Endogenous levels of gibberellins, auxins and cytokinins incrown gall tissues of Catharanthus roseus G. Don. (Vinca roseaL.) of two different tumor types, induced by a nopaline-typeand an octopine-type Ti-plasmid, were examined. The levels of gibberellins were higher in nopaline-type V208cells than in octopine-type V277 cells. GA12 and GA24 were identifiedfrom V208 cells. The level of IAA was 20-fold higher in V208cells than in V277 cells. The level of trans-ribosylzeatin washigher in V277 cells than in V208 cells, whereas trans-zeatinwas present at higher levels in V208 cells than in V277 cells. Our results revealed that the production of gibberellins, aswell as of IAA and cytokinins, in the crown gall tissues ofCatharanthus roseus differed between the octopine- and nopaline-typetumors, even though the crown galls apparently grow as unorganizedaggregates of cells in both cases. 4Present address: Facultad de Ciencias, Pontificia UniversidadJaveriana, Bogota, Colombia. The application of 8[14C]t-zeatin to the cotyledons of germinatingbean seeds demonstrated that cytokinins are not readily exportedfrom the cotyledons to the embryonic axis during the early stagesof this process. In the cotyledons the applied zeatin is metabolizedextensively to metabolites which are polar and which occur atRF 0·2–0·5 on paper chromatograms. Thesemetabolites are stable and are not readily exported from thecotyledons. In contrast the metabolites found at RF 0–0·2are more readily exported. When exported to the radicles andplumules a large proportion of the translocated metaboliteswere converted to compounds which on paper co-chromatographedwith zeatin. This seems to suggest that the embryonic axis hasthe capacity to synthesize cytokinins and that some of the metabolitesformed during its catabolism can also be used for its synthesis. Identification by full-scan GC-MS revealed that [2H6]-teasteronefed to suspension cultured cells of Marchantia polymorpha wasconverted to [2H6]3-dehydroteasterone and [2H6]typhasterol.This indicates that the cells carry out a C3-epimerization inwhich teasterone is converted to typhasterol via 3-dehydroteasterone.In vitro enzymatic conversions of teasterone to typhasterolwere also investigated. A crude cytosolic solution preparedfrom Marchantia cells catalyzed not only the dehydrogenationof teasterone to 3-dehydroteasterone but also the reductionof 3-dehydroteasterone to typhasterol. The major 4-demethysterolin cultured M. polymorpha cells was 24-methylcholesterol, theprecursor of brassinosteroids. These results suggest that enzymessimilar to those involved in the early C-6 oxidation pathwayof the brassinosteroid biosynthesis are present in the liverwort. The rate of net photosynthesis (P) of whole plant stands oftomato (Lycopersicon esculentum Mill.), cucumber (Cucumis sativusL.) and sweet pepper (Capsicum annuum L.) was measured in sixlong-term experiments in large greenhouses under normal operatingconditions and CO2-concentrations between 200 and 1200 µmolmol-1. The objective was to quantify the responses to lightand carbon dioxide and to obtain data sets for testing simulationmodels. The method of measuring canopy photosynthesis involvedan accurate estimation of the greenhouse CO2 balance, usingnitrous oxide (N2O) as tracer gas to determine, on-line, theexchange rate between greenhouse and outside air. The estimatedrelative error in the observed P was about ± 10%, exceptthat higher relative errors could occur under particular conditions. A regression equation relating P to the photosynthetically activeradiation, the CO2 concentration and the leaf area index explained83-91% of the variance. The main canopy photosynthesis characteristicscalculated with the fitted regression equations were: canopyPmax 5-9 g m-2 h-1 CO2 uptake; ratio Pmax/LAI 1·5-3 gm-2 h-1; light compensation point 32-86 µmol s-1 m-2;light use efficiency (quantum yield) at low light 0·06-0·10µmol µmol-1 