A theory is presented which can explain the interaction of the major factors known to influence in vitro extrusion of the microsporidian polar filament. It is proposed that the pH, and concentration and species of cation in the external medium influence the activity of car?ylic ionophore molecules in spore membranes in the following manner: (1) Alkaline environmental conditions establish a proton gradient across the spore plasma membrane, and facilitate the activation of ionophore molecules in this membrane. (2) This proton gradient drives an ionophorically-mediated cation/proton exchange across the plasma membrane. (3) As protons are lost from the sporoplasm its alkalinity increases, so that ionophore molecules in organelle membranes (i.e. in the polaroplast and posterior vacuole) are activated. This initiates a cation/proton exchange between sporoplasm and organelles. (4) Continued movement of cations into organelles in the spore causes major osmotic imbalance across spore membranes. This leads to a rapid inflow of water into the spore and swelling of the polaroplast and posterior vacuole. The associated pressure increase in the spore causes the explosive discharge of the polar filament through the polar cap. This model is used to explain previously published results from the literature, and methods of testing predictions generated by this hypothesis are outlined. 相似文献

Curcumin (C) and its natural analogues demethoxycurcumin (dmC) and bisdemethoxycurcumin (bdmC), known for their potent anti-inflammatory, antioxidant, antimutagenic and anticarcinogenic effects, were tested for their possible inhibitory effects against seven cooked food mutagens (heterocyclic amines): 2-amino-3-methylimidazo[4,5-f]quinoline (IQ), 2-amino-3,4-dimethylimidazo[4,5-f]quinoline (MeIQ), 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx), 3-amino-1,4-dimethyl-5H-pyrido[4,3-b]indole (Trp-P-1), 3-amino-1-methyl-5H-pyrido[4,3-b]indole (Trp-P-2), 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP) and 2-amino-6-methyldipyrido[1,2-a:3',2'-d]imidazole (Glu-P-1), in both TA98 and TA100 strains of Salmonella typhimurium using Ames Salmonella/reversion assay in the presence of Aroclor induced rat liver S9 homogenate. In the present investigations, curcumin as well as its two natural analogues i.e., dmC and bdmC were found to be highly effective in suppressing genotoxicity of all the tested cooked food mutagens in a dose-dependent manner, in both the frame shift (TA98) as well as base pair mutation sensitive (TA100) strains of S. typhimurium. However, bdmC appeared to be a relatively less active antimutagen compared to C and dmC. More than 80% inhibition of mutagenicity was observed at 200 microg/plate in case of C and dmC in both TA98 and TA100 against all tested cooked food mutagens. Where as, bdmC showed 39-79% inhibition in TA100 and 60-80% inhibition in TA98, at a dose of 200 microg/plate. These findings warrant further biochemical, enzymatic and in vivo investigations in animal models as well as in humans to establish the chemoprotective effect of these agents against mutagenic heterocyclic amines found in cooked food. 相似文献

A fraction of polar lipids was isolated from spores of the anaerobic bacterium Clostridium butyricum 35/11 exerting a noticeable radioprotective effect. The main biological activity of spore extracts was associated with this fraction. The fraction of polar lipids inhibited autolysis of the bacterial cell walls. The fraction was found to contain a phenolic glycolipid and a peptide component. The bacteriostatic and radiotherapeutic properties of the fraction are presumed to be due to its membranotropic activity. 相似文献

Polylysine-coated polystyrene beads can nucleate polar assembly of monomeric actin into filamentous form. This nucleation has been demonstrated by a combination of biochemical and structural experiments. The polylysine-coated beads accelerate the rate of actin assembly as detected by two different biochemical assays. Subsequent examination of the beads by electron microscopy reveals numerous actin filaments of similar length radiating from the beads. ATP promotes this bead-induced acceleration of assembly. Decoration of the filaments with the myosin fragment S1 shows that these filaments all have the same polarity, with the arrowhead pattern pointing toward the bead. The relevance of the system to in vitro mechanisms and its usefulness in other studies are discussed. 相似文献

The fine structure of Nosema oryzaephili is very similar to that of the closely related Nosema whitei. The typical number of coils in the polar filament in the spore is slightly different in the two species. The number of coils and their angle of tilt to the long axis of the spore distinguished the two species from many others. 相似文献

This study reports the inhibitory effect of single walled carbon nanotubes (SWCNTs) on biofilm formation from Bacillus anthracis spores. Although the presence of 50 to 100 μg ml?1 of SWCNTs in the suspension increased spore attachment in the wells of 96-well plates, the presence of 200 μg ml?1 of SWCNTs in the germination solution decreased the germination percentage of the attached spores by 93.14%, completely inhibiting subsequent biofilm formation. The inhibition kinetics of 50 μg ml?1 SWCNTs on biofilm formation showed that this concentration inhibited biofilm formation by 81.2% after incubation for 48 h. SWCNT treatment in the earlier stages of biofilm formation was more effective compared to treatment at later stages. Mature biofilms were highly resistant to SWCNT treatment. 相似文献

