Conventional methods for histological preparation of degenerated myelin are time-consuming and difficult. The purpose of our study was to shorten the time required for the procedure and to obtain better quality results for light microscopic demonstration of degenerated myelin in the central and peripheral nervous systems by using microwave irradiation. Rat brain and sciatic nerve were used for the study. The middle cerebral artery was occluded and the sciatic nerve was cut to produce myelin degeneration. Marchi's method was used for staining degenerated myelin. Fixation for light microscopy that would take two days using the conventional procedure was completed in 16.5-18.5 min using microwave irradiation. While staining of degenerated myelin requires 10 days for the conventional Marchi method, we decreased it to 7 h for brain tissue and 1 h for sciatic nerve by using the microwave oven. Moreover, a better quality preparation was achieved in the groups stained under microwave irradiation than those prepared by the conventional method.  相似文献

Unlike F4-neuroprostanes (F4-NeuroPs), which are relatively selective in vivo markers of oxidative damage to neuronal membranes, there currently is no method to assess the extent of free radical damage to myelin with relative selectively. The polyunsaturated fatty acid adrenic acid (AdA) is susceptible to free radical attack and, at least in primates, is concentrated in myelin within white matter. Here, we characterized oxidation products of AdA as potential markers of free radical damage to myelin in human brain. Unesterified AdA was reacted with a free radical initiator to yield products (F2-dihomo-IsoPs) that were 28 Da larger than but otherwise closely resembled F2-isoprostanes (F2-IsoPs), which are generated by free radical attack on arachidonic acid. Phospholipids derived from human cerebral gray matter, white matter, and myelin similarly oxidized ex vivo showed that the ratio of esterified F2-dihomo-IsoPs to F4-NeuroPs was approximately 10-fold greater in myelin-derived than in gray matter-derived phospholipids. Finally, we showed that F2-dihomo-IsoPs are significantly increased in white matter samples from patients with Alzheimer's disease. We propose that F2-dihomo-IsoPs may serve as quantitative in vivo biomarkers of free radical damage to myelin from primate white matter.  相似文献

运用动力学方法研究了微波对青霉素酰化酶(pK1和pK2分别为5.69-6.06和11.56)催化反应性能的影响。结果显示:使用微波解冻档对青霉素酰化酶进行一定时间的预处理后,能够加速酶的水解反应。酶液的最适处理时间为15 s,微波处理后,酶的最适温度为从原来的37℃上升到40℃,操作稳定性基本不变。对最适微波条件处理后的青霉素酰化酶pH值依赖性催化反应进行研究,从logVm和log(Vm/Km)与pH值关系曲线计算得到该酶的pK1和pK2分别为5.66-6.55和11.05。  相似文献

In this work we analyzed variations in the expression of MBPs and P0 in ligated sciatic nerves of young and adult rats at 3, 7, and 14 days postligation (PL), by immunohistochemistry and SDS-PAGE of isolated myelin. A protein redistribution was seen in the distal stump of ligated nerves with the appearance of immunoreactive clusters. Using the KS400 image analyzer, immunostained area values were obtained from the different nerves dissected. In adult rats, there was an increase of the immunostained area for MBP from 3 to 7 days PL, coincident with a reorganization of the marker in clusters, followed by a marked decrease at 14 days. P0 immunolabeling gave similar results without, however, a decrease of the immunostained area at the longer survival time tested. Young animals showed an acceleration in the process of protein redistribution and digestion within ligated nerves, which followed a similar pattern as that of adult animals. Analysis by electrophoresis showed a marked decrease in P0 and MBP at 7 days PL in young rats and 14 days PL in adult rats. The functional significance of protein clustering within myelin in injured nerves deserves further analysis.  相似文献

Intracellular protein trafficking is tightly regulated, and improper trafficking might be the fundamental provocateur for human diseases including neurodegeneration. In neurons, protein trafficking to and from the plasma membrane affects synaptic plasticity. Voltage‐gated potassium channel 2.1 (Kv2.1) is a predominant delayed rectifier potassium (K+) current, and electrical activity patterns of dopamine (DA) neurons within the substantia nigra are generated and modulated by the orchestrated function of different ion channels. The pathological hallmark of Parkinson's disease (PD) is the progressive loss of these DA neurons, resulting in the degeneration of striatal dopaminergic terminals. However, whether trafficking of Kv2.1 channels contributes to PD remains unclear. In this study, we demonstrated that MPTP/MPP+ increases the surface expression of the Kv2.1 channel and causes nigrostriatal degeneration by using a subchronic MPTP mouse model. The inhibition of the Kv2.1 channel by using a specific blocker, guangxitoxin‐1E, protected nigrostriatal projections against MPTP/MPP+ insult and thus facilitated the recovery of motor coordination. These findings highlight the importance of trafficking of Kv2.1 channels in the pathogenesis of PD.

