Archegonium development, beginning with the archegonial initial and culminating in the mature egg, was studied with the electron microscope. The ultrastructural features of the beginning stages in development of the archegonium are relatively similar to one another. Plasmodesmata occur between all adjacent cells at this time. After the secondary central cell is formed these protoplasmic connections are lost, and both axial and parietal cell lineages begin to show signs of ultrastructural differentiation. The mature egg is characterized by cytoplasm rich in ribosomes and larger organelles. Mitochondria and simplified plastids commonly display a juxtaposed association. As far as could be ascertained the numerous plastids and mitochondria in the egg of Marchantia arise through division of preexisting organelles and are not formed anew from evaginations of the nucleus. Blebbing of the nucleus produces polymorphic organelles which appear to be pinched off into the cytoplasm. The mature egg also contains vacuoles and lipid bodies toward its periphery, while dictyosomes and extensive endoplasmic reticulum occur throughout. The space between the wall cells and the mature egg appears to contain an amorphous substance. No extra membrane was observed around the mature egg.  相似文献

In the liverwort Sphaerocarpus donnellii Aust., the behavior of the cell constituents, especially of mitochondria and plastids, was studied by electron microscopy during the development of the egg and its preceding cells. A degeneration and elimination of mitochondria and plastids was not found in any of the developmental stages. In all growth phases of the archegonium, the plastids may deposit starch which becomes especially frequent in the maturing egg cell. No indications have been observed that new mitochondria or plastids generate from the nuclear evaginations, which often penetrate deeply into the cytoplasm of the maturing and fully developed eggs. A quantitative investigation based on general micrographs elucidates the numerical aspects of the cell constituents during oögenesis. With the increase of cell volume, the numbers of dictyosomes, mitochondria, plastids, and lipid bodies increase. From the stages of the mother cell of the axial row up to that of the mature egg, the cell volume enlarges about 8 times and the nucleus volume about 15 times. Simultaneously, the numbers of mitochondria and plastids increase up to 8 to 15 times. On the basis of these findings, mitochondria and plastids with three-dimensional narrow constrictions are interpreted as divisional stages.  相似文献

At maturity the companion cell of the phloem of the sycamore Acer pseudoplatanus has a large nucleus, simple plastids closely sheathed with rough endoplasmic reticulum, and numerous mitochondria. The cytoplasm contains numerous ribosomes, resulting in a very electron-opaque cytoplasm after permanganate fixation. Bodies similar to the spherosomes of Frey-Wyssling et al. (4) are collected in clusters and these also contain bodies of an unidentified nature similar to those found by Buttrose (1) in the aleurone cells of the wheat grain. The pores through the wall between the companion cell and sieve tube are complex and develop from a single plasmodesma. Eight to fifteen plasmodesmata on the companion cell side communicate individually with a cavity in the centre of the wall which is linked to the sieve tube by a single pore about twice the diameter of an individual plasmodesma. This pore is lined with material of an electron opacity equivalent to that of material bounding the sieve plate pores. The development of the cell organelles, the possible role played in the phloem tissue by the companion cell, and the function of the complex pores contained in its wall are discussed.  相似文献

The composition and ultrastructure of the synergids of cotton were studied. The cells were found to be surrounded by a partial wall composed of cellulose, hemicellulose, and pectins. The structure of the wall was observed to consist of an unusual fibrillar arrangement. The filiform apparatus was demonstrated to be an extension of the wall at the micropylar end of the cell. Large amounts of ER surround the filiform apparatus. Also associated with the latter are large numbers of plastids and mitochondria. The nucleus is large and contains a single, large nucleolus and, frequently, 1 or more micronucleoli. The nuclear membrane contains membrane-bound vesicles but has few extensions into the cytoplasm. The ER is oriented parallel to the long axis of the cell and decreases in concentration from the micropylar to the chalazal end of the cell. Dictyosomes are common throughout the cell but are more numerous in the midportion where they are closely associated with the ER. The chalazal end of the cell is occupied by vacuoles rich in an inorganic compound which leaves a considerable residue of ash. Spherosome-like bodies are common throughout the cell. Both the plastids and mitochondria show evidence of division. Ribosomes are numerous and are both free and associated with the ER, nucleus, plastids, and mitochondria. The function of the synergids is proposed to be the absorption, storage, and transport of compounds from the nucellus. On the basis of this function, it is suggested that the synergids act by providing material to the egg and the developing embryo and endosperm and that they are involved in the growth of the pollen tube into the embryo sac.  相似文献

