The nitrogen requirements of 96Gluconobacter, 55Acetobacter and 7Frateuria strains were examined. Only someFrateuria strains were able to grow on 0.5% yeast extract broth or 0.5% peptone broth. In the presence ofd-glucose ord-mannitol as a carbon source, ammonium was used as the sole source of nitrogen by all three genera. With ethanol, only a fewAcetobacter strains grew on ammonium as a sole nitrogen source. Singlel-amino acids cannot serve as a sole source of carbon and nitrogen for growth ofGluconobacter, Acetobacter orFrateuria. The singlel-amino acids which were used by most strains as a sole nitrogen source for growth are: asparagine, aspartic acid, glutamine, glutamic acid, proline and alanine. SomeAcetobacter andGluconobacter strains deaminated alanine, asparagine, glutamic acid, threonine, serine and proline. NoFrateuria strain was able to develop on cysteine, glycine, threonine or tryptophan as a sole source of nitrogen for growth. An inhibitory effect of valine may explain the absence of growth on this amino acid. No amino acid is “essential” forGluconobacter, Acetobacter orFrateuria.  相似文献

The limits of a male moth's ability to resolve closely spaced odor filaments have been investigated. Male Helicoverpa zea normally respond to their conspecific sex pheromone blend by exhibiting an upwind flight, which culminates in source contact by at least 50% of the bioassayed individuals. When loaded onto the same filter paper source containing this hitherto attractive pheromone blend, or onto a separate filter paper and co-emitted from the same pipette source with pheromone, (Z)-11-hexadecenyl acetate severely reduced upwind flight and source contact by male H. zea. A similar level of upwind flight inhibition was recorded when the antagonist (Z)-11-hexadecenyl acetate was emitted from its own point source placed 1 mm upwind of the pheromone point source, both plumes being simultaneously emitted in a continuous mode to form a confluent strand. However, (Z)-11-hexadecenyl acetate was less effective in reducing upwind flight and source contact when it was isolated and pulsed from its own source, placed 1 mm either upwind, downwind or cross-wind of a pipette source from which pheromone was simultaneously being pulsed, such that both filaments were separated in time by 0.001–0. 003 s. These results suggest that male H. zea are able to distinguish between odor sources separated by as little as 1 mm in space and 0.001 s in time. Accepted: 31 March 1999  相似文献

According to thereciprocity law, the total dose absorbed in a specimen irradiated by a point source can be inferred from the reciprocal situation, where the absorber is replaced by a source of the same shape and the radiation is measured at the position of the original point source. In this paper, for a spherical source of homogeneous radioactive material with absorption coefficient μ1, we calculate the intensity of radiation (or flux) at a pointP, as well as the intensity which would be found at the same point if μ1 were zero. The ratio between these two quantities is denoted byk. We assume throughout that absorption follows the simple exponential law. Self-absorption can also be characterized by the numberv, which is defined as the ratio between the total radiation current flowing through the surface of the source and the total current obtained if there were no absorption. For a homogeneous spherical source in a nonabsorbing medium it is shown that the quantityk becomes equal tov if the distance between the pointP and the center of the sphere is sufficiently large. This fact was used by W. K. Sinclair in estimating an absorption coefficient from his experimental results. We also derive an expression for the self-absorption ratio,v, of acylindrical source. In the limiting case of an infinitely long cylinder our result coincides with an equation previously found by S. Kushneriuk. Fumarate was previously known to serve as an anaerobic electron acceptor by E. coli when colonizing the mammalian intestine, but the source of that fumarate was elusive. In this issue, Unden and coworkers demonstrate that l -aspartic acid is the source of fumarate that drives anaerobic respiration by colonized E. coli (Schubert et al., 2021). Moreover, Schubert et al., establish that E. coli is able to grow anaerobically by using aspartate as a sole source of nitrogen. These groundbreaking findings indicate that a single amino acid – aspartate – supports anaerobic respiration and acquisition of nitrogen by E. coli in the intestine.  相似文献

The phototrophic bacterium Rhodobacter capsulatus utilizes the aromatic amino acids L-phenylalanine and L-tyrosine as nitrogen source. L-Phenylalanine is hydroxylated to L-tyrosine, which is further converted into p-hydroxyphenyl pyruvate (pHPP) by a transamination reaction. The bacterium is unable to grow at the expense of these amino acids as the sole carbon source, although it is able to degrade them to homogentisate, probably by unspecific hydroxylation reactions. Metabolization of L-phenylalanine or L-tyrosine as nitrogen source requires phototrophic growth conditions and does not produce free ammonium inside the cells. A low aminotransferase activity with 2-oxoglutarate and L-tyrosine as substrates can be detected in crude extracts of R. capsulatus. Uptake of both amino acids by R. capsulatus was completely inhibited by ammonium addition, which also prevents aminotransferase induction. Received: 21 July 1998 / Accepted: 19 August 1998  相似文献

