The effects of starch granules on the rheological behaviour of gels of native potato and high amylopectin potato (HAPP) starches have been studied with small deformation oscillatory rheometry. The influence of granule remnants on the rheological properties of samples treated at 90 °C was evident when compared with samples treated at 140 °C, where no granule remnants were found. The presence of amylose in native potato starch gave to stronger network formation since potato starch gave higher moduli values than HAPP, after both 90 and 140 °C treatments. In addition, amylose may have strengthened the network of HAPP because higher moduli values were obtained when native potato starch was added to the system. The moduli values of the mixtures also increased with increasing polysaccharide concentration in the system, which is due to an increment in the polysaccharide chain contacts and entanglements. Finally, it was found that a mixture of commercial amylose from potato starch and HAPP resulted in lower values of G′ compared to native potato starch. This indicates that the source of amylose is important for the properties in a blend with native amylopectin.  相似文献

Summary TheZymomonas fermentation for industrial ethanol production has been successfully scaled up. Pilot plant experiments at 100 and 1,000 litre fermentation capacity gave 91–95% conversion efficiencies and up to 10% (v/v) ethanol yields within 17–20 hours using sugar cane syrup, A-, B-, and C-molasses with the addition of sucrose or syrup to a final 15% total sugar concentration.  相似文献

High fructose recovery yields were obtained using sugarcane syrup and C-molasses (equal to blackstrap molasses) and a fructokinase negative mutant ofZymomonas mobilis. The fructose recovery was 95.7% with sugarcane syrup and 99.4% with 300 g/L C-molasses or mixtures of both. High fructose corn syrup of a 48/52 mixture of glucose and fructose gave only a 65–70% fructose recovery due to high sorbitol formation.  相似文献

Summary Fructokinase negativeZymomonas mobilis UQM 2864, was co-immobilised with invertase in alginate and incubated on sucrose-based media in batch and fedbatch culture. The highest fructose concentration achieved was 138 g/l using fed-batch cultivation with sugar-cane syrup-simultaneously producing 79.9 g/l or 10.1% (v/v) ethanol in less than 24 hours. The ethanol and fructose yields were 95 and 84% respectively. Co-immobilisation resulted in faster fermentation times, particularly for the batch fermentations, and complete utilisation of substrate.  相似文献

Rapid visco analysis (RVA) was performed to study the pasting properties of mixtures of wheat flour and tuber starches, i.e., potato starch (PS), sweet potato starch (SPS), yam starch (YS), and cassava starch (CS), at 10–50% starch in the mixtures. Lower phosphorus and higher amylose contents were observed in CS, followed by YS, SPS, and PS. The peak, breakdown, final, and setback viscosities of the control wheat flour were lower than those of the control PS, SPS, YS, and CS. The peak viscosity of wheat–PS mixtures was higher than those of the wheat–SPS, wheat–YS, and wheat–CS because of the higher phosphorus and lower amylose content of PS, which resulted in higher swelling of PS than that of SPS, YS, and CS. The breakdown viscosities increased as the starch content of the PS, SPS, and CS in the mixtures increased to up to the 50%, and the values tended to decrease in the wheat–YS mixture. The setback viscosities of wheat–SPS, wheat–YS, and wheat–CS increased significantly as the starch content increased from 10% to 50%, and that of wheat–PS dropped dramatically at 50%. The findings in this work provide evidence that tuber starches could be used as a partial substitute for wheat flour in some wheat-based products.  相似文献

Wheat and potato starches were hydrolysed with 2·2 n hydrochloric acid at 35°C for a period of time up to 15 days. The residues (lintnerised starches) were washed and freeze dried, and studied by differential scanning calorimetry (DSC), wide-angle X-ray scattering (WAXS), small-angle light scattering (SALS), small-angle neutron scattering (SANS) and small-angle X-ray scattering (SAXS). These techniques showed that profound changes took place in the first day of hydrolysis (during which time the extent of hydrolysis was 7·7% for potato starch and 12·5% for wheat starch). In particular, the gelatinisation enthalpy (ΔH) decreased, the X-ray crystallinity increased and the SANS and SAXS peaks (indicative of a regular spacing between crystalline and amorphous regions) virtually disappeared. The reduction in ΔH is surprising and is discussed at length. It was also shown that freeze drying results in a considerable lowering of the gelatinisation temperature of potato starch (and also of ΔH) while that of wheat starch is only slightly affected.  相似文献

Traditional fermentation of paddy malt mash (containing 18.1% w/v dextrose equivalent) to paddy arrack using paddy husk as source of inoculum yielded very low level of ethanol (4.25% v/v). Use of yeast isolates obtained from paddy husk as well as a potent ethanol producer like Zymomonas mobilis ZM4 and their combinations in the fermentation revealed that a combination of an yeast isolate PH 03 (Saccharomyces cerevisiae) and Z. mobilis ZM4 produced synergistically and statistically more ethanol (10.1% v/v) than the individual and other combination of cultures. In this process, addition of penicillin G at a concentration of 20 U/ml rather than heat sterilization, helped retention of the limited amylase activity in the mash for simultaneous saccharification and fermentation over 7 d at 30°C. About 98.5% of the carbohydrate was accountable in the fermentation which yielded 86.7% of the theoretical yield of ethanol, apart from biomass and acids.  相似文献