1. The structure and distribution of the components of striated muscle cells vary with the species and with the specialization of muscle fiber function. 2. There appear to be two, easily distinguishable, general categories of striated muscle structure. A. High frequency muscle (represented by flight muscle of higher insects and hummingbird, and cicada tympanal muscle) is characterized by widely spaced, non-branching fibrils of large diameter and short period, little endoplasmic reticulum, and large quantities of large mitochondria (low fibril-sarcoplasm ratio). This structure is correlated with heavy tracheolization or vascularization, high oxidative activity, and dark color as compared with other muscles of the same species. B. Low frequency muscle is characterized, in general, by high fibril-sarcoplasm ratio, relatively long period, few mitochondria increasing with activity and decreasing with absolute power of the fiber. Oxidative capacity and color are proportional to the quantity of mitochondria. These fibers are further differentiated into (a) fibrillar arrangement of contractile material which permits a regular pattern of interfibrillar and segmental reticulum, and (b) afibrillar arrangement of contractile material leading to an unsystematic distribution of reticulum. 3. The endoplasmic reticulum appears as a complex coordination system in the muscle fiber. Peripherally, it links the Z and M lines of the fibrils to the sarcolemma and between the fibrils it links the cross-bands, forming the Grundmembran of earlier authors. By longitudinal linkage, it connects with the sarcolemma at the muscle extremity to form a digital arrangement into which the tendon fibrils are spliced. The extent of its development and its position have a definite relationship to the degree and site of fiber shortening. At present the reticulum is the only structure that one can consider to be an internal conducting system. It may distribute the excitation transversely from fibril to fibril, and lengthwise saltatorially to the symmetry centers of the sarcomeres. 4. The nucleus is the mediating element between the cytoplasmic phases within and without the tubular system of the endoplasmic reticulum. A possible mechanism which correlates nucleus, adenylic acid system, ion exchange, and reticulum with the initiation of contraction is postulated.  相似文献

中华绒螯蟹的成熟卵仅有初级卵膜,无次级和三级卵巢,质膜初期厚而多层,且具皱褶,卵核在卵的发育过程中变化很大,未发见中心粒,内质网和高尔基体均始见于蟹卵发育的初期,皮层颗粒先出现于蟹卵深部,随后移到卵的表层,无滋养细胞,蟹卵由卵泡细胞提供物质,形成卵黄,此外,还可直接从血淋巴内摄取卵黄前身物质。  相似文献