The pathway by which glutamate is degraded as a carbon source has not previously been elucidated, but enzymatic analysis of Rhizobium meliloti CMF1 indicated that both glutamate dehydrogenase (GDH) and gamma-aminobutyrate (GABA) bypass activities were present in free living cells. However, when similar studies were performed on R. meliloti CMF1 bacteroids, isolated from alfalfa nodules, only GABA bypass activities were detectable. Both GDH and GABA bypass activities were influenced by the carbon source provided, with maximum activities being detected when glutamate was present as sole carbon and nitrogen source. Addition of a second carbon source, such as succinate, to the growth medium did not influence GDH activity but substantially decreased levels of the first enzyme of the GABA bypass, glutamate decarboxylase (GDC). Cyclic adenosine 3′5′-monophosphate (cAMP) failed to increase GDC activities in R. meliloti CMF1 cells grown in the presence of an additional carbon source. It is proposed that the GABA bypass is a major mechanism of glutamate carbon degradation in R. meliloti CMF1, a system whose enzymatic activities are influenced by the nature of the carbon source present in the growth environment.  相似文献

Summary In a laboratory incubation study the humic acid isolated from a forest soil of Palamau (Bihar) was subjected to biodegradation for a period of six weeks by using nine cultures of fungi. These fungi were tested earlier for their cellulose decomposing ability. The humic acid was used as sole source of carbon, nitrogen and carbon plus nitrogen in Czapek-Dox broth. Of the nine culturesAspergillus awamori (IARI),Penicillium sp. (Ranchi),Humicola insolense (Hissar) were found to be very effective in decomposing humic acid. The humic acid used as sole source of carbon was most efficiently degraded followed by that used as carbon+nitrogen source. When it was used as sole source of nitrogen, it could not be degraded so efficiently. This may be due to unavailability of its nitrogen to these microorganisms.  相似文献

The biodegradation of estradiol (E2), estrone (E1), and ethinylestradiol (EE2) was investigated using mixed bacterial cultures enriched from activated sludge. Enrichments were carried out on E2 or EE2 in batch conditions with acetonitrile as additional carbon source. Degradation experiments were performed both using hormones as sole carbon source or with an additional source. The hormones were completely degraded by these cultures. Estradiol was rapidly converted to E1 within 24 h. Thereafter, E1 degradation began, displaying a lag phase ranging from 3 to 4 days. Estrone depletion took from 48 h to more than 6 days, depending on the culture conditions. For EE2 degradation, when it was the sole carbon source, the lag phase and the time required for its complete removal (7 and 15 days, respectively) were shorter that in cultures with a supplementary carbon source. The specific degradation rates observed for E2 both with and without an additional carbon source were similar. By contrast, the specific degradation rates for E1 and EE2 were, respectively, seven and 20 times faster when these hormones were supplied as the sole carbon source. The bacterial community structure of each culture was characterized by molecular and cultural methods. The mixed cultures were made up of species belonging to Alcaligenes faecalis, Pusillimonas sp., Denitrobacter sp., and Brevundimonas diminuta or related to uncultured Bacteroidetes. The isolated strain B. diminuta achieved the conversion of E2 to E1.  相似文献

The naturally occurring sulfonate N-acetyltaurine was synthesized chemically and its identity was confirmed. Aerobic enrichment cultures for bacteria able to utilize N-acetyltaurine as sole source of fixed nitrogen or as sole source of carbon were successful. One representative isolate, strain NAT, which was identified as a strain of Delftia acidovorans, grew with N-acetyltaurine as carbon source and excreted stoichiometric amounts of sulfate and ammonium. Inducible enzyme activities were measured in crude extracts of this organism to elucidate the degradative pathway. Cleavage of N-acetyltaurine by a highly active amidase yielded acetate and taurine. The latter was oxidatively deaminated by taurine dehydrogenase to ammonium and sulfoacetaldehyde. This key intermediate of sulfonate catabolism was desulfonated by the known reaction of sulfoacetaldehyde acetyltransferase to sulfite and acetyl phosphate, which was further degraded to enter central metabolism. A degradative pathway including transport functions is proposed.  相似文献