The goal of this research was to study the effect of the substitution of wheat starch by potato starch (PS) on the performance, health and digestion of growing rabbits. Three experimental diets were formulated with 0%, 7% and 14% PS (PS0, PS7 and PS14, respectively) and similar starch contents (22% dry matter basis), proteins and fibre. The three diets were administered to three groups of 48 rabbits from weaning (28 days) to slaughter (70 days), and growth and health measurements were made. Another 10 rabbits per diet (30 rabbits at each age), reared under similar conditions, were slaughtered at 6 to 10 weeks of age, and the digesta were collected to analyse the caecal microbial activity (pH, volatile fatty acids (VFA) levels, fibrolytic activity) and the starch concentration in the ileal digesta. At the same ages, the whole tract digestibility coefficients were measured in 10 other rabbits for each treatment (30 rabbits). The feed intake between 28 and 42 days of age (days) increased by 11% (P < 0.05) in PS0 v. PS14. Over the whole growth period (28 to 70 days), weight gain was similar among diets (40.5 g/day), whereas the feed intake and feed conversion increased (8.5% and 5.2%, respectively; P < 0.05) with the PS14 diet. Mortality and morbidity were not affected by the diets. The starch concentration of the ileal contents increased (P < 0.01) with the addition of PS to the diet (0.39%, 0.77% and 1.08% for diets PS0, PS7 and PS14, respectively). Starch digestibility was 0.8 percentage units higher (99.8% v. 99.0%) with the PS0 diet than the PS14 diet (P = 0.04). The bacterial cellulolytic activity in the caecum tended to be higher with the PS14 diet (P = 0.07). The total VFA caecal concentration increased (P < 0.01) only in 6-week-old rabbits with PS7 compared with PS0 (54.7 v. 74.5 mmol/l). Protein digestibility and ileal starch concentration decreased (P < 0.05) with age (6 v. 10 weeks), and hemicelluloses digestibility increased (P < 0.05). At 10 weeks of age, rabbits showed a higher VFA pool (6.25 mol) and proportion of butyrate (15.9%) and a lower proportion of acetate (79.3%), ammonia level (7.5 mmol/l) and C3/C4 ratio (0.31) than at 6 weeks of age. The intake of potato starch had no effect on the performance, caecal microbial activity or digestive health of growing rabbits.  相似文献

Rice, wheat, and potato starches were investigated using ultra-small angle X-Ray diffraction (USXRD) in the range of 100–58,000 Å. The results showed trends consistent with the known sizes of starches. However, the observed Rg values for the scattering substances lie in the 100–300 nm range, very much in the low end of the known starch granule size distributions (and below the resolution of the light microscope) suggesting different, perhaps interesting, structures than those observed by light microscopy. Thus what were detected may possibly be the sizes of the crystalline regions postulated to occur in individual starch granules.  相似文献

A mixed mash of sugar beet roots and potato tubers with a sugar concentration of 23.7% w/v was used as a feedstock for bioethanol production. Enzymatic digestion successfully reduced the viscosity of the mixture, enabling subsequent heat pretreatment for liquefaction/sterilization. An energy-consuming thick juice preparation from sugar beet for concentration and sterilization was omitted in this new process.  相似文献

Samples of oven-dried, air-dried, and moisturised potato starch (5, 13, and 24% w/w moisture content, respectively) were frozen in liquid nitrogen. Samples after thawing were studied by means of cross-polarised light beam microscope (CLBM), Fourier Transformation Infrared Spectroscope (FT-IR), powder X-ray diffractometer, and non-contact Atomic Force Microscope (nc-AFM). Rapid deep-freezing followed by thawing produced changes on the granule surface. They were accompanied by internal alteration manifested by FT-IR spectra and powder X-ray diffractograms. The results depended on the water content in the sample. Deep-freezing of moistened starch resulted in increased crystallinity of granules. It had minor effect on the granule aqueous solubility and characteristics of gelation.  相似文献

Two mutants, Ls1 and Ls2, of Zymomonas mobilis B-806 unable to produce levan were isolated. With native gel electrophoresis and zymogram analysis it was confirmed that the mutants did not synthesize active levansucrase (E2). However, they produced intracellular sucrase (E1) and extracellular invertase (E3). Comparison of these mutants with the parent strain for alcohol production on glucose, fructose and sucrose (100 g/l each) media revealed that the final ethanol concentration achieved in sucrose medium was only about 5 g/l higher with the mutants than with the wild type. The ethanol yield of the mutants increased from 0.48 g/g to 0.50 g/g on sucrose medium.  相似文献

