rod) that comprise the complex cortex of its host Mixotricha paradoxa. Several criteria distinguish Diplocalyx cryptotermitidis sp. nov. isolated from Cryptotermes cavifrons intestine: smaller diameter, fewer flagella, absence of inner and outer coats of the outer membrane, wider angle subtended by its flagella and, most notably, cytoplasmic tubule-associated centers, which are periodic electron dense spheres within the protoplasmic cylinder from which emanate cytoplasmic tubules up to 24 nm in diameter. This is also the first report of abundant populations of Hollandina in Cryptotermes cavifrons (those populations belong to the species H. pterotermitidis). Morphometric analysis of the first thin sections of any spirochetes (published nearly 40 years ago by A.V. Grimstone) permits us to identify the large (0.9 microm diameter) free-swimming intestinal symbiont of Cryptocercus punctulatus also as Hollandina pterotermitidis.  相似文献

Abstract The mycobacterial cell wall visualized by transmission electron microscopy (TEM) of thin sections of resin-embedded specimens is generally believed to consist of an electron-dense peptidoglycan, an electron-transparent arabinogalactan-mycolate layer and an electron-dense outer layer (OL). In addition, a pseudocapsule known as the ‘electron-transparent zone’ (ETZ) has been observed after phagocytosis of mycobacteria by macrophages. TEM of thin sections of Mycobacterium bovis BCG, Tice® substrain, revealed an OL bilayer, each of which measured 2–4 nm in diameter. The intermediate electron-transparent layer varied from 1 to about 250 nm in diameter and appears to be a previously observed oxygen-dependent amorphous integument that consists of hot water-extractable neutral polysaccharides, especially a recently characterized α-glucan, comprising about 12% of the dry cell weight. This and other recent studies of BCG have revealed cell-surface features that may provide a better understanding of the outer mycobacterial cell envelope.  相似文献

对全球大洋氮循环的研究发现,大洋输入和输出的氮存在严重的不平衡,所固定的氮中有相当一部分被还原为N2或N2O从大洋中流失,而海洋最小含氧带(OMZ)被认为是发生氮流失的最主要区域,通过反硝化作用和厌氧氨氧化作用,固定氮在OMZ海区内损失量可达40~450 Tg·a-1.对不同海区OMZ内固定氮损失的两种主要作用总结发现,异养反硝化作用在热带太平洋东部、阿拉伯海的OMZ内以及海洋沉积物内占有显著优势,在智利、秘鲁沿岸海域及阿拉伯海域也已发现自养反硝化作用的存在;而在黑海、非洲西南部的本格拉上升流、智利北部沿岸等地,厌氧氨氧化作用强烈,且其在陆架区的作用强度和面积要大于大洋区.OMZ氮的流失除受氮流失过程自身影响外,固氮作用、硝化作用、硝酸盐异化还原作用等都可能对OMZ海区内氮收支不平衡造成影响.其中固氮作用的影响最不能忽视,其在全球OMZ内固定的氮的总量可达15~40 Tg·a-1,是对OMZ氮流失量的重要补充.区分反硝化作用和厌氧氨氧化作用对OMZ氮流失的相对贡献,明确氮流失的另一产物N2O的形成机制和定量评估方法是当前OMZ氮流失研究中存在的最主要问题.本文针对存在问题提出了相应的研究设想,以期为海洋最小含氧带的研究提供参考.  相似文献

SYNOPSIS Oocysts of Eimeria crotalviridis sp. n. are described from prairie rattlesnakes, Crotalus viridis viridis in New Mexico on the basis of light and electron microscopy and in vitro excystation of sporozoites. Sporulated oocysts of E. crotalviridis are elliptical, 26.4 × 22.3 (23–29 × 20–24) μm with ovoid sporocysts 11.7 × 8.1 (11–13 × 7–9) μm. A micropyle, micropyle cap and polar bodies are absent, but oocyst and sporocyst residua and Stieda and substieda bodies are present. Excysted sporozoites are 12.4 × 2.8 (11–13 × 2–3) μm and have 1 large posterior refractile body and a nucleus with a prominent nucleolus. Ultrastructurally, the oocyst wall has 2 layers, a thick, electron-dense, highly sculptured outer layer composed of a fine granular matrix and a thin, granular, osmiophilic inner layer, separated from the outer layer by at least one unit membrane. These layers are 441 (353–510) and 21.6 (19–29) nm thick, respectively. Within 15 min after exposure to a trypsin-sodium taurocholate fluid, sporozoites of E. crotalviridis excysted from 5-month-old sporocysts.  相似文献

The fine structure of the mature macrogamonts and intracellular oocysts of Eimeria labbeana from the ileal mucosa of experimentally infected Pigeons (Columbia livia) was investigated and described. The macrogamont reached a maximum size of 12.0 x 9.5 mum (average equals 10.8 x 8.8 mum), and was located within a narrow parasitophorus vacuole. Most of the macrogamonts were limited by two membranes. Intravacuolar tubules, 1.2 mum long and 58 nm in diameter, established direct connections between the parasite and the host cell. Each tabule was composed of 9 subunits arranged around the central lumen. Cytoplasmic canaliculi were composed of bundles of microtubule-like structures (8-10 nm wide). Type 1 wall-forming bodies reached a maximum size of 1.8 x 1.5 mum, and many had centric or eccentric electron transparent portions within them. They were frequently seen lodged within peripherally-located mitochondria. Type 2 wall-forming bodies averaged 1.5 mum in diameter. The role of the two types of wall-forming bodies in forming the outer and inner layers of the wall of the oocyst was similar to that in other species of Eimeria. The oocyst wall was 0.2 mum thick and composed of a limiting membrane (20 nm thick), an outer layer (75 nm thick), and an inner layer (100 nm thick).  相似文献

文中对贵州瓮安埃迪卡拉纪陡山沱组磷块岩中一类具螺旋孔的微体球形化石进行形态功能学分析,并与原生动物肉足虫纲的类群进行比较。结果显示它们是一个独立的分类单元,属于单细胞原生动物,并命名为瓮安旋孔虫(新属、新种)(Helico foramina wenganica gen.et sp.nov.),与已报道的胚胎化石很可能没有亲缘关系。虽然这类化石生物很可能属于原生动物肉足虫纲,但还不能归入其已知的目,也许属于新的目一级分类单元。螺旋孔是伪足伸出的孔洞,它们起到了取食、运动和排泄的作用。该类化石生物的个体生长是通过阶段性地脱去外部壳体、再由原生质体分泌出新壳体的方式来实现的。  相似文献

Living Entamoeba histolytica trophozoites were examined by phase-contrast microscopy. Intact critical point dried trophozoites were examined by transmission electron microscopy at an accelerating voltage of 1000 kV (HVEM) and by scanning electron microscopy (SEM). Half and quarter m? thick sections of epoxy-embedded trophozoites were examined by HVEM. Many of the trophozoites of 2 strains examined had surface filopodia, 1 to over 100 pan in length. The cytoplasm of filopodia was continuous with the cytoplasm and bounded by surface plasmalemma bearing a glycocalyx. Structures called “surface-active lysosomes with trigger,”“dendritic plasmalemmal extensions,” and “extra-amebic vesicles” by previous investigators probably represent portions of filopodia demonstrated in the present study. Filopodia appear to be of frequent normal occurrence in E. histolytica and may function in: (a) endocytosis or pinocytosis; (b) exocytosis; (c) attachment to substratum; (d) penetration of tissue; (e) release of cytotoxic substances; or (f) contact cytolysis of host cells.  相似文献
