The epidemiology of clinical cases of campylobacter in temperate climates shows a striking seasonality. In the search for a seasonal environmental reservoir changes in the carriage rate and population size of campylobacters in bovine hosts with time have been measured. Most probable number (MPN) methodology was used to enumerate thermophilic campylobacters in samples taken from the small intestines of beef cattle at slaughter and the fresh faeces of four dairy herds and new-born calves. Statistical analyses revealed significant evidence for seasonal periodicity in the data from dairy herds ( P = 0·044). Not only was there a departure from constancy within a 12-month interval but these data revealed a true seasonality, that is, the same periodicity in numbers from one year to the next. Each herd had two peaks per year, in approximately spring and autumn. Peaks coincided in herds on neighbouring farms but those on farms in the north preceded those on farms in the south by 2 and 1 months, respectively ( P = 0·0057). Intestinal carriage by beef cattle at slaughter was 89·4% ( n = 360) with an average MPN campylobacters per gram fresh weight (MPN gfw−1 ) of 6·1 × 102 . Average MPN gfw−1 in faeces from the dairy herds and calves were 69·9 ( S.D. 3) and 3·3 × 104 ( S.D. 1·7 × 102 ). There was no evidence of seasonal periodicity in the size of the campylobacter population in beef cattle at slaughter. In dairy cows inseminated during the hot months of the year, there is a decrease in fertility. Different factors contribute to this situation; the most important are a consequence of increased temperature and humidity that result in a decreased expression of overt estrus and a reduction in appetite and dry matter intake. Heat stress reduces the degree of dominance of the selected follicle and this can be seen as reduced steroidogenic capacity of its theca and granulosa cells and a fall in blood estradiol concentrations. Plasma progesterone levels can be increased or decreased depending on whether the heat stress is acute or chronic, and on the metabolic state of the animal. These endocrine changes reduce follicular activity and alter the ovulatory mechanism, leading to a decrease in oocyte and embryo quality. The uterine environment is also modified, reducing the likelihood of embryo implantation. Appetite and dry matter intake are both reduced by heat stress thus prolonging the postpartum period of negative energy balance and increasing the calving-conception interval, particularly in high producing dairy cows. The utilization of cooling systems may have a beneficial effect on fertility but dairy cows cooled in this way are still unable to match the fertility achieved in winter. Recent studies suggest that the use of gonadotropins to induce follicular development and ovulation can decrease the severity of seasonal postpartum infertility in dairy cows. 相似文献

AIMS: To determine prevalence and strain diversity of thermophilic campylobacters in healthy ruminants and swine. METHODS AND RESULTS: Faecal samples collected from 343 herds (120 sheep, 124 beef cattle, 82 dairy cattle and 17 swine) in the Basque Country were screened in pools for thermophilic campylobacters. Two hundred and three herds were positive (67.1% dairy cattle, 58.9% beef cattle, 55.0% sheep and 52.9% pig), and species-specific PCR identified Campylobacter jejuni in 20.7% of the herds and Campylobacter coli in 6.4%. Campylobacter coli was isolated from the four production systems and was the most prevalent species in swine, where C. jejuni was not found. Other thermophilic campylobacters were found in all production systems. Four hundred and ninety-three animals from 11 positive herds were individually analysed, detecting significantly higher within-herd prevalences in dairy cattle (66.7%) and swine (57.8%) than in sheep (8.8%) or beef cattle (5.4%). flaA PCR-RFLP and pulsed-field gel electrophoresis analysis of a selection of isolates showed high genetic diversity. CONCLUSIONS: Healthy swine, cattle and sheep are important reservoirs of thermophilic campylobacters of different species and high genetic diversity. SIGNIFICANCE AND IMPACT OF THE STUDY: Efficient farm-based intervention measures are needed to reduce risk of infection. In agrarian economy of developing nations like India, smallholder dairy production is an important enterprise and its sustainability is vital for ensuring livelihood and nutritional security to the masses. Studies on methodological aspects of farm sustainability at micro-level are limited, either confined to a particular dimension of sustainability or based on complex data requirement which is not feasible to obtain in the context of smallholder dairy farms. This study has developed a multi-attribute farm level sustainability assessment method encompassing economic, social and ecological dimensions of sustainability and has applied it to assess the sustainability of dairy farming in north-western part of India. Based on data from 120 dairy farms located in rural area of Jaipur district from the state of Rajasthan, the study computed the composite Sustainable Dairy Farming Index (SDFI). The overall substantiality status of the smallholder dairy farms in the study area was not encouraging, implying that it may not be viable for future generation to take up the enterprise. Among the three dimensions of sustainability, the average scores of ecological dimension were highest followed by the economic and social sustainability scores. Some of the core attributes like feed productivity, management of animal genetic potential and gender equality are particularly weak aspects of the dairy production system in India. The direct relationship of economic sustainability with herd size suggests for farmers with very small herds (one to two) animals, increasing the number of dairy animals to about five to six, would be a good strategy to economize on input costs and generate more marketed surplus of milk. 相似文献

