为探讨宁夏回族原发性膝骨性关节炎(Osteoarthritis, OA)与瘦素受体基因(Leptin receptor, LEPR)A668G位点单核苷酸多态性(SNPs)之间的关系, 文章运用病例-对照研究, 通过聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术, 对148例兼具原发症状和影象证据的宁夏回族膝OA患者以及155名年龄、性别相匹配的对照群体进行LEPR A668G SNPs检测, 并进行测序验证, 分析LEPR基因多态性与膝OA的易感关联。研究表明, 膝OA组瘦素(Leptin, LEP)水平显著高于对照组(P<0.001), 血浆可溶性瘦素受体(sLEPR)水平较对照组明显降低(P<0.001), 膝OA组LEPR A668G位点AG/GA+GG基因型和G等位基因的分布频率和对照组存在差异(P=0.008和P=0.024)。研究结果提示, LEPR A668G位点的多态性可能与宁夏回族人群中膝OA易感性相关, 可以作为预测宁夏回族膝OA发病危险的遗传标记及早期防治的候选基因之一。  相似文献

目的:研究新疆维吾尔族114名运动员(速度力量型项目43人,耐力型项目35人,足球36人)和441名普通人肌型肌酸激酶(CKMM)基因A/G多态性与运动能力相关性。方法:聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)法。结果:1维吾尔族普通人群CKMM基因A/G多态性频率(AA、AG、GG)分别为0.497、0.392、0.111,经检验符合H-W平衡(x~2=2.72,P0.05),具有群体代表性;2速度力量型项目运动员3种基因型频率分别0.442、0.302、0.256,GG基因型和G等位基因频率显著高于对照组,两组差异有显著性(P0.05,df=2);3耐力型项目运动员3种基因型频率分别为0.571、0.400、0.029,A等位基因频率高于对照组,但差异没有显著性(P0.05,df=2);4足球运动员3种基因型频率分别为0.472、0.361、0.167,G等位基因频率高于耐力组低于速度力量组,与对照组相比亦无显著性差异(P0.05,df=2)。结论:CKMM基因A/G多态性与维吾尔族速度力量素质呈显著相关,GG基因型和G等位基因可用于速度力量运动员选材的一个辅助因子,但没有发现A/G多态性与耐力素质和足球成绩存在显著相关性。  相似文献

探讨细胞色素P450 19 (CYP19) 基因Val80多态性及护骨素(OPG) 基因A163G多态性与绝经后女性骨密度 (BMD) 的关系。随机选择居住在重庆的绝经后女性200例, 采用多聚酶链反应-限制性片段长度多态性法检测Val80及A163G多态性, 采用Norland公司XR-46系列双能X线骨密度仪测量股骨近端及腰椎BMD。 200名绝经后女性中Val80基因型GG、GA及AA的频率分别为19.5%、44.5%及36.0%; A163G基因型GG、GC 及CC的频率分别为: 13.0%, 42.0%及45.0%; 基因型频率分布均符合Hardy-Weinberg平衡 (P>0.05)。协方差分析及多元逐步回归分析显示CYP19基因第3外显子Val80多态性与绝经后女性BMD无相关性 (P>0.05)。除大转子外, A163G位点AG/GG/AG+GG基因型者股骨颈、Ward’s三角及腰椎BMD均较AA基因型者低, A163G基因型与股骨颈、Ward’s三角及腰椎BMD有相关性 (P<0.05)。OPG基因启动子区A163G多态性分布存在明显的种族差异, 且与绝经后女性BMD有一定关联, AA型对BMD具有一定的保护作用, G等位基因是BMD降低的危险因素。  相似文献

目的:探讨硫酸基转移酶(sulfotransferase,SULT)lA1、细胞间粘附分子(ICAM5)基因多态性与女性乳腺癌易感性的关系.方法:采外周血DNA后用等位基因特异性扩增法(allele specific amplification,ASA)检测青岛市200例正常对照者和160例乳腺癌患者的SULTIA1、ICAM5基因多态性分布,并进行统计学分析.结果:(1)SULTlA1 Arg/Arg、Arg/His、His/His三种基因型分布在对照组和病例组之间的差异无显著意义(P=0.103);病例组、对照组His等位基因频率分别为19.5%和9.2%(P=0.039),此差别有统计学意义;在淋巴结转移方面SULTIA1基因三种基因型在阴、阳性组间的差异有统计学意义(P=0.038).(2)ICAM5基因各基因型及等位基因分布频率在病例组和对照组间的差异无显著意义(P=0.245,P=0.294);从临床病例分型方面进一步分析,基因型GG与携带变异基因A的GA及AA基因型相比差异均无统计意义.结论:SULTlA1 His等位基因与汉族女性乳腺癌的发生可能相关.  相似文献

