A normalized cDNA library was constructed from the adductor muscle of M. yessoensis and acquired 4595 high quality expressed sequence tags (ESTs). After clustering and assembly of the ESTs, 3061 unigenes containing 654 contigs and 2407 singletons were identified. The contig length ranged from 266 bp to 2364 bp and the average length of these contigs was 544 bp. Blastx nonredundant protein database analysis showed that 1522 unigenes had significant homology to known genes (E value ≤ 10? 5). By comparing to Clusters of Orthologous Groups (COG) categories, 460 unigenes were annotated (E value ≤ 10? 10). Using Kyoto Encyclopedia of Genes and Genomes (KEGG), 345 of 3061 unigenes were assigned into 103 pathways (E value ≤ 10? 5). For InterProScan searches, 1237 unigenes were annotated containing 727 different types of protein domains. 941 of the 1237 unigenes were annotated for Gene Ontology (GO) classification using Uniprot2GO associations in any category (biological, cellular, and molecular). By sequences comparability and analysis of Blastx NCBI nonredundant protein database and KEGG, 66 unigenes were identified that may be involved in genetic information processing based on the known knowledge. The study provides a material basis as useful information for the genomic analysis of shellfish.  相似文献

AimsThis study aimed to investigate the effects of cytidine-5′-diphosphocholine (CDP-choline), an endogenous lipid precursor, on the reactivity of the mouse gastric fundus and to determine the mechanism(s) mediating its effects.Main methodsPossible contractile effect of CDP-choline (10? 5–10? 2 M) was investigated in the absence and presence of a muscarinic receptor antagonist, atropine (3 × 10? 6 M), an acetylcholine esterase inhibitor, physostigmine (10? 6 M), a Na+ channel blocker, tetrodotoxin (TTX, 3 × 10? 6 M), a Rho-kinase inhibitor, Y-27632 (10? 5 M), a purinoceptor antagonist, suramin (2 × 10? 4 M), a nitric oxide synthase inhibitor, NG-nitro-L-arginine (L-NA, 3 × 10? 4 M), a Ca2+ channel blocker, nifedipine (10? 6 M), an α7 nicotinic receptor antagonist, methyllycaconitine citrate (MLA, 10? 6 M) and a G protein (Gi/o) inhibitor, pertussis toxin (PTX, 2 μg/ml). The metabolites of CDP-choline, namely choline (10? 4–10? 2 M), cytidine 5′-triphosphate (CTP, 10? 5–10? 2 M), cytidine (10? 5–10? 2 M) and cytidine monophosphate (CMP, 10? 3–10? 2 M) were also tested. Besides, phosphorylation of MYPT1, which indicates Rho-kinase activity, was also detected.Key findingsCDP-choline produced contractions in a concentration-dependent manner. The contractions were not affected by atropine, physostigmine, TTX, PTX, MLA or L-NA. However, Y-27632, suramin or nifedipine partly reduced these contractions. CDP-choline increased phosphorylation of MYPT1. Among CDP-choline metabolites, cytidine had no contractile effects. However, choline induced considerable contractions, which were sensitive to atropine. CMP and CTP had also contractile activity, comparable to that of CDP-choline.SignificanceThese results suggest that CDP-choline produced contraction through, at least in part, purinoceptors and Rho/Rho-kinase signalling in the mouse gastric fundus.  相似文献

A large quantity of leaf litter was left on soil surface after soybean (Glycine max) harvest in the black soil region, northeast of China, where soybean was planted with the largest area. This paper investigated the effects of different fall tillage practices on soybean leaf litter sequestration into soil, and the subsequently durative effects on soil biological and biochemical properties during the next growing season. Two practices were investigated, fall tillage (T) and no fall tillage (NT) after soybean harvest in autumn. Results showed that the residue biomass on soil surface and in subsoil profile (0–20 cm) after soybean harvest was about 1450 kg ha?1 and 340 kg ha?1, respectively in October 2006. The residue biomass on soil surface and in subsoil profile was about 84 kg ha?1, 1581 kg ha?1 for T, and 423 kg ha?1, 340 kg ha?1 for NT respectively in May 2007. It was obvious that T practice can more effectively sequester leaf litter into soil compared to NT. Results also showed that T practices after soybean harvest eminently improved soil microbial carbon biomass and nitrogen biomass contents, and significantly improved soil urease and acid phosphate activities than NT. No significant difference of dehydrogenase activity was found between N and NT. The positive effects of T treatment on Soil microbial properties and soil enzymes activities among the next growing season due to soybean residues sequestration performed durative profit.  相似文献