Although many indicator-based sustainability monitoring tools for agriculture have been developed in the last decade, considerably less effort has been put on their validation. In the present study, we developed and applied a procedure to validate (i) MOTIFS (Monitoring Tool for Integrated Farm Sustainability), an indicator-based monitoring tool for integrated farm sustainability of Flemish dairy farms and (ii) a selection of ecological indicators (included in MOTIFS), related to nutrient use, energy use, water use and water quality. The procedure considers two steps. The first step is an accuracy evaluation, which consists of a design validation related to the scientific quality of MOTIFS and its selected indicators, and an output validation that is an evaluation of the information supplied by their output. For both validation types, we applied a transdisciplinary approach of stakeholder participation. The second step is a credibility evaluation, which relates to the degree of confidence potential end-users have in MOTIFS and hence their willingness to effectively use it in practice. This involves an end-use validation, for which we designed a test to evaluate (i) the end-use value of the selected indicators as decision aid tools, (ii) the end-use value of MOTIFS as a decision aid tool and communication tool and (iii) the willingness of potential end-users to use MOTIFS in practice. We considered two potential end-user groups: Flemish dairy farmers and ‘sustainability consultants’ (e.g. agricultural advisors assigned by farmer's organisations). Based on the validation results, we made suggestions to improve the tool and its effective application in practice. We concluded that MOTIFS is a potentially effective sustainability monitoring and management tool, since it has major assets that should be incorporated in any indicator-based system: positioning, informing, learning and communicating. 相似文献

Methods and Results: Faecal samples and tank milk samples from three dairy cattle herds were taken five times, and swab samples from drinking troughs once during a 1-year sampling period. The samples were enriched in Bolton broth and subsequently spread on mCCDA. Isolates were then subtyped by pulsed-field gel electrophoresis using SmaI. Campylobacter jejuni was detected in 169 of the 340 faecal samples and in one drinking trough sample. Prevalences between herds and sampling times varied widely. The faecal levels of C. jejuni were mainly low. Between one and four SmaI subtypes were identified from each herd per sampling. Two SmaI subtypes persisted in two of the herds throughout the study.
Conclusions: Dairy cattle can be a long-term reservoir of C. jejuni subtypes similar to clinical isolates. Differences in the colonization potential among C. jejuni strains as well as in the resistance to campylobacter colonization among animals are possible.
Significance and Impact of the Study: The study provides data on contamination dynamics, colonization levels and the persistence of C. jejuni in dairy cattle. 相似文献

AIMS: To investigate the occurrence and numbers of thermophilic campylobacters excreted by cattle in dairy herds, and to assess the strain diversity within herds. METHODS AND RESULTS: Faecal samples from 15 animals at each of 24 cattle farms were cultured quantitatively for thermophilic campylobacters and 23% of animals and 83% of farms were positive for Campylobacter jejuni. Young animals had a higher prevalence and higher faecal concentration than older animals. Serotyping and pulsed field gel electrophoresis (PFGE) of isolates showed that the most common serotypes were 2, 4-complex and 11. Serotype 2 was especially prevalent among calves (68% of the positive calves). In eight of the 20 positive herds, all isolates had the same sero- and PFGE type while, in the other herds, two to five different types were isolated. CONCLUSIONS: Significant differences were found between age groups in relation to the prevalence and numbers of excreted campylobacters, serotype distribution and strain diversity. The relatively few different strains in each herd indicate that transmission between animals is common. SIGNIFICANCE AND IMPACT OF THE STUDY: The high prevalence on cattle farms of the human pathogen C. jejuni and the wide distribution of serotype 2, the most common serotype among Danish patients, indicate that cattle might be an important reservoir for human infections. The ability of this serotype to colonize calves in high numbers further indicates that serotype 2 strains may have an advantage over other serotypes. 相似文献

