The potassium tellurite concentration, 0.01% w/v, in Baird-Parker agar has been recommended for plasma coagulase media such as pig or rabbit fibrinogen agar. Comparative tests have shown that with some strains of Staphylococcus aureus this level of potassium tellurite is too inhibitory and it should be reduced four-fold to 0.0025% w/v to maximize the isolation rate. It is postulated that egg yolk in Baird-Parker agar has a protective effect on staphylococci against the inhibitory action of tellurite.  相似文献

This study is part of an ongoing project aiming at determining the ethnogenesis of an eastern Siberian ethnic group, the Yakuts, on the basis of archaeological excavations carried out over a period of 10 years in three regions of Yakutia: Central Yakutia, the Vilyuy River basin and the Verkhoyansk area. In this study, genetic analyses were carried out on skeletal remains from 130 individuals of unknown ancestry dated mainly from the fifteenth to the nineteenth century AD. Kinship studies were conducted using sets of commercially available autosomal and Y-chromosomal short tandem repeats (STRs) along with hypervariable region I sequences of the mitochondrial DNA. An unexpected and intriguing finding of this work was that the uniparental marker systems did not always corroborate results from autosomal DNA analyses; in some cases, false-positive relationships were observed. These discrepancies revealed that 15 autosomal STR loci are not sufficient to discriminate between first degree relatives and more distantly related individuals in our ancient Yakut sample. The Y-STR analyses led to similar conclusions, because the current Y-STR panels provided the limited resolution of the paternal lineages.  相似文献

Summary A technique for observing the reduction of potassium tellurite byCandidas was developed. Various species of these yeasts reduce it at different concentrations, but that which is most useful for differentiation is 0.02 % added to Sabouraud dextrose agar basal medium. Of the yeasts studied,C. albicans, C. parapsilosis andC. tropicalis all reduced potassium tellurite to the concentration mentioned before, while the growth ofC. krusei, C. parakrusei andC. pseudotropicalis was inhibited. Without exception,C. pseudotropicalis reduced this salt at lower concentrations. The two strains ofC. guilliermondii tested gave contradictory results: one of them grew and reduced potassium tellurite, while the growth of the other was inhibited.Professor of Microbiology.  相似文献

Ribonuclease P (RNase P) is a ubiquitous endonuclease that catalyses the maturation of the 5' end of transfer RNA (tRNA). Although it carries out a biochemically simple reaction, RNase P is a complex ribonucleoprotein particle composed of a single large RNA and at least one protein component. In bacteria and some archaea, the RNA component of RNase P can catalyse tRNA maturation in vitro in the absence of proteins. The discovery of the catalytic activity of the bacterial RNase P RNA triggered numerous mechanistic and biochemical studies of the reactions catalysed by the RNA alone and by the holoenzyme and, in recent years, structures of individual components of the RNase P holoenzyme have been determined. The goal of the present review is to summarize what is known about the bacterial RNase P, and to bring together the recent structural results with extensive earlier biochemical and phylogenetic findings.  相似文献

Natronococcus occultus, a haloalkaliphilic archaeon, was examined for its resistance to potassium tellurite. Cells grown in the presence of 1 mM potassium tellurite reduced it to metallic tellurium resulting in the deposition of intracellular crystals in the cytoplasm. The minimal inhibitory concentration for potassium tellurite was 10 mM. N. occultus had an inducible tellurite reductase activity. Cell-free extracts catalyzed the enzymatic reduction of potassium tellurite in a reaction which was dependent on NADH oxidation and a reduced environment.  相似文献

A previous analysis of tRNA sequences suggested a correlation between the absence of a nucleotide at position 47 (nt 47) in the extra loop and the presence of a U13:G22 base pair in the D-stem. We have evaluated the significance of this correlation by determining the in vivo activity of tRNAs containing either a C13:G22 or a U13:G22 pair in tRNA molecules with or without nt 47. Although this correlation might reflect some malfunction of tRNAs lacking nt 47, but containing the C13:G22, assays of the in vivo suppressor activity showed that this tRNA is actually more active than the tRNA with the features found in the database, i.e., a U13:G22 base pair and no nt 47. Moreover, analogous constructs with a GGC anticodon permitted the growth of an Escherichia coli strain deleted for tRNA(Ala)GGC genes equally well. On the other hand, long-term growth experiments with competing E. coli strains harboring the tRNA lacking nt 47, either with the C13:G22 or the U13:G22 base pair demonstrated that the U13:G22 tRNA overtook the C13:G22 strain even when the starting proportion of strains favored the C13:G22 strain. Thus, the preference for the U13:G22 tRNA lacking nt 47 in the sequence database is most likely due to factors that come into play during extended growth or latency rather than to the ability of the tRNA to engage in protein synthesis.  相似文献

The ultrastructure of strains Pseudomonas putida BS228 and Pseudomonas aeruginosa ML4262 harboring plasmids pBS10, pBS31, and pBS221, which determine resistance to potassium tellurite, was studied. Bacteria were grown in media containing increasing concentrations of potassium tellurite. Crystalline structures containing tellurium appeared in their periplasmic space. The dynamics of crystal growth was studied. Crystals were released into the medium by pinching off of the outer membrane vesicles containing growing crystals. A possible mechanism of this process was described; cytobiochemical peculiarities were discussed.  相似文献

Spliced-leader (SL) trans-splicing is an essential step in pre-mRNA maturation in a variety of lower eukaryotic organisms. However, this processing pathway is absent in mammals, insects, yeast and plants. The patchy phylogenetic distribution of SL trans-splicing is consistent with either ‘multiple gain’ or ‘multiple loss’ evolutionary scenarios. Recent studies show that two additional metazoan phyla carry out SL trans-splicing, significantly increasing its phylogenetic range. However, it remains unclear whether this unusual type of splicing is an ancestral or an acquired trait.  相似文献

Here we compare the physiological state of Escherichia coli exposed to tellurite or selenite by using the noninvasive technique of phosphorus-31 nuclear magnetic resonance (NMR) spectroscopy. We studied glucose-fed Escherichia coli HB101 cells containing either a normal pUC8 plasmid with no tellurite resistance determinants present or the pTWT100 plasmid which contains the resistance determinants tehAB. No differences could be observed in intracellular ATP levels, the presence or absence of a transmembrane pH gradient, or the levels of phosphorylated glycolytic intermediates when resistant cells were studied by 31P NMR in the presence or absence of tellurite. In the sensitive strain, we observed that the transmembrane pH gradient was dissipated and intracellular ATP levels were rapidly depleted upon exposure to tellurite. Only the level of phosphorylated glycolytic intermediates remained the same as observed with resistant cells. Upon exposure to selenite, no differences could be observed by 31P NMR between resistant and sensitive strains, suggesting that the routes for selenite and tellurite reduction within the cells differ significantly, since only tellurite is able to collapse the transmembrane pH gradient and lower ATP levels in sensitive cells. The presence of the resistance determinant tehAB, by an as yet unidentified detoxification event, protects the cells from uncoupling by tellurite.  相似文献

Classification of bacteria evolved from limited subjective groupings to general, more objective arrangements based on overall phenotypic similarities. However, classifications based on phenotypic characters lack stability, whereas those based on genetic relatedness tend to be stable. DNA-DNA hybridization has proven to be extremely useful in resolving taxonomic problems at the species level. Broad relationships among bacteria have been identified by comparing ribosomal RNA cistrons; however, many groups based on ribosomal RNA analysis are not easily definable in terms of phenotypic similarities. Unless resolved, these problems could lead to the establishment of two separate classification systems, one phylogenetic and the other practical.  相似文献
