ABSTRACT Using a compartmentalized treatment technique, the role of arbuscular mycorrhizal fungi (AMF; Acaulospora scrobiculata) on arsenic (As) uptake and translocation in Brachiaria decumbens. Treatments consisted of a factorial arrangement of three As doses (0, 50, and 100 mg kg?1) and the presence/absence of AMF inoculates. In the absence of AMF, B. decumbens did not show As accumulation, indicating the probable presence of tolerance mechanism via As exclusion by the roots. B. decumbens plants showed high AMF colonization levels, especially in the arsenic treatments, with AMF improving shoot and root growth independent of As concentrations. Arsenic accumulation occurred only with AMF inoculation. Phosphorous uptake was reduced in B. decumbens roots in the presence of arsenic with and without inoculation of AMF. Results suggest that B. decumbens can be used in phytoremediation procedures when inoculated with A. scrobiculata, although pasture formation should be strictly avoided in contaminated sites.  相似文献

The purpose of this study was to evaluate the effects of arbuscular mycorrhizal fungi (AMF) on pacara earpod tree (Enterolobium contorstisiliquum) growth and phytoprotection in soil containing coal-mining waste. A greenhouse experiment was carried out with three inoculation treatment groups (non-inoculated, inoculated with Rhizophagus clarus, and inoculated with Acaulospora colombiana) in two substrates (0 or 30% tailings). After 90 days the seedlings were collected to quantify growth parameters, quality, mycorrhizal root colonization rate, and leaf content of chlorophylls and carotenoids. Macronutrients were quantified in the shoots; Cu, Zn, and Mn levels were measured in the shoots and roots; and glomalin content was measured in the rhizosphere. Colonization by A. colombiana (40%) promoted phytoprotection and better growth in seedlings planted in partial tailing substrate, due to the lower Cu (1.04 mg kg?1) and Zn (13.4 mg kg?1) levels in shoot dry mass and reduced translocation of these elements to the shoots. A. colombiana increased soil glomalin concentrations (2.98 mg kg?1) and the accumulation of nutrients necessary for synthesizing chlorophylls and carotenoids in the leaves. Colonization by R. clarus (81%) produced no phytoprotective effects.  相似文献

No previous studies have been conducted on the diversity and population of arbuscular mycorrhizal fungi (AMF) in relation to organically grown chili (Capsicum frutescens L.) in Thailand. This study was carried out to investigate the diversity and status of AMF populations at four organically managed farms in Ubon Ratchathani and Sisaket provinces. The effects of each AMF species on the growth and nutrient uptake of chili grown in sterile, organically managed soil were determined. Fourteen AM fungal taxa belonging to the genera Acaulospora (4 spp.), Entrophospora (1 sp.), Glomus (7 spp.) and Scutellospora (2 spp.) were found. Among these, Glomus was the dominant genus found at all sites, followed by Acaulospora. The spore density and root colonization of AMF on chili did not vary significantly among the sites. The effects of ten selected AMF species on the growth of chili showed that Gl. clarum RA0305 increased the growth, flowering, and fruit production of chili, and also increased the P uptake significantly, compared to non-mycorrhizal plants. This fungus showed the highest potential as a promoter of growth, flowering and yield in organically managed chili production.  相似文献

In this study, the colonization of arbuscular mycorrhizas (AM) and dark septate endophytes (DSE) in 140 specimens of 32 hydrophytes collected from four lakes and four streams in southwest China were investigated. The arbuscular mycorrhizal fungi (AMF) and DSE colonization in these hydrophytes were rare. Typical AM structures were observed in one of the 25 hydrophytic species collected in lakes and six of the 17 species collected in streams. Spores of 10 identified AMF species and an unidentified Acaulospora sp. were isolated from the sediments. The identified AMF came from the four genera, Acaulospora, Gigaspora, Glomus and Scutellospora . Glomus and G. mosseae were the dominant genus and species respectively in these aquatic environments. The presence of DSE in hydrophytes was recorded for the first time. DSE occurred in one of the 25 hydrophyte species collected in lakes and three of the 17 species collected in streams. Lead (Pb) contamination of soils is a widespread problem. Mycorrhizal inoculation and synthetic chelators such as ethylenediaminetetraacetic acid (EDTA) may be useful for improving phytoremediation efficiency in Pb-contaminated soils. A greenhouse experiment was performed to study the influence of inoculation with arbuscular mycorrhizal fungus (AMF), Glomus mosseae, and addition of EDTA on phytoremediation of Pb by sunflowers (Helianthus annuus) in a calcareous soil. The experiment was a completely randomized design in a factorial arrangement with five levels of Pb, two levels of mycorrhizal treatments, and two levels of EDTA. Inoculation increased root colonization as Pb levels increased, but the addition of EDTA decreased it. Shoot and root dry matter yields increased by inoculation; however, they decreased with EDTA and Pb levels in co-application treatments. Pb concentration in shoots was significantly higher than that in roots, indicating a translocation factor greater than 1. Inoculation or addition of EDTA significantly increased Pb in roots and its translocation to shoots. The uptake index (UI) value increased in co-application of EDTA and AMF and the individual application of them; it is, therefore, concluded that both AMF and EDTA are effective in phytoremediation of Pb by sunflowers in the studied soil.  相似文献