Although East Africa is home to one of the most advanced dairy industries in Sub-Saharan Africa, regional annual milk production is insufficient to meet the demand. The challenge of increasing milk yields (MYs) among smallholder dairy cattle farmers (SDCFs) has received considerable attention and resulted in the introduction of various dairy management strategies (DMSs). Despite adoption of these DMSs, MYs remain low on-farm and there is a large discrepancy in the efficacy of DMSs across different farms. Therefore, the present study sought to: (1) identify on-farm DMSs employed by East African SDCFs to increase MYs and (2) summarize existing literature to quantify the expected MY changes associated with these identified DMSs. Data were collected through a comprehensive literature review and in-depth semi-structured interviews with 10 experts from the East African dairy sector. Meta-analysis of the literature review data was performed by deriving four multivariate regression models (i.e. models 1 to 4) that related DMSs to expected MYs. Each model differed in the weighting strategy used (e.g. number of observations and inverse of the standard errors) and the preferred model was selected based on the root estimated error variance and concordance correlation coefficient. Nine DMSs were identified, of which only adoption of improved cattle breeds and improved feeding (i.e. increasing diet quality and quantity) consistently and significantly (P < 0.05) increased daily MYs across the available studies. Improved breeds alongside adequate feeding explained ≤50% of the daily MYs observed in the metadata while improved feeding explained ≤30% of the daily MYs observed across the different models. Conversely, calf suckling significantly (P < 0.05) reduced MYs according to model 2. Other variables including days in milk, trial length and maximum ambient temperature (used as a proxy for heat stress) contributed significantly to decreasing MYs. These variables may explain some of the heterogeneity in MY responses to DMSs reported in the literature. Our results suggest that using improved cattle breeds alongside improved feeding is the most reliable strategy to increase MYs on-farm in East Africa. Nevertheless, these DMSs should not be considered as standalone solutions but as a pool of options that should be combined depending on the resources available to the farmer to achieve a balance between using dairy cattle genetics, proper husbandry and feeding to secure higher MYs. 相似文献

To improve the sustainability of agricultural systems of the Lombardia region (northern Italy), a mixed indicator-model-expert approach was used. Starting from the results of a previous assessment of current management (ACT) in dairy and arable farms, alternative management scenarios at field level were designed in order to reduce nitrogen (N) losses whilst maintaining or improving the environmental and economic sustainability at the farming system level. By working with a group of experts supported by a mechanisation model and a cropping system model, two alternative N management scenarios were defined following a step-by-step decision procedure. The first scenario (FERT) is an improvement of the current fertiliser management scheme, applied at the same crops as in ACT and aimed at maintaining the same yields. The second scenario (ROT) is based on changes in crop rotations by introducing new crops to reduce N losses and to maintain economic profitability. The sustainability of the two scenarios was assessed and compared with agro-ecological and economic indicators. The results of FERT, indicate that the application of adequate N management plans tuned to the production target and the promotion of best management practices may help to reduce N surplus and consequently to save fossil energy and to decrease the costs of production. In the ROT scenario, the introduction of alfalfa cultivation reduces N surplus on maize, whereas intensive double cropping systems (two crops harvested in 12 months) increase N surplus and require higher energy consumptions and production costs compared to cultivating a summer crop only. However, in rotational systems more favourable weed population dynamics are expected compared to ACT. Both alternative scenarios were not implemented in practice, but they are realistic and are consistent with results of experiments where management options similar to those introduced in FERT and ROT were tested.This work indicates that the rational integration between scientific tools (indicators and models) and expert knowledge is adequate to deal with complex farming and cropping systems, which require a multidisciplinary approach. 相似文献

