The effects of high salinity (up to 400 m M NaCl) on photosystem II (PSII) photochemistry, photoinhibition and the xanthophyll cycle were investigated in the halophyte Artimisia anethifolia grown under outdoor conditions. In order to examine the changes in PSII photochemistry, photoinhibition, thermal dissipation associated with the xanthophyll cycle in salt-acclimated plants, the experiments were conducted at midday on a clear day (maximal irradiance 1500 μmol m−1 s−1) and on a cloudy day (maximal irradiance 700 μmol m−1 s−1), respectively. With increasing salt concentration, the accumulation of sodium and chloride in leaves increased considerably while the relative growth rate and CO2 assimilation rate decreased significantly. Salinity induced no effects on PSII photochemistry, thermal energy dissipation, and the contents of the xanthophyll cycle pigments either on a clear day or on a cloudy day. However, when compared with those on a cloudy day, PSII photochemistry decreased and thermal energy dissipation increased significantly in both control and salt-acclimated plants on a clear day. The levels of zeaxanthin and antheraxanthin at the expense of violaxanthin were higher on a clear day than on a cloudy day. The results suggest that photoinhibition and the xanthophyll cycle were not induced by high salinity but by high light only in A. anethifolia plants. The results also suggest that A. anethifolia showed high resistance not only to high salinity, but also to photoinhibition even when it was treated with high salinity and exposed to full sunlight.  相似文献

Previous results in our laboratory indicated that a reduced Mn concentration in the leaves of barley was highly correlated with the reduced relative growth and net assimilation rates of salt-stressed plants. If Mn deficiency limits the growth of salt-stressed barley, then increasing leaf Mn concentrations should increase growth. In the present study, the effect of supplemental Mn on the growth of salt-stressed barley ( Hordeum vulgare L. cv. CM 72) was tested to determine if a salinity-induced Mn deficiency was limiting growth. Plants were salinized with 125 mol m−3 NaCl and 9.6 mol m−3 CaCl2. Supplemental Mn was applied in 2 ways: 1) by increasing the Mn concentration in the solution culture and 2) by spraying Mn solutions directly onto the leaves. Growth was markedly inhibited at this salinity level. Dry matter production was increased 100% in salt-stressed plants treated with supplemental Mn to about 32% of the level of nonsalinized controls. The optimum solution culture concentration was 2.0 mmol m−3, and the optimum concentration applied to the leaves was 5.0 mol m−3. Supplemental Mn did not affect the growth of control plants. Further experiments showed that supplemental Mn increased Mn concentrations and uptake to the shoot. Supplemental Mn increased the relative growth rate of salt-stressed plants and this increase was attributed to an increase in the net assimilation rate; there were no significant effects on the leaf area ratio. Supplemental Mn also increased the net photosynthetic rate of salt-stressed plants. The data support the hypothesis that salinity induced a Mn deficiency in the shoot, which partially reduced photosynthetic rates and growth.  相似文献

The effects of high salinity (0-400 mmol/L NaCl) on photosystem II (PSII) photochemistry and photosynthetic pigment composition were investigated in the halophyte Artimisia anethifolia grown under outdoor conditions and exposed to full sunlight. High salinity resulted in an inhibition in plant growth and a significant accumulation of sodium and chloride in leaves. However, high salinity induced no effects on the actual PSII efficiency, the efficiency of excitation energy capture by open PSII reaction centres, photochemical quenching, and non-photochemical quenching at midday. High salinity also induced neither changes in the maximum efficiency of PSII photochemistry, the efficiency with which a trapped exciton can move an electron into the electron transport chain further than QA and the quantum yield of electron transport beyond QA, nor changes in absorption, trapping and electron transport fluxes per PSII reaction centre. No significant changes were observed in the levels of neoxanthin, lutein, beta-carotene, violaxanthin, antheraxanthin, and zeaxanthin expressed on a total chlorophyll basis in salt-adapted plants. Our results suggest that Artimisia anethifolia showed high resistance not only to high salinity, but also to photoinhibition even if it was treated with high salinity as high as 400 mmol/L NaCl and exposed to full sunlight. The results indicate that tolerance of PSII to high salinity and photoinhibition can be viewed as an important strategy for Artimisia anethifolia, a halophyte plant, to grow in very high saline soil.  相似文献

