Seven groups of three sheep were given pelleted diets as follows: A, 97% hay + 3% molasses; B, C, D, 82% hay + 3% molasses + 15% dried pig faeces; E, F, G, 67% hay + 3% molasses + 30% dried pig faeces. In an attempt to counteract potential toxic effects of copper in the pig faeces (613 mg/kg dry matter), molybdenum was added to diets C and F at the rate of 90 mg/kg diet, and to diets D and G at the rate of 175 mg/kg diet. Sulphate was included in all diets at the rate of 1.08% of diet. Measurements were made of digestibility, copper retention, and plasma glutamic oxaloacetic transaminase (GOT) values during a 70-day feeding period, after which the animals were slaughtered.Dry matter digestibility coefficients were : diet A, 49.2; diets B, C, D, 44.4; diets E, F, G, 42.5, from which the coefficient for pig faeces alone is calculated to be < 30. Mean copper retention over the 70 days ranged from 210 mg for diet A to 1225 mg for diet G (excluding diet E which showed an aberrant retention of 53 ± 185 mg). Plasma GOT values fluctuated between diets and between sheep with a maximum value of 182 units/ml. Mean liver copper concentrations ranged from 718 mg/kg dry matter for diet A to 1740 mg/kg for diet E. Necrotic lesions occurred in some livers. Kidneys were apparently normal. There were no clear differences in copper status of animals receiving 15% versus 30% pig faeces except for a tendency for liver damage to be greater in the latter. There were no apparent effects of additions of molybdenum.The pig faeces were poorly utilized and, because of the high copper content, potentially hazardous to the health of the sheep.  相似文献

Aim: The mammalian intestinal microflora has been shown to impact host physiology. In cattle, intestinal bacteria are also associated with faecal contamination of environmental sources and human illness via foodborne pathogens. Use of wet distillers’ grains with solubles (WDGS) in cattle feed creates a gastrointestinal environment where some bacterial species are enriched. Here, we examine if a diet containing 40% WDGS results in fundamentally different microbial community structures. Methods and Results: The 20 002 16S r‐RNA gene sequences from 20 beef cattle were analysed using Sanger sequencing methods. At the genus level, Prevotella (Gram negative) and Anaerobacter (Gram positive) were the most frequently occurring bacteria in our beef cattle faecal samples. Diet‐associated differences in prevalence were noted for Prevotella but not Anaerobacter. Conclusions: Diet affects community structure. Faecal communities of co‐housed beef cattle are not identical. Significance and Impact of the Study: It is known that a diet of 40% corn‐based WDGS increases the generic Escherichia coli in the faeces and enriches E. coli O157:H7. The results from the current study suggest that in addition to previously observed changes in E. coli, the entire bacterial community structure is different for animals fed 40% corn‐based WDGS compared to a traditional corn‐finishing diet.  相似文献

Sodium hydroxide-treated or untreated wheat straw was included in a basal alfalfamaize diet at 0, 10, 20 and 40%. As the level of straw increased, the apparent digestibility of dry matter (DM) and organic matter (OM) by sheep, decreased linearly (P < 0.01), with a faster decrease (P < 0.05) for diets containing untreated straw. The digestibility of DM decreased by 0.22 and 0.41% and OM by 0.24 and 0.42% for treated and untreated straw diets, respectively, with each 1% increase of straw in the diets.Addition of treated straw increased (P < 0.05) digestibility of cell wall constituents (CWC), acid detergent fibre (ADF) and hemicellulose (HC). However, when untreated straw was added, the digestibility of HC was reduced, whereas the digestibility of CWC and ADF was dependent on the level of straw added. Increasing levels of NaOH-treated straw in the diets produced linear increases (P < 0.05) in digestibility of CWC, ADF and HC. However, significant (P < 0.05) linear or quadratic responses were not noted in the digestibility of CWC, ADF and HC with increasing levels of untreated straw in the diet. Apparent digestibility of crude protein was not affected by addition of either NaOH-treated or untreated straw to the diet.In general, although changes in nutrient digestibility of the basal component small, large changes in the digestibility of nutrients in the straw component were apparent and accounted for the major differences in digestibility of the diets. The absence of curvilinearity in the regression equations suggested that there were no associative effects.  相似文献

