Summary A morphometric analysis has demonstrated ultrastructural changes induced by hypoxia in the epithelial cells and the intracorpuscular nerve endings of the presumed chemoreceptive intrapulmonary neuroepithelial bodies (NEB) of neonatal rabbits.Acute hypoxia stimulates an exocytosis of epithelial dense-core vesicles (DCV) at the level of the morphologically afferent or sensory (type 1 a) intracorpuscular nerve endings of the NEB. Assuming the epithelial cells to be chemoreceptive, this phenomenon could represent a transduction of sensory stimuli.In the morphologically efferent or motor (type 2 and type 1 b) intracorpuscular nerve endings of the NEB, acute hypoxia causes a depletion of synaptic vesicles and an increase in the amount of membrane-bounded cisternae and multivesicular bodies, suggestive of an enhanced synaptic activity of these nerve endings. It is proposed that the chemoreceptor cells could thus in turn be modulated centrifugally by their efferent-like intracorpuscular nerve endings.It has been proposed in our earlier studies that the NEB probably are intrapulmonary chemoreceptors with local secretory activities, reacting to the composition of the inhaled air. By the release of serotonin and peptide substances they may produce a local vasoconstriction in hypoxically aerated lung areas, enabling an intrapulmonary regulation of the V/Q ratio. The present study provides evidence that, in addition to this local effect, NEB could generate centripetal nerve impulses via exocytosis of epithelial DCV at the afferent-like intracorpuscular nerve endings. At the same time they could be modulated by the CNS via their efferent-like intracorpuscular nerve endings.With respect to their innervation, numerous similarities appear to exist morphologically and functionally between the carotid body and the intrapulmonary NEB.  相似文献

Summary Fluorescence excitation and emission spectra of the formaldehyde-induced fluorophore of 5-hydroxytryptamine in a Sephadex model have been examined following exposure to hydrochloric acid or ammonia vapour. Exposure to hydrochloric acid vapour produced excitation spectra with broad maxima centred around 400 nm, whilst exposure to ammonia vapour intensified the maximum normally seen at approximately 450 nm relative to that seen at 400 nm. The emission maximum was generally broad and poorly defined following exposure to hydrochloric acid vapour; exposure to ammonia vapour had little effect on its location. Exposure of the formaldehyde-induced fluorophore in models containing 5-hydroxytryptamine to 300 nm irradiation caused a substantial shift in the position of the emission maximum; a concomitant increase in the fluorescence intensity was also observed. When the fluorescence present in duodenal enterochromaffin cells was examined after similar treatments, a number of differences in the response of the fluorophore were noted.  相似文献

Summary The ultrastructural localization of serotonin in neuroepithelial bodies (NEB) of neonatal rabbits was examined by use of the recently developed immunogold-staining method. In the granulated epithelial cells of NEB serotoninimmunoreactive material could be demonstrated only in dense-cored vesicles (DCV) of the first type. No immunoreactivity was observed in DCV of the second type. The present findings support the previously made suggestion that the corpuscular NEB cells contain two distinctive DCV types which react differently with the immunogold-staining procedure for identifying serotonin. NEB, known as intrapulmonary sources of serotonin (and peptide substances), may play a prominent role in lung physiology.  相似文献

Summary The formaldehyde-induced fluorescence (FIF) of the cytoplasm of individual developing neurons of the main pelvic ganglion was recorded microspectrofluorimetrically in order to follow changes in catecholamine (noradrenaline) content during development. For each ganglion, the fluorescence intensity profile was estimated and shown graphically as columns expressing percentage distribution of relative intensities in different intensity classes.During development, the number of weakly fluorescent neurons increases. Treatment with testosterone shifts the profile towards higher intensities in four- and six-week-old animals. Testosterone affected the main pelvic ganglion but not the superior cervical ganglion.The intensity of the cytoplasmic FIF, which correlates with the catecholamine (noradrenaline) content of the object tissue, showed a tendency to decrease during development. This change was not obvious by visual observation because of the increase in cell size and the toal bulk of the ganglion. Other possible factors affecting visual observation are discussed.  相似文献