By analysis of gas exchange and chlorophyll fluorescence, the effects of NaCl treatment and supplemental CaCl2 on photosynthesis, photosystem II (PSII) photochemistry and photoinhibition were investigated in Rumex leaves. Photosynthesis in Rumex leaves was strongly inhibited by 200 m M NaCl treatment. Such inhibition of photosynthesis was ameliorated by CaCl2 supplement. Neither NaCl treatment nor CaCl2 supplement had any significant effects on the PSII primary photochemical reaction in dark-adapted leaves. In light-adapted leaves, however, 200 m M NaCl treatment significantly decreased photochemical quenching (qp), efficiency of excitation energy capture by open PSII reaction centers (FV'/FM') and quantum yield of PSII electron transport (ΦPSII). These decreases in qp, FV'/FM' and ΦPSII were mitigated by CaCl2 supplement with the maximum of its effect appearing at a concentration of 8 m M CaCl2. A similar mitigating effect was shown in 200 m M NaCl-treated Rumex leaves when susceptibility of PSII to photoinhibition was determined under high irradiance. It is suggested that the mitigation of photoinhibition in NaCl-treated leaves is because of the amelioration of inhibition of photosynthesis.  相似文献

The effects of manganese (Mn) toxicity on photosynthesis in white birch ( Betula platyphylla var. japonica ) leaves were examined by the measurement of gas exchange and chlorophyll fluorescence in hydroponically cultured plants. The net photosynthetic rate at saturating light and ambient CO2 (Ca) of 35 Pa decreased with increasing leaf Mn concentrations. The carboxylation efficiency, derived from the difference in CO2 assimilation rate at intercellular CO2 pressures attained at Ca of 13 Pa and O Pa, decreased with greater leaf Mn accumulation. Net photosynthetic rate at saturating light and saturating CO2 (5%) also declined with leaf Mn accumulation while the maximum quantum yield of O2 evolution at saturating CO2 was not affected. The maximum efficiency of PSII photochemistry (Fv/Fm) was little affected by Mn accumulation in white birch leaves over a wide range of leaf Mn concentrations (2–17 mg g−1 dry weight). When measured in the steady state of photosynthesis under ambient air at 430 μmol quanta m−2 s−1, the levels of photochemical quenching (qP) and the excitation capture efficiency of open PSII (F'v/F'm) declined with Mn accumulation in leaves. The present results suggest that excess Mn in leaves affects the activities of the CO2 reduction cycle rather than the potential efficiency of photochemistry, leading to increases in QA reduction state and thermal energy dissipation, and a decrease in quantum yield of PSII in the steady state.  相似文献

通过林地穿透雨排除的方法模拟降雨减少,测定河南宝天曼自然保护区锐齿栎叶片光合色素含量与反射光谱的季节变化,对减雨处理造成的光合色素变化及其反射光谱的变化进行了定量分析,并探讨了水分控制条件下反射光谱对叶片光合色素变化的响应机制.结果表明: 锐齿栎叶片的光合色素含量和色素比率均呈现明显的季节变化.减雨样地与对照样地叶片的光合色素含量和比率在生长季的各个时期存在差异,其中,叶片叶绿素b(Chl b)含量的差异显著,说明Chl b对减雨处理的敏感性最高,叶片类胡萝卜素(Car)含量的差异较小,说明Car对减雨处理的敏感性相对较弱.550 nm处的光谱反射率对色素季节变化的响应最敏感,以其构造的简单比值指数(SR750,550)与叶片Chl a、Chl b、总Chl和Car含量的正相关关系显著,光化学反射指数(PRI)与叶片Car/Chl的负相关关系显著.550 nm处的光谱反射率对减雨处理造成的色素变化响应最为敏感.SR750,550对减雨处理造成的叶片Chl a、Chl b和总Chl的含量变化表现敏感(P<0.01),对Chl a/b的变化不敏感.PRI对减雨处理造成的叶片Car/Chl变化表现敏感(P<0.01).  相似文献

