Well-nodulated soya bean (Glycine max L.) plants were allowedto assimilate 13CO2 for 10 h in the light, under steady-stateconditions in which CO2 concentration and 13C abundance wereboth strictly controlled at constant levels. The respiratoryevolution of 13CO2 from roots and nodules and 13C incorporationinto various metabolic fractions were measured during the 13CO2feeding and subsequent 48 h chase period. CO2 respired from nodules was much more rapidly labelled with13C than that from roots. The level of labelling (percentageof carbon currently assimilated during the 13COM2 feeding period)of CO2 respired from nodules reached a maximum of about 87 percent after 4 h of steady-state l3CO2 assimilation and thereafterremained fairly constant. The absolute amount of labelled carbonevolved by the respiration of the nodules during the 10 h 13CO2feeding period was 1·5-fold that of root respiration.These results demonstrated that the currently assimilated (labelled)carbon was preferentially used to support nodule respiration,while root respiration relied considerably on earlier (non-labelled)carbon reserved in the roots. Sucrose pools were mostly composed of currently assimilatedcarbon in all tissues of the plants, since the levels of labellingaccounted for 86–91 per cent at the end of the 13CO2 feeding.In the nodules, the kinetics and levels of sucrose labellingwere in fairly good agreement with those of respired CO2, whilein the roots, the level of labelling of respired CO2 was significantlylower than that of sucrose. Succinate and malate were highly labelled in both roots andnodules but they were labelled much more slowly than sucroseand respired CO2. The kinetics and levels of labelling of theseKrebs cycle intermediates resembled those of major amino acidswhich are derived directly from Krebs cycle intermediates. Itis suggested that large fractions of organic acids in noduleswere physically separate from the respiration site. Three methods were used to study N2 fixation and effects ofwater deficit on N2 fixation: C2H2 reduction assay (ARA), 15Ndilution technique and accumulated N content. In addition, 15Ndilution was calculated both in a traditional way and in a modifiedway, which takes into consideration N and 15N content for theplants before the experiment started. The three methods wereapplied on the following Rhizobium-symbioses: Acacia albidaDel (Faidherbia albida (Del) A. Chev.) and Leucaena leucocephala(Lam) de Wit., and the Frankia-symbiosis Casuarina equisetifoliaL. The plants wereabout 4-months-old when they were harvested. Nitrogen derived from N2 fixation in control plants of Acaciaalbida was 54·2 mg as measured with ARA, while it was28·5 mg as measured with the 15N dilution technique,compared to 30·7 mg calculated as accumulated N. In comparison,L. leucocephala fixed 41·6 mg N (ARA), 53·5 mgN(15N dilution technique) and 56·3 mg N (accumulatedN). The Frankia-symbiosis had fixed 27·4 mg N as measuredby ARA, 8·1 mg N as measured by 15N dilution techniqueand 12·3 mg N as accumulated N. There were no differencesbetween the estimates based ontraditional and modified waysof calculating 15N dilution. The immediate effect of water deficit treatment on N2 fixationwas continuously measured inall species with ARA, which startedto decrease approximately 10 d after the initiation of the treatment,and declined to less than 5% of the initial level after 21–28d. The decrease in the amount of N derived from N2 fixation wasstudied in L. leucocephala during the period of treatment. Therewas a 26% decrease in amount of N derived from N2 fixation asresult of water deficit (as measured with ARA), while the decreasewas 23% when measured withboth the 15N dilution method and asaccumulated N. The three different methods for measuring N2 fixation and effectsof water deficit on N2 fixation are discussed. A comparative study of the metabolism of Na2 75SeO4 in Astragalusbisulcatus, luma bean, and wheat has been carried out. The resultsindicate that all three plants metabolize selenium extensively.Important differences were observed in the distribution of radioactivitybetween the various fractions isolated from the plants. Comparedto the protein fraction, the free amino acid fraction from A.bisulcatus contained a higher percentage of radioactivity. Theconverse was true for wheat and lima bean. As A. bisulcatusproteins contained a significant percentage of radioactivity,it is suggested, that the differences in the toxicity of seleniumtowards wheat, lime bean, and A. bisulcatus are difficult toexplain in terms of the differences in its incorporation intothe protein of the three species.  相似文献

