The application of a specific species of willow—Salix amygdalina L., marked by high transpiration ability—is a cheap and effective method of landfill leachate disposal. A 2-year study examined the effectiveness of leachate evapotranspiration from soil–plant systems with willow species S. amygdalina L. Evapotranspiration from soil–plant systems planted with willow was from 1.28 up to 5.12 times higher than evaporation from soil surface barren of vegetation. This proves the usefulness of soil–plant systems with willow in landfill leachate treatment through vaporization. Evapotranspiration efficiency, as opposed to total amount of water added into the lysimeter, was not strong enough to vaporize all input of the landfill leachate in the lysimeters. This may indicate that the ground water requires isolation when soil systems remain under landfill leachate irrigation. Linear dependence between willow biomass growth and transpiration was observed to be significant (p < 0.05). Additionally, the research showed that the application of sewage sludge into the soil caused an increase in vaporization efficiency.  相似文献

Gradients of tritiated water, 35S-sulphate and 32P-phosphate, were established in isolated segments of willow stems. Sieve tube exudate was collected as honeydew from the high activity end of the segment. After girdling the stem a few centimetres from the site of sieve tube puncture, the specific activity of 35S and 32P in the honeydew rose, whilst the specific activity of tritium remained constant. These findings indicate that prior to girdling, unlabelled sulphates and phosphates were contributing to the honeydew, whilst there had been no detectable contribution by unlabelled water from the low activity end of the segment. The data support the conclusions drawn from previous experiments by Peel et al. (1969), that water is relatively immobile in sieve tubes of willow when compared to solutes.  相似文献

Somatic embryos ofHevea brasiliensis can be obtained by culturing thin sections of inner tegument of seed on two successive different media, MH1 and MH3. Histological study showed that in calli cultured on non-renewed medium MH1 for 40 days, the embryogenesis process initiated on the 20th day did not produce results owing to early degeneration of the cells involved in the embryogenic pathway. However, typical embryogenic cells formed when medium MH1 was renewed once during the first phase of culture (between day 20 and day 30). Proembryos developed when the calli were subcultured on medium MH3 10–15 days later. Embryogenic cells did not form when there was frequent renewal of medium MH1 or early subculturing on MH3 after less than 40 days of culture on MH1. Methodical histological monitoring of the development of embryogenic quality of calli thus made it possible to define the optimum culture sequences for the embryogenesis process and which are favourable for regular obtaining of proembryos.  相似文献

Summary The maximum specific growth rate (max) of Hansenula polymorpha MH30 on xylose as the sole source of carbon and energy is 0.175 h–1, on methanol 0.21 h–1, on glycerol 0.27 h–1 and on glucose 0.61 h–1. On mixtures of xylose plus methanol, xylose plus glycerol, xylose plus glucose and glycerol plus glucose H. polymorpha MH30 grows faster: 0.36 h–1, 0.37 h–1, 0.47 h–1 and 0.52 h–1, respectively. Attempts have been made to explain these somewhat surprising results, especially the fact that the growth rates on xylose plus methanol and xylose plus glycerol exceed the specific growth rates of these on even the faster partner in the mixture. Variations in nitrogen fixation (acetylene reduction) by Nodularia spumigena blooms in the Peel-Harvey estuarine system were examined with respect to spatial (sampling station location, and depth) and temporal (seasonal and diurnal) distribution. The annual contributions of nitrogen fixation by the blooms to the nitrogen budget of the estuary were estimated to range from 309 to 713t. Contributions by nitrogen fixation were similar to the riverine inputs in the Harvey Estuary, but lower in the Peel Inlet.The Harvey Estuary had higher biomass and total fixation rates (to 0.4 nmol C2H2 · ml–1 h–1), but the heterocyst nitrogen fixation rates were greater in the Peel Inlet (to 9 × 10–1 nmol C2H2 · heterocyst–1 · h–1). Nitrogen fixation decreased with depth in response to light, though other factors also appeared to be involved. The rates of fixation decreased concurrently with increasing bloom age, total soluble inorganic nitrogen and salinities. Maximum daily fixation rates occurred in the early morning.  相似文献