Developmental phases surrounding the processes of gametic delivery and fusion were examined ultrastructurally in the reduced megagametophyte of Plumbago zeylanica, which lacks synergids. Gametic delivery occurs at the end of pollen tube growth and results in deposition of two male gametes, a vegetative nucleus, and a limited amount of pollen cytoplasm between the egg and central cell. Discharge of these materials from the tube is accompanied by loss of inner and outer pollen tube plasma membranes, loss of sperm-associated cell wall components, and disruption of the formerly continuous cell wall between the egg and central cell. The dispersion of egg cell wall components directly exposes female reproductive cell membranes to the unfused male gametes and pollen tube without disrupting gametic cell plasma membranes. Presence of unfused sperms within the female gametophyte appears to be a transitory phenomenon, lasting less than 5 min at the end of over 8½ hr of pollen tube growth. At the time of gametic deposition, plasma membranes of unfused sperm cells become directly appressed to plasma membranes of both the egg and central cell. Gametic fusion is initiated by a single fusion event between membranes of participating male and female cells, which is rapidly followed by subsequent, secondary fusion events between the same two cells at different locations along their surface. Gametic fusion results in the transmission of male gamete nuclei with co-transmission of nearly the entire sperm cytoplasmic volume and organellar complement, and it is possible to identify heritable male cytoplasmic organelles within both the incipient zygote and endosperm. Paternally originating plastids may be distinguished from maternal plastids by differences in morphology and staining characteristics, whereas paternal mitochondria may be distinguished from maternal mitochondria by populational differences in mitochondrial size which are statistically significant. Such observations further indicate that transmitted paternal mitochondria seem to remain viable, as judged by their ultrastructural appearance, and are transmitted exclusively by sperm cytoplasm rather than discharged pollen cytoplasm. The presence of anucleate, membrane-bounded cytoplasmic bodies between the egg and central cell are identifiable on the basis of their enclosed organelles and indicate that fragmentation of a small amount of the sperm cytoplasm associated with the vegetative nucleus commonly occurs. The presence and identification of sperm cytoplasmic organelles and associated membranes within female reproductive cells following gametic transmission represents strong evidence in support of the cellular basis of nuclear and cytoplasmic transmission during sexual reproduction in Plumbago.  相似文献

云南油杉(Keteleeria evelyniana Mast)在受精前,精核与卵核周围的细胞质鞘不明显。受精后,合子核周围出现细密的新细胞质。应用孚尔根核染色法,可以较清晰地将新细胞质染出,呈现较弱的正反应,而合子的核质及受精前的精核与卵核染色极弱。卵细胞质及其中的蛋白泡均为负反应。原胚形成后,除上层外,其余几层细胞质内开始积累淀粉粒。此时胚原细胞核的孚尔根染色深度有所增加。幼胚形成后,在顶端的胚原细胞群中核的孚尔根染色反应已恢复正常。在原胚及幼胚胚原细胞质中也呈现很弱的正反应。在电镜下,胚原层细胞质及新细胞质中均含有核样电子致密小体或称作染色质小体,而原胚莲座层细胞质及四周套细胞质中的线粒体则不含这种核样小体。因此,大蛋白泡在卵核形成的早期数量不多,当合子形成时含量最高,而随着游离核的分裂进程,蛋白泡以及原卵质均逐渐地解体,在原胚形成后全部消解。  相似文献

1. The eggs of many mammalian species show signs of early parthenogenetic development as they age after ovulation and oocytes may form transplantable terato-carcinomas. These cases of apparently spontaneous parthenogenetic development suggest that the cells of the female germ line have an inherent tendency to divide and differentiate. 2. The ovulated eggs of virgin female mammals may be stimulated to start parthenogenetic development by a wide variety of treatments. Most of these damage the egg so that it does not develop beyond the 4 cell stage. However if the eggs are exposed to electrical activation, hyaluronidase treatment, or temperature shock then in many cases they will develop into blastocysts. 3. These blastocysts may be either haploid or diploid. Haploid blastocysts may be formed either by the egg extruding the nucleus of the second polar body or by the egg dividing in half, so that the female pronucleus is in one cell and the nucleus of the second polar body is in another cell. Diploid blastocysts are formed by the retention of the nucleus of the second polar body within the egg. The way in which the egg develops may be controlled by altering the osmolarity of the culture medium, the age of the egg at the time of activation, or the strain of animal used. 4. The action of the sperm on the egg can be defined by comparing the events of normal fertilization and parthenogenetic activation. Both these stimuli cause the egg to expose binding sites for Concanavalin A to synthesize DNA and to divide. However, the release of cortical granules, which occurs after fertilization, does not appear to be induced by parthenogenetic activation, and it is significant that parthenogenones lack the sperm nucleus and mitochondria. 5. The majority of parthenogenones die soon after implantation. Death at this time occurs with parthenogenones obtained from the activated eggs of both inbred and outbred stocks. Death might be caused by recessive lethal mutations or by extra-genetic effects of the maternal chromosomes. 6. Parthenogenones contain endogenous A-type particles which shows that these bodies are inherited through the female germ line. 7. Parthenogenones may in the future provide both a method for chromosome mapping and a source of haploid cells. At present the use of mammalian parthenogenones in biological research is restricted by the heavy embryonic losses which occur around the time of implantation. This means that the role of the sperm, gene activity and virus expression must be studied during a very limited period. Part of the mortality before implantation is the consequence of the damage which the egg suffers during activation and it should be possible to reduce this loss by improving the techniques for activation. It may also be possible to increase the quantity of cells derived from haploid and diploid mammalian embryos by deriving teratocarcinomas from them.  相似文献

