Two silicone coatings have been evaluated for barnacle adhesion. One coating is an unfilled hydrosilation cured polydimethylsiloxane (PDMS) network, while the other is a room temperature vulcanized (RTV), filled, ethoxysiloxane cured PDMS elastomer, RTV11?. The adhesion strength of one species of barnacle, Balanus eburneus, to the hydrosilation coatings is in the range of 0.37–0.60 kg cm‐2 while the corresponding range for RTV11 is 0.64–0.90 kg cm‐2. The easier release of B. eburneus from the hydrosilation cured network compared to RTV11 is discussed in relationship to differences in bulk and surface properties. Preliminary results suggest bulk modulus may be the most important parameter in determining barnacle adhesion strength. In light or mechanical property analysis, a re‐evaluation of surface properties and chemical stability is presented.  相似文献

Changes in the surface properties of some silicone elastomers (General Electric RTV11, RTV160 and RTV655) and fluorosilicones (‐(Si(CH3)((CH2)3‐O(CH2)2(CF2)XCF3)‐O)n‐, x = 5,7,9) on prolonged immersion in water (distilled water, artificial seawater, filtered and unfiltered seawater) have been investigated using measurements of advancing and receding contact angles, of surface roughness and of water uptake. Considerable increases in hydrophilicity and surface rugosity are attributed mainly to surface structural rearrangements accompanying absorption of water; these effects are greater for the fluorosilicones and the rugosity of RTV160 also increased considerably. Observations of early marine settlement (up to 16 weeks in flowing seawater), visualised by scanning electron microscopy, showed patchy attachment mainly of bacteria and microalgae while mature biofilms were formed on the poly(methylmethacrylate) controls. Although settlement was least on RTV11 and greatest on the fluorosilicones, the range of variation was small relative to the differences in the long‐term fouling resistance of the materials. The design requirements for fouling resistance of a smooth, soft, uniform surface are supported.  相似文献

Experiments were conducted to evaluate a controlled internal drug-release (CIDR) dispenser containing progesterone to control the estrous cycle of ewes. After insertion of CIDR dispensers into the vaginae of ovariectomized ewes (Experiment 1; n = 11), the mean plasma progesterone rose from 0.74 ± 0.2 ng/ml to a peak of 5.5 ± 1.0 ng/ml within 2 h and then declined to 3.0 ± 0.5 ng/ml by 48 h. This was followed by a more gradual decline to 1.7 ± 0.3 ng/ml at the time of removal 12 or 14 d later. Following removal, the levels declined to baseline within 4 h. In Experiment 2, a 12- or 14-d treatment with CIDR dispensers was initiated in ewes 2, 9 and 16 d after synchronization of the estrous cycle with fluorogestone acetate (FGA)-impregnated intravaginal sponges. An intramuscular (i.m.) injection of 500 IU pregnant mare serum gonadotropin (PMSG) was given at the time of removal of the FGA sponge or CIDR dispenser. Based on plasma progesterone profiles, CIDR dispensers inserted 9 or 16 d after FGA sponge removal delayed the onset of a new estrous cycle until they were withdrawn. Following withdrawal, ovulation was effectively synchronized in all treatment groups and accompanied by development of functionally active corpora lutea with a normal lifespan. In Experiment 3, comparison of the mating response of ewes after treatment with CIDR dispensers (n = 192) or FGA sponges (n = 194) showed that 92% and 91% of the treated ewes, respectively, were marked by the ram within 72 h. Fertility and litter size of ewes bred at the synchronized and followup estrus were similar for both treatments. These results indicate that treatment of ewes with CIDR dispensers containing progesterone maintains plasma levels of progesterone within the range found during the normal estrous cycle. The CIDR dispenser is effective in synchronizing the estrous cycle of adult ewes and offers a promising alternative to the FGA-impregnated intravaginal sponge.  相似文献