In the first of two experiments, 40 Friesian steers weighing initially 300 kg were fattened on diets containing barley and soya bean meal alone (C) or with inclusions of 40% untreated straw (WS40) or 40 or 60% alkali-treated straw (AS40 and AS60). The straw was coarsely milled into a horizontal mixer, where sodium hydroxide was applied as a 16% solution providing 80 kg NaOHt straw dry matter (DM). Intakes of DM (kg/d) were: C, 7.88; AS40, 9.67; AS60, 9.23; WS40, 8.60, and empty body weight gains (kg/d, in the same order) were 1.16, 1.03, 0.82 and 0.78.In the second experiment there were four diets, all containing 60% straw and 40% concentrates. The straw was coarsely milled (M) or chopped by forage harvester (C), and treated with alkali (A) or untreated (U). In a trial of latin square design, intake (kg DM/d) was; MU, 7.77; MA, 10.37; CU, 7.44; CA, 10.12. In a longer trial with five steers per diet, liveweight gains (kg/d, in the same order) were 0.70, 1.09, 0.71 and 1.18. The digestibility of DM for the four diets was 60.0, 72.7, 60.3 and 72.0%.The utilization of the energy of the diets, and the economic value of alkali treatment, are discussed.  相似文献

AIM: To determine the effect of different carbohydrate-based finishing diets on fermentation characteristics and the shedding of Escherichia coli and enterohaemorrhagic E. coli (EHEC) virulence genes in cattle faeces. METHODS AND RESULTS: The size of faecal E. coli populations and fermentation characteristics were ascertained in three experiments where cattle were maintained on a range of finishing diets including high grain, roughage, and roughage + molasses (50%) diets. Increased E. coli numbers, decreased pH and enhanced butyrate and lactate fermentation pathways were associated with grain diets, whereas roughage and roughage + molasses diets resulted in decreased concentrations of ehxA, eaeA and stx(1) genes, this trend remaining at lairage. In one experiment, faecal E. coli numbers were significantly lower in animals fed roughage and roughage + molasses, than animals fed grain (4.5, 5.2 and 6.3 mean log10 g(-1) digesta respectively). In a second experiment, faecal E. coli numbers were 2 log lower in the roughage and roughage + molasses diets compared with grain-fed animals prior to lairage (5.6, 5.5 and 7.9 mean log10 g(-1) digesta respectively) this difference increasing to 2.5 log at lairage. CONCLUSIONS: The type of dietary carbohydrate has a significant effect on E. coli numbers and concentration of EHEC virulence genes in faeces of cattle. SIGNIFICANCE AND IMPACT OF THE STUDY: The study provides a better understanding of the impact finishing diet and commercial lairage management practices may have on the shedding of E. coli and EHEC virulence factors, thus reducing the risk of carcass contamination by EHEC.  相似文献

AIMS: To determine the identity and composition of mesophilic Bacillus spp. in faeces sampled from feedlot cattle. METHODS AND RESULTS: Faecal samples from 10 feedlot cattle were analysed. The total aerobic spore count increased from 4.6 x 10(4) CFU g(-1) (before feedlotting, day 0) to 1.6 x 10(6) CFU g(-1) (feedlot for day 76). A total of 150 randomly selected spore isolates (60 each from days 0 and 76 cattle, 30 from feed) were speciated using a Bacillus group-specific PCR-amplified ribosomal DNA restriction analysis technique (Wu et al. 2006). At day 0, Bacillus subtilis and Bacillus cereus predominated with a prevalence of 58.3% and 26.7%, respectively, whereas three species, B. subtilis (50.0%), Bacillus licheniformis (27.6%) and Bacillus clausii (20.0%) predominated in day 76 faecal samples. Of these, only the first two species were present in feed samples at a frequency of 70% and 30% respectively. All B. cereus isolates on day 0, possessed at least one of three enterotoxin genes (nheA, nheB and nheC) but these were completely eliminated after a period of feedlotting. All isolates of B. licheniformis were genotypically heterogeneous according to pulsed-field gel electrophoresis analysis. CONCLUSIONS: Cattle faeces contain large numbers of Bacillus spores representing different mesophilic species. Stable faecal populations of particular Bacillus spp. mimicking those found in feed, were subsequently established by feedlotting. SIGNIFICANCE AND IMPACT OF THE STUDY: The results obtained and methods used in this study will help to investigate the indigenous Bacillus composition in the gastrointestinal tract of cattle and will further guide the administration of Bacillus probiotics.  相似文献