Ammonia Induces Starch Degradation in Chlorella Cells   总被引:3,自引:0,他引:3  
When ammonia was added to cells of Chlorella which had fixed14CO2 photo synthetically, 14C which had been incorporated intostarch was greatly decreased. A similar effect was observedwhen potassium nitrate and sodium nitrite were added. The ammonia-induceddecrease in 14C-starch was observed in all species of Chlorellatested. With cells of C. vulgaris 11h, most of the radioactivityin starch was recovered in sucrose, indicating that ammoniainduces the conversion of starch into sucrose. The percent of14C recovered in sucrose differed from species to species andpractically no recovery in sucrose was observed in C. pyrenoidosa.In most species tested, the enhancing effects of blue lightand ammonia on O2 uptake as well as the ammonia effect on starchdegradation were greater in cells which had been starved inphosphate medium in the dark than in non-starved cells. In contrast,the enhancing effect of ammonia on dark CO2 fixation was muchgreater in non-starved cells. C. pyrenoidosa was unique in thatblue light did not show any effect on its O2 uptake. Growth and N-accumulation rates in leaves, stolons and rootsof individual white clover plants were studied in three experimentsusing two methods. In a growth chamber experiment, the relativedifferences between tissues were found to be almost constantfor a wide range of clover plant sizes. The stolon dry matter(DM) production was 56% and the root DM production 40% of theDM production in leaves. The N yield of stolons was 30% whileN yield in roots was 34% of N yield in leaves. The effect ofN application on these relations was investigated in a glasshouseexperiment. Application of N reduced the root:shoot N ratiofrom 0.50 to 0.28, whereas the stolon+root:leaf N ratio (i.e.for abovevs.below cutting-height tissues) was only reduced from0.97 to 0.80. In a field trial with two contrasting N regimes,growth and N accumulation were measured on individual cloverplants. Dinitrogen fixation was estimated by15N isotope dilutionbased on analysis of leaves-only or by including stolons. Usingleaves-only did not affect the calculation of percentage ofclover N derived from N2fixation (% Ndfa) since the15N enrichmentwas found to be uniform in all parts of the clover. Two membrane fractions were obtained from 16%/26% and 34%/40%interfaces following discontinuous sucrose density gradientcentrifugation of a 10,000–80,000xg pellet from mung bean(Phaseolus mungo L.) roots. The ATPases in the fractions differedfrom each other in their sensitivity toward various inhibitors,activation with salts, dependence of activity on pH, and Kmfor ATP.Mg2+. Judging from their sensitivity toward inhibitors,the ATPases in the low and high density membranes are consideredmainly of tonoplast and plasma membrane origin, respectively.Both ATPases were activated by gramicidin D and nigericin. ATP-inducedquenching of quinacrine fluorescence in both fractions requiredMg2+ and permeant anions such as Cl and quenching wascollapsed by carbonylcyanide p-trifluoromethoxyphenyl hydrazone.The sensitivities of quenching to the inhibitors were essentiallythe same as those of ATPase activity in the membranes. A closed-system flow-through enclosure apparatus was used tomeasure symbiotic nitrogen fixation directly. A legume-basedsystem comprising 6-week-old Trifolium repens L. (white clovercv. Blanca) growing with Lolium perenne L. (perennial ryegrasscv. Trani) in an agricultural soil was incubated for 19 d ina 15N-enriched atmosphere (mean value 3.663 atom%). An actinorhizal-basedsystem comprising 1 -year-old Alnus glutinosa L. (alder) saplingsgrowing with Festuca rubra L. (red fescue) in open-cast coalspoil was incubated for 21 d in a 15N-enriched atmosphere (meanvalue 3.265 atom%). Indirect estimates of N2 fixation were carriedout concurrently using N difference and 15N isotope dilutiontechniques. The theory underlying the three techniques and modificationswhich were adopted for comparative purposes are discussed. Thedirect measurements of N2 fixation were then compared with theindirect estimates using Pinc, the proportion of the N incrementduring the measurement period that was derived from fixation.The simple N difference method gave similar values for Pinc(0.94 and 0.97) as those derived from more complicated isotopemethodologies, both indirect (0.91) and direct (0.90). Valuesfor alder were far more variable, ranging from 0.16 to 0.92;this was due largely to variability within the trees and