钙蛋白酶抑制蛋白基因是影响猪肉质性状的候选基因之一。本研究以125头地方猪和117头外来猪为材料,研究CAST基因的多态性。结果在CAST基因上检测到一个多态性位点(A876G),并引起了氨基酸残基的改变Lys250 Arg。在地方猪种中仅检测到G (Arg)等位基因,而在外来猪种中A (Lys) 和 G (Arg)两个等位基因均检测到。基因型与肉质性状的关联性分析结果表明,CAST基因型与肌肉的嫩度,屠宰45 min后的pH值及滴水损失存在强相关,又由于地方猪种与外来猪种的肉质性状间存在显著差异。因此,在CAST基因上检测到的多态型位点Lys250Arg的基因型效应有待于进一步研究,并将其有效应用于商品猪生产中。  相似文献

为了探讨粘蛋白(MUC1)基因568位点A/G单核苷酸多态性与胃癌遗传易感性的关系, 采用序列特异性引物-聚合酶链反应(Sequence specific primers PCR, PCR-SSPs)检测来自辽宁地区人群138例胃癌患者及与其配比的131例对照个体MUC1 568 位点A/G多态性, 以ELISA法检测血清H. pylori IgG抗体。结果显示:(1)对照人群MUC1基因568位点AA、AG、GG 3种基因型分布频率分别为73.3%、22.1%、4.6%; (2)胃癌组MUC1 AA基因型携带频率显著高于正常对照组(P=0.03), 携带MUC1 AA基因型个体胃癌的发病风险增高到1.92倍; (3)以MUC1 AG+GG基因型并血清幽门螺杆菌(H. pylori)IgG抗体阴性的个体为对照, AG+GG基因型并H. pylori IgG抗体阳性个体、AA基因型并H. pylori IgG抗体阴性个体、AA基因型并H. pylori IgG抗体阳性个体胃癌患病风险增高, 但3组各组间差异均无统计学意义(P>0.05)。说明MUC1基因568位点A/G多态与胃癌的遗传易感性相关; MUC1 A/G基因多态性和H. pylori感染在胃癌发生发展过程未见交互作用。  相似文献

目的:探讨TLR4基因rs10983755 A/G单核苷酸多态性(SNP)与非小细胞肺癌(NSCLC)易感性的相关性。方法:采用病例-对照研究方法纳入160例非小细胞肺癌患者(NSCLC组)和160例健康对照(NC组),利用MassARRAY飞行时间质谱生物芯片系统对TLR4基因rs10983755位点的单核苷酸多态性进行分型检测,并进行统计学分析。结果:rs10983755等位基因频率在中国汉族NSCLC患者和健康人群中的分布差异具有统计学意义(P0.05),A等位基因携带者NSCLC发生风险是G等位基因携带者的1.821倍(95%CI=1.124~2.906);rs10983755基因型频率在NSCLC患者和健康人群中分布差异具有统计学意义(P0.05),AA+AG基因型NSCLC发生风险是GG基因型的2.103倍(95%CI=1.118~3.898)。结论:TLR4基因rs10983755 A/G单核苷酸多态性与NSCLC的易感性显著相关,A是风险等位基因。  相似文献

目的:研究中国北方人群TCF7L2基因rs11196218和rs290487多态性的分布特点及其与血脂谱的相关性。方法:对1255例中国北方人群TCF7L2基因进行单核苷酸多态性检测,同时检测其血脂水平,分析血脂水平与上述基因的相关性。结果:在中国北方人群中TCF7L2基因rs11196218位点AA,AG和GG基因型频率分别为6.61%,39.68%和53.71%,等位基因A、G频率分别为26.45%和73.55%;而rs290487位点TT,CT和CC基因型频率分别为37.45%,45.98%和16.57%,等位基因T,C频率分别为60.44%和39.56%。rs11196218A/G与血清低密度脂蛋白、总胆固醇水平具有相关性(P≤0.05),而rs290487C/T与血脂水平无相关性(P0.05)。结论:在中国北方人群中存在TCF7L2基因rs11196218A/G和rs290487C/T单核苷酸多态性,且其变异频率大,rs11196218A/G与血脂异常相关。  相似文献