AIMS: To determine the production of reuterin by lactobacilli isolates from pig faeces and to evaluate their potential as probiotic bacteria. METHODS AND RESULTS: Twenty-eight of 165 lactobacilli isolates produced reuterin in the presence of glycerol. Six isolates yielding high levels of reuterin with respect to type strain Lactobacillus reuteri CECT 925T were identified as Lact. reuteri. They were able to survive at pH 3 and subsequent exposure to cholic acid or oxgall, and presented bile salt hydrolase and bacteriocin-like activities. CONCLUSIONS: Reuterin production is a frequently found trait among lactobacilli isolated from pig faeces. Selected Lact. reuteri isolates were able to survive at conditions likely to be encountered throughout the gastrointestinal tract. SIGNIFICANCE AND IMPACT OF THE STUDY: High yields of reuterin may be obtained from selected isolates of Lact. reuteri. Probiotic characteristics of isolates studied in the present work suggest their application in food and feed.  相似文献

Digestibility is a key parameter in the evaluation of feeds; however, the measurements on animals require heavy experimental trials, which are hardly feasible when large numbers of determinations are required – for example, in genetic studies. This experiment aimed at investigating the possibility to predict digestibility from NIRS spectra measured on faeces. A total of 196 samples were available from a digestibility experiment investigating the effects of age and genetic background of Large White pigs fed the same diet, rich in fibre (NDF=21.4% DM). Digestibility of dry matter (dDM), organic matter (dOM), nitrogen content (dN), energy (dE) and apparent digestible energy content (ADE) were calculated, as well as total N content of faeces (N). The faeces samples were submitted to reflectance NIRS analysis after freeze-drying and grinding. Calibration errors and validation errors were, respectively, 0.08 and 0.13% DM for total N in faeces, 0.97% and 1.08% for dDM, 0.79% and 1.04% for dOM, 1.04% and 1.47% for dN, 0.87% and 1.12% for dE and 167 and 213 kJ/kg DM for ADE. These results indicate that NIRS can account for digestibility differences due to animal factors, with an acceptable accuracy. NIRS appears to be a promising tool for large-scale evaluations of digestibility. It could also be used for the study of digestibility of different feeds, after appropriate calibration based on a wide range of feed types.  相似文献

The purpose of the present investigation is to compare the ability of chickens, ducks and geese to digest and utilise a diet containing a relatively large amount of barley (40%) rich in beta-glucan (18 g kg(-1)) and NSP (137 g kg(-1)) of which 35 g kg(-1) were soluble non-starch polysaccharides (NSP). The diets were offered to the birds (50 chickens, 40 ducks and 30 geese) in the period from hatching to 42 days of age. The digestibility of NSP was measured during the last week of the growth period using chromic oxide as an indigestible marker. Emphasis was on total NSP, soluble and insoluble NSP and their constituent sugar residues (rhamnose, fucose, arabinose, xylose, mannose, galactose, glucose and glucuronic acid). The degradation of NSP to short chain fatty acids (SCFA) was determined in the small intestine, caeca and large intestine. Although significant differences were found between species to the extent of degradation of individual soluble, insoluble and total NSP residues in the small intestine and caeca, the overall apparent digestibility of total NSP was similar (39-42%). On the basis of the digestibility of the NSP sugar residues and the formation of SCFA in the gut, the energy value of NSP was estimated on 2.8, 3.2 and 2.7 kJ g(-1) NSP ingested (P>0.05) in chickens, ducks and geese, respectively. On average, NSP contributed approximately 3.5% of metabolisable energy (ME) in the three poultry species.  相似文献

