A regioisomer of the known flavanolignan (-)-silandrin (3a), named (-)-isosilandrin (8a), was isolated from the fruits of a white-flowered variant of Silybum marianum L. populated in Hungary. Its structure was established both by spectroscopic methods and total synthesis, and its absolute configuration was determined by means of circular dichroism. This compound showed stronger inhibitory activity on the superoxide anion (O2*-) release by human polymorphonuclear leukocytes (PMNL) than (+)-silybin (1a,b).  相似文献

A novel polysaccharide named CBP-1 was isolated from the fruiting body of cultured Cordyceps militaris by alkaline extraction as well as anion-exchange and gel-permeation chromatography. Its structural features were investigated by a combination of chemical and instrumental analysis approaches, including partial hydrolysis, methylation analysis, HIO4 oxidation-Smith degradation, GC–MS, 13C NMR, HPAEC-PAD and FT-IR. The results indicated that CBP-1 has a backbone of (1  4)-α-d-mannose residues which occasionally branches at O-3. The branches were mainly composed of (1  4)-α-d-glucose residues and (1  6)-β-d-galactose residues, and terminated with β-d-galactose residues. In the in vitro antioxidant assay, CBP-1 was found to possess the hydroxyl radical-scavenging activity with an IC50 value of 0.638 mg/ml.  相似文献

The Gram-negative bacterium under study belongs to the genus Kaistella. It was isolated from a soil sample of the Haian Island in China, and it produces a lipophilic polysaccharide characterised by a branched hexasaccharide repeating unit, counting four 6-deoxy-alpha-l-mannose (Rha) residues, one 2-acetamido-2-deoxy-beta-d-glucose (GlcNAc) and a 2-acetamido-2,6-dideoxy-beta-d-galactose (FucNAc) unit. Three polysaccharide fractions (designated as Fr-I, Fr-II and Fr-III) were successfully purified from the crude exopolysaccharide (EPS) produced from submerged culture of Boletus aereus by gel filtration chromatography on Sepharose CL-6B. The size exclusion chromatography/multi-angle laser light scattering (SEC/MALLS) system showed that the average molecular weights (Mws) of these three fractions were 1.365 × 106, 1.048 × 105 and 2.471 × 104 g/mol, respectively. The SEC/MALLS also revealed that the molecular conformation of the Fr-I was a random coil, with Fr-II being a rigid rod in aqueous solution. Moreover, monosaccharide composition analysis indicated that Fr-I was mainly composed of glucose, while both of Fr-II and Fr-III were mainly composed of mannose and glucose. Then, FT-IR spectral analysis of the purified EPS revealed prominent characteristic groups. Furthermore, thermo gravimetric analysis (TGA) indicated the degradation temperature of the Fr-I (170 °C) was higher than those of Fr-II (156 °C) and Fr-III (155 °C). Finally, on the basis of the antioxidant activity test in vitro, Fr-I exhibited the highest antioxidant ability among these samples, which might be attributed to the monosaccharide composition and molecular weight in the EPS fraction.  相似文献

Structure of a capsular polysaccharide isolated from Salmonella enteritidis   总被引:1,自引:0,他引:1  
Salmonella enteritidis is a food-borne enteric human pathogen that can form a complex protective extracellular matrix. We describe here a component of this matrix which is distinct from other known salmonella extracellular polysaccharides such as cellulose and colanic acid. We have used glycosyl composition and linkage analysis, as well as 1D and 2D NMR spectroscopy to determine the structure of this polysaccharide. We propose that the primary saccharide in the S. enteritidis capsule has a branched tetrasaccharide repeating unit having the following structure: -->3)-alpha-D-Galp-(1-->2)-[alpha-Tyvp-(1-->3)]-alpha-D-Manp-(1-->4)-alpha-L-Rhap-(1-->. This structure is partially substituted on both tyvelose and galactose with a glucose-containing side chain. It further bears considerable similarity to the O antigen from this organism, a feature found in a number of other capsules from Gram-negative bacteria. In addition, we have detected fatty acids at levels that indicate the presence of a lipid anchor.  相似文献

