Insect cells have recently proven to be an excellent platform for the high-level production of functional recombinant proteins. Autophagy is an important mechanism that promotes cell survival by eliminating damaged organelles and protein aggregates, and it also may influence recombinant protein production. In the present study, we compared the effects that autophagy inducers rapamycin, everolimus, and lithium chloride exert on recombinant lepidopteran insect cells that secrete an engineered antibody molecule. Compared with nontreatment, treatment with either rapamycin or everolimus prolonged cell growth to allow high cell density, improved viability in the declining phase, and then increased the yield of secreted antibodies. These positive effects appeared to be induced via autophagy since autophagosomes were clearly detected, particularly in cells treated with rapamycin or everolimus. Unlike rapamycin, another autophagy inducer, FK506, was ineffective in insect cells. The addition of an appropriate autophagy inducer may be effective in increasing the productivity of recombinant proteins in insect cells.

The larval drift behaviour of 23 species representing Ephemeroptera, Plecoptera and Trichoptera was investigated in the laboratory using different current regimes. Mayfly nymphs often performed swimming, while caddis larvae were reluctant to do so. Stonefly nymphs were intermediate. In mayflies swimming seemed to be used to reach the substrate as soon as possible. In contrast most stonefly nymphs by swimming prolonged the time spent in the water column. Modes of swimming and sinking posture differed markedly between the orders. Living passively sinking animals often reached bottom faster than dead control specimens, so consequently behaviour did not always express itself in activity. Some caddis larvae spun adherent anchor lines. Differences among taxa seemed more important in explaining swimming activity compared to preferred habitats (as stream, river and lake) in each species. However, observed differences among closely related species indicated subtle differences related to microhabitat to be of profound importance in explaining the alternative behavioural strategies used.  相似文献

A novel two-compartment bioreactor, BelloCell®, was used to cultivate insect cells and a maximum yield of 4.6 × 109 cells was attained. The cells were immobilized in a packed bed fixed in the upper chamber, and the bellow in the lower chamber was compressed and released in an alternating fashion. The motion resulted in gentle, cyclic movement of the medium that was contained in the lower chamber and consequently exposed the cells to air in an oscillatory manner, thus rendering adequate aeration and uniform cell distribution in the bed. The baculovirus yield produced in BelloCell® could amount up to 3.3 × 1017 pfu using as little as 1.1 l medium in the production run. Besides, BelloCell® was extremely easy to handle and operate. These benefits underline the potential of BelloCell® for simple, economical and high-density cell culture and protein/virus production.  相似文献

The rotavirus nonstructural NSP4 protein, a transmembrane endoplasmic reticulum-specific glycoprotein, has been described as the first viral enterotoxin. Purified NSP4 or a peptide corresponding to NSP4 residues 114-135 induces diarrhea in young mice. NSP4 has a membrane-destabilizing activity and causes an increase in intracellular calcium levels and chloride secretion by a calcium-dependent signalling pathway in eucaryotic cells. In this study, four recombinant baculoviruses were generated expressing the rotavirus NSP4 glycoprotein from the human strains Wa and Ito, the porcine strain OSU, and the simian strain SA11, which belong to two different NSP4 genotypes, A and B. The recombinant glycoproteins, expressed as polyhistidine-tagged molecules, were analyzed by Western blotting and immunoprecipitation. Newborn mice responded with diarrhea after inoculation with each of the recombinant NSP4 proteins.  相似文献