Effects of irradiance and iron on the growth of a typical harmful algal blooms (HABs) causative dinoflagellate, Scrippsiella trochoidea, were investigated under various irradiances (high light: 70 μmol m?2 s?1 and low light: 4 μmol m?2 s?1) and iron concentrations (low iron: 0.063 mg L?1, medium iron: 0.63 mg L?1 and high iron: 6.3 mg L?1), and evaluated by the parameters of algal cell density, specific growth rate, optical density and chlorophyll a content. The results indicated that there was significant difference in the cell density of dinoflagellate S. trochoidea between high light and low light intensity treatments across the entire experiments, 7-fold higher at high irradiance as compared with low irradiance, which was further enhanced by the iron concentration. It was found that the maximum cell density of 25 × 104 cell mL?1 occurred under the combination of high light intensity and high iron concentration, followed by 23 × 104 cell mL?1 in the combination of high light and medium iron, and 20 × 104 cell mL?1 in the combination of high light and low iron. There was no significant effect of iron concentration on the cell density under low light intensity. The cell density maintained about 3 × 104 cell mL?1 across all combinations of iron concentrations and low light in the end of experiments. Such interactive effects of light intensity and iron level dependent were also observed for the specific growth rate, OD680 and chlorophyll a content of S. trochoidea. The maximum values of specific growth rate, OD680 and chlorophyll a content peaked at the condition of high irradiance and high iron, which were 0.22 d?1, 0.282 and 0.673 mg L?1, respectively. In general, their values increased significantly with the increasing of iron concentration at high irradiance, whereas no significant difference was observed among three iron concentrations at low irradiance, all remaining approximately 0.06 d?1, 0.03 and 0.050 mg L?1, respectively. Those results suggest that there may be a strong interactive effect between irradiance and iron on microalgal growth and their physiological characteristics. The combination of high light and high iron concentration may accelerate algal cell growth and pigment biosynthesis, thus leading to massive occurrence of HABs.  相似文献

Stressful situations are a major problem in aquaculture because they affect the immune system. 2-Deoxy-d-glucose (2-DG) is a derivative of a glucose analogue that reduces the availability of energy, thereby inhibiting cell metabolism so that it is unable to enter the glycolysis pathway. In this paper, 2-DG has been administered in order to study if the immune function is compromised during metabolic stress. Blood glucose level was measured as an indicator of the inhibition of glycolysis, and the effects of intraperitoneal administration of 2-DG on the main parameters of the humoral (complement, IgM levels and peroxidase activity in blood plasma) and cellular (respiratory burst, intracellular peroxidase level and phagocytosis activity) immune parameters of gilthead seabream (Sparus aurata, L) were evaluated. Furthermore, the expression levels of immune-associated genes (CSF-1R, NCCRP-1, Hep, TCR-β, IgMH, MHC-IIα, C3 and IL-1β) were analyzed by real-time PCR in head-kidney. A total of 5 intraperitoneal injections were performed at 48 h intervals. Three experimental groups were established: a control group injected with phosphate buffer saline, group 2-DG 500 and group 2-DG 750 injected with 500 mg kg?1 and 750 mg kg?1 2-DG, respectively (N = 15). After the third and fourth injection, some specimens of both DG-treated groups died. Following the first and third injection, the blood glucose levels of both 2-DG treated groups increased to a statistically significant extent with respect to the control group. While the humoral immune parameters were not significantly affected as a consequence of 2-DG administration, the cellular activities of leucocytes were. The injection of 500 mg kg?1 2-DG provoked up- or down-regulation of the immune-relevant genes analyzed, while the injection of 750 mg kg?1 always caused down-regulation of these genes. The results suggest that 2-DG provokes metabolic stress, which reduces the activities carried out by immune cells (leucocytes) and induces down-regulation of the immune-relevant genes analyzed when the energy available to the cell decreases.  相似文献