Mature Arbacia eggs were extracted with cold dilute perchloric acid, the extract concentrated, and the concentrate digested in hot perchloric acid. Thymine was recovered from the digest by paper chromatography, and the amount per egg found to be about 5 times the amount per sperm. This was the amount expected from previous experiments and is believed to represent all or almost all of the thymine in the egg. The result supports previous observations that DNA is absent from the mature egg although present in the nucleus of the egg in the germinal vesicle stage. No thymine could be recovered from a similar extract of 5,000 times as many sperm of the same species. The observations are consistent with the theory that DNA and its derivatives act as metabolic antagonists of the corresponding ribose compounds.  相似文献

The fine structure of the exoerythrocytic cycle of an avian malarial parasite, Plasmodium fallax, has been analyzed using preparations grown in a tissue culture system derived from embryonic turkey brain cells which were fixed in glutaraldehyde-OsO4. The mature merozoite, an elongated cell 3- to 4-µ long and 1- to 2-µ wide, is ensheathed in a complex double-layered pellicle. The anterior end consists of a conoid, from which emanate two lobed paired organelles and several closely associated dense bodies. A nucleus is situated in the mid portion of the cell, while a single mitochondrion wrapped around a spherical body is found in the posterior end. On the pellicle of the merozoite near the nucleus a cytostomal cavity, 80 to 100 mµ in diameter, is located. Based on changes in fine structure, the subsequent sequence of development is divided into three phases: first, the dedifferentiation phase, in which the merozoite loses many complex structures, i.e. the conoid, paired organelles, dense bodies, spherical body, and the thick inner layers of the pellicle, and transforms into a trophozoite; second, the growth phase, which consists of many nuclear divisions as well as parallel increases in mitochondria, endoplasmic reticulum, and ribosomes; and third, the redifferentiation and cytoplasmic schizogony phase, in which the specialized organelles reappear as the new merozoites bud off from the mother schizont.  相似文献

The immature megaspore mother cell of Ginkgo biloba is essentially spherical and is surrounded by a thick, complex wall. A large nucleus occupies the central region of the cell, and the organelles appear to be randomly arranged in the cytoplasm. With approaching maturity and the onset of meiosis, the cell elongates in the direction of the ovular axis. An extensive system of ER develops at the micropylar pole of the cell during elongation, and the plastids and mitochondria migrate to the opposite or chalazal pole. The micropylar end of the mature megaspore mother cell is usually devoid of plastids and mitochondria, but these organelles are densely packed in the chalazal end of the cell below the nucleus. The dictyosomes and dense spherosome-like bodies do not show such polarity in their distribution. At meiosis I plastids and mitochondria are, as a rule, restricted to the chalazal dyad cell that is destined to produce the functional megaspore. The wall of the megaspore mother cell consists of a middle lamella which is irregularly thickened, an outer wall layer resembling the walls of the surrounding nutritive cells, and an inner layer resembling the middle lamella in appearance.  相似文献

