Control of nitrogen and carbon metabolism in root nodules   总被引:4,自引:0,他引:4  
Because legume root nodules have high rates of carbon and nitrogen metabolism, they are ideal for the study of plant physiology, biochemistry and molecular biology. Many plant enzymes involved in carbon and nitrogen assimilation have enhanced activity and enzyme protein in nodules as compared to other plant organs. For all intents and purposes the interior of the root nodule is O2 limited. Both plant and bacterial components of effective root nodules have unique adaptive features for maximizing carbon and nitrogen metabolism in an O2-limited environment. Plant glycolysis appears to be shunted to malic acid synthesis with further reductive synthesis to fumarate and succinate. Nodule bacteroids utilize these organic acids for the energy to fuel nitrogenase activity. Activities of the plant enzymes phosphoenolpyruvate carboxylase (PEPC, EC 4.1.1.31), malate dehydrogenase (MDH, EC 1.1.1.37) and aspartate aminotransferase (AAT, EC 2.6.1.1), which are very high in nodules, may mediate the flux of carbon between organic and amino acid pools. Dark CO2 fixation via nodule PEPC can provide up to 25% of the carbon needed for malate and aspartate synthesis. At least three of the plant proteins showing enhanced expression in root nodules are O2 regulated. Isolation of alfalfa cDNAs encoding PEPC, AAT, NADH-glutamate synthase (NADH-GOGAT, EC 1.4.1.14) and aldolase (EC 4.1.2.13) will offer new tools to assess molecular events controlling nodule carbon and nitrogen metabolism.  相似文献

The responses of three species of nitrogen-fixing trees to CO2 enrichment of the atmosphere were investigated under nutrient-poor conditions. Seedlings of the legume, Robinia pseudoacacia L. and the actinorhizal species, Alnus glutinosa (L.) Gaertn. and Elaeagnus angustifolia L. were grown in an infertile forest soil in controlled-environment chambers with atmospheric CO2 concentrations of 350 μl −1 (ambient) or 700 μl −1. In R. pseudoacacia and A. glutinosa , total nitrogenase (N2 reduction) activity per plant, assayed by the acetylene reduction method, was significantly higher in elevated CO2, because the plants were larger and had more nodule mass than did plants in ambient CO2. The specific nitrogenase activity of the nodules, however, was not consistently or significantly affected by CO2 enrichment. Substantial increases in plant growth occurred with CO2 enrichment despite probable nitrogen and phosphorus deficiencies. These results support the premises that nutrient limitations will not preclude growth responses of woody plants to elevated CO2 and that stimulation of symbiotic activity by CO2 enrichment of the atmosphere could increase nutrient availability in infertile habitats.  相似文献

The increased supply of photosynthate from maternal tissue is known to promote grain growth in several crop species. However, the effect of increasing photosynthate supply on grain growth receives little attention in rice. This study was aimed at evaluating the effect of increasing photosynthate supply through CO2 enrichment (650 μl I-1) on grain growth in three rice cultivars differing in grain size. CO2 enrichment was applied to the pot-grown plants between anthesis and final harvest. The results indicated that high CO2 treatment enhanced the CO2 exchange rate of leaf tissue, and subsequently increased the sucrose level of peduncle exudate, but it did not promote starch accumulation in the developing grains. This phenomenon was linked to the poor CO2 responses for the grain activities of sucrose synthase, UDP-glucose pyrophosphorylase. ADP-glucose pyrophosphorylase, and starch synthases involved in the conversion of sucrose to starch. Significant cultivar differences also existed for the activities of sucrose to starch conversion enzymes with larger grain size cultivars tending to have higher enzymes activities (expressed on a grain basis), resulting in a greater carbohydrate accumulation.  相似文献

The effects of mycorrhiza formation in combination with elevated CO2 concentrations on carbon metabolism of Norway spruce ( Picea abies ) seedlings and aspen ( Populus tremula × Populus tremuloides ) plantlets were analysed. Plants were inoculated for 6 wk with the ectomycorrhizal fungi Amanita muscaria and Paxillus involutus (aspen only) in an axenic Petri-dish culture at 350 and 700 μl l−1 CO2 partial pressure. After mycorrhiza formation, a stimulation of net assimilation rate was accompanied by decreased activities of sucrose synthase, an increased activation state of sucrose-phosphate synthase, decreased fructose-2,6-bisphosphate and starch, and slightly elevated glucose-6-phosphate contents in source leaves of both host species, independent of CO2 concentration. Exposure to elevated CO2 generally resulted in higher net assimilation rates, increased starch as well as decreased fructose-2,6-bisphosphate (aspen only) content in source leaves of both mycorrhizal and nonmycorrhizal plants. Our data indicate only slightly improved carbon utilization by mycorrhizal plants at elevated CO2. They demonstrate however, that both factors which modulate the sink-source properties of plants increase the capacity for sucrose synthesis in source leaves mainly by allosteric enzyme regulation.  相似文献

