We analyzed a functional homologue of the swallow gene from Drosophila
pseudoobscura. The swallow gene of D. melanogaster plays an essential role in localizing bicoid mRNA in oocytes, and swallow mutant embryos show anterior pattern defects that result from the lack of localization of the bicoid morphogen. The pseudoobscura homologue rescues the function of swallow mutants when introduced into the genome of D. melanogaster, and its expression is similar to that of the melanogaster gene. The predicted pseudoobscura and melanogaster proteins are 49% identical and 69% conserved. The coiled-coil domain previously identified in the melanogaster swallow protein is strongly conserved in the pseudoobscura homologue, but the weak similarity of the melanogaster swallow protein to the RNP class of RNA-binding proteins is not conserved in the pseudoobscura homologue. These and other observations suggest a structural role for swallow in localizing bicoid mRNA, perhaps as part of the egg cytoskeleton.

Methionine in the form of free amino acids, small peptides and several proteins is transferred from ejaculate to females in Drosophila melanogaster and D. pseudoobscura. The male contribution is found mostly in the ovary and the sizes of the polypeptides transferred have been established. Some components of the ejaculate are rapidly transferred to the somatic tissues outside the ovary and reproductive system where they co-migrate with the yolk polypeptides. The incorporation of free methionine into polypeptides in non-reproductive tissues is higher in females that have been deprived of a source of protein prior to mating. The molecules transferred from ejaculate may have roles in regulating reproductive behaviour and may also be used as a nutrient source. We suggest that free amino acids in the ejaculate are rapidly utilised in protein synthesis in the somatic tissues outside the ovary and could provide a boost to yolk protein and hence egg production in females in nature, where nutrients may be in limited supply. 相似文献

Since the discovery of Drosophila pseudoobscura in the tropical highlands of the Colombian Andes during the 1960s, this population has been studied by many evolutionary biologists because of its geographical isolation from the main North American range of this species. We used five highly variable microsatellite loci (DPSX001, DPS2001, DPS3001, DPS3002, and DPS4001) to analyze the genetic structure of three Colombian populations and the genetic relationships with four North American populations. We found that the average heterozygosity was consistent among the three tropical Colombian populations (H = 0.665–0.675), but they had less variability than their North American counterparts. Nonetheless, the genie diversity found in the Colombian populations was higher than that found previously using other genetic markers. The average genie heterogeneity estimate among the Colombian populations (Rjr = 0.042), although statistically significant, was substantially lower than that found among the North American populations (RsT = 0.088). We identified alleles in the Colombian populations not reported in North American populations, suggesting further divergence between the populations. We estimated that the populations on the two continents diverged ca 80,000 years ago, consistent with independent sequence analyses of these populations but contrary to some suggestions in the literature. Finally, we estimated an average effective population size of the Colombian populations to be on the order of 100,000. 相似文献

Using 30,000 bp of anonymous sequence data, we note that dinucleotide repeat arrays appear to be much more common in Drosophila pseudoobscura than in D. melanogaster or D. simulans. Repeat arrays bearing five or more units are situated on average once every 3000 bp in D. pseudoobscura, and repeat arrays bearing ten or more units are situated on average once every 7500 bp. We did not detect an association between microsatellite presence and GC-content of flanking regions. 相似文献

We have isolated, characterized and mapped 33 dinucleotide, three trinucleotide and one tetranucleotide repeat loci from the four major chromosomes of Drosophila pseudoobscura. Average inferred repeat unit length of the dinucleotide repeats is 12 repeat units, similar to D. melanogaster. Assays of D. pseudoobscura and populations of its sibling species, D. persimilis, using 10 of these loci show extremely high levels of variation compared with similar studies of dinucleotide repeat variation in D. melanogaster populations. The high levels of variation are consistent with an average mutation rate of approximately 10(-6) per locus per generation and an effective population size of D. pseudoobscura approximately four times larger than that of D. melanogaster. Consistent with allozymes and nucleotide sequence polymorphism, the dinucleotide repeat loci reveal minimal structure across four populations of D. pseudoobscura. Finally, our preliminary recombinational mapping of 24 of these microsatellites suggests that the total recombinational genome size may be larger than previously inferred using morphological mutant markers. 相似文献

