Linoleic acid was differentially catabolized by the various rumen microbial fractions, such as rumen bacteria (B), protozoa (P), and their mixture (BP). The predominant isomer of conjugated linoleic acids (CLA) synthesized by B, P, and BP from linoleic acid was 9c11t-CLA. The formation of 9c11t-CLA was higher (P < 0.05) in P suspension (53.6 μg/mg microbial nitrogen) compared with B (38.3 μg/mg microbial nitrogen) and BP (28.8 μg/mg microbial nitrogen) suspensions by 12 h of incubation. The second most abundant CLA isomer was 10t12c. The accumulation of 10t12c-CLA in BP suspension was 2.3 times lower (P < 0.05) than that in B suspension (84.8 μg/mg microbial nitrogen) by 12 h of incubation. The accumulation of 10t-18:1 in BP suspension during 6- and 12-h incubation periods were not different (P > 0.05) than that in B suspension (6.8 and 14.0 μg/mg microbial nitrogen, respectively). However, the accumulation of 11t-18:1 in BP suspension at 6- and 12-h incubations were 2.7 and 3.3 times higher (P < 0.05), respectively, than that in B suspension. There were no significant accumulations of 11t-18:1, 10t-18:1, and 18:0 in P suspension throughout the incubation period. It was concluded that B, P, and BP metabolized linoleic acid to different isomers of CLA, whereas B, including BP, was only capable of biohydrogenating the CLA isomers to 18:0 by the reduction of 18:1 isomers. P was incapable of biohydrogenating LA, but its association with B in the BP suspension altered the biohydrogenation of LA significantly compared with B alone.  相似文献

 Our isolate, Pseudomonas putida, is known to be capable of utilizing cyanides as the sole source of carbon (C) and nitrogen (N) both in the form of free cells and cells immobilized in calcium alginate. In the present study, the cell-free extract(s) were prepared from the cells of P. putida grown in the presence of sodium cyanide. The ability of enzyme(s) to convert cyanides, cyanates, thiocyanates, formamide and cyanide-containing mine waters into ammonia (NH3) was studied at pH 7.5 and pH 9.5. The kinetic analysis of cyanide and formamide conversion into NH3 at pH 7.5 and pH 9.5 by the cell-free extract(s) of P. putida was also studied. The K m and V max values for cyanide/formamide were found to be 4.3/8 mM and 142/227 μmol NH3 released mg protein-1 min-1 respectively at pH 7.5 and 5/16.67 mM and 181/434 μmol NH3 released mg protein-1 h-1 respectively at pH 9.5. The study thus concludes that the cell-free extract(s) of P. putida is able to metabolize not only cyanides, cyanates, thiocyanates, and formamide but also cyanide-containing mine waters to NH3.  Large-scale exploitation of the potential benefits of ectomycorrhizal fungi in improving plantation yields means that fermentation techniques for these fungi will be required. Starting with a base performance on a rich, complex medium, the effect of variations in some physicochemical culture parameters on biomass yield was studied. It was possible to reduce the amount of phosphate salts (to 1/9th) and other ingredients (to 1/3rd) in the medium. A shaking speed of either 100 rpm or 200 rpm in an orbital incubator was satisfactory and biomass yield responded to an increase in carbon substrate (glucose, from 10 g l-1 and 20 g l-1) though Y x/s declined. An increase in inoculum size shortened culture time but decreased biomass yield. The upper limit of the incubation temperature was between 25°C and 30°C. Biomass