The endemic seagrass Halophila johnsonii grows intertidally to 3 m deep, in both marine and riverine influenced habitats of eastern Florida. Salinity and chromophoric dissolved organic matter (CDOM) levels widely fluctuate across this broad habitat range, changing tidally and with variable influx of freshwater from watershed runoff, river discharge and stochastic storm events. CDOM exponentially absorbs light in the UV to blue wavelengths, affecting optical water quality. H. johnsonii produces 15 flavonoid compounds that maximally absorb in the UV range. These flavonoids are thought to function as UV-protectants (UVP) in high-light and UV-intense environments. This mesocosm study examined the photosynthetic capacity, quantum efficiency and pigment content of H. johnsonii under experimental treatments of three salinities (10, 20 and 30) with and without CDOM. Main treatment effects and possible interactive effects at both short- (1 day to 1 week) and longer-term (1 month) time scales were examined. There were no significant CDOM or CDOM x salinity effects over any of the experimental treatment durations. There was 100% mortality of plants at salinity 10 after 10 days regardless of water color. UVP content of leaves was not affected by CDOM in this study, but there was significant variation in UVP in response to salinity. Our results do not support the primary role of UVP in this species as a sunscreen, but indicate that different salinity environments contribute to changes in the levels of these flavonoids. The UVP response to salinity stress response was not mitigated by a decrease in UV-radiation (increased CDOM) as H. johnsonii continued to put energy into the production of the carbon-rich flavonoids regardless of potential UV-stress. The experimental results indicate that prolonged hypo-salinity conditions are an important environmental factor to manage in the limited geographic range of H. johnsonii.  相似文献