There are several models in the literature for predicting enteric methane (CH4) emissions. These models were often developed on region or country‐specific data and may not be able to predict the emissions successfully in every region. The majority of extant models require dry matter intake (DMI) of individual animals, which is not routinely measured. The objectives of this study were to (i) evaluate performance of extant models in predicting enteric CH4 emissions from dairy cows in North America (NA), Europe (EU), and Australia and New Zealand (AUNZ) and (ii) explore the performance using estimated DMI. Forty extant models were challenged on 55, 105, and 52 enteric CH4 measurements (g per lactating cow per day) from NA, EU, and AUNZ, respectively. The models were ranked using root mean square prediction error as a percentage of the average observed value (RMSPE) and concordance correlation coefficient (CCC). A modified model of Nielsen et al. (Acta Agriculturae Scand Section A, 63 , 2013 and 126) using DMI, and dietary digestible neutral detergent fiber and fatty acid contents as predictor variables, were ranked highest in NA (RMSPE = 13.1% and CCC = 0.78). The gross energy intake‐based model of Yan et al. (Livestock Production Science, 64 , 2000 and 253) and the updated IPCC Tier 2 model were ranked highest in EU (RMSPE = 11.0% and CCC = 0.66) and AUNZ (RMSPE = 15.6% and CCC = 0.75), respectively. DMI of cows in NA and EU was estimated satisfactorily with body weight and fat‐corrected milk yield data (RMSPE < 12.0% and CCC > 0.60). Using estimated DMI, the Nielsen et al. (2013) (RMSPE = 12.7 and CCC = 0.79) and Yan et al. (2000) (RMSPE = 13.7 and CCC = 0.50) models still predicted emissions in respective regions well. Enteric CH4 emissions from dairy cows can be predicted successfully (i.e., RMSPE < 15%), if DMI can be estimated with reasonable accuracy (i.e., RMSPE < 10%). 相似文献

New specimens of Cooksonia and Hostinella are described from the Bertie Group of Ontario and New York State, which is dated by faunas as latest Silurian (Přídolí). The rare plant fossils are unusual in that they are preserved in fine-grained, slightly argillaceous dolostones ('waterlime') rather than clastic rocks. At least two species of Cooksonia are present, one with ± globular sporangial morphology close to C. hemisphaerica Lang. Those with ellipsoidal/discoidal sporangia are compared with C. pertoni Lang, C . paranensis Gerrienne et al . and C. bohemica Schweitzer, the latter represented by a single specimen from the Přídolí of the Czech Republic. However, the paucity of specimens, which prevents assessment of taphonomic influences on shape, combined with the absence of any anatomical features and the gross morphological simplicity of the fossils, precludes specific assignment. Specimens of Hostinella include one in which apices and a lateral basal structure resembling a root are preserved. It is concluded that the Laurentian assemblage of Ontario and New York State is less diverse and disparate than coeval assemblages, which are also preserved in marine rocks. For the three species of Erioderma occurring in North America ( E . mollissimum (Samp.) Du Rietz, E. pedicellatum (Hue) P. M. Jørg. and E. sorediatun D. Gall. & P. M. Jørg), considerable range extensions are being reported together with ecological information that will help to understand the distribution of these lichens. Erioderma sorediatum has been recently discovered in North America by Sherwood and Pike (unpubl.), while E. mollissimum and E. pedicellatum are reported as new to Nova Scotia herein along with a number of new localities for the latter species in Newfoundland. Ongoing work on E. pedicellatum and its symbionts has been summarized and an attempt has been made to outline its possible postglacial history. It is postulated that the presently known ranges have been occupied by the lichen only within the past few thousand years (more recently in Europe than in North America), and that considerable migrations must have taken place prior to that. The sex‐ratio (SR), defined as the proportion of males, has been studied in three North American colonizing populations of Drosophila subobscura (Eureka, Davis and Gilroy). The proportion of sexes under laboratory conditions was studied using the one‐generation serial transfer technique in one‐ and two‐species populations, to infer whether biased SR affects the outcome when competing with Drosophila pseudoobscura, another member of the same group now in sympatry with D. subobscura in North America. The wild samples of D. subobscura yielded a significantly higher number of males than females during those months where the species is more abundant. However, there was no significant deviation in the 1 : 1 proportion of sexes in the descendants of D. subobscura at any of the experimental conditions. On the contrary, D. pseudoobscura produced a higher proportion of females which could be responsible for the exclusion of D. subobscura in laboratory competition experiments with overlapping generations. Thus, if sexes are equal at birth and survival is similar, the preponderance of males of D. subobscura in our wild collections could indicate greater activity and probably greater chance of dispersal of males versus females especially under favourable conditions. 相似文献