yields were about 12 g l-1 dry weight (Y x/s=0.63) when 20 g l-1 glucose was supplied, and about 7 g l-1 (Y x/s=0.74) when 10 g l-1 glucose was supplied. Summary One of the first Azorean endemic vascular taxa chosen for the development ofin vitro multiplication techniques wasHypericum foliosum Aiton, due to its colonizing ability (Sj?gren, 1984), a loss of seed germination capacity after only 1 yr of storage (Maciel, 1994), and the populations' generally low number of individuals. The following culture media were tested usingHypericum foliosum's single node cuttings: Murashige and Skoog (1962), Roest and Bockelmman (1973), Lloyd and McCown (1980), C?rte and Mendon?a (1985), and Cellárová et al. (1992). Further experiments were performed on CM medium supplemented with four different growth regulators: α-naphthaleneacetic acid (NAA), N6-benzyladenine (BA), γ, γ-(dimethylallyl) aminopurine (2iP), and kinetin (KIN). The acclimatization stage was carried out in Jiffy 7? pots and in a 2∶1 or 1∶1 peat/perlite mixture. We found that micropropagation ofHypericum foliosum is possible on CM medium and that the best results with growth regulators were achieved with the following supplements: 0.1 mg/l (0.4 μM) BA and 0.5 mg/l (2.6 μM) NAA+1.0 mg/l (4.4 μM) BA (in the initiation stage), and 0.1 mg/l (0.4 μM) BA (in the elongation stage). As for culture multiplication, 0.1 mg/l (0.4 μM) BA (in the initiation stage) and 0.5 mg/l (2.6 μM) NAA+1.0 mg/l (4.4 μM) BA (both in the initiation and elongation stages), proved to be the most efficient concentrations. (S)(E)-2-{3-[3-[2-(7-chloro-2-quinolinyl)ethenyl]-phenyl]-3-hydroxypropyl} benzoic acid methyl ester,␣a key intermediate in the synthesis of the anti-asthma drug, Montelukast, was prepared from the corresponding ketone (keto ester M) by microbial transformation. The biotransforming organism, Microbacterium campoquemadoensis (MB5614), was discovered as a result of an extensive screening program and was used for the isolation and purification of the responsible enzyme. The enzyme is a soluble cytoplasmic protein which was purified as a complex with a low-molecular-mass molecule that had a visible-light absorption maximum at 460 nm. The purified enzyme has an apparent molecular mass of 60 kDa, when denatured, and is isolated in the native state as an oligomer. The isolated enzyme requires NADPH for its activity and reduces the keto ester M to the desired (S)-hydroxy ester with an enantiomeric excess greater than 95% at the optimum temperature of 30 °C and pH 8. The enzyme was immobilized on oxirane-activated acrylamide beads with some loss of activity, but it was fully active in a two-phase (water/hexane 25:75) solvent system, both as a free solution and in an immobilized form. 为探讨黄河三角洲海岸带湿地不同水盐条件下植物叶片化学计量特征的季节动态及植物生长的限制性营养元素,以滨州贝壳堤岛与湿地国家级自然保护区内的柽柳、杠柳、酸枣3种优势灌木为研究对象,于2017-2018年的生长季(5-10月)每月定期采集叶片样品,测定叶片C、N、P含量。结果表明,生长季内3种灌木叶片C含量呈逐渐上升趋势;叶片N、P含量呈先下降后上升趋势,说明3种灌木采用防御性生活史策略适应盐生和干旱生境。3种灌木叶片C平均含量分别为(399.65±2.66)mg/g、(424.32±1.59)mg/g、(437.47±1.08)mg/g,低于全国(455.1 mg/g)及全球(461.6 mg/g)水平,呈现盐生生境下较低的植物碳储存能力。3种灌木叶片N和P平均含量分别为(30.14±0.26)mg/g和(1.81±0.03)mg/g、(23.18±0.38)mg/g和(2.06±0.04)mg/g、(27.36±0.49)mg/g和(2.01±0.03)mg/g,显著高于全国(N∶20.2 mg/g,P:1.46 mg/g)及全球(N∶19.3-20.1 mg/g,P:1.11-1.42 mg/g)水平。叶片C∶N∶P比呈先上升后下降的趋势,叶片P含量对C∶N∶P比变化具有主导作用。3种灌木叶片C∶N∶P质量比分别为246∶17∶1、224∶12∶1、237∶14∶1,说明柽柳的水盐胁迫适应能力高于杠柳和酸枣。从叶片N、P化学计量特征看,生长季内,柽柳生长一定程度上受土壤P限制,杠柳生长受到土壤N限制,酸枣生长则受土壤N、P共同限制,说明3种灌木的生物地球化学生态位发生了分化,避免了对同种资源的竞争,利于物种共生。  相似文献

