The migration and distribution of Ancylostoma caninum larvae in the tissues of chickens, infected orally with 1,000 larvae, were studied. Larval yield at necropsy from different organs after digestion with artificial gastric juice revealed a 62.9% recovery four hours after inoculation, followed by a sharp decline to 5.4% at 72 hours. Larvae were found in the heart within four hours, the lungs within eight hours and the liver within 12 to 18 hours but no larvae were recovered from the spleen, kidney or brain. Migration in the muscles of head, neck, thorax and abdomen was detected at 12 hours and was maximal at 36 hours. The establishment of patent infection in the definitive host was studied by feeding infected chicks to hookworm-free pups (one chick/pup) 48 hours, 7 days and 14 days after infection. The mean worm burden at necropsy was highest (15) in the pups fed with chicks 48 hours after infection and was three and nil in the other groups respectively.  相似文献

The incorporations of uridine-3H, thymidine-3H, and leucine-3H were studied in the process-forming regions of the anal-fin rays of the ethisterone-treated females of Oryzias latipes. The activity of alkaline phosphatase was also studied. The increased incorporation of uridine-3H was detected between 12 and 24 hours of ethisterone treatment, attaining the maximum at 24 hours. The percentage of thymidine-3H labeled nuclei increased rapidly between 48 and 84 hours. The incorporation of leucine-3H was found to increase during the first 12 hours, attaining a constant level at 24 hours. An additional increase in incorporation of leucine-3H took place at 60 hours, the incorporation coming up to the maximum at 72 hours. In the horny substance secreted by the scleroblast mass, grains in the autoradiograph were detected at and after 72 hours. Alkaline phosphatase activity was manifested between 48 and 72 hours. These results seem to correspond to the histological changes, such as the appearance of the precursor cells of scleroblasts at 48 hours, the formation of scleroblast mass during the next 24 hours, and the initiation of horny substance secretion at 72 hours.  相似文献

The state of hepatocyte chromatin (the area occupied by the regions of condensed chromatin on ultrathin sections and the quantity of perichromatin RNP fibrils which was estimated by the area of the fibrillar zone and the concentration of fibrils within the same zone) were studied within the first hours after partial hepatectomy of guinea pigs. The area occupied by the regions of condensed chromatin on preparations with differentially revealed DNP and RNP components decreased by 12% in 2.5 hours since the operation had been performed, became normal in 5 hours, and again decreased by 30% in 9 hours. Decondensation of chromatin was accompanied with the increase of the number of perichromatin RNP fibrils, products of template activity of chromatin, and the rise of ethidium bromide binding. The binding of ethidium bromide by the chromatin of hepatocytes increased by 39% in 2.5 hours, returned to the control level in 5 hours and again increased by 22% in 9 hours.  相似文献

The effect of biliary obstruction in the rat on several hydroxylations involved in the formation and metabolism of bile acids was studied. The hydroxylations studied were all catalyzed by the microsomal fraction of liver homogenate fortified with NADPH. The rate of 7α-hydroxylation of cholesterol increased two- to threefold between 24 and 48 hours after ligation of the bile duct and remained at this level the next 48 hours. During the first 24 hours of obstruction the rates of 1 2α-hydroxylation of 7α-hydroxy-4-cholesten-3-one and 7α-hydroxylation of taurodeoxycholic acid decreased but returned to control levels between 24 and 48 hours after operation. The rate of 6β-hydroxylation of lithocholic acid and taurochenodeoxycholic acid increased gradually and reached a plateau between 24 and 48 hours at which time the rate was two to three times faster than in the controls. The increase in 6β-hydroxylase activity was reflected in the pattern of the bile acids excreted in urine. After 48 hours of obstruction β-muricholic acid accounted for 50% or more of the bile acids in urine.  相似文献

The effects of gamete coincubation time on porcine in vitro fertilization for 4, 6 or 8 hours in Trial 1 or for 1, 2, 3 or 4 hours in Trial 2 were studied. The 876 oocytes were recovered from oviducts of prepubertal gilts. Ovulation was induced. Excessive spermatozoa attached to zona pellucida were removed by pippeting after coincubation (1 to 8 hours). Optimum results were obtained after 3 to 4 hours of coincubation, when 23 to 29% of the oocytes were fertilized by a single spermatozoa. Shorter intervals of coincubation were characterized by reduced percentages of fertilized oocytes and longer intervals of coincubation by increased rates of polyspermic fertilization. The employed sperm concentration was deliberately high (2 x 10(6) sperm/ml). The results show that a coincubation time of 4 hours may be optimal for pig in vitro fertilization. Under these assay conditions, modification of other factors such as sperm concentration, medium volume and physiological environment may increase the percentage of monospermic fertilization.  相似文献

The disappearance of methotrexate (MTX) from the serum after an intravenous bolus injection and intravenous infusion was studied over 24 hours in eight and four patients respectively. Probenecid given at the same time as the bolus injection delayed the disappearance of MTX from the serum and resulted in enhanced concentrations throughout the 24 hours studied. At 24 hours the mean concentration was four times higher than in patients not given probenecid. Overall serum concentrations were even greater than those in patients who had received MTX by intravenous infusion. We suggest that smaller doses of MTX may be given and treatment costs thereby reduced if probenecid is given in addition.  相似文献