Neospora caninum, a coccidian parasite closely related to Toxoplasma gondii, is one of the major causes of abortion in cattle worldwide. Conventional serological techniques, such as the indirect fluorescent antibody test and enzyme-linked immunosorbent assay (ELISA), are routinely used in adult animals and aborted fetuses for the detection of anti- N. caninum antibodies. In Romania, infection with N. caninum in cattle has been reported recently, but only in limited areas from the north and central parts of the country. Therefore, the aim of this study was to obtain additional seroepidemiological data on infection with N. caninum on dairy farms from the south of Romania. A total of 258 blood samples was analyzed from 230 dairy cows and 28 calves from 9 dairy farms in southern Romania; the presence of specific IgG antibodies against N. caninum was determined using an indirect ELISA test. The average seroprevalence was 40.3%, but the within-herd prevalence ranged between 11.5 and 80.0%; the seroprevalence in dairy cows was 41.7%, while in calves it was 28.6%. Of the positive samples, 74.0% (77/104) had a high positive reaction (S/P ratio more than 1.0), while 26.0% (27/104) had a low positive reaction (S/P ratio between 0.5 and 1.0). This study indicates that N. caninum infection is widespread in the south of Romania, which could explain the causes of abortions registered in some herds in the studied area. However, a serological screening across the country is planned in order to assess the actual national prevalence of N. caninum infection, followed by implementation of a prevention and control program. 相似文献

A survey was conducted between March and October of 1994 to determine the prevalence and identify the sources of serotype O157:H7 isolates of Escherichia coli in Wisconsin dairy herds. A stratified sample of 400 farms was identified, and 70 farms with weaned calves less than 4 months old were included in the study. During the prevalence study, 5 of the 70 farms (herd prevalence, 7.1 +/- 4.5%) and fecal samples from 10 of 560 calves (animal prevalence, 1.8%) tested positive for serotype O157:H7. In a follow-up study, the five O157:H7-positive farms and seven of the O157:H7-negative farms identified in the prevalence study were visited again. An additional 517 fecal samples from cattle of various ages were tested, and a total of 15 animals from four of the five herds that were previously positive and 4 animals from two of seven herds that were previously negative tested positive for E. coli O157:H7. Observations made during the follow-up study suggested that horizontal transmission was an important means of E. coli O157:H7 dissemination on the farms. A total of 302 environmental samples, were examined, and 2 animal drinking water samples from one previously negative farm and 1 animal drinking water sample from a previously positive farm contained E. coli O157:H7. Analyses by the pulsed-field gel electrophoresis technique of contour-clamped homogeneous electric field electrophoresis revealed that isolates from the same farm displayed identical or very similar XbaI restriction endonuclease digestion profiles (REDP), whereas isolates from different farms typically displayed different REDP. However, more than one REDP was usually observed for a given herd over the 8-month sampling period. Analyses of multiple isolates from an animal revealed that some animals harbored O157:H7 strains that had different REDP, although the REDP of isolates obtained from the same fecal sample were very similar. Collectively, 160 bovine isolates obtained from 29 different animals and three water isolates displayed 20 distinct XbaI REDP. Our data revealed that there are several clonal types of serotype O157:H7 isolates in Wisconsin and indicated that there is probably more than one source of this pathogen on the dairy farms studied. However, animal drinking water was identified as one source of E. coli O157:H7 on one farm. 相似文献

Small-scale laboratory research was conducted to compare the effects of different aeration rates and oxic/anoxic phasing on nitrous oxide (N(2)O) formation from dairy manure slurries. Manure slurry samples were incubated in triplicate for three-weeks under a range of continuous sweep gas flows (0.01-0.23Lmin(-1)kg(-1) slurry) with and without oxygen (air and dinitrogen gas). The net release of N(2)O-N was affected by both aeration rates and oxic/anoxic conditions, whereas ammonia volatilization depended mainly on gas flow rates. Maximum N(2)O-N losses after three-weeks incubation were 4.2% of total slurry N. Major N losses (up to 50% of total slurry N) were caused by ammonia volatilization that increased with increasing gas flow rates. The lowest nitrous oxide and ammonia production was observed from low flow phased oxic/anoxic treatment. 相似文献