Summary The spectral distribution of the formaldehyde-induced fluorescence emitted by model solutions and by adenohypophyses after intraperitoneal injection of l-dopa or 5-hydroxytryptophan was analyzed microspectrographically. Based on previously reported studies and on present findings, it seems that dopamine is stored in the strongly fluorescent cells after injection of l-dopa, and that a compound closely related to 5-hydroxytryptophan or serotonin is present in most of the cells after injection of 5-hydroxytryptophan. A non-specific, granular fluorescence appeared after 5-hydroxytryptophan and, to a lesser extend, l-dopa treatment. It probably represents autofluorescence of lysosomes, which are numerous in these circumstances.  相似文献

The effects of 5-hydroxytryptamine (5-HT; serotonin) and dopamine (DA) on tissue carbohydrate metabolism and haemolymph glucose levels in the freshwater prawn, Macrobrachium malcolmsonii, were investigated. Injection of 5-HT and DA produced hyperglycaemia in a dose-dependent and time-dependent manner, with DA being more effective than 5-HT. Interestingly, 5-HT and DA induced hyperglycaemia only in intact prawns but not in bilaterally eyestalk-ablated individuals. Total carbohydrate (TCHO) and glycogen levels decreased and phosphorylase activity increased in the hepatopancreas and muscle of intact prawns after being injected with 5-HT or DA. However, bilateral eyestalk ablation decreased haemolymph glucose and tissue phosphorylase activity and increased TCHO and glycogen levels of the hepatopancreas and muscle. Injection of 5-HT or DA did not cause significant changes in these variables in eyestalk-ablated prawns. It is hypothesized that 5-HT and DA induce hyperglycaemia in prawns by stimulating the release of crustacean hyperglycaemic hormone (CHH) from the X-organ sinus gland (XO-SG) complex located in the eyestalk.  相似文献

Endogenous and deuterium labelled acetylcholine (ACh) and choline (Ch) in the neostriatum were chemically assayed after radio-frequency lesions in the substantia nigra-ventral tegmental area and in the B7 or B8 5-hydroxytryptamine (5-HT) regions of the brainstem. Lesions in the substantia nigra-ventral tegmental region or in the B7 area, which provide dopamine (DA) and 5-HT afferents to the caudate-putamen nucleus, respectively, caused a decrease in 2H9-ACh synthesis, while endogenous levels of ACh and Ch were unchanged. A unilateral lesion of the B8 5-HT region, which projects in part to the substantia nigra, produced an increase in endogenous ACh levels, as well as a decrease in 2H9-ACh synthesis on the side ipsilateral to the lesion.On the basis of these data, we conclude that DA and 5-HT projections to the caudate-putamen nucleus have a net excitatory effect on cholinergic interneurons in that area. Furthermore, we suggest that the putative B8 5-HT projection is excitatory upon nigral neurons that, in turn, project to the neostriatum.  相似文献