The effect of a single antioxidant (dibunol-D) injection (100 mg/kg body weight) on the functional activity of adrenal cortex (AC), thyroid gland (TG) and tropic hormone production by adenohypophysis (AH) has been studied in old and adult rats. For 48 hours following D administration two-phase changes in adrenocorticotropic function of AH and steroidogenesis were detected in the AC: activation during the first hours was followed by suppression 24 hours later, and recovery 48 hours later. Thyrotropic AH function and thyroidogenesis were found to be decreased during the first hours of D effect. Thyroidogenesis recovery by the end of the first day was delayed as compared to the recovery of AH thyrotropic function. It is suggested that the mechanisms of D action are based on its effects mediated by changes in the functional activity of endocrine glands and associated with resetting of endocrine regulation of body functions.  相似文献

The authors studied changes in the rabbit's blood picture in the first 24 hours after the administration of three different doses of endotoxin. The most pronounced changes were observed in the white blood component, particularly the granulocytes, which almost vanished from the blood stream immediately after the endotoxin was injected. In 24 hours granulocytopenia was succeeded by marked granulocytosis. Changes in the lymphocytes were smaller; the lymphocyte count fell slightly about 3 hours after the injection of endotoxin, but by 24 hours it was almost normal again. The platelet count also fell after the administration of endotoxin, but the red blood picture remained virtually the same.  相似文献

通过对小鼠肌母细胞C2C12的培养,研究C2C12细胞的增殖与分化的关系以及胰岛素在细胞分化过程中的作用。在对照组中,C2C12细胞增殖占了明显的优势,细胞形态几乎没有发生变化;而在实验组中,C2C12细胞在换为分化培养基24小时后,就出现了部分细胞衰亡和死亡的现象,尤其是在48小时细胞的死亡率达到最高,存活细胞开始从增殖期进入分化期,72小时出现了少量肌管,在96小时细胞分化效果达到最好。而在添加了胰岛素的分化培养基中的细胞分化效果明显好于没有添加胰岛素的分化培养基中的细胞,结果表明,胰岛素促进C2C12细胞的分化。  相似文献

The changes of adenine nucleotide and adenylate energy charge (AEC) during the development of mitochondria in imbibed mung bean cotyledons and the relationship between these changes and cellular energy status are studied. After cotyledons were imbibed in water for two hours, mitochondrial cristae were not observed, but for 12 hours, they appeared obviously on the inner membrane. With the structural integrity of the mitochondria, the functional mitochondria were graduately shown. For instance, the activity of H+-ATPase of cotyledons imbibed for 24 hours was about twice higher than that of 2 hours. The ATP content and the AEC value in the cotyledons imbibed for 24 hours increased sharply and the AMP decreased, but these were not observed in the mitochondria of the cotyledons imbibed either for 24 hours or 2 hours. When the cotyledons were imbibed in 1 × 10-4 mol/l or 5 × 10-4 mol/l DNP solution for 24 hours, the ATP and the AEC in the Cells exhibited a rapid decrease, but in the mitochondria they remained canstant. In the same DNP solution with cotyledons for 24 hours, the activity of mitochondrial adenylate kinase (AK) not only was not decreased but also increased by about 50% over the control. This result shows that the energy equilibration in the mitochondria seems likely to be regulated by adenylate kinase locating between inner and out membranes of the mitochondria.  相似文献

The effects of 17alpha-hydroxyprogesterone capronate (17-OH-P) on spontaneous uterine motility and on oxytocin-induced uterine contraction during the postabortion period were studied in 22 women. The effects of 17-OH-P were present 8 hours after administration and still appreciable after 12 hours, but were considerably diminished after 24 and 48 hours. The efficiency of 17-OH-P in inhibiting uterine contraction after administration of oxytocin is clearly inferior to that of natural progesterone.  相似文献

Gross changes in protein and nucleic acid were studied in germinating wheat seeds. The nucleic acid fraction was separated on columns of methylated albumin-keiselguhr. It was found that more than 50% of the transfer RNA was lost from the embryo in the first 10 to 15 hours of germination. This was followed by a period of rapid resynthesis of transfer RNA, to the normal level at about 20 hours. The decline and increase in transfer RNA was accompanied by a change in the ratios of certain amino acid acceptor species. Evidence is also presented that an embryo ribonuclease is lost during the first 10 to 15 hours, followed by the appearance of a second seedling ribonuclease between 15 and 30 hours of germination.  相似文献

The possibilities of the transport systems manufactured by Copan (Italy) and Deltalab (Spain) were studied on 15 microbial strains: representatives of the family Enterobacteriaceae (enterobacterial swabs with Cary-Blair medium) and the genus Streptococcus, as well as the species Neisseria meningitidis, Haemophilus influenzae (universal swabs with charcoal-enriched Amies medium). Microorganisms were shown to retain their viability (in colony-forming units, %) for 48 hours in the systems of both firms. H. influenzae exhibited greater viability in the system manufactured by Copan than in that manufactured by Deltalab (respectively, 62% and 28% in 24 hours, 19% and 6% in 48 hours).  相似文献