Isolated trimeric Photosystem I complexes of the cyanobacterium Synechococcus elongatus have been studied with absorption spectroscopy and site-selective polarized fluorescence spectroscopy at cryogenic temperatures. The 4 K absorption spectrum exhibits a clear and distinct peak at 710 nm and shoulders near 720, 698 and 692 nm apart from the strong absorption profile located at 680 nm. Deconvoluting the 4 K absorption spectrum with Gaussian components revealed that Synechococcus elongatus contains two types of long-wavelength pigments peaking at 708 nm and 719 nm, which we denoted C-708 and C-719, respectively. An estimate of the oscillator strengths revealed that Synechococcus elongatus contains about 4–5 C-708 pigments and 5–6 C-719 pigments. At 4 K and for excitation wavelengths shorter than 712 nm, the emission maximum appeared at 731 nm. For excitation wavelengths longer than 712 nm, the emission maximum shifted to the red, and for excitation in the far red edge of the absorption spectrum the emission maximum was observed 10–11 nm to the red with respect to the excitation wavelength, which indicates that the Stokes shift of C-719 is 10–11 nm. The fluorescence anisotropy, as calculated in the emission maximum, reached a maximal anisotropy of r=0.35 for excitation in the far red edge of the absorption spectrum (at and above 730 nm), and showed a complicated behavior for excitation at shorter wavelengths. A newly developed laboratory fluorescence imaging system was used to obtain fluorescence images (FImage) of freshly excised cucumber (Cucumis sativus L.) leaves in spectral bands centered in the blue (F450), green (F550), red (F680), and far-red (F730) spectral regions that resulted from a broad-band (300-400 nm) excitation source centered at 360 nm. Means of relative fluorescence intensities (RFI) from these spectral fluorescence images were compared with spectral fluorescence emission data obtained from excitation wavelengths at 280 nm (280EX, 300-550 nm) and 380 nm (380EX, 400-800 nm) of dimethyl sulfoxide (DMSO) extracts from these leaves. All three fluorescence data types (FImage, 280EX, 380EX) were used to assess ultraviolet-B (UV-B, 280-320 nm) induced physiological changes and the possible use of N-[2-(2-oxo-1-imidazolidinyl) ethyl]-N′-phenylurea (EDU or ethylenediurea) as a chemical protectant against UV-B damage. Plants exhibited well known foliar growth and pigment responses to UV-B exposure (e.g., increased UV-B absorbing compounds and decreased leaf area, chlorophyll a content; and and lower chlorophyll a/b and chlorophyll/carotenoid pigment ratios). Since EDU alone had no effect on foliar variables, there was no evidence that EDU afforded protection against UV-B. Instead, EDU augmented some UV-B effects when provided in conjunction with UV-B irradiation (e.g., reductions in the chlorophyll/carotenoid ratio, total photosynthetic pigments, and chlorophyll b content).Relative fluorescence intensities (RFI) in the longer visible wavelengths (green, red, and far-red) were uncorrelated for comparisons between the FImage and 380EX data sets. However, blue and green RFI were significantly correlated (0.8r0.6; P ≤0.002) for comparisons between FImage and 280EX data sets. UV-B treatment caused an increase in blue RFI (e.g., F450) in both images and 280EX measurements. One explanation is that the UV-B excitation of both 280EX and FImage stimulates processes that produce excess blue fluorescence. The molecules that produce the excess blue fluorescence in both the 280EX and the Fimage data are different electron transfer agents that operate in parallel. For FImage, the UV excitation penetrates leaf surface layers to stimulate fluorescence from compounds in mesophyll and epidermal tissues (as occurs for the extracts of leaf discs), whereas emissions captured at longer, less energetic wavelengths, were primarily from the epidermal layer. UV-B irradiated leaves showed much greater heteorgeneity of RFI in both the green (F550FImag) and the red (F680FImag) bands than unirradiated leaves; this was true irrespective of EDU treatment.Although qualitative responses in individual bands differed between FImage and 380EX data, similar results were obtained in the detection of UV-B induced effects when the red/green and blue/far-red fluorescence ratios of these data were compared. The red/green ratio (either F680/F550FImage or F675/F525380EX) was lower for UV-B exposed plants in both images and 380EX data. UV-B exposure also significantly enhanced the blue/far-red ratio of images (F450/F740FImage) and the comparable 380EX ratio (F450/F730380EX) for the combined UV-B/EDU group. The far-red/red ratios were not useful in separating treatment effects in images or 380EX. Although comparable ratios were not available in 280EX data, the UV/blue ratio (F315/F420280EX) was substantially reduced by UV-B exposure and was inversely related to total photosynthetic pigment content. These findings suggest that the red/green ratio (FImage, 380EX) and the UV/blue ratio (280EX) may be as useful as the blue/far-red ratio (380EX) reported previously in detection of UV-B stress. Furthermore, the results support the validity of the imaging technique as a non-destructive diagnostic tool for assessing UV-B stress damage in plants.  相似文献

Very little is known about the physiological interactions between plants and endophytic bacteria. We investigated the impact of three endophytic bacteria, Bacillus pumilus 2-1, Chryseobacterium indologene 2-2, and Acinetobacter johnsonii 3-1, on the photosynthetic capacity and growth of sugar beet. Endophyte-free plants were obtained first and infected with the bacteria. Measurements of total chlorophyll content revealed very significant differences between endophyte-free beet plants and some infected by endophytic bacteria. The maximum photochemical yield (Fv/Fm) was used to determine any photosynthetic effect on plants caused by biotic or abiotic factors. After 30 days of growth, there was significantly higher Fv/Fm for endophyte-infected than endophyte-free plants. The light response curves of beet showed that photosynthetic capacity was significantly increased in endophyte-infected plants. Photosynthesis of endophyte-free plants was saturated at 1,300 μmol m−2 s−1, whereas endophyte-infected plants were not saturated at the irradiance used. The effect seemed to be due to promotion of electron transport in the thylakoid membranes. Promotion of photosynthetic capacity in sugar beet was due to increased chlorophyll content, leading to a consequent increased carbohydrate synthesis. It is possible that the increased maximum yield of photosynthesis in sugar beet was promoted by phytohormones and produced by the bacteria.  相似文献