Effects of nitrate (2.0, 2.5, and 3.0 g L?1) and salt (0.5, 1.0, 1.5, 2.0 g L?1) concentrations on biomass production by Spirulina platensis was examined in the Schlösser medium. The highest (p < 0.001) biomass yields and chlorophyll a content was observed at 2.5 g L?1 nitrate and 1.5 g L?1 NaCl as 3.495 g L?1 and 29.92 mg L?1, respectively. Increment rate of biomass production was especially found between 72 and 216 h. Modified Richards, Schnute, Logistic and Gompertz models was successfully predicted (r2 > 0.96 and RSS ? 0.003) biomass production by S. platensis as function of nitrate and salt concentrations. Low residual sum of squares (RSS) and high regression coefficients (r2) indicated that used models were well fitted to the experiment data and it could be regarded as sufficient to describe biomass production of Spirulina sp. Biological variables i.e. production rate (μ) and lag time (λ) for S. platensis ranged 0.012–0.034 h?1 and 2.43–5.85 h, respectively from biomass production were successfully predicted by modified Logistic model according to low RSS and F-testing value.  相似文献

Proteins containing reactive cysteine residues (protein-Cys) are receiving increased attention as mediators of hydrogen peroxide signaling. These proteins are mainly identified by mining the thiol proteomes of oxidized protein-Cys in cells and tissues. However, it is difficult to determine if oxidation occurs through a direct reaction with hydrogen peroxide or by thiol–disulfide exchange reactions. Kinetic studies with purified proteins provide invaluable information about the reactivity of protein-Cys residues with hydrogen peroxide. Previously, we showed that the characteristic UV–Vis spectrum of horseradish peroxidase compound I, produced from the oxidation of horseradish peroxidase by hydrogen peroxide, is a simple, reliable, and useful tool to determine the second-order rate constant of the reaction of reactive protein-Cys with hydrogen peroxide and peroxynitrite. Here, the method is fully described and extended to quantify reactive protein-Cys residues and micromolar concentrations of hydrogen peroxide. Members of the peroxiredoxin family were selected for the demonstration and validation of this methodology. In particular, we determined the pKa of the peroxidatic thiol of rPrx6 (5.2) and the second-order rate constant of its reactions with hydrogen peroxide ((3.4 ± 0.2) × 107 M? 1 s? 1) and peroxynitrite ((3.7 ± 0.4) × 105 M? 1 s? 1) at pH 7.4 and 25 °C.  相似文献

It is assumed that small herbivores produce negligible amounts of methane, but it is unclear whether this is a physiological peculiarity or simply a scaling effect. A respiratory chamber experiment was conducted with six rabbits (Oryctolagus cuniculus, 1.57 ± 0.31 kg body mass) and six guinea pigs (Cavia porcellus, 0.79 ± 0.07 kg) offered grass hay ad libitum. Daily dry matter (DM) intake and DM digestibility were 50 ± 6 g kg? 0.75 d? 1 and 55 ± 6% in rabbits and 59 ± 11 g kg? 0.75 d? 1 and 61 ± 3% in guinea pigs, respectively. Methane production was similar for both species (0.20 ± 0.10 L d? 1 and 0.22 ± 0.08 L d? 1) and represented 0.69 ± 0.32 and 1.03 ± 0.29% of gross energy intake in rabbits and guinea pigs, respectively. In relation to body mass (BM) guinea pigs produced significantly more methane. The data on methane per unit of BM obtained in this study and from the literature on the methane output of elephant, wallabies and hyraxes all lay close to a regression line derived from roughage-fed horses, showing an increase in methane output with BM. The regression, including all data, was nearly identical to that based on the horse data only (methane production in horses [L d? 1] = 0.18 BM [kg]0.97 (95%CI 0.92–1.02)) and indicates linear scaling. Because feed intake typically scales to BM0.75, linear scaling of methane output translates into increasing energetic losses at increasing BM. Accordingly, the data collection indicates that an increasing proportion of ingested gross energy is lost because relative methane production increases with BM. Different from ruminants, such losses (1%–2% of gross energy) appear too small in non-ruminant herbivores to represent a physiologic constraint on body size. Nevertheless, this relationship may represent a physiological disadvantage with increasing herbivore body size.  相似文献