Dithiothreitol (DTT)-treated spores of Clostridium perfringens were much more sensitive to lysis by H2 O2 in the presence of Cu2+ than untreated spores. Lysis was greatly inhibited by hydroxyl radical (.OH) scavengers such as thiourea, dimethylthiourea and dimethylsulfoxide, suggesting that lysis of spores by H2 O2 involves formation of OH by Cu2+ -catalysed decomposition of the peroxide. DTT-treated spores took up Cu2+ at almost the same rate and extent as did isolated cortical fragments. Hydrogen peroxide caused both the decrease in optical density and the hexosamine solubilization of cortical fragments which bound Cu2+ . 相似文献

Water from a hypertrophic lake rich in filamentous blue-green algae was passed through a continuous-flow system of aquaria containing Daphnia magna, and a control system without Daphnia. Daphnia caused a significant decrease in the blue-green algal density, and a two-fold reduction in filament length. It is suggested that feeding activity of Daphnia may result in an increase in the availability of blue-green filaments to filter-feeding cladocerans. 相似文献

Inhibitory effect of bilirubin on complement-mediated hemolysis 总被引:1,自引:0,他引:1
We investigated the in vitro action of the bile pigments, unconjugated bilirubin (UB) and bilirubin monoglucuronide (BMG) on complement (C) cascade reaction. Both UB and BMG inhibited hemolysis in the classical pathway (CP) in a dose-dependent manner at low micromolar concentrations, UB showing a stronger effect than BMG. The analysis of the action of UB on the hemolytic activity of the C1, C4, C2 and C-EDTA components of the C cascade revealed that the C1 step was the most inhibited. An enzyme immunoassay was developed to evaluate the effect of UB on the binding of C1q, one of the subcomponents of C1, to human IgM and IgG. The study demonstrated that the unconjugated pigment interferes both the C1q-IgM and -IgG interactions, thus tentatively explaining the inhibitory action of UB on hemolytic activity of C1. We conclude that the anti-complement effect of UB is mainly exerted on the C1 component, the recognition unit of CP. The potential clinical implication of the reported effects in hyperbilirubinemia is discussed. 相似文献

The effect of polyelectrolytes on the stability and catalytic characteristics of oligomeric enzymes--pig muscle lactate dehydrogenase (LDH) and bovine liver glutamate dehydrogenase (GDH)--was studied by fluorescent spectroscopic and steady state kinetic methods. It was shown that the binding of negatively charged polyelectrolytes--polystyrene sulfonate, polymethacrylate, and polyphosphate--destroys the tertiary and partially the secondary structure of LDH and GDH, resulting in their complete inactivation at pH < 7. The concentrations of polyelectrolytes needed for inhibition of the enzymes were in this case by two or more orders of magnitude lower than the corresponding concentrations for monomers--toluene sulfonate, methacrylate, and phosphate. The affinity of the substrate (pyruvate) for LDH did not vary in the presence of the polyelectrolytes, but the inhibition was removed by excess of substrate. We propose that the oligomeric state of enzymes causes polyelectrolytes to act on them in a special manner, this special effect differing significantly from the effect of polyelectrolytes on monomeric enzymes. The effect consists in that polyelectrolytes cleave the oligomeric structure of the enzymes, this "cleaving" effect being higher the greater the hydrophobicity of the polyelectrolyte chain. 相似文献

目的 研究企业常用消毒剂对于洁净室环境监测分离的菌株样本的抑制作用。方法 通过VITEK2-COMPACT全自动细菌鉴定及药敏分析系统鉴定收集到的环境菌株。对3种消毒剂(碘伏、无水乙醇和苯扎溴铵)进行梯度稀释,利用打孔法研究3种消毒剂在不同含量下对环境菌株的抑制作用。结果 共检出革兰阳性菌8种、革兰阴性菌2种、酵母菌1种、芽孢杆菌2种;苯扎溴铵对革兰阳性菌的抑菌能力都较强,随着含量的降低,抑菌作用逐渐减弱;碘伏对革兰阴性菌及酵母菌的抑制作用都非常强,随着含量降低,抑菌作用逐渐降低。3种消毒剂对2种芽孢杆菌的抑制作用均有限。结论 用1.00%苯扎溴铵和0.50%的碘伏抑菌作用都非常强。另外,应配合使用杀孢子剂,避免芽孢杆菌孢子在空气中传播。 相似文献