a verysmall N increment during the measurement period. Barley plants were grown in nutrient solution at two contrastingnitrate concentrations to produce plants of low or high nitrogen(N) status. Leaves were then exposed continuously to either0.3 mm3 dm–3 NO2 or clean air, with the roots and rootingmedium isolated from the polluted air. Uptake of NO2 was measuredin two ways; as depletion from an air stream containing thegas and using 15N-labelled NO2. Results from the two methodsagreed well and demonstrated that the flux of NO2 into the leavesof N-deficient barley was lower than that of N-sufficient plants.Nevertheless, the relative contribution of15N derived from 15NO2to the N status of the plant was greater in the plants suppliedwith low nitrate. A major factor in regulating NO2 uptake bybarley leaves appeared to be stomatal conductance, althoughinternal conductance may also be involved. The effects of NO2exposure of barley on carbon dioxide exchange rates, transpirationand water vapour conductance were also influenced by the N statusof the plant. Nitrogen-fixing activity in two nodulated African legumes, Bambaragroundnut (Vigna subterraneaL.) and Kersting's bean (MacrotylomageocarpumL.), was assessed in the presence of nitrate (NO3-)ions in the rooting medium. Nitrogenase activity was unimpairedby the supply of 5 mol m-3NO3to both species. Also, large concentrationsof ureides dominated the transpiration stream of NO3-fed plants.Compared to other symbiotic legumes cultured with similar NO3concentrations,nodule functioning in the tested landraces of Bambara groundnutand Kersting's bean is tolerant of NO3ions in the rhizosphere.The potential benefits of such naturally occurring NO3-tolerantsymbioses are substantial, as they would permit inorganic Nfertilizer application in intercropping systems without inhibitingN2fixation in the associated legumes.Copyright 1998 Annals ofBotany Company NO3tolerance, Bambara groundnut, Kersting's bean, nitrogenase activity, xylem ureides.  相似文献

Photosynthetic carbon metabolism was studied with Chroomonassp. cells in which the rate of photosynthesis was inhibitedunder both an anaerobic condition and high concentrations ofoxygen. The time course of 14C-incorporation into photosyntheticproducts showed that 3-phosphoglycerate was the initial productof photosynthetic CO2 fixation in Chroomonas sp. cells. During5-min photosynthesis, a considerable amount of 14C was incorporatedinto the insoluble fraction (mostly cryptomonad starch), andoxygen predominantly affected 14C-incorporation into this fraction.Although 14C-incorporation into intermediates of the photorespiratorypathway increased with increasing O2 concentration, the amountswere much less than expected from the degree of oxygen inhibition.It is noteworthy that 14C-dihydroxyacetone phosphate accumulatedduring photosynthesis only under the anaerobic condition, whereasthe levels of the other phosphate esters were scarcely affectedby the oxygen concentration. Ribulose-1,5-bisphosphate carboxylase from Chroomonas sp. wascompetitively inhibited by oxygen, and its Km(CO2) value wassimilar to those of terrestrial C3 plant enzymes. In rice, alday, wheat and tobacco (Nicotiana tabacum L. Samsunand Samsun NN) plants which contained large amounts of ABA,the transpiration rate decreased rapidly with 2 ppm SO2 fumigationand reached 20 to 65% of the initial level after 5- to 30-minexposure depending on their ABA contents. In the cases of broadbean and tobacco (N. glutinosa L.) with low ABA contents, therate slightly increased for 20 and 40 min, respectively, afterthe start of the fumigation and then decreased gradually. Thetranspiration rates of corn and sorghum, in spite of their extremelylow ABA contents, pronouncedly decreased with SO2 fumigationand reached 65 and 50%, respectively, of the initial levelsafter 40-min exposure. Foliar application of 0.04 N HC1 to N.tabacum L. Samsun NN leaves remarkably depressed the transpirationrate, while the application of 0.04 M Na2SO3 decreased the rateonly to the same level as water treatment. Foliar applicationof either HCl or Na2SO3 to N. glutinosa L. leaves exerted littlechange in the transpiration rate. When 10–4M ABA was appliedto broad bean leaves prior to HCl and Na2SO3 treatment, theirtranspiration rate was decreased by HCl, but not by Na2SO3 application.In sonicated epidermal strips peeled from broad bean leaves,Na2SO3 produced a slight increase in the stomatal aperture sizein the absence of ABA, but showed no effect in the presenceof ABA. The aperture size was identical in the pH range