Summary Excised shoot apices (0.6 or 1.0 mm long) of Rosa multiflora were grown in axenic culture on defined media including auxins, gibberellic acid and cytokinins. In the presence of zeatin (10-8–10-7M) and 6-benzylaminopurine (10-7–10-6 M) apices grew into plantlets but two other cytokinins, kinetin and 6-(3-methylbut-2-enyl)aminopurien, were ineffective. Auxins or gibberellic acid were either inhibitory or had no effect on differentiation.  相似文献

The objective of this study was to improve shoot regeneration from grapefruit. Because many commercially grown citrus types are apomictic, important in vitro applications such as Agrobacterium-mediated transformation commonly use epicotyl explants from in vitro seedlings; thus, adequate adventitious shoot production is an important prerequisite for efficient use of these applications. Eight plant growth regulators were studied—six cytokinins (6-benzylaminopurine, kinetin, zeatin trans-isomer, 6-[γ,γ-dimethylallylamino] purine, zeatin riboside trans-isomer and meta-topolin) and two auxins (α-naphthalene acetic acid and indole-3-acetic acid). An iterative design strategy was followed that included mixture and mixture-amount experimental designs suitable for resolving proportional and concentration effects; in vitro effects of cytokinins and auxins are affected by both proportion and concentration. One-centimeter-long explants were excised from the epicotyl of etiolated, in vitro-grown seedlings. Explants were placed onto experimental formulations and cultured in growth cabinets at 27°C over 6 wk, which included 2 wk in the dark followed by 4 wk in the light. The results indicated that (1) 6-benzylaminopurine or zeatin riboside were the most effective cytokinins for inducing shoot regeneration in citrus; (2) zeatin riboside singly or in combination with indole-3-acetic acid resulted in the highest quality, the greatest number of explants with buds/shoots, and the greatest shoot number; and (3) 6-benzylaminopurine and indole-3-acetic acid improved shoot regeneration vs. 6-benzylaminopurine at a considerably lesser cost than zeatin riboside and indole-3-acetic acid.  相似文献

The role played by plant growth regulators in algae is poorly known. In order to increase the knowledge about the function of auxins and cytokinins in seaweeds, explants such as apical and intercalary segments and callus-like structures (CLS) of Grateloupia dichotoma were cultured in semi-solid or liquid artificial media ASP 12-NTA. Two auxins, indole-3-acetic acid (IAA) and 2,4-dichlorophenoxyacetic acid (2,4-D), and one cytokinin, 6-benzylaminopurine (BA), at concentrations of 0.5 and 5.0 mg l–1 were tested. Moreover, IAA and BA were tested together at concentrations of 1:5 and 5:1 mg l–1. All treatments promoted the growth of CLS in intercalary segments; CLS from apical segments were significantly higher in treatments with 2,4-D or IAA:BA (1:5 mg 1–1). The morphogenetic responses for auxins and BA were opposite, auxins inhibited while BA promoted the formation of lateral branches; however, auxins promoted the elongation of such branches. The process of plant regeneration observed on CLS was stimulated significantly by treatment with high concentration of BA or IAA:BA (1:5 mg 1–1) in semi-solid and liquid media. The growth of upright axes was stimulated significantly by treatment with 2,4-D in semi-solid medium, and IAA:BA (1:5 mg l–1) in liquid medium. These results show the importance that plant growth regulators could have in the control of growth, morphogenetic processes and micropropagation in red algae.This paper is part of the PhD thesis of NSY.  相似文献