and CO2 compensation point 18-54 µmolmol-1. The capacity for C4 photosynthesis in Panicum milioides, a specieshaving reduced levels of photorespiration, was investigatedby examining the activity of certain key enzymes of the C4 pathwayand by pulse-chase experiments with 14CO2. The ATP$P1 dependentactivity of pyruvate,P1 dikinase in the species was extremelylow (0.14–0.18 µmol mg chlorophyll–1 min–1).Low activity of the enzyme was also found in Panicum decipiensand Panicum hians (related species with reduced photorespiration)and in Panicum laxum (a C3 species). The antibody to pyruvate,P1dikinase caused about 70% inhibition of the ATP$P1 dependentactivity of the enzyme in P. milioides. The activity of NAD-malicenzyme and NADP-malic enzyme in P. milioides was equally low(approximately 0.1–0.2 µmol mg chlorophyll–1min–1) and similar to the activity in P. decipiens, P.hians and P. laxum. Photosynthetic pulse-chase experiments underatmospheric conditions showed a typical C3-like pattern of carbonassimilation including the labelling of glycine and serine asexpected during photorespiration. During the pulse with 14CO2only about 1% of the labelled products appeared in malate and2–3% in aspartate. During a chase in atmospheric levelsof CO2 for up to 6 min there was a slight increase in labellingin the C4 acids. The amount of label in carbon 4 of aspartatedid not change during the chase, indicating little or no turnoverof the C4 acid via decarboxylation. The results indicate thatunder atmospheric conditions P. milioides assimilates carbondirectly through the C3 pathway. Oikopleura longicauda occurred throughout the year in ToyamaBay, southern Japan Sea, and analysis of its size compositionand maturity revealed that reproduction was continuous overtheyear. Somatic growth production (Pg) varied with season from0.03 to 103 mg carbon (C) m–2day–1 (annual Pg 4.5g C m–2), and house production (Pe) from 0.11 to 266 mgC m–2 day–1 (annualPe 11.3 g C m–2). The annualPg/B ratio was 176. Compared with production data of some predominantzooplankton species in Toyama Bay, it is suggested that despitetheir smaller biomass, appendicularians are an important secondaryproducer.  相似文献

The effects of various plant growth regulators and that of pHon the in vitro growth and development of young inflorescencesof Brassica napus L. cv. Westar were examined. A cytokinin wasrequired for normal maturation of floral buds, including thecompletion of microsporogenesis, and it stimulated the initiationof additional buds on the inflorescence axis. Benzylaminopurine(BAP) was the most effective of the cytokinins tested. Gibberellicacid (GA3) and naphthaleneacetic acid (NAA) alone were ineffective.In combination with BAP, both reduced the positive influenceof the cytokinin but GA3 was more inhibitory than NAA. At alow initial pH (3.9–4.6), the percentage of cultures whichproduced normal buds was significantly higher, especially inthe presence of 10-7 M or 5 ? 10-7 M BAP, in comparison to cultureswith a pH of 5.3-6.0, the standard range for plant tissue culture.  相似文献

Photosynthetic rates of outdoor-grown soybean (Glycine max L.Merr. cv. Bragg) canopies increased with increasing CO2 concentrationduring growth, before and after canopy closure (complete lightinterception), when measured over a wide range of solar irradiancevalues. Total canopy leaf area was greater as the CO2 concentrationduring growth was increased from 160 to 990 mm3 dm–3.Photosynthetic rates of canopies grown at 330 and 660 mm3 CO2dm–3 were similar when measured at the same CO2 concentrationsand high irradiance. There was no difference in ribulose bisphosphatecarboxylase/oxygenase (rubisco) activity or ribulose 1,5-bisphosphate(RuBP) concentration between plants grown at the two CO2 concentrations.However, photosynthetic rates averaged 87% greater for the canopiesgrown and measured at 660 mm3 CO2 dm–3. A 10°C differencein air temperature during growth