Rapeseed meal, a major byproduct of biodiesel production, has been used as a low-cost raw material for the production of a generic microbial feedstock through a consolidated bioconversion process. Various strategies were tested for the production of a novel fermentation medium, rich in free amino nitrogen (FAN): commercial enzymes (CEs) (2.7 mg g?1 dry meal), liquid state fungal pre-treatment (LSF) using Aspergillus oryzae (4.6 mg g?1), liquid state fungal pre-treatment followed by fungal autolysis (LSFA) (9.13 mg g?1), liquid state pre-treatment using fungal enzymatic broth (EB) (2.1 mg g?1), but the best strategy was a solid state fungal pre-treatment followed by fungal autolysis (34.5 mg g?1).The bioavailability of the nitrogen sources in the novel medium was confirmed in fed-batch bioreactor studies, in which 82.3 g dry cell L?1 of the oleaginous yeast Rhodosporidium toruloides Y4 was obtained with a lipid content of 48%. The dry cell weight obtained was higher than that obtained using conventional yeast extract, due to a higher total nitrogen content in the novel biomedium. The fatty acids obtained from the microbial oil were similar to those derived from rapeseed oil.  相似文献

ObjectiveDNA repair pathway genes play an important role in maintaining genomic integrity and protecting against cancer development. This study aimed to identify novel SNPs in the DNA repair-related genes associated with melanoma risk from a genome-wide association study (GWAS).MethodsA total of 8422 SNPs from the 165 DNA repair-related genes were extracted from a GWAS of melanoma risk, including 494 cases and 5628 controls from the Nurses’ Health Study (NHS) and the Health Professionals Follow-up Study (HPFS). We further replicated the top SNPs in a GWAS of melanoma risk from the MD Anderson Cancer Center (1804 cases and 1026 controls).ResultsA total of 3 SNPs with P value <0.001 were selected for in silico replication. One SNP was replicated: rs3902093 [A] in EXO1 promoter region (Pdiscovery = 6.6 × 10?4, Preplication = 0.039, Pjoint = 2.5 × 10?4; ORjoint = 0.80, 95% CI: 0.71, 0.90). This SNP was associated with the expression of the EXO1; carriers of the A allele showed lower expression (P = 0.002).ConclusionOur study found that a promoter region SNP in the editing and processing nucleases gene EXO1 was associated with decreased expression of EXO1 and decreased melanoma risk. Further studies are warranted to validate this association and to investigate the potential mechanisms.  相似文献

In order to obtain high productivity for a cotton crop, one of the major requirements is to establish an adequate plant population. The use of good-quality seed may ultimately be the best approach to attain this goal problem. The objective of this research was to study the effect of N-fertilization (at rates of 95.2 and 142.8 kg of N ha?1), foliar application of K (at rates of 0, 0.38, 0.77, 1.15 kg of K2O ha?1, applied twice during square initiation and boll development stages) and the plant growth retardant (PGR), mepiquat chloride (applied twice, 75 days after planting at 0.0 [control] and 0.048 kg a.i. ha?1, and 90 days after planting at 0.0 [control] and 0.024 kg a.i. ha?1), on seed yield, viability, and seedling vigor of Egyptian cotton (Gossypium barbadense cv. Giza 86). A field experiment was conducted at the Agricultural Research Center, Giza, Egypt in two growing seasons. Growth, mineral uptake, seed yield per plant and per ha, seed weight, seed viability, seedling vigor and cool germination test performance were all found to increase significantly due to the addition of the high N-rate, the foliar application of three potassium concentrations, and the PGR mepiquat chloride. The N and K rates as well as application of mepiquat chloride had no significant effect on the germination rate index in both seasons. Under the conditions of this study, applying N at a rate of 142.8 kg ha?1 combined with spraying cotton plants with K2O at 1.15 kg ha?1 and with mepiquat chloride at 0.048 + 0.024 kg ha?1 were found to improve seed yield as well as seed viability and seedling vigor in the next season.  相似文献

