The effect of copper ions in seawater (0.02 mg/l) on the early stages of development of the sea urchin Strongylocentrotus intermedius was studied. Copper exposure from fertilization or the prism stage retarded development and growth and led to abnormalities in the morphology of the embryos and larvae. However, if development to the pluteus stage proceeded in clean seawater, an increased copper concentration did not inhibit the growth of larvae. If sea urchin embryos at fertilization and the prism stage were maintained for 1–2 days in seawater containing 0.02 mg Cu/l and then transferred to clean seawater, the adverse consequences of this exposure remained present after 48 h.  相似文献

The effects of water temperatures of 7, 15 and 20°C and pollution of sea water by copper ions in concentrations of 0.005 and 0.01 mg/l on the early development of the Pacific mussel Mytilus trossulus were studied. It was shown that copper additives were most toxic at the initial stages of development, from fertilization up to the veliger stage. It was determined that larvae of the Pacific mussel were able to adapt to copper at a concentration of 0.005 mg/l. The copper toxicity increased to a greater extent with an increase in water temperature from its average value of 15°C, than with a temperature decrease.  相似文献

The appearance of muscle proteins in the contractile apparatus of the mussel Mytilus trossulus was subjected to comparative analysis during ontogenesis. It was established, with the use of Western blot analysis and electrophoresis in polyacrylamid gel in the presence of sodium dodecylsulfate, that proteins of the contractile apparatus of mussel muscles express long before the formation of the first functionally active muscle system of the veliger larvae. Paramyosin is present in egg cells; twitchin, myorod, and actin appear at the stage of blastula (12 h after fertilization), and myosin appears at the trochophore stage (17 h after fertilization). The quantitative relation of muscle proteins was studied in actomyosin extracts of larvae obtained from different developmental stages. It was shown that the ratios actin/myosin and paramyosin/myosin at the veliger stage (96 h after fertilization) were found to be similar to those in the striated muscles of invertebrates.  相似文献

Brachymystax tsinlingensis Li is a threatened fish species endemic to China. With the problems of environmental factors and seeding breeding diseases, it is important to further improve the efficiency of seeding breeding and the basis of resource protection. This study investigated the acute toxicity of copper, zinc and methylene blue (MB) on hatching, survival, morphology, heart rate (HR) and stress behaviour of B. tsinlingensis. Eggs (diameter: 3.86 ± 0.07 mm, weight: 0.032 ± 0.004 g) of B. tsinlingensis were selected randomly from artificial propagation and developed from eye-pigmentation-stage embryos to yolk-sac stage larvae (length: 12.40 ± 0.02 mm, weight: 0.03 ± 0.001 g) and exposed to different concentrations of Cu, Zn and MB for 144 h in a series of semi-static toxicity tests. The acute toxicity tests indicated that the 96-h median lethal concentration (LC50) values of the embryos and larvae were 1.71 and 0.22 mg l−1 for copper and 2.57 and 2.72 mg l−1 for zinc, respectively, whereas the MB LC50 after 144-h exposure for embryos and larvae were 67.88 and 17.81 mg l−1, respectively. The safe concentrations of copper, zinc and MB were 0.17, 0.77 and 6.79 mg l−1 for embryos and 0.03, 0.03 and 1.78 mg l−1 for larvae, respectively. Copper, zinc and MB treatments with concentrations greater than 1.60, 2.00 and 60.00 mg l−1, respectively, led to a significantly low hatching rate and significantly high embryo mortality (P < 0.05), and copper and MB treatments with concentrations greater than 0.2 and 20 mg l−1 led to significantly high larvae mortality (P < 0.05). Exposure to copper, zinc and MB resulted in developmental defects, including spinal curvature, tail deformity, vascular system anomalies and discolouration. Moreover, copper exposure significantly reduced the HR of larvae (P < 0.05). The embryos exhibited an obvious change in behaviour, converting from the normal behaviour of emerging from the membrane head first to emerging tail first, with probabilities of 34.82%, 14.81% and 49.07% under copper, zinc and MB treatments, respectively. The results demonstrated that the sensitivity of yolk-sac larvae to copper and MB was significantly higher than that of embryos (P < 0.05) and that B. tsinlingensis embryos or larvae might be more resistant to copper, zinc and MB than other members of the Salmonidae family, which benefits their resource protection and restoration.  相似文献