Boletus edulis is a well-known delicious mushroom. In this study, three crude polysaccharides (BEPF30, BEPF60 and BEPF80) were isolated from the fruiting bodies of B. edulis with boiling water. Chemical and physical characteristics of the three crude polysaccharides were investigated by the combination of chemical and instrumental analysis methods. Their antioxidant activities were investigated in vitro systems including hydroxyl assay, superoxide radical assay, reducing power and chelating activity. Among these three polysaccharides, BEPF60 showed more significant reducing power and chelating activity; and highest inhibitory effects on superoxide radical and hydroxyl radical. These results indicated that polysaccharides extracted from B. edulis might be employed as ingredients in healthy and functional food to alleviate the oxidative stress.  相似文献

The structure of a new phosphoglycolipid from the halophilic Gram-negative bacteria Halomonas elongata ATCC 33173(T), Halomonas eurihalina ATCC 49336(T), Halomonas almeriensis CECT 7050(T), strain Sharm (AM238662), Halomonas halophila DSM 4770(T), and Halomonas salina ATCC 49509(T) was elucidated by NMR and mass spectroscopy studies. In all of the species examined, the polar lipid composition consisted of 1,2-diacylglycero-3-phosphorylethanolamine, 1,2-diacylglycero-3-phosphoryl-glycerol, bisphosphatidyl glycerol, and the new phosphoglycolipid PGL1. The structure of PGL1 was established to be (2-(alpha-D-glucopyranosyloxy)-3-hydroxy-propyl)-phosphatidyl diacylglycerol. C16:0;C18:1 and C16:0;C19:cyclopropane are the most abundant acyl chains linked to the phosphatidylglycerol moiety of each isolated PGL1. All of the species presenting the lipid PGL1 belong to Halomonas rRNA group 1, suggesting that the new phosphoglycolipid could be a chemotaxonomic marker of this phylogenetic group.  相似文献

We report here the isolation of a novel acid-labile yellow chromophore from the enzymatic digest of human lens proteins and the identification of its chemical structure by liquid chromatography-mass spectrometry, liquid chromatography-tandem mass spectrometry, and (1)H, (13)C, and two-dimensional NMR. This new chromophore exhibited a UV absorbance maximum at 343 nm and fluorescence at 410 nm when excited at 343 nm. Analysis of the purified compound by reversed-phase HPLC with in-line electrospray ionization mass spectrometry revealed a molecular mass of 370 Da. One- and two-dimensional NMR analyses elucidated the structure to be 1-(5-amino-5-carboxypentyl)-4-(5-amino-5-carboxypentylamino)-3-hydroxy-2,3-dihydropyridinium, a cross-link between the epsilon-amino groups of two lysine residues, and a five-carbon ring. Because this cross-link contains two lysine residues and a dihydropyridinium ring, we assigned it the trivial name of K2P. Quantitative determinations of K2P in individual normal human lens or cataract lens water-soluble and water-insoluble protein digests were made using a high-performance liquid chromatograph equipped with a diode array detector. These measurements revealed a significant enhancement of K2P in cataract lens proteins (613 +/- 362 pmol/mg of water-insoluble sonicate supernatant (WISS) protein or 85 +/- 51 pmol/mg of WS protein) when compared with aged normal human lens proteins (261 +/- 93 pmol/mg of WISS protein or 23 +/- 15 pmol/mg of water-soluble (WS) protein). These data provide chemical evidence for increased protein cross-linking during aging and cataract development in vivo. This new cross-link may serve as a quantitatively more significant biomarker for assessing the role of lens protein modifications during aging and in the pathogenesis of cataract.  相似文献

The water-soluble polysaccharide (AMP), with a molecular mass of 7.8 × 103 Da as determined by high-performance size-exclusion chromatography (HPSEC), was obtained from the fruiting body of Armillaria mellea. Methylation, Smith degradation, acetolysis, 1H and 13C NMR spectroscopy and acid hydrolysis studies were conducted to elucidate its structure. The results indicated that AMP consisted of a backbone composed of (1→6)-linked-α-d-glucopyranosyl, (1→2,6)-linked-α-d-glucopyranosyl and (1→6)-linked-α-d-galactopyranosyl residues in the ratio of 3:1:1, and terminated with one single terminal (1→)-β-d-glucopyranosyl at the O-2 position of (1→2,6)-linked-α-d-glucopyranosyl, on average, along the main chain. Preliminary tests in vitro showed that AMP has stimulating effects on murine lymphocyte proliferation induced by concanavalin A or lipopolysaccharide in a dose-dependent manner. It is a possible potential immunopotentiating agent for use in health-care food or medicine.  相似文献