应用透射电镜研究了临近受精时天竺葵(Pelargonium hortorum Bailey)胚囊中的卵细胞、助细胞和中央细胞的结构。证明了卵细胞与助细胞以及助细胞与助细胞之间从合点端至珠孔端有很大的面积以质膜分界,仅珠孔端少部分以壁分隔。卵细胞与中央细胞之间同样缺乏细胞壁。在卵细胞的合点端,两质膜不同程度地分离形成宽窄相间的间隙。在间隙的絮状基质中存在小泡,这些小泡的产生似与卵和中央细胞中周质内质网的活动有关。推测小泡为多糖性质,可能为合子新壁的建造提供物质。卵细胞质中含巨大线粒体,质体和内质网也较丰富。基于超微结构的特征,可认为卵细胞具高度的生理合成活动的潜能。中央细胞极核位于珠孔端与卵器细胞毗邻,有利于在双受精作用中同时发生精细胞与卵细胞和精细胞与中央细胞核的融合。中央细胞的侧壁在珠孔端形成内突,具传递细胞的特点,表明这是雌配子体向孢子体摄取营养的重要部位。助细胞的细胞质含丰富的细胞器,这与多数植物中的相似,但具几个明显的特征,即核中存在微核仁,内质网形成圆球体或脂体,线粒体富集在丝状器的附近。传粉后花粉管进入胚囊之前,两个助细胞中一个退化。  相似文献

黄颡鱼精子由头部、中段和鞭毛(尾部)三部分组成。头部的主要结构是细胞核。核中浓缩了的染色质呈颗粒状。染色质中有核泡存在。核泡中有致密颗粒状物。植入窝里井状,从核后端往前深陷入核的中央。中段的中心粒复合体位于植入窝中,结构独特。近端中心粒和基体首尾相对,排在同一直线上。某些精子的近端中心粒的中央腔中能见到一、二个粗大的颗粒状物。基体的中央腔中有一对中央微管。近端中心粒和基体之间有中心粒间体将两者隔开。中段的袖套连接于细胞核之后,其中分布着线粒体和一些囊泡。近袖套内膜处的细胞质中有一层膜与袖套内膜平行。鞭毛细长,其起始端位于袖套腔中。鞭毛上长有两排侧鳍。侧鳍呈波纹状,分居轴丝两侧,大致与轴丝的两条中央微管同在一个平面上。侧鳍的基部有囊泡。  相似文献

小麦(Triticum aestivum )受精前成熟胚囊,除胚囊中央细胞的合点端细胞质中有酸性磷酸酶外,其余部位均未发现酸性磷酸酶。受精时期,以下部位存在酸性磷酸酶活性:卵细胞的细胞核内一部分染色质和细胞质中大部分线粒体;精、卵核融合时两核的核周腔内;退化助细胞合点端细胞质和一些液泡内;进入雌性细胞中的两个精核;胚囊各成员细胞的细胞壁及胚囊周围珠心细胞的细胞壁。二细胞原胚中未见有酸性磷酸酶。早期胚乳游离核染色质上有酸性磷酸酶。小麦受精过程酸性磷酸酶的分布特点可能与卵细胞生理状态的变化和细胞质中线粒体的改组、助细胞的退化、精核的生理状态以及精核与卵核的核膜融合等有关。  相似文献

Ultrastructural observations of the in utero sperm of Ascaris lumbricoides reveal that it consists of a relatively clear, ameboid anterior region and a conical posterior region containing numerous surface membrane specializations, dense mitochondria, a lipid-like refringent body of variable size, and a dense nucleus which lacks an apparent nuclear envelope. No acrosomal complex was observed. Pseudopods emanating from the anterior cytoplasm make first contact with the primary oocytes and appear to be responsible for the localized removal of the extraneous coat covering the oolemma. Subsequently the gamete membranes interdigitate and finally fuse. Because this pseudopodial action appears similar to that reported for the acrosomal filaments in flagellated sperm, the anterior region of the Ascaris sperm is thought to serve an acrosomal function. Following gamete-membrane fusion, the sperm nucleus acquires a particulate appearance and becomes disorganized. Once inside the oocyte, the sperm cytoplasm consists of dense mitochondria, ribosomes, and vesicles derived from the surface membrane specializations. The refringent body, whose contents possibly contribute to the synthesis of ribosomes, is usually absent by the time the sperm cytoplasm attains a central position in the egg.  相似文献