The effect of four different NaCl concentrations (from 0 to 102 mM NaCl) on seedlings leaves of two corn (Zea mays L.) varieties (Aristo and Arper) was investigated through chlorophyll (Chl) a fluorescence parameters, photosynthesis, stomatal conductance, photosynthetic pigments concentration, tissue hydration and ionic accumulation. Salinity treatments showed a decrease in maximal efficiency of PSII photochemistry (Fv/Fm) in dark-adapted leaves. Moreover, the actual PSII efficiency (ϕPSII), photochemical quenching coefficient (qp), proportion of PSII centers effectively reoxidized, and the fraction of light used in PSII photochemistry (%P) were also dropped with increasing salinity in light-adapted leaves. Reductions in these parameters were greater in Aristo than in Arper. The tissue hydration decreased in salt-treated leaves as did the photosynthesis, stomatal conductance (g s) and photosynthetic pigments concentration essentially at 68 and 102 mM NaCl. In both varieties the reduction of photosynthesis was mainly due to stomatal closure and partially to PSII photoinhibition. The differences between the two varieties indicate that Aristo was more susceptible to salt-stress damage than Arper which revealed a moderate regulation of the leaf ionic accumulation.  相似文献

Chilling in the light imposes a considerable level of stress on the photosynthetic apparatus, resulting in a decrease of photosystem II activity and the quenching of maximum and variable fluorescence. We selected in a fah - 1 mutagenized population of Arabidopsis thaliana , which permits a direct visible evaluation of the intensity of chlorophyll (Chl) fluorescence, a monogenic recessive nuclear mutant hypersensitive to photoinhibition induced by light and cold. The major phenotypic trait of the mutant is the appearance of chlorotic areas on developed leaves. Photochemical analyses indicate that the mutant is hypersensitive to photoinhibition in excess light in the cold but also at room temperature. The susceptibility to photoinhibition is a consequence of perturbations in photochemistry already present in unstressed plants. Such perturbations result in a greater fraction of the primary acceptor QA remaining in the reduced state even at low light fluxes. From estimates of the number of total and functional PSII units and measurements of PSII quantum yield and QA reoxidation kinetics, the basic lesion of the mutant seems restricted to PSII photochemistry likely affecting the rate of electron transport from QA to QB.  相似文献

Some processes of excess radiation dissipation have been associated with changes in leaf reflectance near 531 nm. We aimed to study the relations between the photochemical reflectance index (PRI) derived from this signal, and photosynthetic radiation-use efficiency (defined as net CO2 assimilation rate/incident photon flux density) in a cereal canopy. Measurements of reflectance, fluorescence, gas exchange and xanthophyll cycle pigments were made in the morning, midday and afternoon in barley canopies with two levels of nitrogen fertilization. The photosynthetic radiation-use efficiency decreased at midday, mainly in the third leaf, in both treatments, with lower values for the nitrogen deficient leaves. The zeaxanthin content showed the inverse pattern, increasing at midday and in the nitrogen deficient treatment. The photosynthetic radiation-use efficiency was well correlated with the epoxidation state, EPS (violaxanthin + 0.5 antheraxanthin)/(violaxanthin + antheraxanthin + zeaxanthin). The PRI [here defined as (R539 - R570)/(R539+ R570)] was significantly correlated with epoxidation state and zeaxanthin and with photosynthetic radiation-use efficiency. These results validate the utility of PRI in the assessment of radiation-use efficiency at canopy level.  相似文献

Abstract. Plantago maritima L. was grown at three levels of salinity, 50, 200, 350 mol m−3 NaCl, and the effects on growth, ion content and photosynthetic capacity were studied. Shoot and root dry weight, leaf production and leaf length were all substantially reduced in plants grown at high salinity. Total leaf area of plants grown at 350 mol m−3 NaCl was only 20% of that in plants at low salinity. Both the Na+ and K+ content of leaves and roots increased with external salinity. There was no change in the Na+/K+ ratio of leaves or roots at different salinity levels. Despite the large reductions in growth and high accumulation of Na+ ions, leaf photosynthetic rate was only slightly reduced by salinity stress. The reduction in photosynthesis was not caused by reduced biochemical capacity as judged by photosynthetic response to intercellular CO2 and by ribulose-1,5-bisphosphate carboxylase activity, but was due to reduced leaf conductance and low intercellular CO2 concentration. The increased stomatal limitation of photosynthesis resulted in higher water-use efficiency of plants grown at high salinity.  相似文献