A chemiluminescent assay composed of TCPO [bis(2,4,6-trichlorophenyl)oxalate] and harmless rhodamine B is proposed to be superior in the determination of menadione-catalyzed hydrogen peroxide (H2O2) production by viable mammalian cells to that composed of TCPO and harmful pyrene [Anal. Biochem. 207 (1992) 255–260]. In tests, the proposed assay showed that the measurable concentration of H2O2 and the viable cell number ranged from 10?9 to 10?3 M and from 2 × 102 to 2 × 106 cells/100 μl/well in the presence of 10% bovine serum, respectively. The measuring time was approximately 10 min. On the other hand, the measurable cell numbers by the colorimetric WST-1 and MTT assays requiring several hours ranged only from 103 to 104 cells/100 μl/well and from 104 to 105 cells/100 μl/well, respectively. The cytotoxicity of sodium dodecyl sulfate was also observed at intervals of 1 min by the proposed assay, but not by the above colorimetric assays.  相似文献

Background and AimsWith the advent of regenerative therapy, there is renewed interest in the use of bone marrow as a source of adult stem and progenitor cells, including cell subsets prepared by immunomagnetic selection. Cell selection must be rapid, efficient and performed according to current good manufacturing practices. In this report we present a methodology for intra-operative preparation of CD34+ selected autologous bone marrow for autologous use in patients receiving coronary artery bypass grafts or left ventricular assist devices.Methods and ResultsWe developed a rapid erythrocyte depletion method using hydroxyethyl starch and low-speed centrifugation to prepare large-scale (mean 359 mL) bone marrow aspirates for separation on a Baxter Isolex 300i immunomagnetic cell separation device. CD34 recovery after erythrocyte depletion was 68.3 ± 20.2%, with an average depletion of 91.2 ± 2.8% and an average CD34 content of 0.58 ± 0.27%. After separation, CD34 purity was 64.1 ± 17.2%, with 44.3 ± 26.1% recovery and an average dose of 5.0 ± 2.7 × 106 CD34+ cells/product. In uncomplicated cases CD34-enriched cellular products could be accessioned, prepared, tested for release and administered within 6 h. Further analysis of CD34+ bone marrow cells revealed a significant proportion of CD45? CD34+ cells.ConclusionsIntra-operative immunomagnetic separation of CD34-enriched bone marrow is feasible using rapid low-speed Hetastarch sedimentation for erythrocyte depletion. The resulting CD34-enriched product contains CD45? cells that may represent non-hematopoietic or very early hematopoietic stem cells that participate in tissue regeneration.  相似文献

Alanine aminotransferase (ALT), which catalyzes the reversible conversion between l-glutamic acid (l-Glu) and l-alanine (l-Ala), is one of the most active aminotransferases in the clinical diagnosis of liver diseases. This work displays a microanalytical method for evaluating ALT enzyme kinetics using a microchip electrophoresis laser-induced fluorescence system. Four groups of amino acid (AA) mixtures, including the substrates of ALT (l-Glu and l-Ala), were effectively separated. Under the optimized conditions, the quantitative analysis of l-Glu and l-Ala was conducted and limits of detection (signal/noise = 3) for l-Glu and l-Ala were 4.0 × 10?7 and 2.0 × 10?7 M, respectively. In the reaction catalyzed by ALT, enzyme kinetic constants were determined for both the forward and reverse reactions by monitoring the concentration decrease of substrate AAs (l-Ala and l-Glu), and the Km and Vmax values were 10.12 mM and 0.48 mM/min for forward reaction and 3.22 mM and 0.22 mM/min for reverse reaction, respectively. Furthermore, the applicability of this assay was assessed by analysis of real serum samples. The results demonstrated that the proposed method could be used for kinetic study of ALT and shows great potential in the real application.  相似文献