Using a bacterial artificial chromosome (BAC) library, we analysed a 99.5 kb genomic segment containing the major histocompatibility class II genes of a teleost, the three-spined stickleback Gasterosteus aculeatus. Experiments with G. aculeatus have provided direct evidence for balancing selection by pathogens and mate choice driving MH class II beta polymorphism. Two sets of paralogous class II alpha genes and beta genes in a tandem arrangement were identified, designated Gaac-DAA/DAB and Gaac-DBA/DBB. Expression analysis of the beta genes using single-strand conformation polymorphism revealed that both gene copies are expressed. Based on an analysis of pairwise nucleotide polymorphisms, we estimate that the duplication into two paralogous class II loci occurred only 1.2–2.4 million years ago, 1–2 orders of magnitude more recently than in other fish, bird or mammalian species. At the 3-direction of the classical MH loci, we identified another seven genes or gene fragments, two of which (small inducible cytokine, complement regulatory factor) are related to immune function in other vertebrates. None of these genes were associated with MH class II genes in zebrafish, suggesting a markedly different organisation of the MH class II region in sticklebacks, and presumably, across bony fishes. When the nucleotide substitution pattern of the novel class II beta genes was analysed together with a representative sequence sample isolated from fish in northern Germany (n=27), we found that the peptide binding region of the Gaac-DAB and Gaac-DBB loci had undergone an inter-locus gene conversion (P=0.007). In accordance, we found a 10- to 20-fold higher frequency of CpG-islands on the MH class II segment compared to other species, a feature that may be conducive for inter-locus recombination.  相似文献

Utilization of surface localized substrate by non-adhesive marine bacteria   总被引:5,自引:0,他引:5  
Thirty-four marine bacteria were isolated from the eluate of seawater passed through a column of glass beads coated with stearic acid. Irreversible attachment of these isolates to stearic acid-coated glass surfaces ranged from 7.6–100% of the total attached population, with 7 isolates exhibiting less than 10% irreversible adhesion. All 14 isolates tested were able to utilize surface bound14C-stearic acid, even though some showed mostly reversible adhesion to the surface. More detailed studies were made comparing the reversibly adheringVibrio MH3 with the irreversibly adheringPseudomonas NCMB2021. MH3 cells were readily removed from the surface by a gentle shear force, and a significant degree of14C-labeling of MH3 cells, but not of NCMB2021 cells, in the bulk phase was observed. The ecological significance of nutrient scavenging at solid surfaces by reversibly attached bacteria is considered.  相似文献

