The efficacy of some putative attractants for the biting midge Culicoides impunctatus (Goetghebuer) (Diptera: Ceratopogonidae) was assessed using odour-baited 'delta traps' and suction traps. 1-octen-3-ol was confirmed as a potent olfactory attractant for C. impunctatus when released at 0.06mg/h. Acetone (23mg/h) and a mix of six phenolic compounds (phenol, 3-ethylphenol, 4-ethylphenol, 3-methylphenol, 4-methylphenol and 4-propylphenol), at undetermined release rate, also significantly increased delta trap catches compared to unbaited controls. When tested in combination, there was evidence of synergism between CO2 (0.2L/min) and acetone, 1-octen-3-ol or cow urine, trap catches being, respectively, 4.7, 6.2 and 9.3-fold greater than for CO2 alone. Highest catches were obtained with triple bait combinations comprising cow urine + acetone + CO2 or cow urine + 1-octen-3-ol+CO2, which increased trap catches by X 22 and X 24, respectively, compared to CO2 alone. Culicoides impunctatus was found to be extremely sensitive to CO2 and responses, gauged over two field seasons, showed a significant dose-dependent increase in catch across the entire range of release rates (0.2-2.5 L/min). Responses to these release rates, ranging from small to large mammal equivalents, emphasized the important role of CO2 in host location by C. impunctatus. Uses of olfactory attractants for monitoring and control of Culicoides are reviewed on the basis of these results. 相似文献

Abstract.
- 1 Swarms of Culicoides impunctatus males were examined in the field in Scotland. Most swarms were close to midge emergence/breeding grounds over a variety of vegetation, some of which clearly acted as swarm markers. Low light (0–1000 lux) and still/humid conditions favoured swarming.
- 2 Swarm size ranged between less than 10 and more than one thousand midges. The modal size was 200 individuals. The smallest swarms were usually columnar in shape and the larger swarms ovoid.
- 3 Midges behaved individually within swarms, moving in an elliptical manner characteristic of other dipterans in swarms.
- 4 Swarms were classified as either ‘persistent’ or ‘transient’ in terms of their shape, size and stability. Wind was the most influential factor in disrupting swarms.
- 5 Swarms were confirmed as mating sites for C.impunctatus.

Abstract. Electroantennogram (EAG) and y-tube bioassays have been used to demonstrate the repellent properties of five plant compounds with host-seeking parous female Culicoides impunctatus Goetghebuer. The compounds were methyl salicylate and allyl-, butyl-, phenyl- and 2-phenylethyl isothiocyanate. EAG thresholds were 1 times 10-3 to 1 μg. In the bioassays, maximal repellencies occurred with 1 times 10˜2 to lOug. When each compound was combined with 1-octen-3-ol, a confirmed host-odour attractant for C.impunctatus females, additive effects were recorded in EAG assays and in bioassays, all of the compounds either reduced or reversed the attractancy of l-octen-3-ol. Of the isothiocyanates, allyl isothiocyanate was the most potent and when combined with 1-octen-3-ol in field trials, the attractant effect of l-octen-3-ol was reduced. 相似文献

Abstract. An indirect enzyme-linked immunosorbent assay (ELISA) has been developed to identify the bloodmeal sources of Culicoides impunctatus Goetghebuer. Nightly light trapping in Scotland, between April and September 1992, caught a total of 344 blood-engorged female C. impunctatus , all in May, June and July. The insects were stored in 70% ethanol and tested 5–7 months later. Bloodmeal sources were positively identified for 246 (71.5%). Bovine hosts were most common (38.4%), followed by deer (23.0%) and sheep (9.9%). Only one midge had fed on human blood. Relatively freshly fed (<24h), fully engorged females gave the clearest results. The wider applications of the technique to the study of Culicoides host preferences is discussed. 相似文献