Soybean [ Glycine max (L.) Merr. cv. Hobbit] plants nodulated by Bradyhizobium japonicum strain USDA 110 were grown in pot cultures in severely P- and N-deficient soil and either colonized by the vesicular-arbuscular mycorrhizal (VAM) fungus Glomus mosseae (Nicol. & Gerd.) Gerd. and Trappe or fertilized with a high (HP) or low (LP) level of KH2PO4 (0.6 or 0.3 m M , respectively), After 7 weeks of growth, nodule and chloroplast activities (C2H2 reduction and CO2 exchange rate) were determined. Photosynthetic P-use efficiency of CO2 fixation was significantly higher in VAM than in HP plants, while that of nitrogenase activity was lower. The LP plants were intermediate in both respects. The ratio of nodule to chloroplast activity [mol C2H2 reduced (mol CO2 fixed)−1] was highest in HP and lowest in VAM plants. Root colonization by the VAM fungus significantly increased nodule number and dry weight and reduced nodule specific mass and activity in comparison to HP plants. In spite of lower nodule activity, VAM plants were significantly larger and had higher N concentrations than the HP plants. The results suggest nonnutritional. VAM-elicited and host-mediated effects on the symbiotic functions of the legume association.  相似文献

The objective of this investigation was to examine the effect of an elevated atmospheric CO2 partial pressure ( p CO2) on the N-sink strength and performance of symbiotic N2 fixation in Trifolium repens L. cv. Milkanova. After initial growth under ambient p CO2 in a nitrogen-free nutrient solution, T. repens in the exponential growth stage was exposed to ambient and elevated p CO2 (35 and 60 Pa) and two levels of mineral N (N-free and 7·5 mol m–3 N) for 36 d in single pots filled with silica sand in growth chambers. Elevated p CO2 evoked a significant increase in biomass production from day 12 after the start of CO2 enrichment. For plants supplied with 7·5 mol m–3 N, the relative contribution of symbiotically fixed N (%Nsym) as opposed to N assimilated from mineral sources (15N-isotope-dilution method), dropped to 40%. However, in the presence of this high level of mineral N, %Nsym was unaffected by atmospheric p CO2 over the entire experimental period. In plants fully dependent on N2 fixation, the increase in N yield reflects a stimulation of symbiotic N2 fixation that was the result of the formation of more nodules rather than of higher specific N2 fixation. These results are discussed with regard to physiological processes governing symbiotic N2 fixation and to the response of symbiotic N2 fixation to elevated p CO2 in field-grown T. repens .  相似文献

The effects of potassium (K) deficiency on growth, N2-fixation and photosynthesis in white clover ( Trifolium repens L.) were investigated using natural occurring gas fluxes on the nodules in real time of plants under three contrasting relative addition rates of K causing mild K deficiency, or following abrupt withdrawal of the K supply causing strong K deficiency of less than 0.65% in dry matter. A steady-state below-optimum K supply rate led to an increase in CO2-fixation per unit leaf surface area as well as per plant leaf surface. However, nitrogenase activity per unit root weight and per unit nodule weight was maintained, as was the efficiency with which electrons were allocated to the reduction of N2 in the nodules. Abrupt K removals stimulated nodule growth strongly without delay, but as K concentrations decreased in the plant tissue a significant decline in nitrogenase activity per unit root weight as well as per unit nodule mass occurred. Further, the rate of photosynthesis per unit leaf area was unaffected, while the CO2 acquisition for the plant as a whole increased due to an expansion of total leaf area whereas the leaf area per unit leaf weight was unaffected. The ratio between CO2-fixation and N2-fixation increased, although not statistically significant, under short-term K deprivation as well as under long-term low K supply indicating a downregulation of nodule activity following morphological and growth adjustments. This downregulation took place despite a partly substitution of the K by Na. It is concluded that N2-fixation does not limit the growth of K-deprived clover plants. K deprivation induces changes in the relative growth of roots, nodules, and shoots rather than changes in N and/or carbon uptake rates per unit mass or area of these organs.  相似文献