resulted in only a 4°Cleaf temperature difference, which was insufficient to changethe photosynthetic rate or rubisco activity in canopies grownand measured at either 330 or 660 mm3 CO2 dm–3. RuBP concentrationsdecreased as air temperature during growth was increased atboth CO2 concentrations. These data indicate that the increasedphotosynthetic rates of soybean canopies at elevated CO2 aredue to several factors, including: more rapid development ofthe leaf area index; a reduction in substrate CO2 limitation;and no downward acclimation in photosynthetic capacity, as occurin some other species. The utilization of inorganic carbon and role of the coccolithswere investigated in intact cells and protoplasts of a marineunicellular calcareous alga, Emiliania huxleyi. Protoplastswith high photosynthetic activity were obtained by artificialdecalcification with 50 mM MES-NaOH (pH5.5). (1) The kineticsof the photosynthetic evolution of O2 at various concentrationsof externally added NaHCO3 were the same for intact cells andprotoplasts, indicating that the kinetic properties with respectto dissolved inorganic carbon (DIC) were not affected by thepresence or absence of the coccoliths on the cell surface. Double-reciprocalplots and plots of the concentration of substrate divided byvelocity (s/v) against the concentration of substrate (s) werebiphasic in the case of both intact cells and protoplasts. TheCO2-utilization reaction was, therefore, considered to involvetwo processes with different values of Km and Vmax. From thekinetic analyses, Km and Vmax [µmoles O2 (ml PCV)–1h–1] were deduced to be 92 µM and 76.3 for a "low-Km"reaction and 4.1 mM and 252 for a "high-Km" reaction, respectively.(2) In short-term (40-min) experiments, time courses of thetotal uptake of 14C-DIC and the incorporation of 14C into acid-stableproducts of photosynthesis and the internal pool of DIC, determinedas acid-labile compounds, under CO2-limiting conditions (80µM) were very similar for intact cells and protoplasts.However, incorporation of 14C into CaCO3 apparently occurredmore slowly in protoplasts than in intact cells. (3) In longterm (24-h) experiments, patterns of incorporation of 14C werealmost same for intact cells and protoplasts, with the exceptionthat the amount of 14C incorporated into CaCO3 was much smallerin the former than the latter. The production of Ca14CO3 increasedduring the course of 10 h after a 4-h lag. However, after 10h the level of Ca14CCO3 started to decrease. The decrease wasaccompanied by an increase in 14C in the products of photosynthesis,suggesting that CaCO3 was reutilized for the photosyntheticfixation of CO2 and, therefore, that the coccoliths functionas sites of storage of DIC. The addition of Braun and Wood's inorganic supplements (845mg l–1 KCl, 1800 mgl–1 NaNO3, 300 mg l–1 NaH2PO4.2H2O,790 mg l–1 (NH4)2SO4) to White's medium caused markedincreases in the growth of normal tissues of Helianthus annuus,Nicotiana rustica, Daucus carota, and Vinca rosea and crown-galltumour tissues of H. annuus. However, no evidence was obtainedwhich suggested that the presence of these extra salts markedlyinfluenced the essential requirements of normal callus for auxinsand kinetin. In contrast their presence significantly influencedthe hormonal requirements of certain habituated cultures ofH. annuus and V. rosea. These habituated cultures had specificauxin requirements on White's medium while either an auxin orkinetin was sufficient on high-salts medium. These results arediscussed in relation to previous reports which suggested thatthe biosyntheses of auxins and other growth factors in normaland crown-gall cultures are specifically activated by certaininorganic ions.  相似文献