The effect of Cu toxicity on photosynthetic function, chlorophyll and Ca2+ content of Cu-tolerant Silene compacta plants grown in nutrient solution was studied. Since, in plants grown under 8 μ M Cu, the chlorophyll and Ca2+ concentration as well as the photosystem II (PSII) photochemistry were increased, compared to the control, the development of an adaptive mechanism of the Cu-tolerant ecotype of S. compacta to 8 μ M Cu is suggested. Increased Cu tolerance of the S. compacta ecotype reflects modulation of the photosynthetic apparatus to optimize photosynthesis. However, exposure of plants to 160 μ M Cu resulted in a marked increase of the fraction of closed PSII centres and decreased quantum yield of PSII electron transport (ΦPSU) which was accompanied by a significant decline of relative quantum yield for O2 evolution (Aox/Apt). The concentration of chlorophyll and Ca2+ in leaves also decreased significantly under 160 μ M Cu treatment. Photochemical quenching (qp) displayed a reduction as a result of perturbation of the photosynthetic electron transfer chain, while non-photochemical quenching (qN) increased. High Cu treatment reduced photosynthetic productivity of S. compacta plants which can be attributed, in part, to pertubation of photosynthetic process and photosynthetic pigments as well as to Ca2+ loss.  相似文献

Abstract. Three parameters influencing the capacity for carbon accumulation, i.e. photosynthesis, respiration, and leaf extension growth, were studied in Beta vulgaris L. (sugar beet) cultured in nutrient solution containing 0.5 to 500 mol m−3 NaCl. Leaf extension growth was the parameter most sensitive to salinity: the initial rate of leaf extension and final leaf length each declined linearly with increase in external NaCl concentration. Photosynthetic O2 evolution of thin leaf slices did not decline until salinity levels reached 350 to 500 mol m−3 NaCl, while respiratory O2 consumption was not affected by salinity throughout the range. The results suggest that the influence of salinity on the capacity for carbon accumulation in B. vulgaris occurs primarily through reduction in the area of photosynthetic surface.  相似文献

The effect of exogenous application of the cytokinin meta -topolin [mT; N6-( meta -hydroxybenzyl)adenine] on artificial senescence of detached wheat leaves ( Triticum aestivum L. cv. Hereward) was studied and compared in leaves senescing under continuous light (100 µmol photons m−2 s−1) and darkness. Senescence-induced deterioration in structure and function of the photosynthetic apparatus was characterized by reduction in chlorophyll content, maximal efficiency of photosystem (PS) II photochemistry ( F v/ F m) and the rate of CO2 assimilation, by increase in the excitation pressure on PSII (1 −  q P) and a level of lipid peroxidation and by modifications in chloroplast ultrastructure. While in darkened leaf segments mT effectively slowed senescence-induced changes in all measured parameters, in light-senescing segments the effect of mT changed into opposite a few days after detachment. We observed an overexcitation of photosynthetic apparatus, as indicated by pronounced increases in the excitation pressure on PSII and in a deepoxidation state of xanthophyll cycle pigments, marked starch grain accumulation in chloroplasts and stimulation of lipid peroxidation in light-senescing leaf segments in mT. Possible mechanisms of acceleration of senescence-accompanying decrease in photosynthetic function and increase in lipid peroxidation during mT influence are discussed. We propose that protective mT action in darkness becomes damaging during artificial senescence in continuous light due to overexcitation of photosynthetic apparatus resulting in oxidative damage.  相似文献

