Graomys griseoflavus is a South American phyllotine rodent having a remarkable Robertsonian polymorphism which may have produced reproductive isolation between 2n=42–41 and 2n=38–34 karyomorphs. Analysis of nucleolar organizer region (NOR) locations both by silver staining (Ag-NOR) and in situ hybridization revealed that 2n=42 individuals exhibit highly variable Ag-NOR patterns, while specimens of the 2n=38–34 karyomorphic group showed a single Ag-NOR pattern. The latter animals underwent two NOR deletions in reference to the 2n=42 karyomorphs, one of which would be the consequence of a Robertsonian fusion and the other would be produced by the unequal crossing-over mechanism. The differential NOR homogenization supports the hypothesis that G. griseoflavus karyomorphs are evolving separately towards the acquisition of separate species status.  相似文献

The south American rodent Graomys griseoflavus exhibits a remarkable chromosome polymorphism as a consequence of four Robertsonian fusions. Focusing on the genetic analysis of the taxon, genome organization of all karyomorphs was studied at chromosome and molecular organization level. Cytogenetic (G, NOR and Re banding) and molecular (satellite and mitochondrial DNAs) events accompanying chromosome divergence allowed tracing a phylogenetic relationship among all karyomorphs. Available data led to propose that chromosome evolution of G. griseoflavus occurred in a non-random sequence of centric fusions, supporting the hypothesis of single origin for Robertsonian karyomorphs.  相似文献

Cytogenetic analysis was performed on twenty seven specimens of Akodon simulator simulator collected in three different localities of Tucumán Province, Argentina. Diploid number, chromosomal morphology and C and G banding patterns were studied. Eight different karyomorphs were found, with diploid numbers of 2n=38, 39, 40, 41, and 42. All individuals showed the same number of chromosomal arms (FN=42). G-bands enable to identify chromosomal pairs (1, 10, 11, 12, 13, and 14) involved in three centric fusions. C-bands revealed that the heterochromatin is located in centromeric regions of the telocentric and biarmed chromosomes. The present study allowed us to document a new example of a floating multiple Robertsonian fusion polymorphism. The data are discussed in relation to the occurrence of Robertsonian polymorphism in natural populations.
Resumen Polimorfismo autosómico múltiple en poblaciones de Akodon simulator simulator Thomas, 1916 de Tucumán, Argentina (Rodentia, Cricetidae). Se realizó un análisis citogenético de 27 especímenes de Akodon simulator simulator colectados en tres localidades diferentes de la provincia de Tucumán, Argentina. Se determinó el número diploide, la morfología cromosómica y los patrones de bandeo C y G. Se encotraron 8 cariomorfos differentes, con números diploides de 2n=38, 39, 40, 41, y 42. Todos los individuos presentaron el mismo número de brazos cromosómicos (FN=42). Las bandas G permitieron identificar los pares cromosómicos (1, 10, 11, 12, 13, y 14) involucrados en tres fusiones céntricas y las bandas C revelaron que la heterocromatina está localizada en las regiones centroméricas de los cromosomas telocéntricos y de los cromosomas bibraquiados. El presente estudio nos permite analizar un nuevo ejemplo de un polimorfismo robertsoniano. Los datos son discutidos en relación con la presencia de los polimorfismos robertsonianos en la naturaleza.