Many strategies have been developed to improve the fouling release (FR) performance of silicone coatings. However, biofilms inevitably build on these surfaces over time. Previous studies have shown that intentional deformation of silicone elastomers can be employed to detach biofouling species. In this study, inspired by the methods used in soft-robotic systems, controlled deformation of silicone elastomers via pneumatic actuation was employed to detach adherent biofilms. Using programmed surface deformation, it was possible to release > 90% of biofilm from surfaces in both laboratory and field environments. A higher substratum strain was required to remove biofilms accumulated in the field environment as compared with laboratory-grown biofilms. Further, the study indicated that substratum modulus influences the strain needed to de-bond biofilms. Surface deformation-based approaches have potential for use in the management of biofouling in a number of technological areas, including in niche applications where pneumatic actuation of surface deformation is feasible.  相似文献

Plasma concentrations of oxytocin and progesterone have been measured by radioimmunoassay in jugular venous blood obtained daily from 5 sheep during 2 estrous cycles and in early pregnancy.Concentrations of oxytocin were relatively high (15–30 pg/ml) during the luteal phase of the cycle, but fell at estrus (to 1–17 pg/ml). A fall in oxytocin was also observed on day 15 of pregnancy, when, as expected, progesterone levels remained high. It is suggested that raised basal levels of oxytocin are unlikely to cause the increasing uterine release of prostaglandin F which occurs at the end of the estrous cycle.  相似文献

The bioavailability of therapeutic agents from eye drops is usually limited due to corneal barrier functions and effective eye protective mechanisms. Therefore, the current study aims to enhance ocular bioavailability of brimonidine, a potent antiglaucoma drug, through the preparation of ocular inserts. Solvent casting technique was employed to prepare the inserts using polyvinylpyrrolidone K-90 (PVP K-90) as film-forming polymer blended with different viscosity grades of bioadhesive polymers namely hydroxypropyl methycellulose, carbopol, sodium alginate, and chitosan. The prepared ocular inserts were evaluated for various physicochemical parameters, swelling behavior, and in vitro release patterns. Sodium alginate-based ocular inserts revealed the most sustainment in drug release (99% at 6 h), so it was selected for further modifications via coating it, on one side or dual sides, using hydrophobic film composed of either ethylcellulose or Eudragit RSPO. The obtained in vitro release results for the modified ocular inserts revealed that ethylcellulose is superior to Eudragit RSPO in terms of brimonidine release sustainment effect. Ocular inserts composed of 7% PVP K-90, 1.5% low molecular weight sodium alginate with or without ethylcellulose coat were able to sustain the in vitro release of brimonidine. Their therapeutic efficacy regarding intraocular pressure (IOP) lowering effect when inserted in albino rabbits eyes showed superior sustainment effect compared with that of brimonidine solution. Furthermore, due to both the mucoadhesive property and the drug sustainment effect, the one-side-coated ocular insert showed more IOP lowering effect compared with that of its non-coated or dual-side-coated counterpart.  相似文献

This study aimed to validate the measurements of body temperature and fecal progesterone concentrations as minimally invasive techniques for assessing ovarian cycle in a single sexually mature female killer whale. Rectal temperature data, fecal and blood samples were collected in the dorsal position using routine husbandry training on a voluntary basis. The correlations between rectal temperature and plasma progesterone concentration and between fecal and plasma progesterone concentrations were investigated. Fecal progesterone metabolites were identified by a combination of high‐performance liquid chromatography and enzyme immunoassay. Plasma progesterone concentrations (range: 0.2–18.6 ng/ml) and rectal temperature (range: 35.3–35.9°C) changed cyclically, and cycle lengths were an average (±SD) of 44.9±4.0 days (nine cycles) and 44.6±5.9 days (nine cycles), respectively. Rectal temperature positively correlated with the plasma progesterone concentrations (r=0.641, P<0.01). There was a visual trend for fecal progesterone profiles to be similar to circulating plasma progesterone profiles. Fecal immunoreactive progestagen analysis resulted in a marked immunoreactive peak of progesterone. The data from the single killer whale indicate that the measurement of rectal temperature is suitable for minimally invasive assessment of the estrous cycle and monitoring the fecal progesterone concentration is useful to assess ovarian luteal activity. Synchrony of estrous, and consequently of conception and birth of young, may be of adaptive significance for certain mammals. Among the species in which estrous synchrony has been suspected several times are elephants, but clear evidence is still missing. We determined estrous cycles of African elephants (Loxodonta africana) (n=4) at the Vienna Zoo, Austria, between June 2003 and January 2006 by measuring serum progesterone levels from weekly blood samples. Except for the dominant female when she was intensively lactating, all animals showed clear cycles or progesterone release with a mean period of 105.3+/-15.37 days. For most of the study period, estrous cycles were asynchronous between females. However, after re-occurrence of the progesterone cycle in the dominant female following the first period of lactation, all four females showed high synchrony of progesterone release over the two subsequent cycles. Large changes in individual period lengths indicated that synchronization was due to the adjustment of cycle length in subdominants to that of the dominant female. We used a bootstrap procedure, based on resampling measured times of progesterone peaks, to determine if this apparent synchrony could have been caused by chance alone. This statistical analysis indicated that between-individual variances of the timing of progesterone peaks were much smaller that to be expected by chance (P=0.009). This finding represents the first evidence for estrous synchrony between elephants. We discuss various hypotheses to explain the biological function of cycle synchrony in elephants.  相似文献