The changes in PSII photochemistry in Spirulina platensis cells exposed to salinity stress (0–0.8 M NaCl) for 12 h were studied. Salinity stress induced a decrease in oxygen evolution activity, which correlated with the decrease in the quantum yield of PSII electron transport ( Φ PSII). Phycocyanin content decreased significantly while chlorophyll content remained unchanged in salt-stressed cells. Salinity stress induced an increase in non-photochemical quenching (qN) and a decrease in photochemical quenching (qP). Analyses of the polyphasic fluorescence transients (OJIP) showed that with the increase in salt concentration, the fluorescence yield at the phases J, I and P declined sharply and the transient almost levelled off at salt concentration of 0.8 M NaCl. The effects of DCMU on the polyphasic rise of fluorescence transients decreased significantly. Salinity stress resulted in a decrease in the efficiency of electron transfer from QA to QB. The slope at the origin of the relative variable fluorescence curves (dV/dto) and the relative variable fluorescence at phase J (VJ) increased in the absence of DCMU, but decreased in the presence of DCMU. The shape of the relative variable fluorescence transients in salt-stressed cells was comparable to that of the control cells incubated with DCMU. The results in this study suggest that salt stress inhibited the electron transport at both donor and acceptor sides of PSII, resulted in damage to phycobilisome and shifted the distribution of excitation energy in favour of PSI.  相似文献

The characteristics of pigment composition and photosystem II (PSII) photochemistry in the flag leaf and ear parts of wheat (Triticum aestivum L.) grown in the field was compared. At the early stage of flowering, awns and the flag leaf showed the highest values in the maximal efficiency of PSII photochemistry (Fv/Fm), actual PSII efficiency (phi(PSII)), photochemical quenching (qP), and the efficiency of excitation capture by open PSII centres (Fv/F'm), followed by glumes, lemmas, and paleae, respectively except that no differences in F'v/F'm were observed among glumes, leamms, and paleae. With progressing grain filling, there was a change in the photosynthetic pigment stoichiometry. In the ear parts, neoxanthin and antheraxanthin decreased equally with chlorophyll levels. Lutein and zeaxanthin decreased less than chlorophyll levels while beta-carotene and violaxanthin decreased faster than chlorophyll levels. No big differences in pigment composition were observed among different ear parts. For the flag leaf, neoxanthin and beta-carotene decreased concomitantly with chlorophyll, whereas lutein and xanthophyll cycle pigment were less affected, leading to increases in lutein/chlorophyll and xanthophyll cycle pigment/chlorophyll ratios. Fv/Fm, phi(PSII), qP, and F'v/F'm decreased gradually in the flag leaf and ear parts but to different extents. The largest changes were observed in awns, followed by the lemmas of floret 2, the lemmas of floret 1, glumes, and the flag leaf, respectively. The results suggest that during grain filling, a down-regulation of PSII associated with an increase of the de-epoxidation state of the xanthophyll cycle carotenoids occurred in the flag leaf but not in the ear parts.  相似文献

A study was conducted, using rapid time course of chlorophyll (Chl) fluorescence parameters, and light-response curves of Chl fluorescence parameters, to determine the induction requirements and response of photosystem II (PSII) photochemistry and non-photochemical reactions after changes in irradiance in greenhouse mulberry plants. The induction of PSII photochemistry rapidly approached to steady state after leaves were treated from darkness to low irradiance (LI). When irradiance of leaves changed from darkness to high irradiance (HI), a biphasic induction was observed. A slight photoinhibition occurred in the leaves exposed to sunlight coming to the greenhouse, whereas a chronic photoinhibition occurred in the leaves fully exposed to sunlight outside the greenhouse. The chronic photoinhibition was demonstrated by sustained reduction of maximal quantum yield of PSII photochemistry (Fv/Fm). Moreover, the leaves of mulberry plants in greenhouse were sensitive to abrupt changes in irradiance and the sensitivity of leaves suffered in a short-term (1h) high light treatment was reduced, based on the changes in photosynthetic quantum conversion. These results demonstrated an inducible response of photosynthetic quantum conversion to changes in irradiance in mulberry.  相似文献