Bone-marrow karyotypes of 68 specimens of the subterranean octodontid rodent genus Ctenomys from 16 different populations of north east Argentina and one from Paraguay have been studied. A surprising variety of chromosome numbers was found, ranging from 2n=42 to 2n=70. Some of the karyomorphs are clearly assigned to named species by topotypy: C. conoveris 2n=50, FN=56; C. argentinus, 2n=44, FN=54; C. perrensi, 2n=50, FN=84; C. dorbignyi, 2n=70, FN=84; C. roigi, 2n=48, FN=80; C. yolandae, 2n=50, FN=78. Four populations of Corrientes Province similar in morphology to C. perrensi were found to be polymorphic and polytypic; they maintain the same FN=84, but diploid numbers increase from 2n=54 to 2n=58 from SW to the NE, thus suggesting Robertsonian rearrangements. In the middle of this cline, a stable karyomorph of 2n=62, FN=84 was found in two different populations, suggesting to belong to an undescribed species. Another karyomorph of 2n=42, FN=76 found in Curuzú Laurel, Corrientes, may also prove to represent another undescribed species. One karyomorph of 2n=52, FN=74, and another of 2n=56, FN=78 from Paraná and Ubajaý (Entre Ríos Province, Argentina) respectively are close to C. rionegrensis. The relationships among these karyomorphs is considered in light of data on sperm morphology. The hypothesis is advanced that karyotypic rearrangements among the FN=84 group may be the result of Robertsonian repatterning from a 2n=70 original widespread form. Fixation of chromosomal variants is correlated with patchy distribution and small size of unstable demes, and may or may not have resulted in reproductive isolation.  相似文献

We investigated chromosomal evolution in the African killifish species Chromaphyosemion bivittatum using a combination of cytogenetic and phylogenetic methods. Specimens from five populations were examined by conventional Giemsa staining as well as sequential chromosome banding with 4',6-diamidino-2-phenylindole (DAPI), chromomycin A(3) (CMA(3)), AgNO(3)-staining and C-banding. The cytogenetic analysis revealed variability in 2n ranging from 2n = 29 to 2n = 36 and in NF ranging from NF = 38 to NF = 44. Two populations showed an extensive chromosomal polymorphism (2n = 29-34, NF = 44 and 2n = 32-34, NF = 38-42, respectively). Karyotypic variability within and among populations was mainly due to Robertsonian translocations and heterochromatin additions, and chromosome banding patterns suggested that both types of chromosomal rearrangements were related to the presence of AT-rich heterochromatin. A phylogenetic analysis of the partial mitochondrial (mt) cytochrome b gene, using specimens from eleven populations, revealed a low degree of haplotype differentiation, which suggested a relatively recent divergence of the populations examined. This finding conformed to the low degree of morphological differentiation observed among C. bivittatum populations and might indicate fast chromosomal evolution. The high karyotypic variability may be caused by an elevated chromosomal mutation rate as well as certain aspects of the mating system and population dynamics of C. bivittatum facilitating the fixation of new chromosomal variants.  相似文献

We analyzed chromosomes of male and female individuals of Phyllodactylus lanei Smith, 1935 (Squamata, Gekkonidae) from Chamela-Cuixmala Biosphere Reserve, Jalisco state, Mexico. The karyotype constructed for these specimens is composed of 19 pairs of telocentric chromosomes (2n = 38, FN = 38). This karyotype, due to Robertsonian fusions/fissions, differs from the one previously reported in samples from the State of Guerrero, which probably belonged to a different subspecies (2n = 33−34, FN = 40−41). Moreover, a presumed ZW sex chromosome system was not confirmed in the presently studied individuals.   相似文献

1. Thermal freedom, defined as the physiological potential to make full spatiotemporal use of the seasonal thermal macroclimate, was assessed in Graomys griseoflavus, a cricetid rodent inhabiting the Chaco Province in Argentina. 2. The extent of the physiological thermal range (58 degrees C) was larger than the seasonal thermal range (30-50 degrees C), favoring Graomys resistance to low winter temperatures. The upper physiological thermal limit was almost coincident with the highest extreme of summer temperatures. 3. Graomys thermal freedom is facilitated by a high evaporative water loss, high metabolic capability, wide thermal neutral zone and a 4 degrees C lability of body temperature. 4. Behavioral responses (thermal avoidance) are utilized by this species, but with no apparent loss in thermal freedom.  相似文献