Recombinant Escherichia coli cells expressing the toluene dioxygenase (TDO) genes from Pseudomonas putida convert indene to cis-1S,2R-indandiol, a potentially important intermediate for the chemical synthesis of the HIV-1 protease inhibitor, Crixivan. A bioconversion process was developed through optimization of medium composition and reaction conditions at the shake-flask and 23-l fermentor scales. A cis-1,2-indandiol productivity of approx. 1000 mg/l was achieved with construct TDO123, which represents a 50-fold increase over the initial titer. Varying the bioconversion conditions did not change the enantiomeric excess (e.e.) for the 1S,2R enantiomer from about 30%, suggesting that toluene dioxygenase intrinsically converts indene to 1S,2R- and 1R,2S-indandiols at a ratio of 2:1. Further inclusion of the Pseudomonas dehydrogenase gene in construct D160-1 led to the production of chirally pure cis-1S,2R-indandiol (e.e. > 99%) as a result of the selective degradation of the 1R,2S enantiomer, with the overall yield (650 mg/l) proportionally reduced. A single stage process was developed for D160-1 and scaled up to the 23-l fermentor, achieving a cis-1S,2R-indandiol titer of 1200 mg/l.  Diatom composition and biomass were investigated in the nearshore water (<30 m in depth) of Maxwell Bay, Antarctica during the 1992/1993 austral summer. Epiphytic or epilithic diatoms such as Fragilaria striatula, Achnanthes brevipes var. angustata and Licmophora spp. dominated the water column microalgal populations. Within the bay, diatom biomass in surface water was several times higher at the nearshore (2.4–14 μg C l-1) than at the offshore stations (>100 m) (1.2–3.2 μg C l-1) with a dramatic decrease towards the bay mouth. Benthic forms accounted for >90% of diatom carbon in all nearshore stations, while in the offshore stations planktonic forms such as Thalassiosira antarctica predominated (50–>90%). Microscopic examination revealed that many of these diatoms have become detached from a variety of macroalgae growing in the intertidal and shallow subtidal bottoms. Epiphytic diatoms persistently dominated during a 19-day period in the water column at a fixed nearshore station, and the biomass of these diatoms fluctuated from 0.86 to 53 μg C l-1. A positive correlation between diatom biomass and wind speed strongly suggests that wind-driven resuspension of benthic forms is the major mechanism increasing diatom biomass in the water column. Immobilized cells of Delftia tsuruhatensis CCTCC M 205114 harboring R-amidase were applied in asymmetric hydrolysis of (R)-2, 2-dimethylcyclopropane carboxamide (R − 1) from racemic (R, S)-2, 2-dimethylcyclopropane carboxamide to accumulate (S)-2, 2-dimethylcyclopropane carboxamide (S − 1). Maximum R-amidase activity of 13.1 U/g wet cells (0.982 U/g beads) was obtained under conditions of 3% sodium alginate, 2.5% CaCl2, 15 h crosslinking and 2 mm bead size, which was 53.9% of that of free cells (24.3 U/g wet cells). In addition, characterization of the immobilized cells was examined. The optimum R − 1 hydrolysis conditions were identified as follows: substrate concentration 10 mM, pH 8.5, temperature 35°C and time course 40 min. Under optimum conditions, the maximum yield and enantiomeric excess of (R)-2, 2-dimethylcyclopropanecarboxylic acid were 49.5% and >99%, respectively. This afforded S − 1 with a yield >49% and an e.e. of 97.7%. With good operational stability and excellent enanotioselectivity, the immobilized cells could be potentially utilized in industrial production of S − 1.  相似文献