The aim of this study was to determine if the duration of treatment with an intravaginal progesterone releasing insert (IVP(4)) after treatment with oestradiol benzoate (ODB) at the time of insertion and 24 h after removal would affect selected variables including: size of ovarian follicles at the time of removal of inserts, diameter of ovulatory follicles, plasma concentrations of progesterone following ovulation, and duration of the following oestrous cycle. Characteristics of oestrus at a synchronised and spontaneous oestrus were also monitored. Non-lactating Holstein cows were synchronised with an IVP(4) for 5 (n = 10), 7 (n = 10), 8 (n = 9) or 9 (n = 9) days together with injections of ODB at device insertion (2 mg) and 24 h after removal (1 mg). Ultrasonography showed no significant effect of treatment on the day of emergence of preovulatory follicles relative to the day of removal of inserts (overall mean = -4.22 +/- 0.58; P = 0.15) for cows that ovulated within 120 h insert removal (n = 36). Treatment with ODB and an IVP(4) for 5 days reduced the diameter of preovulatory follicles at the time of removal of inserts and for the following 2 days compared to cows treated for 7-9 days (mean difference 2.56 +/- 1.15 mm; P = 0.033) but did not reduce the diameter of the ovulatory follicle (P = 0.21). Day of emergence relative to removal of inserts was associated with the diameter of the ovulatory follicle (R2 = 0.69; P < 0.001). Concentrations of progesterone and the diameter of the corpus luteum following ovulation were not affected by treatment (P > 0.20), but were affected by the diameter of the ovulatory follicle (P < 0.01). Diameter of the ovulatory follicle did not affect interoestrous and interovulatory intervals (P > 0.40). We conclude that treatment with an IVP(4) for 5 compared to 7-9 days with ODB administered at device insertion, and 24 h after removal reduced the diameter of preovulatory follicles at the time of removal of the insert but did not reduce the diameter of the ovulatory follicle or concentrations of progesterone in plasma. Emergence of preovulatory follicles closer to the time of removal of inserts reduced the diameter of the ovulatory follicle when oestrus was induced with ODB. Ovulation of smaller follicles reduced concentrations of progesterone in plasma following ovulation but did not affect oestrous cycle duration.  相似文献

Significantly higher numbers of zoospores of the fouling, green alga Enteromorpha adhered to silicone elastomers cured by dibutyltin dilaurate (DBTDL) than to identical polymers cured by dibutyltin diacetate (DBTDA). Enhanced zoospore adhesion was shown to be due to the presence of DBTDL and the effect was concentration‐dependent. Further experiments revealed that enhanced zoospore adhesion also occurred to glass coverslips coated with lauric acid (C12) and other saturated fatty acids. The possibility that fatty acids may act as chemical cues (chemoattractants), guiding motile zoospores to the substratum for settlement in the natural environment is discussed. Settlement of other fouling organisms to DBTDL‐cured silicone elastomers is currently being investigated.  相似文献