Production of recombinant proteins in high-density insect cell cultures   总被引:1,自引:0,他引:1  
The effect of the growth phase of Spodoptera frugiperda (Sf9) cells on the production of recombinant proteins (beta-galactosidase and glucocerebrosidase) was investigated. Cells infected with the recombinant Autographa californica nuclear polyhedrosis virus at the late exponential and stationary phases yielded low quantities of expressed protein. Highest enzyme yields were obtained using Sf9 cells from the early exponential phase (0.9 mg beta-galactosidase/10(6) cells and 1.7 mug glucocerebrosidase/10(6) cells). Infection of resuspension of cells collected from various phases of growth in fresh medium resulted in 75% restoration of maximal expression levels. This finding suggested either nutrient limitation or waste product accumulation as the cause of the decrease in productivity at the latter phases of growth. Further experiments revealed that the highest productivity levels could be obtained with cultures of Sf9 cells grown in a fermentor to a cell concentration of 4 x 10(6) mL(-1). Protein convertase 1/3 is a serine endoproteinase present in the regulated secretory pathway of endocrine and neuroendocrine cells. It is responsible for the processing of numerous prohormones and proneuropeptides into their biologically active moieties, often following cleavage at pairs of basic residues. The determination of its three-dimensional structure, as well as the understanding of its enzymatic properties, would greatly benefit from the production and availability of large amounts of recombinant enzyme. We report herein improvements in the production of PC1/3 by expressing recombinant mutated forms in both insect cells (Spodoptera frugiperda, Sf9 cells) and larvae (Trichoplusia ni commonly referred to as cabbage looper). On one hand, we deleted the last 135 COOH-terminal residues of mPC1/3 and, on the other hand, we replaced the signal peptide of mPC1/3 by the viral glycoprotein gp67 signal peptide. These modifications were shown to improve markedly (up to 125%) the secretion into the Sf9 cells medium and the amount of enzymatic activity recovered when compared to the original vector. Moreover, intracoelemic injection of the vectors into insect larvae led to the production and purification of enzymatically active enzyme at a level of 30 microg/larva in the case of mPC1/3 and to the production of a high amount of another enzymatically active convertase, PC7. The optimal viral titer for infection of larvae was determined to be 10(6)pfu/ml. Taking into account the purification protocol combined with the ease and efficiency of using larvae, it should now be possible to meet the needs for biochemical and structural studies.  相似文献

The production of an antibody single chain fragment (scFv) in insect cells was accompanied by the formation of an insoluble intracellular precursor even with the inclusion of the bee melittin signal peptide. The presence of the precursor polypeptide suggests a limitation in the processing of the signal peptide so a baculovirus containing a signal peptidase from Bacillus subtilis (SipS) was constructed for expression studies. When the wild type SipS was coexpressed with scFv, preprocessed scFv fragments were no longer detected in insect cell lysates. Conversely, coexpression of scFv alone or with an inactive mutant SipS resulted in at least 30% of the intracellular polypeptide in an unprocessed form at 3 days post infection. Production of scFv in the medium was also enhanced in the presence of SipS; however, low secretion levels indicate the presence of a post-processing bottleneck.  相似文献

It has been shown that severe acute respiratory syndrome-associated coronavirus (SARS-CoV) 3a and 7a proteins, but not membrane (M) protein, induce apoptosis in mammalian cells. Upon expression of SARS-CoV M protein using the baculovirus/insect cell expression system, however, we found that the expressed M protein triggered accelerated apoptosis in insect cells, as characterized by rapid cell death, elevated cytotoxicity, cell shrinkage, nuclear condensation and DNA fragmentation. Conversely, the M protein expressed in mammalian cells did not induce apoptosis. This is the first report describing the induction of apoptosis by SARS-CoV M protein in animal cells and possible implications are discussed.  相似文献

Human sapovirus (SaV) is uncultivable, but expression of the recombinant capsid protein (rVP1) in insect cells results in the formation of virus-like particles (VLPs) that are morphologically similar to the native viruses. However, the SaV rVP1 expression levels are considerably low. We have found that inclusions of short foreign nucleotide sequences inserted directly upstream from the predicted rVP1 AUG start codon lead to increased yield of VLPs. This method allowed us to express a SaV rVP1, which could not have been expressed to measurable or practical levels otherwise.  相似文献

A total of 13 insect cell lines spanning 4 orders (Lepidoptera, Coleoptera, Diptera, and Homoptera) were tested for their ability to replicate the nonoccluded virus Hz-1. Only the Lepidopteran cell lines supported replication of the virus with TN-CL1 and BCIRL-HZ-AM1 producing the highest titers of 2.4 × 108 tissue culture infective dose (TCID)50/ml and 2.0 × 108 TCID50/ml, respectively. A codling moth cell line (CP-169) was the only Lepidopteran cell line that did not replicate the virus and transfection of this cell line with Hz-1 DNA failed to replicate the virus. Also, transfection with DNA from a recombinant baculovirus carrying the red fluorescent protein gene (AcMNPVhsp70 Red) was not expressed in CP-169 cells. The replication cycle of Hz-1 in BCIRL-HZ-AM1 cells showed that this virus replicated rapidly starting at 16 h postinoculation (p.i.) and reaching a peak titer of 1.0 × 108 TCID50/ml 56 h postinoculation. Hz-1 when compared with several other baculoviruses has the widest in vitro host spectrum.  相似文献