 The presence of a codominant AFLP marker, EAA/MCAT10, correlates with the primary source of resistance to root-knot nematodes (Meloidogyne incognita and M. javanica) in rootstock cultivars of peach [Prunus persica (L.) Batsch]. Two allelic DNA fragments of this AFLP marker were cloned, sequenced and converted to sequence tagged sites (STS). Four nucleotide differences (i.e. one addition and three substitutions) were observed between the two clones. Furthermore, there was a diagnostic Sau3 AI cleavage site (GATC) in the large fragment that was absent from the small fragment (GTTC at this site). The applicability of this STS marker system to peach germplasm improvement was evaluated: genomic DNAs of cross parents (i.e. ‘Lovell’ and ‘Nemared’), four F1 hybrids (K62-67, K62-68, P101-40 and P101-41) and two F2 populations (from K62-68 and P101-41), as well as DNA from a test panel of 18 rootstock cultivars or selections, were PCR-amplified with the Mij3F/Mij1R primer pair and then digested with Sau3 AI. The banding patterns showed that the EAA/MCAT10 STS markers can clearly distinguish the three genotypes – homozygous resistant, heterozygous resistant and homozygous susceptible – in the ‘Lovell’בNemared’ cross. In addition, results from the rootstock survey were consistent with each rootstock’s phenotypic response to nematode infection, except for ‘Okinawa’, ‘Flordaguard’ and ‘Yunnan’ where root-knot resistance may have arisen independently. Therefore, the EAA/MCAT10 STS markers will be a useful tool to initiate marker assisted selection studies in peach rootstock breeding for root-knot nematode resistance.  The trophic interaction in the pelagic food web of oligotrophic-dystrophic Lake Shirakoma-ike was investigated by a stable isotope technique. Carbon and nitrogen isotope analyses revealed the trophic interaction among Piona carnea (a water mite), Chaoborus sp. (a phantom midge), Daphnia longispina (a cladoceran), and Acanthodiaptomus pacificus (a calanoida copepoda). The δ13C values for estimating the relative contributions of D. longispina and A. pacificus to carnivores were used, and they were also used for the constraint of the trophic enrichment factor of δ15N. Both Chaoborus sp. and P. carnea were suggested to consume D. longispina and A. pacificus at almost the same ratio (1 : 1). However, both carnivores preferred A. pacificus to D. longispina, because D. longispina was much more abundant in the lake than A. pacificus. Omnivory of P. carnea was also analyzed by the mixing model. Streptomyces viridosporus T7A (ATCC␣39115), during growth in tryptone/yeast extract broth, cometabolized five heterocyclic nitrogen-containing compounds. The metabolites produced from the azaarenes were identified by high-performance liquid chromatography, UV/visible absorption spectroscopy, and mass spectrometry. Isoquinoline was transformed to 1(2H)-isoquinolinone (14%), phenanthridine to 6(5H)-phenanthridinone (25%), phthalazine to 1(2H)-phthalazinone (46%), quinazoline to 2,4(1H,3H)-quinazolinedione (4%), and quinoxaline to 2(1H)-quinoxalinone (8%) and 1-methyl-2(1H)-quinoxalinone (12%). Glomus intraradices , Glomus mosseae, and Gigaspora rosea leads to the accumulation of cyclohexenone derivatives. Mycorrhizal roots of all plants accumulate in