Alpha-1-antitrypsin (AAT) plays an important role in the pathogenesis of emphysema, the pathological lesion underlying the majority of the manifestations of Chronic Obstructive Pulmonary Disease (COPD). In this study we tested the hypothesis that common AAT polymorphisms influence the risk of developing COPDs. We investigated PiM1 (Ala213Val), PiM2 (Arg101His), PiM3 (Glu376Asp), PiS (Glu264Val) and PiZ (Glu342Lys) SERPINA1 alleles in 100 COPD patients and 200 healthy controls. No significant differences were observed in allele frequencies between COPD patients and controls, neither did haplotype analysis show significant differences between the two groups. A cross-sectional study revealed no significant relationship between common SERPINA1 polymorphisms (PiM1, PiM2, PiM3) and the emphysematous type of COPD. In addition, FEV(1) annual decline, determined during a two-year follow up period, revealed no difference among carriers of the tested polymorphisms.  相似文献

Our studies have shown that the genotype and allele frequencies of polymorphisms G(-1607)GG of MMPI gene, C(-1562)T of MMP9 gene and A(-82)G of MMP12 gene do not significantly differ in the samples of chronic obstructive pulmonary disease (COPD) patients (N = 318) and healthy controls (N = 319) dwelling in Bashkortostan Republic. However, association of (-1562)T allele of the MMP9 gene with the severity of COPD disease progression has been revealed. In COPD patients at stage 4 of the disease, the frequency of allele T was significantly higher that in patients with the stages 2 and 3 (15.89% versus 8.38%; chi2 = 7.804, d.f. = 1, P = = 0.005; OR = 2.06 95% CI 1.22-3.49). The distribution of the genotype frequencies of C(-1562)T polymorphism of MMP9 gene significantly differed between the patients with various COPD severity (chi2 = 9.849, d.f. = 2, P = 0.007). The individuals with rare genotype TT were revealed only among patients with severe COPD form (3.97% versus 0%; chi2 = 4.78, P = 0.029, Pcor = 0.058). Analysis of this polymorphism in patients with early COPD onset (younger than 55 years old) has shown a significant increase in the allele Tfrequency in the group of patients with severe COPD (stage 4 according to GOLD) compared to the patients of the same age but with less severe COPD progression (chi2 = 5.26, d.f. = 1, P = 0.022). As the major clinical characteristics of stage 4 COPD is the development of pulmonary emphysema as well as bronchial walls deformation, we suggest that the increased expression of MMP9 gene caused by genetic polymorphism in the gene promoter is important in the early development of serious complications of the disease.  相似文献

目的:探讨内皮型一氧化氮合酶(eNOS)基因894G/T多态性与原发性高血压(EH)合并脑梗塞(CI)的关系。方法:应用聚合酶链反应限制性片段长度多态性方法检测湖北地区汉族74例健康者(NT组)、103例原发性高血压无合并症者(EH组)及70例原发性高血压合并脑梗塞者(EH-CI组)的eNOS基因型;生化技术测定其血脂、一氧化氮代谢物(NOM)水平。结果:EH组及EH-CI组患者的T等位基因频率分别为0.224和0.321,均显著高于NT组(P<0.05);且两者之间的T等位基因频率差异显著性(P<0.05);EH-CI组中,GT+TT基因型者的舒张压显著高于GG基因型者(P<0.05),而NOM显著低于GG基因型者。结论:eNOS基因894位G/T多态性可能与汉族高血压病患者伴脑梗塞有关,该位点多态性可能使T等位基因携带者NOM减少,进而参与EH-CI发病。  相似文献