Shellfish have been identified as a potential source of Cryptosporidium infection for humans. The inactivation of C. parvum and other pathogens in raw molluscan shellfish would provide increased food safety for normal and at-risk consumers. The present study examined the efficacy of two alternative food-processing treatments, e-beam irradiation and microwave energy, on the viability of C. parvum oocysts in Eastern Oysters (Crassostrea virginica), which were artificially infected with the Beltsville strain of C. parvum. The effects of the treatments were evaluated by oral feeding of the processed oyster tissues to neonatal mice. Significant reductions (P<0.05) in infectivity were observed for in-shell and shucked oysters treated with e-beam irradiation at doses of 1.0, 1.5, or 2 kGy vs. untreated controls. A dose of 2 kGy completely eliminated C. parvum infectivity and did not adversely affect the visual appearance of the oysters. Oyster tissue treated with microwave exposures of 1 s (43.2 degrees C), 2 s (54.0 degrees C), and 3 s (62.5 degrees C) showed a reduction in C. parvum mouse infectivity, but the effects were not significantly different (P>0.05) from controls. Microwave energy treatments at 2 and 3 s showed extensive changes in oyster meat texture and color. Thus, because of lack of efficacy and unacceptable tissue changes, microwave treatment of oysters is not considered a viable food-processing method.  相似文献

The vacuolar degeneration of central myelin was produced in Sprague-Dawley rats by oral administration of triethyl tin. The wet weight of brain stems which seems to reflect the degree of accumulation of water increased during the administration of the toxin, whereas the activity of 2, 3-cyclic nucleotide 3-phosphodiesterase altered less remarkably. When TET was withdrawn from the drinking water, the rats showed a dramatic clinical improvement along with reduction in wet weight of brain stems. Treatment with acetazolamide following TET inhibited the clinical improvement and reduction in wet weight of brain stems. The present results indicates that central myelin has plasticity in recovering from the vacuolar degeneration by removing the accumulated fluid and carbonic anhydrase is possibly involved in the dehydration, of myelin in such a recovery phase.  相似文献

Treatment of p-tosyloxybenzaldehyde ( 1 ) with ethyl cyanoacetate afforded ethyl 2-cyano-3-(4-{[(4-methylphenyl)sulfonyl]oxy}phenyl)acrylate ( 2 ) which reacted with some active methylene derivatives under microwave irradiation in presence of ammonium acetate yielded pyridine derivatives 3 – 7 . On the other hand, when treatment of compound 1 with thiosemicarbazide gave 4-tosyloxybenzylidenethiosemicarbazone ( 8 ), which allowed to react with some active methylene compounds, such as: ethyl bromoacetate, chloroacetonitrile or phenacyl bromide derivatives gave thiazole derivatives 9 – 13 . The structure of all products were confirmed by elemental and spectroscopic analyses such as IR, 1H-NMR, 13CNMR and mass spectra. The advanced of this method are short reaction time (3–7 min), excellent yield, pure products, and low-cost processing. In the final category, the toxicological characteristics of all compounds were tested towards Saissetia oleae (Olivier, 1791) (Hemiptera: Coccidae). With respect to the LC50 values. It has been found that compound 3 possesses the highest insecticidal bioefficacy compared with other products, with values of 0.502 and 1.009 ppm, for nymphs and adults female, respectively. This study paves the way towards discovering new materials for potential use as insecticidal active agents.  相似文献

以深黄被孢霉(Mortierella isabellina As3.3410)为出发菌株,经微波诱变和紫外诱变,乙酰水杨酸与低温(15℃)相结合的筛选方法,获得1株高产多不饱和脂肪酸菌株A35-4,其生物量为17.9 g/L,油脂含量为67.8%,油脂产量为12.12 g/L,PUFAs含量为20.3%,PUFAs产量为2.46 g/L,上述指标比原始菌株A0分别增加32.6%、49.8%、98.69%、14.0%和125.7%。连续斜面传代培养证实该菌株具有较好的遗传稳定性。本研究为直接利用该菌株生产PUFAs以及克隆高效PUFAs相关基因,创造高含PUFAs转基因植物材料奠定基础。  相似文献