response to all three fungi blumenin [9-O-(2′-O-glucuronosyl)-β-glucopyranoside of 6-(3-hydroxybutyl)-1,1,5-trimethyl-4-cyclohexen-3-one], 13-carboxyblumenol C 9-O-gentiobioside, nicoblumin [9-O-(6′-O-β-glucopyranosyl)-β-glucopyranoside of 13-hydroxy-6-(3-hydroxybutyl)-1,1,5-trimethyl-4-cyclohexen-3-one] and another, as yet unidentified, cyclohexenone derivative. The accumulation of all four compounds in three tested mycorrhizal plants colonized by the three arbuscular mycorrhizal fungi indicates no fungus-specific induction of these compounds.  Epibenthic brittle star assemblages were investigated on the northwestern Barents Sea shelf between 81° and 77°N in July 1991. At 9 drift stations in water depths between 80 and 360 m, series of 35–71 photographs, each depicting about 1 m2 of the seabed, were taken along transects of about 150- to 300-m length to assess abundances and spatial distribution patterns of adult brittle stars (disc diameter ≥1 mm). Biomass values were derived by combining abundances with size-weight relationships and size frequencies established using specimens from trawl catches. Six brittle star species were identified on the seabed images. Ophiocten sericeum was the most abundant species on shallow shelf banks (≤100 m). Up to 2,800 individuals were counted on a single photograph; median abundances per station ranged from 32 to 524 ind.m-2 and biomass from 0.3 to 5.0 g ash-free dry weight (AFDW) m-2. The spatial distribution along the transects (i.e. on the 100-m scale) was, however, extremely patchy. Disc diameters of O. sericeum ranged between 1.6 mm and 15.4 mm. In deeper shelf habitats (>150 m), O. sericeum was rare or absent, and Ophiacantha bidentata dominated the brittle star fauna with median densities and biomasses of 2–49 ind.m-2 and 0.07–1.9 g AFDW m-2, respectively. Its disc diameters ranged from 2.9 to 14.4 mm. The other species (Ophiura sarsi, Ophiopholis aculeata, Ophioscolex glacialis, Ophiopleura borealis) occurred in distinctly lower numbers. Our findings provide further evidence that brittle stars dominate epibenthic communities on Arctic shelves and locally reach very high abundances.  Mating behavior of the Far Eastern catfish, Silurus asotus (Siluriformes: Siluridae), was observed in a ricefield system facing the shore of Lake Biwa in mid-May to early June in 1990–1997. A set behavioral sequence similar to those of two other silurid fishes, S. biwaensis and S. lithophilus, both endemic to the Lake Biwa system, was observed: “chasing,”“clinging,”“enfolding” while “squeezing” by the male, and “circling” by the spawning pair. This form of mating behavior is quite different from that of S. asotus reported from the Ooi River system in Kyoto Prefecture, which mainly spawns in running water in ditches.  The influence of culture chamber capacity, medium volume and culture density on the growth yields of lettuce (Lactuca sativa L.) and spearmint (Mentha spicata L.) shoots were determined in an environment containing either 350 or 10,000 μmol mol–1 CO2 after 8 weeks of incubation. High positive correlations occurred between the culture vessel capacity and spearmint fresh weight, leaf number, root number, and shoot number. Similarly, high positive correlations occurred between culture vessel capacity and lettuce fresh weight, leaf number, and root number. Higher fresh weights, leaf numbers, and root numbers were obtained from lettuce and spearmint shoots