of 3.0to 7.0 in the incubation medium. In the presence of ABA in themedium, the aperture size was small in the acidic region ofpH with a minimal value at pH 4.0. ABA decreased the aperturesize at concentrations above 10–9 M at pH 4.0 and 10–6M at pH 7.0 in the medium. [2–14C] ABA uptake by epidermalstrips was large in the acidic region, especially at pH 4.0. An investigation was made to study the assimilation and transportof 15N-labelled nitrate nitrogen in rice plant (Oryza sativaL.). Nitrogen from labelled nitrate at the end of plant feedingwas found mainly in nitrate form, and was more prevalent inroots, stem and leaf sheaths. The nitrite fraction had the nextlargest 15N enrichment. The 15NO3 assimilation in the newlyemerged panicle was mainly in amide and amino acid. The 15N-incorporation at day 0 was greatest in amino acid andnitrate of roots and decreased towards the stem and leaves.Incorporation in these fractions considerably decreased fromday 0 to day 10. Probably most of the nitrogen from the nitratesource was transported from the roots to the shoot in nitrateand amino acid forms. A decrease of 15N-incorporation in the soluble N fraction andincrease in the insoluble N fraction from day 0 to day 10 inplant parts, particularly the blades, suggested that proteinsynthesis occurred mostly in young parts of the shoot duringthis period. The marked variation in 15N distribution in differentparts of the plant during the 10 days indicated that the nitrogenin roots and tillers was probably remobilized and transportedto other parts, particularly the upper leaf blades. Cyclopenin (C17H14O3N2) and cyclopenol (C17H14O4N2), isolatedfrom an abberent strain of Penicillium cyclopium (NRRL 6233),significantly inhibited the growth of etiolated wheat (Triticumaestivum) coleoptile segments. The former inhibited at 10–3and 10–4 M, the latter at 10–3 M. Cyclopenin producedmalformation of the first set of trifoliate leaves in bean (Phaseolusvulgaris) at 10–2 M and necrosis and stunting in corn(Zea mays) at 10–2 M. Cyclopenol induced no apparent effectsin bean or corn plants. Neither compound changed the growthor morphology of tobacco (Nicotiana tabacum) plants. Cyclopenininduced intoxication, prostration and ataxia in day-old chicksat 500 mg/kg, but they recovered within 18 hours. Cyclopenolwas inactive against chicks when dosed at levels up to 500 mg/kg. Cultures of the water fern Azolla pinnata R, Br. exposed for1 week to atmospheric NO2 (50, 100 or 200 nl l-1) induced additionallevels of nitrate reductase (NaR) protein and nitrite reductase(NiR) activity. At low concentrations of NO2 (50 nl l-1), nitratederived from NO2 provides an alternative N source for Azollabut does not affect rates of acetylene reduction. However, thesymbiotic relationship between Azolla and its endosymbiont,Anabaena azollae is only affected adversely by high concentrations(100 and 200 nl l-1) of atmospheric NO2. The resultant decreasesin rate of growth, nitrogen fixation, heterocyst formation,and overall nitrogen cycling are probably due to the additionalaccumulation of N products derived from higher levels of atmosphericNO2. Sixteen legumes were grown in N-free media so that N was suppliedentirely by symbiotic N2 fixation. The plant tissues were analyzedfor natural 15N abundance (expressed as 15N per mil relativeto air N2) with a ratio mass spectrometer. The nodules of desmodium,centro, siratro, soybean and winged bean showed high enrichmentin 15N (+9), while red clover showed slight enrichment (+2).The nodules of 9 other forage legumes (Townsville stylo, whiteclover, alsike clover, common vetch, Chinese milk vetch, senna,alfalfa, ladino clover, and hairy vetch) showed little enrichmentin 15N. In all the legumes investigated, particularly in the ureide-transportingplants such as desmodium, centro, siratro, soybean, winged beanand field bean, the 15N value of the shoots was negative (–3.2).The 15N value of the shoots in winged bean and field bean variedby about 1 depending on the Rhizobium strains used. The isotopicmass balance of 13 legumes indicated that isotopic fractionationoccurs during N2 fixation by the legume-rhizobia symbiosis witha preference for 14N over 15N, resulting in a 15N value of –0.2to –2 in the whole plant. Carbonic anhydrase (CA) activity was detected in homogenatesfrom Anabaena variabilis ATCC 29413, M-2 and M-3, but not inthe suspension of the intact cells. Activity was higher in cellsgrown in ordinary air (low-CO2 cells) than in those grown inair enriched with 2–4% CO2 (high-CO2 cells). Fractionationby centrifugation indicated