We evaluated the ability of microemulsions containing medium-chain glycerides as penetration enhancers to increase the transdermal delivery of lipophilic (progesterone) and hydrophilic (adenosine) model drugs as well as the effects of an increase in surfactant blend concentration on drug transdermal delivery. Microemulsions composed of polysorbate 80, medium-chain glycerides, and propylene glycol (1:1:1, w/w/w) as surfactant blend, myvacet oil as the oily phase, and water were developed. Two microemulsions containing different concentrations of surfactant blend but similar water/oil ratios were chosen; ME-lo contained a smaller concentration of surfactant than ME-hi (47:20:33 and 63:14:23 surfactant/oil/water, w/w/w). Although in vitro progesterone and adenosine release from ME-lo and ME-hi was similar, their transdermal delivery was differently affected. ME-lo significantly increased the flux of progesterone and adenosine delivered across porcine ear skin (4-fold or higher, p < 0.05) compared to progesterone solution in oil (0.05 ± 0.01 μg/cm2/h) or adenosine in water (no drug was detected in the receptor phase). The transdermal flux of adenosine, but not of progesterone, was further increased (2-fold) by ME-hi, suggesting that increases in surfactant concentration represent an interesting strategy to enhance transdermal delivery of hydrophilic, but not of lipophilic, compounds. The relative safety of the microemulsions was assessed in cultured fibroblasts. The cytotoxicity of ME-lo and ME-hi was significantly smaller than sodium lauryl sulfate (considered moderate-to-severe irritant) at same concentrations (up to 50 μg/mL), but similar to propylene glycol (regarded as safe), suggesting the safety of these formulations.  相似文献

This study compared the shear adhesion strength of barnacles, oysters and tubeworms on eight RTV 11‐based silicone fouling‐release coatings containing different silicone oil additives. It was found that adhesion strength differed among species and coating types. In most cases, oysters and tubeworms had higher adhesion strengths than barnacles. Barnacle adhesion strength was reduced on all coatings containing oil additives; however, this was not generally true for oysters and tubeworms. The difference in the adhesion strength among the three organisms tested in this study emphasizes the importance of understanding the fundamental interaction between marine invertebrate adhesives and the substratum.  相似文献

A series of experiments were conducted to evaluate the effects of mode and frequency of administration and estrous cycle stage on the response of the cycling ewe to PGF. The effects of dexamethasone, arachadonic acid and prostaglandin synthetase inhibitors on estrous cycle length and plasma progesterone levels were also determined.Intramuscular administration of 5 or 10 mg of PGF, on days 8 and 9 after estrus (5 ewes/group), significantly (p<.01) shortened the mean length of the estrous cycle and the interval from the end of treatment to estrus. Mean plasma progesterone levels, 24 hours after initial injection, were significantly (p<.01) lowered. When administered on day 8 only, these doses were considerably less effective in shortening estrous cycle length or lowering plasma progesterone levels. Intravaginal administration of PGF, by polyurethane tampon, was also largely ineffective.Treatment of ewes with 10 mg of PGF i.m., on days 3 and 4 of the estrous cycle, resulted in a return to estrus in 2 days in 25% of the treated animals. Plasma progesterone levels of PGF-treated ewes were significantly lower than controls on the second, third and fourth days after the start of dosing. It would appear that PGF exerts a retarding effect on developing CL functionality.The prostaglandin synthetase inhibitors, aspirin, flufenamic acid and 1-p-chlorobenzylidene-2-methyl-5-methoxy-3-indenylacetic acid, were administered orally or parenterally for 16 days beginning on day 8 of the estrous cycle. These compounds failed to prolong estrous cycle length. Parenteral administration of dexamethasone did not result in PGF release in the cycling ewe, at least not in quantities sufficient to induce luteolysis. The prostaglandin precursor, arachadonic acid, also was not luteolytic when given parenterally to cycling ewes.  相似文献