The effects of light-induced non-photochemical quenching on the minimal Fo, and variable Fv, fluorescence emissions at 690 and 730 nm in leaves were determined. Non-photochemical quenching of Fo, but not Fv, was found to be dependent upon the wavelength of emission, and was greater at 690 nm than at 730 nm. For emission at 730, compared to at 690 nm, approx. 30% of Fo was not affected by non-photochemical quenching processes in leaves of C3 plants; in maize leaves this was found to be approx. 50%. The data indicate that a substantial proportion of the pigments contributing to Fo emission at 730 nm are not quenched by light-induced, non-photochemical quenching processes and that there are large differences in the pigment matrices contributing to Fo and Fv emissions at 730 nm, compared to those at 690 nm. These findings have important implications for the accurate estimation and interpretation of non-photochemical quenching of fluorescence parameters and their use in the calculation of photochemical efficiencies in leaves. Measurements of fluorescence emissions at wavelengths above 700 nm are likely to give rise to significant errors when used for determinations of photochemical and non-photochemical quenching parameters.  相似文献

Phototaxis of the blue-green alga Anabaena variabilis was studied using both population method and observation of single trichomes by microscope. The trichomes react positively at low and negatively at high illuminance. The inversion point lies at about 1000 1x. The action spectrum of positive phototaxis indicates that the photosynthetic pigments chlorophyll a, C-phycocyanin and allo-phycocyanin are involved in the absorption of the active light. The same range of wavelengths is active in negative phototaxis, but in addition, wavelengths between 500 and 560 nm and between 700 and 750 nm are also effective. Obviously pigments of unknown chemical nature are sharing in light absorption. Two alternatives are discussed. 以闽江河口鳝鱼滩湿地互花米草(Spartina alterniflora)的实测冠层高光谱反射率和叶片光合色素含量(LPPC)为数据源,在分析LPPC与原始光谱反射率、一阶导数光谱反射率、22种已报道光谱指数和14种新构建的植被指数相关性的基础上,利用直线回归、指数回归、对数回归以及乘幂回归方法,系统地比较了36种植被指数在估算互花米草LPPC中的表现。研究表明:(1)一阶导数光谱反射率组合的植被指数用于估算互花米草的LPPC优于原始光谱反射率;(2)红边区域一阶导数光谱是估测互花米草LPPC的最佳波段;(3)对于单一色素含量的估算,叶绿素a(Chla)的最佳估算指数为FDNDVI[723,703];叶绿素b(Chlb)的最佳估算指数为FDRVI[723,525];类胡萝卜素(Cars)的最佳估算指数为FDNDVI[723,703];(4)对于使用统一参量同时估算Chla、Chlb、Cars,由FDRVI[723,703]建立的对数估算模型效果最佳。研究成果可为湿地植物生化参量反演提供参考,也可为闽江河口湿地入侵种互花米草的动态监测和生态评估管理提供有力的科学依据。  相似文献

We measured the responses of pigments and chlorophyll a fluorescence parameters of the Antarctic leafy liverwort Cephaloziella varians to snowmelt during austral spring 2005 at Rothera Point on the western Antarctic Peninsula. Although no changes to the concentrations of UV-B photoprotective pigments were detected during snowmelt, chlorophyll and carotenoid concentrations and maximum photosystem (PS)II yield (F v /F m) were respectively 88, 60 and 144% higher in the tissues of the liverwort that had recently emerged from snow than in those under a 10 cm depth of snow. A laboratory experiment similarly showed that effective PSII yield increased rapidly within the first 45 min after plants sampled from under snow were removed to an illuminated growth cabinet. The pigmentation and PSII yields of plants during snowmelt were also compared with those of plants in January, during the middle of the growing season at Rothera Point. During snowmelt, plants had lower F v /F m values, chlorophyll a/b ratios and concentrations of UV-B photoprotective pigments and carotenoids than during mid-season, suggesting that although there is some recovery of PSII activity and increases in concentrations of photosynthetic pigments during snowmelt, the metabolism of C. varians is restricted during this period.  相似文献