黑水虻Hermetia illucens(L.)是一种重要的资源昆虫。本文旨在筛选出适合黑水虻血细胞观察的染色方法,明确黑水虻血细胞类型、数量及组成,为黑水虻血细胞免疫研究奠定基础。采用Giemsa和Giemsa-Wright's染色方法和血球计数板法,对黑水虻血细胞染色方法和血细胞数量及形态进行研究。结果表明,甲醇固定4 min,Giemsa-Wright's染液染色9 min、pH 7.2磷酸盐缓冲液分色10 min是黑水虻幼虫血细胞最佳染色方法;黑水虻幼虫血细胞包括原血细胞、浆血细胞、粒血细胞、类绛血细胞、珠血细胞5类;4龄黑水虻幼虫血细胞数量大约为2917个/μL,其中浆血细胞占53.20%±2.78%,粒血细胞占37.49%±3.96%,原血细胞占7.97%±1.51%,类绛血细胞占1.02%±0.24%,珠血细胞占0.62%±0.08%。Giemsa-Wright's染色法为黑水虻幼虫血细胞最佳染色方法,黑水虻幼虫血细胞可分为5类10种。  相似文献

微波辐射浸软法是一种简单、快速制备高分辨植物扫描电镜样品的方法。经冰冻断裂处理后的样品,利用断续微波辐射浸软,既可阻止浸软溶液温度陡然升高,不使植物结构受到破坏,又可使浸软时间显著缩短,由原来的72小时变为30分钟。因为断续的微波辐射能保持较低的炉内温度,所以它不仅能很好地适用于扫描电镜样品制备过程中的浸软,而且也适用于样品固定、导电染色等。此法大大缩短了O-D-O法的制样周期。  相似文献

Mice heterozygously deficient in the p0 gene (P0(+/-)) are animal models for some forms of inherited neuropathies. They display a progressive demyelinating phenotype in motor nerves, accompanied by mild infiltration of lymphocytes and increase in macrophages. We have shown previously that the T lymphocytes are instrumental in the demyelination process. This study addresses the functional role of the macrophage in this monogenic myelin disorder.In motor nerves of P0(+/)- mice, the number of macrophages in demyelinated peripheral nerves was increased by a factor of five when compared with motor nerves of wild-type mice. Immunoelectron microscopy, using a specific marker for mouse macrophages, displayed macrophages not only in the endoneurium of the myelin mutants, but also within endoneurial tubes, suggesting an active role in demyelination. To elucidate the roles of the macrophages, we crossbred the myelin mutants with a spontaneous mouse mutant deficient in macrophage colony-stimulating factor (M-CSF), hence displaying impaired macrophage activation. In the P0-deficient double mutants also deficient in M-CSF, the numbers of macrophages were not elevated in the demyelinating motor nerves and demyelination was less severe. These findings demonstrate an active role of macrophages during pathogenesis of inherited demyelination with putative impact on future treatment strategies.  相似文献

Herein, we make an effort to enhance the antimicrobial activity of levofloxacin (LVX) antibiotic via conjugation to a cell‐penetrating peptide (CPP) including Cys‐Gly‐Ala‐Phe‐Pro‐His‐Arg. For this purpose, cysteine is used as a linker between the LVX and CPP chain, and two heterogeneous nanoscale catalytic systems are employed as the substantial alternatives for traditional peptide coupling reagents like N,N,N′,N′‐tetramethyl‐O‐(benzotriazol‐1‐yl)uronium tetrafluoroborate (TBTU). Briefly, it has been found out that the antimicrobial potency of the synthesized CPP‐LVX conjugate (on the gram‐positive and gram‐negative bacteria) is noticeably enhanced (~20% more). It has been revealed via zone of inhibition (ZOI) and optical density (OD) evaluations. As a convenient method for making this type of the effective conjugations, ultrasound waves (with a specific frequency and power density) activate the catalytic sites of the heterogeneous nanoparticles. Through this synergistic effect, peptide/amide bond is formed during a short time (10 min), and high reaction yield (>90%) is obtained under mild conditions. Moreover, as a simple purification process, the catalyst nanoparticles are collected and separated through their high magnetic property.  相似文献

采用微波炉加热,在敞口容器中,进行甲壳素脱乙酰反应,制备壳聚糖。考察了碱溶液浓度和微波加热时间对壳聚糖脱乙酰度的影响。固定微波加热时间30min,随NaOH溶液浓度增加,脱乙酰度先增加,后减小;NaOH溶液浓度为45%时,壳聚糖的脱乙酰度最高。固定NaOH溶液浓度为45%,随着微波加热时间延长,壳聚糖的脱乙酰度增加。微波加热的最佳时间为30min。加热时间继续延长,壳聚糖变黑。碱溶液浓度和微波加热时间对壳聚糖的粘均分子量影响都不大。本文试图从微波场的能量分布和微波加热机理方面解释实验结果。  相似文献