The chromosomes of Echiichthys vipera (Trachinidae) and Uranoscopus scaber (Uranoscopidae) were analyzed by means of various banding methods and fluorescence in situ hybridization (FISH) with telomeric and major rDNA probes, respectively. The karyotype of E. vipera was composed of 48 acrocentric chromosomes and NOR sites, as revealed by all detection methods, were situated pericentromerically on a single pair of middle-sized chromosomes. Blocks of constitutive heterochromatin were present in the pericentromeric regions of all pairs of chromosomes. The karyotype of U. scaber showed three karyomorphs: 2n = 30 (18 m + 12 a/st [m = metacentric, a = acrocentric and st = subtelocentric]), 2n = 28 (20 m + 8 a/st), 2n = 27 (21 m + 6 a/st). NORs, as revealed by FISH, were situated pericentromerically on a single pair of middle-sized chromosomes in spite of Ag-positive signals in the centromeres of all pairs of chromosomes. In this study, genetic differentiation between karyomorphs A (2n = 42) and D (2n = 39/40) of the wolf fish Hoplias malabaricus, which is comprised of several cryptic species that present a wide variety of diploid chromosome numbers and sex chromosome systems, resulting in the identification of seven distinct karyomorphs (A–G), was investigated using a combination of molecular and cytogenetic tools. Deep sequence divergences for both karyomorphs were observed and indicate a long period of reproductive isolation between karyomorphs A and D. Additionally, one individual with 61 chromosomes was identified, which, as far as is known, is the first case of natural triploidy resulting from the hybridization between these highly differentiated karyomorphs of H. malabaricus. Molecular and cytogenetic analyses revealed that this allotriploid specimen carries two sets of maternal chromosomes from karyomorph D (2n = 40) and one set of chromosomes from karyomorph A (n = 21). Moreover, ribosomal sites and active nucleolus organizer regions from both parental contributors were found in the triploid hybrid. Considering the significant genetic distances between karyomorphs A and D, one of the primary reasons for the lack of recurrent reports of hybridization in the H. malabaricus species complex may be due to post‐zygotic barriers, such as hybrid sterility or unviability.  相似文献

Two new species of Nippostrongylinae, Hassalstrongylus puntanus n. sp., and Stilestrongylus franciscanus n. sp. are described from the intestine of the grey leaf-eared mouse Graomys griseoflavus (Waterhouse, 1837) (Sigmodontinae) from the Province of San Luis, Argentina. Hassalstrongylus puntanus n. sp. is distinguished from the most closely related species H. dollfusi (Díaz-Ungría, 1963), a parasite of Mus musculus from Venezuela by longer rays 2, shorter rays 4, proximal half of the dorsal ray non-enlarged and a non-retractile female tail. Stilestrongylus franciscanus n. sp. is distinguished from the most closely related species S. flavescens Sutton & Durette-Desset, 1991, a parasite of Oryzomys flavescens from Argentina, by rays 2 and 3 diverging separately from common trunk of rays 2 to 6, and by right ray 3 arising from this trunk more distally than ray 6. Stilestrongylus azarai Durette-Desset & Sutton, 1985, a parasite of Akodon azarae from Buenos Aires, Argentina, was also found parasitizing G. griseoflavus, representing new host and locality records. The order Anguilliformes comprises 15 families, 141 genera and 791 fish species. Eight families had at least one karyotyped species, with a prevalence of 2n = 38 chromosomes and high fundamental numbers (FN). The only exception to this pattern is the family Muraenidae, in which the eight species analyzed presented 2n = 42 chromosomes. Despite of the large number of Anguilliformes species, karyotypic reports are available for only a few representatives. In the present work, a species of Ophichthidae, Myrichthys ocellatus (2n = 38; 8m+14sm+10st+6a; FN = 70) and four species of Muraenidae, Enchelycore nigricans (2n = 42; 6m+8sm+12st+16a; FN = 68), Gymnothorax miliaris (2n = 42; 14m+18sm+10st; FN = 84), G. vicinus (2n = 42; 8m+6sm+28a; FN = 56) and Muraena pavonina (2n = 42; 6m+4sm+32a; FN = 52), collected along the Northeastern coast of Brazil and around the St Peter and St Paul Archipelago were analyzed. Typical large metacentric chromosomes were observed in all species. Conspicuous polymorphic heterochromatic regions were observed at the centromeres of most chromosomes and at single ribosomal sites. The data obtained for Ophichthidae corroborate the hypothesis of a karyotypic diversification mainly due to pericentric inversions and Robertsonian rearrangements, while the identification of constant chromosome numbers in Muraenidae (2n = 42) suggests a karyotype diversification through pericentric inversions and heterochromatin processes.  相似文献