The growth and development of the nuclear population from intersegmental dorsal longitudinal muscles (IDLM) of Bombyx mori and Pieris brassicae during larval development were studied. Using whole muscle mounts, examined with an image analysis system, we observed increases in muscle size but a constant number of fibres and concomitant increases in the number and size of fibre nuclei. The gradual growth of the nuclei throughout larval life, corresponding very probably to an increase in their DNA content, is unequal and results in heterogeneity of the nuclear population. This suggests periodic incorporation of myoblasts into the larval muscles; the nuclei arising from these myoblasts would therefore undergo endomitotic processes and grow polyploid. The possibility of somatic reduction divisions of the polyploid muscle nuclei is discussed.  相似文献

Self-incompatibility is a phenomenon that involves recognition of self versus non-self pollen, leading to the rejection of self-related pollen and preventing self-fertilization. In this study, we used a baculovirus-infected insect cell culture system to express two Brassica oleracea stigma-specific proteins required for self-incompatibility: the S-locus glycoprotein, a soluble cell wall-localized glycosylated protein, and the S-locus receptor kinase, a receptor-like integral plasma membrane glycoprotein with serine/threonine kinase activity. Insect cells expressing the S-locus receptor kinase were used in conjunction with immunofluorescence and a whole cell enzyme-linked immunosorbant assay to demonstrate that the receptor is targeted to the cell surface and is oriented with its N-terminal S domain towards the outside of the cell. Human haptoglobin (Hp2) was synthesized in insect cells using the baculovirusAutographa california nuclear polyhedrosis virus (AcNPV) as an expression vector. Viruses carrying the proHp2 cDNA, either fused or non fused to viral polyhedrin DNA sequences, expressed intracellularly low levels of unglycosylated and non maturated haptoglobin. On the contrary, recombinant viruses containing the preoroHp2 cDNA directed the expression of high levels of prohaptoglobin. To a large extent, the uncleaved product was found in the culture medium as a glycosylated molecule. Despite the lack of maturation into subunits, the secreted recombinant prohaptoglobin was able to bind hemoglobin in vitro, although less efficiently than plasma-derived haptoglobin.  相似文献

The production of C595 diabody fragment (dbFv) in Escherichia coli (E. coli) HB2151 clone has been explored. The comparison of fermentation processes mode demonstrated that a higher biomass inoculum operation enhanced C595 dbFv production. It was demonstrated that a concentration of 12.1 mg l−1 broth of dbFv and a cell concentration of 23.6 g l−1 broth were achieved at the end of 75 l fermentation.  相似文献

Biologically active recombinant human erythropoietin has been expressed at high levels in an insect cell background. Expression involved the preparation of a human erythropoietin cDNA, the transfer of this cDNA to the Autographa californica nuclear polyhedrosis virus (AcNPV) genome under the polyhedrin gene promoter, and the subsequent infection of Spodoptera frugiperda cells with recombinant AcNPV. Erythropoietin cDNA was prepared through the expression of the human erythropoietin gene in COS cells using pSV2 and the construction of a COS cell cDNA library in bacteriophage Lambda GT10. Prior to transfer to the AcNPV genome, erythropoietin cDNA isolated from this library was modified at the 3′-terminus in order to replace genomic erythropoietin for SV40 cDNA derived from pSV2. Transfer of this cDNA to AcNPV and the infection of S. frugiperda cells with cloned recombinant virus led to the secretion of erythropoietin: based on bioassay, rates of hormone secretion (over 40 U/ml per h) were 50-fold greater than observed for COS cells. The purified recombinant product possessed full biological activity (at least 200000 U/mg), but was of lower Mr (23000) than human erythropoietin produced in COS cells (30000) or purified from urine (30000 to 38000). This difference was attributed to the glycosylation of erythropoietin in S. frugiperda cells with oligosaccharides of only limited size. Further removal of N-linked oligosac-charides from this Mr 23000 hormone using N-Glycanase yielded an apo-erythropoietin (Mr 18000) which possessed substantially reduced biological activity. These results indicate that glycosylation, but not the normal processing of oligosaccharides to complex types, is required for the full hormonal activity of human erythropoietin during red cell development.  相似文献