Biosynthesis of poly(3‐hydroxybutyrate) (PHB) from raw starch as the carbon source by the polysaccharide‐digesting bacteria Saccharophagus degradans was investigated in a fed‐batch culture. The production and properties of the PHB synthesized from starch were compared to those obtained using glucose as carbon source. In fed‐batch cultures, S. degradans accumulated 21.35 and 17.46% of PHB, using glucose or starch as carbon source, respectively. The physical properties of the biopolymer produced from each carbon source were similar between them. Molecular mass, melting temperature and heat of fusion were 54.23 kDa, 165.61°C and 59.59 J/g, respectively, using glucose; and 57.07 kDa, 174.31°C and 67.66 J/g, respectively, using starch. This is the first work describing the capability of S. degradans to utilize raw starch as the sole carbon source for the production of PHB.  相似文献

Lipid accumulation by Gordonia sp. DG using sodium gluconate as carbon source in comparison with Rhodococcus opacus PD630 was studied. Maximum lipid content 80% was observed at the beginning of the stationary phase for R. opacus and 72% at the end of stationary phase for Gordonia sp. Different agro-industrial wastes were used as carbon source. The cells of the two organism accumulated lipid more than 50% of the biomass with most tested agro-industrial wastes. The maximum value was in presence of sugar cane molasses (93 and 96%) for R. opacus and Gordonia sp. respectively. Maximum triacyglycerols (TAGs), 88.9 and 57.8 mg/l, was obtained using carob and orange waste by R. opacus and Gordonia sp. respectively. The use of orange waste as carbon source by R. opacus, increased lipid unsaturation with C18:3 as the major unsaturated fatty acid. On the other hand, C22:0 and C6:0 were the dominant fatty acids (54.5% of the total identified fatty acids) produced by Gordonia sp. in presence of orange waste as carbon source. Statistical optimization of the medium revealed that maximum lipid content was achieved with 60% orange waste, 0.05 g/l ammonium chloride and 0.2 g/l magnesium sulphate.  相似文献

A chemically defined medium has been developed to support the growth and the production of mosquito larvicidal factor(s) (MLF) of Bacillus sphaericus 1593M. On the basis of the data of steady-state continuous cultures, it has been understood that acetate can serve as a sole carbon and energy source for B. sphaericus 1593M. Utilization of acetate by B. sphaer-icus 1593M and the production of MLF are further enhanced by the addition of glutamate at low concentrations, both in steady-state continuous as well as in total-cell-retention cultures (TCRC). A two-step TCRC procedure resulted in better biomass and MLF production by B. sphaericus 1593M. It was also found that glutamate can serve as a carbon source as well as a growth factor in the presence of acetate and hence is a partially substitutable carbon source. A strain of the genus Rhodococcus, designated isolate S45-1, was isolated from an environmental water sample by enrichment, using the chlorinated paraffin Cereclor S45 as the sole carbon and energy source. This is the first report of microbial utilisation of chlorinated paraffins as sole source of carbon and energy. Biochemical studies of isolate S45-1 revealed little similarity with other Rhodococcus species. Isolate S45-1 was able to utilise 1-chloroalkanes of chain-length 12–18C as sole source of carbon and energy. Gas chromatography-mass spectrometry of the reaction medium indicated that γ-butyrolactone was formed as a product of 1-chlorotetradecane metabolism.  相似文献

Human long-latency auditory evoked potentials were studied during simulation with variable-amplitude pulse sequences from a sound source moving to and from the subject. The N1 peak parameters were shown to depend on an accurate estimate of the direction of the change in the distance to the sound source. Differences in the processing of signals that simulated the approaching and/or distancing of the sound source were found in the N1 and P2 component parameters of on- and off-responses as was a more pronounced long negative potential shift in the evoked response to the approaching source as compared to the distancing source.  相似文献

Summary Carbon and nitrogen sources were investigated for improving peroxidase production by Arthromyces ramosus, a hyperproducer of peroxidase. Glucose as carbon source and a mixture of yeast extract and polypeptone at the ratio of 3 to 5 as nitrogen source in a production medium were shown to give the highest peroxidase activity. During the culture amino acids such as alanine, arginine, methionine, leucine, tyrosine and tryptophan were depleted. Therefore, glucose supplemented nitrogen source fed-batch culture was carried out and a peroxidase activity of 73 U/ml was obtained. This activity was 1.7 times higher than that of glucose fed-batch culture. This indicates that an adequate nitrogen source supply during the culture is effective for improving the peroxidase production by A. ramosus.  相似文献