Plant cell and suspension cultures have been established from stem cuttings of Picrasma quassioides Bennett. The effect of 244 different types/concentrations of plant growth regulators on growth and quassin accumulation in callus tissue was investigated. Best growth, in terms of wet/dry weight after four weeks growth, was obtained on B5 media supplemented with 2% glucose, 10% coconut milk, 0.5 mg.l–1 zeatin riboside and 1.5 mg.l–1 IBA. The highest yields of quassin (0.014–0.018%) were detected on this same media supplemented with 1.0 mg.l–1 IBA and varying concentrations of zeatin riboside. Suspension cultures were easily established on B5 media supplemented with 2% glucose, 1.0 mg.l–1 2,4-D and 0.5 mg.l–1 kinetin. The carbon source had a marked effect on quassin accumulation with 0.32% quassin being detected when cells were grown in 2% galactose. Within 3 weeks of culture, excised cotyledon expiants of Camellia sinensis (L.) O. Kuntze produced somatic embryos without intermediate callus when cultured in Murashige and Skoog's basal medium with 30 g–1 sucrose. In medium without plant growth regulators, up to 60% of the cultures developed somatic embryos. Embryogenic competence was reduced by increasing concentrations of plant growth regulators tested (i.e. kinetin, 6-benzylaminopurine, and indole butyric acid). The somatic embryos developed, grew to maturity without being subcultured within 6–8 weeks. Secondary embryogenesis was not observed. Germination of isolated mature somatic embryos was low in medium without plant growth regulators. Up to 53% and 60% germination occurred when medium impregnated with kinetin at 1.8 mgl–1 or 1.0 mgl–1 6-benzylaminopurine were used respectively. Callus was also routinely produced when cotyledons were cultured in MS basal medium with auxins (2,4-dichlorophenoxyacetic acid and indole acetic acid). Callus induction was however, also achieved in plant growth regulator free medium. The present work deals with the effect of some auxins [2,4-D,IAA, NAA and ß-naphthoxyacetic acid (NOA)] and cytokinins[6-benzylaminopurine (BAP), 6-(,-dimethylallylamino)purinc (2iP),KN and 6-(4-hydroxy-3-methylbut-2-enylaimno)purine (zeatin)]on growth and gametangial formation in the liverwort Ricciagangetica. Of these, 2,4-D and IAA stimulate vegetative growth,induce the formation of rhizoids on dorsal surface of thallusand shorten the time required for gametangial initiation. NAAand NOA inhibit growth, and do not affect the time of gametangialproduction. The auxins increase archegonial number without affectingantheridial induction and, among those tested, IAA is the mosteffective. Archegonial number is enhanced by the cytokinins,but antheridial production decreases. The maximum number ofarchegonia is initiated in response to 2iP, and this effectappears to be specific. Plants of Solanum melongena were propagated under in vitro conditions (27°C, 12h/day illumination at 62 Em-2s-1, 60% humidity) by subculture of terminal and lateral cuttings on MS medium +20 gl-1 sucrose + Morel and Wetmore vitamins at 1/8 strength and 7 gl-1 agar. Lamina, petioles and stems of 3-week-old cuttings were used as sources of protoplasts. The best mean yield of protoplasts was obtained from the lamina with 9,030×103 protoplasts per gram of tissue. Petioles and stems yielded respectively 3,144×103 and 1,220.4×103 protoplasts per gram of tissue. first division of petiole and stem protoplasts occurred within 48 h, while lamina protoplasts underwent division after 3–4 days of culture in KM8p medium +2,4-D(0.2 gl-1) + zeatin (0.5 mgl-1) + NAA (1 mgl-1) and 0.35M glucose as osmoticum. The highest percentage of dividing cells was obtained from petiole material, estimated at 33.4% after 7 days, compared to 23.8% and 19.4% respectively for stem and lamina protoplasts. When BAP replaced zeatin in KM8p, the division percentage of lamina protoplasts was reduced to 10–15%. When transferred to regeneration medium, all calli derived from KM8p + zeatin formed deep-green spots identified as embryo-like structures, while only few calli from KM8p + BAP underwent shoot organogenesis without formation of green spots. Some of embryo-like structure developed into plantlets with a frequency of 1–2 plantlets per callus especially on MS medium + zeatin (4 mgl-1) + IAA (0.2 mgl-1). The ipt gene from the T-DNA of Agrobacterium tumefaciens was transferred to tobacco (Nicotiana tabacum L.) in order to study the control which auxin appears to exert over levels of cytokinin generated by expression of this gene. The transgenic tissues contained elevated levels of cytokinins, exhibited cytokinin and auxin autonomy and grew as shooty calli on hormone-free media. Addition