Heterozygosity at eight nuclear enzymatic loci and mitochondrial DNA control region (D-loop) sequence polymorphism was compared between North and South American nine-banded armadillos (Dasypus novemcinctus: Xenarthra, Dasypodidae). All markers revealed a striking genetic homogeneity amongst Texas, Louisiana, and Mississippi individuals, vs. the usual level of polymorphism for the French Guiana population. This may reflect a founder effect during colonization of North America. Occurrence of polymorphism in the D-loop microsatellite motif of North American armadillos suggests a recent recovery of mitochondrial variability. Phylogeographic analyses using Dasypus kappleri as outgroup provides evidence for a clear separation between North and South American control region haplotypes. 相似文献

This article presents a nucleotide sequence analysis of 500 bp determined in each of five X-linked genes, runt, sisterlessA, period, esterase 5, and Heat-shock protein 83, in 40 Drosophila pseudoobscura strains collected from two populations. Estimates of the neutral migration parameter for the five loci show that gene flow among D. pseudoobscura populations is sufficient to homogenize inversion frequencies across the range of the species. Nucleotide diversity at each locus fails to reject a neutral model of molecular evolution. The sample of 40 chromosomes included six Sex-ratio inversions, a series of three nonoverlapping inversions that are associated with a strong meiotic drive phenotype. The selection driven by the Sex-ratio meiotic drive element has not fixed variation across the X chromosome of D. pseudoobscura because, while significant linkage disequilibrium was observed within the sisterlessA, period, and esterase 5 genes, we did not find evidence for nonrandom association among loci. The Sex-ratio chromosome was estimated to be 25,000 years old based on the decomposition of linkage disequilibrium between esterase 5 and Heat-shock protein 83 or 1 million years old based on the net divergence of esterase 5 between Standard and Sex-ratio chromosomes. Genetic diversity was depressed within esterase 5 within Sex-ratio chromosomes, while the four other genes failed to show a reduction in heterozygosity in the Sex-ratio background. The reduced heterogeneity in esterase 5 is due either to its location near one of the Sex-ratio inversion breakpoints or that it is closely linked to a gene or genes responsible for the Sex-ratio meiotic drive system. 相似文献

The inverted gene arrangements of Drosophila pseudoobscura were used by Th. Dobzhansky in pioneering analyses of natural selection. Recent experiments have shed light on the mechanisms of selection contributing to the balanced polymorphism for the gene arrangements. In experimental populations, both major components of fitness, viability and fertility, are frequency dependent, and rare genotypes often have a selective advantage. Viabilities are also density dependent. The frequency dependence and density dependence of the fitness components are not universal. Some karyotypes are strongly influenced by frequency or density, some are slightly influenced, and some do not appear to be influenced at all. The role of heterozygote advantage in the selection on the gene arrangements is not clear. It is probably one important element in the overall selection, but viability and fertility do not always show a heterozygote advantage. Viability and fertility components of selection seem to be about equally important in changing inversion frequencies. Male mating success is an important component of selection in natural populations, and in one population rare male karyotypes have been found to have a pronounced mating advantage. 相似文献