A continuous process involving simultaneous saccharification and fermentation of liquefied starch has been developed using Zymomonas mobilis. Amyloglucosidase retention and cell recycle have been effected by using an Amicon hollow-fiber membrane system with a MW cutoff of 5000. Relatively high productivities of up to 60 g L(-1) h(-1) have been achieved at ethanol concentrations of 60-65 g/L. The system also offers the potential for reduced enzyme requirements for saccharification.  相似文献

Halloysite/potato starch composites were prepared by adding modified natural halloysite nanotubes into potato starch matrices to reenforce the mechanical properties of potato starch films. The halloysite/potato starch films were characterized by X-ray diffraction, scanning electron microscope and infrared spectrometry. Meanwhile, the mechanical properties and transparency of the films were studied. The results show that the modified halloysite nanotubes can be well distributed in the starch matrix and thus the tensile strength of the films was clearly enhanced. The flexibility of the films could be improved through adding glycerol although at the cost of reducing tensile strength. But incorporation of PVA could further improve the tensile strength of the halloysite/potato starch films.  相似文献

Summary Cassava and sago starch were evaluated for their feasibilities as substrates for ethanol production using Zymomonas mobilis ZM4 strain. Before fermentation, the starch materials were pretreated employing two commercial enzymes, Termamyl (thermostable -amylase) and AMG (amyloglucosidase). Using 2 l/g of Termamyl and 4 l/g of AMG, effective conversion of both cassava and sago starch into glucose was found with substrate concentration up to 30%(w/v) dry substances. Fermentation study performed using these starch hydrolysates as substrates resulted in ethanol yield at an average of 0.48g/g by Z. Mobilis ZM4.  相似文献

Silication of potato starch was performed by microwave irradiation and convectional heating of starch with sodium metasilicate. The study has shown that microwaves offered more selective silication than convectional heating. Depending on the dose of metasilicate products of either monoesterification or crosslinking esterification were formed. Increase in the amount of the silicating agent favoured crosslinking of starch. In the case of microwave irradiation, the C–O–SiO2Na moieties were formed, whereas the convectional heating generated the C–O–Si–O–Si–O–C crosslinks.  相似文献

The fermentation kinetics Zymomonas mobilis were studied near zero growth rate in fed-batch cultures and continuous cultures with complete cell recycle. The results show the ethanol enhances that specific substrate conversion rate under these conditions. The maximum achievable ethanol concentration in continuous cultures with cell recycle (66 g/L) was significantly lower than in fed-batch cultures (100 g/L). The results indicate that growth-rate-independent metabolism is not instantaneous and can lag behind steadily increasing ethanol concentrations in fed-batch fermentations. A model is proposed to account for this slow adaptation.  相似文献

Summary Zymomonas mobilis was grown in batch concentrations between 200 and 400 g/l sucrose. The fermentation pattern revealed that the efficiency of sucrose hydrolysis dropped only from 94 to 78.6% whereas the efficiency with which the hydrolyzed products were converted to ethanol decreased from 94 to 43%. The ethanol yields were relatively constant for final concentrations which lay between 80 and 132 g/l. Fermentation times increased to 72 hours at the higher sucrose concentrations. Sorbitol and fructose were identified as the major by-products. Preliminary evidence suggests that the ratio between the two by-products depends on the pH of the culture medium. Results suggest the possibility of processes producing ethanol plus fructose, ethanol plus fructose and sorbitol, or ethanol plus sorbitol in a single-stage batch fermentation.  相似文献

Wheat starch was cross-linked at 40 °C and pH 11.0 by slurrying the starch (30% solids) in a solution of sodium trimetaphosphate (STMP), sodium hydroxide, and sodium sulfate. The extent of cross-linking was determined by an increase in alkaline fluidity or by a decrease in alkaline clarity. Response surface analysis showed that cross-linking increased with increasing levels of STMP (0.5-1.5%, based on starch, bos) and sodium sulfate (0–4.0%, bos) over a reaction period of 120–720 min. A regression equation with first and second order terms showed that STMP and sodium sulfate concentrations and the reaction time accounted for 99% of the variability in alkaline fluidity. Wheat starch (37% slurry) was hydroxypropylated by reaction with propylene oxide (8%, bos) for 24h at 45 °C in alkali (pH 11.5) containing 16.0% sodium sulfate (bos). The hydroxypropylated (4.5wt%) wheat starch (DS 0.12-0.13) was not isolated but was cross-linked with STMP (0.1-0.5%, bos) over a 10–40 min reaction period. A comparison of pasting curves at pH 3.5 showed that some of the cross-links produced by STMP were less stable than those produced by phosphoryl chloride, indicating a low level of pyrophosphate as well as monophosphate cross-links. The less stable cross-links were diminished by changing reaction conditions with STMP.  相似文献