Plants of four isolines of soyabean [Glycine max(L.) Merrill]‘Clark’, viz‘L71-920’ (maturity genecomplemente1e2e3 ), ‘L80-5914’ (E1e2e3), ‘Clark’(e1E2E3), and ‘L65-3366’ (E1E2E3), were grown inshort (12.25 h d - 1natural light) and long days (12.25 h d- 1natural light supplemented with 2.75 h d - 1low-irradianceartificial light) from first flowering to maturity in a polythenetunnel maintained at 30/24°C (day/night). Whereas therewere few differences among the isolines grown in short days,in long days the dominant alleles increased crop duration, biomassand seed yield substantially. Increases in biological and economicyield were not solely a consequence of longer crop duration:the dominant alleles also increased crop growth rate and radiationuse efficiency in long days (from 1.3 g MJ - 1total radiationine1e2e3 to 2.8 g MJ - 1inE1E2E3 ). Greater radiation use efficiencyresulted from a relatively longer leaf area duration, betterdistribution and orientation of a larger mass of leaves withinthe canopy, and smaller partitioning of assimilates to reproductivestructures. The work reveals the substantial effects of thethree lociE1 / e1, E2/ e2and E3/e3 on the response of plantgrowth, as well as development, to environment. The cell quotas of microcystin (Qmcyst), protein (Qprot), chlorophylla (Qchloro) and carbohydrate (Qcarbo), as well as the net productionrates of these parameters, were determined during the exponentialand stationary phases in nine batch cultures of Microcystisaeruginosa (CYA 228) at light regimes from 33 to 53 µmolphotons m–2 s–1. The following results were obtained.(i) A parallel pattern was found in the changes of Qmcyst, Qprot,Qchloro and Qcarbo during the entire growth cycle and significantcorrelations were recorded between Qmcyst and Qprot, Qchloroand Qcarbo. (ii) The net microcystin production rate (µmcyst)was positively correlated with the specific cell division rate(µc), the chlorophyll production rate (µchloro)and the protein production rate. (iii) A significant inverselinear relationship was found between µc and Qmcyst, i.e.cultures with a positive µc had a Qmcyst between 110 and400 fg microcystin cell–1, while declining cultures hadQmcyst values >400 fg microcystin cell–1. Maximum variationin Qmcyst within cultures was 3.5-fold. Collectively, the resultsshow that cells produced microcystin at rates approximatingthose needed to replace losses to daughter cells during divisionand that microcystin was produced in a similar way to proteinand chlorophyll, indicating a constitutive microcystin production.  相似文献

Pulmonary vasoconstriction and vascularmedial hypertrophy greatly contribute to the elevated pulmonaryvascular resistance in patients with pulmonary hypertension. A rise incytosolic free Ca2+ ([Ca2+]cyt)in pulmonary artery smooth muscle cells (PASMC) triggers vasoconstriction and stimulates cell growth. Membrane potential (Em) regulates[Ca2+]cyt by governing Ca2+influx through voltage-dependent Ca2+ channels. Thusintracellular Ca2+ may serve as a shared signaltransduction element that leads to pulmonary vasoconstriction andvascular remodeling. In PASMC, activity of voltage-gated K+(Kv) channels regulates resting Em. In thisstudy, we investigated whether changes of Kv currents[IK(V)], Em, and[Ca2+]cyt affect cell growth by comparingthese parameters in proliferating and growth-arrested PASMC. Serumdeprivation induced growth arrest of PASMC, whereas chelation ofextracellular Ca2+ abolished PASMC growth. Resting[Ca2+]cyt was significantly higher, andresting Em was more depolarized, inproliferating PASMC than in growth-arrested cells. Consistently, wholecell IK(V) was significantly attenuated in PASMCduring proliferation. Furthermore, Emdepolarization significantly increased resting[Ca2+]cyt and augmented agonist-mediatedrises in [Ca2+]cyt in the absence ofextracellular Ca2+. These results demonstrate that reducedIK(V), depolarized Em, and elevated [Ca2+]cyt may play a criticalrole in stimulating PASMC proliferation. Pulmonary vascular medialhypertrophy in patients with pulmonary hypertension may be partlycaused by a membrane depolarization-mediated increase in[Ca2+]cyt in PASMC.