Acclimation of plants to an increase in atmospheric carbon dioxide concentration is a well described phenomenon. It is characterized by an increase in leaf carbohydrates and a degradation of ribulose 1, 5-bisphosphate carboxylase protein (Rubisco) leading in the long term to a lower rate of CO2 assimilation than expected from the kinetic constants of Rubisco. This article summarizes studies with transgenic plants grown in elevated pCO2 which are modified in their capacity of CO2 fixation, of sucrose and starch synthesis, of triosephosphate and sucrose transport and of sink metabolism of sucrose. These studies show that a feedback accumulation of carbohydrates in leaves play only a minor role in acclimation, because leaf starch synthesis functions as an efficient buffer for photoassimilates. There is some evidence that in elevated pCO2, plants grow faster and senescence is induced earlier.  相似文献

Dry weight (DW) and nitrogen (N) accumulation and allocation were measured in isolated plants of Danthonia richardsonii (Wallaby Grass) for 37 d following seed imbibition. Plants were grown at ≈ 365 or 735 μ L L–1 CO2 with N supply of 0·05, 0·2 or 0·5 mg N plant–1 d–1. Elevated CO2 increased DW accumulation by 28% (low-N) to 103% (high-N), following an initial stimulation of relative growth rate. Net assimilation rate and leaf nitrogen productivity were increased by elevated CO2, while N concentration was reduced. N uptake per unit root surface area was unaffected by CO2 enrichment. The ratio of leaf area to root surface area was decreased by CO2 enrichment. Allometric analysis revealed a decrease in the shoot-N to root-N ratio at elevated CO2, while the shoot-DW to root-DW ratio was unchanged. Allometric analysis showed leaf area was reduced, while root surface area was unchanged by elevated CO2, indicating a down-regulation of total plant capacity for carbon gain rather than a stimulation of mineral nutrient acquisition capacity. Overall, growth in elevated CO2 resulted in changes in plant morphology and nitrogen use, other than those associated simply with changing plant size and non-structural carbohydrate content.  相似文献

The effects of short-term NaCl-salinity on nodules of soybean ( Glycine max L. cv. Kingsoy) were studied on hydroponically-grown plants. Both acetylene reducing activity (ARA) and nodule respiration (O2 uptake and CO2 evolution) were immediately inhibited, and the stimulation of them by rising the external partial pressure of O2 (pO2) was diminished by the application of 0.1 M NaCl in the nutrient solution. The permeability of the nodule to O2 diffusion, estimated by O2 consumption or CO2 evolution, was significantly lower in the stressed nodules than in the cootrol ones. The respiratory quotient of intact nodules and the ethanol production of excised nodules were increased by low pO2 and by salt stress. These data confirm that in salt-stressed soybean nodules, O2 availability is reduced and fermentative pathways are stimulated.  相似文献

Plants grown in an environment of elevated CO2 and temperature often show reduced CO2 assimilation capacity, providing evidence of photosynthetic downregulation. The aim of this study was to analyse the downregulation of photosynthesis in elevated CO2 (700 µmol mol−1) in nodulated alfalfa plants grown at different temperatures (ambient and ambient + 4°C) and water availability regimes in temperature gradient tunnels. When the measurements were taken in growth conditions, a combination of elevated CO2 and temperature enhanced the photosynthetic rate; however, when they were carried out at the same CO2 concentration (350 and 700 µmol mol−1), elevated CO2 induced photosynthetic downregulation, regardless of temperature and drought. Intercellular CO2 concentration measurements revealed that photosynthetic acclimation could not be accounted for by stomatal limitations. Downregulation of plants grown in elevated CO2 was a consequence of decreased carboxylation efficiency as a result of reduced rubisco activity and protein content; in plants grown at ambient temperature, downregulation was also induced by decreased quantum efficiency. The decrease in rubisco activity was associated with carbohydrate accumulation and depleted nitrogen availability. The root nodules were not sufficiently effective to balance the source–sink relation in elevated CO2 treatments and to provide the required nitrogen to counteract photosynthetic acclimation.  相似文献

Respiratory responses of higher plants to atmospheric CO2 enrichment   总被引:5,自引:0,他引:5  
Although the respiratory response of native and agricultural plants to atmospheric CO2 enrichment has been reported over the past 75 years, only recently have these effects emerged as prominent measures of plant and ecosystem response to the earth's changing climate. In this review we discuss this rapidly expanding field of study and propose that both increasing and decreasing rates of leaf and whole-plant respiration are likely to occur in response to rising CO2 concentrations. While the stimulatory effects of CO2 on respiration are consistent with our knowledge of leaf carbohydrate status and plant metabolism, we wish to emphasize the rather surprising short-term inhibition of leaf respiration by elevated CO2 and the reported effects of long-term CO2 exposure on growth and maintenance respiration. As is being found in many studies, it is easier to document the respiratory response of higher plants to elevated CO2 than it is to assign a mechanistic basis for the observed effects. Despite this gap in our understanding of how respiration is affected by CO2 enrichment, data are sufficient to suggest that changes in leaf and whole-plant respiration may be important considerations in the carbon dynamics of terrestrial ecosystems as global CO2 continues to rise. Suggestions for future research that would enable these and other effects of CO2 on respiration to be unravelled are presented.  相似文献