Summary The present study investigates some of mechanisms for tumor-specific Lyt-1+2 T cell-mediated tumor cell eradication in vivo through analyses of tumor specificity in the afferent tumor recognition and efferent rejection phases. When C3H/He mice which had acquired immunity against syngeneic MH134 hepatoma were challenged with other syngeneic X5563 plasmacytoma cells, these mice failed to exhibit any inhibitory effect on the growth of X5563 tumor cells. However, the inoculation of X5563 tumor cells into the MH134-immune C3H/He mice together with the MH134 tumor cells resulted in appreciable growth inhibition of antigenically distinct (bystander) X5563 tumor cells. Although the growth of X5563 cells was inhibited in an antigen-nonspecific way in mice immunized to antigenically unrelated tumor cells (bystander effect), the activation of Lyt-1+2 T cells leading to this effect was strictly antigen-specific. Such a bystander growth inhibition also required the admixed inoculation of the bystander (X5563) and specific target (MH134) tumor cells into a single site in mice immunized against the relevant MH134 tumor cells. Furthermore, the results demonstrated that Lyt-1+2 T cells specific to MH134 tumor cells were responsible for mediating the growth inhibition of antigenically irrelevant (bystander) and relevant tumor cells. Rapid leaf decay and nutrient release in a chinese tallow forest   总被引:6,自引:0,他引:6  
Summary The Chinese tallow tree, Sapium sebiferum, was introduced to the Texas Gulf Coast in the early 1900's and has spread into coastal prairie habitats. Since coastal prairie contains few deciduous trees, we studied leaf fall dynamics, rate of decomposition, and the quantity and rate of nutrient input from decomposing tallow leaves. Among-year leaf fall averaged 382.6 g·m-2·yr-1, similar to other south temperat deciduous forests and about as predicted by Meentemeyer et al.'s (1982) AE-lignin model. Decay of tallow leaves (k=-4.33) was much more rapid than native black willow (k=-0.35) and than other temperate deciduous trees (k=-0.77). The ratio of lignin to initial nitrogen concentration, highly correlated with rate of decomposition for hardwood trees, was low for Chinese tallow and may contribute to rapid leaf decay. Taking AE and lignin content into account, Meentemeyer's (1984) model predicted k=-1.39 for Chinese tallow and k=-0.88 for black willow. Decay of tallow was much faster but decay of willow was slower than predicted, suggesting that decay on the coastal prairie may be controlled by factors other than lignin content and climate. N, P, and K characteristically accumulate as leaves decay. However, these elements did not accumulate as tallow leaves decayed, possibly because high densities of Armadillidium vulgare, a detritivore, reduced immobilization of elements by microbes. This would result in increased turnover of these elements. Accumulation of Al, Fe, Zn, and S in decaying tallow litter may be related to flood-drain cycles on coastel prairie clay soils. Ca, N, K, Mg, and S were added to forest soil in greatest amounts from decaying tallow leaves. Concentrations of P, K, NO3–N, Zn, Mn, and Fe were significantly higher and Mg and Na were significantly lower in forest than in prairie soil, raising the possibility that Chinese tallow trees altered the distribution of nutrients in the soil profile. We conclude that the Chinese tallow tree may enhance productivity in ecosystems to which it has been introduced by addition of nutrients from rapid decay of leaves.  相似文献

We studied the relationship between variation in age and shoot characteristics of the host plant Salix exigua Nuttall (coyote or sandbar willow) and the attack and survival of Euura sp. (an unnamed leaf-midrib galling sawfly). Variation in shoot characteristics resulted from reduced growth as willow ramets aged. Mean shoot length per ramet and mean longest leaf length per shoot decreased by 95% and 50% respectively between 1- and 9-year-old willow ramets. All measured shoot characteristics-shoot length, longest leaf length, number of leaves per shoot, and mean internode length-were significantly negatively correlated with ramet age (r 2 ranged from –0.23 to –0.41). Correlations between shoot characteristics were highly positive, indicating that plants also grew in a strongly integrated fashion (r 2 ranged from 0.54 to 0.85). Four hypotheses were examined to explain sawfly attack patterns. The host-plant hypothesis was supported in explaining enhanced larval sawfly survival through reduced plant resistance. As willow ramets aged, the probability of Euura sp. attack decreased over 10-fold, from 0.315 on 1-year-old ramets to 0.024 on 2- to 9-year-old ramets. As shoot length increased, the probability of sawfly attack increased over 100-fold, from 0.007 on shoots <100 mm, to 0.800 on shoots in the 1001–1100 mm shoot length class. These attack patterns occurred even though 1-year-old ramets and shoots >500 mm each represented less than 2% of the total shoots available for oviposition. Host plant induced mortality of the egg/early instar stage decreased by 50% on longer leaves and was the most important factor determining survival differences between vigorous and non-vigorous hosts. Sawfly attack was not determined by the resource distribution hypothesis. Although shoots <200 mm contained 82% of the total leaves available, they contained only 43% of the galls initiated. The attack pattern also was not explained by the gall volume hypothesis. Although gall volume increased on longer shoots, there was no significant variation in mid or late instar mortality over shoot length, as would be expected if food resources within smaller galls were limited. The natural enemy attack hypothesis could not explain the pattern of oviposition since predation was greater on longer shoots and leaves. In addition, larval survival was related to oviposition behavior. Due to a 69% reduction in late instar death and an 83% reduction in parasitism, survival of progeny in galls initiated close to the petiole base was 2.8 times greater than in galls initiated near the leaf tip. A 75% reduction in gall volume over this range of gall positions may account for the observed increases in late instar mortality and parasitism.  相似文献