We conducted a pilot survey of genetic diversity among 37 karyotyped individuals of the black rat Rattus rattus (sensu lato) from six localities on the Japanese Islands, using complete gene sequences of mitochondrial cytochrome b (cyt b) and nuclear interphotoreceptor retinoid binding protein (IRBP). Our sampling included two previously documented karyotypic groups: 'Oceanian' with 2n = 38 and 'Asian' with 2n = 42. Cyt b sequences for most individuals clustered according to their karyotypic groups, with an average between-group divergence of 3.8%. One exception was that individuals from Kagoshima (Kyushu Island) showed 'Asian' karyotypes combined with a cyt b haplotype that differed by a single nucleotide substitution from the haplotype of the 'Oceanian' karyotypic group. Six IRBP haplotypes were identified. They belonged to three distinct IRBP lineages (I-III), with an average inter-lineage divergence of 1%. Among homozygous individuals, these lineages showed good association with the karyotypic groups: IRBP lineage I occurred only with 'Oceanian' karyotypes, while IRBP lineages II and III both occurred with 'Asian' karyotypes. Individuals from Kagoshima all possessed IRBP of 'Asian' lineages, despite the presence of an 'Oceanian' mitochondrial type. The Chichijima population (Ogasawara Islands) featured exclusively 'Asian' karyotypes and cyt b sequences, but various combinations of all three IRBP lineages. The Kagoshima and Chichijima populations thus provide strong evidence of viable hybridization and genetic introgression between the two karyotypic groups, but with variable genetic outcomes. Our results demonstrate the potential of combined analysis of karyotypes and mitochondrial and nuclear gene sequences to elucidate the complex dispersal and population history of the black rat.  相似文献

Rodents of the tribe Phyllotini represents one of the main radiations of South American Sigmodontini. Phylogenetic relationships among species of this highly diversified group are poorly known. In this paper we analyse evolutionary relationships among eight phyllotine species belonging to the genera Calomys , Graomys , Phyllotis and Eligmodontia , on the basis of allozymic polymorphisms. Most of the differences among species were in allele frequencies and not of allele class. Neighbour-joining and maximum likelihood methods place P. xanthopygus in the same group as E. typus and G. griseoflavus , in agreement with results obtained by several authors on the basis of morphological characters. Parsimony analysis of 0–9 coded data suggest that the genus Calomys is paraphyletic, but with a low bootstrap support. In the tree based on genetic distance data, the genus also appears as paraphyletic. The maximum likelihood method yields a tree where Calomys is monophyletic, but this phylogeny is supported by only two out of 78 alleles analysed. Calomys hummelincki and C. venustus occupy a basal position among Calomys species. Calomys musculinus and C. lepidus are the most closely related species of the genus, with C. laucha as sister to them. These relationships are strongly supported by bootstrap percentages.  相似文献

Two diploid numbers and five karyomorphs were found in ten specimens of Rhipidomys (Sigmodontinae, Rodentia) from three states in Brazil: 2n = 50 from Amazonas, and 2n = 44 from Mato Grosso and Bahia. CBG, GTG, and RBG-banding and Ag-NOR analyses were performed, as well as fluorescence in situ: hybridization with (T2AG3)7 probes. The new diploid number of 2n = 50 was associated with two different fundamental numbers (FN = 71 and 72) as a result of pericentric inversions and addition/deletion of constitutive heterochromatin. The samples from two localities (Aripuan? and Vila Rica) in the state of Mato Grosso shared 2n = 44 and FN = 52, but their karyotypes differed because of pericentric inversions. Although the single specimen from Bahia had the same diploid number as the samples from Mato Grosso, its karyotype and FN were completely distinctive. Karyological comparison of GTG-banding patterns revealed total homology between the karyotypes of the specimens from Bahia and Mato Grosso, implying the occurrence of 14 autosomal pericentric inversions. Homologies between ten of the autosomes in the karyotypes with 2n = 50 (FN = 72) and 2n = 44 (from Vila Rica, MT) were demonstrated. The differentiation between 2n = 44 and 2n = 50 involved five pericentric inversions, addition/deletion of constitutive heterochromatin in both autosomes and sex chromosomes, at least one Robertsonian rearrangement and other not detected rearrangements. Despite the remarkable number of rearrangements, interstitial telomeric sites (ITS) were not detected. Sex chromosomes also exhibited polymorphism in size and morphology.  相似文献