Integrins play a key role in the intermediation and coordination between cells and extracellular matrix components. In this study, we first determined the presence of the β integrin‐like protein and its presumptive ligand, fibronectin‐like protein, during development and in some adult tissues of the bivalve mollusc Mytilus trossulus. We found that β integrin‐like protein expression correlated with the development and differentiation of the digestive system in larvae. Besides the presence of β integrin‐like protein in the digestive epithelial larval cells, this protein was detected in the hemocytes and some adult tissues of M. trossulus. The fibronectin‐like protein was detected firstly at the blastula stage and later, the FN‐LP‐immunoreactive cells were scattered in the trochophore larvae. The fibronectin‐like protein was not expressed in the β integrin‐positive cells of either the veliger stage larvae or the adult mussel tissues and the primary hemocyte cell culture. Despite the β integrin‐ and fibronectin‐like proteins being expressed in different cell types of mussel larvae, we do not exclude the possibility of direct interaction between these two proteins during M. trossulus development or in adult tissues.  相似文献

Studies on the avoidance behavior of aquatic organisms to contaminants have confirmed that such behavior can be relevant in field situations. However, almost all toxicity tests involve the forced exposure of organisms to toxicants. In particular, despite the importance of Chironomus riparius Meigen larvae in sediment toxicity testing, only a few studies on avoidance behavior have been performed. This study investigated the ability of different life stages of C. riparius, including ovipositing females, first-, second-, and fourth-instar larvae, to avoid copper-contaminated environments. Ovipositing females were given a choice between a control and copper solution (1.3 mg Cu l? 1). First-instar larvae were provided with a choice between a control and a copper (2.0 mg Cu l? 1)-spiked sediment. Both second- and fourth-instars were exposed to a copper gradient (0.38–3.4 mg Cu l? 1) in a flow-through system. None of the life stages avoided copper, even though the highest concentrations caused lethal effects on midges. The avoidance behavior of C. riparius is not a sensitive endpoint to assess copper sublethal toxicity.  相似文献

本实验采用暴露重金属的方法,研究了不同浓度硫酸铜(Cu2+ 分别为0、0.005、0.01、0.02、0.05 mg/L)在不同暴露时间(0—14d)下对梨形环棱螺(Bellamya purificata)过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽硫转移酶(GST)的活性、还原性谷胱甘肽(GSH)和金属硫蛋白(MT)含量的影响,以探讨Cu2+ 对梨形环棱螺的氧化损伤及其防御作用的机理,并为水环境Cu2+ 污染的早期诊断及生态风险评价提供科学的依据。结果表明:Cu2+对梨形环棱螺肝脏和鳃中CAT、SOD、GST、GSH 和MT 均有明显影响,表现出时间剂量效应。SOD在前4天、CAT在前3天酶活性总体上表现出诱导趋势, GST在前4天酶活性处于诱导状态,随着暴露时间的延长,酶活性下降,到第5天时表现出抑制趋势;随着时间的进一步增长,至14d时, 0.005 mg/L剂量组酶活性维持在正常值附近波动, 0.01 mg/L剂量组酶活性被诱导, 0.02 mg/L剂量组酶活性在肝脏中表现为诱导而在鳃中则被抑制,0.05 mg/L剂量组酶活性被抑制。肝脏和鳃GSH含量的变化与GST相似,在短时间内表现出诱导效应,肝脏GSH在暴露的前5天、鳃GSH在暴露的前4天均处于诱导状态,随着暴露时间的延长,0.005 mg/L剂量组表现出诱导,0.05 mg/L剂量组则受到抑制。MT在整个实验期间均处于诱导状态,各剂量组在0.5d被极显著诱导,随后MT含量出现起伏波动,有上升和下降,至第14天时达到一稳定水平。其中,0.01 mg/L剂量组肝脏的MT在整个实验期间均被极显著地诱导(P <0.01),0.01 mg/L 剂量组的鳃组织MT除第10天外也被极显著诱导(P <0.01)。在暴露14d时,除0.05 mg/L剂量组的肝脏MT外,其余处于极显著诱导状态(P <0.01)。  相似文献

