Phytochemicals are constitutive metabolites that enable plants to overcome temporary or continuous threats integral to their environment, while also controlling essential functions of growth and reproduction. All of these roles are generally advantageous to the producing organisms but the inherent biological activity of such constituents often causes dramatic adverse consequences in other organisms that may be exposed to them. Nevertheless, such effects may be the essential indicator of desirable properties, such as therapeutic potential, especially when the mechanism of bioactivity can be delineated. Careful observation of cause and effect, followed by a coordinated approach to identify the responsible entities, has proved extremely fruitful in discovering roles for phytochemical constituents. The process is illustrated by selected examples of plants poisonous to animals and include the steroidal alkaloid toxin of Veratrum californicum (Western false hellebore), piperidine alkaloids of Lupinus species (lupines), and polyhydroxy indolizidine, pyrrolizidine and nortropane alkaloids of Astragalus and Oxytropis species (locoweeds), Castanospermum australe (Moreton Bay chestnut) and Ipomoea species (morning glories).  相似文献

Pathological aggregation of the microtubule-associated protein tau is a common feature of many neurodegenerative diseases. Although tau aggregation is associated with abnormal tau phosphorylation, the role of phosphorylation in the initiation of neurodegeneration has been unclear. Now, several animal models and data from human patients provide converging evidence that aberrant tau phosphorylation can cause a neurodegenerative phenotype similar to that seen in human neurodegenerative diseases.  相似文献

A total of 842 white sea bream (Diplodus sargus), sampled in Banyuls-sur-Mer, were analysed to test 'genotype-phenotype' relationship for various characters related to the fitness. The results show significant differences (MLH and FIS) for the age according to the sex between females carrying out and not carrying out sexual inversion. This suggests an overdominance for old females and a genetic sex determination. The individuals laying very early during the period of reproduction are also differentiated from the individuals reproducing later in the season. These results suggest either a stable calendar of laying in time separating the individuals genetically reproducing precociously from the others and this by differential selection and/or genetic drift either a Wahlund effect among cohorts.  相似文献

Anthropology should deal with man holistically, but has failed to do so. The field is unfortunately split between mentalistic and materialistic schools and has not developed a theory to cope with the complexity of large contemporary societies. Energy similarly has been little studied by anthropology because there has been no theory sufficient for the purpose. The thermodynamics of structures far from equilibrium and the dissipative structure model of Ilya Prigogine offer a method for both studying energy in society and dealing with society as a whole. Anthropologists are urged to pursue study of this area because it is critical for an understanding of human destiny . Aulisio and Arora argue that the moral significance of value imposition explains the moral distinction between traditional conscientious objection and non-traditional conscientious objection. The former objects to directly performing actions, whereas the latter objects to indirectly assisting actions on the grounds that indirectly assisting makes the actor morally complicit. Examples of non-traditional conscientious objection include objections to the duty to refer. Typically, we expect physicians who object to a practice to refer, but the non-traditional conscientious objector physician refuses to refer. Aulisio and Arora argue that physicians have a duty to refer because refusing to do so violates the patient’s values. While we agree with Aulisio and Arora’s conclusions, we argue value imposition cannot adequately explain the moral difference between traditional conscientious objection and non-traditional conscientious objection. Treating autonomy as the freedom to live in accordance with one’s values, as Aulisio and Arora do, is a departure from traditional liberal conceptions of autonomy and consequently fails to explain the moral difference between the two kinds of objection. We outline how a traditional liberal understanding of autonomy would help in this regard, and we make two additional arguments—one that maintains that non-traditional conscientious objection undermines society’s autonomy, and another that maintains that it undermines the physician-patient relationship—to establish why physicians have a duty to refer.  相似文献

Recent advances have highlighted the complex web of biological mechanisms and pathways involved in oncogenic transformation and maintenance of the cancer phenotype. To that end, a number of key factors have been identified and thoroughly investigated over the past couple of decades, such as redox regulation of cell fate decisions, cellular metabolism and bioenergetics, autophagy induction as a survival signal, and how these pathways interplay with oncogene-induced transformation. This has been particularly well documented for oncoprotein Ras-induced carcinogenesis, and recent reports provide ample evidence to indicate a well-coordinated crosstalk between these diverse cellular pathways in the process of cancer initiation and progression. Here we provide a brief summary of the recent advances in the field to illustrate the dual role of autophagy as a tumor suppressor and as a survival mechanism required for cancer maintenance as well as its implication in the complex relationship between Ras-mediated carcinogenesis, mitochondrial metabolism, cellular redox status and bioenergetics.  相似文献

The leaf-beetle Leptomona russica (Gmelin, 1790) known from the Caucasus, Kazakhstan, and Middle Asia was found in Orenburg Province for the first time. It is the first record of this species from Europe and the first record of the genus Leptomona from European Russia. Ultrastructure of stemmata (larval eyes), stemmatal nerves, and the optic neuropils of 5th-instar larvae of cotton bollworm, Heliothis armigera (Hübner) (Lepidoptera : Noctuidae), were examined with scanning and transmission electron microscopes. Six stemmata are on each side of the head. Each stemma consists of 7 retinula cells arranged into 2 tiers. Stemmata I and II have 4 distal retinula cells and 3 proximal cells, the other 4 stemmata (III–IV) have 3 distal cells and 4 proximal cells. Stemmata I and IV have a short proximal rhabdom and the rhabdomere of each proximal cell has its microvilli projecting in only one direction. On the other hand, each stemma (in stemmata II–V) has a long proximal rhabdom and the rhabdomere of each proximal cell has microvilli pitched in several different directions relative to the horizontal plane. An axon projects proximally from each retinula cell body. The stemmatal nerve is composed of the 42 retinular axons from all of the 6 stemmata on the same side of the head. Each stemmatal nerve projects to the ipsilateral optic neuropil. Axons from each stemma are in a fasicle (within the stemmatal nerve), which consists of 7 axons, 3–4 of them are thick and terminate synaptically in the proximal neuropil; the others are thinner and terminate in the distal neuropil. Organelles, particularly lysosomes, undergo ultrastructural transformations relative to ambient light levels. The functional significance of abovementioned structures are discussed in light of current knowledge.  相似文献