Summary The ability of embryos at different developmental stages to form plants in vitro has been studied in cultivated Cucumis sativus L. and in the wild species C. zeyheri 2 x Sond. and C. metuliferus Naud. On MS medium containing 3.5% sucrose, 0.1 mg 1–1 kinetin (Kn) and 0.01 mg 1–1 indoleacetic acid (IAA), proembryos (0.03–0.05 mm) and early globular embryos (0.05–0.08 mm) from the wild species developed into plants in low frequencies of 8% and 21%, respectively. These embryos should be surrounded by the embryo sac tissue. On the same medium late globular (0.08–0.1 mm) and early heart-stage embryos (0.1–0.3 mm) developed into plants in moderately high and high frequencies of 48% and 83%, respectively. The presence of the embryo sac at these stages was still beneficial, but no longer a prerequisite. Late heart-stage embryos (0.3–0.8 mm) also showed high frequencies of plant formation, 63%, if Kn was applied at a concentration of 1 mg 1–1. From the early cotyledon stage onwards, the frequency of plant formation gradually decreased, reaching a minimum at the late cotyledon stage. Subsequently it began to increase again up to the late maturation stage. The poor plant formation shown by the intermediate-aged embryos could be improved slightly by lowering the sucrose concentration to 0.5% and by increasing the Kn concentration to 10 mg 1–1. Relative to the wild species, embryos of C. sativus showed lower percentages of plant formation. The optimum sucrose concentration was 2% for the heart-stage C. sativus embryos. In all three species the ability to form plants strongly decreased with increasing embryo age, from early to late cotyledon. This is thought to be caused by the increasing tendency of the embryos at these stages to continue in vitro the normal embryo development.  相似文献

Summary Water willow and grey alder were grown on a raised sphagnum bog in central Sweden. The stands were intensively treated by daily irrigation and fertilization during the growing period in order to improve site fertility. After a 2-year establishment period high production rates were achieved in willow stands, 0.8 kg stem dry weight m–2 year–1 on current plus one (C+1) year old shoots. In these stands the canopy was closed with a leaf area index (LAI) that peaked at approximately 7. The canopy in the alder stand did not close during the initial 3 years of growth and the measured production rate was relatively low, at approximately 0.4 kg dry weight m–2 year –1 in the last year. The leaf nitrogen content was 3%–4% of dry weight during the summer and the other studied mineral elements were in almost optimal proportion to nitrogen. This was considered to be an effect of the intensive fertilization regime. Above-ground production close to maximum yield was attained in the prevailing conditions of soil, climate and biomass partitioning.  相似文献

Cadmium accumulation and tolerance in Populus nigra and Salix alba   总被引:1,自引:0,他引:1  
Rooted cuttings of Populus nigra L. clone Poli and Salix alba L. clone SS5 were treated for three weeks with 50 μM CdSO4 in nutrient solution. The willow showed a far higher Cd tolerance, expressed as tolerance index (Ti), than the poplar in both roots and leaves. The root Cd content was higher in poplar than in willow, whereas in leaves the opposite was found. As a consequence, the translocation factor (Tf) revealed a greater ability of Cd transport in willow than in poplar. Cd treatment enhanced cysteine, γ-glutamylcysteine and reduced glutathione contents in roots of both species, whereas in leaves they were only enhanced in poplar. Furthermore, only poplar leaves showed an enhanced content of phytochelatins, whereas malic and citric acids rose in response to Cd only in the willow leaves. Cd treatment increased putrescine, spermidine and spermine contents in both roots and leaves of the willow, whereas in poplar only the putrescine content was enhanced in roots.  相似文献