The effects of Haemoproteus belopolskyi, Haemoproteus fringillae, and Haemoproteus lanii on the survival of their vector the biting midge Culicoides impunctatus were examined. Wild-caught females were infected experimentally by allowing them to feed on naturally infected birds. A group of flies, which fed on an uninfected bird, was used as a control. There was a highly significant difference in the survival rate between the noninfected group and all groups of infected flies for postfeeding (PF) periods 1-2, 3-4, 5-6, and 7-8 days. For days 7-8, the percentage of noninfected surviving flies was 4.4 times higher than the percentage of infected surviving flies, testifying to the detrimental effects of avian Haemoproteus infections on the longevity of C. impunctatus. The mortality rates of the infected flies were highest for days 1-2 and 3-4 PF, indicating possible negative effects of ookinetes and early oocysts on the vector. 相似文献

Responses of host-seeking female Culicoides impunctatus (Goetghebuer) (Diptera: Ceratopogonidae) to acetone, carbon dioxide and 1-octen-3-ol were measured in a wind tunnel. Carbon dioxide, presented as a filamentous plume, increased upwind flight in a dose-dependent manner, up to 0.09% concentration. A homogenous CO2 plume elicited similar upwind responses at concentrations up to 0.09%, whereas higher plume concentrations (> 0.1%) induced erratic responses with a suppression of upwind flight. Bovine equivalent concentrations of acetone (1.5 x 10(-6)g/l) and 1-octen-3-ol (1.3 x 10(-8)g/) failed to induce any significant upwind response when tested alone. In the presence of CO2, however, 1-octen-3-ol showed highly significant increases in upwind responses at concentrations of 1.3 x 10(-1) - 10(-8)g/l. Mixtures of CO2+ acetone also enhanced upwind flight at 1.5 x 10(-9)g/l. High tunnel concentrations of both 1-octen-3-ol and acetone inhibited upwind responses. These findings are discussed in relation to host finding by C. impunctatus and known mechanisms by which upwind flight is initiated and arrested at high odour concentrations. 相似文献

Abstract. 1-octen-3-ol, a component of the body odour of ruminants, is demonstrated to be an attractant for host-seeking, parous female Culicoides impunctatus Goetghebuer (Diptera, Ceratopogonidae).This compound was electrophysiologically and behaviourally active and, in the field, released at 0.11 ± 0.01 mg/day, increased female C.impunctatus catches over a seven-day period compared with control catches.When 1-octen-3-ol was combined either with live female midges or an extract of female midges in an electroantennogram assay, additive effects were recorded.Behaviourally, the same combination resulted in both enhanced attraction and repellency, depending on concentration.Attraction was recorded mainly when sub-threshold concentrations of the separate components were combined.The potential of such a combination as an effective field bait is discussed. 相似文献

Abstract. Electrophysiological, behavioural and field evidence is presented for a volatile attractant produced by host-seeking, parous female Culicoides impunctatus Goetghebuer. Attraction of host-seeking females to other live parous females or to extracts of parous females was demonstrated in Y-tube bioassays, as well as in field trials using baited sticky traps. Threshold numbers were 100–200 females, both in the laboratory and field. Antennal receptors to this pheromone were revealed by recording electroantennograms (EAGs) in response to odour from live parous females and to the extracts of female midges. The larval stage of the trombidioid mites Parafeiderium stuarti n. sp. (family Microtrombidiidae Thor, 1935) and Centrotrombidium blackwellae n. sp. (family Johnstonianidae Newell, 1957) are described. Both species were found parasitising Culicoides impunctatus Goetghebuer, the highland midge (Diptera: Ceratopogonidae), in Scotland. A revised diagnosis for Parafeiderium Vercammen-Grandjean & Cochrane, 1974 and a key to the world species of larval Centrotrombidium Kramer, 1896 are given. 相似文献

Herein, we describe the first case of a natural infection of biting midges by a kinetoplastid protozoan. Flagellates from a female Culicoides kibunensis captured in a bird's nest were introduced into culture and characterized by light and electron microscopy. However, because the morphological data were inconclusive, the novel endosymbiont-free trypanosomatid was assigned into Herpetomonas primarily on the basis of the 18S and 5S ribosomal RNA (rRNA) gene sequences. 相似文献