that the CA from A. variabilis ATCC29413 is soluble, whereas both soluble and insoluble forms existin A. variabilis M-2 and M-3. The addition of dithiothreitoland Mg2 $ greatly decreased the CA activity of A. variabilisATCC 29413. The specific activity of the CA from A. variabilis ATCC 29413was increased ca. 200 times by purification with ammonium sulfate,DEAE-Sephadex A-50 and Sephadex G-100. Major and minor CA peaksin Sephadex G-100 chromatography showed respective molecularweights of 48,000 and 25,000. The molecular weight of the CAdetermined by polyacrylamide disc gel electrophoresis was 42,000?5,000.The activity of CA was inhibited by ethoxyzolamide (I50=2.8?10-9M), acetazolamide (I50=2.5?10-7 M) and sulfanilamide (I50=2.9?10-6M). Daucus carota tissues were grown on Murashige-Skoog medium (MS)at different concentrations with abscisic acid (ABA). Sevenbands of chloroplast fractions were obtained on a sucrose gradient.At 10–5M, ABA highly increased chlorophyll and proteinnitrogen content of medium density chloroplasts. With increasingage of the tissues, the most active chloroplasts according totheir 14CO2 fixation were found in smaller numbers. When treatedwith 10–5 M ABA, 34 day-old tissues cultivated in vitroshowed the chloroplast pattern of 110 day-old tissues. The effectof ABA—given to the tissues during a short pretreatmentor continuously present in the culture medium—on the ribulosediphosphate carboxylase activity was analysed. Salinity Reduces Water Use and Nitrate-N-use Efficiency of Citrus   总被引:1,自引:0,他引:1  
Five-month-old Cleopatra mandarin (Citrus reticulata Blanco)(CM) and Volkamer lemon (Citrus volkameriana Ten. and Pasq.)(VL) seedlings were grown in a glasshouse in 2·3-1 potsof Candler fine sand. Plants were irrigated with either non-saline(ECe = 0·23 dS m-1) or saline (6·13 dS m-1) waterusing 3:1 NaCl:CaCl2 solution over a 4-week period. A singleapplication of K15NO3 (19·64 atom % excess 15N) at 212mg N1-1, was substituted for a normal weekly fertilization after3 weeks and plants were harvested 7 d later. The transpirationrate, uptake of nitrogen, growth and nitrogen-use efficiency(NUE) on a dry weight basis (mg d. wt mg-1 N) of both specieswas reduced by salinity. Based on growth, water-use and chloride(Cl) accumulation in leaves, VL was more salt-sensitive thanCM, but 15N uptake was equally reduced by salinity in both species.Salinity reduced 15N uptake relatively more than shoot growthover the 7-d period, such that the 15NUE (mg d. wt µg-115N) of new shoot growth of both species increased. There wasno evidence of Cl antagonism of nitrate (NO3) uptake but totalplant 15NO3 uptake was positively correlated with whole planttranspiration in both species. Nitrogen fixation activity in common bean is generally thoughtto be low and sensitive to soil drying and, consequently, droughtcan have important negative effects on N accumulation and yieldpotential. The objectives of this research were to examine theresponse of N2fixation to drought stress in common bean, andto test the hypothesis that drought sensitivity of N2fixationin common bean is linked to ureide levels in the plants. Twoglasshouse experiments were conducted to compare the responsesof leaf transpiration and acetylene reduction activity (ARA)to soil water contents. ARA decrease during soil dehydrationwas found to lag behind the decline in transpiration. This indicatesthat ARA is relatively less sensitive to soil dehydration comparedto leaf gas exchange. Further, in comparing two cultivars therewas no consistent difference in the relative response of ARAand transpiration to soil drying. The ureide concentrationsmeasured in common bean plants were low, ranging from 0.1 to1.0 mmol l-1in xylem sap exudates. Komatsuna (Brassica campestris L. var. rapa) plants were grownhydroponically under various conditions with respect to thesupply of nitrate, and the variations in levels of natural 15Nabundance (15N) in nitrogenous fractions of leaf blades, petiolesplus midribs, and roots in these plants were analyzed. The fractionation of nitrogen isotopes during uptake of nitratewas null irrespective of the concentrations of nitrate in theculture medium. The roots had lower 15N values than that inthe nitrate applied to plants. The nitrate in the three tissuesexamined had higher 15N values than that in the nitrate applied:the values were highest in the leaf blades which were presumedto have highest activities in terms of reduction of nitrate.In contrast, the amino acids and residual fractions had lower15N values than those in the nitrate applied.