The rapid (2 min) nongenomic effects of aldosterone (ALDO) and/or spironolactone (MR antagonist), RU 486 (GR antagonist), atrial natriuretic peptide (ANP) and dimethyl-BAPTA (BAPTA) on the intracellular pH recovery rate (pHirr) via NHE1 (basolateral Na+/H+ exchanger isoform), after the acid load induced by NH4Cl, and on the cytosolic free calcium concentration ([Ca2+]i) were investigated in the proximal S3 segment isolated from rats, by the probes BCECF-AM and FLUO-4-AM, respectively. The basal pHi was 7.15 ± 0.008 and the basal pHirr was 0.195 ± 0.012 pH units/min (number of tubules/number of tubular areas = 16/96). Our results confirmed the rapid biphasic effect of ALDO on NHE1: ALDO (10?12 M) increases the pHirr to approximately 59% of control value, and ALDO (10?6 M) decreases it to approximately 49%. Spironolactone did not change these effects, but RU 486 inhibited the stimulatory effect and maintained the inhibitory effect. ANP (10?6 M) or BAPTA (5 × 10?5 M) alone had no significant effect on NHE1 but prevented both effects of ALDO on this exchanger. The basal [Ca2+]i was 104 ± 3 nM (15), and ALDO (10?12 or 10?6 M) increased the basal [Ca2+]i to approximately 50% or 124%, respectively. RU 486, ANP and BAPTA decreased the [Ca2+]i and inhibited the stimulatory effect of both doses of ALDO. The results suggest the involvement of GR on the nongenomic effects of ALDO and indicate a pHirr-regulating role for [Ca2+]i that is mediated by NHE1, stimulated/impaired by ALDO, and affected by ANP or BAPTA with ALDO. The observed nongenomic hormonal interaction in the S3 segment may represent a rapid and physiologically relevant regulatory mechanism in the intact animal under conditions of volume alterations.  相似文献