AIM: To study the diversity of commensal Escherichia coli populations shed in faeces of cattle fed on different diets. METHODS AND RESULTS: Thirty Brahman-cross steers were initially fed a high grain (80%) diet and then randomly allocated into three dietary treatment groups, fed 80% grain, roughage, or roughage + 50% molasses. Up to eight different E. coli isolates were selected from primary isolation plates of faecal samples from each animal. Fifty-two distinct serotypes, including nine different VTEC strains, were identified from a total of 474 E. coli isolates. Cattle fed a roughage + molasses diet had greater serotype diversity (30 serotypes identified) than cattle fed roughage or grain (21 and 17 serotypes identified respectively). Cluster analysis showed that serotypes isolated from cattle fed roughage and roughage + molasses diets were more closely associated than serotypes isolated from cattle fed grain. Resistance to one or more of 11 antimicrobial agents was detected among isolates from 20 different serotypes. Whilst only 2.3% of E. coli isolates produced enterohaemolysin, 25% were found to produce alpha-haemolysin. CONCLUSIONS: Diverse non-VTEC populations of E. coli serotypes are shed in cattle faeces and diet may affect population diversity. SIGNIFICANCE AND IMPACT OF THE STUDY: This study provides new information on the serotype diversity and phenotypic traits of predominant E. coli populations in cattle faeces, which could be sources of environmental contamination.  相似文献

Faecal samples of 52 hill cattle fed largely on oak leaves were screened for tannin-protein complex-degrading micro-organisms under different culture conditions using tannin-treated brain heart infusion agar medium. None of the animals were found to harbour tannin-protein complex-degrading enterobacteria. However, a fungus identified as Aspergillus niger van Tieghem having tannin-protein complex-degrading activity, was consistently isolated from the faeces of such animals. Optimum growth and sporulation was noticed under aerobic conditions at 30°C on Czapek Yeast Agar medium.  相似文献

A total of 92 enterococci, isolated from the faeces of minipigs subjected to an in vivo feeding trial, were screened for the production of antimicrobial substances. Bacteriocin production was confirmed for seven strains, of which four were identified as Enterococcus faecalis and three as Enterococcus faecium, on the basis of physiological and biochemical characteristics. The bacteriocins produced by the Ent. faecalis strains showed a narrow spectrum of activity, mainly against other Enterococcus spp., compared with those from the Ent. faecium strains showing a broader spectrum of activity, against indicator strains of Enterococcus spp., Listeria spp., Clostridium spp. and Propionibacterium spp. The bacteriocins of all seven Enterococcus strains were inactivated by alpha-chymotrypsin, proteinase K, trypsin, pronase, pepsin and papain, but not by lipase, lysozyme and catalase. The bacteriocins were heat stable and displayed highest activity at neutral pH. The molecular weight of the bacteriocins, as determined by tricine SDS-PAGE, was approximately 3.4 kDa. Only the strains of Ent. faecalis were found to contain plasmids. PCR detection revealed that the bacteriocins produced by Ent. faecium BFE 1170 and BFE 1228 were similar to enterocin A, whereas those produced by Ent. faecium BFE 1072 displayed homology with enterocin L50A and B.  相似文献

Digestibility studies were carried out with cattle to determine coefficients of digestibility for residual nutrients in wet cage layer excreta (CLE), and the effect of incorporating CLE in a maize silage-based diet, on nutrient digestibility. Two methods were used and compared — total collection, and the use of the recently identified internal marker, acid insoluble ash (AIASH).Digestibility coefficients for cage layer excreta were: dry matter, 74.0; organic matter, 73.1; crude protein, 76.8; energy 71.3; acid detergent fibre, 71.8%. Substituting CLE for soya bean meal or urea in the maize silage-based diet did not adversely affect nutrient digestibility. Comparable digestion coefficients were obtained with both the total collection method and the AIASH marker technique, confirming the results of others that the AIASH marker gives satisfactory estimates of nutrient digestibility.  相似文献