Pyrimidine ribonucleoside catabolic enzyme activities of the opportunistic pathogenPseudomonas pickettii were examined. Of the pyrimidine and related compounds tested, only dihydrouracil (nitrogen source) and ribose (carbon source) supported growth. Thin-layer chromatographic separation of the uridine and cytidine catabolities produced byP. pickettii extracts indicated that this pseudomonad contained nucleoside hydrolase activity. Its presence was confirmed by enzyme assay. Hydrolase activity was elevated in both glucose- and ribose-grown cells relative to succinate-grown cells. Nucleoside hydrolase activity was depressed when dihydrouracil served as a nitrogen source. Cytosine deaminase activity was present in extracts prepared from succinate-, glucose- or ribose-grown cells when (NH4)2SO4 served as the nitrogen source although cells grown on glucose or ribose exhibited a higher enzyme activity. Cytosine deaminase activity was not detected in extracts prepared from cells grown on dihydrouracil as a nitrogen source. Both dihydropyrimidine dehydrogenase and dihydropyrimidinase activities were measurable inP. pickettii. The dehydrogenase activity was higher with NADH than with NADPH as its nicotinamide cofactor when uracil served as its substrate. Carbon source did not affect dehydrogenase or dihydropyrimidinase activity greatly but both activities were diminished in cells grown on the nitrogen source dihydrouracil.  相似文献

Production of ammonia by urea-grown Chlorella ellipsoidea was investigated. Ammonia was produced during the stationary growth phase in cultures with urea as sole nitrogen source and glucose as supplementary carbon source. Ammonia was produced only in medium containing excess urea and limiting amounts of glucose. Ammonia production was accompanied by increase in pH. In cultures with nitrate as sole nitrogen source and glucose as supplementary carbon source, growth and pH changes were similar to those in urea-glucose medium, but no ammonia was detected. Cultures grown in urea-acetate medium were similar to those grown in urea medium without additional organic carbon source. No ammonia was produced under these circumstances and growth was significantly lower than that achieved in glucose-supplemented cultures. C. ellipsoidea evidently produces an enzyme or enzyme system which forms ammonia from urea. This organism was reportedly urease-free because previous workers did not detect ammonia formation from urea. Our findings indicate that special circumstances are required to produce detectable amounts of ammonia from urea. These findings are in agreement with a recent report of urea-splitting, cofactor-requiring enzyme in cell-free extracts of Chlorella.  相似文献

Field experiments were done to determine whether drenching plants with two systemically active neonicotinoid insecticides, thiamethoxam and imidacloprid, suppresses spread of Tomato spotted wilt virus (TSWV) by thrips vectors. Separate treatments to TSWV ‘infector’ tomato (source) and healthy lettuce (recipient) plants provided information on the relative importance of targeting control at virus acquisition by nymphs versus virus transmission to healthy plants by adults. Drenches were applied either to seedlings just before transplanting or to soil around plants. The thrips vectors recorded were Frankliniella occidentalis, F. schultzei and Thrips tabaci, but F. schultzei and T. tabaci predominated. Overall ratios of external to internal TSWV spread into and within plots without insecticide ranged from 1 : 2.3 to 1 : 2.8 between field experiments. Applying thiamethoxam as a soil drench to both young source plants and recipient seedling transplants suppressed TSWV incidence by 86%, while such application to either young source or recipient seedlings diminished incidence by 67–70%. When thiamethoxam was applied either as a soil drench to old source plants and concurrently as a seedling drench to recipient plants or as a seedling drench to recipient plants alone, incidence was suppressed by 65–73% and 54–73%, respectively. Thiamethoxam applied as a soil drench to old source plants diminished incidence by only 33% or not significantly. Imidacloprid applied either as a soil drench to old source plants and concurently as a seedling drench or as a seedling drench alone, suppressed TSWV incidence by 90–92% and 80% respectively. Although adult vector thrips and nymph numbers were low, fewer adults and/or nymphs were sometimes recorded due to insecticide application. Drenching healthy seedlings with neonicotinoid insecticides just before transplanting can be an effective chemical control measure to include in integrated disease management strategies to suppress TSWV epidemics in short‐duration crops like lettuce.  相似文献

A bacterium capable of degrading propoxur (2-isopropoxyphenyl-N-methylcarbamate) was isolated from soil by enrichment cultures and was identified as a Pseudomonas species. The organism grew on propoxur at 2 g/l as sole source of carbon and nitrogen, and accumulated 2-isopropoxyphenol as metabolite in the culture medium. The cell free extract of Pseudomonas sp. grown on propoxur contained the activity of propoxur hydrolase. The results suggest that the organism degraded propoxur by hydrolysis to yield 2-isopropoxyphenol and methylamine, which was further utilized as carbon source.  相似文献