Radix Paeoniae Alba is widely used in Chinese traditional medicine to treat various diseases such as gastrointestinal disorders, immunomodulatory, cancer, and other diseases. In this paper, a novel acidic polysaccharide RPAPS purified from Radix Paeoniae Alba was evaluated for its structural features and potential of immunomodulatory and antioxidant activities. RPAPS (molecular weight: 1.0× 105 Da) was mainly composed of α-(1 → 4)-Glcp, α-Arap, α-Galp, α-Rhap, β-D-Glcp, α-(1 → 6)-linked Glcp and GalA. Immunological tests indicated that RPAPS could improve RAW264.7 phagocytic activity and LPS-induced splenocyte proliferation. For antioxidant activities, RPAPS showed reducing power and DPPH scavenging activity in dose dependent. Moreover, RPAPS could significantly protect the PC12 cells from H2O2 damage. These data implied polysaccharides RPAPS had the potential to be novel natural antioxidative and immunopotentiating agents for using in functional foods or medicine.  相似文献

Three polysaccharide fractions (DDP1-1, DDP2-1 and DDP3-1) were successfully purified from the crude polysaccharide of Dendrobium denneanum by DEAE-Cellulose and Sephadex G-200 column chromatography. The average molecular weights (Mws) of these fractions were 51.5, 26.1 and 6.95 kDa, respectively. Monosaccharide components analysis indicated that DDP1-1 and DDP2-1 were composed of arabinose, xylose, mannose, glucose and galactose in a molar ratio of 1.00:2.82:57.11:140.82:7.76 and 1.00:1.62:1.18:77.5:7.79. DDP3-1 was composed of arabinose, mannose, glucose and galactose in a molar ratio of 1.00:1.03:8.84:2.00. On the basis of antioxidant test in vitro, DDP2-1 exhibited the highest antioxidant ability among these samples.  相似文献

A water-soluble polysaccharide (FUP-1) was obtained from Fritillaria ussuriensis Maxim through warm water extraction, ethanol precipitation, anion-exchange and gel-permeation chromatography. Structural analysis of this compound revealed that the average molecular weight of FUP-1 was 4.1×10(4)Da, and the major monosaccharide component was xylose, followed by glucose and galactose, at low levels. In vitro antioxidant assays showed that FUP-1 exhibited strong hydroxyl, superoxide anion and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging activities. During in vivo antioxidant testing, the administration of FUP-1 could largely attenuate the increase in d-gal-induced malondialdehyde (MDA) and renew the activities of superoxide dismutase (SOD) in serums as well as glutathione peroxidase (GSH-Px) and the total antioxidant capacity (T-AOC) in the livers of d-gal-treated mice. Overall, the results of the present study indicate that FUP-1 exerts antioxidant activity not only through its own radical-scavenging activity but also by boosting the enzymatic and nonenzymatic antioxidant defense system of the host.  相似文献

Immunomodulatory activity of polysaccharide isolated from Angelica sinensis   总被引:3,自引:0,他引:3  
The immunomodulatory activities of an Angelica sinensis polysaccharide (AP), purified from the fresh root of A. sinensis Diels, were investigated in vitro in relation to the specificity to immune cells. AP consisted of rhamnose, arabinose, mannose, glucose, galactose with the molar ratio of 1.00:4.54:2.98:11.09:7.45. Cell proliferation results showed that proliferation of total spleen cells, macrophages and T cells were promoted by the action of AP. The treatment of AP increased the production of IL-2 and IFN-γ, while that of IL-4 was decreased. RT-PCR analysis displayed that the IL-2 and IFN-γ gene expression were enhanced but the IL-4 gene expression was decreased. Some differences in cytokines secretion pattern were also detected, the expression of IFN-γ was rapidly augmented while that of IL-2 responded later. The flow cytometry results showed that the percentage of CD4+T cell in total spleen cells was remarkably increased by AP, while that of CD8+T cell was slightly decreased. In conclusion, AP has immunomodulatory activity by regulating expression of Th1 and Th2 related cytokines. The time–effect relation of cytokines response also suggests that macrophages and natural killer cells involved in nonspecific immunity were primary activated, and helper T cell were secondarily affected by AP.  相似文献