Well-nodulated soybean ( Glycine max L. Merr. cv. Akisengoku) plants were allowed to assimilate 13CO2. Plant cytosol and bacteroid fractions were isolated from nodules, and the kinetics of [13C]-labelling of soluble carbohydrates, organic acids and amino acids were investigated.
The concentrations of all metabolites, with the exception of trehalose and 3-hydroxy-butyrate, were 10- to 1000-fold higher in plant cell cytosol than in bacteroids. The major portion of trehalose was found in bacteroids and 3-hydroxybutyrate only in bacteroids. Sucrose was most highly labelled with 13C in nodules, and the levels and time-course of labelling of sucrose were in good agreement with those of respired CO2 from the nodules. The levels and time-courses of labelling of sucrose were closely similar in cytosol and bacteroids. Glucose was less labelled than sucrose and the level of labelling was consistently higher in cytosol than in bacteroids. The levels of [13C]-labelling of organic acids and amino acids in nodules were lower than those of sucrose and of respired CO2. Tricarboxylic acid cycle intermediates, particularly succinate, were considerably less labelled in bacteroids than in the cytosol. All amino acids detected were also much more rapidly labelled in the cytosol. The results are discussed in relation to the utilization and possible compartmentation of carbon substrates in nodule tissues.  相似文献

Carob seedlings ( Ceratonia siliqua L. cv. Mulata), fed with nitrate or ammonium, were grown in growth chambers containing two levels of CO2 (360 or 800 μl l−1), three root temperatures (15, 20 or 25°C), and the same shoot temperature (20/24°C, night/day temperature). The response of the plants to CO2 enrichment was affected by environmental factors such as the type of inorganic nitrogen in the medium and root temperature. Increasing root temperature enhanced photosynthesis rate more in the presence of nitrate than in the presence of ammonium. Differences in photosynthetic products were also observed between nitrate- and ammonium-fed carob seedlings. Nitrate-grown plants showed an enhanced content of sucrose, while ammonium led to enhanced storage of starch. Increase in root temperature caused an increase in dry mass of the plants of similar proportions in both nitrogen sources. The enhancement of the rates of photosynthesis by CO2 enrichment was proportionally much larger than the resulting increases in dry mass production when nitrate was the nitrogen source. Ammonium was the preferred nitrogen source for carob at both ambient and high CO2 concentrations. The level of photosynthesis of a plant is limited not only by atmospheric CO2 concentration but also by the nutritional and environmental conditions of the root.  相似文献

The hypothesis is tested that crassulacean acid metabolism (CAM) in isoetids is a mechanism which not only conserves inorganic carbon but also plays a role in nitrogen economy of the plants. This hypothesis was tested in an outdoor experiment, where Littorella uniflora (L.) Aschers. were grown at two CO2 and five inorganic nitrogen concentrations in a crossed factorial design. The growth of Littorella responded positively to enhanced nitrogen availability at high but not at low CO2 indicating that growth was limited by nitrogen at high CO2 only. For the nitrogen-limited plants, the capacity for CAM (CAMcap) increased with the degree of nitrogen limitation of growth and an inverse coupling between CAM and tissue-N was found. Although this might indicate a role of CAM in economizing on nitrogen in Littorella , the hypothesis was rejected for the following reasons: (1) although CAMcap was related to tissue-N no relationship between tissue-N and ambient CAM activity (CAMambient) was found whereas a close relationship would be expected if CAM was regulated by nitrogen availability; (2) the photosynthetic nitrogen use efficiency for high CO2-grown plants declined with increased CAMambient and with CAMcap; and (3) growth per unit tissue-N per unit time declined with increased CAMambient and CAMcap.  相似文献

Changes in carbon metabolism and δ13C value of transgenic potato plants with a maize pyruvate,orthophosphate dikinase (PPDK; EC 2.7.9.1) gene are reported. PPDK catalyzes the formation of phospho enol pyruvate (PEP), the initial acceptor of CO2 in the C4 photosynthetic pathway. PPDK activities in the leases of transgenic potatoes were up to 5.4‐fold higher than those of control potato plants (wild‐type and treated control plants). In the transgenic potato plants, PPDK activity in leaves was negatively correlated with pyruvate content (r2= 0.81), and was positively correlated with malate content (r2= 0.88). A significant increase in the δ13C value was observed in the transgenic potato plants, suggesting a certain contribution of PEP carboxylase as the initial acceptor of atmospheric CO2. These data suggest that elevated PPDK activity may alter carbon metabolism and lead to a partial operation of C4‐type carbon metabolism. However, since parameters associated with CO2 gas exchange were not affected, the altered carbon metabolism had only a small effect on the total photosynthetic characteristics of the transgenic plants.  相似文献