when cultured in 1-quart Mason jars containing 100- or 150-ml aliquots of medium compared to jars containing 25- or 50-ml aliquots of medium within an environment containing either 350 or 10,000 μmol mol–1 CO2. High culture density decreased growth yields, and this phenomenon could only be slightly off-set by the employment of an elevated CO2 environment or larger culture vessels. Methyl ent-17-hydroxy-16β-kauran-19-oate was fed to a 2-day-old culture of the fungus Rhizopus stolonifer, fermenting at room temperature (25 °C) in an orbital shaker (2 l). After 11 days, both broth and mycelia were extracted with ethyl acetate. Two novel compounds were isolated from this experiment: methyl ent-9α,17-dihydroxy-16β-kauran-19-oate and methyl ent-7α,17-dihydroxy-16β-kauran-19-oate. We examined trophic positions and fatty acid concentrations of riverine, lacustrine, and aquaculture diet and fish in Austrian pre-alpine aquatic ecosystems. It was hypothesized that dietary fatty acid (FA) profiles largely influence the FA composition of the salmonids Salvelinus alpinus, Salmo trutta, and Oncorhynchus mykiss. We analyzed trophic positions using stable isotopes (δ15N) and tested for correlations with polyunsaturated fatty acid (PUFA) concentrations. Gut content analysis revealed benthos (rivers), pellets (aquaculture), and zooplankton (lakes) as the predominant diet source. Results of dorsal muscle tissues analysis showed that the omega-3 PUFA, docosahexaenoic acid (DHA; 22:6n − 3), was the mostly retained PUFA in all fish of all ecosystems, yet with the highest concentrations in S. alpinus from aquaculture (mean: 20 mg DHA/g dry weight). Moreover, we found that eicosapentaenoic acid (EPA; 20:5n − 3) in fish of natural habitats (rivers, lakes) was the second most abundant PUFA (3–5 mg/g DW), whereas aquaculture-raised fish had higher concentrations of the omega-6 linoleic acid (18:2n – 6; 9–11 mg/g DW) than EPA. In addition, PUFA patterns showed that higher omega-3/-6 ratios in aquacultures than in both riverine and lacustrine fish. Data of this pilot field study suggest that salmonids did not seem to directly adjust their PUFA to dietary PUFA profiles in either natural habitats or aquaculture and that some alterations of PUFA are plausible. Finally, we suggest that trophic positions of these freshwater salmonids do not predict PUFA concentrations in their dorsal muscle tissues.  相似文献

沉水植物是水生生态系统中重要的初级生产者。当前有关沉水植物生态化学计量特征的研究主要集中在非喀斯特区,而在喀斯特区的相关研究较为缺乏。因此,以广西澄江喀斯特典型湿地中的7种沉水植物为研究对象,分析沉水植物地上部分及底泥的碳(C)、氮(N)、磷(P)化学计量特征。结果显示,(1)7种沉水植物地上部分总碳、总氮和总磷平均含量最大值均出现在小茨藻(Najas minor)分别为(325.4±5.01) g/kg、(33.07±1.59) g/kg和(3.79±0.16) g/kg;C : N、C : P、N : P平均值分别为10.14±0.18、96.23±3.56和9.47±0.32,C : N最大值(11.89±0.54)出现在苦草(Vallisneria natans),C : P和N : P最大值(113.27±18.14和11.13±1.63)均出现在穗花狐尾藻(Myriophyllum spicatum)。(2)底泥有机碳、总氮、总磷、碱解氮和速效磷含量平均值分别为(15.05±0.56) g/kg、(2.06±0.08) g/kg、(0.58±0.01) g/kg、(162.53±9.16) mg/kg和(21.73±0.86) mg/kg,有机碳、总氮、总磷、碱解氮和速效磷的平均含量最大值均出现在小茨藻(N.minor)分别为(18.54±1.04) g/kg、(2.55±0.25) g/kg、(0.66±0.03) g/kg、(214.82±32.05) mg/kg和(26.37±3.31) mg/kg;底泥的C : N、C : P和N : P平均值分别是7.33±0.14、25.7±0.72和3.53±0.09,C : N、C : P和N : P最大值分别出现在金鱼藻(Ceratophyllum demersum)(7.45±0.32)、小茨藻(N. minor)(28.29±1.29)和黑藻(Hydrilla verticillata)(3.89±0.25)。(3)沉水植物的地上部分总碳与底泥的有机碳和总氮均呈显著负相关性;沉水植物的地上部分总氮与底泥有机碳、碱解氮、C : P均呈显著正相关性;沉水植物地上部分C : N、C : P均与底泥有机碳、总氮、总磷、碱解氮、C : P、N : P呈显著负相关,表明不同喀斯特湿地沉水植物和底泥之间的养分耦联性不同。本研究为喀斯特湿地生态系统生态化学计量学研究提供理论依据。  相似文献