To determine whether leptin receptor (LEPR) 223A>G polymorphism has an effect on the plasma leptin levels and the macroangiopathic complications in type 2 diabetes mellitus (T2DM). The genotypes and allelic frequencies of the LEPR 223A>G were examined with polymerase chain reaction and restriction fragment length polymorphism in 301 patients with T2DM and 172 unrelated healthy subjects. The plasma concentrations of leptin were determined in all subjects. The mean plasma leptin levels in the T2DM group were significantly higher than that of controls and the plasma levels of leptin were higher in diabetic patients with macroangiopathy than in patients without macroangiopathy (P < 0.05). The genotype (GG, AG and AA) distribution of 223A>G polymorphism was 58.3, 32.5, and 9.2% in diabetic patients with macroangiopathy, 75.3, 22.1, and 2.6% in patients without macroangiopathy, and 70.3, 27.5, 2.2% in controls respectively, a significant difference was found between diabetic patients with and without macroangiopathy (P < 0.05). The frequency of the allele A was higher in patients with macroangiopathy than in patients without macroangiopathy (25.6 vs. 16.3%; P < 0.05). Moreover, the plasma leptin levels were markedly higher in patients with AA genotype than those with AG or GG genotype in patients with macroangiopathy (P < 0.05). The LEPR 223A>G gene polymorphism associated with a predisposition to increased plasma leptin levels could constitute a useful predictive marker for diabetic macroangiopathy.  相似文献

Objective: To investigate the association between leptin levels, polymorphisms in the leptin receptor (LEPR) gene, and weight gain. Research Methods and Procedures: From two large prospective cohorts in The Netherlands (n = 17, 500), we compared the baseline leptin of 259 subjects who had gained an average of 12.6 kg (range 5.5 to 33 kg) with 277 subjects who kept stable weight (range ?2.6 to 3.1 kg) after a mean follow‐up of 6.8 years. Three polymorphisms in the LEPR gene (Lys109Arg, Gln223Arg, and Lys656Asn) were determined. Results: Weight gainers had significantly higher baseline leptin levels than those who kept stable weight (odds ratio = 1.27, 95% confidence interval 1.1 to 1.5, per SD increase in loge‐transformed leptin). Weight gainers with the Arg109 or the Arg223 alleles had higher leptin levels compared with the noncarriers of these alleles. Only among men, the association between leptin and weight gain tended to be stronger among those with an Arg223 allele compared with those without this mutation. Discussion: Relatively high leptin levels predict weight gain, suggesting that leptin resistance plays a role in the development of obesity in the general population. Higher leptin levels for those with a Lys109Arg or Gln223Arg mutation (or a linked other marker) may imply that these subjects have a modified functional leptin receptor. However, the role of these mutations on weight gain is limited.  相似文献

Based on the protease-antiprotease hypothesis in the pathogenesis of chronic lung diseases, we investigated the influence of Z and S mutations and TaqI-polymorphism of alpha1-antitrypsin gene (PI) and Ala - 15Thr polymorphism in signal peptide of alpha1-antichymotrypsin gene (AACT) in patients with COPD (n = 239), nonobstructive chronic bronchitis (n = 34), brochiectases (n = 33), chronic infant lung disease (n = 151) and cystic fibrosis (n = 57). The allele and genotype frequency of any genetic polymorphism was not statistically different between the groups and control subjects (n = 225). Furthermore, promoter polymorphism G - 1607GG in interstitial collagenase gene (MMP1) was determined in patients with COPD, chronic nonobstructive bronchitis and bronchiectases. In patients with COPD it was marked prevalence of mutant homozygous genotype GG/GG compared to controls (30.6 % and 18.0%, respectively, OR = 1.99; 95% CI 1.11-3.59). These findings suggest that genetic polymorphism in the promoter of MMPI gene may be associated with individual susceptibility to the development of COPD.  相似文献

Genetic polymorphisms of p53 and its negative regulator murine double minute 2 homolog (MDM2) have been shown to be closely associated with tumorigenesis in a variety of human cancers. In the present study, single nucleotide polymorphism (SNP) at p53 codon 72 and MDM2 promoter 309 was examined for germline DNA samples from 102 endometrial cancer cases and 95 controls using polymerase chain reaction-based fragment analysis. There were no significant differences in the genotype and allele prevalence between control subjects and endometrial cancer patients for p53 codon 72. The GG genotype frequency of MDM2-SNP309 was statistically higher in endometrial cancer patients than that in normal healthy women when compared with the TG genotype ( P = 0.0088). However, no statistically significant differences were found between the TT and TG or GG genotype frequencies and allele prevalence. Interestingly, the combination of the homozygous Arg/Arg genotype of p53 codon 72 and homozygous GG genotype of MDM2 SNP309 polymorphisms was significantly associated with the risk of endometrial cancer (odds ratio = 3.28, 95% confidence interval = 1.13 to 9.53, P = 0.0212). The homozygous variants of wild p53 codon 72 and mutant MDM2 promoter 309 may cooperatively increase the risk of endometrial cancer in a Japanese population.  相似文献