The influence of the arbuscular mycorrhizal (AM) fungus, Glomus mosseae, on characteristics of growth, photosynthetic pigments, osmotic adjustment, membrane lipid peroxidation and activity of antioxidant enzymes in leaves of tomato (Lycopersicon esculentum cv Zhongzha105) plants was studied in pot culture under low temperature stress. The tomato plants were placed in a sand and soil mixture at 25°C for 6 weeks, and then subjected to 8°C for 1 week. AM symbiosis decreased malondialdehyde (MDA) content in leaves. The contents of photosynthetic pigments, sugars and soluble protein in leaves were higher, but leaf proline content was lower in mycorrhizal than non-mycorrhizal plants. AM colonization increased the activities of superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and ascorbate peroxidase (APX) in leaves. The results indicate that the AM fungus is capable of alleviating the damage caused by low temperature stress on tomato plants by reducing membrane lipid peroxidation and increasing the photosynthetic pigments, accumulation of osmotic adjustment compounds, and antioxidant enzyme activity. Consequently, arbuscular mycorrhiza formation highly enhanced the cold tolerance of tomato plant, which increased host biomass and promoted plant growth.  相似文献

Leaves of spinach (Spinacia oleracea, cv. Ispolinskii) were preilluminated by low-intensity light (1.0 and 1.5 W/m2, 0.5?C3.0 h) with wavelengths ranging from 530 to 730 nm to study the effect of this pretreatment on the activity of photosystem II (PS II), content of photosynthetic pigments, and peroxidase activity in excised leaves exposed to UV-A irradiation. Irradiation of leaves with UV-A suppressed the activity of PS II, reduced the content of chlorophylls (a + b) and carotenoids, and increased the peroxidase activity. Preillumination of leaves with red light (RL, 620?C660 nm) alleviated the inhibitory action of UV-A on PS II activity and reduced the pigment losses but increased the peroxidase activity in leaves and thylakoid membrane preparations, as compared to the respective effects of UV-A light applied without preillumination. The preexposure of leaves to red light alternating with far-red light (FR, 730 nm) removed partly the influence of RL on the parameters under study, which indicates the involvement of phytochrome active form, PFR into stress-induced defense responses in leaves. It is supposed that elevated resistance of photosynthetic apparatus to UV-A radiation was formed with the involvement of PFR and the antioxidant system induced by oxidative stress after preillumination of leaves with red light  相似文献

Endophyte-infected (EI) seeds of Lolium perenne L. were used to attain endophyte-free (EF) population by heating the seeds at 43°C for 15 min and then 57°C for 25 min. Relative water content (RWC), chlorophyll, soluble sugar and starch content of EI and EF populations under normal and drought stress conditions were compared to investigate the effect of endophyte infection on the host plant. Under severe stress, RWC of EI leaf was significantly higher than that of EF leaf, i.e. EI plants took more advantages over EF plants in water-holding ability. Under mild stress, endophyte could enhance soluble sugars in host plants to improve their osmotic ability. With stress intensification, the improvement of endophyte no longer existed, and more photosynthetic products (such as starch) accumulated in EI plants to survive through the undesirable conditions. In the next spring, EI populations will recover more rapidly than EF populations. The biomass of a population is closely related to its photosynthesis. Under severe stress, EI population significantly accumulated more biomass than EF population. In the present study, ameliorative capabilities of wuxal amino (bio stimulant) under salt stress has been investigated through adaptive mechanisms and antioxidant potential in tomato plants. In the experiment, two different concentrations (2 cm L-1 and 3 cm L-1) of wuxal amino through foliar application and soil irrigation were applied to the salt (150 mM) treated tomato plants and then morphological traits, photosynthetic pigments, osmolytes, secondary metabolites, oxidative stress and antioxidant enzymes activity were assessed at 60 days after planting. The results revealed that salt stress decreased the growth parameters, photosynthetic pigments, soluble sugars and soluble protein whereas, content of proline, ascorbic acid, total phenols, malondialdehyde, hydrogen peroxide and the activity of antioxidant enzymes activity increased under salt stress. Moreover, Wuxal amino application through foliar or soil to salt stressed plants improved morphological traits, photosynthetic pigments, osmolytes, total phenol and antioxidant enzymes activity. Interestingly, the deleterious impact of salinity on tomato plants were significantly reduced and it can be evident from reduced MDA and H2O2 levels. These responses varied with the mode (foliar or soil) of application of Wuxal amino under different concentrations (2 cm L-1 and 3 cm L-1). It was concluded that application of Wuxal amino (2 cm L-1, foliar) and (3 cm L-1; soil) proved best and could be commercially used as eco-friendly tool for the protection of tomato plants grown under salinity stress.  相似文献