The decline in dairy herd fertility internationally has highlighted the limited impact of traditional veterinary approaches to herd fertility. The role of the veterinarian in fertility management on dairy farms has evolved from addressing individual clinical conditions to analyzing suboptimal herd metrics. However, this paradigm shift has only successfully occurred in some dairy industries and less so in others. Needs analyses indicate that the critical constraints to change are veterinary practice size, client motivation and data quality and availability. In addition, this review identified the inability of veterinarians to demonstrate and to market the cost-benefit of their fertility management services as important impediments to change. In many cases change is not being managed but is imposed by the growth of paraprofessionals. Some veterinarians still see their role as an animal clinician while others have evolved into leaders of the herd fertility management team. The core role of dairy veterinarians remains individual animal examinations but this must be supplemented with systematic herd fertility investigation and veterinarian-led herd fertility management. This new role encompasses leading the change from clinical calls only to a planned approach to herd fertility, demonstrating the cost-benefits of the program, scheduling fertility management consultations, assisting the farmer in setting specific, measurable, attainable, relevant and time-limited (SMART) goals, drawing up standard operating procedures (SOPs), training and auditing staff in fertility management practices, encouraging a team approach, implementing veterinary fertility management and monitoring performance. Veterinarians who fail to engage in this process of change risk being marginalized by others keen to promote their herd fertility services. 相似文献

通过历史时期地图数字化和遥感图像解译得到三江平原1954~2005年的6期土地利用/覆被数据。根据IPCC《2006指南》提供的方法,评估土地利用/覆被变化对三江平原植被碳储量的影响。结果表明:三江平原1954~2005年土地利用/覆被变化显著,农田大面积增加,沼泽湿地、林地、草地面积锐减;土地利用/覆被变化主要发生在农田、沼泽湿地、林地和草地之间;农田是沼泽湿地、林地、草地的主要转出对象,林地的主要转入来源为农田和草地,沼泽湿地的主要转入来源为农田和林地。1954~2005年共有1.07×103km2林地、5.73×103km2草地和2.59×104km2沼泽湿地转出为农田。土地利用/覆被变化导致三江平原植被碳储量不断减少,1954~2005年三江平原植被碳储量共减少57.48Tg。林地、沼泽湿地、草地向农田的转化及林地向草地、沼泽湿地的转化导致植被碳储量减少97.06Tg,农田向林地、沼泽湿地、草地的转化及草地、沼泽湿地向林地转化导致植被碳储量增加39.58Tg。 相似文献

Prevalence of anti-Neospora caninum antibodies was determined in sera of 320 dogs from Argentina using the indirect fluorescent antibody test (IFAT). Antibodies to N. caninum were found in 121 of 320 (37.8%) sera with titers of 1:50 (21 dogs), 1:100 (23 dogs), 1:200 (23 dogs), 1:400 (17 dogs), 1:800 (23 dogs), and > or = 1:1.600 (14 dogs). The seropositivity (IFAT, > or = 1:50) was higher in dogs from dairy (48% of 125) and beef (54.2% of 35) farms than in dogs from urban areas (26.2% of 160). Prevalence of anti-N. caninum antibodies was higher in dogs more than 12 mo of age (47.7%, 105 of 222) versus in 12-mo-old or younger dogs (12.7% of 86), suggesting postnatal exposure of N. caninum infection in dogs. 相似文献

Multilocus sequence typing (MLST), an accurate and phylogenetically robust characterization method for population studies of Campylobacter, was applied to Campylobacter jejuni isolates (n = 297) from the fecal samples of cattle from five dairy farms in Cheshire, United Kingdom, collected throughout 2003. The population dynamics of the C. jejuni strains, as identified by the occurrence of sequence types and clonal complexes, demonstrated variations within and between cattle populations over time. Three clonal lineages have emerged to predominate among the cattle isolates, namely, the ST-61 complex (24.2%), ST-21 complex (23.6%), and ST-42 complex (20.5%). This provided further evidence that the ST-61 clonal complex may present a cattle-adapted C. jejuni genotype. In addition, the ST-42 clonal complex may also represent an important cattle-associated genotype. Strong geographical associations for these genotypes were also found among the farms. This is the first longitudinal study and the largest study to date for C. jejuni involving cattle populations using MLST for accurate strain characterization. This study shows the important associations between cattle and C. jejuni clonal complexes ST-61, ST-21, and ST-42, and it suggests that cattle and/or dairy products are likely to be a source of the human Campylobacter gastroenteritis caused by such genotypes. The reported findings have significant implications for the design of effective intervention strategies for disease control and prevention. 相似文献