运用SKW-3微量呼吸仪对卵形鲳鲹(Trachinotus ovatus)胚胎和早期仔鱼的耗氧量进行测定,研究温度、盐度及pH的变化对胚胎耗氧量和Cu2+、Cd2+等重金属离子对早期仔鱼的耗氧量的影响。结果表明:在(25±0.5)℃条件下,卵形鲳鲹胚胎和早期仔鱼的耗氧量均随着发育时间的延长总体上呈上升的趋势,胚胎耗氧量在出膜前达到最大值,其中以原肠期和出膜期耗氧量变化最为显著。胚胎耗氧量随着温度、盐度、pH的增大而逐渐升高,在水温25℃、盐度35和pH为8时耗氧量到达最大值,随后逐渐减小。随着Cu2+浓度的升高,早期仔鱼耗氧量呈先增大后减小,在0.01mg·L-1时耗氧率最大:胚胎耗氧量随着Cd2+浓度的升高而逐渐降低。  相似文献

Salinity adaptations of the gastropodsLittorina mandshurica andL. squalida were studied at different stages of their life cycles.L. mandshurica exhibited a higher degree of adaptation to variations in salinity thanL. squalida at all developmental stages. Egg masses did not protect embryos from desalination; the larvae inside the egg masses were capable of adapting to diluted seawater. As larvae developed in egg masses, their adaptive capabilities increased. At the time of hatching, the salinity tolerance of larvae differed little, if at all, from that of adults. It is assumed that pelagic egg capsules provide an advantage in the distribution of a species.  相似文献

We investigated the behavior of larvae of the sea urchin Strongylocentrotus intermedius at the blastula and early pluteus stages in the water column at a temperature of 20, 22, and 23.5°C and in the presence of copper in concentrations of 0.01 and 0.02 mg/l. In clean sea water, at all tested temperatures, sea urchin larvae preferred the subsurface layer to where they rose from the bottom of the vessel to form dense aggregations. In water supplemented with copper, the behavior of larvae at 20 and 22°C was not different from that in clean water. An increase in temperature to 23.5°C and the addition of copper altered the behavior of the larvae; they did not leave near-bottom water, did not rise to the surface, although they continued moving.  相似文献

The possibility of transferring exogenous DNA into eggs by mussel Mytilus galloprovincialisLam. sperms both with the use of certain methods of transfection and without them was studied. The efficacy of egg fertilization by sperms treated with foreign DNA and the development of larvae at early stages of embryogenesis were evaluated. Negative effects of the contact between mussel sperms and exogenous DNA on fertilization and subsequent development were noted. The proportion of developing larvae decreased with increasing DNA concentration and sperm exposure. Transfer of plasmids pCMVlacZ and pMTbGHinto eggs was observed in group crosses. With the use of PCR, foreign DNA sequences were found in the larvae at the stage of veliger 48 h after fertilization. An intense signal was recorded after sperm electroporation in 10% DMSO.  相似文献

The development of contractile apparatus was subjected to comparative analysis during ontogenesis of the mussel Mytilus trossulus. Indirect immunofluorescence with the polyclonal antibody against mussel twitchin, a protein of thick filaments, and fluorescent phalloidin as a marker of filamentous cell actin were used to monitor changes in the developing muscle system at different larval stages. The first definitive muscle structures were found at the late trochophore stage (36 h after fertilization) and starting from the midveliger stage (96h), striated muscles, which are never present in adult mussels, were distinctly seen. The striated muscle periodicity was 1.25 μm in both mussel larvae and adult scallop. The contractile activities of veliger and adult muscles were measured using an electronic signal-processing video workstation. This work is the first complex study of morphological, biochemical, and physiological characteristics of the muscle system in the larvae and adult molluscs.  相似文献