The kinetics of product formation by cytochrome P450 2B4 were compared in the presence of cytochrome b(5) (cyt b(5)) and NADPH-cyt P450 reductase (CPR) under conditions in which cytochrome P450 (cyt P450) underwent a single catalytic cycle with two substrates, benzphetamine and cyclohexane. At a cyt P450:cyt b(5) molar ratio of 1:1 under single turnover conditions, cyt P450 2B4 catalyzes the oxidation of the substrates, benzphetamine and cyclohexane, with rate constants of 18 +/- 2 and 29 +/- 4.5 s(-1), respectively. Approximately 500 pmol of norbenzphetamine and 58 pmol of cyclohexanol were formed per nmol of cyt P450. In marked contrast, at a cyt P450:CPR molar ratio of 1:1, cyt P450 2B4 catalyzes the oxidation of benzphetamine congruent with100-fold (k = 0.15 +/- 0.05 s(-1)) and cyclohexane congruent with10-fold (k = 2.5 +/- 0.35 s(-1)) more slowly. Four hundred picomoles of norbenzphetamine and 21 pmol of cyclohexanol were formed per nmol of cyt P450. In the presence of equimolar concentrations of cyt P450, cyt b(5), and CPR, product formation is biphasic and occurs with fast and slow rate constants characteristic of catalysis by cyt b(5) and CPR. Increasing the concentration of cyt b(5) enhanced the amount of product formed by cyt b(5) while decreasing the amount of product generated by CPR. Karyotypes of 39 Japanese raccoon dogs (NPV) which appeared in the literature and of 7 previously unreported specimens were examined. Thirty four individuals showed the standard karyotype 2K = 26M + 10A + (M)X + (A)Y + Bs (2n = 38 + Bs), where Bs are supernumerary chromosomes. The remaining 11 individuals had 2K = 25M + 12A + XY + Bs (2n = 39 + Bs) and one was 2K = 23M + 16A + XY + Bs (2n = 41 + Bs). The G- and C-banding analyses of both somatic and germ cells revealed that these karyotypes with odd numbers are heterozygous (M/A) for a single Robertsonian rearrangement of chromosomes 2, 5, 6, 8, or 11, and one is M/A heterozygous for three autosomes: 5, 6, and 11.  相似文献

Fifteen chromosome forms of Ellobius talpinus (from 2n = 31 to 2n = 54) were found in the small area in the Pamirs. Low-chromosome karyotypes evolved from 54-chromosomal ancestral form by Robertsonia centric fusions. The DNA reassociation kinetics of 34- and 54-chromosome forms of E. talpinus have been studied. For comparison DNA of E. lutescens (2n = 17) the karyotype of which seems to have arisen from 54-chromosome ancestor by Robertsonian and other types rearrangements was examined. Reassociation profiles of Ellobius DNA suggest the existence of several repeated sequences families with different frequences of repetitions. The reassociation curves of DNA from 34- and 54-chromosome forms were identical. These data indicate absence of changes in DNA molecular organization during the evolution of E. talpinus karyotypes by Robertsonian fusions. Comparative analysis of DNA reassociation kinetics of E. talpinus and E. lutescens showed identical characteristics of highly repeated sequences and of one from the three intermediate fractions, however Cot 1/2, complexity and repetitive frequencies of two intermediate fractions of E. talpinus and E. lutescens were different. It is possible that non-robertsonian rearrangements of E. lutescens karyotype affected only intermediate repetitions. The alternative explanation of these data is a simple divergence of repeated sequences during the evolution of E. lutescens DNA.  相似文献