Fluence-rate response curves for wavelengths from 640 nm to 730 nm were constructed for the day-extension promotion of flowering in green, light-grown, wheat (Triticum aestivum L., cv. Alexandria), a long-day plant. The resultant action spectrum had action maxima at 660 nm and 716 nm and resembles spectra for the high-irradiance reaction (HIR) seen in etiolated plants. The response of barley (Hordeum vulgare L. cv. Akcent) to various photosynthetic photon flux densities (PPFDs) and elevated [CO2] [700 μmol (CO2) mol−1; EC] was studied by gas exchange, chlorophyll (Chl) a fluorescence, and pigment analysis. In comparison with barley grown under ambient [CO2] [350 μmol (CO2) mol−1; AC] the EC acclimation resulted in a decrease in photosynthetic capacity, reduced stomatal conductance, and decreased total Chl content. The extent of acclimation depression of photosynthesis, the most pronounced for the plants grown at 730 μmol m−2 s−1 (PPFD730), may be related to the degree of sink-limitation. The increased non-radiative dissipation of absorbed photon energy for all EC plants corresponded to the higher de-epoxidation state of xanthophylls only for PPFD730 barley. Further, a pronounced decrease in photosystem 2 (PS2) photochemical efficiency (given as FV/FM) for EC plants grown at 730 and 1 200 μmol m−2 s−1 in comparison with AC barley was related to the reduced epoxidation of antheraxanthin and zeaxanthin back to violaxanthin in darkness. Thus the EC conditions sensitise the photosynthetic apparatus of high-irradiance acclimated barley plants (particularly PPFD730) to the photoinactivation of PS2. During the photosynthetic process, highly organized membranal assemblies convert light into biochemical energy with high efficiency. We have used whole-mount cryo-electron tomography to study the intracellular architecture of the photosynthetic membranes of the anaerobic purple photosynthetic bacterium Rhodopseudomonas viridis, as well as the organization of the photosynthetic units within the membranes. Three-dimensional reconstruction demonstrates a continuity of the plasma membrane with the photosynthetic membranes that form tunnel-like structures with an average diameter of 31 nm ± 8 nm at the connection sites. The spacing between the photosynthetic membranes at their cytoplasmic faces was found to be 11 nm, thus enforcing a highly close packaging of the photosynthetic membranes. Analysis of successive tomographic slices allowed for derivation of the spacing between adjacent photosynthetic core complexes from a single-layered photosynthetic membrane, in situ. This analysis suggests that most, if not all, photosynthetic membranes in R. viridis are characterized by a similar two-dimensional hexagonal lattice organization.  相似文献

The visual pigments and oil droplets in the retina of the diurnal gecko Gonatodes albogularis were examined microspectrophotometrically, and the spectral sensitivity under various adapting conditions was recorded using electrophysiological responses. Three classes of visual pigments were identified, with max at about 542, 475, and 362 nm. Spectral sensitivity functions revealed a broad range of sensitivity, with a peak at approximately 530–540 nm. The cornea and oil droplets were found to be transparent across a range from 350–700 nm, but the lens absorbed short wavelength light below 450 nm. Despite the filtering effect of the lens, a secondary peak in spectral sensitivity to ultraviolet wavelengths was found. These results suggest that G. albogularis does possess the visual mechanisms for discrimination of the color pattern of conspecifics based on either hue or brightness. Attenuation of UV radiation by plant cuticles from woody species   总被引:8,自引:0,他引:8  
Transmittance spectra of isolated plant cuticles were measured in the wavelength range from 270 to 600 nm. The cuticles were enzymatically isolated from the leaves of 27 species (26 evergreen or deciduous woody, one succulent herbaceous) and from four species of fruits. With the exception of subtropical and tropical species all plants were cultivated in the field. The cuticles of the species studied strongly attenuated ultraviolet (UV) radiation at wavelengths < 400 nm while they were practically translucent in the visible range. Relatively broad transmittance minima occurred at wavelengths from 280 to 320 nm (UV-B). Spectral transmittances at 300 nm ranged from 0.004 (Ilex aquifolium) to 0.50 (Prunus avium) for leaf cuticles and from 0.00023 (Cydonia oblonga) to 0.005 (Mains domestica) for fruit cuticles. The constitutive UV protection by cuticular pigments may be supplemented, to varying degrees, by pigments located in the epidermal cell wall and protoplast. Thus, it is concluded that only a small fraction of incident UV-B radiation may actually reach the sensitive tissues of the leaves of non-herbaceous species and of fruits.  相似文献