The objective of the present study was to determine whether treatment of postpartum multiparous and primiparous anestrous beef cows with an intravaginal progesterone-releasing insert (CIDR) and PGF(2alpha), with and without the addition of GnRH or estradiol cypionate (ECP) at the time of CIDR insertion, is effective in stimulating onset of estrous cycles. Postpartum lactating Angus primiparous (n=47, 2 years of age, 495+/-6 kg) and multiparous (n=76, >or=3 years of age, 553+/-9 kg) cows were assigned by calving date to four blocks spaced 21-day apart. Cows were assigned sequentially by calving date to four treatment groups: (1) PGF(2alpha) (n=30), (2) CIDR-PGF(2alpha) (n=30), (3) GnRH-CIDR-PGF(2alpha) (n=33), and (4) ECP-CIDR-PGF(2alpha) (n=27). Intravaginal CIDR inserts were in place from days -7 to 0. A single 100 microg injection of GnRH or 2 mg ECP were administered on day -7, and 25mg PGF(2alpha) was administered on day 0. Day 0 averaged 38+/-1 day postpartum. Blood samples were collected on days -19, -9, 0, 5, 9, 12, 16, 19, 23, 26, and 30 for determination of plasma progesterone concentrations. Pre-treatment luteal activity (progesterone>or=1 ng/ml) was detected in 19% of primiparous and 8% of multiparous cows. Progesterone concentrations on day 0 were greater (P<0.001) in primiparous (3.2+/-0.3 ng/ml) than multiparous (2.0+/-0.2 ng/ml) cows. Following CIDR withdrawal, progesterone concentrations from days 5 to 30 were used to categorize response profiles as either: (1) treatment-induced onset of estrous cycles, (2) continued anestrus, or (3) spontaneous ovulation and subsequent formation of a CL. Incidence of treatment-induced onset of estrous cycles, which was defined as progesterone concentrations >or=1 ng/ml in three or more consecutive samples from days 9 to 19, was influenced by treatment and parity. Percentages of cows initiating estrous cycles were greater (P<0.001) in the three CIDR-treated groups than in the PGF(2alpha) group (55 and 8%, respectively). Percentages of cows initiating estrous cycles in the CIDR-PGF(2alpha), GnRH-CIDR-PGF(2alpha), and ECP-CIDR-PGF(2alpha) groups were 55, 58, and 52%, respectively. Incidence of treatment-induced estrous cycles in the three CIDR-treated groups of cows was greater (P=0.008) in primiparous (76%) than multiparous (43%) cows. Treatment of postpartum anestrous primiparous and multiparous beef cows with CIDR-PGF(2alpha) approximately 40-day postpartum provides an approach to increase the percentage of cows that have reinitiated estrous cycles by the start of the breeding season.  相似文献

Exogenous PGF failed to consistently alter estrous cycle length of the guinea pig. A wide range of dose levels were administered with varying frequency, at different stages of the estrous cycle, in different vehicles and by various routes. Massive doses of PGF (5.0–10.0 mg) produced a significant (p<.05), although transient, lowering of plasma progesterone levels. Smaller doses were ineffectual. An i.p. injection of 25 mg of PGF was toxic in four of five treated animals. It would appear that the intact guinea pig is extremely resistant to the luteolytic effects of parenterally administered PGF.Estradiol-17β, administered s.c. on days 3 to 10 of the estrous cycle, significantly (p<.05) reduced corpus luteum diameter and plasma progesterone levels. Estrous cycle length was unaffected. Clomiphene, in the same experiment, caused premature vaginal opening in some treated animals, but corpus luteum size and plasma progesterone levels were unaffected and no ovulations occurred.The prolactin secretion inhibitor, CB-154, administered early in the estrous cycle, did not have any effect on estrous cycle length of the guinea pig alone or in combination with PGF. The prostaglandin precursor, arachidonic acid, also failed to influence estrous cycle length when administered on days 8 and 9 of the cycle. Plasma progesterone levels remained unaltered.Oral administration of a prostaglandin synthetase inhibitor, (MK-715), caused a small, but non-significant (p>.05) prolongation of the estrous cycle. The progesterone biosynthesis inhibitors, aminoglutethimide and 6β-hydroxy-3α, 5α-cyclo-androstane-17-one did not effect estrous cycle length or plasma progesterone levels of the guinea pig.  相似文献