Welfare and management of calves is of increasing interest and also influences performance of these animals in later life. The aim of this study was to assess management and environmental conditions under which pre-weaned dairy calves are reared on commercial Irish dairy farms. We included 47 spring-calving, pasture-based herds in this study. Herd and animal-specific data, such as mortality rate, age and breed, were gathered from all participants via the HerdPlus® database. Information pertaining to management practices was collected by conducting an interview with the principal calf rearer, while an assessment of calf housing facilities was conducted to identify conditions calves were reared in. The environmental assessment included measurements of space allowance per calf, as well as feeding equipment hygiene. To assess calf behaviour video observations were used, while accounting for the number of calves present in a group and the space available per calf. Faecal samples were also collected to determine the presence of enteric pathogens among calves. To compare calf space allowance, group size and presence of enteric pathogens early and late in the calving season each farm was visited twice. Calf mortality was not associated with either herd size, space allowance per calf or post-colostrum feeding practices. Higher calf mortality was identified among herds which reported experiencing an on-set of calf pneumonia during weeks 8 to 10 of the calving season. This study demonstrates that factors associated with calf welfare on commercial Irish dairy farms (e.g. space allowance, mortality rate) are independent of herd size. Some management practices however, such as methods used for treating health issues can affect rates of calf mortality experienced. Calf mortality, for example, was lower in herds which treated diarrhoea cases by administering electrolytes, while continuing to offer milk. Behavioural observations indicate that smaller group sizes could promote expression of positive behaviours, potentially resulting from an overall improvement in welfare. Space allowance per calf was not associated with observed behaviour frequencies. We also identified that similar rates of calf mortality are experienced across herds of different sizes. 相似文献

Aims: To determine the counts and/or prevalence in fresh bovine faeces of Escherichia coli, enterococci, Campylobacter, Salmonella, shiga toxin‐producing E. coli (STEC), Giardia and Cryptosporidium, as inputs to numerical models designed to estimate microbial loadings on pasture grazed by cattle in New Zealand. Methods and Results: In each season over one year, samples of freshly deposited bovine faeces were collected from four New Zealand dairy farms (n = 155), and enumerated for E. coli, enterococci, Campylobacter, Giardia and Cryptosporidium. They were also tested for the presence of Salmonella and STEC. The overall median bacterial counts (g?1 wet weight) were E. coli– 5·9 × 106; enterococci – 1·3 × 104; Campylobacter– 3·9 × 105. All counts were highly variable within and between samplings, and few seasonal or regional patterns emerged. However, mean Campylobacter counts were consistently higher in spring. No Salmonella spp. was detected, and only two samples were positive for STEC. Cryptosporidium and Giardia were isolated from 5·2% and 4·5% of the samples, respectively, yielding low numbers of (oo)cysts (1–25 g?1 and 1–17 g?1, respectively). Conclusions: Fresh bovine faeces are a significant source of E. coli, enterococci and Campylobacter on New Zealand pastures, although numbers are likely to vary markedly between faecal samples. Significance and Impact of the Study: The study provides the first significant set of indicator and pathogen counts for one of the largest sources of faecal contamination of natural waters in New Zealand, and will be used to model these inputs. 相似文献

While both annual and perennial plants store nitrogen resources during the growing season, seasonal N cycling is a hallmark of the perennial habit. In Populus the vegetative storage proteins BSP, WIN4 and PNI288 all play a role in N storage during active growth, whereas BSP is the major form of reduced N storage during winter dormancy. In this review we explore cellular and molecular events implicated in seasonal N cycling in Populus, as well as environmental cues that modulate both the phenology of seasonal N cycling, and the efficiency and proficiency of autumn N resorption. We highlight recent advances that have been made using Populus genomics resources to address processes germane to seasonal N cycling. Genetic and genetological studies are enabling us to connect our understanding of seasonal N cycling at molecular and cellular levels with that at ecophysiological levels. With the genomics resources and foundational knowledge that are now in place, Populus researchers are poised to build an integrative understanding of seasonal N cycling that spans from genomes to ecosystems. 相似文献