We propose that a fruitful way to understand the extension of molecular biology into clinical practice is through the notion of biomedical platform. In this article, we analyze the development of the new genetics in clinical haematology since the mid‐1980s, focusing on two recent instantiations of the molecular biology platform: RT‐PCR and DNA microarrays. We show how clinical research is more closely entwined with “fundamental” biology than is often imagined and simultaneously caution that the use of techniques in clinical research does not automatically entail their use in routine clinical practice. The article calls attention to the work of articulation and regulation as constitutive aspects of a platform that enters standard clinical use.  相似文献

Progenitor cell based therapies have emerged for the treatment of ischemic cardiovascular diseases where there is insufficient endogenous repair. However, clinical success has been limited, which challenges the original premise that transplanted progenitor cells would orchestrate repair. In this review, we discuss the basics of endothelial progenitor cell therapy and describe how microenvironmental changes (i.e., trophic and mechano-structural factors) in the damaged myocardium influence progenitor cell plasticity and hamper beneficial therapeutic outcome. Further understanding of these microenvironmental clues will enable optimization of cell therapy at all levels. We discuss current concepts and provide future perspectives for the enhancement of progenitor cell therapy, and merge these advances into a combined approach for ischemic tissue repair.  相似文献

Atherogenesis requires and is highly influenced by the interaction between lipoproteins and macrophages. Most of the focus to date has been on the ability of atherogenic lipoproteins (such as low-density lipoproteins, LDL) to promote and of anti-atherogenic lipoproteins (such as high-density lipoproteins, HDL) to prevent the development of the cholesteryl ester-enriched macrophage-derived foam cell. However, lipoprotein-macrophage interactions have the potential to modulate macrophage function in a variety of additional ways that may impact on atherosclerosis. These include modulating cellular cholesterol and oxysterol content, providing fatty acids as ligands for PPARs, and acting as ligands for macrophage scavenger and Toll-like receptors. We suggest that atherogenic lipoproteins promote and anti-atherogenic lipoproteins inhibit atherogenesis by modulating macrophage function in a variety of ways beyond cholesteryl ester accumulation and foam cell formation.  相似文献

The kinetochore is the proteinaceous complex that governs the movement of duplicated chromosomes by interacting with spindle microtubules during mitosis and meiosis. Faithful chromosome segregation requires that kinetochores form robust load-bearing attachments to the tips of dynamic spindle microtubules, correct microtubule attachment errors, and delay the onset of anaphase until all chromosomes have made proper attachments. To understand how this macromolecular machine operates to segregate duplicated chromosomes with exquisite accuracy, it is critical to reconstitute and study kinetochore–microtubule interactions in vitro using defined components. Here, we review the current status of reconstitution as well as recent progress in understanding the microtubule-binding functions of kinetochores in vivo.  相似文献

Central nervous system (CNS) development involves neural patterning, neuronal and axonal migrations, and synapse formation. DSCAM, a chromosome 21 axon guidance molecule, is expressed by CNS neurons during development and throughout adult life. We now report that DSCAM and its chromosome 11 paralog DSCAML1 exhibit inverse ventral-dorsal expression patterns in the developing spinal cord and distinct, partly inverse, expression patterns in the developing cortex, beginning in the Cajal-Retzius cells. In the adult cortex, DSCAM predominates in layer 3/5 pyramidal cells and DSCAML1 predominates in layer 2 granule cells. In the cerebellum, DSCAM is stronger in the Purkinje cells and DSCAML1 in the granule cells. Finally, we find that the predicted DSCAML1 protein contains 60 additional N-terminal amino acids which may contribute to its distinct expression pattern and putative function. We propose that the DSCAMs comprise novel elements of the pathways mediating dorsal-ventral patterning and cell-fate specification in the developing CNS.  相似文献

Translocations or other movements of wildlife sometimes accomplish their intended objectives, but unforeseen consequences may arise and disrupt locally adapted ecological communities, restructure or dilute genetic integrity of populations or subspecies of the moved organism, and otherwise negatively influences a species’ long‐term fitness. Two historical populations of Mottled Ducks (Anas fulvigula) exist and are endemic to (1) Mexico and the West‐Gulf Coast (A. f. maculosa) regions of the United States and (2) Florida (A. f. fulvigula). From 1975 to 1983, 1285 Mottled Ducks from Florida, Louisiana, and Texas were released to coastal South Carolina, primarily to ultimately establish a legally harvestable population. This movement stirred mixed reactions amid the conservation community. Contemporary information suggests an increasing Mottled Duck population in South Carolina and possibly dispersing into Georgia. Herein, I objectively discuss the potential consequences of this new population per the birds’ evolution, ecology, and management. Ultimately, I suggest that this translocation is a long‐term benefit to the species.  相似文献