An earlier paper showed that in Saccoglossus the acrosomal tubule makes contact with the egg plasma membrane. The present paper includes evidence that the sperm and egg plasma membranes fuse to establish the single continuous zygote membrane which, consequently, is a mosaic. Contrary to the general hypothesis of Tyler, pinocytosis or phagocytosis plays no role in zygote formation. Contact between the gametes is actually between two newly exposed surfaces: in the spermatozoon, the surface was formerly the interior of the acrosomal vesicle; in the egg, it was membrane previously covered by the egg envelopes. The concept that all the events of fertilization are mediated by a fertilizin-antifertilizin reaction seems an oversimplification of events actually observed: rather, the evidence indicates that a series of specific biochemical interactions probably would be involved. Gamete membrane fusion permits sperm periacrosomal material to meet the egg cytoplasm; if an activating substance exists in the spermatozoon it probably is periacrosomal rather than acrosomal in origin. The contents of the acrosome are expended in the process of delivering the sperm plasma membrane to the egg plasma membrane. After these membranes coalesce, the sperm nucleus and other internal sperm structures move into the egg cytoplasm.  相似文献

The development of giant cells induced by the nematode Meloidogyne in tomato roots has been followed under controlled growth conditions and the ultrastructure and histochemistry of these structures have been examined. Entry of the nematode larvae into the roots took place within 24 hours; giant cell formation started on the 4th day and involved breakdown of the cell walls accompanied by thickening of a surrounding giant cell wall and an increase in density and area of the cytoplasm. The nuclei increased in number by simultaneous mitosis throughout a single giant cell. The peak of cytoplasmic density was reached after moulting and during egg production. The rate of protein synthesis in the giant cell is correlated with the rate of growth of the nematode. The giant cell wall is a thick, irregularly surfaced structure which contains all the normal polysaccharide components of a cell wall. The cytoplasm is rich in protein and RNA and contains mitochondria, proplastids, Golgi bodies, and a dense endoplasmic reticulum. The nuclei are large and irregular in shape and contain large nucleoli and a number of Feulgen-positive bodies scattered irregularly along the nuclear envelope. The nucleolus contains RNA and fat as well as Feulgen-positive granules which are revealed after treatment with ribonuclease. It consists of a dense outer cortex surrounding a much lighter central core and is connected at times with the Feulgen-positive bodies in the nucleus. Speculation is provided on the role of these bodies in cytoplasmic protein synthesis.  相似文献

Oocytes (future egg cells) of various animal groups often contain complex organelle assemblages (Balbiani bodies, yolk nuclei). The molecular composition and function of Balbiani bodies, such as those found in the oocytes of Xenopus laevis, have been recently recognized. In contrast, the functional significance of more complex and highly ordered yolk nuclei has not been elucidated to date. In this report we describe the structure, cytochemical content and evolution of the yolk nucleus in the oocytes of a common spider, Clubiona sp. We show that the yolk nucleus is a spherical, rather compact and persistent cytoplasmic accumulation of several different organelles. It consists predominantly of a highly elaborate cytoskeletal scaffold of condensed filamentous actin and a dense meshwork of intermediate-sized filaments. The yolk nucleus also comprises cisterns of endoplasmic reticulum, mitochondria, lipid droplets and other organelles. Nascent lipid droplets are regularly found in the cortical regions of the yolk nucleus in association with the endoplasmic reticulum. Single lipid droplets become surrounded by filamentous cages formed by intermediate filaments. Coexistence of the forming lipid droplets with the endoplasmic reticulum in the cortical zone of the yolk nucleus and their later investment by intermediate-sized filamentous cages suggest that the yolk nucleus is the birthplace of lipid droplets.  相似文献