Three Arbuscular mycorrhizal fungi (AMF) from Glomus, Acaulospora and Scutellospora, and four plant growth promoting rhizobacteria (PGPR) isolates related to genera Streptomyces, Azotobacter, Pseudomonas and Paenibacillus were found to be effective in phytoremediation of Fe3+ contaminated soil where Pennisetum glaucum and Sorghum bicolor were growing as host plants. Co-inoculation of AMF and PGPR showed better results in comparison to either, AMF and PGPR under pot conditions. Both AMF and PGPR were able to produce siderophores. AMF and PGPR associated to P. glaucum and S. bicolor plants increased the extent of iron absorption. AMF and PGPR combination exhibited superior (p < 0.01) phytoremediation efficiency with P. glaucum compared to S. bicolor. These findings warrant further investigations of these synergistic interactions and large-scale in situ studies for bioremediation of iron-contaminated soils.  相似文献

The arbuscular mycorrhizal status of fifteen mangroves and one mangrove associate was investigated from 27 sites of three inundation types namely, diurnal, usual springtide and summer springtide. Roots and rhizospheric soil samples were analysed for spore density, frequency of mycorrhizal colonization and some chemical characteristics of soil. Relative abundance, frequency and spore richness of AMF were assessed at each inundation type. All the plant species except Avicennia alba exhibited mycorrhizal colonization. The study demonstrated that mycorrhizal colonization and spore density were more influenced by host plant species than tidal inundation. Forty four AMF species belonging to six genera, namely Acaulospora, Entrophospora, Gigaspora, Glomus, Sclerocystis and Scutellospora, were recorded. Glomus mosseae exhibited highest frequency at all the inundation types; Glomus fistulosum, Sclerocystis coremioides and Glomus mosseae showed highest relative abundance at sites inundated by usual springtides, summer springtides and diurnal tides, respectively. Spore richness of AMF was of the order usual springtide > diurnal > summer springtide inundated sites. The mean spore richness was 3.27. Diurnally inundated sites had the lowest concentrations of salinity, available phosphorus, exchangeable potassium, sodium and magnesium. Statistical analyses indicated that mycorrhizal frequency and AMF spore richness were significantly negatively correlated to soil salinity. Spore richness was also significantly negatively correlated to available phosphorus. The soil parameters of the usual springtide inundated sites appeared to be favourable for the existence of maximum number of AMF. Glomus mosseae was the predominant species in terms of frequency in the soils of the Sundarbans.  相似文献

The controlled disposal of tannery sludge in agricultural soils is a viable alternative for recycling such waste; however, the impact of this practice on the arbuscular mycorrhizal fungi (AMF) communities is not well understood. We studied the effects of low-chromium tannery sludge amendment in soils on AMF spore density, species richness and diversity, and root colonization levels. Sludge was applied at four doses to an agricultural field in Rolandia, Paraná state, Brazil. The sludge was left undisturbed on the soil surface and then the area was harrowed and planted with corn. The soil was sampled at four intervals and corn roots once within a year (2007/2008). AMF spore density was low (1 to 49 spores per 50 cm3 of soil) and decreased as doses of tannery sludge increased. AMF root colonization was high (64%) and unaffected by tannery sludge. Eighteen AMF species belonging to six genera (Acaulospora, Glomus, Gigaspora, Scutellospora, Paraglomus, and Ambispora) were recorded. At the sludge doses of 9.0 and 22.6 Mg ha−1, we observed a decrease in AMF species richness and diversity, and changes in their relative frequencies. Hierarchical grouping analysis showed that adding tannery waste to the soil altered AMF spore community in relation to the control, modifying the mycorrhizal status of soil and selectively favoring the sporulation of certain species.  相似文献