A study was conducted on 34 Surti buffalo cows to determine the feasibility of synchronizing oestrus using prostaglandin F and a 12-day progesterone intravaginal device. Eighteen cycling buffalo cows having palpable corpora lutea were treated with a single intramuscular injection of 30 mg of prostaglandin F. Three cows exhibited oestrus approximately 54 h after treatment and two of these were diagnosed pregnant 90 days after natural breeding. Sixteen randomly selected post partum cows were treated for 12 days with a progesterone intravaginal device. Ten mg of oestradiol 17β in 5 ml of ether was also injected at the time of insertion of the device. Thirteen cows retained the device for 12 days and 10 of them returned to oestrus 4–5 days after its removal. Eight animals were diagnosed pregnant 90 days after natural breeding. The results indicate that short term progesterone intravaginal device treatment is more reliable than prostaglandin for synchronizing oestrus in buffaloes.  相似文献

This study compared the effect of double and single ovulation on serum progesterone concentrations and luteal characteristics in Sanjabi ewes at different days of the estrous cycle. The estrous cycles of 197 Sanjabi ewes were synchronized by a 12-day treatment with intravaginal sponges (Chronogest®). Estrus was detected in 144 ewes 27–39 h after sponge removal. Daily blood samples were taken every morning and analyzed for serum progesterone (P4). Ewes were then transported to a local abattoir, where nine ewes were slaughtered on each experimental day (days 1–16 after estrus) for ovary collection. The ovarian follicles were measured and categorized by size (very small <2 mm; small 2–3.5 mm; medium 3.5–5 mm; large >5 mm). On each slaughter day, the number of corpora lutea per ewe was classified as single and double ovulation. The results show that the effect of dominant follicles was less during the mid-luteal phase. Ovulation rate of right, left and both ovaries were (54.9%), (23.6%) and (21.5%), respectively. The incidence of double ovulations was 40.2%. In the case of ewes exhibiting double ovulation, 46.6% occurred unilateral (ewes exhibited both ovulations on the right ovary); whereas 53.4% occurred bilateral (ewes exhibited ovulations on the right and left ovaries). Unilateral double ovulation was not observed in the left ovary. The right ovary appeared to play a significantly greater role in ewes showing single and double ovulations than the left ovary (P < 0.05). Serum progesterone concentration showed minimum and maximum levels of 0.29 ± 0.15 and 5.51 ± 0.75 ng/ml on days 16 and 11 post-estrous, respectively (P < 0.001). The mean volume of individual corpus lutea in ewes with single ovulations was significantly higher than in ewes with double ovulations (P < 0.01). However, the total volume of corpus lutea in ewes with single ovulation was significantly lower than in ewes with double ovulations in some days of estrous cycle (P < 0.01). The serum progesterone concentration was significantly higher in double than single ovulating animals on days 1–16 of the estrous cycle (P < 0.001). These results indicated a relatively high incidence of double ovulation in ewes associated with increasing total luteal volume and high circulating concentrations of progesterone.  相似文献

Patterns of luteal lipid and arachidonic acid accumulation were examined in relation to luteal progesterone and prostaglandin F synthesis in 30 sows and gilts between days 8 and 18 of the estrous cycle. Net in vitro release of progesterone from luteal tissue declined from 722 ng/100 mg tissue at day 8 to 81 ng/100 mg tissue at day 18. Although statistical significance was not present, net prostaglandin F release increased slightly from 8.6 to 13.9 ng/100 mg tissue. Luteal free cholesterol, esterified cholesterol, and free fatty acid contents did not change between days 8 and 18 whereas triglycerides accumulated rapidly between days 14 and 18 of the estrous cycle. Phospholipids increased between days 8 and 12, plateaued at 20.2 mg/g between days 14 and 16, and decreased to 15.4 mg/g on day 18. Between days 12 and 18, arachidonic acid increased from 19.4 to 34.8% in cholesterol esters, from 10.1 to 22.5% in triglycerides, and from 12.3 to 27.2% in luteal free fatty acids. Arachidonic acid in luteal phospholipids increased from 21.3 to 25.1% between days 14 and 16 of the estrous cycle. Luteal regression was associated with conservation of arachidonic acid. Based on blood plasma lipid fatty acid compositions, the corpus luteum elongated and desaturated essential fatty acids. Within porcine corpora lutea, calculated free arachidonic acid content was adequate for maintenance of prostaglandin synthesis.  相似文献