The use of 5-deazaFAD T491V cytochrome P450 reductase has made it possible to directly measure the rate of electron transfer to microsomal oxyferrous cytochrome (cyt) P450 2B4. In this reductase the FMN moiety can be reduced to the hydroquinone, FMNH(2), while the 5-deazaFAD moiety remains oxidized [Zhang, H., et al. (2003) Biochemistry 42, 6804-6813]. The rate of electron transfer from 5-deazaFAD cyt P450 reductase to oxyferrous cyt P450 was determined by rapidly mixing the ferrous cyt P450-2-electron-reduced 5-deazaFAD T491V reductase complex with oxygen in the presence of substrate. The 5-deazaFAD T491V reductase which can only donate a single electron reduces the oxyferrous cyt P450 and oxidizes to the air-stable semiquinone, with rate constants of 8.4 and 0.37 s(-1) at 15 degrees C. Surprisingly, oxyferrous cyt P450 turns over more slowly with a rate constant of 0.09 s(-1), which is the rate of catalysis under steady-state conditions at 15 degrees C (k(cat) = 0.08 s(-1)). In contrast, the rate constant for electron transfer from ferrous cyt b(5) to oxyferrous cyt P450 is 10 s(-1) with oxyferrous cyt P450 and cyt b(5) simultaneously undergoing spectral changes. Quantitative analyses by LC-MS/MS revealed that the product, norbenzphetamine, was formed with a coupling efficiency of 52% with cyt b(5) and 32% with 5-deazaFAD T491V reductase. Collectively, these results suggest that during catalysis a relatively stable reduced oxyferrous intermediate of cyt P450 is formed in the presence of cyt P450 reductase but not cyt b(5) and that the rate-limiting step in catalysis follows introduction of the second electron.  相似文献

Karyotypes (chromosome number and shape) of four species of the subtribe Liarina were studied. The chromosome numbers and NF (Fundamental Number) in this group of species range from 2n Male = 33 (34) to 27(30): Liaromorpha buonluoiensis 2n Male = 33 (34), Sialaiana transiens 2n Male = 29 (34), Liara tramlapensis 2n Male = 29 (32), and Anelytra (Perianelytra) propria 2n Male = 27 (30). Cyto-taxonomy analysis indicates an intensive karyotype evolution among species belonging to three different groups of the genera. Differences of karyotypes are connected with Robertsonian fusion and tandem fusion in autosomes. Additionally, C-banding distribution and location of the NORs were studied.  相似文献

The South American group of rodents known as Graomys griseoflavus comprises two sibling species differing only in diploid chromosomal complement: G. griseoflavus (2n = 36, 37 and 38) and G. centralis (2n=42). Reproductive barriers comprising postzygotic as well as precopulatory mechanisms prevent gene exchange between these species. We have studied genetic polymorphism in two populations of G. centralis and four of G. griseoflavus by means of gel electrophoresis of enzymes and other proteins giving information on a total of 30 loci. Values of interspecific genetic identity were similar to those obtained for conspecific populations, suggesting that fixation of Robertsonian fusions would have occurred without significant bottlenecks. It would also indicate that the cladogenetic process must be relatively recent. FIS values showed no evidence of inbreeding. Fixation indices (FST) for the ancestral species showed a tendency to form demes with very low gene flow among them, while in the derived species such tendency was not shown. However, because of the characteristics of the region they occupy, populations are of moderate size, and genic flow is low. Lack of correlation between gene flow levels and geographical distance between population pairs would indicate a recent and fast colonization of its distribution areas by the derived species. It is possible that fixation of Robertsonian fusions occurred in a marginal deme of the ancestral species, e.g. in a parapatric geographical context.  相似文献