Arbuscular mycorrhizal fungi (AMF) were surveyed for species richness and abundance in sporulation in six distinct land uses in the western Amazon region of Brazil. Areas included mature pristine forest and sites converted to pasture, crops, agroforestry, young and old secondary forest. A total of 61 AMF morphotypes were recovered and 30% of them could not be identified to known species. Fungal communities were dominated by Glomus species but Acaulospora species produced the most abundant sporulation. Acaulospora gedanensis cf., Acaulospora foveata, Acaulospora spinosa, Acaulospora tuberculata, Glomus corymbiforme, Glomus sp15, Scutellospora pellucida, and Archaeospora trappei sporulated in all land use areas. Total spore numbers were highly variable among land uses. Mean species richness in crop, agroforestry, young and old secondary forest sites was twice that in pristine forest and pasture. fungal communities were dominated in all land use areas except young secondary forest by two or three species which accounted for 48% to 63% of all sporulation. Land uses influenced AMF community in (1) frequency of occurrence of sporulating AMF species, (2) mean species diversity, and (3) relative spore abundance. Conversion of pristine forest into distinct land uses does not appear to reduce AMF diversity. Cultural practices adopted in this region maintain a high diversity of arbuscular mycorrhizal fungi.  相似文献

The arbuscular mycorrhizal (AM) status of 67 plant species in a savanna community in the hot, dry valley of Jinsha River, southwest China was surveyed. It was found that about 95% of the plant species formed AM and 5% possibly formed AM. The composition of AM fungi (AMF) in the rhizosphere soils was also investigated. The AMF spore density ranged from 5 to 6,400 per 100 g soil, with an average of 1,530, and these spores/sporocarps were identified as belonging to six genera. Fungi belonging to the genera Glomus and Acaulospora were the dominant AMF. High densities of AMF spores in the rhizosphere soils, and the intensive colonization of the plant roots, indicated that plants grown in this valley-type savanna may be highly dependent on AM.  相似文献

The symbiosis between arbuscular mycorrhizal fungi (AMF) and mangrove plant species was investigated in two mangrove swamps in south China. AMF were mostly found in the form of hyphae and were commonly associated with all the mangrove species we investigated. Six AMF species belonging to the genera Glomus or Acaulospora were identified. Multiple step-wise linear regression analyses showed that hydrological conditions and phosphorus levels in the rhizosphere were the main abiotic factors affecting the colonization of mangrove species by AMF. A greenhouse experiment was conducted to evaluate the effects of AMF inoculation on the growth and nutrient uptake of a true mangrove plant species, Sonneratia apetala B. Ham. The inoculated AMF significantly improved growth, resulting in greater plant height, diameter at ground level and plant biomass, as well as increased absorption of N, P and K. These findings suggest that AMF play important roles in mangrove ecosystems.  相似文献

Alsophila firma is a deciduous tree fern considered as an emblematic species of Mexican tropical montane cloud forest (TMCF). We studied spores diversity, structure and colonization by arbuscular mycorrhizal fungi (AMF) within the roots of the Alsophila firma in rainy and dry season. Eighteen species of the genera Acaulospora (5), Gigaspora (4), Glomus (4), Funneliformis (2), Sclerocystis (2) and Scutellospora (1) were identified. The species F. constrictum, F. geosporum, Gigaspora albida, G. decipiens, Glomus microaggregatum and Sclerocystis coremioides are reported for the first time in TMCF. The dominant genera were Funneliformis and Acaulospora. In rainy season, a higher richness (H′ t0.005(2)9?=?4.78) and evenness (E) of AMF spores was recorded, compared to the dry season. However, the degree of colonization was statistically significant higher in the dry season. This study is the first to estimate the species richness of AMF associated with the rhizosphere of a fern in Mexico as well as for A. firma.  相似文献

采用土壤培养方法,研究了不同含砷水平土壤中添加外源磷或有机质对砷在板蓝根地下部和地上部累积与分配的影响。结果表明,在外源添加磷或者有机质的情况下,与自然土相比含砷土对板蓝根的生长有一定的促进作用;在自然土(13.4 mg/kg)中,外源磷没有明显影响板蓝根地下部对砷的累积,却显著降低了砷由地下部向地上部的转运,并且添加200 mg P2 O5/kg显著降低了砷在地上部的累积。然而,在含砷土(33.4 mg/kg)中,100 mg P2O5/kg处理显著降低了砷在地下部的累积,但随磷用量的增加反而促进了地下部砷的累积;在添加有机质试验中,10 g/kg的有机质显著降低了自然土中板蓝根地下部和地上部对砷的累积,并且砷的吸收能力也明显下降。在含砷土(23.4 mg/kg)中,添加5 g/kg的有机质不仅降低了砷在板蓝根中的富集,而且降低了其对砷的吸收能力,提高了砷由地下部向地上部的转运,但是随着有机质施用量增至10 g/kg,地下部砷含量及其吸收砷的能力均有一定程度的增大。因此,在砷水平较低的自然土壤上种植板蓝根添加200 mg P2O5/kg和10 g/kg的有机质是控制砷在该草药体内积累的适宜用量,而在砷水平较高的土壤上100 mg P2O5/kg和5 g/kg的有机质是降低板蓝根体内砷累积的适宜用量。  相似文献