Estimates of global riverine nitrous oxide (N2O) emissions contain great uncertainty. We conducted a meta‐analysis incorporating 169 observations from published literature to estimate global riverine N2O emission rates and emission factors. Riverine N2O flux was significantly correlated with NH4, NO3 and DIN (NH4 + NO3) concentrations, loads and yields. The emission factors EF(a) (i.e., the ratio of N2O emission rate and DIN load) and EF(b) (i.e., the ratio of N2O and DIN concentrations) values were comparable and showed negative correlations with nitrogen concentration, load and yield and water discharge, but positive correlations with the dissolved organic carbon : DIN ratio. After individually evaluating 82 potential regression models based on EF(a) or EF(b) for global, temperate zone and subtropical zone datasets, a power function of DIN yield multiplied by watershed area was determined to provide the best fit between modeled and observed riverine N2O emission rates (EF(a): R2 = 0.92 for both global and climatic zone models, n = 70; EF(b): R2 = 0.91 for global model and R2 = 0.90 for climatic zone models, n = 70). Using recent estimates of DIN loads for 6400 rivers, models estimated global riverine N2O emission rates of 29.6–35.3 (mean = 32.2) Gg N2O–N yr−1 and emission factors of 0.16–0.19% (mean = 0.17%). Global riverine N2O emission rates are forecasted to increase by 35%, 25%, 18% and 3% in 2050 compared to the 2000s under the Millennium Ecosystem Assessment's Global Orchestration, Order from Strength, Technogarden, and Adapting Mosaic scenarios, respectively. Previous studies may overestimate global riverine N2O emission rates (300–2100 Gg N2O–N yr−1) because they ignore declining emission factor values with increasing nitrogen levels and channel size, as well as neglect differences in emission factors corresponding to different nitrogen forms. Riverine N2O emission estimates will be further enhanced through refining emission factor estimates, extending measurements longitudinally along entire river networks and improving estimates of global riverine nitrogen loads.  相似文献

Sub-aerial biofilms (SABs) are ubiquitous microbial communities that develop at the interface between hard surfaces and the atmosphere. Inherent SAB “core-settlers” include phototrophic algae, cyanobacteria, heterotrophic bacteria and microcolonial fungi (MCF). SABs do not simply cover hard surfaces; they interact with them in myriads of ways and bind to the underlying substrate. Secretion of extracellular mucilage aids adhesion, while organic acids and acidic polysaccharides weather the surface. As protection against solar radiation, many members of the SAB consortia produce shielding pigments while the phototrophic inhabitants are laden with photosynthetic pigments. All absorb light of many wavelengths and in addition, the cells themselves scatter light. Both effects change the spectra of incoming radiation (including wavelengths that are converted to electricity by photovoltaic cells) and decrease its intensity. To quantify these effects on SABs as complex entities of organisms and pigments, we measured the spectral properties of model and natural biofilms transferred to glass. Here we show that SABs growing on solar panels and other substrates scatter incident radiation between 250 nm up to 1800 nm and block up to 70% of its transmission. Model biofilms have the advantage that their microbial components can be “tuned” to resemble natural ones of different compositions thus providing a novel materials-testing tool.  相似文献