of 1-naphthylacetic acid to this culture medium reduced the total level of cytokinins by 84% while 6-benzylaminopurine elevated the cytokinin level when added to media containing auxin. The cytokinins in the transgenic tissue were labelled with 3H and auxin was found to promote conversion of zeatin-type cytokinins to 3H-labelled adenine derivatives. When the very rapid metabolism of exogenous [3H]zeatin riboside was suppressed by a phenylurea derivative, a noncompetitive inhibitor of cytokinin oxidase, auxin promoted metabolism to adenine-type compounds. Since these results indicated that auxin promoted cytokinin oxidase activity in the transformed tissue, this enzyme was purified from the tobacco tissue cultures. Auxin did not increase the level of the enzyme per unit tissue protein, but did enhance the activity of the enzyme in vitro and promoted the activity of both glycosylated and non-glycosylated forms. This enhancement could contribute to the decrease in cytokinin level induced by auxin. The influence of plant growth regulators on biomass growth and the accumulation of medicinally-relevant isoflavone phytoestrogens, derivatives of genistein and daidzein (8 compounds including aglycones, glucosides and glucoside esters) in callus cultures of Genista tinctoria (Fabaceae) was examined. The experiments included 10 auxins [2,4-dichlorophenoxyacetic acid (2,4-D), p-chlorophenoxyacetic acid, indole-3-acetic acid, indole-3-butyric acid, indole-3-propionic acid, 1-naphthaleneacetic acid, β-naphthoxyacetic acid, picloram, 2,3,5-triiodobenzoic acid (TIBA), 2,4,5-trichlorophenoxyacetic acid (2,4,5-T)] and 7 cytokinins [6-benzylaminopurine, forchlorfenuron, 1,3-diphenylurea, 2-isopentenyladenine, kinetin (KIN), thidiazuron, zeatin] applied at 0.5 and 5.0 mg l?1, jointly with 5.0 or 0.5 mg l?1 KIN or 2,4-D (for auxins and cytokinins, respectively—36 phytohormone combinations in total). Statistical analysis of the relationships between callus growth [expressed as growth index (Gi)] and the accumulation of isoflavones showed positive correlation in the cytokinin group (rxy values from 0.13 to 0.61) and negative correlation within auxins (rxy values from ?0.31 to ?0.39). Among the cytokinins tested, the highest isoflavone content (6,436.26 mg/100 g dry weight) and the fastest biomass growth (Gi = 892.46 %) were obtained for 0.5 mg l?1 KIN used jointly with 5.0 mg l?1 2,4-D. In the group of auxins, the combination of 0.5 mg l?1 TIBA and 5.0 mg l?1 KIN provided the fastest culture growth (Gi = 983.07 %) and the isoflavone concentration of 10,474.23 mg/100 g dry weight, which is so far the highest amount of these metabolites achieved in callus cultures of higher plants.  相似文献

Micropropagation of Limonium cavanillesii Erben, a threatened and endemic statice species from Valencia Community (Eastern Spain), was successfully achieved using inflorescence stem pieces as initial explants. Segments 20 mm long from basal parts of immature inflorescences and with axillary buds were cut, sterilised and established in vitro. Shoots obtained from indifferentiated buds were sectioned and then transferred to Murashige and Skoog (MS) medium with 2 mg l–1 kinetin to provide a plant stock.Shoot multiplication was achieved on MS medium with different cytokinins. The best results for shoot formation were obtained with 2–5 mg l–1kinetin, 5 mg l–1 6---dimethylallylaminopurine or 0.1 mg l–1 6-benzylaminopurine, without significant differences between them. High shoot rooting (80–85%) was obtained within four weeks with indolebutyric acid or indoleacetic acid (0.1 or 0.5 mg l–1), and also on medium without plant growth regulators. Plant survival to hardened greenhouse conditions was 90% four weeks after plantlet removal from in vitro conditions.This protocol for micropropagation of Limonium cavanillesii is very useful for conservation purposes of endangered statice species, because by using inflorescence stem as initial material it is easier to establish aseptic cultures while preserving the mother plant.  相似文献