A novel water-soluble polysaccharide (POPS-1) was obtained from the fruiting bodies of Pleurotus ostreatus by hot water extraction, ethanol precipitation, and fractionated by DEAE-cellulose ion exchange chromatography and sepharose CL-6B gel filtration chromatography using an ATKA explore 100 purifier. The structure characterization and antitumor activity of the POPS-1 were evaluated in this paper. According to GC analysis, HPGPC, FT-IR, partial acid hydrolysis, periodate oxidation and Smith degradation, methylation and GC-MS analysis, the results indicate POPS-1 (M(w)=31 kDa) was composed of Man; Gal; Glc with a molar ratio of 1:2.1:7.9, it had a backbone of beta-(1-->3)-linked glucose residues, which occasionally branches at O-6. The branches were composed of (1-->3)-linked Glc, (1-->4)-linked Gal, (1-->4)-linked Man, and terminated with Glc and Gal residues. Cytotoxicity assay showed POPS-1 presented significantly higher antitumor activity against Hela tumor cell in vitro, in a dose-dependent manner, and exhibited significantly lower cytotoxicity to human embryo kidney 293T cells than Hela tumor cells compared with 5-Fu. The results suggest POPS-1 may be considered as a potential candidate for developing a novel low toxicity antitumor agent.  相似文献

Three new flavonoids-5,7,4'-trihydroxy-6-[1-hydroxy-2-methylbuten-2-yl]isoflavone (isogancaonin C), 7,2'-dihydroxy-4'-methoxyisoflav-3-ene (bolusanthin III), 6,6'-dihydroxy-4'-methoxy-2-arylbenzofuran (bolusanthin IV), in addition to eight known flavonoids; derrone, medicarpan, genistein, wighteone, lupiwighteone, gancaonin C, 7-hydroxy-4'-methoxyisoflavone and 7,3'-dihydroxy-4'-methoxyisoflavone were isolated from the root wood of Bolusanthus speciosus. The compounds showed strong antimicrobial activity against Escherichia coli, Bacillus subtilis, Staphylococcus aureus and Candida mycoderma. The isolated compounds also showed moderate to strong radical scavenging properties against DPPH radical with the highest activities shown by the 2-arylbenzofuran, the isoflav-3-ene and 7,3'-dihydroxy-4'-methoxyisoflavone in decreasing order.  相似文献

本研究对水溶醇沉法提取的鸡腿菇粗多糖脱蛋白方法进行了比较,最终确定Sevage法为最优的脱蛋白方法。经DEAE-纤维素52离子交换及Sephadex G-200分子层析柱分级、纯化,最终得到2个主要的多糖组分Ccp-I-A和Ccp-I-B。理化性质测定结果表明,二者均为白色絮状固体,能溶于水,不溶于无水乙醇、丙酮等有机溶剂;与斐林试剂,CTAB、硫酸-咔唑、碘-碘化钾及三氯化铁反应均为阴性。GC测定结果可知Ccp-I-A主要由甘露糖、葡萄糖和半乳糖组成,摩尔比为2.03∶9.52∶1;Ccp-I-B主要由岩藻糖和半乳糖组成,其摩尔比为1∶5.21。另外,Ccp-I-A和Ccp-I-B对DPPH和·OH显示出良好的清除能力,而且,相比于Ccp-I-B,Ccp-I-A的清除能力更高,当浓度为300μg/mL,其对DPPH和·OH的清除能力可分别达到72.1%和55.3%。  相似文献

用热水浸提、乙醇沉淀、Sevag法除蛋白分离得到水溶性莲子粗多糖。经十六烷基三甲基溴化铵(CTAB)沉淀,硼酸反应以及SephadexG-150层析纯化,得到3种莲子多糖SN1、SN2、SN3。用气相色谱、红外光谱、紫外光谱分析其化学结构。结果表明:SN1主要由葡萄糖、半乳糖组成,含a—D-吡喃葡萄糖环;SN2主链为葡聚糖、并含有其他4种单糖;SN3含7种单糖,并推测是一种酸性氨基多糖或蛋白多糖。体外抗氧化活性实验表明莲子多糖具有一定的清除自由基能力,清除·O2-的能力较强。  相似文献