Plant species capable of hyper-accumulating heavy metals are of considerable interest for phytoremediation, and differ in their ability to accumulate metals from environment. Using two brassica species (Brassica juncea and Brassica napus), nutrient solution experiments were conducted to study variation in tolerance to cadmium (Cd) toxicity based on (1) lipid peroxidation and (2) changes in antioxidative defense system in leaves of both plants (i.e., superoxide dismutase (SOD EC 1.15.1.1), catalase (CAT EC 1.11.1.6), ascorbate peroxidase (APX EC 1.11.1.11), guaiacol peroxidase (GPX EC 1.11.1.7), glutathione reductase (GR EC 1.6.4.2), levels of phytochelatins (PCs), non-protein thiols (NP-SH), and glutathione. Plants were grown in nutrient solution under controlled environmental conditions, and subjected to increasing concentrations of Cd (0, 10, 25 and 50 μM) for 15 days. Results showed marked differences between both species. Brassica napus under Cd stress exhibited increased level of lipid peroxidation, as was evidenced by the increased malondialdehyde (MDA) content in leaves. However, in Brassica juncea treated plants, MDA content remained unchanged. In Brassica napus, with the exception of GPX, activity levels of some antioxidant enzymes involved in detoxification of reactive oxygen species (ROS), including SOD, CAT, GR, and APX, decreased drastically at high Cd concentrations. By contrast, in leaves of Brassica juncea treated plants, there was either only slight or no change in the activities of the antioxidative enzymes. Analysis of the profile of anionic isoenzymes of GPX revealed qualitative changes occurring during Cd exposure for both species. Moreover, levels of NP-SH and PCs, monitored as metal detoxifying responses, were much increased in leaves of Brassica juncea by increasing Cd supply, but did not change in Brassica napus. These results indicate that Brassica juncea plants possess the greater potential for Cd accumulation and tolerance than Brassica napus.  相似文献

Hevea brasiliensis is one of the important economic trees with a great economic value for natural rubber production. Symbiosis between roots of H. brasiliensis and arbuscular mycorrhizal fungi (AMF) is widely recognized, and can provide a range of benefits for both of them. Hainan Island harbors is one of the largest plantations of H. brasiliensis in China, whereas the information regarding the diversity of AMF in the rhizosphere of H. brasiliensis on this island is scarce. The diversity of AMF species in the rhizosphere of rubber tree plantations in Hainan was investigated in this study. A total of 72 soil samples from the rhizosphere of H. brasiliensis RY7-33-97 were collected. These included 48 samples from plantations in 11 cities or counties that had been planted for 15–25 years, and 24 samples from a demonstrating plantation site of the China National Rubber Tree Germplasm Repository representing plantations with tree plantation ages from one to 40 year-old. Collectively, a total of 68 morphotypes of AMF, belonging to the genera of Archaeospora (1), Glomus (43), Acaulospora (18), Entrophospora (3), Scutellospora (2), and Gigaspora (1) were isolated and identified, as per morphological characteristics of spores presented in the collected soil samples. Glomus (Frequency, F = 100%) and Acaulospora (F = 100%) were the predominant genera, and A. mellea (F = 63.9%) and A. scrobiculata (F = 63.9%) were the predominant species. AMF species differed significantly among collected sites in spore density (SD, 290.7–2,186.7 spores per 100 g dry soil), species richness (SR, 4.3–12.3), and Shannon-Weiner index of diversity (H, 1.24–2.24). SD was negatively correlated with available phosphorus level in the soil; SR was positively correlated with soil total phosphorus content; and H was positively correlated with levels of soil organic matter and total phosphorus. Similarly, SD, SR, and H were also correlated with H. brasiliensis plantation age, and an increasing trend was observed up to 40 years. These results suggest that the AMF community was complex and ubiquitous in the island plantation ecosystems of H. brasiliensis, with high species abundance and diversity. Soil factors and plantation age dramatically affected AMF diversity at species level.  相似文献