The physiological effects of three auxins [indole-3-butyric acid (IBA), α-naphthaleneacetic acid (NAA) and 2,4-dichlorophenoxyacetic acid (2,4-d)] and two cytokinins [thidiazuron (TDZ) and N6-benzylaminopurine (NAA)] on in vitro morphogenesis of Doryanthes excelsa were measured. Longitudinal bud sections derived from immature inflorescences were used as a source of explants. Callus regeneration was observed at the highest frequencies (46.2%) when grown on media containing 50 μmol L-1 NAA and 0.5 μmol L−1 TDZ. Adventitious shoot organogenesis was observed at the highest frequency (56.8%) when grown on media containing 0.5 μmol L−1 NAA and 50 μmol L−1 TDZ. Regenerated shoots were rooted ex vitro after 6 weeks when dipped in a solution of 50 μmol L−1 NAA or no plant growth regulators were applied.  相似文献

Single node explants of 'Koroneiki' olive trees werecultured for one month on a modified Driver-Kuniyuki for Walnut medium, lackinggrowth regulators. The explants were subcultured once a month on a mediumsupplemented with zeatin riboside, 6-(--dimethylallylamino)purine,6-benzyladenine or thidiazuron. Zeatin riboside proved to be superior to othercytokinins in inducing shoot proliferation. The combination of olive knotextract at 25 or 50 mg l–1 with cytokininssuppressed shoot proliferation. After two months at the proliferation stage,theexplants were cultured for one week in the dark in 1 ml liquidWoody Plant Medium supplemented with IBA, -NAA or IBA+-NAA. Theexplants were then transferred to the same solid medium lacking growthregulators, with a small layer of perlite on the surface. The combination ofthetwo auxins at 1+1 mg l–1 resulted in almost 76%rooting. The combination of olive knot extract at 50 mgl–1 with auxins increased the rooting percentage up toalmost 87%. Artificial infection of explants with the bacteriumPseudomonas savastanoi pv. savastanoiinhibited rhizogenesis, even in the presence of auxins. Rooted explants weresuccessfully acclimatised under a mist system, with the survival rate reachingalmost 75%.  相似文献

Studies were conducted to test the effects of various auxins, cytokinins, carbohydrates and amino acids on somatic embryogenesis from shoot apices of pea (Pisum sativum L.) cultured on a sole medium. Picloram (4.5 M) and 4-chlorophenoxyacetic acid (45 M) were the most effective auxins. Addition of cytokinins (benzyladenine, zeatin, kinetin) to auxin-containing medium reduced embryo production. Amino acids (glutamine, alanine, proline) did not improve somatic embryogenesis. Carbohydrate seemed to be a critical factor. Embryogenic efficiency and embryo development were promoted by high carbohydrate concentration. The best results were obtained with fructose (252–504 mM); the number of somatic embryos per cultured explant was 3- to 4-fold higher compared to the control (84 mM sucrose). Summary The effect of a non-indole compound, phenylacetic acid (PAA), on the induction of somatic embryogenesis in tissue cultures of geranium (Pelargonium x hortorum Bailey cv. Scarlet Orbit Improved) was investigated. Hypocotyl explants derived from young, dark-grown seedlings were cultured on Murashige and Skoog (1962) medium (MS) supplemented with PAA or IAA (0.01–120 M) alone or in combination with BAP (8 M). Somatic embryogenesis was induced by both PAA and IAA at 0.01–20 M with 8 M BAP, however, the optima differed considerably for the two compounds. Maximal activity of IAA for somatic embryogenesis was found at 0.1–2.5 M, whereas PAA gave best results at 10 and 20 M under identical culture conditions. Isolated embryos ofKarwinskia humboldtiana were cultured in vitro. The growth of embryos and development to plantlets on woody plant medium supplemented with indole-3-acetic acid 6.10-2 mol l–1, gibberellic acid (GA3) 3.10-2 mol l–1, and 6-benzylaminopurine (BA) 2 mol l–1 was obtained. Multiplication of shoots and rooting of excised shoots has been achieved. The effects of plant growth regulators, light intensity, and end-of-day (EOD) light quality treatments on node and microtuber induction (% of cultures with microtubers) and development (fresh weight of microtubers) in yam (Dioscorea alata L. cv. Oriental) cultures were investigated. Nodal segments were excised from plantlets cultured on tuberization medium