Three year data on the effect of water- and mannitol (4%) priming of chickpea seeds (12 h at 25°C) showed higher number and biomass of nodules in the plants from primed seeds than from non-primed seeds. The biomass of nodules increased to 75 DAS but decreased by 90 DAS. Activities of sucrose metabolism enzymes (sucrose synthase (SS) and alkaline invertase) and of nitrogen metabolism (glutamine synthetase (GS), glutamate synthase (GOGAT) and glutamate dehydrogenase (GDH)) in nodules of primed and non-primed crops during development are reported. SS and alkaline invertase activities increased to 70 DAS and then decreased. In primed plants, the higher SS activity in nodules at 60 and 70 DAS might be responsible for providing more energy and carbon skeleton for nitrogen fixation and for ammonium assimilation in primed plants. At 85 DAS, though the SS activity decreased in comparison with the earlier growth stages, it was still higher in nodules of the primed crops than the non-primed crop. Activity of alkaline invertase was maximum at 70 DAS in the nodules of primed and non-primed crops. Priming increased nodule GS activity at 70 and 85 DAS. GOGAT activity was unaffected by priming but GDH activity was greater in nodules from primed crops at 50 DAS. Elevated SS and GS nodule activities in primed chickpeas might be responsible in increasing nodule biomass and metabolic activity thereby increasing seed fill.  相似文献

The effect of genetic factors in Rhizobium on host plant biomass production and on the carbon costs of N2 fixation in pea root nodules was studied. Nine strains of Rhizobium leguminosarum were constructed, each containing one of three symbiotic plasmids in combination with one of three different genomic backgrounds. The resulting strains were tested in symbiosis with plants of Pisum sativum using a flow-through apparatus in which nodule nitrogenase activity and respiration were measured simultaneously under steady state conditions. Nodules formed by strains containing the background of JI6015 had the lowest carbon costs of N2 fixation (7.10–8.10 μmol C/μmol N2), but shoot dry weight of those plants was also smaller than that of plants nodulated by strains with the background of B151 or JI8400. Nodules formed by these two strain types had carbon costs of N2 fixation varying between 11.26 and 13.95 μmol C/μmol N2. The effect of symbiotic plasmids on the carbon costs was relatively small. A time-course experiment demonstrated that nodules formed by a strain derived from JI6015 were delayed in the onset of nitrogenase activity and had a lower rate of activity compared to nodules induced by a strain with the background of B151. The relationship between nitrogenase activity, carbon costs of N2 fixation and host plant biomass production is discussed.  相似文献

Carbon and water fluxes in a calcareous grassland under elevated CO2   总被引:3,自引:2,他引:1  
1. As part of a long-term study of the effects of elevated CO2 on biodiversity and ecosystem function in a calcareous grassland, we measured ecosystem carbon dioxide and water-vapour fluxes over 24-h periods during the 1994 and 1995 growing seasons. Data were used to derive CO2 and H2O gas-exchange response functions to quantum flux density (QFD).
2. The relative increase in net ecosystem CO2 flux (NEC) owing to CO2 enrichment increased as QFD rose. Daytime NEC at high QFD under elevated CO2 increased by 25% to 60%, with the greatest increases in the spring and after mowing in June when above-ground biomass was lowest. There was much less stimulation of NEC in early June and again in October when the canopy was fully developed. Night-time NEC was not significantly altered under elevated CO2.
3. Short-term reversal of CO2 concentrations between treatments after two seasons of CO2 exposure provided evidence for a 50% downward adjustment of NEC expressed per unit above-ground plant dry weight. However, when expressed on a land area basis, this difference disappeared because of a c. 20% increase in above-ground biomass under elevated CO2.
4. Ecosystem evapotranspiration (ET) was not significantly altered by elevated CO2 when averaged over all measurement dates and positions. However, ET was reduced 3–18% at high QFD in plots at the top of the slope at our study site. In summary, CO2 enrichment resulted in a large stimulation of ecosystem CO2 capture, especially during periods of a large demand of carbon in relationship to its supply, and resulted in a relatively small and variable effect on ecosystem water consumption.  相似文献