Summary When labelled uracil was supplied to the solution bathing the cambial surfaces of bark strips of willow, uptake into the sieve elements took place as evidenced by the appearance of radioactivity in the honeydew of aphids feeding on the strips. When maleic hydrazide (MH) was also applied an inhibition of uracil uptake was observed. However uracil seemed to have no effect on MH uptake. Biomass production and Cd uptake in Salix (willow) is generally high. The aim of this study was to quantify the amounts of Cd in different plant and soil compartments in commercial Salix stands grown on different soil types and to use these data for estimation of the long-term effects of Cd removal by Salix at stem harvest on the soil Cd content by calculating Cd balances. Ten commercial short rotation willow coppice stands, planted with clone 78183 of Salix viminalis (L.) were investigated in order to include three different soil types with respect to pH. Stands with high stem biomass production generally had lower Cd concentrations in the plant and grew on more alkaline soils with low Cd availability when compared to stands with low stem biomass production. The highest output of Cd by Salix is therefore not necessarily achieved by stands with the highest yields or the highest Cd concentrations, as both these parameters have to be considered. The net removal of Cd from the plough layer by the Salix crop under different conditions varied between 2.6 and 16.5 g Cd ha–1 year–1. The long-term effects of Salix cultivation on soil Cd content, i.e. after 6–7 cutting cycles (25 years), were calculated and we reached the conclusion that there is a high potential in Salix to remove significant amounts of Cd at stem harvest. Salix can therefore presumably be used to counteract Cd pollution of agricultural soils in Sweden provided that the Cd in furnace ashes is taken care of.  相似文献

14C-labelled sugars (sucrose, glucose, and fructose) were suppliedto the cambial surface of bark strips of willow, and in conjunctionwith the aphid stylet technique (Weatherley, Peel, and Hill,1959), the movement into and distribution of activity in thesieve tube sap was examined. As well as sucrose, free hexosesand sugar phosphates were found to contain the 14C-label, andrapid interconversions occurred between these compounds somewherealong the entry route. Sucrose entry into the sieve elementswas accompanied by at least a partial breakdown into its componenthexoses, and the involvement of a sugar phosphate pool was alsodemonstrated. Possible transformation sites on the entry routebetween the solution bathing the cambial surface and the sieveelement vacuole are discussed.  相似文献

Poplar and willow are economically-important, fast-growing tree species with the ability to colonize nutrient-poor environments. To initiate a study on the possible contribution of endophytes to this ability, we isolated bacteria from within surface-sterilized stems of native poplar (Populus trichocarpa) and willow (Salix sitchensis) in a riparian system in western Washington state. Several of the isolates grew well in nitrogen-limited medium. The presence ofnifH, a gene encoding one of the subunits of nitrogenase, was confirmed in several of the isolates including species ofBurkholderia, Rahnella, Sphingomonas, andAcinetobacter. Nitrogenase activity (as measured by the acetylene reduction assay) was also confirmed in some of the isolates. The presence of these diazotrophic microorganisms may help explain the ability of these pioneering tree species to grow under nitrogen limitation.  相似文献

FourRhizobium meliloti bacteriophages belonging to theMyoviridae family of tailed phages were studied. Burst sizes (50–100 virulent particles per infected cell), adsorption rates (2.6–4.1×10–9 ml/min), and latent periods (2–4 h) made this group heterogeneous. However, these characteristics indicate an important infection ability compared with other rhizobiophages. In vivo morphogenesis of phage CM1, studied by electron microscopy, seems to have steps similar to those of some other tailed phages such as coliphages.  相似文献

The presence of a large number of pollutants, including mutagenic agents in the environment is a problem of a major concern. Rapid progress in plant biotechnology, especially in the development of cell transformation methods, including the production of transformed roots – ‘hairy roots’ – has opened new possibilities to use transformed root cultures in plant bioassays for the evaluation mutagenic effects of different agents. We have used Crepis capillaris hairy roots for evaluation of cytogenetic effects of mutagenic treatment. Effects of maleic acid hydrazide (MH) and X-ray treatment were analysed in chromosomal aberration, sister chromatid exchange (SCE) and TUNEL tests. Comparison of cytogenetic effects in hairy roots and roots of seedlings showed a much higher sensitivity of hairy roots, which makes them convenient material for monitoring DNA damage after mutagenic treatment.  相似文献