A model is constructed to describe gregariousness of colonial insect larvae which occure in discrete natural habitat units. The model is based on the assumption that the distribution of individuals in a clump is determined by a balance between aggregative and dispersing movements of individuals. The model includes three parameters, i.e., clump size, carrying capacity of habitat units and intensity of aggregation against dispersal. The modes of effects of these parameters on the distribution of individuals are analyzed. Several methods are developed to estimate these parameters from observed data, and their applicabilities are investigated. Application of these methods are described on the larval distribution of a colonial insect Pryeria sinicaMoore .  相似文献

The renin-angiotensin (RA) system is important for the regulation of blood pressure and electrolyte balance, and renin is the rate-limiting enzyme in this system. The recent discovery of (pro)renin receptor (PRR) has reinforced the functional role of the RA system. PRR non-proteolytically activates prorenin and its role has attracted the attention of researchers towards the RA system. However, there is insufficient information on the biochemical structure and biological functioning of PRR due to the difficulty of measuring PRR expression. In this work, human PRR (hPRR) with intact transmembrane and C-terminal domain (hPRR-wTM) and PRR without this domain (hPRR-w/oTM) were expressed in insect cells using baculovirus expression system (BES). Both hPRR-wTM and hPRR-w/oTM were fused with FLAG peptide by its N-terminus. Most of the hPRR-wTM was expressed in cell fraction and hPRR-w/oTM was secreted into the culture medium. hPRR-wTM was solubilized from the membrane fraction of recombinant baculovirus-infected cells by various detergents, suggesting that hPRR-wTM might be a transmembrane protein. hPRR-wTM was purified from the solubilized fraction using anti-FLAG M2 antibody agarose. Binding of purified hPRR-wTM to renin immobilized onto sensor chips was directly proportional to the hPRR-wTM concentration. Approximately 225 μg of functional hPRR-wTM was purified from 80 ml of baculovirus-infected cell culture. Scale-up of this system will lead to mass production and crystallization of hPRR-wTM and determination of its biochemical structure and biological function.  相似文献

A new technique for the direct production of recombinant baculovirus in the silkworm larvae is described. To assess the utility of this method, a combination of Bombyx mori nucleopolyhedroviral genome, transfer vector and Lipofectin was co-injected directly into newly ecdysed fifth instar, silkworm larvae. The recombinant virus was obtained from the hemolymph of injected larvae and the hemolymph then re-injected into the larvae as an inoculum. This resulted in a high-level production of foreign protein in the silkworm larvae. This technique produces easy and rapid recombinant protein production in silkworms.  相似文献

Recombinant bovine cardiac sodium-calcium exchange (NCX1) in a baculovirus construct was used to infect cabbage looper larvae (Trichoplusia ni). Infected larvae were homogenized and larvae membrane vesicles were purified. Western blot analysis indicated the presence of recombinant NCX1 protein in vesicles from infected larvae but not in controls. Vesicles from infected larvae expressed high levels of NCX1 activity (1.7 nmol Ca2+/mg protein/s) while vesicles from control larvae had no activity. NCX1 in larvae vesicles was bidirectional. Kinetic analysis yielded a Vmax of 3.6 nmol Ca2+/mg protein/s and a Km for Ca of 4.2 microM. NCX1 activity was inhibited by the exchange inhibitory peptide with an IC50 of 4 microM. These data demonstrate a novel and efficient method for the expression of large amounts of active recombinant NCX1 protein that has general application for expression and analysis of recombinant membrane proteins.  相似文献

Summary

There is a latitudinal gradient in the species richness of Decapoda with pelagic larvae. For example, only two species of Brachyura are found around Svalbard, 54 species are known from the English Channel and ~100 from the Atlantic coast of the Iberian peninsula. The distributions of many species are limited by effects of temperature. Intermoult times of larvae are inversely related to temperature. In the field the size of larvae is inversely related to temperature, the relationship tending to increase in significance in successive larval stages. The timing of the seasonal occurrence of larvae of decapod species in the plankton is also related to temperature, to a greater extent than are seasonal cycles of the holoplankton. These effects of temperature on larval development influence the biomass and production of the larvae in the plankton. Some potential effects of climate change on the distributions and dynamics of planktonic larvae of decapods and consequent changes in their diversity, biomass and production can be predicted insofar as temperature is a limiting factor.  相似文献