To investigate the relationships between plant composition in a complex diet mixture and intake by sheep, blood and faeces were analysed by Fourier transform mid-infrared (FT-IR) spectroscopy. Each of four mature Welsh Mountain ewes was maintained, for a period of 9 days, on one of three diets comprising a mixture of heathland plants containing either 100 g/kg (H100), 200 g/kg (H200) or 300 g/kg (H300) heather (Calluna vulgaris) in the fresh matter. Spectra of blood and faeces from each animal were acquired and the data analysed using principal component analysis (PCA) and hierarchical cluster analysis (HCA). We also investigated multinomial regression as an extension beyond the pattern recognition state. Animals offered the H300 diet could be consistently separated from animals fed on the H100 and H200 diets by PCA and HCA of plasma data, and these results were consistent with animals on the H300 diet consuming greater quantities of heather. Using faecal spectra, PCA and HCA discriminated between animals on the H200 diet and those on the H100 and H300 diets. Analysis of the plasma and faecal data by the multinomial model indicated that when PCs 4, 5 and 6 of the plasma data and PCs 3 and 4 of the faecal data were selected as covariates, leave-one-out cross-validation (LOO-CV) achieved an accurate dietary classification for 9 out of the 11 animals. In the present study, we have shown the ability to discriminate animals offered closely related diets on the basis of metabolic fingerprints of plasma and faeces.  相似文献

An HPLC method with diode array detection (DAD) is proposed for the detection of sulphamethazine (SMZ) residues in pig and cattle hair. Hair samples were extracted under alkaline conditions (NH4OH 0.2M for calf samples and NaOH 0.1M for piglet samples) and purified with a dual solid-phase extraction (SPE) cartridge system (reverse phase/strong-cation exchange). Recovery of SMZ in fortified samples varied from 70 to 85%, with a limit of quantification of 0.155 ng/mg. Residues of SMZ (7.2-59.2 ng/mg) were detected both in calf and piglet hairs after a therapeutic treatment with SMZ, while no interfering peak was observed in samples from untreated animals.  相似文献

AIMS: Identification and characterization of obligately homofermentative and facultatively heterofermentative strains of Lactobacillus spp. isolated from the faeces of pigs that had been raised under different conditions. METHODS AND RESULTS: The phenotypic relatedness of the isolated strains and reference strains were determined by numerical analysis of total soluble cell protein patterns and simple physiological and biochemical tests. Of the 23 strains isolated from faeces, nine were obligately homofermentative and 14 facultatively heterofermentative. The strains clustered at r > or = 0.61 with Lactobacillus amylovorus (seven strains), Lactobacillus crispatus (one strain), Lactobacillus plantarum (14 strains) and Lactobacillus intestinalis (one strain). CONCLUSIONS: Results obtained from the physiological and biochemical tests confirmed the identity of the isolates as determined by numerical analysis of total soluble cell protein profiles. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first report on the association of Lact. crispatus and Lact. intestinalis with the gastro-intestinal tract of pigs.  相似文献

A commerically prepared sample of pig hair was evaluated in semi-purified and practical diets as a source of essential amino acids for poultry. Amino acid (total) supplementation of pig-hair protein in semi-purified diets indicated that the amino acids were utilized by poultry and that satisfactory performance was obtained when the protein was supplemented with the essential amino acids that analysis had shown to be lacking. The performance of broiler chickens fed on diets containing up to 6% pig hair, and of growing pullets fed on diets containing up to 9% of this material, suggests that properly processed pig hair can be used in rations along with other keratin proteins.  相似文献

Effective pond production of tench [Tinca tinca (L.)] requires the use of formulated diets. However, insufficient information is available regarding the nutrition of this species. Therefore, for the first time, the digestibility of macronutrients by tench was investigated using a sieving method of faeces collection and chromic oxide. A pelleted experimental diet rich in untreated wheat starch revealed the following mean apparent digestibility coefficients (ADCs): dry matter 77.2%, crude protein 89.4%, crude fat 94.3%, ash 30.5%, nitrogen‐free extracts 78.2%, organic matter 83.3% and energy 85.1%. Generally, these ADCs indicate that the ability of tench to digest the macronutrients from pelleted feed correlates with reports of other fish species. However, the tench is more able to digest untreated wheat starch than are carnivorous and coldwater fish species. Compared with carp (Cyprinus carpio L.), the digestibility of untreated wheat starch is less in tench. The faeces sieving method as described in this paper is a useful procedure for further digestibility trials with tench. However, compared with the approach presented here, several adaptations are recommended, e.g. long adaptation times to the rearing facility, high stocking densities, usage of more waterproof food pellets and the addition of formalin to the collection pans.  相似文献