containing growth regulators and exposed to various light treatments. Absciscic acid (1 M) stimulated and cytokinins (2.5 M) inhibited microtuber development from yam nodal segments cultured on Mantell's and Hugo's full-strength tuberization medium under 8-h photoperiods. EOD far-red (FR) light inhibited microtuber induction and development and enhanced node formation. EOD FR light effects were nullified by immediately following the FR treatment with red light. This suggested the involvement of phytochrome in these processes. The lowest light intensity evaluated (12 mol m–2 s–1) inhibited microtuber, root and shoot production as compared to light intensities of 42, 72 and 102 mol m–2 s–1. Protocols were established for achieving plant regeneration from stem internode, callus, and cell suspension cultures of Solanum lycopersicoides Dun. Two accessions of S. lycopersicoides exhibited different responses as to callus formation on various media, requirement of gibberellic acid for shoot regeneration, and ability to grow in suspension culture. The optimum medium for initiation and maintenance of cell suspension cultures was Murashige and Skoog [9] medium with 15 mg l NAA. For shoot regeneration, of three cytokinins tested, zeatin was found most effective relative to number, rapidity of response and overall quality of shoots. The effects of serial combinations of either indole-3-acetic acid, indole-3-butyric acid or -naphthaleneacetic acid (0.5–10.0 mg/l) with either kinetin, 6-benzyl-amino-purine, zeatin or 6-methylaminopurine (0.5–5.0 mg/l) have been investigated to assess the morphogenetic potential of foliar explants of Duboisia myoporoides. Shoot buds developed either directly or via a callus interphase. Combinations involving indole-3-acetic acid with any of the cytokinins were more effective in inducing shoot bud formation compared to those containing indole-3-butyric acid or -napthalenacetic acid as an auxin. Among cytokinins, zeatin, kinetin and 6-benzylamino-purine were equally effective for shoot formation. However, optimum response with zeatin could be achieved at low concentrations (0.5–2.0 mg/l), while kinetin and 6-benzylamino-purine exhibited comparable efficacy at higher levels (3.0–5.0 mg/l). 6-Methylaminopurine proved least effective in all concentrations and combinations tested. Rooting of the differentiated shoots was readily achieved with -naphthaleneacetic acid alone (0.5 mg/l) after changing the physical form of the medium from gel to static liquid. Mutants altered in their response to auxins and cytokinins have been isolated in the moss Physcomitrella patens either by screening clones from mutagenized spores for growth on high concentrations of cytokinin or auxin, in which case mutants showing altered sensitivities can be recognized 3–4 weeks later, or by non-selective isolation of morphologically abnormal mutants, some of which are found to have altered sensitivities. Most of the mutants obtained selectively are also morphologically abnormal. The mutants are heterogeneous in their responses to auxin and cytokinin, and the behaviour of some is consistent with their being unable to make auxin, while that of others may be due to their being unable to synthesize cytokinin. Physiological analysis of the mutants has shown that both endogenous auxin and cytokinin are likely to play important and interdependent roles in several steps of gametophytic development. An obligate fungus Albugo candida (Pers. ex Lév.) Ktze. (race unidentified) was successfully grown on host callus tissues of Brassica juncea cv. Varuna. Of the various type of diseased explants used, young (green) hypertrophied inflorescence axis bearing non-erumpent zoosporangial blisters allowed the fungus to multiply asexually over the host calli on modified MS-medium (Murashige and Skoog, 1962). The dual cultures were maintained up to 6–8 subcultures without loss of viability of zoosporangia on MS-medium supplemented with 10.0 mg L–1 IBA, 0.05 mg L–1 kinetin, 25.0 mg L–1 AA, 1.0 mg L–1 biotin, 1.0 mg L–1 thiamine-HCl and 1.0 g L–1 casein hydrolysate. The fungus grew only on the callus cells and not axenically on the medium.