We aimed to investigate whether polymorphisms LEP G-2548A and LEPR Q223R in the human leptin (LEP), and leptin receptor (LEPR) genes are associated with obesity and metabolic traits in a sample of Romanian population. Two hundred and two subjects divided in obese (body mass index, BMI ? 30 kg/m2), and non-obese were included in this study. The polymorphisms were genotyped using polymerase chain reaction (PCR) followed by restriction fragment length polymorphism (RFLP) analysis. The results showed no significant differences in LEP and LEPR genotype and allele frequencies between obese and non-obese subjects. Logistic regression analysis showed that LEP -2548GG genotype presented an increased risk of obesity (p = 0.013, OR = 1.003, 95% CI = 1.000-1.007), after adjusting for age and gender. The association analysis with metabolic syndrome quantitative traits showed that homozygous for LEP -2548G allele had significantly higher leptin levels (17.2 ± 6.6 ng/ml vs. 13.2 ± 4.9 ng/ml, p = 0.011), and carriers of R allele had higher levels of triglycerides (p = 0.017) and glucose (p = 0.040), and enhanced systolic (p = 0.015) and diastolic blood pressure (p = 0.026), after adjustment for age, gender, and BMI. These results indicate that LEP G-2548A and LEPR Q223R SNPs may not be considered as genetic risk factors for obesity in a sample of Romanian population. However, LEP -2548GG genotype appear to be important in regulating leptin levels, whereas the LEPR 223R allele might predispose healthy subjects to develop metabolic disturbances.  相似文献

采用病例.家系对照和随机病例.对照两种设计,分析了603例样本脂联素基因(Adiponectin,APMl)单核苷酸多态性(SNP)rs13061862(T45G)与湖北汉族人群2型糖尿病的相关性.在所有样本中,2型糖尿病病人的G等位基因及GG基因型频率显著高于正常人(G:42.0%比21.7%,P<0.001;GG:13.6%比4.5%,P=0.032);在180个病例.家系对照中,2型糖尿病患者的GG基因型频率显著高于对照组(GG:17.8%比5.6%,P=0.011);在423个随机病例.对照中,2型糖尿病患者GG基因型频率也显著高于对照组(GG:12.2%比3.9%,P=0.025);单因素Logistic回归分析显示,GG基因型是2型糖尿病的危险因子(OR=3.58,95%C/=1.70-7.54).这些结果表明,脂联素基因SNPT45G多态性与湖北汉族人群2型糖尿病的发生发展相关,GG基因型是中国湖北汉族人2型糖尿病的遗传危险因素.  相似文献

目的:探讨老年慢性阻塞性肺疾病(COPD)患者的营养状况以及血清甲状腺素、瘦素水平变化,并分析其相关性。方法:选择我院2015年6月至2017年12月收治的64例老年COPD患者作为研究对象,按照营养状态分为营养不良组(34例)和非营养不良组(30例)两组,并选择同期来院检查的32例健康人作为对照组,比较三组患者的营养状况以及血清甲状腺素、瘦素水平,并分析血清瘦素水平与营养状况以及血清甲状腺素的相关性。结果:营养不良组患者的体重、体质量(BMI)、血清清蛋白(ALB)、血红蛋白(Hb)、肩胛下皮褶厚度(SSF)、三头肌皮褶厚度(TSF)、上臂周径(MAC)、上臂肌围长(MAMC)、理想体质量百分比(IBW%)营养状态指标以及血清瘦素、三碘甲状腺原氨酸(T3)、四碘甲状腺原氨酸(T4)、促甲状腺素(TSH)水平明显低于对照组和非营养不良组,差异具有统计学意义(P0.05),而非营养不良组和对照组两组患者的各项营养状态指标差异无统计学意义(P0.05)。老年COPD患者血清瘦素与BMI、TSF、MAC、IBW%营养指标以及血清TSH水平呈正相关(P0.05)。结论:营养不良的老年COPD患者的血清瘦素以及甲状腺素水平明显降低,瘦素与COPD患者相关营养状态指标以及TSH呈正相关,与甲状腺素通过下丘脑-垂体-甲状腺轴,共同参与机体能量代谢、体质量以及饮食的调节。  相似文献