The relationships between non‐radiative energy dissipation and the carotenoid content, especially the xanthophyll cycle components, were studied in sun and shade leaves of several plants possessing C3 ( Hedera helix and Laurus nobilis ) or C4 ( Zea mays and Sorghum bicolor ) photosynthetic pathways. Sun‐shade acclimation caused marked changes in the organisation and function of photosynthetic apparatus, including significant variation in carotenoid content and composition. The contents of zanthophyll cycle pigments were higher in sun than in shade leaves in all species, but this difference was considerably greater in C3 than in C4 plants. The proportion of photoconvertible violaxanthin, that is the amount of violaxanthin (V) which can actually be de‐epoxidised to zeaxanthin, was much greater in sun than in shade leaves. The amount of photoconvertible V was always linearly dependent on the chlorophyll a/b ratio, although the slope of the relationship varied especially between C3 and C4 species. The leaf zeaxanthin and antheraxanthin contents were correlated with non‐radiative energy dissipation in all species under different light environments. These relationships were curvilinear and variable between sun and shade leaves and between C3 and C4 species. Hence, the dissipation of excess energy does not appear to be univocally dependent on zeaxanthin content and other photoprotective mechanisms may be involved under high irradiance stress. Such mechanisms appear largely variable between C3 and C4 species according to their photosynthetic characteristics.  相似文献

The effects of NaCl stress on the growth and photosynthetic characters of Ulmus pumila L. seedlings were investigated under sand culture condition. With increasing NaCl concentration, main stem height, branch number, leaf number, and leaf area declined, while Na+ content and the Na+/K+ ratio in both expanded and expanding leaves increased. Na+ content was significantly higher in expanded leaves than in those just expanding. Chlorophyll (Chl) a and Chl b contents declined as NaCl concentration increased. The net photosynthetic rate, intercellular CO2 concentration, stomatal conductance, and transpiration rate also declined, but stomatal limitation value increased as NaCl concentration increased. Both the maximal quantum yield of PSII photochemistry and the effective quantum yield of PSII photochemistry declined as NaCl concentration rose. These results suggest that the accumulation of Na+ in already expanded leaves might reduce damage to the expanding leaves and help U. pumila endure high salinity. The reduced photosynthesis in response to salt stress was mainly caused by stomatal limitation.  相似文献

Abstract. Kosteletzkya virginica (L.) Presl., a dicotyledonous halophyte native to brackish tidal marshes, was grown on nutrient solution containing 0. 85, 170 or 255 mol m-3 NaCl, and the effects of external salinity on shoot growth and ion content of individual leaves were studied in successive harvests. Growth was stimulated by 85 mol m-3 NaCl and was progressively reduced at the two higher salinities. Growth suppression at high salinity resulted principally from decreased leaf production and area, not from accelerated leaf death. As is characteristic of halophytic dicots. K. virginica accumulated inorganic ions in its leaves, particularly Na+ and K+. However, the Na+ concentration of individual leaves did not increase with time, but remained constant or even declined, seeming to be well-coordinated with changes in water content. A striking feature of the ion composition of salinized plants was the development of a dramatic gradient in sodium content, with Na+ partitioned away from the most actively growing leaves. Salt-treated plants exhibited a strong potassium affinity, with foliar K+ levels higher in salinized plants than unsalinized plants after an initial decrease. These results suggest that selective uptake and transport, foliar compartmentation of Na+ and K+ in opposite directions along the shoot axis, and the regulation of leaf salt loads over time to prevent build-up of toxic concentrations are whole-plant features which enable K. virginica to establish favourable K+-Na+ relations under saline conditions.  相似文献

Parameters for the evaluation of the effects of photoinhibition on photosynthetic carbon gain were studied in Chenopodium album leaves. The light-response curve of photosynthetic rate was determined at 36 Pa CO2 partial pressure and fitted by a non-rectangular hyperbola. Both the initial slope of the curve and the light-saturated rate decreased in photoinhibited leaves, although the decrease in the latter was small. The convexity of the curve was also smaller in photoinhibited leaves. The capacities of ribulose-1,5-bisphosphate carboxylation ( V cmax) and electron transport ( J max) were estimated from the CO2-response curves. V cmax and J max decreased similarly with increasing photoinhibition. Energy partitioning in photosystem II (PSII) was estimated using chlorophyll fluorescence parameters. The fraction of energy that was consumed by photochemistry decreased with increasing photoinhibition. However, an increase in inactive PSII, decreasing energy partitioning to active PSII, relaxed the excitation pressure in PSII, and led to a reduction in the fraction of excess energy that was neither consumed by photochemistry nor dissipated as heat.  相似文献