以陕西延长县石油污染区常见的13种人工种植林木为材料,测定了各人工种植林木根际丛枝菌根(AM)真菌发育状况、污染土壤的理化性质、土壤酶活性和球囊霉素含量,探讨AM真菌在石油污染土壤生态修复中的作用。结果表明,13种林木均能形成AM,其定殖率平均为63.2%,孢子密度平均为1.93个.g-1干土,其中受污染程度最低的柠条AM真菌定殖率和孢子密度最高,分别为91.6%和4.73个.g-1干土;毛白杨、狼牙刺和刺槐的根际土壤养分(有机碳、碱解氮、速效磷)含量相对较高;各种人工种植林木的根际土壤球囊霉素含量、多酚氧化酶和过氧化氢酶活性随根际土壤石油烃污染浓度的增加而明显升高,其中刺槐、狼牙刺和酸枣根际土壤的过氧化氢酶和多酚氧化酶活性都较高,同时这3种林木的球囊霉素含量也较高。因此,林木根际土壤球囊霉素含量、多酚氧化酶和过氧化氢酶活性可以作为石油污染的敏感指标。  相似文献

明确丛枝菌根真菌(arbuscular mycorrhizal fungi, AMF)侵染率对喀斯特坡地坡位与物种的响应及其关键影响因子,是合理利用土壤AMF促进喀斯特植被恢复的前提。该研究在充分调查喀斯特峰丛洼地典型灌丛坡地环境背景信息的基础上,采用曲利苯蓝染色-镜检法检测并计算典型灌木黄荆(Vitex negundo)、红背山麻杆(Alchornea trewioides)和火棘(Pyracantha fortuneana)根系AMF侵染频度、侵染强度和丛枝丰度等侵染率参数。结果表明:(1)相同坡位火棘的侵染频度、侵染强度和丛枝丰度均显著低于红背山麻杆和黄荆;下坡位黄荆和火棘的侵染强度均高于中、上坡位,而中坡位红背山麻杆的丛枝丰度显著高于上、下坡位(P<0.05)。(2)物种显著影响AMF侵染频度、侵染强度和丛枝丰度,坡位显著影响AMF侵染强度,物种与坡位的交互作用仅对AMF丛枝丰度有显著影响(P<0.05)。(3)影响AMF侵染率的关键土壤因子是土壤深度和全钾含量(P<0.05)。因此,合理利用土壤AMF促进喀斯特地区植被恢复需要考虑地形与物种的选择,并应注重兼...  相似文献

Efficient N2-fixing Leguminosae nodulating bacteria resistant to As may facilitate plant growth on As-contaminated sites. In order to identify bacteria holding these features, 24 strains were isolated from nodules of the trap species Crotalaria spectabilis (12) and Stizolobium aterrimum (12) growing on an As-contaminated gold mine site. 16S rRNA gene sequencing revealed that most of the strains belonged to the group of α-Proteobacteria, being representatives of the genera Bradyrhizobium, Rhizobium, Inquilinus, Labrys, Bosea, Starkeya, and Methylobacterium. Strains of the first four genera showed symbiotic efficiency with their original host, and demonstrated in vitro specific plant-growth-promoting (PGP) traits (production of organic acids, indole-3-acetic-acid and siderophores, 1-aminocyclopropane-1-carboxylate deaminase activity, and Ca3(PO4)2 solubilization), and increased resistance to As, Zn, and Cd. In addition, these strains and some type and reference rhizobia strains exhibited a wide resistance spectrum to β-lactam antibiotics. Both intrinsic PGP abilities and multi-element resistance of rhizobia are promising for exploiting the symbiosis with different legume plants on trace-element-contaminated soils.  相似文献

In vitro breeding and somaclonal variation were used as tools to improve the potential of Indian mustard (Brassica juncea L.) to extract and accumulate toxic metals. Calli from B. juncea were cultivated on a modified MS medium supplemented with 10–200 μM Cd or Pb. Afterwards, new B. juncea somaclones were regenerated from metal-tolerant callus cells. Three different phenotypes with improved tolerance of Cd, Zn and Pb were observed under hydroponic conditions: enhanced metal accumulation in both shoots and roots; limited metal translocation from roots to shoots; reduced accumulation in shoots and roots. Seven out of thirty individual variants showed a significantly higher metal extraction than the control plants. The improvement of metal shoot accumulation of the best regenerant (3× Cd, 1.6× Zn, 1.8× Pb) and metal extraction (6.2× Cd, 3.2× Zn, 3.8× Pb) indicated a successful breeding and selection of B. juncea, which could be used for phytoremediation purpose.  相似文献