Although our understanding of neuronal development in Trochozoa has progressed substantially in recent years, relatively little attention has been paid to the bivalve molluscs in this regard. In the present study, the development of FMRFamide-, serotonin- and catecholamine-containing cells in the mussel, Mytilus trossulus, was examined using immunocytochemical and histofluorescent techniques. Neurogenesis starts during the trochophore stage at the apical extreme with the appearance of one FMRFamide-like immunoreactive (lir) and one serotonin-lir sensory cell. Later, five FMRFamide-lir and five serotonin-lir apical sensory cells appear, and their basal fibres form an apical neuropil. Fibres of two lateral FMRFamide-lir apical cells grow posteriorly and at the time that they reach the developing foot, the first FMRFamide-lir neurons of the pedal ganglia also appear. Subsequently, FMRFamide-lir fibres grow further posteriorly and reach the caudal region where neurons of the developing visceral ganglia then begin to appear. In contrast, the five apical serotonin-lir neurons do not appear to project outside the apical neuropil until the late veliger stage. Catecholamine-containing cells are first detected in the veliger stage where they appear above the oesophagus, and subsequently in the velum, foot, and posterior regions. Though neural development in M. trossulus partly resembles that of polyplacophorans in the appearance of the early FMRFamidergic elements, and of scaphopods in the appearance of the early serotonergic elements, the scenario of neural development in M. trossulus differs considerably from that of other Trochozoa (bivalves, gastropods, polyplacophorans, scaphopods and polychaetes) studied to date. Summary

Nectochaete larvae of the ecologically and economically important ragworm, Nereis virens, were exposed to cadmium, chromium, copper, lead and zinc dissolved in seawater to nominal concentrations ranging from 0 to 5000 μg l?1. Copper was the most toxic (mean LC50 of 76.5 μg l?1 ± 95% CI 73.8–79.2 after 96 h exposure) and so was used for subsequent experiments. Exposure of gametes to greater than 500 μg l?1 copper for 2 or 4 h at 10°C prior to fertilization, or a 10 min exposure during fertilization, significantly reduced embryo developmental success. The effect of copper on larval settlement was also assessed using sediment spiked to a range of concentrations (0, 50, 250, 500, 1000 mg kg!1 dry weight). Significantly fewer larvae were found in sediment of $250 mg kg!1 in comparison to the control or the 50 mg kg!1 treatment. Assessment of living larvae also confirmed a significant reduction in settlement, but in all treatments compared to the control, although the number of dead larvae also increased as the concentrations increased. These effects may have important implications for reproductive success and recruitment of N. virens to polluted sediments.  相似文献

The oyster Ostrea chilensis develops in estuaries and incubates its embryos inside the pallial cavity. Reduced external salinity triggers valve closure and isolation of the brood chamber, which in turn alters conditions in the pallial fluid, including pH reductions ranging from 7.65 (SD = 0.05) to 6.97 (SD = 0.02) in non-brooding females and 6.96 (SD = 0.10) in brooding females after 12 h of isolation. Exposing veliger larvae to acidic pH's (5 and 3) decreased the thickness of the embryonic shell valves. Calcium, sodium, chlorine, tin, sulphur, and magnesium were the most important (> 99%) elements constituting the veliger shells. Calcium content in the pallial cavity fluid increased with continued isolation in both brooding and non-brooding females, but calcium remained as the primary component (about 94%) of embryonic shells and maintained its normal proportion with respect to the rest of the elements at all tested pH's. The increased calcium in the pallial fluid was identified mainly as coming from the shells of brooding females. If any neutralizing elements were coming from embryonic shell valves, all elements in those shells must have been solubilising to the same degree. Veligers ceased all further shell growth while isolated within the female's brood chamber. However, veliger shell growth resumed once females were returned to high-salinity seawater. Our study shows that female isolation of the mantle cavity, often considered an adaptive response to the periodically reduced salinities of estuarine waters, can in fact generate adverse effects on brooded embryos.  相似文献