Administration of 100 ng/hr 5-hydroxytryptamine for 48 hr significantly suppressed the PHA wattle response. 3. Summary By means of a histochemical reaction using formaldehyde vapour (Falck and Owman 1965), biogenic monoamines and precursor substances, i.e., L-DOPA, dopamine, noradrenaline, adrenaline, 5-hydroxytryptophan and 5-hydroxytryptamine, may be converted into fluorophores with specific spectral characteristics, i.e., the emission spectrum, excitation spectrum and fading curve. The registration and correction of the spectral properties and changes induced by acidification with hydrochloric acid vapour or treatment with ammonia vapour, of these formaldehyde-induced fluorophores, are performed by an automated microspectrofluorimeter, developed by modification of a Leitz MPV 2 system. This work deals with the instrumental configuration and certain methodological features in order to identify the fluorogenic biogenic monoamines and precursor substances in models and tissue sections. Registrations of excitation peak values, for the first time extended to a wavelength range from 240–460 nm, are discussed, which enable the calculation of peak ratio values 410/260, 380/320, 320/260 or 385/315, suitable as identification parameters for formaldehyde-induced fluorophores of biogenic monoamines and precursor amino acids.Dedicated to Mrs. Prof. Dr. M.H.A. Synopsis The influence of concentration of dopamine and noradrenaline on the spectral characteristics of their formaldehyde-induced fluorophore, together with the influence of duration of reaction with formaldehyde, has been investigated in a model system. No substantial differences between fluorescence spectra of the amine fluorophores were observed. Accordingly, the influence of hydrochloric acid and ammonia vapours on the fluorophores was investigated. A shift to shorter wavelengths in the excitation maximum of each fluorophore was observed after a brief exposure to hydrochloric acid vapour; more prolonged exposure resulted in a pronounced shift back to longer wavelengths in the case of noradrenaline but no substantial change was observed with dopamine. Brief exposure to hydrochloric acid vapour resulted in a substantial increase in the rate of fading of the noradrenaline fluorophore on exposure to exciting radiation. It is suggested that this offers a convenient way of differentiating the amines.  相似文献

Liquid chromatography with electrochemical detection has been used to determine endogenous dopamine (DA) and serotonin (5-HT) in a striatal crude synaptosomal fraction isolated by centrifugal methods. The electrochemical determinations are accomplished with a new type of electrode material, pressure annealed pyrolytic graphite. The analysis scheme permits direct quantitation of DA and 5-HT with minimal sample pretreatment. The distribution of endogenous 5-HT in each subfraction throughout the centrifugation procedure is found to be approximately parallel to that of endogenous DA. The release of DA and 5-HT from the crude synaptosome preparation exhibits the properties expected for stimulus-coupled, depolarization-induced release. The release of both neurotransmitters is Ca2+ dependent and is induced by high K+ or veratridine in the external medium. The latter is blocked by tetrodotoxin. Release is independent of ascorbic acid, but is dependent on the temperature of incubation.  相似文献

Endocrine cells (apudocytes) and neuroepithelial bodies (NEBs) were revealed in the lungs of rats of different ages by Grimelius' argyrophilic method. Solitary apudocytes were found among the bronchial epitheliocytes, they had the oval, columnar or triangular shape. NEBs comprise groups of argyrophilic cells, in some cases the penetration of neural fibers into the bodies and their branching in the terminals are observed. Apudocytes and NEBs in 1-, 7-, and 15-day-old rats are more numerous in the epithelium of small than big bronchi. In 21- and 30-day-old and in adult animals apudocytes and NEBs are very scarce. The elements under study are likely to take part in the early postnatal development of the rat lungs.  相似文献

The concentrations of dopamine (DA), 5-hydroxytryptamine (5-HT) and noradrenaline (NA) in the rat vas deferens divided in eight or four sections were determined by high performance liquid chromatography with electrochemical detection. Dopamine and NA had the same regional distribution; their concentrations were maximal near the prostatic end and decreased towards the epididymis. The concentration of 5-HT also decreased from the prostatic to the epididimal end, but 5-HT did not follow the same regional distribution as DA and NA. Reserpine (0.02 or 0.2 mg/kg, i.p., 24 hr) and 6-hydroxydopamine (2×80 mg/kg, i.v., 6 days) decreased the contents of DA and NA; the concentrations of both amines were modified to a similar extent. Reserpine also diminished the content of 5-HT. Pargyline (200 mg/kg, i.p., 2 hr) increased the concentration of 5-HT whilep-chlorophenylalanine (300 mg/kg, oral, 3 days) decreased the contents of the amine in some sections of the vas deferens. This study suggests that DA and NA co-exist in the same sympathetic neurons. Some of the 5-HT could be stored in mast cells as previously proposed, but the finding that tissue content of 5-HT changes after inhibiting the deamination or synthesis of the amine suggests that other source(s) of 5-HT distinct from mast cells exist in the rat vas deferens.  相似文献