In cardiac-specific Na+-Ca2+ exchanger (NCX) knockout (KO) mice, the ventricular action potential (AP) is shortened. The shortening of the AP, as well as a decrease of the L-type Ca2+ current (ICa), provides a critical mechanism for the maintenance of Ca2+ homeostasis and contractility in the absence of NCX (Pott C, Philipson KD, Goldhaber JI. Excitation-contraction coupling in Na+-Ca2+ exchanger knockout mice: reduced transsarcolemmal Ca2+ flux. Circ Res 97: 1288–1295, 2005). To investigate the mechanism that underlies the accelerated AP repolarization, we recorded the transient outward current (Ito) in patch-clamped myocytes isolated from wild-type (WT) and NCX KO mice. Peak Ito was increased by 78% and decay kinetics were slowed in KO vs. WT. Consistent with increased Ito, ECGs from KO mice exhibited shortened QT intervals. Expression of the Ito-generating K+ channel subunit Kv4.2 and the K+ channel interacting protein was increased in KO. We used a computer model of the murine AP (Bondarenko VE, Szigeti GP, Bett GC, Kim SJ, and Rasmusson RL. Computer model of action potential of mouse ventricular myocytes. Am J Physiol Heart Circ Physiol 287: 1378–1403, 2004) to determine the relative contributions of increased Ito, reduced ICa, and reduced NCX current (INCX) on the shape and kinetics of the AP. Reduction of ICa and elimination of INCX had relatively small effects on the duration of the AP in the computer model. In contrast, AP repolarization was substantially accelerated when Ito was increased in the computer model. Thus, the increase in Ito, and not the reduction of ICa or INCX, is likely to be the major mechanism of AP shortening in KO myocytes. The effect of -adrenergic stimulation on cardiac Na+/Ca2+ exchange has been controversial. To clarify the effect, we measured Na+/Ca2+ exchange current (INCX) in voltage-clamped guinea pig, mouse, and rat ventricular cells. When INCX was defined as a 5 mM Ni2+-sensitive current in guinea pig ventricular myocytes, 1 µM isoproterenol apparently augmented INCX by 32%. However, this increase was probably due to contamination of the cAMP-dependent Cl current (CFTR-Cl current, ICFTR-Cl), because Ni2+ inhibited the activation of ICFTR-Cl by 1 µM isoproterenol with a half-maximum concentration of 0.5 mM under conditions where INCX was suppressed. Five or ten millimolar Ni2+ did not inhibit ICFTR-Cl activated by 10 µM forskolin, an activator of adenylate cyclase, suggesting that Ni2+ acted upstream of adenylate cyclase in the -adrenergic signaling pathway. Furthermore, in a low-extracellular Cl bath solution, 1 µM isoproterenol did not significantly alter the amplitude of Ni2+-sensitive INCX at +50 mV, which is close to the reversal potential of ICFTR-Cl. No change in INCX amplitude was induced by 10 µM forskolin. When INCX was activated by extracellular Ca2+, it was not significantly affected by 1 µM isoproterenol in guinea pig, mouse, or rat ventricular cells. The effect of photosynthetic photon flux density (PPFD) on carboxylationefficiency, estimated as the initial slope (IS) of net CO2 assimilationrate versus intercellular CO2 partial pressure response curve,as well as on ribulose-1, 5-bisphosphate carboxylase (Rubisco)activation was measured in Trifolium subterraneum L. leavesunder field conditions. The relationship between IS and PPFDfits a logarithmic curve. Rubisco activation accounts for theIS increase only up to a PPFD of 550 µmol photons m-2s-1. Further IS increase, between 550 and 1000 µmol photonsm-2 s-1, could be related to a higher ribulose fcwphosphate(RuBP) availability. The slow, but sustained IS increase above1000 µmol photons m-2 s-1 could be explained by the mesophyllCO2 diffusion barriers associated with the high chlorophylland protein content in field developed leaves. Rates of CO2 and HCC3 fixation in cells of various Chlorellaspecies in suspension were compared from the amounts of 14Cfixed during the 5 s after the injection of a solution containingonly 14CO2 or H14CO3. Results indicated that irrespectiveof the CO2 concentration during growth, Chlorella vulgaris 11h and C. miniata mainly utilized CO2, whereas C. vulgaris C-3,C. sp. K. and C. ellipsoidea took up HCO3 in additionto CO2. Cells of C. pyrenoidosa that had been grown with 1.5%CO2 (high-CO2 cells) mainly utilized CO2, whereas those grownwith air (low-CO2 cells) utilized HCO3 in addition toCO2. Cells that utilized HCO3 had carbonic anhydrase(CA) on their surfaces. The