The development of mitochondria in the course, of imbibition ofmung bean cotyledons was studied. Mitochondria were prepared by differential centrifugation, and purified by discontinuous sucrose, density gradient centrifugation. Theelectron micrographs revealed that the mitochondria isolated from dry seeds cotyledonslook like small vesicles, when cotyledons were imbibed for two hours, the mitochondrial cristae were not observed, but after four hours, a few cristae appeared on the innermembrane. Till 12 hours, the inner membrane systems wore well-developed. Al this time.all the space in mitochondria are filled with cristae. With the structural integrity of the mitochondria, the functions of oxidation and phosphorylation were graduallyshown, For instance, ADP/O ratio and RCR were not able to be measured in the imbibition of 2 and 4 hours, but at 6th hours, ADP/O Was increased by 0.6, RCR nearly 2.0, After 24 hours imbibition, ADP/O and RCR were increased to 1.5 and 3.5 respectively. The activity of cytochrome oxidase reached the highest after imbibition for 3 hours (2.54 OD/mg protein/min). If cotyledons were imbibed continuously, the activityof this enzyme in mitochondria remained constant. The activity of ATPase located onthe inside of the mitochondrial inner membrane was gradually enhanced from the beginnning of imbibition. These results suggest that the assembly of cytochrome oxidaseand ATPase on mitochondrial membrane may not be synchronous.  相似文献

A correlation between the length of the olfactory cilia and the electrical activity of the olfactory epithelium was studied in newts living in water and on land. The olfactory cilia grew when newts were transferred onto land. The cilia in the olfactory bud became longest in 108 hours after the transfer and then became shorter, while those in the interstitium only gradually elongated. Slow potentials were evoked in the epithelium by the application of odorous fluids but not by odorous vapors for 20 hours after the transfer. Thereafter, the slow potential began to appear in response to odorous vapors and reached maximal magnitude between 60 and 70 hours after the transfer, while it was not evoked by odorous fluids in this period. In the later stage, the slow potential to odorous vapors decreased in magnitude and disappeared 120 hours after the transfer, while it began to reappear in response to odorous fluids. When these changes in the slow potential were compared with those in the cilium, a discrepancy was found between the period of maximal potential magnitude and that of maximal cilium length.  相似文献

Utilizing histamine electrophoresis, the degree and duration of "antihistaminic" effect of varying doses of Pyribenzamine(R) administered by different routes were studied. Delayed action Pyribenzamine in 50 mg. and 100 mg. dosage exerted a small but measurable anti-whealing effect five hours after ingestion. The larger dose exerted approximately twice the effect of the smaller. The anti-whealing activity lasted five to six hours. Pyribenzamine given intravenously affected the whealing response one hour after administration. A peak of activity was reached two to two and a half hours after injection, and "antihistaminic" effect continued for a total of five hours. The inunction of 250 mg. and 500 mg. of Pyribenzamine in an ointment base resulted in sufficient absorption of the drug to produce a measurable anti-whealing effect. The "antihistaminic" activity was noted three hours after application and lasted 10 to 12 hours from the time of inunction.  相似文献

The immune response of guinea pigs to Q fever vaccine following 75 to 250 R (60 to 180 rads) of acute whole-body irradiations was investigated. Complement-fixing (CF) antibody titers and protection against febrile response to challenge with virulent Coxiella burnetii were studied. Exposures ranging from 75 to 250 R, 24 hours prior to inoculation, did not detectably alter the CF antibody response. Similar results were observed with 175 R delivered 48 or 72 hours before immunization. Protection against febrile response to challenge with 10(3) median fever doses of C. burnetii was seen in animals irradiated with 175 R, 24 or 72 hours before immunization. Significant protection was detectable at 14, 21, and 42 days after immunization in both irradiated and nonirradiated animals. Acute irradiation of the degree studied increases the mortality in normal animals infected 15 to 17 days later with virulent C. burnetii. The lethal effect could be prevented by use of Q fever vaccine.  相似文献

Post-mortem changes of cytoplasmic RNP particles from rat skeletal muscles are studied using sucrose density gradient. The polysome destruction is found to beign within 1-2 hours after the animal death. However, a part of polysomes turned to be stable at least within 48-60 hours after the death. These polysomes incorporated 14C-leucine into acid insoluble fraction in cell-free system and were decomposited by RNase treatment (10 mkg/ml). The data suggest that within 24-48 hours of the after-death destruction the decay begins to free monoribosomes while there are still active polysomes in muscles.  相似文献

Diurnal variations in serum testosterone levels were studied in six adult maleMacaca radiata. Though the testosterone levels fluctuated considerably during the 24-hr period, a distinctive diurnal pattern characterized by lower concentrations during day hours and higher concentrations during night hours was observed.  相似文献