A microsporidium with ultrastructural characteristics of the genus Vavraia was found in the fat body of an adult specimen of Culicoides edeni (Diptera: Ceratopogonidae) collected in northern Florida. The sporogonial stages developed within sporophorous vesicles, which contained variable numbers of oval spores at maturity. The wall of the sporophorous vesicle was composed of two electron-dense outer layers and an electron-lucent intermediate layer. Sporonts contained haplokaryotic nuclei and divided by rosette formation. Mature spores had anisofilar polar filaments and measured 3.8 +/- 0.28 microns in length and 2.2 +/- 0.16 microns in width in thick sections of resin-embedded material. This is the first report of a Vavraia sp. from a species of Culicodes. 相似文献

Salivary proteins of Culicoides biting midges are thought to play a key role in summer eczema (SE), a seasonal recurrent allergic dermatitis in horses. The present study describes the identification, expression and clinical relevance of a candidate allergen of the North American midge Culicoides sonorensis. Immunoblot analysis of midge saliva revealed a 66 kDa protein (Cul s 1) that was bound by IgE from several SE-affected (SE+) horses. Further characterisation by fragmentation, mass spectrometry and bioinformatics identified Cul s 1 as maltase, an enzyme involved in sugar meal digestion. A cDNA encoding Cul s 1 was isolated and expressed as a polyhistidine-tagged fusion protein in a baculovirus/insect cell expression system. The clinical relevance of the affinity-purified recombinant Cul s 1 (rCul s 1) was investigated by immunoblotting, histamine release testing (HRT) and intradermal testing (IDT) in eight SE+ and eight control horses. Seven SE+ horses had rCul s 1-specific IgE, whereas only one control animal had IgE directed against this allergen. Furthermore, the HRT showed rCul s 1 induced basophil degranulation in samples from seven of eight SE+ horses but in none of the control animals. rCul s 1 also induced immediate (7/8), late-phase (8/8) and delayed (1/8) skin reactivity in IDT on all SE+ horses that had a positive test with the whole body extract (WBE) of C. sonorensis. None of the control horses showed immediate or delayed skin reactivity with rCul s 1, and only one control horse had a positive late-phase response, while several non-specific late-phase reactions were observed with the insect WBE. Thus, we believe rCul s 1 is the first specific salivary allergen of C. sonorensis to be described that promises to advance both in vitro and in vivo diagnosis and may contribute to the development of immunotherapy for SE in horses. 相似文献

The biting midge Culicoides imicola Kieffer (Diptera: Ceratopogonidae) is the most important Old World vector of African horse sickness (AHS) and bluetongue (BT). Recent increases of BT incidence in the Mediterranean basin are attributed to its increased abundance and distribution. The phylogenetic status and genetic structure of C. imicola in this region are unknown, despite the importance of these aspects for BT epidemiology in the North American BT vector. In this study, analyses of partial mitochondrial cytochrome oxidase subunit I gene (COI) sequences were used to infer phylogenetic relationships among 50 C. imicola from Portugal, Rhodes, Israel, and South Africa and four other species of the Imicola Complex from southern Africa, and to estimate levels of matrilineal subdivision in C. imicola between Portugal and Israel. Eleven haplotypes were detected in C. imicola, and these formed one well-supported clade in maximum likelihood and Bayesian trees implying that the C. imicola samples comprise one phylogenetic species. Molecular variance was distributed mainly between Portugal and Israel, with no haplotypes shared between these countries, suggesting that female-mediated gene flow at this scale has been either limited or non-existent. Our results provide phylogenetic evidence that C. imicola in the study areas are potentially competent AHS and BT vectors. The geographical structure of the C. imicola COI haplotypes was concordant with that of BT virus serotypes in recent BT outbreaks in the Mediterranean basin, suggesting that population subdivision in its vector can impose spatial constraints on BT virus transmission. 相似文献