在实验室条件下人工孵化扁玉螺(Neverita didyma)的卵块,观察了其胚胎发育和幼虫发育过程.扁玉螺的早期发育属间接发生型,其胚胎发育包括卵裂期、囊胚期、原肠胚、膜内担轮幼虫、膜内面盘幼虫;幼虫发育包括面盘幼虫、后期面盘幼虫和匍匐幼虫;匍匐幼虫经变态后发育为稚螺.在水温25~26℃条件下,受精卵发育至膜内面盘幼虫约需38h,5~6d后面盘幼虫冲破卵膜而孵化.扁玉螺面盘幼虫的显著特点是具有1对眼点和1对平衡囊,面盘呈双叶状;后期面盘幼虫的面盘为4叶,呈蝴蝶状,足发达,幼虫既能浮游,又能爬行.后期面盘幼虫进一步生长发育,逐渐转入匍匐生活.  相似文献

Somatic embryogenesis was obtained from cotyledon and mature zygotic embryo callus cultures of Terminalia chebula Retz. Callus cultures of cotyledon and mature zygotic embryo were initiated on induction medium containing Murashige and Skoog (MS) nutrients with 1.0 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D) either 0.01 or 0.1 mg/l Kinetin and 30 g/l sucrose. Induction of somatic embryogenesis, proliferation and development was obtained through different culture passages. Embryogenic cotyledon callus with globular somatic embryos was obtained on MS basal medium supplemented with 50 g/l sucrose. Globular somatic embryos were observed from mature zygotic embryo callus on induction medium. Different stages of somatic embryo development from cotyledon and mature zygotic embryo calluses were observed on MS basal medium supplemented with 50 g/l sucrose after 4 weeks of culture. Histological studies have revealed the developmental stages of somatic embryos. A maximum of 40.3±1.45 cotyledonary somatic embryos/callus was obtained from mature zygotic embryo compared to 7.70±0.37 cotyledonary somatic embryos/callus initiated from cotyledons. Germination of somatic embryos and conversion to plants were achieved. Highest frequency of germination (46.66±0.88) of somatic embryos was obtained on MS basal medium containing benzyladenine (0.5 mg/l) with 30 g/l sucrose.  相似文献

Toxicity of copper to T. pseudonana (formerly Cyclotella nana, clone 13-1) wax examined in inshore seawater using a 96-hr bioassay method. Raw unenriched seawater wax filtered through a 0.22-μ membrane filter and then pasteurized, for 30 min at 60 C. Following this treatment, samples contained 0.68–1.14 μg Cu/liter. Copper was added as the chloride in 5-μg increments over the range of 5 to 30 μg/liter (about 0.1–0.5 μM). Population densities, mean cell, volume, and 14C bicarbonate uptake were measured. Population growth, and 14C uptake by T. pseudonana displayed inhibition over the entire range of added copper. Growth rate constant (k) of T. pseudonana decreased with increasing copper concentration and during the course of growth at each concentration. Correspondingly, mean cell volumes increased with copper concentration and time. Copper toxicity varied in different water samples. The presence of decomposed natural plankton and detritus decreased toxicity. In the absence of enrichment, bacteria had little effect on copper toxicity. H exults were influenced by glassware treatment, collection and storage of seawater, and absence of enrichments.  相似文献

The effectiveness of the protection of embryos and larvae in egg masses from reduced salinity and desiccation was investigated in the gastropod Epheria turrita, which inhabits the intertidal and upper subtidal zones. Embryos and larvae developing inside egg masses were shown not to be protected against changes in environmental salinity. Viable larvae hatched from egg masses at a salinity of 24–26. However, if free-swimming veliger larvae, rather than egg masses, were transferred to water of reduced salinity, the range of salinity tolerated by the larvae was wider, and its lower limit was 18–20. Egg masses did not protect against desiccation either. A 3-h exposure of egg masses to drying resulted in larval mortality. Larvae hatched from egg masses did not rise to the surface and displayed an early tendency towards gregarious behavior, which apparently enhances their ability to settle on a proper substrate near parental populations.  相似文献