AimTo investigate the mechanism through which the extracellular alkalinization promotes relaxation in rat thoracic aorta.MethodsThe relaxation response to NaOH-induced extracellular alkalinization (7.4–8.5) was measured in aortic rings pre-contracted with phenylephrine (Phe, 10?6 M). The vascular reactivity experiments were performed in endothelium-intact and -denuded rings, in the presence or and absence of indomethacin (10?5 M), NG-nitro-l-arginine methyl ester (L-NAME, 10?4 M), N-(6-Aminohexyl)-5-chloro-1-naphthalenesulfonamide/HCl (W-7, 10?7 M), 2,5-dimethylbenzimidazole (DMB, 2 × 10?5 M) and methyl-β-cyclodextrin (10?2 M). In addition, the effects of NaOH-induced extracellular alkalinization (pH 8.0 and 8.5) on the intracellular nitric oxide (NO) concentration was evaluated in isolated endothelial cells loaded with diaminofluorescein-FM diacetate (DAF-FM DA, 5 μM), in the presence and absence of DMB (2 × 10?5 M).ResultsThe extracellular alkalinization failed to induce any change in vascular tone in aortic rings pre-contracted with KCl. In rings pre-contracted with Phe, the extracellular alkalinization caused relaxation in the endothelium-intact rings only, and this relaxation was maintained after cyclooxygenase inhibition; completely abolished by the inhibition of nitric oxide synthase (NOS), Ca2+/calmodulin and Na+/Ca2+ exchanger (NCX), and partially blunted by the caveolae disassembly.ConclusionsThese results suggest that, in rat thoracic aorta, that extracellular alkalinization with NaOH activates the NCX reverse mode of endothelial cells in rat thoracic aorta, thereby the intracellular Ca2+ concentration and activating the Ca2+/calmodulin-dependent NOS. Carbon fiber microelectrodes and carbon fiber composite minielectrodes (CFM/CFCM) have been generally used for measurements of nitric oxide (NO) concentration in chemical and biological systems. The response time of a CFM/CFCM is usually from milliseconds to seconds depending on the electrode size, the thickness of coating layers on the electrode, and NO diffusion coefficients of the coating layers. As a result, the time course of recoded current changes (It curves) by the CFM/CFCM may be different from the actual time course of NO concentration changes (ct curves) if the half-life of NO decay is close to or shorter than the response time of the electrode used. This adds complexity to the process for determining rate constants of NO decay kinetics from the recorded current curves (It curves). By computer simulations based on a mathematical model, an approximation method was developed for determining rate constants of NO decay from the recorded current curves. This method was first tested and valuated using a commercial CFCM in several simple reaction systems with known rate constants. The response time of the CFCM was measured as 4.7 ± 0.7 s (n = 5). The determined rate constants of NO volatilization and NO autoxidation in our measurement system at 37 °C are (1.9 ± 0.1) × 10?3 s?1 (n = 4) and (2.0 ± 0.3) × 103 M?1 s?1 (n = 7), which are close to the reported rate constants. The method was then applied to determine the rate of NO decay in blood samples from control and smoking exposed mice. It was observed that the NO decay rate in the smoking group is >20% higher than that in control group, and the increased NO decay rate in the smoking group was reversed by 10 μM diphenyleneiodonium chloride (DPI), an inhibitor of flavin enzymes such as leukocyte NADPH oxidase.  相似文献

An online immobilized glucose oxidase (GOx) capillary microreactor was developed based on an enzymatic redox reaction with 1,4-benzoquinone as an acceptor of electrons, replacing the molecular oxygen typically used in a GOx reaction to achieve direct ultraviolet detection without derivation. A high efficiency of enzymolysis was obtained at 1 mg ml?1 1,4-benzoquinone for 5 min of incubation at 25 °C, and baseline separation of the substrate and product could be achieved with a resolution of 3.85 by employing 20 mM phosphate buffer (pH 8.0) containing 40 mg ml?1 sulfated β-cyclodextrin as an additive, a constant voltage of 15 kV, and a detection wavelength of 220 nm. In addition, an online enzyme inhibition study was performed on the immobilized GOx microreactor with metal ions Ag+ and Cu2+ used as model inhibitors. The results indicate that Ag+ (IC50 = 69.16 μM) has a markedly higher inhibitory effect than Cu2+ (IC50 = 1.33 mM). The protocol described can be applied in high-throughput screening of enzyme reactions and inhibitors.  相似文献

We investigated the suitability of surface plasmon resonance (SPR) for providing quantitative binding information from direct screening of a chemical library on protein tyrosine phosphatase 1b (PTP1B). The experimental design was established from simulations to detect binding with KD < 10?4 M. The 1120 compounds (cpds) were injected sequentially at concentrations [C(cpd)] of 0.5 or 10 μM over various target surfaces. An optimized evaluation procedure was applied. More than 90% of cpds showed no detectable signal in four screens. The 30 highest responders at C(cpd) = 10 μM, of which 25 were selected in at least one of three screens at C(cpd) = 0.5 μM, contained 22 promiscuous binders and 8 potential PTP1B-specific binders with KD  10?5 M. Inhibition of PTP1B activity was assayed and confirmed for 6 of these, including sanguinarine, a known PTP1B inhibitor. C(cpd) dependence studies fully confirmed screening conclusions. The quantitative consistency of SPR data led us to propose a structure–activity relationship (SAR) model for developing selective PTP1B inhibitors based on the ranking of 10 arylbutylpiperidine analogs.  相似文献