effects of Diamox and CA on the rates of CO2 and HCO3fixation are in accord with the inference that HCO3 wasutilized after conversion to CO2 via the CA located on the cellsurface. CA was found in both the soluble and insoluble fractions;the CA on the cell surface was insoluble. Independent of the modes of utilization, the apparent Km (NaHCO3)for photosynthesis was much lower in low-CO2 cells than in high-CO2ones. The role of nitric oxide (NO) in the occurrence of intracellular Ca2+ concentration ([Ca2+]i) oscillations in pituitary GH3 cells was evaluated by studying the effect of increasing or decreasing endogenous NO synthesis with L-arginine and nitro-L-arginine methyl ester (L-NAME), respectively. When NO synthesis was blocked with L-NAME (1 mM) [Ca2+]i, oscillations disappeared in 68% of spontaneously active cells, whereas 41% of the quiescent cells showed [Ca2+]i oscillations in response to the NO synthase (NOS) substrate L-arginine (10 mM). This effect was reproduced by the NO donors NOC-18 and S-nitroso-N-acetylpenicillamine (SNAP). NOC-18 was ineffective in the presence of the L-type voltage-dependent Ca2+ channels (VDCC) blocker nimodipine (1 µM) or in Ca2+-free medium. Conversely, its effect was preserved when Ca2+ release from intracellular Ca2+ stores was inhibited either with the ryanodine-receptor blocker ryanodine (500 µM) or with the inositol 1,4,5-trisphosphate receptor blocker xestospongin C (3 µM). These results suggest that NO induces the appearance of [Ca2+]i oscillations by determining Ca2+ influx. Patch-clamp experiments excluded that NO acted directly on VDCC but suggested that NO determined membrane depolarization because of the inhibition of voltage-gated K+ channels. NOC-18 and SNAP caused a decrease in the amplitude of slow-inactivating (IDR) and ether-à-go-go-related gene (ERG) hyperpolarization-evoked, deactivating K+ currents. Similar results were obtained when GH3 cells were treated with L-arginine. H+ translocation driven by NO3, NO2 and N2O reductionswith endogenous substrates in cells of Rhodopseudomonas sphaeroidesforma sp. denitrificans was investigated by the oxidant pulsemethod. Upon injection of nitrogenous oxides to anaerobic cellsin darkness, an alkaline transient in the external medium wasobserved, followed by acidification. The alkaline transientwas enhanced by carbonyl cyanide m-chlorophenylhydrazone. When a viologen dye was used as an electron donor in the presenceof 1 mM Af-ethylmaleimide and 0.1 mM 2-n-heptyl-4-hydroxyquinoline-N-oxideto preclude respiration-linked H+ extrusion, addition of KNO3,KNO2 and N2O caused only a rapid alkalinization. The H+ consumptionstoichiometries, H+/2e ratios for NO3 reductionto NO2, NO2 reduction to 1/2 N2O and N2O reductionto N2 were –1.90, –3.18 and –2.04, respectively.These values agreed well with the fact that all reductions ofnitrogenous oxides in denitrification occur on the periplasmicside of the cytoplasmic membrane. When corrected for H+ consumption in the periplasm, the H+ extrusionstoichiometries, H+/2e ratios with endogenous substratesin the presence of K+/valinomycin for NO3 reduction toNO2, NO2 reduction to 1/2 N2O and N2O reductionto N2 were 4.05, 4.95 and 6.01, respectively. Etiolated seedling tissues of aseptically grown squash (Cucurbitamaxima Duch) contain indole-3-acetamide (IAM) as a natural endogenouscompound, conclusively identified by gas chromatography-massspectrometry (GC-MS). Roots of aseptically raised seedlingsalso contain amide hydrolysing activity, which converts IAMto IAA, indoleacetonitrile (IAN) to IAM and IAA, and 1-naphthaleneacetamideto 1-naphthaleneacetic acid. This activity was enriched 48-foldby fractional precipitation with ammonium sulphate, Sephadexgel nitration and anion exchange chromatography. Being hydrolytic,it works equally well in air and in vacuo, without added cofactors.The partially purified enzyme works optimally between pH 7 and7.5, and a Km value of 80 µM was calculated with IAM asthe substrate. The product of this reaction was definitivelyidentified as IAA by GC-MS. The temperature optimum of thisamidohydrolase lies around 45°C, and it is stable to freezing.A comparison of its properties with the amidohydrolase of Agrobacteriumor crown gall tissue, shows it to be different. In view of thenatural occurrence of both IAM and the amidohydrolase, it issuggested that the IAM pathway of IAA biogenesis is feasiblein etiolated squash seedlings. 4Deceased 2/2-1993.  相似文献