A study was conducted to determine the effect of suprabasal plasma concentrations of progesterone on the release of prostaglandin F (PGF) at luteolysis and oestrus. Heifers received silicone implants containing 2.5 (n = 4), 5 (n = 4), 6 (n = 3), 7.5 (n = 3), 10 (n = 4), or 15 (n = 3) g of progesterone, or an empty implant (controls, n = 4) between Days 8 and 25 post ovulation. Blood was collected frequently between Days 14 and 28 and assayed for progesterone and 15-ketodihydroprostaglandin F. Basal progesterone concentrations in control heifers did not differ from those in heifers with 2.5- or 5-g implants and remained around 0.4−0.5 nmol l−1 until ovulation in all three groups. In the heifers treated with 6–15 g of progesterone, basal concentrations were maintained at higher (P < 0.05) levels compared with those in the controls, ranging from 0.8 to 1.6 nmol 1−1. The effect of these elevated progesterone levels was to delay ovulation by prolonging the growth of the ovulatory follicle, which continued growing until the implant was removed. In all experimental groups, the first significant increase of the PGF metabolite occurred between Days 15.3 and 16.3 (P > 0.05) and was associated with the onset of a decrease in progesterone concentrations, which had reached levels below 3 nmol 1−1 by Days 17.4−19.1. PGF metabolite peaks associated with luteolysis were frequent until Day 20. In the period from Day 20 until implant removal, sporadic peaks were observed, ranging in number from 1.0 ± 1.2 (mean ± SEM) in the control group to 3.0 ± 1.4 peaks in the heifers treated with 7.5 g of progesterone (P > 0.05). The number of PGF metabolite peaks during that period was higher (P < 0.05) in heifers treated with 10 and 15 g than in controls. A positive correlation was found between the basal concentration of progesterone and the number of PGF peaks after luteolysis (r = 0.54; P < 0.01). Plasma progesterone concentrations above approximately 1.4 nmol l−1 were able to maintain the release of PGF until the progesterone implants were removed and plasma levels decreased to basal values. These heifers had a preovulatory PGF release pattern resembling that found in repeat breeder heifers.  相似文献

The objectives of this experiment were to determine if subnormal levels of progesterone (P4) indicative of luteal insufficiency influence (1) pulsatile release of luteinizing hormone (LH), (2) the interval to the preovulatory surge of LH after removal of P4, and (3) the secretion of P4 during the estrous cycle subsequent to administration of subnormal levels of P4. On Day 5 (Day = 0 day of estrus) of the estrous cycle, cows received P4-releasing intravaginal devices (PRID) to produce normal (2 PRIDs; n = 7) or subnormal (0.5 PRID; n = 6) concentrations of P4. Five cows served as controls. On Day 10, serial blood samples were collected from all cows. Collection of blood samples was again initiated on Day 17 in cows receiving PRIDs. The PRIDs were removed and blood collection continued for 78 h. Daily blood samples were collected from all animals for 42 days subsequent to estrus (estrous cycles 1 and 2, respectively). During estrous cycle 1, mean concentration of P4 was lower (p less than 0.05) and frequency of pulses of LH was higher (p less than 0.05) in cows receiving subnormal P4 than in cows receiving normal P4 and control cows. Plasma concentrations of estradiol (E2) were higher (p less than 0.05) on Days 9-16 of estrous cycle 1 in cows receiving subnormal P4 than in cows receiving normal P4 or in control cows. Concentrations of E2 were greater (p less than 0.05) at 6, 18, and 30 h following removal of PRIDs in cows receiving subnormal P4 than in cows receiving normal P4.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
